Serum vitamin E and blood glutathione peroxidase values were determined in 40 horses with a histologically confirmed diagnosis of degenerative myeloencephalopathy and in 49 age-matched control horses with normal neurologic function. Significant differences were not detected in serum vitamin E or blood glutathione peroxidase values between horses affected with degenerative myeloencephalopathy and control horses. These findings fail to support a reported role of vitamin E deficiency as a cause of equine degenerative myeloencephalopathy.  相似文献

For the investigation of the cause of white muscle disease (WMD), tocopherol (Toc) and selenium (Se) levels and blood glutathione peroxidase (GSH-Px) activities were examined using lambs with WMD and their ewes. Serum Se levels of 4 lambs with WMD were low under 30 ppb, lambs showing very low levels below 15 ppb. The serum Se level was correlated with blood GSH-Px activity showing remarkably low activities in the lambs with WMD. Se contents in the organs of lambs with WMD were lower than those of control lambs, and particularly liver Se contents were deficient levels below 50 ppb. Serum Toc levels were normal, but alpha-Toc contents in organs showed very low levels, especially in the liver. The serum Toc and Se levels and blood GSH-Px activities of their ewes and other sheep kept in the same farm revealed similar results to those of lambs with WMD. Feedstuffs supplied on the farm showed the deficient level of the Se content below 50 ppb and a very low level of alpha-Toc. It was concluded that WMD of lambs in Hokkaido was nutritional muscular dystrophy resulted from deficiencies of Toc and Se to their ewes.  相似文献

Blood serum glutathione peroxidase activity and blood selenium concentration were measured in blood samples from pigs subjected to experimentally induced selenium deficiency and dietary selenium supplementation on graded levels. A highly significant correlation between blood selenium and serum GSH-Px activity in pigs, especially in selenium deficient pigs, was demonstrated. There was also a strong relationship between blood selenium concentration and serum GSH-Px activity in pigs receiving dietary selenium at graded levels. Serum GSH-Px activity exhibited an excellent close-response relationship to dietary selenium. Linear regression analysis showed that the increased serum GSH-Px activity was a function of the dietary selenium concentration. The fitness of serum in monitoring slight changes of the selenium status of pigs with help of the estimation of GSH-Px activity was discussed. The measurement of serum GSH-Px activity seems to provide a useful and rapid means for defining selenium requirements and for identifying selenium deficiency in growing pigs.  相似文献

For the purpose of clarifying the cause of white muscle disease (WMD) in calves, tocopherol and selenium levels and blood glutathione peroxidase (GSH-Px) activity were measured on 10 calves with WMD and nine of their dams. The main clinical symptoms of the 10 calves with WMD were motor disturbances including recumbency and stiffness. Serum enzyme activities (GOT, GPT, CPK, LDH) in calves with WMD increased markedly, and this increase was also observed in some of their dams. Serum tocopherol levels of calves with WMD were low, 70% of which showing deficient levels of less than 70 micrograms/100 ml. Serum selenium levels of all the calves were lower than 35 ppb, indicating a deficiency, and were accompanied by low blood GSH-Px activity. alpha-Tocopherol and selenium concentrations in organs were very low. Dams of calves with WMD showed low serum tocopherol levels, 22% of which indicating deficient levels below 150 micrograms/100 ml. Serum selenium levels in dams showed a marked decrease to under 20 ppb, and also low blood GSH-Px activity. Feedstuffs supplied in the farms to affected calves indicated very low alpha-tocopherol contents (below 3 mg/100g DM) and low selenium concentrations below 50 ppb in DM. It was concluded that WMD in calves was attributable to nutritional muscular dystrophy caused by deficiencies in tocopherol and selenium in feedstuffs supplied to their dams.  相似文献

Blood glutathione peroxidase (GSH-Px) levels in 709 normal dromedary camels (442 females and 267 males) were assessed in the Canary Islands. All animals were intensively reared, and three different nutritional systems were evaluated, depending on selenium content of the diet. Mean GSH-Px level in the total population was 288.5+/-157.2 IU x g(-1) Hb. Reference ranges were estimated and enzymatic activities below 51 IU x g(-1) Hb were considered inadequate. GSH-Px activities obtained in females (298.1+/-155.7 IU x g(-1) Hb) were significantly (P = 0.037) higher than in males (272.6+/-157.2 IU x g(-1) Hb). When age groups were compared, only males between 6 and 12 months old exhibited significantly lower mean GSH-Px (P = 0.006) than females. A high correlation (r = 0.88) between serum selenium concentration and blood GSH-Px activity was estimated, and the regression equation was y = 2.5101x + 42.423. Selenium content of the diet above 0.1 mg x kg(-1) DM seems to supply adequate selenium requirements for dromedaries under intensive husbandry.  相似文献

Tissues of 27 pigs with spontaneous dietetic microangiopathy (DM) and of 27 control pigs that died of causes unrelated to vitamin E and selenium (E-Se) deficiency were analyzed for alpha-tocopherol, Se and polyunsaturated fatty acid (PUFA) concentrations and for glutathione perioxidase (GSH-Px) activity. These variables (except for GSH-Px) also were measured in rations fed to control pigs and pigs with DM. Swine with DM had lower heart and liver alpha-tocopherol concentrations than did control pigs. Heart and kidney Se concentrations and heart and liver PUFA concentrations were similar in pigs of either group. Diets fed to both groups of pigs contained similar content of alpha-tocopherol, Se, oil, and PUFA; alpha-tocopherol and Se concentrations in the diets of both groups of pigs were high. In spite of apparently adequate amount of dietary alpha-tocopherol, results indicate that pigs with DM had lower tissue alpha-tocopherol concentration than did control pigs. Spontaneous DM is associated with altered alpha-tocopherol metabolism, but is unrelated to alterations in dietary Se and PUFA contents and tissue Se and PUFA concentrations and GSH-Px activity.  相似文献

The distribution of glutathione peroxidase (GSH-Px) activity and selenium concentrations among several components in the blood of dairy cows was examined. Approximately 98% of the GSH-Px activity in peripheral blood was associated with the erythrocytes when enzyme activity was expressed as units per milliliter of blood. The GSH-Px activity expressed per cell was approximately fourfold greater for peripheral leukocytes than for erythrocytes. The cellular component contained a greater proportion (approx 73%) of whole blood selenium than did the plasma. A positive linear relationship (r = 0.958) between blood GSH-Px activity and blood selenium concentrations was found in dairy cattle under practical field conditions.  相似文献

Vitamin E requirements of adult Standardbred horses were evaluated by tissue depletion and repletion. All the horses used in the study were given the same basal feed low in vitamin E during the eight months of the experiment. After an initial depletion period of two-and-a-half months the horses were divided into groups according to the amounts of DL alpha-tocopheryl acetate given (0 mg, control; 200, 600, 1800 and 5400 mg, respectively) as a daily oral supplement. The supplement study was followed by a second depletion period. Total vitamin E content and individual natural tocopherol isomers and tocotrienol isomers were measured both in the feed (hay and oats) and in tissue (serum, liver, skeletal muscle and adipose tissue) using high performance liquid chromatography with fluorescent detection. Tissue vitamin E response to different dietary vitamin E levels were studied. The serum total lipid content remained unchanged during the experiment; serum vitamin E levels were expressed per gram serum lipid. The total vitamin E levels in serum, liver, skeletal muscle and fat reflected the supplement levels. The highest vitamin E levels were seen in fat tissue, followed by the liver and by skeletal muscle. In spite of the wide occurrence of the different vitamin E isomers in the feed, alpha-tocopherol was almost the only isomer detected in the tissues. To ensure nutritional adequacy, 600 and 1800 mg of DL alpha-tocopheryl acetate was suggested as an optimal oral daily supplement of vitamin E to adult Standardbred horses given feed low in vitamin E; this corresponds to 1.5 to 4.4 mg/kg bodyweight.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

BACKGROUND: Chronic pulmonary diseases (recurrent airway obstruction [RAO]) have been reported to alter skeletal muscle cells in humans. The purpose of this study was to evaluate a potential relationship between pulmonary and muscle variables in horses with a clinical diagnosis of RAO. Muscle biopsies from healthy horses and from horses with RAO were investigated and the relationship between the severity of lung disease and the degree of muscular changes was determined. HYPOTHESIS: We hypothesized that chronic pulmonary disease can lead to changes of the skeletal muscle in horses. ANIMALS: Fifteen healthy horses (control) and 50 horses with RAO were examined. METHODS: In a prospective clinical trial, a complete lung examination was performed in all horses. In all horses, muscle enzyme activity at rest and after exercise and muscle biopsies from the M. gluteus medius were examined. RESULTS: None of the horses had clinical or histologic signs of primary or neurogenic myopathies. According to the clinical, endoscopic, and radiographic findings and with a scoring system, the horses with RAO were grouped according to the severity of pulmonary findings (15 horses mild, 24 horses moderate, 11 horses severe RAO). Pathologic changes of the skeletal muscle (fiber atrophy or fiber hypertrophy, myofibrillar degeneration, hyperplasia of mitochondria, and ragged-red-like fibers) were identified in most horses with RAO but in only a few individual control horses. In addition, a marked depletion of muscle glycogen storage was evident in the RAO horses but not in the control group. Other pathologic changes of skeletal muscle such as centralized nuclei and regenerating fibers were rare, but were more frequent in horses with lung diseases than in the control group. The degree of muscle cell changes was also graded with a scoring system and correlated with the severity of pulmonary disease (r= 0.55). CONCLUSION: Chronic pulmonary disease in horses is associated with structural changes in skeletal muscle. CLINICAL IMPORTANCE: Because chronic pulmonary disease may affect muscles, early and effective therapy may prevent these changes. This finding could be of clinical importance but requires further studies.  相似文献

Blood glutathione peroxidase activity and selenium levels were found to correlate well, indicating that glutathione peroxidase activity can be used to assess blood selenium levels in beef cattle. The glutathione peroxidase activity of blood is less stable than is the selenium concentration but when blood was stored at 4 degrees C, the glutathione peroxidase activity remained constant for seven days.  相似文献

Pigs from sows fed a diet deficient in Se and low in vitamin E were fed a Torula yeast diet supplemented with 100 IU dl-alpha-tocopheryl acetate/kg of diet. Dietary treatments were levels of supplemental Se of 0, .025, .050, .075 or .100 ppm. Some death loss occurred in pigs receiving no supplemental Se at approximately 5 wk of age. Autopsy revealed liver and heart lesions typical of vitamin E-Se deficiency. Selenium supplement had no significant effect on average daily gain, feed intake or gain to feed ratio for the 4-wk experiment. Selenium status of pigs was determined by serum Se concentration and serum glutathione peroxidase (GSH-Px) activity. Serum Se increased linearly (P less than .01) with increasing supplemental Se. Serum GSH-Px activity increased linearly (P less than .01) and quadratically (P less than .05) with increasing supplemental Se. With time, the level of serum Se and GSH-Px activity decreased in unsupplemental pigs, but increased in pigs fed diets supplemented with Se and resulted in significant interactions (P less than .01) between dietary Se level and time on experiment. The dynamics of vitamin E in the blood serum of sows of the Slovak Large White breed was studied from the 10th day of gravidity to the 20th day of lactation. A slight variation was observed in the average values of serum vitamin E from the 10th to the 50th day of gravidity (from 0.134 to 0.122 and 0.129 mg per 100 ml). A rise to the absolutely highest average values of serum concentration of vitamin E, 0.166 mg per 100 ml, was ascertained in the period from the 70th to the 90th day of gravidity. In the subsequent period, i.e. 110th day of gravidity, the value dropped to 0.120 mg per 100 ml, and on the 5th day of lactation the absolutely lowest value (0.095 mg per 100 ml) for the whole study was obtained. On the 20th day of lactation the serum concentration of vitamin E increased on an average to 0.103 mg per 100 ml. These data indicate the critical periods in the levels of vitamin E; these are from the 90th to the 100th day of gravidity and the first days of lactation when the concentration of vitamin E markedly decreases. If vitamin E is not supplemented to the sows during gravidity in sufficient amounts, preclinical and clinical vitamin E deficiency can be expected to occur.  相似文献

Ronnel [0,0-dimethyl 0-(2,4,5-trichlorophenyl) phosphorothioate] is an organophosphate pesticide with growth-promoting properties. Experiments were conducted to determine effects of ronnel on oxidation of and fatty acid synthesis from acetate and glucose as indices of metabolic activity in subcutaneous adipose tissue and skeletal muscle from 6-, 12- and 18-mo-old steers. Ronnel depressed metabolic activity in adipose tissue from 6- and 12-mo-old steers without concomitantly decreasing metabolic activity in skeletal muscle. Production of CO2 and fatty acids from acetate and glucose in tissues from 18-mo-old steers was influenced less by ronnel than in tissues from younger steers. Interactions of ronnel with thyroxine or growth hormone on acetate oxidation and conversion to fatty acids in adipose tissue also were investigated. Thyroxine increased acetate oxidation and decreased fatty acid synthesis. Ronnel interfered with the metabolic effects of thyroxine. Growth hormone, with or without ronnel, did not affect metabolic activity of adipose tissue. Ronnel seemingly alters the partitioning of acetate and glucose between major metabolic processes in adipose tissue and skeletal muscle.  相似文献

Twenty mature geldings, averaging 535 kg, were used to determine the influence of dietary selenium (Se) on the blood levels of Se and Se-dependent glutathione peroxidase (SeGSH-Px) activity in the horse. Horses were randomly assigned within breed to four treatments consisting of five horses each and fed a basal diet containing .06 ppm of naturally occurring Se. Diets were supplemented with .05, .10 and .20 ppm Se, as sodium selenite. Blood was drawn for 2 wk before, and for 12 wk following, the inclusion of supplement Se in the diets. Whole blood and plasma Se concentrations and plasma SeGSH-Px activities were determined from all blood samples. Selenium concentrations in plasma and whole blood increased linearly from wk 1 to wk 5 and 6, respectively, in Se-supplemented horses. After these times, no significant changes in Se concentration were observed in Se-supplemented or in unsupplemented horses throughout the remainder of the 12-wk trial. Plasma Se reached plateaus of .10 to .11, .12 to .14, and .13 to .14 micrograms/ml in horses supplemented with .05, .10 and .20 ppm Se, respectively. Whole blood Se reached plateaus of .16 to .18, .19 to .21, and .17 to .18 micrograms/ml in horses supplemented with .05, .10 and .20 ppm Se, respectively. Plasma SeGSH-Px activity was not significantly affected by dietary treatment. Therefore, this enzyme was not a good indicator of dietary Se in these mature horses.  相似文献

The levels of selenium (Se) and glutathione peroxidase (GPX) in the blood of sheep, cows and pigs under farm conditions were examined. Sheep appear to form two distinct groups, namely high Se and GPX and low Se and GPX. The high group gave ranges of 133-249 ng/ml and 77-179 iu/g Hb for blood Se and GPX respectively, while the low group showed levels of 21-67 ng/ml and 2-20 iu/g Hb. Overall sheep blood showed a high correlation between Se and GPX (r = 0-92, P less than 0-001). Cow bloods formed one group, all having low Se and GPX levels except for a single outlier. Omitting this animal, the overall ranges were 9-72 ng/ml and 6-36 iu/g Hb for Se and GPX respectively. Blood Se and GPX activity were significantly correlated (r = 0-59, P less than 0-001). Pigs formed a single group also, with the difference that while their blood Se was high, the corresponding blood GPX activities were relatively low. Overall ranges were 93-193 ng/ml and 17-69 iu/g Hb for Se and GPX respectively. Correlation between blood Se level and GPX activity in this species was not significant (r = 0-27, P more than 0-1).  相似文献
