The objective of this study was to isolate and identify native entomopathogenic fungi from different components of maize agroecosystem, and evaluate their virulence against Tribolium confusum, Rhyzopertha dominica, and Sitophilus zeamais, three insect pest vectors of aflatoxigenic fungi. Paecilomyces and Metarhizium were the most abundant genera isolated from the soil. Identification of fungal cultures by DNA extraction, amplification, and sequencing showed that all isolates macro- and micromorphological identified as P. lilacinus were Purpureocillium lilacinum. The isolate JQ926223 showed the lowest LT50 for T. confusum (4, 66 days) and R. dominica (9, 38 days), and the isolate JQ926212 demonstrated similar LT50 for the three insects evaluated between the range of 11, 7 to 14, 95 days. Maximum mortality rate was observed for the isolate JQ926223. The isolates of Purpureocillium lilacinum JQ926223 and JQ926212 may be considered good candidates for biologic control in the ecosystem of stored maize.  相似文献

In the present work, the insecticidal effect of two pirimiphos-methyl formulations [emulsifiable concentrate (EC) and capsule suspension (CS)] as surface treatment on concrete, plywood, galvanized metal, plastic, and ceramic tile was evaluated in laboratory bioassays against Sitophilus granarius (L.), Rhyzopertha dominica (F.), and Tribolium confusum Jacquelin du Val. Pirimiphos-methyl was applied at two-dose-rates, 0.01 and 0.1 mg/cm2, and insect mortality was assessed after 1, 2, 3, 4, 5, 6, and 7 days of exposure. After the final mortality count, alive individuals were transferred to untreated dishes, and delayed mortality was evaluated after an additional period of 7 days. In another series of laboratory bioassays, the residual efficacy of these two pirimiphos-methyl formulations on concrete was evaluated against S. granarius, R. dominica, and T. confusum. Pirimiphos-methyl was applied at two-dose-rates, 0.01 and 0.1 mg/cm2, on concrete dishes which were either kept in continuous darkness or exposed to light. A bioassay was initiated at the day of storage and every 4 weeks for 4 consecutive months (5 bioassays in total). Mortality of the exposed adult beetles was measured after 3 and 7 days of exposure. Both formulations were highly effective, with S. granarius being the most susceptible species. In general, mortality of the exposed beetles was not significantly affected by the surface type, with the exception of the significantly lower mortality counts of S. granarius adults on ceramic tile. CS appeared to be much more persistent on concrete than EC for all three species tested. Moreover, CS was not affected by light. In contrast, in some of the combinations tested, EC performance was poor in dishes that were exposed to light, in comparison with dishes that were in continuous darkness. We conclude that the surface type does not significantly and consistently affect S. granarius, R. dominica, and T. confusum mortality after surface application of the two pirimiphos-methyl formulations tested. Furthermore, CS can provide long-term protection when applied on concrete surfaces.  相似文献

Agrobacterium tumefaciens-mediated transformation (AMT) was used in order to create fluorescent conidia of Beauveria bassiana (Balsamo) Vuillemin (Ascomycota: Hypocreales). From the fungal transformants, one Enhanced Green Fluorescent Protein B. bassiana strain indicated stability (maintained the GFP characteristics) in fluorescent signal, while its pathogenicity did not differ significantly from the wild B. bassiana type. After exposure on wheat treated with the transformed conidia, fluorescence was evident on all body parts but especially on metasternon of adults of Tribolium confusum Jacquelin du Val (Coleoptera: Tenebrionidae) and on elytra in of Sitophilus oryzae (L.) (Coleoptera: Curculionidae). No significant differences were noted in fluorescence levels in the case of T. confusum between the transformed and the wild strain. However, the addition of diatomaceous earth (DE) increased attachment, especially at increased intervals after exposure. For S. oryzae, fluorescence of the transformed strain was decreased with the increased post-exposure time, but the simultaneous presence of DE increased fluorescence. The results of this study demonstrate that fluorescent conidia of B. bassiana can be utilized as a tool for the evaluation of parameters that affect the attachment of entomopathogenic fungi in insects?? cuticle.  相似文献

Toxicity and repellency effects of several pure compounds (of plant origin) against the confused flour beetle (Tribolium confusum) were investigated in a filter paper test. The compounds were α-pinen, β-pinen, 4-allyl-anisol, camphor, 1,8-cineol, eugenol, linalool, menthol, piperin, terpineol, thymol and zimtaldehyde. Five compounds (4-allyl-anisol, linalood, terpineol, thymol and zimtaldehyde) caused enough mortality of this insect to allow the determination of LD50 and LD95 values. Zimtaldehyde and 4-allyl-anisol showed the strongest toxicity effects with LD50 values of 0.04–0.05 μl/cm2 within an exposure period of 24 h. When the isolated fumigant effects were tested by separating the insects from the filter paper, the toxicity of zimtaldehyde (LD50=0.29 μl/cm2) was much lower than that of 4-allyl-anisol (LD50=0.10μl/cm2).  相似文献

In this study, gamma radiation was applied between 10–120 Gy at 10 dose levels for 2–4 days-old pupae; 10–150 Gy at 11 dose levels for 12–14 days-old adults ofT. confusum. All experiments were carried out in incubators maintained at 27±0.5°C and 70±5% relative humidity. The development of adults reared on different flours from irradiated pupae was not prevented by exposing these stages investigated doses. Rapidly developing confused flour beetle were more susceptible to killing by gamma radiation. Adults that emerged from treated pupa had elytral and other deformities. In barley flour, rate of deformed individuals were higher than the other rearing media. LD50, LD99,9 and SD99,9 values for wheat flour were high compared with both barley and maize flour. For adult treatment, there were differences in survival among rearing diets especially at doses of 40, 50 and 60 Gys. But there were no differences for LD50 and LD99,9 values at adult stage, indicating little influence of rearing medium on susceptibility of adults. The extensive use of synthetic insecticides and fumigants for control stored-product insects has led to the development of resistance. Essential oils from aromatic plants may provide proper alternatives to currently used insect control agents. Essential oils from 20 Egyptian plants were obtained by hydrodistillation. The chemical composition of the oils was identified by gas chromatograph/mass spectrometer. Fumigant and contact toxicities of the essential oils were evaluated against Sitophilus oryzae. The inhibitory effects of the essential oils on acetylcholinesterase and adenosine triphosphatases activities were examined. The oils were composed of monoterpene hydrocarbons (i.e., limonene, sabinene, β-pinene and γ-terpinene) and oxygenated monoterpenes (i.e., terpinen-4-ol, β–thujone, 4-terpineol, α-citral and 1,8-cineole) with the exception of the oil of Schinus terebinthifolius which was contained sesquiterpenes, and the oil of Vitex agnus-castus which contained similar amounts of monoterpenes and sesquiterpenes. In the fumigation assay, the oils of Origanum vulgare (LC50 = 1.64 mg/L air), Citrus lemon (LC50 = 9.89 mg/L air), Callistemon viminals (LC50 = 16.17 mg/L air), Cupressus sempervirens (LC50 = 17.16 mg/L air), and Citrus sinensis (LC50 = 19.65 mg/L air) showed high toxicity to S. oryzae. In the contact assay, the oils of Artemisia judaica, C. viminals, and O. vulgare caused the highest toxicity to S. oryzae with LC50 values of 0.08, 0.09, and 0.11 mg/cm2, respectively. The oil of A. judaica (I50 = 16.1 mg/L) invoked the highest inhibitory effect on AChE activity, while the oils of C. viminals and O. vulgare were the most potent inhibitors to ATPases activity with I50 values of 4.69 and 6.07 mg/L, respectively. The results indicate that the essential oils of A. Judaica, O. vulgare, C. limon, C. viminals, and C. sempervirens could be applicable to the management of populations of S. oryzae.  相似文献

Survival and growth were investigated for 10Eucalyptus species and 2 fodder tree species planted for salinity control in the wheatbelt of Western Australia. After two years of growth the trees were harvested to determine fodder biomass production and yields of cineole from the eucalyptus leaf oil. Subsequent harvests were conducted at three and five years after planting.At each harvest, biomass production from fodder species was greater than from most species of eucalypts. Biomass yields from eucalypts were variable, and there were no consistent trends in the productivity of the different species for the three harvests.Leaf cineole concentrations and cineole yields were low after two years of growth, but after three and five years cineole yields were generally higher from all species.E. kochii subspp.plenissima andkochii, E. horistes, E. radiata andE. angustissima produced consistently high cineole yields after three and five years. These species appear to have potential for the production of high grade eucalyptus oil in the wheatbelt of Western Australia.  相似文献

Field experiments have been carried out to evaluate the potency of the biological control agentBacillus thuringiensis (Dipel), the pyrethroid Fenvalerate and their combinations for the control of the lesser cotton leafwormSpodoptera exigua on soybeans in Qualubia governorate. Molasses was combined with all treatments to enhance their effectiveness. One spray application with either Dipel or Fenvalerate showed an obvious reduction in larval counts ofS. exigua, associated with a significant increase in the crop yield but in varying degrees, in correlation with the tested dose. Treatments with combinations of both biological and chemical insecticides at the lowest tested doses (62 g ofB. t. +50 ml of Fenvalerate/feddan) showed to be highly potential, and caused 2.8 fold increase in the crop yield. It may be recommended that a combination of both preparations may be used for the control ofS. exigua.  相似文献

Persistence of neem-based products against sweetpotato whitefly Bemisia tabaci Gennadius (Homoptera: Aleyrodidae) was tested in air conditioned rearing rooms and tropical netted greenhouses (GHs). Two commercial neem products, NeemAzal®-T/S (1% azadirachtin) and NeemAzal®-U (17% azadirachtin), were used. Foliar application, under room conditions at dose-rates of 7 and 10 ml NeemAzal®-T/S, induced an immature mortality of 32 and 44%, respectively, whereas 7 days post-application, under GH conditions, mortality rates declined to 5 and 7%, respectively. This result indicated rapid dissipation of the active ingredients. However, systemic application by soil drenching resulted in more stable effects under both laboratory and GH conditions. After soil drenching with solutions of 3.0 g NeemAzal®-U until 7-day, immature mortality declined from 88% for the first day to almost half (45%) by day-7 in the GH, and from 90% on first day to 64% by day-7 under laboratory condition. Similar response trends for B. tabaci were obtained for other parameters such as adult colonisation, egg deposition, and egg hatch. The loss of efficiency of the neem products was clearly related to dose-rate, methods of application, and environment (temperature and UV). Soil application is therefore, a convenient approach to achieve high efficiency and persistence with neem products under the conditions in tropical GH environments for whitefly management.  相似文献

The effectiveness of radio frequency (RF) treatment in the control of wood decay fungi (Gloeophyllum trabeum, Ganoderma lucidum, and Irpex lacteus) and sapstain fungus (Ceratocystis fimbriata) in red oak (Quercus spp.), poplar (Populus alba), and southern yellow pine (Pinus spp.) was evaluated in the laboratory as an alternative to methyl bromide (MB) treatment. Wood samples (15.5 x 10 x 10 cm) were inoculated with fungi from a 7-day culture by dipping them to a depth of one face deep (2 cm) into inoculum and incubating them at 25°C for 14 days. Identical wood samples were left uninoculated as controls. Subsequent to incubation, the wood blocks were exposed to RF radiation in an industrial 40-kW dielectric oven at temperatures between 60° and 70°C for 2 min. The test fungi were recovered and reisolated from all of the control wood blocks but not from RF-treated wood blocks. RF treatment resulted in complete inhibition of the fungus in 98%-100% of the wood samples. Moisture content loss (≥1%) was noted after wood had been exposed to RF treatment. Moisture content may be an important factor to consider with RF treatments. RF treatment can, therefore, potentially provide an effective and rapid quarantine treatment as an alternative to MB fumigation for certain pathogen-wood combinations. 为了解退耕还林工程对黄土高原生态脆弱区米脂县综合效益的影响,采用生态经济分析法、问卷调查法、修正的Bossel定位理论方法,2009年10~11月,对米脂县退耕还林工程实施前后的生态、社会和经济效益进行评价。研究结果表明,(1)自1998~2009年,退耕造林地土壤有机质(0~20 cm)平均含量和土壤有机碳密度分别比临近(﹤30 m)的坡耕地高出0.478%和0.574 kg/m2;(2)退耕还林经济净效益主要体现在退耕造林后劳动力转移劳务创收。即:69%的被调查农户家庭纯收入上涨了2~5倍,56%被调查农户种植业纯收入上涨0~1倍,44.5%的农户劳务收入占了家庭纯收入的50%以上,36.6%农户劳务收入超过了家庭纯收入的10%~20%,87.5%农户的退耕补贴占家庭纯收入的10%以下;(3)退耕还林提高了社会整体协调性0.81。国家实施退耕还林工程以来,初步改善了区域生态环境,整体提高了农户经济收入,优化了经济收入结构,但农业产业结构调整空间潜力很大。  相似文献

A study has been carried out on the presence of pesticide residues (deltamethrin, fenthion, formothion and their metabolites) on olives, oil and processing waste waters, after adulticide and larvicide treatments for the control ofDacus oleae (Gmel.). Deltamethrin was found to have a faster degradation rate than fenthion, leaving no traces on olives 18 days after treatment. Fenthion, on the contrary, was still present on the drupes at harvest (28 days after the treatment). Formothion was not found in definable quantities 3 days after treatment, while its metabolities, dimethoate and omethoate, were found on olives at harvest in levels of 4–23 ppb.  相似文献
