不同方法测定反刍动物饲料NDF、ADF和木质素含量的比较   总被引:4,自引:0,他引:4  
采用传统抽滤、ANKOM滤袋和CAU滤袋技术测定16种反刍动物饲料的中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)和木质素(PL,高锰酸钾法)含量。结果表明:3种方法测定的大部分饲料的NDF、ADF和PL含量间没有差异(P>0.05),但在测定谷物性饲料NDF含量时,ANKOM滤袋技术测定值明显低于其他2种方法(P<0.05);在测定动物性蛋白质饲料时,CAU滤袋测定的NDF值明显高于其他2种方法(P<0.05)。3种方法测定的NDF、ADF和木质素含量均具有显著的线性相关关系(R2=0.990~0.996),其中2个滤袋技术测定值之间的相关程度最高(R2=0.992~0.996);2个滤袋技术测定值的变异系数(CV=2.20%~3.96%)均小于传统抽滤(CV=3.21%~4.91%)。  相似文献

Adhesive tape strip and dry swab sampling techniques were compared for the detection of Malassezia pachydermatis on the skin of dogs with chronic dermatitis. One hundred and four dogs were sampled by each of the techniques. Two methods, a culture method and a stain method, were used to assess the sampling techniques. By the adhesive tape strip sampling technique, M. pachydermatis was detected on 83 (80%) dogs using the culture method and on 45 (43%) dogs using the stain method. By the dry swab sampling technique, M. pachydermatis was detected on 55 (53%) dogs using the culture method and on 33 (32%) dogs using the stain method. The study showed that the adhesive tape strip sampling technique, using the culture method, detected Malassezia on the skin of significantly more dogs (P<0.001) than the same technique using the stain method and also significantly more than the dry swab sampling technique, using either the culture or stain methods. It was also shown that an adhesive tape sample could be used to transfer cells to a slide for staining and microscopy prior to being used for culturing Malassezia.  相似文献

Experiments were conducted to investigate the influence of the adaptation of rumen micro-organisms on the degradation of native potato starch (PS) in the rumen. Cows were fed with rations used for gas production (GP) analysis (dry cows, 1.6% starch) and for the nylon bag (NB) technique (lactating cows, 23% starch, mainly maize starch) and a ration containing 19% native PS (lactating cows). Fermentation characteristics of 13 samples were investigated with the GP technique using rumen fluid from cows fed each of the three rations. The same samples were investigated with the NB technique in the cows obtaining the NB ration and the PS ration. The results showed that the rate of GP was influenced by the source of the rumen fluid. The fermentation rate of PS was considerably enhanced by using rumen fluid adapted to the fermentation of native PS instead of using the other rumen fluids. Incubating in cows fed the PS ration, the rate of PS degradation determined with the NB technique, was higher compared with cows fed other rations. Using the PS ration the observed lag period for PS was shorter. The results show a clear influence of ration on the degradation characteristics of starch, determined with both the GP technique and the NB technique. However, these changes in behaviour did not explain observed differences in amounts of rumen escape PS measured in vivo in animal experiments and in situ, using the NB technique.  相似文献

Between days 24 and 32 after mating/insemination, 881 pigs (785 pregnant and 96 not-pregnant) were tested for pregnancy on a commercial farm with a linear-array real-time ultrasound scanner. 5-7 Days later, 785 of these animals (708 pregnant and 77 not-pregnant) were tested again with A-mode equipment by farm employees. Confirmation of pregnancy was based on recorded farrowings or abortions; confirmation of non-pregnancy was based on return to oestrus and rebreeding, recorded non-farrowing, or inspection of the uterus of culled animals at the slaughterhouse. From the number of correct positive (a), incorrect positive (b), correct negative (c) and incorrect negative (d) diagnoses, a sensitivity (a/a + d) of 100% versus 97.5%, a specificity (c/c + b) of 90.6 versus 55.8%, a positive predictive value (a:a + b) of 98.9% versus 95.3% and a negative predictive value (c:c + d) of 100% versus 70.5% were calculated for the real-time ultrasound technique versus A-mode technique. It was concluded that real-time ultrasound scanning provides a very accurate technique for pregnancy diagnosis in pigs, enabling immediate decision making on treatment or culling of animals diagnosed as non-pregnant.  相似文献

The accuracy of diagnoses based on examination of frozen sections was determined by comparing the results to those obtained by examination of tissues prepared using conventional methods (formalin fixation, paraffin-embedded tissue). One hundred ninety-four specimens were examined using the frozen section technique; 37 were examined to confirm a tentative diagnosis or to document lymph node metastasis and the remainder were examined to diagnose an unknown pathologic process. Of the 194 specimens examined, an accurate, specific diagnosis was obtained in 161 (83%); in 19 (10%), the pathologic process was correctly identified, but a specific diagnosis was not obtained; and in 2 (1%) the diagnosis was deferred. The remaining 12 (6%) were incorrectly diagnosed by the frozen section technique. When the number of specimens in which a specific diagnosis was obtained was combined with the number of specimens in which the pathologic process was correctly identified, the overall accuracy rate of the frozen section technique was 93%. There was no difference in the accuracy of the frozen section technique based on the reason for submission of the sample, source of tissue submitted, or the type of pathologic process (i.e., inflammatory or neoplastic). Of the 12 incorrect diagnoses, 4 (33%) were because of sampling errors and 8 (67%) were caused by interpretation errors. The proposed indications for the use of intraoperative frozen sections are: 1) to determine the nature of a pathologic process for which a preoperative diagnosis has not been established, 2) to determine the extent of spread of neoplastic tissue to lymph nodes and other organs, 3) to evaluate resection margins of a neoplastic process, and 4) to clarify situations where a discrepancy exists between the preoperative cytologic or histologic diagnosis and intraoperative gross pathology.  相似文献

It was concluded that real‐time ultrasound scanning provides a very accurate technique for pregnancy diagnosis in pigs, enabling immediate decision making on treatment or culling of animals diagnosed as non‐pregnant.  相似文献

A cranial technique for entry of the medial femorotibial (MFT) joint in the horse was validated. It was hypothesised that the frequency of correct placement into the MFT joint using the described cranial technique would be similar to using a standard medial approach. Twenty‐four limbs from 15 horses were injected with a latex mixture. Limbs with stifle joint effusion and/or known stifle pathology were grouped separately to those without. When effusion/pathology was not present the medial technique (93%) was almost twice as successful as the cranial technique (53%), including when extremes of size were removed from the population. With the presence of femoropatellar or medial femorotibial effusion, the cranial technique (20%) was 5 times less successful than using the medial technique (100%). When extremes of size were removed from the population of horses with effusion, the success of the cranial technique was marginally improved (33%). When an ‘average size’ of horse is used with or without joint effusion, the cranial technique is successful in entering the MFT joint in approximately 50% of cases, and may hence be of use in a fractious animal without sedation, a stallion, or in the presence of a wound/skin infection at the site of the medial needle entrance.  相似文献

Immunoperoxidase technique was applied for pathological study on naturally occurring pneumonic tissues of calves from which Pasteurella haemolytica was isolated. Multifocal necrosis occurred in the lungs of 25 out of 42 calves (59.5%) and P. haemolytica antigen was detected in 22 out of the 25 calves (88.0%). The calves were divided into 3 groups according to the number of P. haemolytica isolated. The positive rate of the bacterial antigen detected by the technique was 66.6% (28/42) on the average, reaching up to 85.7% (18/21) in the group from which the largest number of P. haemolytica was isolated.  相似文献

Bilateral thyroidectomy was performed in 106 cats with hyperthyroidism by one of three techniques: original intracapsular, modified intracapsular, or modified extracapsular. Hypocalcemia was detected in the first 3 days after surgery in 11 (22%) of 50 cats treated by the intracapsular technique, 10 (33%) of 30 cats treated by the modified intracapsular technique, and 6 (23%) of 26 cats treated by the modified extracapsular technique. Hypocalcemia was classified as mild or severe. No signs of hypoparathyroidism developed in any of the 13 cats with mild hypocalcemia. Of the 14 cats with severe hypocalcemia, 8 had clinical signs of hypoparathyroidism before and during treatment with calcium and vitamin D, 3 were treated and no clinical signs developed, 2 were not treated but no clinical signs developed, and 1 was lost to follow-up. No cat required permanent calcium or vitamin D supplementation after surgery. Severe hypocalcemia and clinical signs of hypoparathyroidism occurred in 3 (6%) of the 50 cats treated by the intracapsular technique, 4 (13.3%) of the 30 cats treated by the modified intracapsular technique, and 1 (3.8%) of the 26 cats treated by the modified extracapsular technique. Twelve cats had recurrence of hyperthyroidism at a median time of 23 months. The intracapsular technique was used in 11 of these cats, and the modified extracapsular technique was used in 1. No clinical signs of hypothyroidism were detected in any of the cats. The modified intracapsular and modified extracapsular techniques of bilateral thyroidectomy are effective procedures for the treatment of feline hyperthyroidism.  相似文献

An in vitro technique and an in situ technique were used to measure release of Ca, Mg, P and Zn over time from hand-clipped alfalfa (Medicago sativa L.). Maximal release of P and Zn was greater using the in situ technique. 'Florida 77' alfalfa (Medicago sativa L.), 'Florigraze' rhizoma peanut (Arachis glabrata Benth.), 'Mott' dwarf elephantgrass (Pennisetum purpureum Schum.), 'Tifton 78' bermudagrass (Cynodon dactylon (L) Pers.), 'Pensacola' bahiagrass (Paspalum notatum Flugge) and 'Floralta' limpograss (Hemarthria altissima (Poir.) Stapf and Hubbard) were used to examine the ruminal release of Ca, Mg, P, K, Cu and Zn by means of the in situ technique. Minerals were ranked from highest to lowest for extent of immediate and maximal release in the rumen. Averaged across forage species, ranking was K at 100% and 100%, Mg at 82% and 95%, Cu at 71% and 84%, P at 66% and 80%, Zn at 26% and 69% and Ca at 29% and 70%. A large proportion of Ca was associated with the plant cell wall, based on the duration of incubation time needed to maximize its release and the large fractinal amount of Ca slowly released. A large portion of the Zn was released slowly as well (mean, 43.8%), but release of Zn was maximal in less than 24 h. Most of the Mg, K, P and Cu probably reside in the cell contents. Forages with high NDF concentrations can have high release coefficients for their minerals.  相似文献

In this study five parasitological methods and a polymerase chain reaction (PCR) were compared for the diagnostic sensitivity for Trypanosoma evansi in experimentally infected water buffaloes over a period of 15 weeks. The combined estimates of sensitivity (CE(se)) of the PCR proved to be highest at 78.2%, closely followed by the mouse inoculation (MI), the micro-haematocrite centrifugation technique (MHCT) and the mini-anion-exchange centrifugation technique (MAECT) with CE(se) of, respectively, 74.0, 69.6 and 62.4%. The CE(se) of the buffy-coat technique (BCT) at 38.6% and the sodium dodecyl sulfate (SDS) clarification technique at 25.1% were considerably lower. PCR detected consistently all buffaloes infected from week 3 post-infection (PI) onwards. For MI this occurred after 5 weeks PI while for MHCT and MAECT these sustainable high levels were reached in the 7th week PI. BCT and SDS never detected all buffaloes infected. The influence of time and temperature on the viability of T. evansi in heparinized blood from water buffalo was also studied. In general we observed that the survival time tends to be longer when blood is kept at 4 degrees C. In samples kept in direct sunlight parasites became undetectable with the MHCT after 30min. After treatment of the water buffaloes with diminazene aceturate, the PCR signal disappeared within 24h.  相似文献

OBJECTIVE: To compare the monotonic tensile and fatigue strength of 16-gauge stainless steel wire (SSW) to ultra-high-molecular weight polyethylene (UHMWPE) cable using a transfixed cerclage technique in an in vitro midbody sesamoid osteotomy model. Endoscopic modifications to Martins transfixed cerclage technique were developed. A new suture technique of fixation was compared with the transfixed cerclage technique by measuring gap formation after cyclic testing. STUDY DESIGN: An in vitro biomechanical paired equine cadaver limb study. SAMPLE POPULATION: Twenty-one paired cadaveric adult equine forelimbs. METHODS: Uniaxial medial midbody sesamoid osteotomies were created in paired adult equine forelimbs. Monotonic tensile strength was measured on 10 forelimbs repaired by a transfixed cerclage technique using wire or cable. Fatigue testing to failure was performed on 4 forelimbs repaired using the transfixed cerclage technique by cycling the limbs between 500 N and 2,000 N. The limbs were initially repaired with wire, cycled until the wire broke, then repaired with cable and cycled again to failure. Fatigue testing for gap displacement was performed on 8 limbs repaired with either the transfixed cerclage technique or the suture technique. Limbs were cycled between 500 N and 2,000 N for 10,000 cycles. The limbs were repaired with wire initially, tested, and then repaired with cable and tested again. Twenty-two limbs were used for mechanical testing. The remaining limbs (20) were used to develop and practice the endoscopic transfixed cerclage (10 limbs) and suture (10 limbs) techniques. RESULTS: Ultimate tensile strength (UTS) of UHMWPE cable constructs was 34% greater than the UTS of SSW constructs. Fatigue strength was 2 to 20 times greater for UHMWPE cable constructs than SSW constructs. Separation of fragments was 153% less for limbs repaired by the suture technique compared with those repaired by the transfixed cerclage technique. CONCLUSIONS: UHMWPE cable shows promise for this clinical application because of its greater tensile and fatigue strength. The newly described suture technique significantly reduced gap formation compared with the transfixed cerclage technique. Osteotomy gap formation occurred early in cycling, suggesting that rigid support in the form of a cast may be needed during the early postoperative period for wiring techniques. CLINICAL RELEVANCE: Clinical testing of UHMWPE cable should eliminate problems of wire breakage seen with SSW. The endoscopic transfixed cerclage technique can be used by surgeons familiar with arthroscopic surgery. However, the suture technique needs to be tested in vivo to determine whether there is a clinical advantage compared with the transfixed cerclage technique.  相似文献

A technique that did not require use of a bronchoscope for bronchoalveolar lavage (BAL) in dogs was developed. An inexpensive, readily available 16-F Levin-type stomach tube was modified for the procedure. The technique was effective for collecting BAL fluid in 9 dogs that ranged from 9.3 to 26.2 kg (20.5 to 57.6 lb). Recovered fluid was consistent with fluid collected bronchoscopically. Mean recovery volume was 84/125 ml (67%), mean WBC counts were high (> 300 cells/microl), and > 70% of cells were macrophages. Complications from use of the technique were not detected on the basis of pulse oximetry, thoracic radiography, and clinical observation. This effective, simple, and safe technique for BAL can be readily performed in clinical settings that do not have bronchoscopic capabilities. It also provides a less costly alternative than bronchoscopic BAL.  相似文献

Equine cryptorchidism was examined by a review of the literature and a retrospective study of 350 horses over a 14-year period. The incidence of left vs right testis retention was nearly equal. On the left side, 75.2% of the retained testes were retained abdominally and 24.8% inguinally; on the right side, 41.8% of the retained testes were retained abdominally and 58.2% inguinally. Preoperative diagnosis by rectal palpation of the vaginal rings was considered a valuable technique, with 87.9% accuracy in 190 horses. Invasive and nonivasive surgical techniques for abdominal cryptorchidectomy and associated complications were compared. The results supported the technique of traction on gonadal structures outside the abdominal cavity (noninvasive) as superior to techniques requiring intraabdominal manipulation (invasive).  相似文献

A simple diagnostic technique was developed to improve the estimated prevalence of tapeworms on farms, by examining post-deworming fecal samples. All horses (n=162) on one farm were dewormed with a triple dose of pyrantel pamoatea and a fecal sample was collected before deworming and 24 hours after deworming. The results showed that the Cornell-Wisconsin centrifugal fecal floatation technique on pre-deworming fecal samples had a 32% sensitivity and 98% specificity when compared to the same technique on post-deworming fecal samples. One barn of horses (n=24) had fecals collected at 17, 24, 39 and 48 hours to determine the optimum time to collect fecals after deworming. The results suggested that the optimum time to collect fecals was 24 hours post-deworming.  相似文献

A technique for extracting Toxocara spp eggs from soil had a recovery efficiency ranging from 53% to 68%. Using the technique, 50 of 232 (22%) soil samples from city and university properties were found to be contaminated with eggs of Toxocara spp. Of 50 samples from highway rest areas, 8 (16%) were contaminated. From a public health standpoint, it was particularly noteworthy that 9 of 23 (39%) samples from sandboxes in student family quarters were contaminated with Toxocara spp eggs.  相似文献

本试验应用体外培养技术研究了不同氮源对反刍动物饲料可利用粗蛋白(utilizable crude protein,uCP)的影响,以及不同氮源在培养发酵过程中降解损失的情况。将4种氮源,即尿素、赖氨酸、蛋氨酸和酪蛋白水解物,分别添加到肉牛的典型基础日粮中(粗蛋白,CP14.1%),配制成4种日粮,并使各日粮的粗蛋白水平相近(约20%),作为试验日粮。以未添加氮源的基础日粮作为对照日粮。应用Zhao和Lebzien(2002)的体外培养法测定了4种试验日粮和对照日粮的uCP。结果表明,添加尿素、赖氨酸、蛋氨酸和酪蛋白水解物,均能够有效地提高基础日粮的uCP(P<0.05)。在这4种氮源中,赖氨酸提高uCP的效果最好。在培养发酵过程中,尿素、赖氨酸、蛋氨酸和酪蛋白水解物的氨态氮损失分别占粗蛋白质的72.9%、22.8%、55.2%和70.3%。  相似文献

The objective of this study was to describe a new technique for urinary catheterisation of female dogs using a novel catheterisation device (NCD) and to compare the time taken to place a catheter using this technique with traditional techniques. A secondary objective was to survey participants on which of the techniques they preferred. Female canine cadavers of varying sizes were utilised and veterinary students who had not previously placed a urinary catheter were enrolled. Each participant performed three catheterisation techniques, Visual with speculum (SPEC), Blind Palpation (BP) and catheterisation with NCD on three sizes of dog. Time required using each technique was compared using Kaplan–Meier plots and mixed models Cox Proportional Hazards regression. Median times to catheterisation were 300 s (IQR 261–417 s) with the SPEC method, 420 s (IQR 253–545 s) with the NCD method and 725 s (574–1032s) with the BP method. Both SPEC and NCD methods were significantly faster compared to the BP method, with Hazard Ratios of 3.66 (95% CI 1.94–6.91, P < 0.001) and 3.57 (95% CI 1.87–6.81, P < 0.001), respectively. Six of nine participants found the NCD the easiest technique, 5/9 of the participants found the palpation technique most difficult and 4/9 found the speculum technique most difficult. BP appears to be the technique of least preference and increased time requirement. The novel urinary catheterisation device may provide a simpler method of visualisation of the urethral papilla and may provide a more sterile way of placing the catheter, although further investigation is needed to confirm this.  相似文献

Mapping the distribution and abundance of invasive plants is a high priority, but establishing cost-effective and practical techniques at appropriate scales remains elusive. Mesquite is a highly invasive shrub that cannot currently be reliably distinguished from other plant species using remote sensing technologies, at least not at accuracies necessary for mapping mesquite at very low densities. This paper describes and tests an alternative method. A visual, aerial technique was used to map a large mesquite (Leguminoseae: Prosopis spp.) population in Australia; 216 654 ha was surveyed in 18.5-ha grid cells to include the entire population. The objective was to test the ability of this technique to detect and map mesquite at very low densities for surveillance and to assist in prioritizing management effort and, where mesquite was well established, to categorize mesquite into broad canopy cover classes for change detection and to identify habitat associations. The survey technique was very effective at detecting isolated mesquite plants (&spilt; 0.6% canopy cover across a grid cell), which is considerably better than existing remote sensing technologies. Detection of low-density mesquite was particularly important, as most occupied grid cells (55%) had isolated mesquite, and their management may offer the best return on investment. The technique was also competitive cost wise ($0.39 USD per hectare) and required relatively little expertise. Grid cells with moderate (20%–50%) to dense (&spigt; 50%) canopy covers were almost all restricted to a 32 500-ha area on the floodplain delta of the Fortescue River, where the original introductions occurred. Cover class estimates appeared to be well calibrated between observers within a survey; however, they were poorly calibrated between independently conducted surveys, suggesting that further methodological refinement is necessary if this technique is to be reliable for change detection.  相似文献

Seventy-nine diarrheic calf fecal samples were examined by electron microscopy, immunodiffusion and the fluorescent antibody technique for the presence of rotavirus (reovirus-like agent). Thirty-eight (48%) of the samples were positive by electron microscopy, 59% by immunodiffusion and 20% positive by fluorescent antibody technique analyses. Another 9% were suspect-positive by fluorescent antibody technique. Chymotrypsin treatment of the fecal samples increased the ease of observing the viral particles by electron microscopy and also intensified the immunodiffusion arcs obtained. Immunodiffusion analyses using specific antisera to the virus would appear to be a practical method of detecting rotavirus in diarrheic fecal samples.  相似文献
