Observations of feed intake, digestibility, production and selected blood values were made during the winter housing period on 10 lactating dairy cows, naturally infected with gastrointestinal roundworms, which were either left untreated or dosed on two occasions with thiabendazole in-feed. While treatment had no significant effect on appetite or production, there was a 9.8 per cent improvement in cellulose digestibility (P less than 0.05). An increase in circulating cholesterol in treated cows was associated with the improved utilisation of cellulose. It is considered unlikely that thiabendazole's primary anthelmintic activity was responsible for this effect. Treated cattle also exhibited a significant improvement in thyroid function during the course of the study compared with control animals. This might account for the increase in milk production reported in several trials following anthelmintic treatment of dairy cows.  相似文献

Five Finnish Ayrshire cows in mid or end-lactation were treated with 40 mg sulphadiazine/kg and 8 mg trimethoprim/kg using intravenous (i.v.), intramuscular (i.m.) and subcutaneous (s.c.) routes. Elimination of sulphadiazine was not affected by the route of administration (median t1/2 4.4-5.0 h) while elimination of trimethoprim was strongly limited by slow absorption from the injection site after s.c. and i.m. administration (median for apparent t1/2 21-25 h) compared to that after i.v. administration (median t1/2 1.2 h; p < 0.05). The median bioavailability of trimethoprim was also decreased, being 37% and 55% after s.c. and i.m. administration, respectively. When i.v. administration was used, trimethoprim concentration exceeded 0.1 mg/l in milk between 0.15-8 h while sulphadiazine concentrations above 2 mg/l were maintained from 0.5-2 h to 8 h. After s.c. and i.m. administration sulphadiazine in milk behaved similar to that after i.v. administration, while trimethoprim time-concentration curves were flat and trimethoprim concentrations were around 0.1 mg/l for an extended period of time (8-12 h). Median Cmax values in milk were only 0.07 mg/l and 0.10 mg/l for s.c. and i.m. administrations, respectively. After s.c. administration, 4 out of 5 cows showed signs of pain. After i.m. administration, 2 of the cows showed clear signs of pain and one had some local tenderness at the site of injection.  相似文献

Introduction   A number of systems based on metabolizable protein, such as that adopted in the UK (A gricultural and F ood R esearch C ouncil 1992) have been developed to improve the accuracy of protein rationing for ruminants. Quantification of microbial protein synthesis in the rumen is a fundamental requirement of all such systems. In the UK system, microbial protein supply is predicted from an estimate of fermentable metabolizable energy intake, using a correction for the effects of level of feeding on the energetic efficiency of microbial protein synthesis. Use of such an approach is however subject to considerable error due to large variations in the energetic efficiency of microbial protein synthesis (A gricultural R esearch C ouncil 1984). Consequently there is an urgent requirement for an on-farm diagnostic marker of microbial protein supply as a basis for adjusting diets to maximize efficiency of dietary nitrogen utilization by dairy cows (D ewhurst et al. 1996). Urinary purine derivative excretion has been proposed as a noninvasive index of microbial protein supply in ruminant animals (T opps and E lliot 1965). Use of this microbial marker is based on the assumption that purines entering the duodenum are essentially microbial in origin (M c A llan 1982), and that following metabolism, their derivatives are quantitatively recovered in the urine (C hen et al. 1990; V erbic et al. 1990). Purine metabolites excreted in ruminant urine are primarily derived from the metabolism of absorbed purines, but as a consequence of tissue adenosine triphosphate and nucleic acid turnover, a proportion of purine bases are not salvaged and re-utilized, but enter catabolic pathways, constituting an endogenous loss.  相似文献

As a prelude to mammary gland challenge experiments, this investigation was implemented to assess the hematologic changes in lactating dairy cattle induced by two dosage regimes of human recombinant colony stimulating factor (Hr-GCSF). This study documents the capability of the human recombinant colony stimulating factor to produce hematologic changes in both a time and dose dependent manner when administered to the adult lactating bovine. A screening dose of 1 microgram/kg of Hr-GCSF administered to three study subjects produced a three- to four-fold increase in peripheral blood mature neutrophil counts (P less than 0.043) by day 12 of the trial. The priming dose treatment group of four lactating cows (3 micrograms/kg of Hr-GCSF) exhibited a three- to five-fold increase in peripheral blood mature neutrophil counts (P less than 0.05) and two- to three-fold increases in white blood cell counts by day 5 of the trial. Hematologic examinations of the control group (n = 4; no Hr-GCSF administration) did not detect significant changes in their neutrophil counts over baseline values. The milk somatic cell counts did not statistically shift over baseline values in any of the control or Hr-GCSF treatment groups. When attempting to alter the course of infectious disease processes, potential applications of colony stimulating factors provide interesting speculations about new therapeutic modalities.  相似文献

Sixty Holstein cows were paired by parity and sire, and one of each pair was allocated at random to treatment or control; 17 cows were injected with 7.5 mg/100 kg dexamethasone trioxa undecanoate 14 days before the predicted date of calving, 13 cows received the same dose five days before term and 30 cows were left untreated. The treatment significantly advanced parturition and 29 of the 30 induced cows calved within 72 hours of the injection. Induction at day 14 before term was safe for calf and dam, the calves were 3.2 kg lighter than control calves and there was a high incidence of retained placenta. Treatment for this condition resulted in increased veterinary costs of 14.50 pounds per cow exclusive of dexamethasone treatment. Treatment at this stage was also associated with low pregnancy rates in the next breeding season. Calves born after induction at five days before term were not significantly lighter than calves from control cows, the problem of retained placenta was less marked and there were no subsequent effects on fertility. There were no significant effects of induction on milk yield or milk quality up to 200 days of lactation.  相似文献

Pharmacokinetics of sodium cephapirin in lactating dairy cows   总被引:1,自引:0,他引:1  
Sodium cephapirin was administered (10 mg/kg of body weight, IM) at 8-hour intervals in 4 consecutive doses to each of 6 lactating dairy cows. Blood, normal milk, mastitic milk, urine, and endometrial tissue samples were collected serially. Mean peak cephapirin concentrations in serum were 13.3 micrograms/ml 10 minutes after the 1st injection and were 15.8 micrograms/ml 20 minutes after the 4th injection (post[initial]injection hour [PIH] 24.33). The overall elimination rate constant value was 0.66/h and plasma clearance was 760 ml/h/kg. Mean peak cephapirin concentration in normal milk was 0.11 microgram/ml at PIH 2 and mean peak cephapirin concentration in mastitic milk was 0.18 microgram/ml at PIH 4. Cephapirin was not detected in the endometrium. The highest concentration of cephapirin in urine was 452 micrograms/ml, 2 hours after the 4th dose (PIH 26).  相似文献

Samples of serum and urine were obtained simultaneously from 56 healthy lactating cows to determine ranges of fractional excretion (FE) of calcium (Ca), phosphate (PO4), magnesium (Mg), sodium (Na), potassium (K), and chloride (Cl). Samples were obtained at 3 stages of lactation: period 1 = 1 to 7 days, 2 = 83 to 112 days, and 3 = 175 to 197 days. The FE of electrolytes were significantly different among periods 1, 2, and 3 for Ca (P less than 0.001), PO4 (P less than 0.025) and Mg (P less than 0.025), but were not significantly different for Na, K, and Cl. Least squares mean FE of Ca was lowest in period 1 and not significantly different for periods 2 and 3, whereas mean FE values for PO4 and Mg were highest in period 2 and not significantly different for periods 1 and 3. The mean FE values of Na, K, and Cl did not change with stage of lactation. Age and category of milk production (high, medium, and low) did not influence the FE values of the electrolytes.  相似文献

Plasma concentration stability of glucose, free fatty acids, ketone bodies, growth hormone, insulin were determined in lactating dairy cows. Concentrations of these metabolites and hormones were measured during a 36- to 48-hour period in 3 normal, mature dairy cows in the 2nd month of lactation. Samples were taken at 30-minute intervals; also, intensive sampling (every 10 minutes) was done at varying times in relation to feeding and milking. Of the 5 components measured, glucose concentration was the most stable, easiest to assay, and most reliable for use as a diagnostic aid in assessing metabolic carbohydrate disturbances in dairy cattle.  相似文献

Maternal behavior of beef cows at parturition   总被引:1,自引:0,他引:1  
Over an 11-yr period maternal behavior, calf birth weight and calf condition were recorded within 24 h for 2,684 parturitions. Maternal behavior was rated on a scale from 1 to 11, where 1 indicated extreme aggressiveness and 11 indicated no maternal attentiveness. Birth condition was rated on a scale from 1 to 9, with 9 being fattest and 1 thinnest. Respective breed means of Hereford, Angus, Charolais and Red POll were 6.2, 5.3, 6.0, and 5.7 for maternal behavior rating; 30.6, 27.3, 39.2 and 32.6 kg for birth weight, and 5.5, 4.7, 5.6 and 5.5 for birth condition. Heritability estimates for maternal rating, birth weight and birth condition were .06 +/- .01, .48 +/- .02 and .19 +/- .01, respectively. Genetic correlations between maternal rating and birth weight and condition were .04 +/- .05 and -.02 +/- .06, respectively. These results suggest that, in the population of cows studied, nongenetic influences were the primary cause of differences in aggressiveness of cows at parturition.  相似文献

Phenylbutazone was administered intravenously (i.v.) to a group of four lactating cows at a dosage of 6 mg/kg body weight. Whole plasma, protein-free plasma and milk were analysed for phenylbutazone residues. Pharmacokinetic parameters of total and free phenylbutazone in plasma were calculated using a non compartmental method. In regards to whole plasma data, the mean volume of distribution at steady state ( V ss), was 147 mL/kg body weight, with a mean (± SEM) terminal elimination half-life ( t 1/2) of 40 ± 6 h. The mean clearance ( Cl ) was 3 mL/h/kg body weight. The V ss as determined from the protein-free plasma fraction was 50 021 mL/kg body weight. This larger V ss of free phenylbutazone compared to total plasma phenylbutazone was attributed to a high degree of plasma protein binding, as well as the greater penetration of free phenylbutazone into tissues. The mean t 1/2 of free phenylbutazone was 39 ± 5 h. This similarity to the t 1/2 estimated from total plasma phenylbutazone data is attributed to an equilibrium between free and plasma phenylbutazone during the terminal elimination phase. Mean t 1/2 as determined from milk, applying a urinary excretion rate model, was 47 ± 4 h. Milk clearance of phenylbutazone was 0.009 mL/h/kg body weight, or about 0.34% of total body clearance. Furthermore, evidence suggests that phenylbutazone either binds to milk proteins, or is actively transported into milk, as its concentration in milk was greater than that predicted due to a simple partitioning from plasma into milk.  相似文献

选用6头2~3胎泌乳中期健康荷斯坦奶牛,采用3×3有重复拉丁方试验设计,通过消化试验就菊芋粕对泌乳奶牛的营养价值进行评定。试验设3个日粮处理,处理1为基础日粮,处理2为菊芋粕替代15%基础日粮(干物质基础),处理3为菊芋粕替代30%基础日粮(干物质基础)。结果表明,菊芋粕风干样品中干物质、粗蛋白、粗脂肪、粗纤维、中性洗涤纤维、酸性洗涤纤维、粗灰分、钙、磷和产奶净能分别为92.43%,12.98%,0.81%,12.67%,25.93%,21.44%,3.92%,0.27%,0.20%和 5.18 MJ/kg;菊芋粕对泌乳奶牛干物质消化率、粗蛋白消化率、中性洗涤纤维消化率、酸性洗涤纤维消化率分别为61.78%,73.21%,51.65%,61.14%,消化能和产奶净能分别为10.13和5.18 MJ/kg;3个处理组试牛产奶量和乳成分无显著差异(P>0.05),表明菊芋粕是一种营养价值略低但养分消化率较高的能量饲料。  相似文献

For a period of 150 days, an Escherichia co/; infection in a single quarter of the udder of 13 cows occurred on average every 9 days, in a herd of 120 cows kept at pasture under temperate environmental conditions. Most cases were clinically mild, affecting cows in lactations 1 to 10 and at all stages of lactation; none were evident at calving. Samples of secretion from infected quarters sometimes had few demonstrable organisms. Eleven different serotypes in 8 (0) groups were recovered; one isolate was multiply resistant to antibiotics and one infection persisted for 120 days. A precise explanation for this episodic exacerbation of the new infection rate was not established, but probable contributing factors were recognised.  相似文献

Abstract

Two herds of approximately 50 dairy cows were observed for oestrous behaviour for 6 weeks, 12 times a day for 30 minutes. It appeared that 3.08% of the pregnant cows showed oestrous behaviour during pregnancy (EBP) in such an intensity that they would have been considered in oestrus. With a less rigid oestrus detection threshold, 10.8% of the pregnant cows would have been considered to be in oestrus. Animals showed EBP during all months of pregnancy, but most of the behavior was observed in the middle of the gestation period.  相似文献

The bovine blood neutrophil phagocytosis and the blood and milk lymphocyte proliferative response upon stimulation with Phytohaemagglutinin, Concanavalin A and Pokeweed mitogens was studied from 3 weeks prior to calving until 3 weeks after calving. Neutrophil phagocytosis and the total and differential blood leukocyte counts were performed by flow cytometry. A gradual increase in the percentage of phagocytized bacteria and the average number of bacteria per phagocyte was observed before calving followed by a sharp fall on the first postpartum. This was followed by a steady increase in the above parameters reaching the highest levels at two weeks postpartum. There was a gradual increase in the number of neutrophils in blood as calving approached followed by a sharp decrease after calving. The number of lymphocytes in blood dropped before calving, being at the lowest level on the day before calving. The proliferative response of blood and milk lymphocytes upon stimulation with the three mitogens was low during the week preceding parturition with the lowest value on the day before calving. The response of blood lymphocytes returned to a higher level the second week after calving while that of milk lymphocytes remained at a low level during the first and the second postpartum weeks.  相似文献

Three-year-old Angus x Gelbvieh beef cows nutritionally managed to achieve a BCS of 4 +/- 0.07 (479.3 +/- 36.3 kg of initial BW) or 6 +/- 0.07 (579.6 +/- 53.1 kg of initial BW) at parturition were used in a 2-yr experiment (n = 36/yr) to determine the effects of BCS at parturition and postpartum lipid supplementation on cow adipose tissue lipogenesis. Beginning 3 d postpartum, cows within each BCS were randomly assigned to be fed hay and a low-fat control supplement or supplements with either cracked high-linoleate safflower seeds or cracked high-oleate safflower seeds until d 60 of lactation. Diets were formulated to be isonitrogenous and isocaloric, and safflower seed diets provided 5% DMI as fat. Adipose tissue biopsies were collected near the tail-head region of cows on d 30 and 60 of lactation. Dietary treatment did not affect (P > or = 0.43) adipose tissue lipogenesis. Body condition score at parturition did not affect acetate incorporation into lipid (P = 0.53) or activity of acetyl CoA carboxylase (P = 0.77) or fatty acid synthase (P = 0.18). Lipoprotein lipase activity and palmitate incorporation into triacyl-glycerol tended to be greater (P = 0.06), and palmitate esterification into total acylglycerols was greater (P = 0.01) in cows with a BCS of 4 at parturition. Mean activity of acetyl-CoA carboxylase (P < 0.001), lipoprotein lipase (P = 0.01), and rate of palmitate incorporation into monoacylglycerol (P = 0.02), diacylglycerol (P = 0.001), triacylglycerol (P = 0.003), and total acylglycerols (P = 0.002) were greater at d 30 than d 60, suggesting a greater proclivity for fatty acid biosynthesis and esterification by adipose tissue at d 30 of lactation. Although dietary lipid supplementation did not affect adipose tissue lipogenesis, results suggest that cows with a BCS of 4 at parturition have a greater propensity to deliver exogenously derived fatty acids to the adipocyte surface and incorporate preformed fatty acids into acylglycerols as stored adipocyte lipid. Additionally, cows in early lactation seemed to be able to synthesize and incorporate more fatty acids into stored lipid than cows during peak lactation.  相似文献
