Cured sweetpotato roots were stored at different temperatures (4.5, 15.6, and 24 degrees C) for 7 weeks and assayed for invertase activities and reducing sugar levels during two separate years. Invertase activities and reducing sugar concentration significantly increased in the roots kept at low temperature. Of the three types of invertases assayed, acid invertase specific activity was the highest. Acid invertase was the most influential in determining reducing sugar levels in stored sweetpotato. Cultivar differences were found in invertase specific activities and reducing sugar concentration. Reducing sugar content was highly correlated to acid and total invertase activity, regardless of cultivar.  相似文献

若尔盖湿地土壤酶活性和理化性质与微生物关系的研究   总被引:10,自引:2,他引:10  
本文旨在研究若尔盖湿地土壤退化过程中四种典型土壤(泥炭土、沼泽土、草甸土、风沙土)中脲酶、过氧化氢酶、转化酶、多酚氧化酶活性及其与理化性质、微生物数量的相关性分析。结果表明:随着土壤深度增加,脲酶、转化酶活性逐渐降低,过氧化氢酶、多酚氧化酶活性逐渐升高;随着土壤退化程度的加剧,脲酶和转化酶活性总体呈下降趋势;泥炭土、沼泽土、草甸土中过氧化氢酶活性显著高于风沙土;多酚氧化酶活性随退化程度加剧呈上升趋势,但在沼泽土中活性最低。泥炭土、沼泽土、草甸土中转化酶及风沙土中过氧化氢酶与土壤理化性质、微生物数量的相关性较显著。因此土壤酶活性(脲酶、转化酶)、理化性质(含水量、有机质、全氮、全磷、全钾)、微生物数量能较好地指示当地土壤退化状况。  相似文献

以塿土为研究对象,探讨不同有机无机肥料配比对土壤酶活性和作物产量的影响。结果表明,不同有机无机肥料配比条件下,蔗糖酶、脲酶和碱性磷酸酶活性的动态变化特征没有明显改变;但对蔗糖酶、脲酶和碱性磷酸酶活性的影响显著,显著影响小麦干物质量和子粒产量。2/3CN+3/5OM处理蔗糖酶活性最高,达29.51 mg/g,分别比3/3CN、3/3CN+1/5OM、1/3CN+5/5OM和5/5OM处理提高33%、32%、89%和146%;2/3CN+3/5OM处理脲酶活性最高为0.75 mg/g,与其余处理差异达显著水平。2/3CN+3/5OM处理碱性磷酸酶活性则与其它处理差异不显著;但小麦子粒产量为5.7 t/hm2,比其它处理分别提高22%、18%、27%和45%。  相似文献

本研究基于鹰潭农田生态系统国家野外科学观测研究站24年的长期定位试验,揭示对照(不施肥,CK)、有机肥(C)、化学氮磷钾肥(NPK)、化学氮磷钾肥+有机肥(NPKC)等不同施肥处理对红壤水稻土酶活性及土壤养分的影响。于晚稻收获后采集各小区耕层土壤,测定红壤水稻土中转化酶、脲酶活性(测定时并设加0.5 ml甲苯与不加甲苯处理)及转化酶动力学特征,同时测定土壤养分含量及微生物生物量碳,分析酶活性与养分含量及微生物生物量碳间的关系,明确土壤中累积酶活性及土壤养分对长期不同施肥处理的响应。结果发现,与对照相比,施肥处理下土壤转化酶活性显著提高了31.3%~131.7%,微生物生物量碳显著提高了84.9%~125.1%;在没有甲苯抑制微生物活性下,施肥处理的转化酶底物蔗糖转化速率增加量提高了89.5%~153.7%,脲酶底物尿素转化增加量提高了59.2%~98.9%,表明微生物显著影响两种累积酶表观酶活性;转化酶活性、脲酶活性与微生物生物量碳呈显著正相关。与对照处理相比,施肥处理显著增加了土壤有机碳(30.1%~36.3%)、全磷(28.6%~102.9%)、速效磷(62.2%~445.0%)、碱解氮(35.9%~56.4%)含量;统计分析显示,转化酶活性、脲酶活性均与碱解氮、有机碳含量显著正相关。与对照相比,各施肥处理土壤的转化酶米氏常数(Km)差异并不显著,而转化酶表观活性(Vmax)及转化系数(Vmax/Km)均显著增加。长期施肥处理增加了土壤养分含量和微生物生物量碳,提高了土壤中累积酶的活性。  相似文献

Activities of invertase and amylase and respiratory activities of samples of 11 soils from New Hebrides were determined. The soils mostly were under forest and were acid with medium to low C/N ratios. Invertase activities were rather low but amylase activities were similar to those found in New Zealand soils. The ratios of invertase to amylase activities were mostly low.O2 uptakes mostly responded markedly to glucose. Most values of respiratory quotients were about 1.0. Most, but not all, dehydrogenase activities were strongly related to O2 uptakes. On an organic C basis, these respiratory activities declined with the depths to which the soils were sampled.Biochemical activities were mostly similar in forest soils derived from basalt and from andesite. Invertase activities were lower in soils under forest than under grassland covers.All biochemical activities were correlated significantly with contents of soil moisture and organic C, less with numbers of aerobic bacteria, and negatively with soil pH. On an organic C basis, none of the biochemical activities was significantly correlated with either soil moisture content or pH.  相似文献

The activities of three extracellular hydrolytic enzymes, soil invertase, urease, and alkaline phosphatase (AlP), were measured across seasons and with the experimental addition of nitrogen (N) in the soil of the Gurbantunggut Desert, Northwest China. Seasonal fluctuations in hydrolytic enzyme activities were not correlated with seasonal variations in soil temperature, water content, pH, conductance, and organic carbon. Invertase and AlP activities increased with low rates of N addition, peaked at a N addition rate of 3.0 g N m?2 y?1, and then decreased at higher N addition rates. Urease activity decreased with increasing N addition. Higher organic matter content in the upper depths of soil resulted in higher hydrolytic enzyme activity at depths of 0–5 cm in soil samples and hydrolytic enzyme activity at that depth was more sensitive to N addition and seasonal environmental factors than that at depths of 5–10 cm in soil samples.  相似文献

The mineral content and color characteristics of 77 honey samples were analyzed. Eighteen minerals were quantified for each honey. Multiple linear regression (MLR) was used to establish equations relating the colorimetric CIELAB coordinates to the mineral data. The results obtained shown that lightness (L) was significantly correlated with S, Ca, Fe, As, Pb, and Cd for the dark honey types (avocado, heather, chestnut, and honeydew). For the light and brown honey types (citrus, rosemary, lavender, eucalyptus, and thyme), C(ab) and b showed the lower correlation with the mineral content of the honeys; their regression functions involve a few independent variables (Mg and Al for b and only Al for C(ab)). Furthermore, by means of application of linear discriminant analysis to the mineral content, it was possible to obtain a model that classifies the honeys by their lightness. The prediction ability of the built model, determined with the test set method, was 85%.  相似文献

This paper reports the application of near-infrared (NIR) reflectance spectroscopy to determine the concentration in honey of perseitol, a sugar that is specific to avocado honey. Reference values for perseitol were obtained by high-performance liquid chromatography analysis in 109 honey samples. Although the average concentration of perseitol in honey samples was only 0.48%, accurate prediction equations were successfully developed. The regression model of modified partial least squares was superior to that of principal component regressions. Calibrations based on the first or second derivative of Log(1/R) were equally good (R(2) > 0.95). Using half of the samples for calibration and the second half for validation, the correlation between actual and predicted values of the second half was satisfactory (R(2) = 0.87), the slope did not differ from 1, bias was low (0.005%), and the standard error of prediction was relatively low (0.13%). It was concluded that NIRS analysis may be used to detect to what extent honeybees have harvested avocado nectar but not to authenticate avocado honey as unifloral.  相似文献

土壤酶驱动土壤生态系统养分的循环和控制生态系统的功能。本研究以生长年限为10、20年和30年的柑橘林0~20 cm和20~40 cm土层土壤为研究对象,主要探讨了土壤脲酶、转化酶和过氧化氢酶活性随着柑橘林龄的延长和土壤深度的增加的变化规律。结果表明,随着柑橘林龄的延长,0~20 cm土层土壤过氧化氢酶活性10年和20年样地之间无显著性差异,但都显著大于30年的样地;而0~20 cm土层土壤转化酶和脲酶活性逐渐提高,20年时达到最大值,其后又降低。随着土壤深度的增加,过氧化氢酶、转化酶和脲酶活性在3个林龄的柑橘林中都呈现显著的下降趋势。相关性分析的结果表明,土壤转化酶活性、脲酶活性都与土壤有机碳和微生物量碳氮之间都具有显著的正相关性,而过氧化酶氢活性与土壤理化特性及其微生物量之间都无显著的相关性。主成分分析结果进一步显示,土壤转化酶活性、脲酶活性、有机碳和微生物生物量碳氮均在第一主成分中具有较大的载荷,对第一主成分的贡献最大。以上结果表明脲酶和转化酶活性能够做为柑橘土壤质量变化的敏感指标。  相似文献

Research continues to differentiate the impact of water activity (a(W)) and the glass transition temperature (T(g)) on chemical reactions. Invertase with and without sucrose was incorporated into low and high molecular weight poly(vinylpyrrolidone) model systems (PVP-LMW and PVP-K30, respectively). Invertase activity and sucrose hydrolysis were monitored during storage at a(W) = 0.32-0.75 and 30 degrees C. Pseudo-first-order rate constants for activity loss in PVP-K30 were not different, regardless of the system being glassy or rubbery. In PVP-LMW, invertase stability decreased with increasing a(W). An a(W) > 0.62 was required for sucrose hydrolysis to occur in PVP-LMW. PVP molecular weight appeared to affect invertase stability and reactivity. No dramatic change around T(g) was found in either invertase stability or sucrose hydrolysis, suggesting that T(g)-dictated mobility has a minimal effect on these reactions in amorphous solids.  相似文献

调查了宁夏御马酿酒葡萄基地不同栽培年限下土壤的蔗糖酶、磷酸酶、脲酶、过氧化氢酶的活性,并与土壤的基本理化性质进行了回归分析。结果表明,供试土壤为强碱性反应,耕作施肥对土壤pH和全盐含量影响不显著;土壤有机质、全氮、铵态氮、速效磷、阳离子代换量大都处于最低的六级水平,但肥力随耕种年限的延长显著增加;随栽培年限的延长,表层土壤磷酸酶活性、蔗糖酶和脲酶活性显著增加;蔗糖酶、脲酶活性随剖面深度的加深而显著减小;表土过氧化氢酶活性随栽培年限的延长而显著下降,但不同土地利用下表层以下各层次土壤过氧化氢酶活性总体上高于表层,且差异显著。磷酸酶活性与土壤有机质、全氮、全磷、速效磷、缓效钾、速效钾之间存在着极显著相关关系;磷酸酶活性、蔗糖酶和脲酶活性与土壤有机质、全氮、全磷、速效磷、缓效钾、速效钾之间存在着极显著相关关系;四种酶之间,磷酸酶与脲酶之间存在极显著相关关系,脲酶与蔗糖酶之间存在极显著相关关系。由此表明,磷酸酶、脲酶、蔗糖酶活性的大小可以代表宁夏贺兰山东麓酿酒葡萄栽培区土壤肥力的高低。  相似文献

Invertase (EC 3.2.1.26) catalyzes the hydrolysis of sucrose into D-glucose and D-fructose. Insoluble acid invertase (INAC-INV) was purified from pea (Pisum sativum L.) by sequential procedures entailing ammonium sulfate precipitation, ion exchange chromatography, absorption chromatography, reactive green-19 affinity chromatography, and gel filtration. The purified INAC-INV had a pH optimum of 4.0 and a temperature optimum of 45 °C. The effects of various concentrations of Tris-HCl, HgCl(2), and CuSO(4) on the activities of the purified invertase were examined. INAC-INV was not affected by Tris-HCl and HgCl(2). INAC-INV activity was inhibited by 6.2 mM CuSO(4) up to 50%. The enzymes display typical hyperbolic saturation kinetics for sucrose hydrolysis. The K(m) and V(max) values of INAC-INV were determined to be 4.41 mM and 8.41 U (mg protein)(-1) min(-1), respectively. INAC-INV is a true member of the β-fructofuranosidases, which can react with sucrose and raffinose as substrates. SDS-PAGE and immunoblotting were used to determine the molecular mass of INAC-INV to be 69 kDa. The isoelectric point of INAC-INV was estimated to be about pH 8.0. Taken together, INAC-INV is a pea seedling invertase with a stable and optimum activity at lower acid pH and at higher temperature than other invertases.  相似文献

锌铬复合污染对水稻不同生育期土壤酶活性的影响   总被引:5,自引:0,他引:5  
通过盆栽试验,研究了锌铬复合污染对水稻(Oryza sativaL.)不同生育期土壤酶活性的影响。结果表明,锌铬复合污染对水稻生长有抑制作用,随土壤中锌铬浓度的增加,不同生育期水稻生物量均呈下降的趋势,而根冠比呈一定的升高趋势,差异极显著(P<0.01)。在锌铬污染下,土壤过氧化氢酶活性随土壤中锌铬浓度增大而降低。与对照相比,在水稻分蘖期、孕穗期及灌浆结实期土壤过氧化氢酶活性的最小值分别降低了34.17%、23.02%和4.89%;除在灌浆结实期锌与铬浓度互作间的差异不显著外,其他生育期的锌浓度间、铬浓度间及锌与铬浓度互作间的差异均极显著(P<0.01)。在水稻分蘖期,土壤脲酶活性、转化酶活性随土壤锌铬浓度的增加而降低,但在孕穗期及灌浆结实期则升高,其差异均极显著(P<0.01)。线性回归及偏相关分析表明,在水稻不同生育期,土壤过氧化氢酶活性与土壤中锌、铬浓度均有极显著的线性回归关系,并呈显著或极显著负相关关系,这说明锌铬复合污染对土壤过氧化氢酶活性可能产生协同或加和的抑制效应。土壤脲酶活性在水稻灌浆结实期与土壤中锌浓度有极显著的线性回归关系,偏相关系数为0.7253**;土壤转化酶活性在水稻分蘖期、灌浆结实期与土壤中铬浓度均有极显著的线性回归关系,偏相关系数分别为-0.7426**、0.8056**,这说明锌铬复合污染对土壤脲酶活性及转化酶活性未产生复合效应。  相似文献

The effect of earthworms on soil hydrolases (protease, urease, invertase, and alkaline phosphatase) and dehydrogenase activities was investigated in maize residue amended rice–wheat rotation agro-ecosystem. Experimental plots in the rotation had five treatments, i.e. incorporation or mulching of maize residues with or without added earthworms and an untreated control. The application of maize residues to soil without earthworms significantly enhanced the five soil enzyme activities compared with the control treatment during rice and wheat cultivation. The presence of earthworms further significantly enhanced protease activity in the soils with both incorporated and mulched maize residues during two cultivation seasons, but only significantly increased alkaline phosphatase activity in the soil with incorporated maize residue during the rice cultivation season. Invertase activity was significantly enhanced by the presence of earthworms in the soil with maize residue incorporation during two cultivation seasons. There were no changes in dehydrogenase activity when earthworms were present. Additionally, the five enzyme activities in earthworm casts were significant higher than those in the surrounding soil, especially dehydrogenase and invertase activities. Whatever the treatment, the values obtained for the enzyme activities in both soil and casts, except for dehydrogenase activity in earthworm casts, were significantly higher under wheat than those in rice-cultivated soil. These results indicate that the presence of earthworms strongly affected soil enzyme activities, depending on the method of organic residue application, and the enhanced enzyme activities of earthworm casts probably contributed to the surrounding soil enzyme activities.  相似文献

基于中国科学院海伦农业生态实验站长期定位试验,研究不同土地利用方式下4月份和7月份黑土脲酶、转化酶、磷酸酶活性的变化.结果表明:土地利用方式不同,黑土酶活性有较大差异.草地与裸地相比,4月份脲酶、转化酶、磷酸酶活性分别增加24.66%、29.56%、34.67%,7月份分别增加56.12%、128.40%和30.42%,表明经过自然植被恢复,土壤生物活性增强,土壤肥力提高;农田3种施肥方式相比,脲酶、转化酶、磷酸酶活性大小均为化肥+有机肥处理>化肥处理>无肥处理.3种不同生态系统相比,转化酶、磷酸酶活性大小依次为草地生态系统>农田生态系统>裸地生态系统,脲酶活性大小依次为农田化肥+有机肥处理>草地生态系统>农田化肥处理>农田无肥处理>裸地生态系统.表明采取一定的施肥措施和草地自然植被恢复可增加黑土酶活性,提高土壤肥力.土壤脲酶、磷酸酶和转化酶活性与土壤有机质含量和全氮含量均有显著相关关系,能较好地反映土壤肥力水平.  相似文献

采用盆栽试验,研究大棚蔬菜(津优1号黄瓜)在轮作(15、18和21年)和连作(2、7年)条件下,对土壤过氧化氢酶、脲酶、转化酶和多酚氧化酶活性的影响。结果表明,轮作土壤的过氧化氢酶、脲酶和转化酶的活性显著地高于连作7年的土壤(P0.05);随着连作年限的增加,过氧化氢酶、脲酶和转化酶的活性显著地降低(P0.05),多酚氧化酶的活性显著升高(P0.05)。通过对产量的分析表明,连作2年、7年土壤的黄瓜产量分别为每3盆7.91和3.14.kg,两者差异显著;轮作15、18和21年土壤的黄瓜产量分别为6.65、6.85和7.16kg/3盆,差异不显著;连作2年土壤的黄瓜产量显著高于其它处理(P0.05),连作7年土壤的黄瓜产量显著低于其它处理,黄瓜产量与土壤酶活性的变化趋势基本一致。过氧化氢酶、脲酶、转化酶和多酚氧化酶可以作为设施蔬菜敏感的土壤酶学指标。  相似文献

Thirteen metal elements were determined in 40 honey samples from Galicia with different environmental origins: rural, urban, and industrial areas. The data set of the honey metallic profiles was studied with a double purpose: first, to make a preliminary evaluation of honey as an environmental indicator in Galicia with the aim of monitoring pollution and, second, to compare the different capabilities of diverse pattern recognition prediction procedures for modeling the environmental surrounding of the hive. A certain level of similarity for urban and industrial samples was obtained using principal component analysis and cluster analysis, whereas significant differences for urban and industrial honeys were found in relation to rural honey samples. Different classification rules to associate metal content of honeys with their environmental surrounding were obtained by chemometric pattern recognition procedures. In general, the classification methods developed by neural networks provided better results than the traditional pattern recognition procedures. The metal profiles of honey seem to provide sufficient information to enable categorization criteria for classifying samples according to their environmental surrounding. Thus, honey could be a potential pollution indicator for the Galician area.  相似文献

The potential of near-infrared (NIR) spectroscopy to determine the geographical origin of honey samples was evaluated. In total, 167 unfiltered honey samples (88 Irish, 54 Mexican, and 25 Spanish) and 125 filtered honey samples (25 Irish, 25 Argentinean, 50 Czech, and 25 Hungarian) were collected. Spectra were recorded in transflectance mode. Following preliminary examination by principal component analysis (PCA), modeling methods applied to the spectral data set were partial least-squares (PLS) regression and soft independent modeling of class analogy (SIMCA); various pretreatments were investigated. For unfiltered honey, best SIMCA models gave correct classification rates of 95.5, 94.4, and 96% for the Irish, Mexican, and Spanish samples, respectively; PLS2 discriminant analysis produced a 100% correct classification for each of these honey classes. In the case of filtered honey, best SIMCA models produced correct classification rates of 91.7, 100, 100, and 96% for the Argentinean, Czech, Hungarian, and Irish samples, respectively, using the standard normal variate (SNV) data pretreatment. PLS2 discriminant analysis produced 96, 100, 100, and 100% correct classifications for the Argentinean, Czech, Hungarian, and Irish honey samples, respectively, using a second-derivative data pretreatment. Overall, while both SIMCA and PLS gave encouraging results, better correct classification rates were found using PLS regression.  相似文献

NIR transflectance spectroscopy was used to analyze fructose, glucose, and moisture in honey. A total of 161 honey samples were collected during 1992 (46), 1995 (58), and 1996 (57). Samples were analyzed by instrumental, enzymatic (fructose and glucose), and refractometric (moisture) methods. Initially, different calibrations were performed for each of the 3 years of sampling. Good predictions were obtained for all three components with equations of the particular year. But good predictions were not always obtained when the equations calculated one year were applied to samples from another year. To perform a lasting calibration, unique calibration (121 samples) and validation (40 samples) sets were built; honeys of the 3 years were included in both sets. Good statistics (bias, standard error of validation (SEV), and R(2)) were obtained for all three components of the validation set. No statistically significant differences (p = 0.05) were found between instrumental and reference methods.  相似文献
