The purpose of this study was to determine the pharmacokinetics of baicalin after intravenous and intramuscular administration of sodium baicalin at 50 mg/kg to piglets. Plasma baicalin levels were determined by high‐performance liquid chromatography. The plasma concentration–time data of baicalin for both administration routes were best described by two‐compartmental open model. The area under the plasma concentration–time curve and the elimination half‐lives were 77.47 ± 6.14 µg/ml × h and 1.73 ± 0.16 hr for intravenous and 64.85 ± 5.67 µg/ml × h and 2.42 ± 0.15 hr for intramuscular administration, respectively. The apparent volume of distribution and body clearance were 1.63 ± 0.23 L/kg and 2.74 ± 0.30 L h?1 kg?1 for intravenous and 0.51 ± 0.10 L/kg and 0.78 ± 0.08 L h?1 kg?1 for intramuscular routes, respectively. An intramuscular injection of sodium baicalin in piglets resulted in rapid and complete absorption, with a mean maximal plasma concentration of 77.28 ± 7.40 µg/ml at 0.17 hr and a high absolute bioavailability of 83.73 ± 5.53%.  相似文献

The dose-related antinociceptive effects of intravenous (IV) buprenorphine were evaluated in cats. Thermal (TT) and mechanical threshold (MT) devices were used for nociceptive stimulation. After baseline threshold recordings, buprenorphine was administered IV (0.01, 0.02 or 0.04 mg/kg; B1, B2 and B4, respectively) in a randomised, blinded and cross-over study. Data were analysed by ANOVA (P < 0.05) using 95% confidence intervals (CI). TT increased 15, 30, 45 min and 1 (5.2 ± 2.7 °C), 2, 3 and 4 h after B1; 15, 30, 45 min and 1 (5.1 ± 3.9 °C) and 2 h after B2, and 15, 30, 45 min and 1 (5.4 ± 3.3 °C), 2, 3, 6 and 8 h after B4. MT increased 15 and 45 min after B2 (260 ± 171 mmHg), and 30 (209 ± 116 mmHg) and 45 min and 1 and 2 h after B4. At 45 min, MT values were significantly higher after B2 compared to B1 (P < 0.05). With MT, B2 and B4 produced more antinociception and longer duration of action than B1, respectively. No dose response to thermal stimulation was detected.  相似文献

The objective was to assess the effect of banding or burdizzo castration performed on farms on plasma testosterone, acute-phase proteins, scrotal circumferences, growth, and well-being of bulls. 243 Continental bulls (12 months; 399.2 ± 5.72 kg) from three different farms were allocated at random, after stratification on weight within breed type, to one of three treatment groups: banding castration (BAND; n = 80), burdizzo castration (BURD; n = 83), or controls (CON; n = 80). The castration methods were conducted under local anaesthesia, and tetanus toxoid vaccine and antibiotic were also injected at castration. BAND and BURD castrates had lower (P < 0.001) plasma testosterone concentration than control bulls, with no difference between BAND and BURD castrates on 28 d post-castration. From days 0 to 14 post-castration, BAND (P = 0.0002) and BURD (P < 0.0001) castrates had lower average daily gain (ADG) than CON bulls, no difference (P = 0.46) was found between BAND and BURD castrates. From days 15 to 28, BAND castrates had lower ADG compared with BURD castrates (P = 0.03) and CON bulls (P = 0.01), while no difference (P = 0.76) was found between BURD and CON. From days 29 to 56, BAND (P = 0.01) and BURD (P = 0.002) castrates had lower ADG than CON bulls, no difference (P = 0.55) was found between BAND and BURD. From days 57 to 84, the ADG of BAND castrates was not different compared with BURD castrates (P = 0.12) and CON bulls (P = 0.38), while BURD had lower (P = 0.02) ADG compared with CON. The integrated ADG from day 0 to 112 of BAND (P = 0.0001) and BURD (P = 0.02) groups were lower compared with CON, while there was no difference (P = 0.09) between BAND and BURD castrates. On d 14 post-castration, BAND castrates had lower scrotal temperature than BURD (P < 0.0001) and CON (P < 0.0001), and BURD castrates had greater (P < 0.006) scrotal temperature than CON; BAND castrates had lower scrotal latitudinal and longitudinal circumferences than BURD castrates (P < 0.001) and CON bulls (P < 0.001), and BURD castrates had greater (P < 0.001) scrotal latitudinal and longitudinal circumferences than CON bulls. BAND (P < 0.0001) and BURD (P = 0.01) castrates had greater glucose concentration than CON bulls, and BAND castrates had greater (P = 0.04) glucose concentration than BURD. In conclusion, BAND or BURD castration significantly reduced plasma testosterone concentration; reduced average daily weight gain mainly during the first 2 weeks, which was not compensated during the subsequent 16 weeks; increased withdrawal of stored energy and increased plasma protein concentration. BURD showed an advantage over BAND in growth during days 15 to 28 following castration.  相似文献

Probiotics of Lactobacillus acidophilus, Saccharomyces cerevisiae, and Aspergillus niger and three commercial exogenous fibrolytic enzymes (EFE) were tested in vitro to select best source and optimum dose followed by in vivo studies on male buffalo calves. Bacterial (P < 0.001) and protozoal population were increased significantly (P < 0.001) with probiotics and EFE. In vitro dry matter digestibility was more (P < 0.001) on L. acidophilus and then on S. cerevisiae. Dose required for L. acidophilus and S. cerevisiae probiotics was 1 × 109 and 3 × 109 cfu/flask, respectively. Cellulase and xylanase were effective at 4,000 and 12,500 IU/kg DM. In vitro cell wall digestibility was increased (P < 0.001) when probiotics and EFE were used together. Best source and optimum dose of probiotics and EFE were fed to 18 male buffalo calves with concentrate supplement (CS). Calves were randomly divided into three groups either without probiotics and EFE (CG) or with probiotics (EG1) or probiotics combined with EFE (EG2) on wheat straw diet. Organic matter, neutral detergent fiber, and acid detergent fiber digestibility was improved significantly. Average daily weight gain (ADG) and feed efficiency were significantly higher (P < 0.001) in EG2 than EG1 or CG. Final body weight was 4% and 12% and feed efficiency was 2.6% or 1.6% more (P < 0.001) in EG2 compared to CG or EG1, respectively. Fortification of CS with probiotics and EFE together had more impact on FE and ADG in buffalo calves.  相似文献

ObjectiveTo determine the specific lung elastance (SEL) in anesthetized dogs and to evaluate the efficacy of a SEL-based recruiting airway pressure (RPaw) at improving global and regional lung aeration.Study designRetrospective and prospective clinical study.AnimalsA total of 28 adult dogs were included in the retrospective study and six adult dogs in the prospective study.MethodsRetrospective study: SEL and SEL-based RPaw were determined using previously published data. In mechanically ventilated dogs undergoing thoracic computed tomography (CT), SEL was calculated as ΔPL/(VT/EELV), where ΔPL is the driving transpulmonary pressure, VT is the tidal volume and EELV is the end-expiratory lung volume. The ratio of lung to respiratory system elastance (EL/Ers) was determined. SEL and EL/Ers were used to calculate the SEL-based RPaw. Prospective study: dogs underwent thoracic CT at end-expiration and at end-inspiration using the SEL-based RPaw, and global and regional aeration was determined. For analysis of regional aeration, lungs were divided into cranial, intermediate and caudal regions. Regional compliance was also calculated. A p value <0.05 was considered significant.ResultsThe SEL and EL/Ers were 12.7 ± 3.1 cmH2O and 0.54 ± 0.07, respectively. The SEL-based RPaw was 29.1 ± 7.6 cmH2O. In the prospective study, the RPaw was 28.2 ± 1.3 cmH2O. During RPaw, hyperinflation increased (p = 0.0003) whereas poorly aerated (p < 0.0001) and nonaerated (p = 0.01) tissue decreased. Normally aerated tissue did not change (p = 0.265). Regional compliance was higher in the intermediate (p = 0.0003) and caudal (p = 0.034) regions compared with the cranial region. Aeration did not differ between regions (p > 0.05).Conclusions and clinical relevanceAn SEL-based RPaw reduces poorly and nonaerated lung tissue in anesthetized dogs. In nonsurgical anesthetized dogs, an RPaw near 30 cmH2O is effective at improving lung aeration.  相似文献

Cardiopulmonary and sedative effects of intravenous or epidural methadone were compared. Six beagles were randomly assigned to group MIV (methadone 0.5 mg/kg IV + NaCl 0.9% epidurally) or MEP (methadone 0.5 mg/kg epidurally + NaCl 0.9% IV). Cardiopulmonary, blood gas and sedation were assessed at time (T) 0, 15, 30, 60, 120, 240 and 480 min after drug administration. Compared to T0, heart rate decreased at T15–T120 in MIV (< .001) and T15–T240 in MEP (< .05); mean arterial pressure was reduced at T15–T60 in MEP (< .01); respiratory rate was higher at T15 and T30 in both groups (< .05); pH was lower at T15–T120 in MIV (< .01) and T15, T30 and T120 in MEP (< .05); PaCO2 was higher at T15–T60 in MIV (< .01) and T15, T30 and T120 in MEP (< .01); sedation scores were higher at T15 and T30 in MIV and T15–T60 in MEP (< .05). At T120 and T240, sedation score was higher in group MEP compared with group MIV (< .01) In conclusion, cardiopulmonary and sedative effects of identical methadone doses are similar when administered IV or epidurally to conscious healthy dogs.  相似文献

This study determined the pharmacokinetics and compared the clinical effects of xylazine and dexmedetomidine in horses recovering from isoflurane anesthesia. Six healthy horses aged 8.5 ± 3 years and weighing 462 ± 50 kg were anesthetized with isoflurane for 2 hr under standard conditions on two occasions one-week apart. In recovery, horses received 200 μg/kg xylazine or 0.875 μg/kg dexmedetomidine intravenously and were allowed to recover without assistance. These doses were selected because they have been used for postanesthetic sedation in clinical and research studies. Serial venous blood samples were collected for quantification of xylazine and dexmedetomidine, and the pharmacokinetic parameters were calculated. Two individuals blinded to treatment identity evaluated recovery quality with a visual analog scale. Times to stand were recorded. Results (mean ± SD) were compared using paired t tests or Wilcoxon signed-ranked test with p < .05 considered significant. Elimination half-lives (62.7 ± 21.8 and 30.1 ± 8 min for xylazine and dexmedetomidine, respectively) and steady-state volumes of distribution (215 ± 123 and 744 ± 403 ml/kg) were significantly different between xylazine and dexmedetomidine, whereas clearances (21.1 ± 17.3 and 48.6 ± 28.1 ml/minute/kg), times to stand (47 ± 24 and 53 ± 12 min) and recovery quality (51 ± 24 and 61 ± 22 mm VAS) were not significantly different. When used for postanesthetic sedation following isoflurane anesthesia in healthy horses, dexmedetomidine displays faster plasma kinetics but is not associated with faster recoveries compared to xylazine.  相似文献

Cefuroxime axetil pharmacokinetic profile was investigated in 12 Beagle dogs after single intravenous and oral administration of tablets or suspension at a dose of 20 mg/kg, under both fasting and fed conditions. A three-period, three-treatment crossover study (IV, PO under fasting and fed condition) was applied. Blood samples were withdrawn at predetermined times over a 12-hr period. Cefuroxime plasma concentrations were determined by HPLC. Data were analyzed by compartmental analysis. No statistically significant differences were observed between formulations and feeding conditions on PK parameters. Independently of the feeding condition, absorption of cefuroxime axetil after tablet administration was low and erratic. The drug has been quantified in plasma in 3 out of 6 and 5 out of 6 dogs in the fasted and fed groups. For this formulation, the bioavailability (F), peak plasma concentration (Cmax), and area under the concentration–time curve (AUC) of cefuroxime axetil were significantly enhanced (p < .05) by the concomitant ingestion of food (32.97 ± 13.47–14.08 ± 7.79%, 6.30 ± 2.62–2.74 ± 0.66 µg/ml, and 15.75 ± 3.98–7.82 ± 2.76 µg.hr/ml for F, Cmax, and AUC in fed and fasted dogs, respectively), while for cefuroxime axetil suspension, feeding conditions affected only the rate of absorption, as reflected by the significantly shorter absorption half-life (T½(a)) and time to peak concentration (Tmax) (0.55 ± 0.27–1.15 ± 0.19 hr and 1.21 ± 0.22–1.70 ± 0.30 for T½(a) and Tmax in fed and fasted dogs, respectively). For cefuroxime axetil tablets, T > MIC (≤1 µg/ml) was <2 hr in fasted and ≈4 hr in fed animals, and for cefuroxime axetil suspension, T > MIC (≤1 µg/ml) was ≈5 hr and for T >MIC (≤4 µg/ml) was ≈2.5 hr for fasted and fed dogs, respectively. Cefuroxime axetil as a suspension formulation seems to be a better option than tablets. However, its short permanence in plasma could reduce its clinical usefulness in dogs.  相似文献

Pharmacokinetics and milk levels of ceftriaxone were studied in healthy and endometritic cows following single intravenous administration. The drug was detected up to 8 h of dosing in plasma of healthy and endometritic cows and the drug disposition followed three-compartment open model. The values of Vdarea, AUC, t1/2β, ClB, MRT and P/C ratio were 0.50 ± 0.19 L.kg−1, 62.2 ± 23.3 μg.ml−1.h, 1.02 ± 0.07 h, 0.30 ± 0.09 L.kg−1.h−1, 1.55 ± 0.25 h and 0.52 ± 0.27, respectively, in healthy and 1.55 ± 0.52 L.kg−1, 37.0 ± 17.1 μg.ml−1.h, 1.56 ± 0.25 h, 0.56 ± 0.14 L.kg−1.h−1, 2.14 ± 0.34 h and 1.44 ± 0.60, respectively, in endometritic cows. The drug was detected in milk for 36 h after administration. For MIC90 of 0.5 μg.ml−1 the most appropriate dosage for ceftriaxone, would be 9.0 mg.kg−1 repeated at 6 h intervals for the treatment of endometritis in cows.  相似文献

The aim of this work was to evaluate the effect of relaxin on fertility parameters of buffalo frozen/thawed sperm. Sperm were incubated in the absence of capacitating agents (negative control), with a known capacitating agent such as heparin (positive control) and with 50 and 100 ng/ml relaxin for 2 and 4 h. Sperm viability, motility, capacitation and the effect of relaxin on the fertilizing ability after heterologous IVF were evaluated. Although viability was not affected, relaxin increased (p < 0.05) sperm motility compared to the negative and positive controls both after 2 h (60.0 ± 2.0, 60.0 ± 3.1, 68.3 ± 1.7 and 69.4 ± 2.7, respectively, in negative control, positive control, 50 and 100 ng/ml relaxin) and 4 h (55.0 ± 2.5, 53.3 ± 3.0, 62.2 ± 3.0 and 65.0 ± 3.2, respectively, in negative control, positive control, 50 and 100 ng/ml relaxin) incubation. When sperm were incubated with both 100 ng/ml relaxin and heparin, a decrease (p < 0.01) of pattern A, that is low capacitation level, was observed compared to the negative control both after 2 h (54.4, 34.3 and 36.4%, respectively, in negative control, positive control and 100 ng/ml relaxin) and 4 h (51.9, 35.0 and 34.3%, respectively, in negative control, positive control and 100 ng/ml relaxin). Moreover, an increase (p < 0.01) of pattern EA, that is high capacitation level, was recorded with 100 ng/ml relaxin and heparin compared to the negative control both after 2 h (44.1, 59.3 and 57.7%, respectively, in negative control, positive control and 100 ng/ml relaxin) and after 4 h (43.0, 54.4 and 56.0%, respectively, in negative control, positive control and 100 ng/ml relaxin). Finally, relaxin increased (p < 0.01) cleavage rate compared to the negative control (57.1 ± 4.4, 72.5 ± 6.0, 71.4 ± 5.5 and 73.6 ± 2.9, respectively, in negative control, positive control, 50 and 100 ng/ml relaxin). In conclusion, relaxin has a beneficial effect on motility, capacitation and fertilizing ability of frozen–thawed buffalo sperm.  相似文献

Effect of extended storage on egg quality, embryo mortality and hatchability in FUNAAB-ɑ chickens was determined. Hatchable eggs (n = 288; weighing 53.2 ± 4.67 g) collected from a flock of FUNAAB-ɑ layer breeder hens aged 32 weeks were stored in egg tray with broad end up under 16 ± 1.5°C for either 0, 4, 8, 12, 16 or 20 d. Before incubation, eight eggs from each group were evaluated for internal and external quality traits. Remaining eggs were set in an incubator and transferred into hatcher on embryonic day 18. Data collected were subjected to one-way analysis of variance. Egg weight loss (EWL; p < .001), surface area (p < .001), yolk diameter (p < .001), inner and outer blastoderm diameters (p < .05) and dead in germ (DIG; p < .001) increased with storage duration while yolk height (p < .001), yolk index (p < .001), albumen weight (p < .05), albumen height (p < .05), albumen index (p < .01), Haugh's unit (HU; p < .05), fertility (p < .001), hatchability of set (HATCHS; p < .001) and fertile eggs (p < .05) decreased. Weight losses of 0, 1.2, 2.2, 3.4, 4.6 and 6.1% were recorded in egg stored for 0, 4, 8, 12, 16 and 20 days respectively. Eggs stored beyond 8 days exhibited higher DIG and lower HATCHS. Shell percentage in 4 days storage (11.4%) was lower (p < .05) than in 16 days storage (13.4%). Shell thickness was similar in eggs stored for 0 to 12 days, but 8 days storage (0.60 mm) had thinner (p < .01) shell than day 16 (0.71 mm) and day 20 (0.73 mm) storage. Internal quality unit (IQU) was higher (p < .05) in fresh eggs (180.4) than in 12 days (167.8) and 20 days (167.8) stored eggs. Extended storage of FUNAAB-ɑ eggs caused EWL, surface area shrinkage, lowered HU and IQU, loss of yolk and albumen quality, increased blastoderm diameters and DIG, and decreased egg fertility and HATCHS from day 8 forward. Storing FUNAAB-ɑ eggs beyond 8 days reduced quality parameters; therefore, other mitigating factors are recommended when storing beyond 8 days.  相似文献

This study was to investigate the effect of oxidized wheat gluten (OG) on growth performance, gut morphology and its oxidative states of broilers. One hundred and eighty‐day‐old male broilers (10 chicks/pen) were randomly allocated into three dietary treatments: control diet (CON), diet with 8% wheat gluten (WG) and diet with 8% OG with six pens/treatment. Body weight (BW) (21 and 35 days) and average daily gain (ADG) (1–21 days and 22–35 days) decreased (p < .05) and feed conversion ratio (FCR) (1–21 days and 22–35 days) increased (p < .05) in OG treatment. Feed intake (FI) decreased (p < .05) in WG and OG treatments during 22–35 days. However, FI was not influenced by dietary treatments during 1–21 days (p > .05). The OG‐fed broilers had a lower faecal pH value (p < .05) and higher faecal moisture content (p < 05) at 14, 21, 28 and 35 days. Villus height, crypt depth and V/C value were not different (p > .05) among treatments at 21 and 35 days. Lipid peroxidation (LPO) (21 and 35 days) and malondialdehyde (MDA) (35 days) content in crop of OG treatment increased (p < .05). Oxidized glutathione (GSSG) (21 days), LPO (21 and 35 days) and MDA (21 and 35 days) content in ileum of OG treatment increased (p < .05). The reduced glutathione/oxidized glutathione (GSH/GSSG) (21 days) and (GSH) (35 days) in ileum of OG treatment decreased (p < .05). The present findings indicate that OG might be a stressor for broiler gut, which could induce oxidative stress both in crop and in ileum, and the diarrhoea as well. The growth performance of broiler was consequently depressed.  相似文献

Effect of Foot and Mouth disease (FMD) vaccination was studied on semen quality parameters of 19 Karan Fries (KF) and eight Murrah (MU) breeding bulls during the period 2002 to 2004 at Artificial Breeding Complex, NDRI, Karnal. A total of non-vaccinated 155 KF and 72 MU bulls' ejaculates were taken as control, while 169 KF and 51 MU bulls' ejaculates, collected after vaccination, were used to study the effect of vaccination stress. The results showed that FMD vaccination had no significant (P > 0.05) effect on ejaculate volume and total volume per day of semen in both KF and MU bulls. Volume of semen increased slightly during post-vaccination period in both the breeds. After FMD vaccination, there was significant (P < 0.01) decrease in mass activity (2.27 ± 0.06 vs. 1.67 ± 0.07 and 2.49 ± 0.09. vs. 1.75 ± 0.10, for KF and MU, respectively), initial motility (56.89 ± 0.03% vs. 44.62 ± 0.02% and 62.26 ± 0.04% vs. 47.08 ± 0.05%, for KF and MU, respectively), sperm concentration (754.19 ± 23.96 vs. 554.14 ± 22.95 × 106/ml and 848.61 ± 33.65 vs. 571.57 ± 39.99 × 106/ml, for KF and MU, respectively), and total sperm output per ejaculate (3,685.94 ± 158.40 vs. 2,781.54 ± 151.70 × 106 and 2,218.75 ± 133.14 vs. 1,582.84 ± 158.20 × 106, for KF and MU, respectively). Application of FMD vaccine had significantly (P < 0.05) adverse effect on most of the seminal attributes during post-vaccination in KF and MU buffalo bulls. So, the spermiograms affected following vaccination suggest that in bovines, the semen collection and preservation should be suspended till normal fertility of sperm is restored to avoid the failure of conception from artificial insemination using such semen.  相似文献

Basal concentration, production rate, metabolic clearance rate and T12 for oxytocin were measured in Holstein cows at three lactational stages and during mammary involution. At each lactational stage, oxytocin was given intravenously at .5 IU/min or 1.0 IU/min for 60 min. Infusions were preceded by priming. During involution, the high dose was used. Mean basal concentrations of oxytocin ranged from 8.7 to 21.4 uU/ml. Mean basal values at early, middle, late lactation and involution were 17.57, 12.33, 15.15, 21.13 uU/ml, respectively and differed significantly. The mean “rapid” T12 was 3.87 ± .1 min. Early, middle, late lactation and involution T12s were 4.2, 3.7, 4.0 and 3.5 min, respectively. The rapid T12 was not affected by lactational stage. The mean “slow” T12 was 25.53 ± 1 min. Early lactation and dry period means differed significantly. The overall mean oxytocin clearance rate was 8.41 ± .1 ml/kg·min. Clearance rate declined through lactation and into the dry period. Mean values of 9.3, 8.8, 7.8 and 7.1 ml/kg·min were obtained at early, middle, late lactation and involution, respectively. Clearance rates at late lactation and involution differed significantly from one another and from the early and middle stages. Mean entry rates for oxytocin at early, middle, late lactation and involution were 168.79, 106.03, 111.1 and 146.8 uU/kg·min, respectively. Measurements at early lactation and involution were greater than values for middle and late lactation. To summarize, basal oxytocin concentrations can be measured in cows that are lactating or undergoing mammary involution and changes in concentrations during lactation are related to hormone production (entry rate) and metabolic clearance rates.  相似文献

1. The objective of this work was to determine the effect of slaughter age and stunning method on the quality of turkey meat from poultry processing plants.

2. One hundred B.U.T. Premium turkeys were divided into 4 groups of 25 animals according to slaughter age (15 or 17 weeks) and CO2 stunning procedure (G1 stepwise: step1: 30% CO2, 15 s; step 2: 55% CO2, 40 s; step 3: 70% CO2, 45 s; G2: progressive increase of the CO2 concentration at a rate of 0.8% per s for 100 s). The quality of the breast meat was determined in fillets taken at different post-mortem times.

There were differences between the stunning groups for several variables (pH: 6.01 ± 0.01 and 5.95 ± 0.02; a*: ?1.84 ± 0.05 and ?2.21 ± 0.04; b*: 4.99 ± 0.15 and 4.68 ± 0.16; drip loss: 0.85 ± 0.02 and 0.71 ± 0.02 for G1 and G2, respectively), while no significant differences were found for L*, cooking loss and texture analysed with a Warner Bratzler Shear Force cell (WBSF).

Slaughter age had a significant effect on all the parameters studied (pH: 6.01 ± 0.01 and 5.95 ± 0.01; a*: ?2.21 ± 0.05 and ?1.88 ± 0.05, b*: 5.50 ± 0.17 and 4.42 ± 0.15; drip loss: 0.71 ± 0.02 and 0.86 ± 0.02, cooking loss: 12.56 ± 0.22 and 14.69 ± 0.16 for turkeys slaughtered at 15 and 17 w, respectively) except on L* and WBSF.

The ageing of the meat affected pH, colour values, drip loss and WBSF, with differing degrees of evolution: mean values of L* (39.36 ± 0.35. 45.77 ± 0.20 and 46.30 ± 0.24, for 20 min, 24 h and 7 d post mortem, respectively) and drip loss (0.75 ± 0.03 and 0.84 ± 0.02 for 24 h and 7 d post mortem, respectively) increased, those of a* (?1.77 ± 0.08, ?1.94 ± 0.07 and ?2.22 ± 0.05 for 20 min, 24 h and 7 d post mortem, respectively) and WBSF decreased (3.73 ± 0.06 and 2.63 ± 0.04 for 24 h and 7 d post mortem, respectively), those of pH decreased in the first 24 h and remained stable for the next 6 d (6.19 ± 0.02, 5.87 ± 0.01 and 5.88 ± 0.01), and those of b* increased in the first 24 h post-mortem and remained stable for the next 6 d (3.26 ± 0.31, 5.86 ± 0.16 and 5.47 ± 0.08).

Oxidative stress is an imbalance between radical-generating and radical-scavenging activity, resulting in oxidation products and tissue damage. This study was aimed to evaluate the status of oxidative stress indices in blood of camels naturally infested with S. scabiei. Forty-seven male camels (Camelus dromedaries) were divided according to the extent of the infested area with Sarcoptes scabiei into four groups, mild (MID, n = 12), moderate (MOD, n = 10), severely infested (SEV, n = 10) and healthy control group (n = 15). Blood was used for determination of red cell count (RBC), hemoglobin (Hb), packed cell volume (PCV), serum nitric oxide (NO, a free radical), ascorbate and albumin concentrations, and erythrocytic values of malondialdehyde (MDA, a marker of lipid peroxidation), protein carbonyls (PC, an indicator of protein oxidation), glutathione (GSH) superoxide dismutase (SOD) and catalase (CAT). Decreased levels (P < 0.05) of RBC, Hb, PCV, albumin and ascorbate were noticed in MOD and SEV compared to controls with the lowest values (P < 0.05) in SEV except for ascorbate, where MOD did not differ from SEV. Compared to controls, NO gradually increased (P < 0.05) in MID followed by MOD and SEV, whereas MDA and PC were higher (P < 0.05) in MOD and SEV. PC was higher (P < 0.05) in MOD than SEV. In addition, the antioxidants GSH, SOD and CAT were higher (P < 0.05) in MID and lower (P < 0.05) in MOD and SEV compared to controls. GSH was lower (P < 0.05) in SEV compared to MOD. Besides, Hb was negatively correlated with NO (r = −0.68, P < 0.001), MDA (r = −0.53, P < 0.001) and PC (r = −0.73, P < 0.001). In conclusion, dromedary sarcoptosis is accompanied by a state of oxidative stress process, which increased by increasing the area of infestation, and may contribute to the pathogenesis of the disease.  相似文献

The effect of extruding the cereal and/or the protein supplement of a compound feed on its nutritive value and on the performance of intensively reared male calves was studied. The compound feed was formulated with 0.65 of a cereal blend (60:40 maize:barley), 0.25 of a protein blend (1/3:1/3:1/3 raw soybeans:peas:lupins), and 0.08 of urea to contain 0.17 of crude protein. It was tested without extruding (NE) and with the cereal blend (CE), the protein blend (PE) or both (CPE) extruded. Another non-extruded compound feed with mainly soybean meal as the protein supplement (NE-SBM) also was studied. The five experimental compound feeds together with barley straw were offered ad libitum to 50 male Friesian calves (112 kg initial live weight) for 13 weeks, recording individual feed intake and live weight gain. Crude protein (CP) solubility and in vitro and in situ CP degradation of experimental compounds, and in vitro gas production kinetics, in vivo digestibility and urinary allantoin excretion of diets were determined. Cereal extrusion promoted a greater gas production at all incubation times (p < 0.001), associated to the increase in starch gelatinisation. PE and NE-SBM compounds showed lower in vitro (p < 0.01) and in situ (p < 0.001) CP degradability than the other feeds. Ingredient extrusion did not affect apparent DM, OM and CP digestibilities, but diet CPE showed a lower NDF digestibility (p < 0.05) than the others (0.293 vs. 0.420, 0.387, 0.390 and 0.407 with CPE vs. NE, CE, PE and NE-SBM, respectively). The apparent EE digestibility increased (p = 0.053) with the inclusion of extruded ingredients, (0.799, 0.749 and 0.794 vs. 0.719 and 0.702 with CE, PE and CPE vs. NE and NE-SBM, respectively). Daily allantoin excretion was lower (p < 0.01) in calves receiving the CPE diet than in those receiving the NE, PE and NE-SBM diets. Treatments did not affect the average daily gains (1.61 ± 0.022 kg d− 1), however calves on the CPE diet showed a lower (p < 0.01) concentrate and total conversion ratios than those fed NE, PE and NE-SBM (2.60 vs. 2.79, 2.85 and 2.98 kg concentrate DM kg− 1 daily gain and 2.93 vs. 3.09, 3.13 and 3.26 kg total DM kg− 1 daily gain, respectively). The CE diet resulted in numerically higher (p > 0.05) concentrate and total DM conversion ratios (2.72 and 3.02) than CPE and lower (p < 0.05) than NE-SBM. Improvement in feed conversion ratio after extrusion would be related to a better starch utilisation and not to changes in microbial or dietary protein flow to the duodenum, although a better utilisation of fat energy cannot be disregarded.  相似文献

The effects of two levels of concentrate and ruminally undegradable protein (RUP) on performance, intake, digestibility, carcass characteristics, meat quality traits, and commercial cuts yield were assessed. Twenty crossbred heifers (240 kg average body weight) were used. At the beginning of the trial, four animals were slaughtered as reference group and the 16 remaining animals were randomly assigned to four treatments, in a 2 × 2 factorial design: two levels of concentrate (40% and 80%, dry matter (DM) basis) and two levels of RUP (48.79% and 27.19% of CP). At the end of the trial, all the animals were slaughtered. There was no interaction (P > 0.05) between concentrate and RUP levels. Dry matter intake and nutrients digestibility was not affected (P > 0.05) by RUP level. Heifers fed the highest RUP level had greater (P < 0.05) average daily gain (ADG). Concentrate level did not affect (P > 0.05) DMI and ADG. Heifers fed diets with 80% concentrate had greater intake of TDN and EE, and lower intake of NDF (P < 0.05). The digestibilities of all nutrients, except the NDF, were greater (P < 0.05) for the 80% concentrate diets. The RUP levels did not affect (P > 0.05) the carcass characteristics and carcass gain composition. Heifers fed 80% concentrate diets had larger (P < 0.05) longissimus muscle area and lower gain of bones (P < 0.05). Concentrate level did not affect (P > 0.05) the composition of carcass gain. There was no effect (P > 0.05) of RUP and concentrate levels on meat quality traits and commercial cut yields.  相似文献
