Mycorrhiza helper bacteria (MHB) can promote the formation and functioning of arbuscular mycorrhizal (AM) symbiosis, but their role and application potential in coping with soil-borne diseases are still unclear. A 14-week greenhouse pot experiment was conducted to obtain several actinomycete strains helping AM symbiosis in suppressing the Phytophthora blight of pepper (Capsicum annuum L.), using a soil inoculated with Phytophthora capsici after sterilization. Five Streptomyces strains, including S. pseudogriseolus, S. albogriseolus, S. griseoaurantiacus, S. tricolor, and S. tendae, as well as the AM fungus (Funneliformis caledonium) were tested. The Phytophthora blight severity reached 66% at full productive stage in the uninoculated control, and inoculation of F. caledonium, S. griseoaurantiacus, and S. tricolor alone significantly decreased (P < 0.05) it to 47%, 40%, and 35%, respectively. Compared to F. caledonium alone, additional inoculation of S. tricolor or S. tendae, which were isolated from the rhizosphere of a healthy individual in an infected field, significantly elevated (P < 0.05) root mycorrhizal colonization, root biomass, fruit yield, and total K acquisitions of pepper and further significantly decreased (P < 0.05) blight severity. According to the feature of enhancing disease-suppression by AM symbiosis, both S. tricolor and S. tendae were confirmed as MHB strains here. Specifically, S. tendae had a stronger performance in directly accelerating mycorrhization, while S. tricolor antagonist to the pathogenic P. capsici. Furthermore, S. griseoaurantiacus with the independent disease-suppression function was not an MHB strain here. The redundancy analyses demonstrated that when AM fungus was present, root mycorrhizal colonization replaced soil pH becoming the main factor affecting pepper Phytophthora blight. Thus, S. tricolor and S. tendae seemed to have the value of preparation and application in the future to help AM symbiosis against pepper Phytophthora blight.

In sustainable agriculture, arbuscular mycorrhizal (AM) fungal inoculation in agronomical management might be very important, especially when the efficiency of native inocula is poor. Here, we assessed the effect of native and exotic selected AM fungal inocula on plant growth and nutrient uptake in a low input Trifolium alexandrinum-Zea mays crop rotation. We evaluated the effects of four exotic AM fungal isolates on T. alexandrinum physiological traits in greenhouse. Then, the field performances of T. alexandrinum inoculated with the exotic AMF, both single and mixed, were compared to those obtained with a native inoculum, using a multivariate analysis approach. Finally, we tested the residual effect of AM fungal field inoculation on maize as following crop. Multivariate analysis showed that the field AM fungal inoculation increased T. alexandrinum and Z. mays productivity and quality and that the native inoculum was as effective as, or more effective than, exotic AM fungal isolates. Moreover, the beneficial effects of AMF were persistent until the second year after inoculation. The use of native AMF, produced on farm with mycotrophic plants species, may represent a convenient alternative to commercial AM fungal inocula, and may offer economically and ecologically important advantages in sustainable or organic cropping systems.

The effects of soil disturbance and residue retention on the functionality of the symbiosis between medic (Medicago truncatula L.) and arbuscular mycorrhizal fungi (AMF) were assessed in a two-stage experiment simulating a crop rotation of wheat (Triticum aestivum L.) followed by medic. Plants were inoculated or not with the AMF, Glomus intraradices and Gigaspora margarita, separately or together. The contribution of the arbuscular mycorrhizal (AM) pathway for P uptake was determined using 32P-labeled soil in a small hyphal compartment accessible only to hyphae of AMF. In general AM colonization was not affected by soil disturbance or residue application and disturbance did not affect hyphal length densities (HLDs) in soil. At 4 weeks disturbance had a negative effect on growth and phosphorus (P) uptake of plants inoculated with G. margarita, but not G. intraradices. By 7 weeks disturbance reduced growth of plants inoculated with G. margarita or AMF mix and total P uptake in all inoculated plants. With the exception of plants inoculated with G. margarita in disturbed soil at 4 weeks, the AM pathway made a significant contribution to P uptake in all AM plants at both harvests. Inoculation with both AMF together eliminated the negative effects of disturbance on AM P uptake and growth, showing that a fungus insensitive to disturbance can compensate for loss of contribution of a sensitive one. Application of residue increased growth and total P uptake of plants but decreased 32P in plants inoculated with the AMF mix in disturbed soil, compared with plants receiving no residue. The AMF responded differently to disturbance and G. intraradices, which was insensitive to disturbance, compensated for lack of contribution by the sensitive G. margarita when they were inoculated together. Colonization of roots and HLDs in soil were not good predictors of the outcomes of AM symbioses on plant growth, P uptake or P delivery via the AM pathway.

Soil compaction is of great importance, due to its adverse effects on plant growth and the environment. Mechanical methods to control soil compaction may not be economically and environmentally friendly. Hence, we designed experiments to test the hypothesis that use of plant symbiotic fungi, arbuscular mycorrhiza (AM) may alleviate the stressful effects of soil compaction on corn (Zea mays L.) growth through enhancing nutrient uptake. AM continuously interact with other soil microorganisms and its original diversity may also be important in determining the ability of the fungi to cope with the stresses. Hence, the objectives were: (1) to determine the effects of soil compaction on corn nutrient uptake in unsterilized (S1) and sterilized (S2) soils, and (2) to determine if inoculation of corn with different species of AM with different origins can enhance corn nutrient uptake in a compacted soil. Using 2 kg weights, soils (from the field topsoil) of 10 kg pots were compacted at three and four levels (C1, C2, C3 and C4) (C1 = non-compacted control) in the first and second experiment, respectively. Corn (cv. 704) seeds were planted in each pot and were inoculated with different AM treatments including control (M1), Iranian Glomus mosseae (M2), Iranian G. etunicatum (M3), and Canadian G. mosseae, received from GINCO (Glomales In Vitro Collection), Canada (M4). Corn leaf nutrient uptake of N, P, K, Fe, Mn, Zn and Cu were determined. Higher levels of compaction reduced corn nutrient uptake, however different species of AM and soil sterilization significantly increased it. The highest increase in nutrient uptake was related to P (60%) and Fe (58%) due to treatment M4S2C3. Although it seems that M3 and M4 may be the most effective species on corn nutrient uptake in a compacted soil, M2 increased nutrient uptake under conditions (C3 and C4 in unsterilized soil) where the other species did not. Through increasing nutrient uptake AM can alleviate the stressful effects of soil compaction on corn growth.

A field experiment was conducted to study and compare the effectiveness of two arbuscular mycorrhizal fungi (AMF), Glomus macrocarpum (GM) and Glomus fasciculatum (GF) on three accessions of Artemisia annua. The AM inoculation significantly increased the production of herbage, dry weight of shoot, nutrient status (P, Zn and Fe) of shoot, concentration of essential oil and artemisinin in leaves as compared to non-inoculated plants. The extent of growth, nutrient concentration and production of secondary plant metabolites varied with the fungus–plant accession combination. The mycorrhizal dependency of the three accessions was related to the shoot: root ratio. Comparing the two fungal inoculants in regard to increase in essential oil concentration in shoot, the effectiveness of GF was more than that of GM. While in two accessions, GM was more effective in enhancing artemisinin concentration than GF. Increase in concentration of essential oil was found to be positively correlated to P-status of the plant. Conversely, no correlation was found between shoot-P and artemisinin concentration.

Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) in combination with sequencing of amplified partial 18S ribosomal DNA was used to assess the effects of transgenic Bt corn 5422Bt1 (event Bt 11) and 5422CBCL (event MON810) on the community structure of a non-target microorganism, namely the arbuscular mycorrhizal fungi (AMF) Glomus in corn roots and rhizosphere soils, relative to their non-Bt isolines 5422 (conventional parent) and 5422wx (conventional hybrid). AMF colonization in roots of different corn genotypes was also assessed using microscopic visualization. No adverse effect was detected on the indigenous AMF colonization of the roots of Bt hybrids 5422Bt1 and 5422CBCL. Two-way indicator species analysis (TWINSPAN®) and detrended correspondence analysis (DCA) of the DGGE data from corn roots presented differences between Bt and non-Bt corn isolines (5422Bt1 vs. 5422wx, and 5422CBCL vs. 5422wx). However, differences were also recognized between the two non-Bt corn cultivars (5422 vs. 5422wx), and between the two Bt corn lines (5422Bt1 vs. 5422CBCL) in roots. Our results suggest that corn genotypes may have a greater influence on the AMF community structure of plant roots and rhizosphere soils than other factors, such as the age of the growing plants.

Plants can mediate interactions between aboveground herbivores and belowground decomposers as both groups depend on plant-provided organic carbon. Most vascular plants also form symbiosis with arbuscular mycorrhizal fungi (AMF), which compete for plant carbon too. Our aim was to reveal how defoliation (trimming of plant leaves twice to 6 cm above the soil surface) and mycorrhizal infection (inoculation of the fungus Glomus claroideum BEG31), in nutrient poor and fertilized conditions, affect plant growth and resource allocation. We also tested how these effects can influence the abundance of microbial-feeding animals and nitrogen availability in the soil. We established a 12-wk microcosm study of Plantago lanceolata plants growing in autoclaved soil, into which we constructed a simplified microfood-web including saprotrophic bacteria and fungi and their nematode feeders. We found that fertilization, defoliation and inoculation of the mycorrhizal fungus all decreased P. lanceolata root growth and that fertilization increased leaf production. Plant inflorescence growth was decreased by defoliation and increased by fertilization and AMF inoculation. These results suggest a negative influence of the treatments on P. lanceolata belowground biomass allocation. Of the soil organisms, AMF root colonization decreased with fertilization and increased with defoliation. Fertilization decreased numbers of bacterial-feeding nematodes, probably because fertilized plants produced less root mass. On the other hand, bacterial feeders were more abundant when associated with defoliated than non-defoliated plants despite defoliated plants having less root mass. The AMF inoculation per se increased the abundance of fungal feeders, but the reduced and increased root AM colonization rates of fertilized and defoliated plants, respectively, were not reflected in the numbers of fungal feeders. We found no evidence of plant-mediated effects of the AM fungus on bacterial feeders, and against our prediction, soil inorganic nitrogen concentrations were not positively associated with the concomitant abundances of microbial-feeding animals. Altogether, our results suggest that (1) while defoliation, fertilization and AMF inoculation all affect plant resource allocation, (2) they do not greatly interact with each other. Moreover, it appears that (3) while changes in plant resource allocation due to fertilization and defoliation can influence numbers of bacterial feeders in the soil, (4) these effects may not significantly alter mineral N concentrations in the soil.

Exudates of Rhodotorula mucilaginosa, a yeast commonly found in the rhizosphere, increased hyphal length of the arbuscular mycorrhizal (AM) fungi Gigaspora rosea and Gigaspora margarita. Rhodotorulic acid (RA), a siderophore compound obtained from R. mucilaginosa exudates, increased hyphal length and branching. Thus, the increase in the number of entry points and the higher AM root colonization of tomato plants in the presence of RA can at least partially be explained by the positive effect of RA on the pre-symbiotic stages of the AM fungi.

The rose of an isolate of the arbuscular mycorrhizal (AM) fungusGlomus mosseae in the protection ofMedicago sativa (+Rhizobium meliloti) against salt stress induced by the addition of increasing levels of soluble salts was studied. The interactions between soluble P in soil (four levels), mycorrhizal inoculum and degree of salinity in relation to plant growth, nutrition and infective parameters were evaluated. Salt stress was induced by sequential irrigation with saline water having four concentrations of three salts (NaCl, CaCl2, and MgCl2).15N-labelled ammonium sulphate was added to provide a quantitative estimate of N2 fixation under moderate to high salinity levels. N and P concentration and nodule formation increased with the amount of plant-available P or mycorrhizal inoculum in the soil and generally declined as the salinity in the solution culture increased from a moderate to a high level. The mycorrhizal inoculation protected the plants from salt stress more efficiently than any amount of plant-available P in soil, particularly at the highest salinity level applied (43.5 dS m–1). Mycorrhizal inoculation matched the effect on dry matter and nutrition of the addition in the soil of 150 mg P kg–1. Nevertheless the highest saline solution assayed (43.5 dS m–1) affected more severely plants supplemented with phosphorus than those with the addition of mycorrhizal inoculum. Such a saline-depressing effect was 1.5 (biomass), 1.4 (N) and 1.5 (P) times higher in plants supplied with soluble phosphate than with AM inoculum. Mechanisms beyond those mediated by P must be involved in the AM-protectioe effect against salinity. The15N methodology used allowed the determination of N2 fixation as influenced by different P applications compared to mycorrhizal inoculation. A lack of correlation between nodule formation and function (N2 fixation) was evidenced in mycorrhizal-inoculated plants. In spite of the reduced activity per nodule in mycorrhizal-inoculated In spite of the reduced activity per nodule in mycorrhizal-inoculated plants, the N contents determined indicated the highest acquisition of N occurred in plants with the symbiotic status. Moreover, N and P uptake increased while Ca and Mg decreased in AM-inoculated plants. Thus P/Ca ratios and cation/anion balance in general were altered in mycorrhizal treatments. This study therefore confirms previous findings that AM-colonized plants have optional and alternative mechanisms available to satisfy their nutritive requirements and to maintain their physiological status in stress situations and in disturbed ecosystems.

Dendrobaena octaedra (Lumbricidae) and Cognettia sphagnetorum (Enchytraeidae) are the two most dominating soil invertebrates in terms of biomass in boreal coniferous forest soils. A microcosm experiment was set up in order to study the influence of pH, moisture and resource addition on D. octaedra and C. sphagnetorum when both species are simultaneously present. Two kinds of coniferous forest humus were used as substrate, pine stand humus (pH 4.2), and spruce stand humus (pH 4.6); in the third treatment the pine stand humus was adjusted with slaked lime (CaOH2) to the same initial pH as the spruce stand humus. Each substrate was adjusted to water contents of 25%, 42.5% and 60% of WHC (referred to as ‘dry’, ‘moist’ and ‘wet’). In the second part of the experiment, spruce needle litter and birch leaf litter were separately added into the pine stand humus (‘moist’, unlimed) and compared with a control without litter. The microcosms were plastic jars with 75 g (d.m.) of humus, into which 4 specimens of D. octaedra and 70 specimens of C. sphagnetorum were added. D. octaedra showed the highest biomass and C. sphagnetorum the lowest biomass in the spruce stand humus with higher pH. Moisture did not affect earthworms, while C. sphagnetorum thrived best at the highest moisture. Addition of both kinds of litter increased the numbers and biomass of D. octaedra, while on C. sphagnetorum resource addition had little effect. The results help to explain the abundance of these two species in coniferous forests differing in soil acidity, moisture and fertility.

Interactions between the biocontrol fungus Clonostachys rosea IK 726 and a tomato/Glomus intraradices BEG87 symbiosis were examined with and without wheat bran, which served as a food base for C. rosea. In soil without wheat bran amendment, inoculation with C. rosea increased plant growth and altered shoot nutrient content resulting in an increase and decrease in P and N content, respectively. Inoculation with G. intraradices had no effect on plant growth, but increased the shoot P content. Dual inoculation with G. intraradices and C. rosea followed the pattern of C. rosea in terms of plant growth and nutrient content. Wheat bran amendment resulted in marked plant growth depressions, which were counteracted by both inoculants and dual inoculation increased plant growth synergistically. Amendment with wheat bran increased the population density of C. rosea and reduced mycorrhizal fungus colonisation of roots. The inoculants were mutually inhibitory, which was shown by a reduction in root colonisation with G. intraradices in treatments with C. rosea and a reduction in colony-forming units (cfu) of C. rosea in treatments with G. intraradices, irrespective of wheat bran amendment. Moreover, both inoculants markedly influenced soil microbial communities examined with biomarker fatty acids. Inoculation with G. intraradices increased most groups of microorganisms irrespective of wheat bran amendment, whereas the influence of C. rosea on other soil microorganisms was affected by wheat bran amendment. Overall, inoculation with C. rosea increased and decreased most groups of microorganisms without and with wheat bran amendment, respectively. In conclusion, despite mutual inhibition between the two inoculants this interaction did not impair their observed plant growth promotion. Both inoculants also markedly influenced other soil microorganisms, which should be further studied in relation to their plant growth-promoting features.

Elevated aluminum (Al) availability limits plant growth on acidic soils. Although this element is found naturally in soils, acidic conditions create an environment where Al solubility increases and toxic forms of Al impact plant function. Plant resistance to Al is often attributed to organic acid exudation from plant roots and the chelation of cationic Al in the rhizosphere. The association of arbuscular mycorrhizal (AM) fungi with the roots of plants may alleviate Al toxicity by altering soil Al availability or plant exposure through the binding of Al to fungal structures or through the influence of fungi on exudation from roots. Diverse communities of AM fungi are found in soil ecosystems and research suggests that AM fungi exhibit functional diversity that may influence plant performance under varying edaphic environments. In the present study, we evaluated acidic isolates of six AM species in their responses to Al. Andropogon virginicus (broomsedge), a warm-season grass that commonly grows in a range of stressful environments including acidic soils, was used as a plant host for Acaulospora morrowiae, Glomus claroideum, Glomus clarum, Glomus etunicatum, Paraglomus brasilianum, and Scutellospora heterogama. Fungal spores were germinated and exposed to 0 or 100 μM Al on filter paper in sand culture or were grown and exposed to Al in sand culture in association with A. virginicus. Short- and long-term responses to Al were evaluated using direct measurements of fungal spore germination, hyphal elongation, and measurements of A. virginicus colonization and plant growth as a phytometer of AM function in symbio. Spore germination and hyphal elongation varied among AM species in response to Al, but patterns were not consistent with the influences of these AM species on A. virginicus under Al exposure. Exposure to Al did not influence colonization of roots, although large differences existed in colonization among fungal species. Plants colonized by G. clarum and S. heterogama exhibited the least reduction in growth when exposed to Al, produced the highest concentrations of Al-chelating organic acids, and had the lowest concentrations of free Al in their root zones. This pattern provides evidence that variation among AM fungi in Al resistance conferred to their plant hosts is associated with the exudation of Al-binding organic acids from roots and highlights the role that AM fungal diversity may play in plant performance in acidic soil environments.

Earthworms and arbuscular mycorrhizal fungi (AMF) might interactively impact plant productivity; however, previous studies reported inconsistent results. We set up a three-factorial greenhouse experiment to study the effects of earthworms (Aporrectodea caliginosa Savigny and Lumbricus terrestris L.) and AMF (Glomus intraradices N.C. Schenck & G.S. Sm.) on the performance (productivity and shoot nutrient content) of plant species (Lolium perenne L., Trifolium pratense L. and Plantago lanceolata L.) belonging to the three functional groups grasses, legumes and herbs, respectively. Further, we investigated earthworm performance and plant root mycorrhization as affected by the treatments. Our results accentuate the importance of root derived resources for earthworm performance since earthworm weight (A. caliginosa and L. terrestris) and survival (L. terrestris) were significantly lower in microcosms containing P. lanceolata than in those containing T. pratense. However, earthworm performance was not affected by AMF, and plant root mycorrhization was not modified by earthworms. Although AMF effectively competed with T. pratense for soil N (as indicated by δ15N analysis), AMF enhanced the productivity of T. pratense considerably by improving P availability. Remarkably, we found no evidence for interactive effects of earthworms and AMF on the performance of the plant species studied. This suggests that interactions between earthworms and AMF likely are of minor importance.

The fungal pathogen Batrachochytrium dendrobatidis (Bd), which causes the disease chytridiomycosis, has been associated with declines and extinctions of montane amphibians worldwide. To gain insight into factors affecting its distribution and prevalence we focus on the amphibian community of the Klamath Mountains in northwest California. The Cascades frog (Rana cascadae), one of the most common amphibians in these mountains, experienced increased mortality as a result of Bd exposure in laboratory trials and has experienced recent, dramatic declines in other parts of California. We surveyed 112 sites in the Klamaths, all of which supported R. cascadae between 1999 and 2002, for amphibians and Bd to (1) determine the distribution of Bd, (2) evaluate changes in the distribution of R. cascadae, and (3) assess associations between potential biotic and abiotic drivers and Bd infection. Bd was widely distributed in the Klamath Mountains – we detected the pathogen at 64% of sites. R. cascadae was found at 79% of sites, and was often infected with Bd. These results suggest that Bd has not caused dramatic declines in R. cascadae in the Klamaths in recent years. Subadult R. cascadae had a higher Bd prevalence than other R. cascadae life stages (subadults: 36%, adults: 25%, metamorphs: 4%, larvae: 1%), and while the probability of infection decreased over the season for adults, it did not for subadults, suggesting that subadults may be more vulnerable to chytridiomycosis than other R. cascadae life stages. Bd prevalence in R. cascadae was highest early in the season at high-elevation sites, which may indicate that populations inhabiting high elevation sites may have a greater risk of being affected by chytridiomycosis. Three other common amphibian species also tested positive for Bd: Pacific chorus frog (Pseudacris regilla), western toad (Anaxyrus boreas), and rough-skinned newt (Taricha granulosa).

Sečovlje salterns are an important protected area of biotic diversity in the Mediterranean. They represent an extreme environment with high salinity and drought that severely influence the growth of organisms. In the present study, diversity of plant halophytes and their mycorrhizal status were screened at eight different locations, which were mostly dikes and salt ponds, and which were deliberately selected for their distinct properties (e.g. soil salinity ranging from 105 to 2627 μS cm−1, vegetation type and management practice of the salterns).Twelve different halophytic plant species were recorded, of which eleven are designated as vulnerable. With few exceptions, they were found at the abandoned (Fontanigge) and sustained (Lera) locations of the Sečovlje salterns, distributed according to their tolerance to the salinity and waterlogging. The highest diversity of halophytes was listed at Fontanigge, in the abandoned, periodically flooded and gradually overgrown salt ponds. All of the examined species were colonised with either arbuscular mycorrhizal fungi (AMF) and/or dark septate endophytes (DSEs). High levels of colonisation were however detected only for species belonging to the Asteraceae and Plantaginaceae families. Higher root colonisation frequencies were generally seen for plants growing in the abandoned parts, when compared to the managed parts, whereas there was little correlation of the colonisation parameters with physicochemical parameters of rhizospheric soil properties.Molecular analysis by temporal temperature gradient gel electrophoresis (TTGE) of roots of halophytic plant species with confirmed AMF colonisation (arbuscules present) revealed the occurrence of at least six different AMF species, related to Glomus geosporum, Glomus caledonium and Glomus intraradices, and to different Glomus sp. clades and the Diversispora clade. This is to the best of our knowledge the first report of AMF and DSE mycorrhizal status of most of the halophyte plant species examined and of the brother scale identification of AMF species based on molecular analyses of roots of diverse halophytes from high saline environments.

Plants inoculated with arbuscular mycorrhizal (AM) fungi utilize more soluble phosphorus from soil mineral phosphate than non-inoculated plants. However, there is no information on the response of soil microflora to mineral phosphate weathering by AM fungi and, in particular, on the catabolic diversity of soil microbial communities. The AM fungus, Glomus intraradices was examined for (i) its effect on the growth of Acacia holosericea, (ii) plant-available phosphate and (iii) soil microbial activity with and without added rock phosphate. After 4-months culture, AM fungal inoculation significantly increased the plant biomasses (by 1.78× and 2.23× for shoot and root biomasses, respectively), while mineral phosphate amendment had no effect in a sterilized soil. After 12-months culture, the biomasses of A. holosericea plants growing in a non-sterilized soil amended with mineral phosphate were significantly higher than those recorded in the control treatment (by 2.5× and 5× for shoot and root biomasses, respectively). The fungal inoculation also significantly stimulated plant growth, which was significantly higher than that measured in the mineral phosphate treatment. When G. intraradices and mineral phosphate were added together to the soil, shoot growth were significantly stimulated over the single treatments (inoculation or amendment) (1.45×). The P leaf mineral content was also higher in the G. intraradices+mineral phosphate treatment than in G. intraradices or rock phosphate amendment. Moreover, the number of fluorescent pseudomonads has been significantly increased when G. intraradices and/or mineral phosphate were added to the soil. By using a specific type of multivariate analysis (co-inertia analysis), it has been shown that plant growth was positively correlated to the metabolization of ketoglutaric acid, and negatively linked to the metabolisation of phenylalanine and other substrates, which shows that microbial activity is also affected.G. intraradices inoculation is highly beneficial to the growth of A. holosericea plants in controlled conditions. This AM symbiosis optimises the P solubilization from the mineral phosphate and affects microbial activity in the hyphosphere of A. holosericea plants.
