The objective of the present study was to determine the temporal relationships among luteal adenylate cyclase activity, luteal phosphodiesterase activity, luteal progesterone concentration and plasma progesterone concentration during prostaglandin F2 alpha (PGF2 alpha)-induced luteolysis in ewes. Corpora lutea (CL) were removed from cycling ewes on d 9 (d 0 = first day of estrus) at 0, 2, 4, 6, 12 and 24 h (seven to eight ewes/group) after PGF2 alpha administration (im). Jugular blood samples were collected at the time of enucleation of CL and analyzed for progesterone. Plasma and luteal progesterone concentrations were decreased (P less than .05) by 4 and 12 h after PGF2 alpha injection, respectively. Basal adenylate cyclase, luteinizing hormone (LH)-activated adenylate cyclase, guanylylimidodiphosphate [Gpp(NH)p]-activated adenylate cyclase and LH plus Gpp(NH)p-activated adenylate cyclase activities were decreased (P less than .05) by 2 h after PGF2 alpha injection. The decrease in adenylate cyclase activity paralleled the decrease in plasma progesterone concentration over time. Luteinizing hormone stimulated (P less than .05) adenylate cyclase activity relative to basal activity at 0, 2, 12 and 24 h post-PGF2 alpha; whereas, Gpp(NH)p stimulated (P less than .01) adenylate cyclase activity relative to basal activity at each time point. In contrast to the decrease in adenylate cyclase activity, phosphodiesterase activity was increased (P less than .05) at 2 and 4 h post-PGF2 alpha. These results suggest that a decrease in adenylate cyclase activity coupled with an increase in phosphodiesterase activity may decrease the intracellular adenosine 3',5' cyclic monophosphate (cAMP) concentration.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Corpus luteum (CL) function following spontaneous or prostaglandin F2 alpha-induced estrus was studied in 27 Brahman cows and 16 Brahman heifers. Females received one injection of 0, .38, .75 or 2.25 mg alfaprostol (PGF)/100 kg body weight at 12 +/- .1 d post-estrus. Serum progesterone (P4) during CL formation, following the subsequent estrus, increased over time (P less than .001, 1 to 13 d post-estrus) and was influenced by PGF (P less than .06) and a PGF X time interaction (P less than .02). Low serum P4 concentrations were observed during CL formation (d 3, 4, 10, and 12) in cattle that received 2.25 mg PGF. On d 13 post-estrus, CL were removed. No differences in weight, P4 content or number of luteinizing hormone (LH) receptors was evident between CL derived 13 d following spontaneous vs PGF-induced estrus. The CL formed following PGF-induced estrus had fewer large (P less than .002) and small (P less than .09) luteal cells in heifers, a lower (P less than .008) percentage composition of histological type I and II luteal cells in cows and a lower (P less than .0001) in vitro P4 response to LH in both cows and heifers when compared with the CL formed following spontaneous estrus. Cows had heavier (P less than .002) CL with a higher (P less than .05) number of large cells, a higher (P less than .09) percentage composition of histological cell type I and II luteal cells, and a higher (P less than .03) in vitro basal and stimulated P4 secretion. It is concluded that 1) the events associated with artificially shortening the estrous cycle with PGF altered subsequent CL function in Brahman females; 2) cows had heavier CL, which were composed of more steroidogenically functional luteal cells, than heifers and 3) age of animal interacted with PGF to alter subsequent CL function in Brahman females.  相似文献

The effect of the uterus on luteal lifespan and pattern of secretion of progesterone following early weaning of calves from anestrous beef cows was studied. Calves were weaned from 15 anestrous beef cows 23 to 33 d postpartum, and cows were allotted to a control (sham surgery, n = 8) or a hysterectomy (n = 7) group, with surgery performed at weaning. Cows in the hysterectomy group were injected (im) with 25 mg prostaglandin F2 alpha (PGF2 alpha) approximately 20 d after first estrus (d 0). The interval from weaning to estrus was longer (P less than .05) for the hysterectomy group (10.4 +/- 1.6 d) than the control group (6.2 +/- .5 d). In the control group, the first estrous cycle (8.8 +/- .3 d) was shorter (P less than .01) than the second estrous cycle (20.2 +/- .5 d). Following first estrus in the hysterectomy group, cows were not detected in estrus until after injection of PGF2 alpha and did not return to estrus. From d 0 to 5, mean concentrations of plasma progesterone were similar (P greater than .05) between groups for both estrous cycles; after d 5 of estrous cycle 1, concentrations of plasma progesterone decreased in the control group. Within the hysterectomy group, the pattern of secretion of progesterone from d 0 to 16 was similar after the first and second estrus. In Trial 1, 26 heifers were allotted randomly to a control group or one of four groups to receive a single injection of 3,000 IU hCG on d 1, 4, 7 or 10 of an estrous cycle. Heifers next completed a nontreated cycle, and at their third estrus were reallotted to one of the five groups described previously. Estrous cycle length was extended in cycle 1 but was not altered during the nontreated cycle or in cycle 3. Administration of hCG on d 4 or 7 increased (P less than .05) mean serum progesterone (P4) over the first 16 d of the estrous cycle by .9 and .8 ng/ml, respectively. In Trial 2, 23 heifers were allotted randomly to one of two groups to receive a placebo or a single injection of 3,000 IU hCG on d 4 of an estrous cycle. Heifers were inseminated artificially at subsequent estrus. On d 4 postbreeding, treatments were repeated. Administration of hCG on d 4 of the prebreeding estrous cycle increased (P less than .05) mean P4 over the first 16 d by .9 ng/ml, whereas mean P4 over the first 16 d postbreeding was not affected by a second injection of hCG on d 4 postbreeding. Administration of hCG increased (P less than .05) first-service pregnancy rate (92 vs 55%). In conclusion, progesterone concentrations were enhanced by hCG given on d 4 or 7 of the estrous cycle, and pregnancy rate was increased when hCG was administered both on d 4 of the prebreeding cycle and d 4 postmating.  相似文献

Effects of an increased level of dietary energy (flushing) on plasma concentrations of FSH, LH, insulin, progesterone and estradiol-17 beta and ovulation rate were studied in 16 gilts. Gilts received 5,400 kcal ME/d for one estrous cycle and the first 7 d of a second. On d 8 of the second estrous cycle, gilts received either 5,400 kcal ME/d (control [C], n = 8) or 11,000 kcal ME/d (flushed [F], n = 8) for the remainder of the estrous cycle. Blood was collected daily at 15-min intervals for 6 h from d 8 through estrus. Gilts were examined by laparotomy 6 d after estrus. Ovulation rate was greater (P less than .05) in F than C gilts (16.0 vs 9.4). Mean daily concentrations of FSH were greater (P less than .05) in F gilts at 5 d, 4 d and 3 d prior to estrus compared with C females. In both C and F gilts, FSH decreased (P less than .05) prior to estrus. Mean daily concentrations of LH and LH pulse amplitude were not different (P greater than .05) between treatments. Mean number of LH pulses/6 h at 4 d, 3 d and 2 d prior to estrus were greater (P less than .05) in F than in C gilts. In both treatments, LH pulse amplitude decreased (P less than .05) and pulse frequency increased (P less than .07) prior to estrus. Early weaning of calves from anestrous cows results in formation of short-lived corpora lutea (CL) unless the animals are pretreated with a progestagen (norgestomet). This study was conducted to investigate the relationship between pre- and post-ovulatory gonadotropin secretion and luteal lifespan. Postpartum beef cows were assigned randomly into two groups, control (n = 5) and norgestomet (implant given at weaning for 9 d; n = 7). Calves from all cows were weaned 30 to 33 d postpartum. Coccygeal artery cannulas were placed into cows in the control group 1 d prior to weaning and 2 d before implant removal in cows in the norgestomet group. Plasma for determination of luteinizing hormone (LH), follicle stimulating hormone (FSH), estradiol-17 beta (E) and progesterone (P) was collected daily at 10-min intervals for 6 h from weaning (control) or the day prior to implant removal (norgestomet) to estrus (d 0) and on d 2, 4 and 6 following estrus. Average interval (X +/- SE; P less than .05) from weaning to estrus or implant removal was 4.2 +/- .8 and 2.3 +/- .2 d for the control and norgestomet groups, respectively. Estrous cycle length for the control group was 12.4 +/- 1.8 d compared with 20.4 +/- .3 d for the norgestomet group (P less than .05). The postpartum (PP) period in the Pelibuey ewe was studied. Laparotomies were performed on 14 ewes in the first year at d 10, 20 and 30 PP, and at d 10 and 20 PP in the second year on 17 ewes. Progesterone concentrations were determined in serum taken daily, from 4 to 7 d after parturition until estrus. Temporal fluctuation of luteinizing hormone (LH) was determined in samples taken at 30-min intervals for 4 h weekly. The mean interval from lambing to first ovulation was longer (P less than .001) in 1980 (59 +/- 4.9 d) than 1979 (26 +/- 3.1 d), the mean interval from lambing to first estrus was also longer (P less than .001) in 1980 (91 +/- 5.6 d) than 1979 (51 +/- 5.5 d). Follicles were present on the ovaries of the majority of the ewes at d 10. The mean diameter of the largest follicles on each ovary was reduced (P less than .025) in ewes in 1980 (6 mm) compared with 1979 (7.7 mm). Corpora lutea (CL) occurred in 67 and 75% of the ewes by d 20 and 30, respectively in 1979; no CL were found by d 20 in 1980. Progesterone profiles suggested that the PP period was composed of a period of anestrus, and a period of cyclic ovarian activity with one, two or three ovulations without behavioral estrus. In some ewes, the first cycle was of shorter duration, and its CL secreted less progesterone (P less than .05) relative to CL of silent and regular estrous cycles. Luteinizing hormone peaks were recorded as early as 6 d PP. When progesterone concentrations were elevated to luteal phase levels, the frequency, but not magnitude, of LH peaks per 4-h bleeding period was reduced (P less than .05) relative to anestrus. It is concluded that there are periods of anestrus and of silent cycles, which precede the first postpartum estrus in Pelibuey ewes.  相似文献

Pituitary-ovarian function was analyzed in a strain of miniature swine previously shown to produce a low ovulation rate resulting in the formation of only 8.6 corpora lutea (CL)/animal. Five multiparous (M) and four nulliparous (N) miniature pigs with a mean inbreeding coefficient of .39 were monitored for estrous behavior through four consecutive estrous cycles. Daily blood samples were collected from 5 d before to 5 d after the onset of the second, third and fourth estrus and at 48-h intervals during the remainder of the second and third estrous cycle. Laparoscopy was used to examine the ovaries 1 and 5 d after onset of the third estrus and 2 d after the beginning of the fourth estrus. For the entire group, temporal fluctuations among serum estradiol-17 beta, luteinizing hormone (LH) and progesterone concentrations and sexual behavior were similar to previously published data in standard swine breeds. Although the mean lengths of the estrous cycle were not different (P greater than .05) between parity subgroups (M, 23 +/- 1.3 vs N, 22 +/- .7 d), multiparous pigs were in estrus longer (P less than .05) than nulliparous females (M, 3.7 +/- .2 vs N, 2.2 +/- .4 d). Parity subgroups were similar with respect to the mean number of follicles forming CL (M, 8.8 +/- .7 vs N, 9.2 +/- .2). This study was undertaken to determine changes in follicular populations on ovaries of dairy cows during three stages of the estrous cycle and their steroidogenic capacity in vitro. Numbers of small (2.0 to 5.0 mm), intermediate (5.1 to 10 mm) and large (greater than 10 mm) antral follicles on ovaries of multiparous cows and heifers (n = 31) in the early luteal (d 4), mid-luteal (d 12) and follicular phase (d 19) of the estrous cycle were determined (d 0 = estrus), and steroidogenic capacity of intermediate and large follicles was measured in vitro. Total number of follicles and number of small follicles were greatest (P less than .05) on d 19 compared with d 12, with numbers on d 4 not different from either d 12 or 19. Intermediate follicles were fewer (P less than .05) on d 19 compared with d 4 or 12. Numbers of large follicles did not change. The proportion of estrogen active (EA) follicles was greater (P less than .05) on d 19 compared with d 4 or 12. Accumulation of estradiol-17 beta (E) into culture medium by intermediate follicles decreased (P less than .05) with increasing days of the estrous cycle, while accumulation of progesterone (P) was greater on d 19. In large follicles, accumulation of E into culture medium was greatest (P less than .05) on d 19 and the lowest on d 12 (P less than .05). Peripheral serum progesterone concentrations were evaluated in beef heifers following breeding collected on d 6 +/- 1, 9 +/- 1 collected on d 6 +/- 1, 9 +/- 1 and 12 +/- 1 (estrus = d 0) after the puberal estrus of all heifers and after the third estrus of E3 heifers. Progesterone concentrations were higher (P less than .05) for heifers in E1 compared with heifers in E3 on d 6, 9 and 12 after breeding to a fertile bull. Progesterone concentrations on d 6, 9 and 12 did not differ (P greater than .10) between pregnant heifers in E1 and E3; however, non-pregnant heifers in E1 had higher (P less than .05) concentrations of progesterone compared with non-pregnant heifers in E3 on each day. Concentrations of progesterone did not differ (P greater than .10) between non-pregnant heifers in E1 and heifers of E3 during their puberal cycle. Pregnant heifers in E1 and E3 had higher (P less than .05) concentrations of progesterone on each day than non-pregnant heifers in their respective treatments. There were no interactions (P greater than .10) between treatment, pregnancy status and day-of-estrous cycle for concentrations of progesterone. Results of this study indicated that luteal function differed between heifers that failed to conceive at their puberal estrus and heifers that failed to conceive at third estrus. However, concentrations of progesterone did not differ between heifers that conceived at puberal or third estrus. The relationship of changes in luteal function from the puberal through the third estrous cycle and pregnancy is not clear.  相似文献

During the fourth week postpartum, 443 healthy Holstein cows milked thrice daily were randomly divided among four groups to receive 0, 2, 8 and 32 micrograms of the GnRH analog HOE 766. Intervals from calving to first estrus and to first breeding, from breeding to conception and conception rates at first breeding were calculated to measure treatment response, and progesterone was measured in the fat-free portion of milk samples collected twice weekly during the first 4 wk following treatment. Uterine involution at the time of treatment was estimated by palpation per rectum. Twenty percent of the cows examined were classified as having delayed uterine involution (abnormal). By analyzing milk progesterone patterns it was determined that 38% of the animals were in the luteal phase of an estrous cycle when treated. Cows without luteal tissue (less than 1 ng of progesterone/ml milk) given 8 or 32 micrograms of HOE 766 increased in progesterone to greater than or equal to 1 ng/ml within 7 d in 77 and 72% of the cows compared with 40 and 57% for cows receiving 0 and 2 micrograms (P less than .05). This increase in progesterone was followed by a normal estrous cycle within 4 wk in a higher proportion of cows treated with the two higher doses of GnRH analog (87 and 86%) compared with 67 and 70% of those receiving 0 or 2 micrograms of the analog (P less than .005). There were no treatment differences (P greater than .05) for other traits analyzed, but cows with a normal progesterone cycle were observed in estrus and were bred sooner (P less than .01) than those with irregular progesterone patterns. It was concluded that the GnRH analog hastened the onset of normal ovarian cycles in cows milked thrice daily.  相似文献

The effect of pretreatment with flurogestone acetate (FA) on the lifespan of corpora lutea induced with pregnant mare serum gonadotropin (PMS) was examined in cycling and anestrous ewes. Cycling ewes received one of three treatments: 750 IU PMS 2 d before expected estrus (P), FA-impregnated vaginal sponges for 16 d (F), and FA sponges for 16 d and 750 IU PMS 2 d before sponge removal (FP). A fourth group served as controls (C). When compared with d 12 means within treatment, plasma progesterone means were lower (P less than .05) on d 16 in control ewes, on d 15 in P and F ewes, and on d 14 in FP ewes. Only 44% of ewes receiving FA treatment alone exhibited estrus (P less than .05) compared with 100% of untreated ewes. The FP treatment increased ovulation rate compared with controls (P less than .01). The decrease in luteal lifespan observed in cycling ewes suggests a possibility of asynchrony between the uterus and embryo, which could result in failure of an embryo to prevent luteal regression, thus resulting in reduced fertility. None of the seasonally anestrous ewes that received PMS alone and only 55% of those treated with FA sponges for 8 d before PMS injection exhibited estrus. Ewes pretreated with FA exhibited higher plasma progesterone concentrations on d 10 through 16 after PMS injection. There were no differences in luteal lifespan as measured by peripheral plasma progesterone patterns. Although FA treatment did not alter luteal lifespan in anestrous ewes, the increased plasma progesterone concentrations observed with FA treatment suggest that progestogen pretreatment may be essential for optimal luteal function.  相似文献

The objectives of this study were to determine 1) the ability of charcoal-extracted bovine follicular fluid (bFF) to suppress endogenous follicle stimulating hormone (FSH) at various stages of the estrous cycle and 2) the effects of suppression of FSH on luteal function and lengths of the current and subsequent estrous cycles. Twenty-six mature ewes were assigned randomly to receive 5 ml of either bFF or saline, subcutaneously, at 8-h intervals on d 1 through 5 (bFF n = 6; saline n = 3), d 6 through 10 (bFF n = 6; saline n = 3) or d 11 through 15 (bFF n = 6; saline n = 2) of the estrous cycle (d 0 = estrus). Blood was collected daily beginning at estrus and continued until the third estrus (two estrous cycles) or 40 d; more frequent samples were collected 2 h prior to initiation of treatment (0600), hourly for the first 8 h of treatment, then every 4 h until 0800 on the first day after treatment, and finally at 1600 and 2400 on that day. Plasma concentrations of FSH were lower (P less than .001) in bFF-treated than in saline-treated ewes. Treatment with bFF reduced (P less than .05) plasma concentrations of progesterone during the current but not during the subsequent estrous cycle. Treatment with bFF did not affect plasma concentrations of estradiol-17 beta. Administration of bFF on d 11 through 15 of the estrous cycle lengthened the interval from the decline in progesterone to estrus and the inter-estrous interval by approximately 3 and 4 d, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The frequency of occurrence of behavioral estrus without subsequent development of functional luteal tissue (termed nonpuberal estrus, NPE), was determined in 43 Simmental X Hereford-Brahman heifers. Blood samples were collected weekly from the start of the study to first behavioral estrus and then daily from d 1 (d 0 = estrus) through d 14 following first and subsequently observed estrous behaviors. All blood samples were analyzed for serum progesterone (P4) concentrations by radioimmunoassay. More heifers (62.8%) exhibited NPE than had luteal development after their first behavioral estrus (37.2%). There was a tendency for fewer light-weight heifers (less than or equal to 240 kg at the start of the experiment) to exhibit a puberal first estrus compared with the heavy-weight (greater than 240 kg at the start of the experiment) heifers (31.2% vs 68.8%, respectively; P = .12). Heifers that had a puberal first estrus were older (376 +/- 12 d vs 334 +/- 9 d, P less than .05) compared with heifers that had NPE. Weight at first behavioral estrus was similar between heifers that had a puberal first estrus and those that had NPE (298 +/- 8 kg and 289 +/- 6 kg, respectively). More heifers that had a puberal first estrus also had an elevation in serum P4 concentrations before that first estrus (64.3% vs 20.0%, P less than .05), and the serum P4 elevation was greater (2.5 +/- .4 ng vs 1.2 +/- .1 ng, P less than .05) than heifers that had NPE. Delaying the onset of estrus after weaning and adding fat to the postweaning diet were studied for their effects on estrus and fertility in 232 crossbred primiparous sows on a commercial swine farm. Sows were assigned randomly to the following treatments after weaning in June, July, August, or September, 1983: 1) altrenogest (20 mg/d) was fed for 7 d after weaning (n = 76), 2) altrenogest was fed for 7 d plus .53 kg dried animal and vegetable fat product (.45 kg actual fat/d) for 14 d after weaning (n = 78), or 3) no treatment (controls, n = 78). While a similar proportion of sows came into heat after weaning (lactation length = 4 wk), sows fed altrenogest (14.4 +/- .2 d) returned to estrus about 9 d later (P less than .01) than controls (5.6 +/- .2 d). Serum progesterone concentration was assayed in blood samples collected from a subgroup (74%) of the cows not observed in estrus by 3 wk after weaning to determine possible causes of anestrus. If serum progesterone (greater than 5 ng/ml) was elevated, we assumed that sows had ovulated without expressing estrus (behavioral anestrus) or ovulated with undetected estrus (less overt estrus or error in estrous detection), whereas low progesterone (less than or equal to 5 ng/ml) indicated that sows were anovulatory. About 53% of the sows not observed in estrus across all treatments had luteal function, probably resulting from post-weaning ovulation. Incidence of anovulation without estrus was 47%. Changes in follicular and luteal structures were assessed and concentrations of estradiol and progesterone were measured in 13 Hereford X Angus suckled beef cows during resumption of estrous cycles. Transrectal ultrasonography was used to monitor follicular size, ovulation, and formation and regression of the corpus luteum (CL). The interval from parturition to first postpartum ovulation (FO) was 82 +/- 4.7 d. Serum progesterone remained low before FO. One cow exhibited standing estrus, two cows showed other signs of estrus, and 10 displayed no signs of behavioral estrus preceding FO. All cows exhibited standing estrus before the second postpartum ovulation (SO). All cows had a short luteal phase after FO, with an average interval of 8.5 +/- .2 d between FO and SO. Concentrations of estradiol in serum during the 8 d preceding ovulation were similar before FO and SO. Maximal diameter of the preovulatory follicle was similar before FO and SO. However, the ovulatory follicle was larger in diameter at 2 d (P = .02) and 3 to 8 d (P less than .005) before FO than before SO. The time from detection until ovulation was less (P = .005) for the ovulatory follicle preceding SO than for the follicle associated with FO (8.5 vs 10.2 d, respectively, SE = .4). The second-largest follicle was larger (P less than .005) in diameter during the 8 d preceding the FO than before the SO. Thirty-four multiparous, lactating, cyclic beef cows which calved in moderate body condition were used to determine effects of restricted nutrition on corpus luteum (CL) development and endocrine status. At 78 d postpartum, six cows were assigned to a control (CON) diet (26.0 Mcal ME), fed to increase bodyweight (BW) and body condition score (BCS), and the remaining 28 cows were fed to lose BW and BCS on a restricted (RES) diet (14.0 Mcal ME). Following a 40-d adjustment period on respective diets, estrous cycles were synchronized and cows bled daily for determination of progesterone (P4), luteinizing hormone (LH) and insulin (INS) beginning at the synchronized estrus. Ultrasonography was used to determine the ovulatory follicle and CL development. Control cows were maintained for one estrous cycle and were ovariectomized on day 11 of their second cycle. Ten cows on restricted diet (RES-C) continued to form a functional CL (P4 > 1.5 ng/ml at day 10 of an estrous cycle) through as many as 5 cycles, after which observations were discontinued. Fourteen cows on restricted diet (RES-A) were ovariectomized on day 11 of a cycle when a CL was identified by ultrasonography, but was subfunctional (P4 < 1.5 ng/ml on day 10 of that cycle). Four additional RES-A cows which had subfunctional CL were not ovariectomized but were bled for an additional 25 d. At ovariectomy, CL and ovarian weights were collected. Luteal tissue was prepared for evaluation of P4 synthesis, LH responsiveness in vitro, and for determination of P4 content and total LH receptors. Bodyweight and BCS increased in CON cows; whereas, RES cows lost BW and BCS (P < .05). In the cycle prior to ovariectomy, serum P4 and LH were not different in 18 RES-A cows which developed subfunctional CL in comparison to CON cows. Four RES-A cows not ovariectomized but bled for an additional 25 d neither exhibited estrus, ovulated, nor had P4 concentrations greater than .3 ng/ml. Serum INS was lower in RES-A cows during the cycle prior to ovariectomy than in CON cows (P < .05). During the 11-d period prior to ovariectomy, mean serum P4 and INS were lower in RES-A cows than in CON cows (P < .05); however, serum LH was not different. Furthermore, CL and ovarian weights, P4 content of CL, secretion of P4 by luteal tissue in response to LH in vitro and LH receptor number were not different between CON and RES-A cows. In conclusion, nutritional anestrus may be preceded by the formation of a CL with lower steroidogenic output in vivo. However, luteal tissue, collected from RES-A cows, did not appear to be subfunctional during in vitro incubation when substrate availability and gonadotropin support were equal between diets.  相似文献

Effect of transportation on estrous behavior, duration of the estrous cycle, ovulation, pregnancy rates and concentrations of serum cortisol, plasma ascorbic acid (AA), LH, estradiol and progesterone in mares was investigated. Fifteen mares were transported for 792 km (12 h) during the preovulatory stage of estrus. Transported mares were bled immediately before transport (baseline), at midtrip and 0, 12, 24, 48 and 72 h post-transport and twice daily from d 1 before transport to d 1 (estrogen) or 3 (LH) post-ovulation. Blood samples also were taken for progesterone on d 0, 2, 6, 10, 15, 16, 17, 18, 19 and 20 post-ovulation. Nontransported control mares (n = 15) were bled on the same schedule as transported mares. There was no difference (P greater than .05) in number of mares ovulating, estrous behavior, duration of the estrous cycle or pregnancy rate between groups. Cortisol in transported mares increased to concentrations greater (P less than .05) than those in control mares at midtrip and 0 h post-transport. Concentrations of AA in transported mares also increased (P less than .05) at midtrip, then decreased (P less than .05) below baseline at 24 h post-transport. Concentrations of LH and estradiol increased (P less than .05) above baseline throughout the blood-sampling period. Increases apparently were due to preovulatory surges of these hormones. This study was designed to determine if luteal cell receptors for luteinizing hormone/human chorionic gonadotropin (LH/hCG) contribute to the previously demonstrated abnormal function of induced corpora lutea (CL) in gilts. Twenty-five prepuberal (P) gilts, induced to ovulate with 1,500 IU pregnant mare serum gonadotropin followed 72 h later with 500 IU hCG (d 0 = day of hCG), and 22 mature (M) gilts that had displayed two or more estrous cycles were ovariectomized (OVX) on d 10, 14, 18, 22 or 26 after the onset of estrus. All gilts except those OVX on d 10 were hysterectomized between d 6 and 9 to ensure luteal maintenance. The CL were stored at -196 degrees C until determination of LH/hCG receptor number and dissociation constant (KD) by saturation analysis. Receptor number was greater for M than for P gilts on d 14 (P less than .07) and d 18 (P less than .01). The KD was greater in M than in P gilts on d 14 (P less than .01) and d 18 (P less than .0001). The LH/hCG receptor number and KD of P gilts remained the same throughout the days studied. The LH/hCG receptor number (fmol/mg protein) of M gilts was elevated on d 10, 14, and 18 (50.8, 50.4 and 51.4, respectively) and decreased on d 22 (26.5) and d 26 (25.4) to values similar to those of P gilts. In M gilts, KD increased on d 14, remained high on d 18 and decreased on d 22. We suggest that abnormal function of induced CL in P gilts may be due to an elevated LH receptor number.  相似文献

This study was conducted to examine the effects of metestrus administration of SyncroMate-B (SMB) on PGF2alpha secretion and corpus luteum (CL) development. In a study replicated over 2 yr, cows were observed for spontaneous estrus in yr 1, and cows received an injection of 25 mg of PGF2alpha and were observed for subsequent estrus in yr 2. At standing estrus (estrus = d 1), cows were randomly allotted to receive either the standard SMB regimen (n = 40) on d 3 of the estrous cycle or no treatment (n = 8). Fifty percent (n = 20) of SMB-treated cows were administered PGF2alpha on d 10 of the estrous cycle 48 h prior to implant removal. Twice-daily blood samples were collected in the morning (AM) and evening (PM) from d 2 AM through d 14 AM of the treated estrous cycle and subsequently analyzed for progesterone (P4) and PGF2alpha metabolite (PGFM). Prior to statistical analysis, SMB- and SMB/PGF2alpha-treated cows were sorted according to P4 concentration at d 10 of the treated estrous cycle to either a CL functional group (P4 > or = 1 ng/mL; n = 20) or a CL nonfunctional group (P4 < 1 ng/mL; n = 17). Following d 10 AM administration of PGF2alpha, functional and nonfunctional groups were further subdivided based on treatment. The groups were as follows: untreated control cows (n = 8); SMB-treated cows retaining a functional CL (SMB-F; n = 8); SMB-treated cows with a nonfunctional CL (SMB-N; n = 11); SMB/PGF2alpha-treated cows retaining a functional CL (SMB/PG-F; n = 12); and SMB/PGF2alpha-treated cows with a nonfunctional CL (SMB/PG-N; n = 6). Of all SMB-treated cows, 54% retained a functional CL through d 10 AM of the treated estrous cycle. Mean serum P4 concentrations increased for cows in all groups until d 7, after which P4 concentrations increased for cows in SMB/PG-F, SMB-F, and control groups and decreased for cows in SMB/PG-N and SMB-N groups. Following PGF2alpha administration on d 10, mean serum P4 concentrations remained < 1 ng/mL for cows in SMB/PG-N and SMB-N groups, decreased to < 1 ng/mL for cows in the SMB/ PG-F group, and remained > 1 ng/mL for cows in SMB-F and control groups. Mean serum PGFM concentrations tended (P = .06) to increase in cows with nonfunctional CL compared with control cows on d 8 AM and were greater (P < .05) in cows with functional CL on d 8 PM through d 9 PM. These results indicate that retention of a functional rather than a nonfunctional CL following metestrus administration of SMB is dependent on a premature release of uterine PGF2alpha.  相似文献
