Chrysanthemum stunt disease has been detected in Spain. An imprint-hybridization method using digoxigenin-labelled probes has been evaluated for quick detection of chrysanthemum stunt viroid (CSVd). The sensitivity of the method has been evaluated with several cultivars grown under different environmental conditions. The method has been successfully used in the eradication of CSVd.  相似文献

A viroid was detected for the first time in symptomless petunia plants (Petunia spp.) and identified as Tomato chlorotic dwarf viroid (TCDVd) based on an analysis of the complete genomic sequence. These petunia plants are a likely source of inoculum for tomato or potato plants because TCDVd induces severe symptoms on these plants. The genomic sequence of this petunia isolate from Japan shared 100 % identity with petunia isolates from the Netherlands and United Kingdom and a tomato isolate from Japan. Phylogenetic analysis showed that all petunia isolates and the tomato isolate from Japan formed a monophyletic clade.  相似文献

For the first time, Chrysanthemum stunt viroid (CSVd) was detected in commercial dahlia bulbs in Japan. CSVd was found in 77.2% of the tested plants (Dahlia spp.). In nucleotide sequence analysis, a CSVd variant was detected consisting of 354 nucleotides, which differed slightly from previously reported CSVd variants. The nucleotide sequence data reported is available in the DDBJ/EMBL/GenBank databases under accession number AB255879.  相似文献

In Belgium pospiviroids are routinely detected in various hosts. The most frequently found pospiviroids are: Citrus exocortis viroid (CEVd), Chrysanthemum stunt viroid (CSVd), Potato spindle tuber viroid (PSTVd), Tomato apical stunt viroid (TASVd) and Tomato chlorotic dwarf viroid (TCDVd). Apart from the high incidence of pospiviroids in latently-infected ornamentals, viroids have also been found in plants where they cause disease: PSTVd and TCDVd in tomatoes and CSVd in chrysanthemum. In order to gain more epidemiological data on these infections, this study has conducted phylogenetic analyses of Belgian isolates for each of these five pospiviroid species. PSTVd and CEVd-isolates show a clustering depending on host plant identity. This was not observed for TCDVd and TASVd. A very high degree of sequence similarity was noticeable for CSVd-isolates from various hosts. During the past decade, PSTVd and CSVd-infected mother plants have been systematically eradicated in Belgium after positive detection results, also when found in symptomless plants, leading to a decreased trend of these quarantine pests in the past few years. However, other non-quarantine pospiviroid species are still ubiquitously present in many ornamentals. Since these pospiviroids can be equally harmful to crops as the two quarantine pests PSTVd and CSVd, there is still a risk that transmission occurs from symptomless-infected ornamental plants to economically important crops in Belgium such as tomato, pepper and chrysanthemum.  相似文献

A new viroid was detected in hops cultivated in Akita Prefecture, Japan where it is prevalent in many hops fields. In a survey of hop samples collected during the 1986–2002 growing seasons, the new viroid was present in the major Japanese hop-cultivating areas as early as the 1980s. A single-stranded circular RNA of 368–372 nucleotides that assume a highly basepaired, stable, rod-like secondary structure, shares 93%–98% sequence homology with Apple fruit crinkle viroid (AFCVd) isolated from apple and 85%–87% with Australian grapevine viroid (AGVd) isolated from grapevine. Taking into account the present concept of viroid species, we conclude that the viroid is AFCVd. Circumstantial evidence suggests that AFCVd from apples and hops were endemic in Japan only where cultivation of the two host plants overlapped, thereby strongly supporting the possibility that AFCVd (or an ancestral viroid) was transmitted across the species barrier from apples to hops or hops to apples somewhere in the region. Phylogenetic analysis of AFCVd from hops, AFCVd from apples, and AGVd together with the other members of the genus Apscaviroid revealed that the Akita isolates of AFCVd from hops (AFCVd-hop) formed a cluster that is distinct from AFCVd-apple and AGVd. Accumulation of host-specific sequence variation following their isolation in different host species may be leading to the formation of two viroid species from a common ancestor.  相似文献

A comparison was made of methods for viroid detection. Molecular hybridization using cDNA is a very sensitive method that can handle large quantities of samples at the same time but it has the disadvantage that only small amounts of the sample can be applied to the nitrocellulose filter. The method therefore can only detect viroid in plants when its concentration is 10–20 ng g-1 of leaves, using 32P as a marker system. Bi-directional electrophoresis can detect viroid in plants when its concentration is 10 ng g-1 of leaves, because it uses larger samples. It does not need hazardous chemicals like 32P and formamide, and the reading of the results of the test is less liable to failures because it is based on two criteria (position and intensity of RNA band). The Dutch Plant Protection Service and the Dutch General Inspection Service for Ornamentals therefore use a modified bi-directional electrophoresis method to detect potato spindle tuber viroid and chrysanthemum stunt viroid, respectively.  相似文献

In studies to identify genotypes resistant to infection with citrus viroids, Eremocitrus glauca and Microcitrus australis were selected because their evolution in their habitat in Australia and New Guinea may have led to the selection of unusual traits. The movement and accumulation of Citrus exocortis viroid (CEVd), Hop stunt viroid, Citrus bent leaf viroid, Citrus dwarfing viroid, Citrus bark cracking viroid and Citrus viroid V (CVd‐V) in self‐rooted as well as in graft‐ propagated E. glauca and M. australis plants was assessed by northern hybridization, RT‐PCR and by topworking to the sensitive selection 861‐S1 of Etrog citron. In both plant species the inoculated viroids were undetectable unless these plants were grafted to a susceptible Citrus partner, the rough lemon rootstock and/or the topworked Etrog citron, which acted as viroid sources. The results obtained indicate that M. australis and in particular E. glauca are poor viroid hosts in which viroid replication/accumulation does not occur or is extremely inefficient. However, viroid downward and upward movement to grafted Citrus partners in which viroid replication and accumulation occurs efficiently was not impaired. Eremocitrus glauca and M. australis showed differences regarding their properties as viroid hosts, but for both species CEVd seemed to have the lowest affinity among the viroid species tested and CVd‐V the highest. Even though E. glauca and M. australis do not appear to be truly resistant to viroid infection, they are interesting genotypes for further characterization of the mechanisms involved in viroid infection.  相似文献

啤酒花矮化类病毒属是重要的植物类病毒属,目前尚无有效的筛查方法。通过对该属类病毒的核苷酸序列进行分析筛选,设计了8条用于该属类病毒筛查的属级特异性探针并制备了寡核苷酸芯片。应用啤酒花矮化类病毒标准样品对该芯片进行验证,结果表明所建立的属级芯片可以特异性检测啤酒花矮化类病毒,可检测到2ng/μL的总RNA。该芯片可用于啤酒花矮化类病毒属类病毒的筛查,为该属类病毒的检疫与防控提供技术支撑。  相似文献

A tobamovirus has been identified as being involved in a devastating disease of trailing petunia. Results from indicator plants and ELISA suggested that the tobamovirus was a strain of Tobacco mosaic virus (TMV). This was confirmed from the full sequence of the coat protein gene and a partial sequence of the replicase gene. Sequence analysis revealed that TMV isolated from diseased petunia had high identity (ca. 98–99) with TMV vulgare type sequences reported from Korea and Japan. Mechanical inoculation of 23 varieties, representing 21 species of pot and bedding plants with the petunia isolate of TMV confirmed that 11 were infected by the petunia isolate of TMV, although several species remained symptomless after three weeks. This highlights a clear risk to a number of commercially important pot and bedding plant species from TMV infected trailing petunias.  相似文献

The polyacrylamide gel electrophoresis (PAGE) test of Morris & Smith (1977) was evaluated for detection of potato spindle tuber viroid (PSTV) in breeding material. Number, density and mobility of nucleic acid bands in the electropherograms were influenced by genotype and growing temperature. So direct testing of genotypes was not reliable. After an intermediate viroid multiplication in tomato host plants at about 30oC and high irradiance, PSTV was reliably detectable with PAGE in inocula of potato samples of diverse origin. A 4-week incubation period proved to be suitable for inocula with low and high concentrations of a mild strain of PSTV (m-PSTV) as well as a severe strain of PSTV (s-PSTV). If incidence of PSTV is expected to be low, testing can be speeded up by bulking samples. With the combined tomato-intermediate/ PAGE assay, one m-PSTV or one s-PSTV infected leaf disk in 200 healthy ones was consistently detectable. Occasionally gels with a nucleic acid band of about the same relative mobility as the viroid band were found. Evidence that these bands were not caused by viroid is presented. A procedure to resolve such questionable test results is described. Infectivity of s-PSTV was higher than that of m-PSTV. Concentration of viroids in the inoculum influenced appearance of mild or severe symptoms and the rate of symptom production.  相似文献

A viroid etiology for citrus gummy bark (CGB) disease of sweet orange is supported by the similarity of symptom expression to cachexia disease of mandarins and tangelos caused by the hop stunt viroid (HSVd) related citrus viroid II (CVd-II), as well as the detection of CVd-II variants in CGB infected Washington navel and Dörtyol sweet orange, a Turkish cultivar. A survey was made of 67 clones of CVd-II related variants recovered from severe CGB symptomatic and non-symptomatic trees of the same cultivars growing in close proximity. Only CVd-IIa, a non-cachexia inducing variant, was found in non-symptomatic Washington navel trees and no CVd-II variants were recovered from the Dörtyol control. CGB infected sources contained a number of CVd-II related variants with the predominant species detected closely related to CVd-IIc, a known cachexia inducing viroid. Biological activity of representactive variants from CGB sources was determined by transmission to citron (Citrus medica) as well as by bioassay on the indexing host for cachexia, Parson's Special mandarin (Citrus reticulata).  相似文献

Using the sequential PAGE method for detection of small circular RNA molecules we isolated a viroid from greenhouse-grown tomato plants exhibiting severe stunting in Israel. The viroid was transmitted to tomato and to several other solanaceous plants by graft and mechanical inoculation, but only tomato plants showed symptoms of disease. Cloning and sequencing revealed that the viroid RNA is composed of 363 nucleotides, has 92% identity with the type strain (Ivory Coast strain) ofTomato apical stunt viroid (TASVd) and 99% identity with the Indonesian strain of this viroid. The experimental host range of TASVd-Is differs significantly from that of the type strain of TASVd. ABSTRACT Citrus exocortis viroid (CEVd), Citrus bent leaf viroid (CBLVd), a noncachexia variant of Hop stunt viroid (HSVd), Citrus viroid III (CVd-III), and Citrus viroid IV (CVd-IV) were co-inoculated as two-, three-, four-, and five-viroid mixtures to Clementine trees grafted on trifoliate orange to evaluate their effect on symptom expression, tree growth, and fruit yield. Most trees infected with CEVd-containing viroid mixtures developed exocortis scaling symptoms, as did CEVd alone, whereas most trees infected with HSVd- or CVd-IV-containing mixtures developed bark-cracking symptoms. Trees infected with mixtures containing both CEVd and CVd-IV revealed the existence of antagonism between these two viroids in terms of the expected bark-scaling and cracking symptoms. Synergistic interactions also were identified in trees infected with certain viroid combinations that, in spite of lacking CEVd, expressed exocortis-like scaling symptoms. Viroid interactions also affected the expected response of trees in terms of vegetative growth and fruit yield. Trees infected with viroid combinations containing CEVd or CVd-III were smaller and produced less fruit than trees infected with mixtures not containing these viroids. Viroid interactions on scion circumference and cumulative fruit yield, in terms of additivity of their effects, were statistically confirmed using a factorial analysis of variance model with two mean estimation approaches. In single-viroid infections, CEVd, CVd-III, and, to a lesser extent, CBLVd consistently and significantly reduced tree size and fruit yield. Conversely, HSVd and CVd-IV slightly increased fruit yield and reduced scion circumference. Rare and not consistent significant interactions were detected with the five-, four-, and three-viroid combinations. Antagonistic interactions between CEVd and CVd-III or CBLVd and CVd-III were revealed over the years with consistent significance. The antagonistic interaction between CEVd and CVd-IV was highly significant over the years when additional viroids were present; however, this antagonism appeared much later in the case of an exclusive interaction. HSVd and CVd-IV showed a consistent and significant synergistic interaction on yield only when both viroids were exclusively present. These results demonstrate antagonistic or synergistic relationships between citrus viroids depending on the viroid mixtures present in the host.  相似文献

Despite numerous studies, viroid elimination from infected plants remains a very challenging task. This study introduces for the first time a novel ‘suspension depletion’ approach for exemption of Solanum jasminoides plants from viroids. The proposed method implies initial establishment of suspension cultures of the infected plant cells. The suspended cells were then physically treated (mild thermotherapy, 33 °C), which presumably delayed the replication of the viroid. The viroid concentration in the treated biomass was monitored weekly using pospiviroid‐specific PCR. After 10–12 weeks of continuous treatment, a sufficient decrease in viroid concentration was observed such that the infection became undetectable by PCR. The treated single cells then gave rise to microcolonies on a solid culture medium and the obtained viroid‐negative clones were further promoted to regenerate into viroid‐free plants. Three years of accumulated experimental data suggests feasibility, broad applicability, and good efficacy of the proposed approach.  相似文献

A 303-nucleotide viroid was isolated from an apple tree (Malus × domestica, ‘Fuji’) cultivated in Japan. The viroid had 84.9% overall nucleotide sequence homology to Apple dimple fruit viroid (ADFVd), a member of Pospiviroidae, reported from Italy. This viroid differed from the Italian variant by 47 mutations (38 substitutions, six deletions and three insertions), and most of these mutations occurred on either side of the central conserved region. The leaves and branches of the infected trees did not have any disease symptoms, but the fruits were dimpled and yellow. The infected scions were top-grafted onto a healthy ‘Fuji’ apple tree, which tested positive for this viroid in a northern hybridization analysis, and yellow dimple fruits were produced in the second growing season. We propose that this viroid is a new variant of ADFVd and causes yellow dimple fruit formation in ‘Fuji’ apple trees.  相似文献
