The aim of this study was to investigate the effect of prehandling stress on the flesh quality of Atlantic cod (Gadus morhua). In order to stress the animal, water was reduced in the tank before a total of 30 fish were caught five at a time using a dipnet and held for 3 min, causing stress by hypoxia. This fish was compared with a control group (n=30) of fish exposed to anaesthetics directly in the tank. All fish were killed by a percussive blow to the head and exsanguinated and stored on ice before flesh quality was measured. Immediately after death and after ice storage for 3 and 8 days, 10 fish per group were measured for muscle pH, texture, fillet gaping, colour, drip loss and cathepsin D. Handling stress resulted in an initial increased fillet lightness, drip loss and decreased fillet shear force, although these differences did level off during ice storage. Stress caused by handling resulted in earlier onset of rigour mortis as compared with the control group, which reached maximum rigour tensions within 26 and 36 h postmortem respectively. We conclude that handling before harvest results in reduced time before entering rigour. After 8 days of ice storage, no effect of handling stress was seen on the muscle pH, flesh colour, fillet shear force, gaping score, drip loss or cathepsin D activity.  相似文献

Turbot and Atlantic halibut are highly valued fish species. However,very little is known about fillet shelf-life characteristics associated withboth species. Thus, fillet -tocopherol content and proximate compositionof wild turbot (1.5 kg) and Atlantic halibut (1.1 kg)caught off the south coast of Ireland and the north-west coast of Iceland,respectively, were investigated. In addition, the susceptibility of fillets, storedunder retail conditions, to lipid oxidation and colour change was studied.Proximate composition analysis showed that turbot had significantly highermoisture (P < 0.001) and lower protein (P < 0.001) contents compared toAtlantic halibut. Atlantic halibut incorporated significantly higher (P <0.001) levels of -tocopherol into fillets than turbot. Over 14 days ofstorage on ice, fillets from Atlantic halibut exhibited significantly lower (P =0.020) levels of lipid oxidation than those of turbot. However, malondialdehyde(MDA) concentrations were generally very low, never exceeding 0.6 gg–1 fillet. Turbot maintained a significantly higher (P< 0.001) pH over the storage period. The lightness (L* values) offillets from both species increased over 14 days of storage, but wassignificantly higher (P < 0.001) in Atlantic halibut than in turbot. Turbotdeveloped a relatively intense yellow colour during storage (decrease in hueangle and increase in b* values), whereas this was not the case forAtlantic halibut. The results of this study demonstrate that fillets of wildAtlantic halibut stored on ice, were less prone to lipid oxidation anddiscolouration than those of wild turbot. However, quality changes in turbotwere very small showing that both fish have tremendous shelf-life capacities interms of lipid oxidation. To determine the effect of maturation on flesh quality, 20 Atlantic halibut (Hippoglossus hippoglossus) consisting of mature and immature fish of both sexes were killed on site, exsanguinated and stored on ice. After 6 days of storage, the fish was gutted and filleted before colour, texture hardness and shear force were evaluated to determine the effect of maturation. Results show that mature males excreted black mucus from the skin during ice storage, while a greyish mucus was observed from immature fish. Mature fish had approximately 2% lower slaughter yield and 6% lower fillet yield compared with immature fish. The fillets from mature males were significantly harder compared with fillets from immature fish and the muscle structure proved to be stronger as an increased fracturability was observed in mature fish. In colour, fillets from mature fish proved to have a whiter appearance (L*) than immature fish, while no difference was seen in a* and b* values. We conclude that the physiological changes associated with maturation affects the end quality, and may be related to slower growth.  相似文献

Improved slaughtering procedures of farmed fish may provide sufficient time so that filleting can be performed pre‐rigor while the muscle pH is still high. Such filleting not only reduces fillet gaping but also lowers the transportation costs and makes fresh fillets available to the markets at an earlier stage. The aim of our work was to determine the weight reduction of the fillets due to liquid loss and to study the proteins and enzymes in the drip. After 11 days of cold storage, fillets of farmed Atlantic cod produced pre‐rigor had a weight loss of 10% and a fillet contraction of 19% while for fillets produced post‐rigor the values were 5% and 4% respectively. At the same time, approximately twice the amount of proteins had been lost from the pre‐rigor‐produced fillets. Sodium dodecyl sulphate polyacrylamide gel electrophoresis analysis showed that the proteins in the drip were similar to the sarcoplasmatic proteins extracted from the muscle. Specific analysis of proteolytic enzymes indicated that they are less stable in the expelled liquid than in the flesh during storage. The extensive loss of weight and proteins from pre‐rigor‐produced fillets during subsequent storage must be taken into account if such processing is considered for farmed cod.  相似文献

Discoloration profiles of dark muscle of skinned tilapia fillets were examined during iced storage after pretreatment with lactic acid (LA) or sodium carbonate (SC). During the subsequent storage, the a* values decreased gradually, and changed more rapidly when the pH was lower than 6.3. The fillet pretreated with 10% (v/v) LA exhibited the highest metmyoglobin formation ratio (MetMb%), followed by the fillet pretreated with 5% (v/v) LA, the control fillet, and the fillet pretreated with 10% (w/v) SC. The sample pretreated with 10% LA showed a marked decrease in the a* value. Discoloration of the control was not observed until the ninth day of iced storage, and no discoloration was observed up to the 11th day for the fillet pretreated with 10% SC. These fillet discoloration profiles were subsequently verified using the myoglobin (Mb) fraction prepared from the dark muscle. MetMb% of the Mb fraction gradually increased during storage, and this increase accelerated at pH values of <6.3. Discoloration of the Mb fraction also showed a similar tendency, and no significant discoloration was observed at pH values of >6.5. These results suggest that pH greatly affects the discoloration rate of the dark muscle of skinned fillet, and the critical pH for the accelerated autooxidation of tilapia Mb is in the range 6.3–6.5.  相似文献

Fish fillet quality has been shown to be influenced by the level of antioxidants in preslaughter diet. Thus, an experiment was conducted to study the effect of different levels of vitamin E and C on the fillet quality of market‐size reared turbot (Scophthalmus maximus). Turbot of a mean initial weight of 347 ± 20 g were divided into four groups and fed commercial turbot diets (60% protein, 12% fat), supplemented with α‐tocopheryl acetate (mg kg?1) and ascorbyl‐2 monophosphate (mg kg?1) at the following dietary levels: 500/100, 1000/100, 100/1000, 100/100 respectively. Over a dietary supplementation period of 15 weeks, fish were fed to satiation and reached a final mean weight of 916 ± 29 g. α‐Tocopherol levels increased significantly (P < 0.001) in tissue (i.e. muscle, liver, heart and kidney) of fish fed diets containing elevated levels of α‐tocopheryl acetate. In ice storage, fillets of these fish exhibited significantly lower (P < 0.001) levels of lipid oxidation, and showed significantly less (P < 0.001) colour deterioration (higher hue angle and lower chroma). Elevated dietary α‐tocopheryl acetate levels had a negative effect (P ≤ 0.001) on the concentration of ascorbic acid in muscle tissue. An increase in dietary vitamin C did not have any detectable effect on fillet quality. Prolonged feeding times had a negative effect on lipid oxidation (P < 0.001) and colour deterioration (P < 0.01). These results suggest that increased dietary α‐tocopheryl acetate could prevent colour deterioration and lipid oxidation of turbot fillets in retail storage on ice.  相似文献

An industrial and experimental electrical method for stunning farmed Atlantic cod in air and seawater (SW), respectively, were compared. The impacts of sedation with AQUI‐S? and exercise to exhaustion before electrical stunning were also assessed to monitor the possible depletion of rested muscle energy levels by electrical stunning. Stress (blood glucose, haematocrit, muscle pH, muscle excitability, high‐energy phosphates and rigor mortis) and flesh quality (fillet texture, colour, liquid leakage (LL), gaping, residual blood and K‐value) were assessed. For the industrial stunning method, an average of 41 V, 0.2 A dc was applied to individual cod for 18–27 s. For the SW method, a bipolar square wave current (170 Hz, 33% duty cycle) was applied for 5 s. After stunning, recovery was prevented by exsanguination in chilled SW. There were no differences (P>0.05) between the two stunning methods except for a higher ultimate fillet pH for cod stunned in air 8 days postmortem. Exercise before stunning depleted muscle energy levels at slaughter, increased LL and fillets had redder and darker flesh after storage on ice for 8 days. Electrical stunning (in air) of AQUI‐S?‐treated fish partly depleted muscle energy levels (pH 7.3, ATP 18.7 μmol g?1, PCr 70.1 μmol g?1). However, flesh quality was not affected. Unless pre‐rigor filleting is the chosen processing strategy, electrical stunning of cod seems to be a promising stunning method.  相似文献

The determination of post mortem muscle pH are often carried out in studies on how pre-slaughter handling influences the development of rigor mortis and the quality of the fish products. The analysis might be performed by inserting a pH-electrode into the intact flesh or using a suspension containing a minced muscle sample. Although it has been known for a long time that mincing muscle tissue accelerates glycolysis and breakdown of adenosine triphosphate (ATP), more recent studies quite commonly determine pH after mincing pre rigor muscle without including an enzyme inhibitor that arrests glycolysis. This may have resulted in anomalously low pre rigor muscle pH. The aim of the present work was to compare the two methods of pH determination in pre rigor fish muscle and to highlight the importance of including the enzyme inhibitor when homogenizing pre rigor fish muscle prior to pH analysis.  相似文献

The effects of filleting method, pre‐slaughter stress, storage and season on drip loss, water content, water‐holding capacity, rigour contraction and colour in Atlantic cod, Gadus morhua L., were examined. A total of 182 fish were sampled under commercial conditions. To test the effects of filleting method and stress, stressed (S) and unstressed (C) fish were filleted either pre (Pr) or post rigour (Po) at 1 and 48 h post mortem, receptively. The muscle pH significantly decreased from 0 to 144 h of storage in all groups. The onset of the rigour contraction was more pronounced in the Pr‐S group as compared with the Pr‐C group, but after 144 h of storage, no difference in degree of contraction was observed. Filleting method, stress and storage time influenced the colour of the fillets. Post‐rigour filleting caused a significantly increased in measured Lightness (L*) during storage. Stress caused a significant increase in measured redness (a*). No significant difference was found for water‐holding capacity. The water content was influenced by filleting method, where the Pr‐S group had a significantly lower water content compared with the Pr‐C groups; water content changed also according to season. The findings of this study show that stress, filleting method, time of storage and season had a significant effect on the drip loss, where a combination of all factors will determine the total loss.  相似文献

Assessment of flesh colour in Atlantic salmon, Salmo salar L.   总被引:1,自引:0,他引:1  
The degree of pigmentation in muscle of Atlantic salmon, Salmo salar L., fillets of fish that were fed eight diets fortified with 10, 20, 40, 60, 80.100, 150 and 200 mg astaxanthin kg?1 and a non-supplemented control diet from 3 to 21 months was assessed using different methods. A tristimulus colorimeter (Minolta Chroma Meter) was used to measure the colour composition of the fillets instrumentally. The colour was also determined using the Roche Colour Card for Salmonids. The concentration of astaxanthin in the muscle was measured by chemical analyses. All measurements for colour were done directly on the epaxial muscle anterior to the dorsal fin. The lightness factor (L *). the red/green chromaticity (a*), the yellow/blue chromaticity (b*) and the saturation C* of the colorimetric readings and the Colour Card scores were compared with the chemical analyses. The astaxanthin concentration in the flesh varied from 1 to 10 mg kg?1 and the visual appearance of the fillets varied from yellowish-white to red. The relationship between the a*, b* and C* values and the astaxanthin concentration in the muscle was non-linear. Non-linear regression lines were found between the a* value and the astaxanthin concentration in the flesh (r2= 0.974) and the b* value and the astaxanthin concentration in the flesh (r2= 0.984). The instrument was not able to detect differences in astaxanthin concentration at astaxanthin levels above 3-4 mg kg?1 using the presented method directly on the fillet. The instrument might be useful for rejecting groups of salmon with poor pigmentation. A good linear regression was found between the Colour Card score and the mean astaxanthin concentration in the flesh (r2 - 0.992). The Colour Card provided a better prediction of the astaxanthin concentration at higher astaxanthin levels than the Chroma Meter. None of the methods provided a satisfactory prediction of the astaxanthin concentration in the muscle of individual fish using the presented methods.  相似文献

Growth performance, muscle cellularity, flesh quality, and plasma ghrelin were investigated in 0+ and 1+ farmed Atlantic salmon (Salmo salar L.) from 40 g to 4.3 kg. Reduced meal frequency was introduced in both smolt groups from ~1.5 kg; one meal per second day (<5 °C) to one daily meal (>5 °C), while control groups were fed one to three daily meals. Results show that 0+ salmon had higher final fibre number and density, pigment content, red colour intensity, firmer flesh, and lower fillet fat content than 1+ salmon at 4.3 kg, affected by season and smolt type. Muscle fibre recruitment was an important determinant of fillet firmness and colour, possibly influenced by the prenatal temperature regime. Fish fed reduced meal frequency showed temporal reduced feeding ration, but growth performance was not compromised in any smolt groups at harvest. However, fillet fat, gaping, and colour decreased by less frequent feeding, with permanent effects in 1+ salmon for gaping and fat. Reduced meal frequency is therefore considered to be a promising tool for managing important flesh quality attributes in salmon without compromising growth performance. It is also suggested that ghrelin stimulates short‐term appetite, and perhaps also in the longer term.  相似文献

Trials were conducted to determine effects of AQUI-S™ sedation during harvest (rested-harvest) on physiological responses and fillet quality of channel catfish, Ictalurus punctatus. Rested-harvest is defined as application of an anesthetic immediately before harvest to reduce fish activity associated with a normal harvest. Doses of 25–35 ppm AQUI-S™ were effective for rested-harvest of catfish (loss of equilibrium in 3 to 10 min and 100% survival following recovery). Time to loss of equilibrium and time to recovery following sedation with 35 ppm AQUI-S™ increased as water temperature decreased from 30 °C to 10 °C. Catfish exposed to 25 ppm AQUI-S™, 35 ppm AQUI-S™, 100 ppm trincaine methanesulfonate, and 8 ppm metomidate had lower blood lactate, cortisol, and glucose and higher blood pH than unsedated fish exposed to a low-water stress. Rested-harvest (RH) catfish had higher muscle and blood pH, lower blood and muscle lactate, and higher muscle ATP levels than catfish exposed to a 45 min low-water stress. Rates of muscle pH decrease, ATP decrease, and lactate increase accelerated as storage temperature decreased from 15 °C to 5 °C in RH fish acclimated to summer temperatures (33 °C), conversely these rates accelerated as storage temperature increased from 5 °C to 15 °C in RH fish acclimated to winter temperatures (7 °C). Based on physiological response (higher muscle pH, lower blood lactate, delayed time to rigor), post-sedation euthanasia by CO2 was superior to post-sedation euthanasia by AQUI-S™ overdose (150 ppm), nitrogen gas, or electrical stunning. Compared to fillets from fish exposed to simulated industry transport conditions, fillets from RH/CO2 euthanised fish had higher pH 1 h post-slaughter, and less drip-loss and lower L and a color values during 7 days of iced storage. RH/CO2 and control fillets were not different for shelf-life based on bacterial counts. Rested-harvest with AQUI-S™ followed by CO2 euthanasia has potential to improve catfish fillet quality, but AQUI-S™ approval, development of rested-harvest strategies, and demonstration of economic benefits of rested-harvest will be required for adoption of rested-harvest to commercial catfish production.  相似文献

Production of triploid turbot is a relevant goal for the aquaculture industry due to the benefits associated with their sterility and growth. In order for a better understanding of the final quality of the species, muscle cellularity and quality parameters of the flesh were studied in seven diploid and seven triploid turbot after sexual maturation. White muscle cellularity was evaluated throughout white muscle cross‐sectional area from the epaxial left muscle section of each fish. To ascertain the flesh quality, several physico‐chemical parameters (moisture, ash, protein, total fat, fatty acids and minerals) were analysed, and textural mechanical properties (springiness, hardness, chewiness, cohesiveness, gumminess and adhesiveness) were determined objectively with a texturometer. Muscle cellularity was different between both groups, such that muscle fibre density was higher for diploid specimens (< 0.001) and muscle fibres diameters were higher for triploids (< 0.001). Both ploidy groups showed no significant differences in moisture, ash, protein and total fat content (> 0.05). All textural properties analysed showed no significant differences between both ploidy groups (> 0.05). In both ploidy groups, fibre density and fibre diameter showed no significant correlations (> 0.05) with textural parameters such as springiness, cohesiveness, chewiness and adhesiveness. Ploidy did not affect fatty acid composition, with the exception of eicosapentaenoic acid (C20:5ω3) which was higher in the triploid group. No significant differences (> 0.05) existed between diploid and triploid turbot in content of all minerals analysed. According to our results, the ploidy status determines significant variations on muscle cellularity but does not influence the flesh quality of the turbot.  相似文献

This study analysed the effect of cleaning intensity of the abdominal cavity and storage temperature from slaughter to the end of processing on the quality of farmed salmon (Salmo salar L.) fillets. These two parameters were manipulated in an experimental setup using in total thirty salmon with an average weight of 4.2 kg. The experiment was designed to imitate realistic scenarios in a normal production process in the Faroe Islands. The salmon stored at low temperatures had an average muscle temperature of 4.65°C, whereas the salmon stored at ambient temperature had an average muscle temperature of 11.27°C. After the salmon were gutted to remove all viscera except the kidney, the abdominal cavity was either rinsed lightly or meticulously cleansed of kidneys, all blood and bodily fluids. A wide range of quality and production parameters were measured either straight after cleaning or after the salmon had been stored in chipped ice at 1.5°C for 7 days. All measured parameters were analysed for possible correlations by principal component analysis (PCA). Blood and remains left in the abdominal cavity were shown to have a significant negative effect on fillet firmness (P < 0.01) and gaping (P < 0.01). The different storage temperatures between slaughter and gutting, tested in this experiment, did not significantly affect fillet firmness or gaping. However, the fillet colour showed significant negative correlation (P < 0.01) with the storage temperatures applied.  相似文献

Supranutritional dietary selenium (Se) has been demonstrated beneficial for fish health, however, its effects on fish flesh quality remain unclear. This study investigated the effects of supranutritional dietary Se on the flesh quality of rainbow trout (Oncorhynchus mykiss) and their potential mechanism. Fish were fed a basal diet supplemented with or without graded Se yeast for 10 weeks. Results showed that Se supplementation significantly increased fillet crude protein and enhanced fillet water‐holding capacity as well as fillet firmness. Supplementing with both 2 and 4 mg/kg Se significantly downregulated the expressions of two autophagy–lysosome‐related genes (autophagy‐related 12‐like and gamma‐aminobutyric acid type A receptor‐associated protein‐like 1) in fish muscle, while supplementing with 4 mg/kg Se also significantly downregulated the expression of two ubiquitin–proteasome‐related genes (muscle RING finger 2 and F‐box protein 25). Correlation analysis indicated that the improved fillet quality parameters were closely correlated with the expressions of these differentially expressed genes. This study revealed that dietary Se was effective for the improvement of rainbow trout flesh quality, and the improved fish flesh quality was associated with the inhibited protein degradation in fish muscle.  相似文献

Sufficient firmness is essential for consumer appreciation and the suitability for processing of fish fillets. The objective of this study was to investigate the effect of functional amino acids (AA) on fillet texture and muscle development of Atlantic salmon. Triplicate net pens of 105 g salmon were fed a standard diet, or the same diet with added 15 g/kg arginine or 15 g/kg glutamate during a 5‐month rearing period. The growth rate and FCR (0.91–0.92) showed no significant dietary effects (body weight 864–887 g). Glutamate supplementation resulted in delayed postmortem glycogen degradation (pH drop) and rigour development, along with improved fillet firmness and intercellular myofibre integrity. An in vitro study with salmon myosatellite cells showed that exogenous glutamine or arginine increased the expression of muscle growth markers (myog, tnnl2, myl) at both 8 and 16°C culture temperature. The expression of a marker for proteolysis (ctsb), myl and myog were highest for the glutamine treatment at 16°C. Significant interaction between exogenous AA and temperature indicated elevated AA requirement when growth is accelerated. It is concluded that AA from the glutamate family are vital for fillet firmness. The dispensable glutamine and glutamate appear more critical compared to arginine, particularly during high‐performance periods.  相似文献

Eels (Anguilla anguilla L.) were slaughtered using two procedures. The commercial method consisted of desliming the fish in dry salt followed by evisceration. The alternative method consisted of stunning and killing in water with a combination of electricity and oxygen removal. Eels slaughtered as commercially exhibited aversive reactions and consequently a higher level of muscular activity before death. In raw fillets, differences appeared clearly between the two batches. The alternative slaughter by electricity and gas led to redder, firmer flesh with a higher pH. Myofibrillar proteolysis, lipid oxidation and loss of freshness (as evaluated by K‐value) were reduced. After hot‐smoking, which is a process that greatly modifies the fish flesh properties, differences between batches were less pronounced. However, eels slaughtered by the alternative method were characterized by a higher pH and a redder colour of the dark muscle, a desirable property form a commercial point of view. When assessed by sensory difference tests, appearance seemed to be the only attribute that allowed panellists to discriminate the two batches. Overall, it is concluded that slaughter by electricity and gas improved the quality of raw and smoked eels compared with the commercial method.  相似文献

Fish fillet quality may be influenced by the antioxidant level in preslaughter diet. Thus, the effects of dietary α‐tocopheryl acetate supplementation and feeding time on the flesh quality of farmed Atlantic halibut Hippoglossus hippoglossus L. were investigated. Halibut of mean initial weight of 312 ± 12.3 g were divided into two groups and fed commercial diets, supplemented with different levels of α‐tocopheryl acetate at the dietary inclusion levels of 189 and 613 mg kg?1 diet. Fish were sampled after 6,9,12 and 24 weeks. Over the experimental period, they reached a final mean weight of 1320 ± 108.4 g. Tissue α‐tocopherol of fillet and liver was significantly affected by the levels of α‐tocopheryl acetate given with the diets (P < 0.001). In storage on ice, fillets of fish fed the diets high in α‐tocopheryl acetate exhibited significantly lower (P < 0.001) levels of lipid oxidation. The colour of fillets in all groups deteriorated slightly, but diet did not affect this process. Halibut fed the supplemented diets for longer periods were better protected against lipid oxidation (P < 0.001) and colour deterioration (P < 0.01) than those fed for shorter periods. However, after 9 days of storage, lipid oxidation levels were still extremely low [< 0.6 µg malondialdehyde (MDA) g?1 fillet], even in fillets of fish fed the low α‐tocopheryl acetate diet for a short period preslaughter. Different slaughtering methods tested at the end of the trial showed that percussive stunning can delay the onset of rigor mortis by 8–12 h compared with bleeding of the fish. These results suggest that halibut fillets have enhanced shelf‐life stability even at low doses of dietary α‐tocopheryl acetate, and that other factors in the antioxidant defence mechanisms of the species might play a major role in the prevention of lipid oxidation.  相似文献

The effects of dietary α‐tocopheryl acetate supplementation and different slaughtering methods were investigated on the flesh quality of farmed market‐size turbot Scophthalmus maximus (L). Turbot were divided into three groups and fed commercial diets, supplemented with different levels of α‐tocopheryl acetate at the following dietary inclusion levels: 72 (100), 547 (500), 969 (1000) (mg of α‐tocopheryl acetate kg?1 diet, analytical values with diet codes in brackets). After 5 months, fish (mean weight 1056 ± 19.7 g) from each dietary treatment were sampled, applying three different slaughtering methods: (A) bleeding in ice water; (B) thermal shock, no bleeding; (C) percussion followed by bleeding in ice water. The time course of rigor mortis was evaluated, using pH, rigor index and mechanical compression tests. The results showed that the three parameters corresponded very well. Percussive stunning resulted in higher initial post‐mortem pH (P < 0.01) and a significantly delayed onset of rigor mortis (P < 0.05). Diet significantly affected shelf‐life, with fillets from fish fed diets 500 and 1000 having lower TBARS (thiobarbituric acid reactive substances) numbers from day 2 (P < 0.001) and less colour deterioration from day 7 of storage on ice onwards (P < 0.05). These results suggest that an increase in dietary α‐tocopheryl acetate before slaughter as well as careful selection of the slaughtering method may greatly enhance the flesh quality of market‐size turbot.  相似文献
