利用随机扩增多态DNA(RAPD)技术对野生14个和笼养18个绿孔雀(Pavomuticus)个体进行了种群遗传多样性分析。用23个随机引物对野生与笼养绿孔雀基因组DNA扩增,分别获得161和166个扩增片段。计算发现野生与笼养绿孔雀的种群内平均相对遗传距离分别是0.0555和0.1355,两种群间的平均相对遗传距离为0.1635;野生绿孔雀和笼养绿孔雀种群的Shannon多样性指数平均分别是0.4348和1.0163,表明有显著性差异。以上分析都显示野生绿孔雀的遗传多样性很低。利用UPGMA法聚类显示两个种群都是分别来源于两个家系,可据此进行绿孔雀的繁育管理。图1表3参5。 相似文献

The genetic diversity of the populations for 14 wild green peafowls (Pavo muticus) and 18 captive green peafowls was investigated by using the technology of random amplified polymorphic DNA (RAPD). Totally 161 and 166 amplified bands were obtained by using 23 arbitrary primers to amplify the genomic DNA of wild and captive green peafowls respectively. The results showed that the average relative genetic distance of the wild and captive green peafowls populations was 0.0555 and 0.1355, respectively, and difference of the average relative genetic distances between the two populations was 0.1635. The Shannon diversity index for the wild and captive green peafowl populations was 0.4348 and 1.0163, respectively, which means that there exists significant difference in genetic diversity between the two populations, and the genetic diversity of wild green peafowl was low. The two populations originated from two different families according to analysis by the UPGMA method. 以改进的CTAB高盐法提取油茶嫩叶总DNA,进行简单重复序列间扩增(ISSR)分析,分别测试了退火温度、模板DNA浓度、M g2+浓度、dNTP s浓度、T aqDNA聚合酶用量和是否加入去离子甲酰胺对反应结果的影响.油茶ISSR分析的最适反应体系为:在20μL PCR反应体积中,含20 ng模板DNA,2.5 mm o l.L-1M gC l2,0.2 mm o l.L-1dNTP s,1UT aqDNA聚合酶,0.5pm o l.μL-1引物,1%去离子甲基酰胺.扩增程序为:94℃预变性2 m in;94℃变性30 s,52℃退火30 s,72℃延伸90 s,反应38个循环;最后在72℃延伸7 m in. 相似文献

A micropropagation protocol was established for a medicinal plant Vitex negundo. Genetic stability of micropropagated plants was investigated. Multiple shoots were induced from nodal explants cultured on Murashige and Skoog (MS) medium with 0.53 μM naphthalene acetic acid (NAA) and 11.0 μM benzyl aminopurine (BAP) along with additives (ascorbic acid, 283.9 μM; citric acid, 130.1 μM; and arginine, 143.6 μM). Shoots were further multiplied by repeated transfer of the mother explant. The shoots were further multiplied on MS medium + 0.57 μM indole-3-acetic acid (IAA) and 6.6 μM BAP. The micropropagated shoots were pulse treated with 122.5 μM indole-3-butyric acid (IBA), in liquid MS medium and then transferred to autoclaved soilrite. These rooted ex vitro. Shoots were also rooted in vitro on a half-strength MS medium + 2.45 μM IBA. The survival rate of in vitro rooted plantlets was poor during hardening compared to ex vitro rooted plantlets. About 95% of the ex vitro rooted, hardened plantlets survived in the field. Genetic stability of micropropagated plants was tested by using 25 random amplified polymorphic DNA primers. 采用油茶上胚轴、下胚轴和子叶作外植体进行油茶组织离体培养诱导愈伤组织发生和不定芽分化的实验.实验结果表明,0.2%的HgCl2消毒处理油茶种胚外植体10 min效果较好;油茶上胚轴、下胚轴和子叶组织在MS+2,4-D2mg/L+KT0.5 mg/L的培养基上,愈伤组织诱导率分别为95.0%、66.7%和30.0%;在MS+6- BA2.5 mg/L+ KT0.1 mg/L+ NAA0.5 mg/L培养基上,不定芽诱导率分别是66.7%、33.3%和11.1%;以MS+6- BA 2 mg/L+ NAA 0.2 mg/L继代培养,上胚轴、下胚轴和子叶所形成愈伤组织芽增殖系数40天时为5.8、5.6和5.3. 相似文献
