The interstitial cells of Cajal (ICC) are c-kit immunoreactive cells of the gastrointestinal tract which are suggested to have a role in the control of intestinal motility. Cells with c-kit immunoreactivity have not been previously described in the gastrointestinal tract of the horse. Immunoreactivity for c-kit was revealed using immunohistochemical labelling with an anti-c-kit polyclonal antibody. Sections of normal gastrointestinal tissue were examined from 13 anatomically defined sites from stomach to small colon taken from horses free from gastrointestinal disease. Three types of c-kit immunoreactive cells were identified: spindle-shaped cells in the region of the myenteric plexus, stellate or bipolar cells in the circular muscle layer, and round cells in the submucosa. The round cells were shown to be mast cells with the use of toluidine blue staining, whereas the other c-kit immunoreactive cells did not exhibit metachromasia and were classified as ICC. This study will serve as a basis for future pathological studies in the horse. 相似文献

In this study we investigated the histological changes of the myenteric plexuses and interstitial cells of Cajal (ICC) in gut samples from horses with colic to try to find results useful in the prognostic evaluation of enteric lesions. A morphologic and quantitative study of myenteric ganglia, ganglion cells and neuronal chromatolytic and necrotic changes of 24 horses with colic was performed. For ganglion cells, enteroglial cells and ICC immunolabeling was also performed to identify cell functional disorders. A significant increase of neuronal chromatolysis and necrosis occurred in horses suffering from colic throughout the gut. The neuron-specific enolase (NSE) and synaptophysin immunoreaction quantified with image analysis showed a significant loss of neuronal activity in all intestinal tracts of the animals under study associated with a significant loss of ICC immunoreactivity. The results supports immunohistochemical evaluation of ENS and ICC as a useful tool along with morphometric investigations in the evaluation of gut lesions produced during colic syndrome. 相似文献

Chronic ingestion of yellow star thistle (Centaurea solstitialis) or Russian knapweed (Acroptilon repens) causes nigropallidal encephalomalacia (NPE) in horses with an abrupt onset of neurologic signs characterized by dystonia of lips and tongue, inability to prehend food, depression, and locomotor deficits. The objectives of this study were to reexamine the pathologic alterations of NPE and to conduct an immunohistochemistry study using antibodies to tyrosine hydroxylase and α-synuclein, to determine whether NPE brains show histopathologic features resembling those in human Parkinson disease. Results confirm that the NPE lesions are located within the substantia nigra pars reticulata, sparing the cell bodies of the dopaminergic neurons in the substantia nigra pars compacta, and in the rostral portion of the globus pallidus, with partial disruption of dopaminergic (tyrosine hydroxylase-positive) fibers passing through the globus pallidus. No abnormal cytoplasmic inclusions like the Lewy bodies of human Parkinson disease were seen in these NPE brains. These findings indicate that equine NPE may serve as a large animal model of environmentally acquired toxic parkinsonism, with clinical phenotype directly attributable to lesions in globus pallidus and substantia nigra pars reticulata rather than to the destruction of dopaminergic neurons. 相似文献

Obesity is becoming a major health concern in horses because of its associations with insulin resistance, oxidative stress/inflammation, and laminitis. However, there is limited information on the prevalence of obesity in horses. The USDA National Animal Health Monitoring System (NAHMS) 1998 Equine study estimated that approximately 1.4% of the U.S. horse population is overweight or obese. The purpose of this study was to determine the prevalence of overweight and obesity in a subpopulation of horses in Virginia. A random sample of 300 mature, Light Breed horses (140 mares, 151 geldings, and 9 stallions) was selected from the VMRCVM Equine Field Service practice. Horses were evaluated during summer 2006. Body Condition Score (BCS) was assigned using a scale of 1 (emaciated) to 9 (obese) by two independent scorers. Neck circumference (cm) was measured at three locations and averaged (ANC). Girth, body length, and height at the withers were measured and used to calculate body weight (BW) and Body Mass Index (BMI). ANC and height at the withers were used to calculate Neck Circumference to Height Ratio (NCHR). Horses were categorized based on BCS as under condition (BCS <4), optimal condition (BCS 4–6), over condition (BCS 6.5–7), and obese condition (BCS 7.5–9). Six horses (2%) were under condition, 141 horses (47%) were in optimal condition, 96 horses (32%) were over condition, and 57 (19%) were obese. BW (p = 0.047), ANC, BMI and NCHR (all p < 0.001) increased with BCS. The prevalence of overweight and obesity in these horses was much higher than previously reported. 相似文献

Antithrombin-III (AT-III) activity was measured at the time of admission in 46 horses referred with colic. Differences in AT-III activities between animals treated medically or surgically and survivors or non-survivors were compared. The mean AT-III values for the horses treated medically (76.2 per cent), surgical survivors (69.5 per cent) and surgical non-survivors (55.9 per cent) were significantly different from the reference value for healthy adults (92 to 108 per cent). The mean AT-III activity of the survivors was significantly (P less than 0.01) greater than that of the nonsurvivors. The majority of the survivors (11 to 13 horses) had AT-III activities exceeding 60 per cent of normal, whereas that of 10 of the 14 non-survivors was less than 60 per cent of normal. There was an apparent distinction between the survivors and non-survivors at approximately 60 per cent of normal AT-III activity. 相似文献

This study highlights the importance of interstitial cells of Cajal (ICs) in gastrointestinal disease. Human research is already considering IC pathologies but in veterinary research IC pathologies are rarely studied. Nevertheless, recent studies of ICs show a growing interest in the pathophysiology of gastrointestinal diseases and emphasize the consideration of this cell type in the pathophysiology of veterinary gastrointestinal malfunctions. 相似文献

Objective: To determine the physiologic response of the equine navicular bone to core decompression surgery in healthy horses. Study Design: Experimental in vivo study. Materials: Healthy adult horses (n=6). Methods: Core decompression was completed by creating three 2.5‐mm‐diameter drill channels into the navicular bone under arthroscopic control. The venous (P V), arterial (P A), articular (P DIPJ), and intraosseous pressures (IOP) were recorded before and after decompression drilling. Each IOP measurement consisted of a baseline (IOP B) and a stress test (intramedullary injection of saline solution, IOP S) recording. Lameness was assessed subjectively and using force plate gait analysis. Fluorochrome bone labeling was performed. Horses were euthanatized at 12 weeks. Navicular bone mineral density (BMD) was measured, and bone histology evaluated. Results: Peak IOP (IOP max) after stress testing was significantly ( P<.05) reduced immediately after core decompression; however, the magnitude of these effects was decreased at 3 and 6 weeks after decompression. A significant ( P<.05) correlation existed between IOP max and BMD. No lameness was observed beyond the first week after surgery. Substantial remodeling and neovascularization was evident adjacent the surgery sites. Conclusion: Navicular bone core decompression surgery reduced IOP max, and, with the exception of a mild short‐lived lameness, caused no other adverse effects in healthy horses during the 12‐week study period. 相似文献

Many causal hypotheses for acute equine abdominal disease (colic) have been proposed. However, few epidemiologic studies have been undertaken to identify risk factors for the syndrome. Risk factors for equine colic were identified using a hospital-based, multi-center, case-control study. Horses admitted with abdominal pain to Cornell, Guelph, Ohio State, Pennsylvania or Tufts university veterinary hospitals between March and December 1991 were selected. Control horses were randomly selected from a list of all other non-colic admissions. The person(s) responsible for the day-to-day care of the horse completed a telephone questionnaire. Risk factors were identified using multivariable logistic regression analysis. A total of 406 colic and 406 control horses were used in the final analysis. Age, breed, outdoor access (pasture or drylot), outdoor water supply, use of a daily worming product, amount of concentrates and whole-grain corn consumed, type of person responsible for the day-to-day care of the horse and previous colic history were all significantly associated with colic risk. Arabian horses were more than twice as likely to be colic cases (OR = 2.0, 95% CI = 1.0–3.9), whereas Standardbred horses were nearly half as likely (OR = 0.6, 95% CI = 0.4–1.0) to be colic cases compared with the comparison Thoroughbred group. Horses that had access to outdoor enclosures without a continuous supply of water were more than twice as likely (OR = 2.2, 95% CI = 1.2–4.3) to be colic cases compared with horses that had an adequate supply of water on outside enclosures. The use of a daily worming product for at least 60 days during the 12-month period prior to presentation was associated with a significantly lower colic risk (OR = 0.11, 95% CI = 0.02–0.55). Colic risk increased with greater consumption of whole-grain corn; however, when all non-roughage concentrate feeds were combined, colic risk decreased with increased intake of concentrates. Significant interactions were identified between age and drylot use and between the type of care-person responsible for the day-to-day care of the horse (owner or non-owner) and previous colic history. The findings of this study illustrate the complex multi-factorial nature of equine colic and point towards the importance of the overall quality of stable management and care, as well as the type of outdoor enclosure (pasture or drylot), water access, carbohydrate intake, parasite control and intrinsic (breed-related) factors. 相似文献

OBJECTIVE: To identify apoptosis in equine intestines and determine whether apoptosis is associated with gastrointestinal tract disease or a specific tissue layer of intestine. ANIMALS: 38 horses that underwent surgery or were euthanatized for small or large intestine obstruction, strangulation, or distension and 9 control horses euthanatized for reasons other than gastrointestinal tract disease or systemic disease. PROCEDURE: Specimens were collected at surgery from intestine involved in the primary lesion and distant to the primary lesion site or at necropsy from several sites including the primary lesion site. Histologic tissue sections were stained with H&E, and apoptosis was detected by use of the terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling technique. The number of apoptotic cells per hpf was counted in the mucosa, circular muscle, longitudinal muscle, and serosa. RESULTS: Apoptotic nuclei were seen in all layers of intestine. An increased number of apoptotic cells was found in the circular muscle of the intestine from horses with simple obstruction, compared with strangulating obstruction or healthy intestine. Intestine distant from a primary strangulating lesion had higher numbers of apoptotic cells than did intestine distant from a simple obstructive lesion or intestine taken at the site of a strangulating or simple obstructive lesion. CONCLUSIONS AND CLINICAL RELEVANCE: Intestine from horses with obstructing or strangulating lesions in the small intestine and large colon had high numbers of apoptotic cells possibly because of ischemic cell injury and subsequent inflammation. Whether substantial apoptosis affects intestinal function is not yet known. 相似文献

Interstitial cells of Cajal (ICC) have been described in the gastrointestinal tract of different mammals including humans,
horses, pigs, rats, dogs, mice and guinea-pigs. In the present study, ICC were identified in the jejunum of Bos taurus using polyclonal anti-c-Kit antibodies in immunohistochemical assays. Vimentin and desmin intermediate filaments were also
determined using monoclonal antibodies. ICC were found in the tunica muscularis either in a palisade distribution pattern
between the outer longitudinal and the inner circular layers (ICC-MP) or freely distributed in clusters in the longitudinal
layer (ICC-LM). Morphometric studies determined that ICC have a fusiform shape presenting cytoplasmic prolongations. ICC were
positive to c-Kit and vimentin antigens but negative to desmin. We have observed and described for the first time the presence
of ICC in a ruminant. As observed in the aforementioned mammals, bovine ICC were associated with the myenteric plexus. Nevertheless,
the presence of widespread ICC in the longitudinal muscular layer of the jejunum differs from previously described studies

REASONS FOR PERFORMING STUDY: Circumferential mural band (CMB) in the small intestine is an emerging condition in Ireland. The clinical presentation is a simple obstruction of the small intestine of unknown aetiology. HYPOTHESIS: Horses affected with CMB in the small intestine have a good prognosis following surgical decompression into the caecum. METHODS: A retrospective study was carried out and 28 cases identified from 559 colic surgeries performed over a 5.5 year period. An exploratory celiotomy was performed in every case. A simple obstruction at the mid or distal jejunum was identified caused by a CMB, which was multiple in some cases. All of the horses except 2 had undergone a regular anthelmintic programme. RESULTS: Manual decompression of the small intestine into the caecum resulted in resolution of the simple jejunal obstruction. A constricted segment of small intestine was resected in one case, as the degree of constriction was severe. A full thickness biopsy was taken from another case. Histopathology was performed on both samples. All horses recovered from surgery and were discharged from the hospital 7-32 days post operatively. Short-term complications were observed. Survival rate was 100% at long-term follow-up. Histopathology showed inflammatory infiltration in the submucosa, muscularis and serosa with eosinophils predominating. Parasites were not detected. CONCLUSION AND POTENTIAL RELEVANCE: Small intestine circumferential mural bands have a good prognosis after surgical decompression of the small intestine into the caecum. 相似文献

Thirty horses were admitted to the University of Georgia Veterinary Medical Teaching Hospital with colic severe enough to warrant surgery and/or euthanasia. Gastrointestinal tracts of these 30 horses were histologically examined for morphologic changes. The horses were grouped according to cause of the colic (ie, simple obstruction, strangulation obstruction, thromboemboli, and inflammation). Lesions were graded as to severity, and grade scores were correlated with survival or nonsurvival. Mucosal changes developed distal and proximal to the primary lesion site and, although there were some differences between groups, changes characteristic of ischemia were common to all groups. The predictability of lesion grades of 2 or higher for nonsurvivability (90%) indicates that intestinal biopsy may have prognostic value in the postsurgical evaluation and management of equine colic. 相似文献

The article provides hands-on advice and scientific background information regarding the management of the equine acute colic patient. It summarizes essential information on the patient's history, clinical examination, and legal aspects. The scope of the history is based on the clinical appearance of the patient. In horses with violent and barely controllable signs of pain, it is limited to colic-associated information such as onset, course and severity of colic signs, and the last time of defecation, as well as prior therapeutic intervention. In these patients, the clinical examination is focussed primarily on the assessment of cardiovascular parameters, rectal temperature, and the patient's behaviour. The nasogastric intubation is an essential part of the examination of a horse with colic to prevent a gastric rupture due to gastric distension. Transrectal palpation is equally important and should always be performed unless there are important reasons to object to this procedure. In most cases, a thorough patient history and clinical examination of an acute colic patient allows the examining veterinarian to make a tentative diagnosis and a prognostic evaluation. This helps with the decision to start a conservative or a surgical therapy. Due to the existing obligatory documentation requirement and the duty to inform patient owners, it is of utmost importance to promptly document one's findings, therapeutic measures, consultations, and information of the patient owner in written form. This supports further therapy and it can also be of relevance in a potentially resulting lawsuit. 相似文献
