Silver carp Hypophthalmichthys molitrix (Valenciennes) were co‐cultured with channel catfish Ictalurus punctatus (Rafinesque) in 0.4 ha earthen ponds to determine the impacts of carp grazing on pond phytoplankton communities and cyanobacterial off‐flavours in catfish. Carp were stocked at densities of 0, 75, or 250 fish ha?1 in seven replicate ponds per treatment. The mean chlorophyll a concentrations (a measure of phytoplankton standing crop) steadily increased in all treatments from about 100 μg L?1 in April to more than 400 μg L?1 by mid‐October. Silver carp had no affect (P>0.1) on chlorophyll a concentrations across all sampling dates (April though October) or for sampling dates late in the growing season (August–October) when the prevalence of cyanobacterial off‐flavours among catfish populations is usually greatest. Silver carp did not eliminate odour‐producing cyanobacteria from pond phytoplankton communities: on sampling dates in September and October, three to six ponds in all treatments contained populations of the odour‐producing cyanobacteria Oscillatoria perornata, Anabaena spp., or both. Failure of silver carp to eliminate odour‐producing cyanobacteria resulted in a relatively high incidence in all treatments of ponds with off‐flavoured catfish. On sampling dates in September and October, catfish in three to five ponds in each treatment were tainted with either musty (2‐methylisoborneol) or earthy (geosmin) off‐flavours. The presence of silver carp had no obvious effect on off‐flavour intensity: on each sampling date, at least three ponds in each treatment contained catfish described as distinctly to extremely off‐flavored. Apparently, hypertrophic conditions in catfish ponds overwhelm the effect of silver carp grazing at the low carp densities used in this study.  相似文献

The study was conducted to investigate the effect of dietary protein on nitrogenous excretion in the form of ammonia in Indian major carp, Labeo rohita (Ham.), fingerlings (3.0 ± 0.25 g). Fish were fed to satiation in a metabolism chamber. Approximately isoenergetic diets containing 230, 280, 330 and 380 g kg−1 protein were used at 28°C. Ammonia-N excretory energy ( U ) as a percentage of ingested food energy ( C ) increased with an increase in dietary protein level: from 4.3% in fish fed 230 g kg−1 to 6.0% in fish fed 380 g kg−1 dietary protein. The relationship between excretory energy and dietary protein level is modelled.  相似文献

从白鲢(Hypophthalmichthys molitrix)骨骼肌中分离纯化到一种丝氨酸蛋白酶抑制剂,该抑制剂的分子晕大约50kD,用PAGE-明胶电泳证明,该抑制剂对胰蛋白酶具有抑制作用;当温度超过60℃时,抑制剂的活性下降70%左右,但在pH3~11的范围内均有活性。用MTT法分析白鲢骨骼肌丝氨酸蛋白酶抑制剂对急性T细胞白血病细胞Jarkat、人骨髓瘤细胞Sp20和人骨瘤细胞MG63生长的影响,结果发现该抑制剂对以上3种肿瘤细胞的增殖都有抑制作用,而且抑制作用呈剂量依赖性。本研究旨为进一步研究白鲢骨骼肌丝氨酸蛋白酶抑制剂的纯化及其抗肿瘤作用提供科学依据。  相似文献

The effects of introducing common carp (CC) and of adding artificial feed to fertilized rohu ponds on water quality and nutrient accumulation efficiency were studied. All ponds were stocked with 15 000 rohu ha?1. Treatments included ponds with rohu alone, rohu plus 5000 common carp ha?1 and rohu plus 10 000 CC ha?1. A comparison was also made between supplementally fed and non‐fed ponds. The overall highest nitrogen (N) and phosphorus (P) concentrations were observed in ponds with 5000 CC ha?1, followed by ponds with 10 000 and 0 CC ha?1. The largest fractions of N and P inputs accumulating in fish, phytoplankton and zooplankton were observed in ponds with 5000 CC ha?1, followed by ponds with 10 000 CC ha?1 and subsequently ponds without CC. Relatively more nutrients accumulated in benthic organisms in ponds without than in ponds with CC. A smaller fraction of the nutrient input was retained in fish, plankton and benthic organisms in ponds without CC compared with ponds with CC. Compared with 5000 CC ha?1, stocking 10 000 CC ha?1 can be considered as overstocking, because this leads to lower fish production and relatively less nutrients retained in plankton and benthic organisms.  相似文献

研究了硫酸铜、福尔马林、高锰酸钾和敌百虫4种渔药对鲢(Hypophthalmichthys molitrix)鱼种的急性毒性。结果显示:硫酸铜的24 h LC50为7.71 mg/L,48 h LC50为6.11 mg/L,安全浓度为1.15 mg/L;福尔马林的24 hLC50为48.17 mg/L,48 h LC50为36.25 mg/L,安全浓度为6.16 mg/L;高锰酸钾的24 h LC50为3.87 mg/L,48 hLC50为2.75 mg/L,安全浓度为1.74 mg/L;敌百虫的24 h LC50为11.96 mg/L,48 h LC50为9.39 mg/L,安全浓度为0.42 mg/L。结果表明,敌百虫可安全使用,硫酸铜和福尔马林需慎用,高锰酸钾不适宜在鲢鱼种的防病中使用。  相似文献

Diel rhythmicity of grazing, swimming, resting and social interactions of rohu (Labeo rohita) (weight 66.5–68.3 g) and common carp (Cyprinus carpio) (79.9–82.0 g) were observed in 1 m2 simulated ponds using video images. Fish behaviour was monitored during a full 24‐h period, starting at 08:00 hours with a 15‐min recording, which was repeated every 3 h. Rohu spent more time grazing during the day than at night. Grazing activity peaked at the beginning and at the end of the day. Grazing and resting of rohu were negatively correlated. Common carp grazed day and night, showing no distinct grazing peak. Considering the grazing time of rohu and common carp, a feeding schedule with two feeding times at 07:30 and 16:30 hours might be appropriate for better food intake and conversion in rohu–common carp polyculture system.  相似文献

Rohu, Labeo rohita (Ham.), fingerlings (average wt 3.38 g) maintained in 25 m2 cement tanks were fed fish-meal-based, 300 g kg−1 protein diets containing different levels (0, 0.25, 0.5, 0.75 and 1.0 g kg−1) of L -carnitine in triplicate over a period of 126 days. Fish that received 0.5 g kg−1 carnitine showed significantly higher growth than those fed the basal diet or the rest of the carnitine diets. Fat digestibility, food conversion efficiency and protein efficiency ratio were better in carnitine-fed fish; however, viscerosomatic and hepatosomatic indices decreased. Muscle proximate composition was affected as a result of carnitine treatment. The results of the present study demonstrate a positive effect of carnitine on the growth and body composition of rohu.  相似文献

A 60‐day experiment was carried out to delineate stress mitigating and immunomodulatory role of dietary pyridoxine (PN) in Labeo rohita fingerlings exposed to endosulfan. Two hundred and seventy fingerlings were randomly distributed into six treatments in triplicates. Five iso‐caloric and iso‐nitrogenous purified diets were prepared with graded levels of pyridoxine. Six treatment groups were T0 (10 mg PN+without endosulfan), T1 (0 mg PN+endosulfan), T2 (10 mg PN+endosulfan), T3 (50 mg PN+endosulfan), T4 (100 mg PN+endosulfan) and T5 (200 mg PN+endosulfan). The role of pyridoxine on stress mitigation and immunomodulation was assessed by biochemical and haemato‐immunological parameters like aspartate aminotransaminase, alanine aminotransaminase, lactate dehydrogenase, malate dehydrogenase, superoxide dismutase and catalase were significantly (P<0.05) lower while acetylcholinesterase was significantly (P<0.05) higher in pyridoxine‐fed groups. Erythrocytes count, haemoglobin content and total serum protein, albumin, globulin, nitroblue tetrazolium and lysozyme activity were significantly (P<0.05) higher while cortisol and blood glucose were decreased significantly (P<0.05) in pyridoxine‐fed groups. Percentage survival after challenge with Aeromonas hydrophila was highest in T0 group. The results obtained in present study indicate that dietary pyridoxine supplementation at 100 mg PN kg?1 diet reduces the endosulfan‐induced stress and triggers immune response in L. rohita fingerlings.  相似文献

Six isonitrogenous (350 g kg−1 crude protein) and isoenergetic (17573 kJ kg−1) experimental diets incorporating raw and fermented sesame ( Seasamum indicum ) seed meal at 200, 300, and 400 g kg−1 into a fishmeal based diet were fed to rohu Labeo rohita fingerlings for 60 days and the growth performance and feed utilization efficiency of the fish was studied. The antinutritional factor phytic acid, from raw sesame seed meal, could be reduced below detection limit by fermentation with lactic acid bacteria ( Lactobacillus acidophilus ). Fermentation of the oilseed meal resulted in reduction of the tannin content from 20 to 10 g kg−1. In terms of growth response, feed conversion ratio and protein efficiency ratio, a diet containing 400 g kg−1 fermented sesame seed meal resulted in a significantly ( P  < 0.01) best fish performance. In general, growth and feed utilization efficiencies of fish fed fermented sesame seed meal diets were superior to those fed raw oilseed meal diets. Apparent protein digestibility (APD) values decreased with increasing levels of raw oilseed meal. APD was, however, significantly ( P  < 0.01) higher at all levels of incorporation of fermented sesame seed meal, while diets containing raw oilseed meal resulted in poor protein and lipid digestibility. Carcass protein and lipid contents of fish fed fermented sesame seed meal diets increased with increasing level of incorporation, being highest with 400 g kg−1 fermented oilseed meal-containing diet. The results showed that sesame seed meal may be incorporated in carp diets up to 200 g kg−1 and 400 g kg−1 in raw and treated (fermented) forms respectively.  相似文献

The genetic variations of rohu (Labeo rohita, Hamilton) sampled from five hatchery populations (Arabpur, Brahmaputra, Comilla, Kishorganj and Natore) and three major river populations (the Halda, the Jamuna and the Padma) were analysed by allozyme electrophoresis. Ten enzymes encoded by 11 loci were screened, and six were polymorphic. Alleles at three loci (Est‐1*, Gpi‐1* and Gpi‐2*) proved variable for hatchery and river populations, and the Mdh‐2* locus exhibited heterozygous genotypes for river populations only. Polymorphic loci per population (27.3±5.3%), heterozygous loci per individual (15.5±1.2%) and relative gene diversity (0.27±0.08) in river populations were higher than those for hatchery populations (25.5±1.8%, 10.7±1.6% and 0.25±0.01 respectively). Also, the observed heterozygosity (Ho) and expected heterozygosity (He) (0.09±0.03 and 0.14±0.04 respectively) in river populations were higher than those in hatchery populations (0.08±0.01 and 0.11±0.01 respectively). The lower levels of genetic variability in hatchery populations suggested the occurrence of inbreeding and/or genetic drift. The pairwise population differentiation (FST) values showed a lower level of genetic differentiation between hatchery and river population pairs. The unweighted pair‐group method with arithmetic mean dendrogram of Nei's genetic distances showed a relationship between the genetic distance and geographic distance. The populations were clustered into three groups: the Padma in one group, the Halda in second group and the Jamuna, including five hatcheries, in the third group. Highly diversified rohu individuals were observed in the Padma and Halda Rivers, whereas less genetically variable individuals were found in the Jamuna River and five hatcheries. These findings can be useful for rohu hatchery propagation to enhance the sustainable aquaculture production.  相似文献

Different laboratory synthesized metal nanoparticles viz. Copper oxide (CuO), Zinc oxide (ZnO) and silver doped titanium dioxide (Ag‐TiO2) were studied for their effect on hatching and survival of larvae and fry of Indian major carp, rohu, Labeo rohita both in direct application in tank water & coated onto tanks. Among these nanoparticles, CuO and ZnO nanoparticles exhibited highest percentage of hatching in both direct addition (78.0 ± 3.1% and 78.05 ± 4.2%, respectively) and coating onto tanks (58.6 ± 2.1% and 61.2 ± 2.7%, respectively) at 1 mg mL?1 while least percentage of hatching was recorded in Ag‐TiO2 nanoparticles irrespective of its concentration & mode of supplementation. Highest survival of L. rohita fry (50.13 ± 2.2%) was observed after 15 days post hatching in CuO coated tanks followed by ZnO coated tanks (38.6 ± 2.8%) while least was recorded in Ag‐TiO2 coated tanks (22.53 ± 3.0%). However in control tanks coated with Poly‐Urethane base with hardener and uncoated control tanks, the survival was 42.4 ± 1.2% and 41.36 ± 1.8% respectively. Further, significantly lower microbial load of water was recorded in CuO nanoparticles coated tanks (1.5 × 1010 CFU L?1) as compared to uncoated control tanks (1.1 × 1016 CFU L?1) without affecting water quality parameters. On the other hand, in Ag‐TiO2 coated tanks, significantly lower microbial load (1.0 × 106 CFU L?1) as compared to uncoated control tanks at 15 days post hatching was recorded. However, Ag‐TiO2 was toxic to L. rohita larvae & fry both in direct application and coating onto tanks. Considering the beneficial effects of CuO nanoparticle application, it has the scope of being used in a more eco‐friendly way in hatchery operations.  相似文献

A 6‐month feeding trial was conducted in field condition using 10 farm ponds (400–600 m2) to investigate the effect of mixed feeding schedules on the growth of sutchi catfish, Pangasius hypophthalmus with silver carp, Hypophthalmichthys molitrix. Fish were stocked at a ratio of 80:20 (sutchi catfish, 4.9±0.5 g: silver carp, 12.0±0.8 g) at the total rate of 25 000 ha?1. Two diets of high protein (30%, HP) and low protein (16%, LP) were prepared using locally available feed ingredients. Five different feeding schedules of high‐protein diet continuously (HP), low‐protein diet continuously (LP), 1‐day low–protein/1‐day high‐protein diet (1LP/1HP), 7 days low–protein/7 days high‐protein diet (7LP/7HP) and 14 days low–protein/14 days high‐protein diet (14LP/14HP) were tested. The fish were fed twice daily at the rate of 15%, 10%, 8% and 5% of their body weight for first, second, third month and rest of experimental period respectively. Feeding rate was calculated only on the basis of sutchi catfish weight only and was adjusted every 2 weeks according to weight gain. Fish fed LP and HP on alternate day (1LP/1HP) resulted in significantly (P<0.05) higher growth rate, feed utilization and production among the treatments. However, there were no significant differences (P>0.05) between the growth rates and production of fish fed HP regularly and fish fed 7 days LP followed by 7 days HP (7LP/7HP). Fish maintained on LP grew the least. The feed conversion ratio (FCR) values for sutchi catfish ranged between 2.04 and 2.79 with feeding schedule 1LP/1HP showing the best FCR. The total production of fish (including silver carp) ranged between 8310 and 12 422 kg ha?1 6 months?1 with 1LP/1HP feeding schedule resulting in the highest production and net profit. The study demonstrated that feeding fish continuously with HP is less economical. Thus, for profitable sutchi catfish culture with silver carp, farmers can use the mixed feeding schedule of alternate day feeding of LP and HP as a means of reducing feed costs.  相似文献

Growth studies were conducted to quantify requirements for the essential amino acids (EAA), arginine and histidine in Labeo rohita (Hamilton). Diets incorporating casein and gelatine, as sources of intact protein, supplemented with crystalline amino acids were formulated to a crude protein content of 400 g kg?1. Diets with six graded levels of arginine (14.6, 17.0, 19.0, 21.0, 23.0 and 25 g kg?1) and histidine (3.2, 5.0, 7.0, 9.0, 11.0 and 13.0 g kg?1) were formulated and fed to triplicate groups of juvenile rohu twice a day up to satiation for 60 days. Dietary requirements for arginine and histidine for rohu, estimated using break point analysis, were 23 and 9 of the diet respectively (57.5 and 22.5 g kg?1 of dietary protein). Food conversion rate, specific growth rate and survival were better in treatments with diets containing optimum levels of EAA.  相似文献

根据已经获得的两种鲢肌球蛋白重链同工型基因(低温型sc-w和高温型sc-s)在3′端展现的明显差异,设计了2个特异性的反向引物,以鲤科鱼类肌球蛋白重链5′端的保守序列为正向引物,通过Long-PCR对编码鲢两种肌球蛋白重链同工型的球状结构域(Subfragment-1,S1)的全长基因进行了克隆和测序,并推断出它们一级结构的氨基酸序列。研究结果表明,sc-w与sc-s在S1的初级结构上显示80.5%的同源性、与已经报道的草鱼低温型(gc10)有97.2%的高同源性;sc-s则与草鱼中间型(gcI)和高温型(gc30)显示了分别为98.4%和97.1%的高同源性。低温型的sc-w和gc10在S1初级结构上展现的特有变异主要发生在43个氨基酸残基位点,其中15个属保守性残基。对S1区域中两个功能性的表面环loop1(与ATP结合位点有关)和loop2(与肌动蛋白结合位点有关)的结构解析发现,sc-w和gc10在两个表面环的长度、残基电荷分布和氨基酸组成等方面与其它同工型之间存在明显差异,揭示了这两个表面环的结构差异可能影响了栖息于不同环境温度下的淡水鱼的肌球蛋白分子马达功能。分子系统树的分析结果进一步证明,鱼类栖...  相似文献

以鲢(Hypophthalmichthys molitrix)肌肉cDNA为模板, 利用小清蛋白特异性引物进行PCR扩增, 克隆得到β小清蛋白两种不同亚型, 型、型编码区基因。将目的基因片段连接到pET28a (+)表达载体, 并在大肠杆菌[E.coli BL21 (DE3)]中诱导表达。结果表明, 经诱导的小清蛋白重组质粒菌株有特异的蛋白表达。SDS-PAGE分析显示, 目的蛋白的分子量约为13 kD, 与预期大小一致。菌体超声破碎后发现2种亚型的小清蛋白均为可溶表达。利用Ni2+亲和层析柱对重组蛋白进行纯化, 得到高纯度的重组小清蛋白PVⅠPVⅡ。经Western Blot 鉴定, 重组小清蛋白PVⅠPVⅡ均能与抗鲢小清蛋白单克隆抗体反应。本研究为进一步分析小清蛋白的结构与致敏性的关系提供了重要的基础。

Abstract. The ultimate upper lethal temperature (UULT) was 43.0 or 46.0°C for common carp and 43.5 or 46.5°C for silver carp, depending on the method of calculation. The influence of fish age on lethal temperature values was low. Using the relationship between the UULT and the optimum growth temperature the latter was calculated as being 38°C for common carp and 39°C for silver carp. No significant differences were found in survival of fish reared at lower and higher temperatures when proper food was used.  相似文献

鱼糜制品(如火锅鱼丸)的风味是消费者关心的质量属性之一,而关键气味活性物质的吸附释放规律并不明确。现有气味研究主要在配置溶液中进行,与真实的气味活性物质—固态鱼糜之间的相互作用存在一定差异,因此,基于固态鱼糜进行气味研究是十分必要的,其关键在于一个无气味或低气味的鱼糜本底模型,从而可进一步研究各气味成分与鱼糜本底模型的互作关系。本研究考察了8种不同漂洗介质对鱼糜本底模型气味残留的影响。结果表明,白鲢(Hypophthalmichthys molitrix)鱼糜经SPME-GC-MS共检出65种挥发性物质,气味活性物质(OAV>1)有18种;经8种漂洗介质处理后,鱼糜样品中分别含有6、8、7、9、6、12、9和9种气味活性物质,挥发性气味物质的残留率依次为(0.380±0.120)%、(0.610±0.086)%、(0.280±0.033)%、(0.480±0.037)%、(0.150± 0.018)%、(4.330±0.160)%、(18.680±0.081)%和(0.490±0.003)%。综合SPME-GC-MS、电子鼻和感官评价结果比较,1% NaCl (W/W) + 1% Na2CO3 (W/W) + 4.0% C2H5OH (V/W)漂洗介质处理后,白鲢鱼糜的挥发性气味物质残留少,总含量降低为(6.57±0.77) μg/kg,17种气味活性物质的OAV<1,仅壬醛的OAV为1.34±0.05,可构建出低气味的鱼糜本底模型。  相似文献

小清蛋白是鱼类的主要过敏原,对该蛋白的研究不仅有利于过敏原检测方法的建立也可为低致敏性水产品的开发提供理论依据。通过组织捣碎、冷冻离心、热处理、Superdex75凝胶过滤等方法从鲢白色肉中纯化得到过敏原小清蛋白。Tricine-SDS-PAGE显示,在非还原条件下,该蛋白呈分子量分别为12ku、14ku、24ku的3个条带。而在还原条件下,仅有分子量为12ku的条带。Western-blotting分析表明,分子量为12ku、14ku和24ku的这3个条带都与小鼠抗蛙小清蛋白单克隆抗体(PARV-19)发生特异性反应,提示它们均为小清蛋白的不同形态。用纯化的小清蛋白制备多克隆抗体,经Protein A Sepharose亲和层析纯化得到高纯度的免疫球蛋白G(IgG)。Dot-blot检测发现,抗体稀释至1/51200时仍能与纯化的小清蛋白有显色反应。用制备的多克隆抗体进行Western-blotting分析,能特异地检测4种鱼(鲤、鲢、鲫、黄鳍鲷)中的小清蛋白。  相似文献

A variety of parameters associated with breathing and feeding of silver carp were measured in response to declines in dissolved oxygen (DO) levels when the fish were fed microalgae Padorina morum (PM group) and Chlorella ellipsoidea (CE group), respectively, to explore the relationship between breathing and feeding while filtering microalgae particles at different DO levels. The results indicated that (i) respiratory frequency (fR) of the fish in CE group were higher significantly than those in PM group at DO levels of 0.98–7.49 mg/L (< 0.05); respiratory stroke volume (VS.R), gill ventilation (VG) and VG/VO2 the fish in two groups began to increase sharply while DO levels declined to below 4.04 mg/L (< 0.05); oxygen extraction efficiency (EO2) of the fish in two groups increased significantly with decline of DO levels from 7.49 mg/L to 0.98 mg/L (< 0.05); (ii) the infiltration rate (FR) and filtering efficiency (E) of the fish in two groups began to decrease significantly while DO levels declined to below 4.04 mg/L (< 0.05); clearance rate (CR) of fish in PM group was sixfold higher than that in CE group at DO levels of 0.98–7.49 mg/L (< 0.05). In addition, antifiltering response appeared at DO levels of 2.21 mg/L and 0.98 mg/L in both groups. This study indicated that ‘filtration effect’ is dominant when the fish filter algae particles larger than its gill raker gaps, while the ‘food sinking effect’ is dominant when the fish filter algae particles smaller than its gill raker gaps.  相似文献
