Lolium spp., ryegrass, variants from Australia, Brazil, Chile, and Italy showing differing levels of glyphosate resistance were examined by (31)P NMR. Extents of glyphosate (i) resistance (LD(50)), (ii) inhibition of 5-enopyruvyl-shikimate-3-phosphate synthase (EPSPS) activity (IC(50)), and (iii) translocation were quantified for glyphosate-resistant (GR) and glyphosate-sensitive (GS) Lolium multiflorum Lam. variants from Chile and Brazil. For comparison, LD(50) and IC(50) data for Lolium rigidum Gaudin variants from Italy were also analyzed. All variants showed similar cellular uptake of glyphosate by (31)P NMR. All GR variants showed glyphosate sequestration within the cell vacuole, whereas there was minimal or no vacuole sequestration in the GS variants. The extent of vacuole sequestration correlated qualitatively with the level of resistance. Previous (31)P NMR studies of horseweed ( Conyza canadensis (L.) Cronquist) revealed that glyphosate sequestration imparted glyphosate resistance. Data presented herein suggest that glyphosate vacuolar sequestration is strongly contributing, if not the major contributing, resistance mechanism in ryegrass as well.  相似文献

Abstract

The effect of plant age and cold hardening on resistance to pink snow mould caused by Microdochium nivale was studied in perennial ryegrass. Resistance to M. nivale was estimated as relative regrowth after inoculation and incubation under artificial snow cover at 2°C. Resistance increased with increasing plant age. Cold hardened and unhardened plants of the same age displayed identical resistance. Preliminary studies indicate that expression of genes coding for the PR proteins chitinase and PR-1a increased during incubation of inoculated perennial ryegrass, but no clear difference in expression of these genes was found between plants of different ages, or in hardened versus unhardened plants.  相似文献

Esterases from a fenitrothion-resistant strain (VOSF) of the saw-toothed grain beetle, Oryzaephilus surinamensis (L.), are presumed to play a role in conferring resistance to malathion, fenitrothion, and chlorpyrifos-methyl. Colorimetric assays showed a significant positive correlation between increased resistance to fenitrothion in strains of O. surinamensis examined and elevated esterase hydrolytic activity to substrates of p-nitrophenyl acetate, alpha-naphthyl acetate, and beta-naphthyl acetate. Esterase zymograms showed different banding patterns between VOSF and an insecticide-susceptible strain, VOS48. A major esterase in the VOSF strain, not detected in VOS48, was purified and characterized by chromatographic and electrophoretic techniques. On the basis of SDS-polyacrylamide gel eletrophoresis, the molecular mass of the purified esterase from VOSF was 130 kDa and consisted of two 65 kDa subunits. A set of wheat cv. Chinese Spring (CS)-Lophopyrum elongatum addition and substitution lines has been analyzed for its potential to offer new sources of stripe rust resistance for wheat. The adult plants of these lines, together with CS-L. elongatum amphiploid and CS, were inoculated by new physiological races CYR-30 and CYR-31 of stripe rust in China. The resistance investigation indicated that chromosome 7E1 of L. elongatum was responsible for the adult resistance in CS background. Moreover, the expression of the adults plant resistance derived from chromosome 7E1 in wheat background was dependent on the wheat genotype. In addition, the different rust resistance and glutenin composition were observed in 2 CS-L. elongatum amphiploids, indicating the genetic diversity existed in different origins of L. elongatum.  相似文献
