The aim of this study was to evaluate the effect of cloprostenol administration on the blood flow of pre-ovulatory follicle (PF) and corpus luteum (CL), progesterone secretion and pregnancy outcome in buffaloes subjected to AI. The trial was performed on 75 Italian buffaloes at 182 ± 8 days in milk. Synchronized animals were randomly divided into two groups on the day of oestrus: Group T (n = 37) received a 0.524 mg intramuscular injection of cloprostenol and Group C (n = 38) received saline. Ultrasound examinations of the ovaries were performed 5 h after AI on the PF and 10 and 20 days after AI on the CL. Resistive (RI) and pulsatily index (PI) were calculated by colour-Doppler mode in each examination. Blood samples were collected on days 10, 20 and 25 after AI for progesterone assay and 25 days after AI, ultrasonography was performed to assess pregnancy, which was confirmed on day 45. Subjects pregnant on day 25 but not on day 45 were considered to have undergone late embryonic mortality (LEM). Statistical analysis was performed by anova. No differences were found in PF dimensions, CL size and blood flow on day 10 and 20 after AI between treated and control groups. Pre-ovulatory follicle area was higher in buffaloes that resulted pregnant on day 25 after AI compared to those that were non-pregnant (2.13 vs 1.66 cm in pregnant and non-pregnant buffaloes, respectively), while non-pregnant buffaloes showed higher values of RI (0.49 vs 0.30; p < 0.05) and PI (1.0 vs 0.37; p = 0.07) compared to pregnant subjects. Treatment by cloprostenol did not influence pregnancy rate both on day 25 (31/75; 41.3%) and 45 (27/75; 36.0%), progesterone levels and incidence of LEM (4/31; 12.9%). In conclusion, cloprostenol administration at the time of AI does not seem to affect PF and CL blood flow.  相似文献

一、前言从外周血中PGFM水平的测定结果可知,分娩前黄体消退并伴有PGF_2α的暂时性升高,黄体的最终消退是PGF_2α溶体黄作用引起的;产后1~4天外周血中PGFM增加,15天左右恢复到基础水平。试验证实,产后早期PGFM主要来源于子宫。因为切除子宫后,不出现产后正常的PGFM水平的上升。子宫动、静脉血中PGFM水平的差异,也表明子宫是PGF_2α的合成、代谢部位。子宫复旧是产后卵巢恢复周期性活动的先决条件,产后子宫复旧时间过长与产后早期内源性PGF_2α的分泌不足有关。PGF_2α能收缩子宫平滑肌,增加子宫紧张性。产后PGFM高  相似文献

A study was undertaken to determine the oestrus response and fertility rates of zebu cows treated with PRID alone or in combination with PGF2alpha. A total of 184 non-suckled cycling Bunaji cows were allotted randomly to four treatment groups of 46 animals per group as follows: group 1 (PRID-12), PRID was inserted for 12 days; group 2 (PRID+7+PGF2alpha-6, PRID was inserted for 7 days and PGF2alpha was administered intramuscularly 1 day prior to PRID withdrawal; group 3 (PRID-7+PGF2alpha-7, PRID was inserted for 7 days and PGF2alpha was administered intramuscularly on the day of PRID withdrawal; group 4 (2 x PGF2alpha-13, two intramuscular injections of PGF2alpha 13 days apart. At the end of each treatment period, the cows were observed for 7 days for behavioural oestrus and were inseminated 12 h following detection of oestrus. Pregnancy was diagnosed by rectal palpation 30-40 days post-insemination. The respective oestrus response rates were 78.3%, 76.1%, 87.0% and 89.1% for groups 1-4. While the corresponding pregnancy rates were 39.1%, 41.3%, 52.2% and 52.2%, the conception rates were 50.0%, 54.3%, 60.3% and 58.6% for groups 1-4. Although individual variations in progesterone levels were observed, the progesterone profiles were generally typical and normal. The results of the study have confirmed the effectiveness of the four regimes in synchronizing and controlling oestrus and ovulation in Bunaji cows. However, groups 3 and 4 showed some superiority over the other treatments. The results of this study provide feasible options from which clinicians involved in intensive breeding programmes and herd health fertility programmes can choose.  相似文献

旨在研究不同黄体期注射前列腺激素(PGF2α)对湖羊育成母羊黄体组织形态、PGF2α受体及程序性坏死通路基因表达的影响。本研究选择健康、体况良好、体重相近经同期发情处理的母羊48只,随机均分为6组,分别为黄体前期试验组和对照组、中期试验组和对照组和末期试验组和对照组母羊,试验组在发情周期第6天(黄体前期)、11天(黄体中期)、16天(黄体末期)注射1 mL PGF2α(0.1 mg),对照组在相同时间注射1 mL生理盐水,每次注射后3 h采集黄体组织,用于基因表达检测及HE组织染色检测黄体组织形态变化。结果表明,注射PGF2α后试验羊黄体组织形态发生了变化,出现炎症细胞浸润与细胞凋亡,其中黄体中期退化程度最高。未注射PGF2α的母羊,黄体前期、中期FPA mRNA表达水平显著高于黄体末期(P<0.05);黄体前期FPB表达水平显著高于黄体末期(P<0.05),与黄体中期无显著差异(P>0.05);黄体中期TNFR1表达水平显著高于黄体前期和末期(P<0.05);黄体中期RIPK3表达水平显著高于黄体末期(P<0.05),黄体中、末期与黄体前期差异均不显著(...  相似文献

The changing concentrations and temporal relationships among a PGF2α metabolite (PGFM), progesterone (P4), LH, and estradiol-17β (E2) before, during, and after luteolysis were studied in 10 mares. Blood samples were collected every hour for ≥4 d beginning on day 12 after ovulation. The luteolytic period extended from a decrease in P4 at a common transitional hour (Hour 0) at the end of preluteolysis and beginning of luteolysis to a defined ending when P4 reached 1 ng/mL. The length of luteolysis was 22.9 ± 0.9 h, contrasting with 2 d in published P4 profiles from sampling every 6 to 24 h. In mares with complete data for Hours −40 to −2 (n = 6), PGFM concentrations remained below assay sensitivity (n = 2) or two or three small pulses (peak, 29 ± 4 pg/mL) occurred. During luteolysis, the pulses became more prominent (peak, 193 ± 36 pg/mL). Rhythmicity of PGFM pulses was not detected by a pulsatility program during preluteolysis but was detected in seven of nine mares during luteolysis and postluteolysis combined. The nadir-to-nadir interval for LH pulses and the peak-to-peak interval between adjacent pulses were longer (P < 0.05) during preluteolysis than during luteolysis (nadir to nadir, 5.2 ± 0.3 h vs 3.6 ± 0.4 h; peak to peak, 9.4 ± 1.0 h vs 4.7 ± 0.5 h). Unlike reported findings in cattle, concentrations of P4 decreased linearly within the hours of each PGFM pulse during luteolysis, and a positive effect of an LH pulse on P4 and E2 concentration was not detected. The reported balancing of P4 concentrations between a negative effect of PGF2α and a positive effect of LH in heifers was not detected in mares.  相似文献

Prostaglandin F2α (PGF) is considered to be the main luteolysin in cattle. We have previously demonstrated that cortisol (Cr) suppresses PGF production in non-pregnant bovine endometrium. This study was carried out to test whether exogenous PGF increases ovarian and/or uterine PGF production and to determine the temporal relationship between PGF and Cr in ovarian and uterine circulations during PGF-induced luteolysis in cows. Catheters were inserted into the ovarian vein (OV), uterine vein (UV) and jugular vein (JV) of 10 cows on Day 9 of the oestrous cycle (Ovulation = Day 0) for frequent blood collection. On Day 10, the cows were divided randomly into two groups and treated with a luteolytic dose of a PGF analogue (cloprostenol) or saline solution. Blood samples were collected at -0.25, 0, 0.25, 0.5, 1 and 2 h and then at 2-h intervals until 12 h after treatment (0 h). The basal concentrations of PGF and Cr in OV and UV plasma were not significantly different. Injection of a PGF analogue induced more than twofold increases in the levels of PGF between 0.25 and 1 h in UV plasma, but not in OV plasma. PGF increased (p < 0.05) the concentrations of Cr in OV, UV and JV plasma between 0.5 and 1 h. The Cr levels in OV, UV and JV plasma were similar. The PGF levels in UV plasma decreased after Cr reached its highest levels. The overall results suggest that the uterus rather than the ovary increases PGF production in response to PGF injection. Based on the temporal changes of PGF and Cr in the ovarian and uterine circulations, Cr may act to reduce uterine PGF production in non-pregnant cows in vivo.  相似文献

为提高母水牛情期受胎率,促进安化县水牛品种改良工作的开展,文章就氯前列烯醇不同处理方式对本地水牛同期发情效果进行了比较研究试验。结果表明:灌注一次PG、注射一次PG、灌注两次PG和注射两次PG的同期发情发情率分别为55.56%、63.64%、60.00%和76.92%,总体受胎率分别为22.22%、27.27%、33.33%和38.46%;一次PG处理组同期发情率为60%,二次PG同期发情率为69.56%,总体受胎率分别为25.00%和34.78%;灌注组的同期发情率同期发情发情率为57.89%,显著高于肌注PG组的57.89%,总体受胎率分别为33.33%和26.32%。利用PG进行滨湖水牛同期发情处理时,子宫内两次灌注法能得到较好的发情率和受胎率。  相似文献
