Considering overcrowding as a stressor, the effects of a short-term exposure on the immune responses of Atlantic cod, Gadus morhua L. were determined at 2, 24 and 72 h post-crowding. While plasma glucose and total antioxidant capacity were quantified as stress responses, immune responses were based on humoral indices. Bactericidal capacity of plasma was also assessed. There was a significant increase in plasma glucose and total antioxidant capacity at 2 h post-crowding, however the former returned to pre-stress levels 24 h later and the latter remained elevated until 72 h. The plasma protein did not reveal any significant change, but there was a transient upregulation of globulin at 24 h post-crowding. The myeloperoxidase activity increased at 2 h after the stress, but returned to its pre-stress levels at 24 h. The lysozyme activity was upregulated even from the first time point after stress, although the differences were significant only at 24 and 72 h compared to the activity prior to stress. At 72 h post-crowding there was a significant increase in alkaline phosphatase activity while haemolytic activity showed a significant decrease. An increase in the proliferation of the bacterial pathogens, Vibrio anguillarum and Aeromonas salmonicida was seen from 24 h in the plasma of the stressed fish. Thus, our finding suggests that short-term crowding stress modulates the immune responses in Atlantic cod and therefore husbandry procedures should consider these effects while addressing better management strategies.  相似文献

To ascertain whether growth rate modifies the oxidative capacity of fish white muscle, we examined the effects of individual growth rate on the activities of four mitochondrial enzymes in white muscle of the fast growing Atlantic cod,Gadus morhua. Growth rates were individually monitored in cod held at three acclimation temperatures during experiments repeated in four seasons. The size dependence of citrate synthase (CS), cytochrome C oxidase (CCO) and β-hydroxyacyl CoA dehydrogenase (HOAD) activities was established using wild cod ranging from 115 to 17,350 g. Given their negative allometry, CS and CCO activities in the experimental cod were corrected to those expected for a 1.2 kg animal. HOAD activities did not change with size. The specific activities of CCO and CS were positively correlated with growth rate. However, for both enzymes, season explained more of the variability than growth rate or temperature. Season was the only factor to significantly affect the activity of HOAD, while temperature and season interacted to determine glutamate dehydrogenase activity. CS activity was positively correlated with the initial condition of the cod, which differed among the seasons. The other enzymes did not show this relationship. The independent changes of these enzymes suggest that mitochondria undergo qualitative modifications with changes in growth rate, season and size. Although growth rate and the activities of CCO and CS are positively correlated, the activity of the mitochondrial enzymes is more affected by size, physical condition and season.  相似文献

Is there a genetic basis to growth in Atlantic cod?   总被引:4,自引:0,他引:4  
There is still much disagreement and debate about whether or not genetically based growth differences occur in Atlantic cod, and there is evidence on both sides. In this review, data on genetically based growth differences in cod will be presented to shed light on this hypothesis. Motivated by the hypothesis that growth patterns may reflect specific genotype adaptations, we review stock‐specific responses on growth. An example of genetically based differences between the population units at two spawning localities off south Iceland is discussed. Here, significant differences in growth performance of the different SypI genotypes were found. Also, the cod sampled at Loftstaðahraun displayed higher mean weight and length compared to the cod from Kantur indicating that these population units may display different life histories. Other studies have shown conflicting results depending on which side of the Atlantic the problem has been investigated. We propose that a common‐garden meta‐analysis with several cod stocks from both sides of the Atlantic is needed to give any reasonable answer to the question of genetically based growth differences. Until such studies have been conducted, it is premature to conclude one way or the other. In this review, we have not tried to quantify how large the environmental part of growth regulation versus the genetic part is, as this information is not available in the published literature on cod. Based on recent research on two flatfish species (turbot and Atlantic halibut), approximately 30% of growth variation is caused by genetical factors, but it remains to be seen if this is similar in cod. A fruitful way to continue this research might be to conduct controlled experiments, where performance (growth, food intake, feed conversion efficiency, feeding behaviour, etc.) and environmental factors (e.g. temperature, oxygen, photoperiod, predation risk, food availability) are studied simultaneously for different genotypes and different stocks.  相似文献

Atlantic cod, Gadus morhua, were maintained on a diet of sandeel and after a 6-day fast were refed a single meal. Concentrations of free amino acids (AAs) were measured in hepatic portal and cardiac blood as well as in the stomach and white muscle at intervals of 6h up to 24h post-feeding. The appearance of both essential and non-essential AAs in the hepatic portal blood was significantly correlated, up to 12h after feeding, to their abundance in the diet. There was a significant decline in total AA concentration in cardiac blood after 6h, followed by a significant increase at 12h. No significant changes in total AA concentration were observed in the other tissues, although mean concentration increased at 12 or 18h. At a more detailed level, the post-prandial changes in concentration of some essential AAs were consistent with their having a role in the stimulation of protein synthesis after feeding.  相似文献

The growth and feed utilization of Atlantic cod (Gadus morhua) (437 g), Atlantic salmon (Salmo salar) (485 g) and rainbow trout (Oncorhynchus mykiss) (413 g) fed a diet (170 g kg−1 fat, 600 g kg−1 crude protein; LE) similar to that used in commercial cod production or one that was top dressed with additional fat (280 g kg−1 fat, 530 g kg−1 crude protein; HE), were compared in an 11‐week trial. In the cod, relative feed intake was 41–58% and thermal growth coefficient 63% of that in the salmonids, but the feed efficiency ratio (FER) was 38% better (P ≤ 0.05). In contrast to the cod where there was no effect of diet on feed intake, growth or FER, both the salmon and trout fed the HE diet had greater feed intake than those fed the LE diet, but the effect of this was only positive for growth in the salmon. The cod retained more of the digested nitrogen (44.9 ± 2.7%) than the salmon (39.4 ± 0.8%), and both of these species retained more than the trout (33.6 ± 1.1%) (P ≤ 0.05). The retention of digested energy was significantly higher in the salmon (52.2 ± 0.9%) than in the trout (44.8 ± 1.1%), with the cod (44.9 ± 4.9%) not different from either of the other species. There were no differences between the species in the retention of absorbed phosphorus (65.9 ± 3.6%). There were very few dietary effects on nutrient utilization in this trial and, for the cod, this indicates that higher energy diets may be feasible for use in production.  相似文献

Microsatellite DNA loci and the Pantophysin locus (Pan I) were used to investigate levels of genetic diversity within farmed strains of Atlantic cod Gadus morhua and to compare them with the wild source population. A total of 282 farmed samples originating from a spawning ground off the south-west coast of Iceland were sampled in the years 2002 and 2003, and 258 wild cod were collected at the same spawning ground in the same years. The farmed strains exhibited a lower mean number of alleles and allelic diversity than the wild samples at the microsatellite loci. Significant differences were observed between wild and farmed samples both in allele and genotype frequencies at the Pan I locus. We argue that the genetic divergence of wild and farmed samples of Atlantic cod may be due to a small number of effective founding breeders contributing to the genetic variation of the farmed strains, inducing a reduction in allelic diversity. We discuss the potential effect of breeding practices on the genetic diversity of Atlantic cod.  相似文献

The objectives of this work were to evaluate how dietary soybean meal (SBM) or a soy meal made by bioprocessing the SBM (BPSBM) to remove anti-nutritional factors affected hydrolytic capacity, amino acid absorption, intestinal morphology, and microflora along the intestinal tract of Atlantic cod at two life stages. Three fish meal based standard cod diets were formulated to contain no soy (FM control), 25% SBM, or 22% BPSBM. Prior to sampling the diets were fed to duplicate groups of 0.5 kg (1-year old) and 1.7 kg (2-year old) cod for a period of 3 months, and the groups reached 0.9 and 2.5 kg, respectively. Digesta was then sampled from different intestinal sections for analyses of trypsin and amylase activity as well as absorption of amino acids, nitrogen, and sulphur. Gastrointestinal sections were sampled for measurements of relative weight (kg− 1 body weight), and tissues from these sections were sampled for analyses of brush border enzyme (alkaline phosphatase (ALP), leucine aminopeptidase (LAP), and maltase) activity and histological examination. Microflora was sampled from both digesta and the intestinal wall. The SBM diet stimulated relative growth of all gastrointestinal sections except the distal intestine in both age classes. Relative growth of the pyloric intestine was also stimulated by BPSBM. The pyloric caeca and the upper mid intestine were found to be the major sites for enzymatic hydrolysis of protein and starch and for amino acid absorption. Dietary SBM and BPSBM did not alter the activity of trypsin and LAP, but the activity of these enzymes in the proximal intestine was affected by age, being higher in 1-year old than in 2-year old cod. The rate of amino acid, nitrogen, and sulphur absorption along the gastrointestinal tract was not affected by SBM, but was slowed by BPSBM. Dietary SBM or BPSBM did not alter the morphology of the intestinal mucosa in any sections of the cod intestine. The distal-most structure of the intestine, a compartment with inlet and outlet (anus) valves, showed very high microbial colonisation in the mucosal brush border. Inclusion of SBM in the diet changed the intestinal microflora, increasing the population level of transient bacteria in the pyloric and mid intestine, but reducing the population level of adherent bacteria throughout the intestine. To conclude, Atlantic cod appeared to have a robust and flexible digestive system able to adjust to high dietary levels of soy protein meals.  相似文献

Juvenile Atlantic cod (Gadus morhua) were fed extruded feeds formulated to contain 360–660 g kg?1 protein, 80–280 g kg?1 lipid and 80–180 g kg?1 starch at feeding frequencies of either once per day or every second day to satiation. The trial was conducted at 8 °C and lasted for 28 weeks during which fish were weighed five times at regular intervals. Sampling for proximate analysis was performed at the start, after 12 weeks and at the end of the trial. Fish grew from an average weight of 192 g to between 750 and 866 g, with growth being negatively affected by low dietary protein concentration. High dietary starch concentrations had some negative effects on growth, whereas changes in dietary fat concentration had no significant effect on growth. Liver indices (at the end of the experiment) varied between 80 and 170 g kg?1, and there was a negative correlation between the ratio of protein to fat and liver index. Feed conversion ratio (FCR) ranged between 0.74 and 0.88, and feed utilization improved with increasing concentrations of dietary protein and fat. Increasing dietary starch concentrations resulted in poorer feed utilization. To achieve good growth and protein retention, and avoid excessive liver size in juvenile cod, feeds should contain 500–600 g kg?1 crude protein, 130–200 g kg?1 lipid and <150 g kg?1 starch.  相似文献

Aggression in groups of 0+ Atlantic salmon (Salmo salar) was monitoredat weekly intervals in two tanks containing 100 fish each. Three 1 + salmonparr were added to one of these. After 5 weeks, fish weights were measuredin both tanks and the conditions reversed. At ten weeks, weights of fish inboth tanks were measured again. In both populations, levels of aggressionamong the smaller fish were significantly lower and growth ratessignificantly higher when the large fish were present. Although the largefish attacked the small ones, the rate at which they did so was an order ofmagnitude lower that the rate at which small fish attacked each other in theabsence of larger conspecifics. This raises the possibility that levels ofaggression among farmed salmon might be reduced by the addition of a fewlarge conspecifics.  相似文献

A mark and recapture study of cod in the western Gulf of Maine was conducted to study the seasonal movements of fish, particularly as they related to areas closed to commercial fishing. A total of 27,772 cod were tagged, and 1334 (4.8%) were recaptured with sufficiently detailed recapture location and date to be included in the study. Results indicated that the group is resident to the area and sedentary. Although there were a small percentage of fish (2.5%) that traveled long distances (>100 km), most were recaptured near their release location. There was no linear relationship between fish length and linear distances traveled, and linear distance traveled was only weakly related to days-at-large. For all groups of cod tagged and released in particular areas and months, mean distances traveled were small (<65 km), rates of travel were slow (<0.2 km/day), and rates of group dispersion were <50 km2/day. There were no recognizable spatial or temporal patterns in the mean angles of travel for groups released in various month/area combinations, and the angular deviations associated with the mean angles were quite large. Although there seems to be little pattern in the movement of cod in our study area, temporal and spatial changes in abundance indicate that movements are occurring.

The general pattern was a concentration of large cod into one small, inshore area (Area 133) in both the spring and winter, and dispersion from this area in the ensuing months. Monthly percentages of ripe cod in Area 133 provide evidence for two spawning groups; a winter group that spawns from November through January, and a spring group that spawns from April through July. Thus it is likely that the observed spring and winter concentrations of fish in Area 133 were associated with spawning. Fish in the spring group were the largest encountered in the study. The study also provided some evidence of natal homing, i.e. a return to the same spawning grounds year after year, for the spring spawning group. Although there is some exchange of fish between most of the closed areas, the timing of the closures appears to protect the largest aggregations of cod.  相似文献

The cod fishery at the Faroes has been managed since 1996 by an effort management system where it is assumed that there is a direct relationship between fishing mortality and fishing effort. We show that this relationship is weak. Due to a factor five variation in the primary production on the Faroe Plateau, the annual growth rate of cod may vary by a factor of five. Commercial catch-per-unit-effort data in combination with stock assessment estimates showed that there was a negative correlation between the annual growth rate of cod and their catchability with longlines, as well as with the total fishing mortality of cod on the Faroe Plateau. Furthermore, mark-recapture experiments showed that longline-caught cod in comparison with trawl-caught cod had a lower condition factor, a lower stomach content of natural prey, and a higher content of longline baits. During the feeding season in September–December, longline-caught cod exhibited a larger displacement distance than trawl-caught cod. These results were largely confirmed by storage tag data. Consequently, during low-productive periods, (i) the longline-dominated cod fishery at the Faroes exerted fishing mortalities that by far exceeded safe biological limits, and (ii) longline catch-per-unit-effort series might overestimate stock size considerably.  相似文献

Digestive capacity and compensatory growth in Atlantic cod (Gadus morhua)   总被引:1,自引:0,他引:1  
The objective of this study was to examine whether digestive capacity correlates with growth rate in Atlantic cod (Gadus morhua). Ninety fish (1035±240 g; 478±36 mm) were assigned to one of three treatments: deprived of food for 5 weeks and then re-fed for 24 days, deprived for 10 weeks and then re-fed for 24 days, or fed for 24 days without deprivation (controls). Three times a week during feeding, the fish were provided with meals of capelin (Mallotus villosus) in excess. Within each treatment, ten fish were sacrificed before and twenty after the feeding period. The relative masses of the pyloric caeca, intestine, and white muscle were determined. Cytochrome c oxidase (CCO), citrate synthase (CS), and nucleoside diphosphate kinase (NDPK) activities were assayed in the pyloric caeca, intestine, and muscle as was trypsin activity in the pyloric caeca. During the re-feeding period, fish that had been deprived of food for 10 weeks showed compensatory growth, growing 1.9 times faster than the controls (0.94±0.26 versus 0.50±0.31% body mass ⋅ day−1). Fish that displayed compensatory growth had a lower relative white muscle mass than controls (66.5±4.1% vs. 69.6±7.4%) while the relative masses of the pyloric caeca (2.21±0.49% vs. 1.78±0.31%) and intestine (0.95±0.20% vs. 0.77±0.15%) were greater than the controls, suggesting that the sizes of these digestive tissues might correlate with compensatory growth capacity. Citrate synthase in the pyloric caeca was the only enzyme that showed higher activity during compensatory growth (8.57±0.94 U ⋅ g tissue−1 compared to 7.13±1.03 U ⋅ g tissue−1 in the control group). This suggests that aerobic catabolic capacity of pyloric caeca could be related to growth capacity during recovery, possibly via the energetic cost of digestive enzyme synthesis. Fluorescein isothiocyanate dextran (FITC-dextran) exchange between the primary (PCS) and secondary (SCS) circulatory systems in the Atlantic cod, Gadus morhua (Linnaeus, 1752), were studied using 20-kDa (n = 4) and 500-kDa (n = 4) FITC-dextran. In order to give a qualitative perspective of the general connection between the PCS and SCS, distribution of plasma-borne tracers (FITC-dextran) in the PCS and SCS were examined. In this study, a total of eight cod were cannulated in the ventral aorta (PCS) and dorsal cutaneous vessel (SCS), for investigation of FITC-dextran disappearance in the PCS and its subsequent appearance in the SCS. FITC-dextran of both sizes was found to be in equilibrium between the PCS and SCS in less than 20 min. This indicates a profound connection between the PCS and SCS in the Atlantic cod, and rapid mixing of tracers between the PCS and SCS. The destination of the injected 500-kDa FITC-dextran was also examined, and it was observed that of the 500-kDa FITC-dextran lost from the primary and secondary vascular systems, 63.0 ± 9.2% could be recovered from the liver  相似文献

During the development of breeding programme for Atlantic cod Gadus morhua L., in Iceland, genetic parameters were estimated for 1402 individuals, which were assigned with DNA profiling to 140 dams and 70 sires. The cod was reared in cages on the eastern and western coasts of Iceland from 2004 to 2005. At the average body weight of 1.8 kg, the estimated heritability (h2 ± SE) for body weight, gutted weight and the condition factor (CF) were 0.31 ± 0.06, 0.34 ± 0.04 and 0.24 ± 0.06 respectively. Genetic correlation (rG) in body weight between the two rearing locations was estimated as 0.95, which reflects a low G × E interaction. The estimated heritability for hepatosomatic index (HSI) and fillet yields was 0.061 ± 0.04 and 0.04 ± 0.04 respectively. The HSI and fillet yields were highly genetically correlated with body weight or 0.67 and 0.82 respectively. The genetic correlation between the CF and body weight was estimated as 0.31. There appears to be substantial amount of additive genetic variation for body weight suggesting that selection is likely to be successful. Low heritability for fillet yields and the HSI indicates less promise of genetic improvement. Assigning of parentage to individuals with DNA profiling was 80% successful.  相似文献

A deoxyribonuclease (DNase) of pancreatic origin has been purified from extracts of the pyloric caeca from Atlantic cod (Gadus morhua L.). The crude extract was prepared by mincing frozen caeca tissue in equal volumes of buffer. The enzyme was isolated from the supernatant after streptomycin sulfate precipitation and centrifugation. The purification scheme further included chromatography on Q-Sepharose Fast Flow and hydroxyapatite columns. Affinity adsorption chromatography of the hydroxyapatite fraction on 8-(6-aminohexyl)-amino-5′-AMP-Sepharose, revealed an apparently homogeneous protein with molecular weight of 35,000 Da as judged by NaDodSO4-PAGE. In sum a 644-fold enzymatic enrichment and 3.5% total enzyme recovery was achieved. The cod enzyme resembles DNase I-type enzymes with an alkaline pH activity optimum and shows dependency for Mg2+. The pI of the enzyme is 6.5 as determined by isoelectric focusing and DNase-zymography. Our findings suggest that the nuclease is a member of the cod's digestive enzymes secreted from the connective tissue surrounding the caeca.  相似文献

Escapes of cod (Gadus morhua) from sea cages represent an economic problem for farmers and a potential environmental problem. We estimate that 0–6% of cod held in sea‐cage farms in Norway were reported to have escaped each year from 2000 to 2005, which is a high proportion compared with salmon. We interviewed employees at 19 coastal sea‐cage cod farms in Norway to investigate both how and why cultured cod escape and to document cage handling and management strategies that were effective in minimizing escapes. Based on the interviews, we describe five working hypotheses that may explain why a greater proportion of cod than salmon escape: (1) cod are more willing to escape than salmon; (2) cod bite the net cage and create wear and tear; (3) net cages have insufficient technical standards for cod culture; (4) cod are placed in sea cages at considerably smaller sizes than salmon; and (5) cod are more popular feed for predators. Preliminary testing of the hypothesis that cod bite netting and create holes was done by placing pre‐damaged net panels with cut twines and control panels inside sea cages. Holes in the pre‐damaged net panels increased in size over a period of 3 months. The type of damage indicated that biting of netting twines was the likely cause.  相似文献

This study has investigated the muscle growth of diploid and triploid Atlantic cod (Gadus morhua) juveniles raised in replicate tanks over a period of 29 weeks and analysed at three sampling points (February, June and September). Data for weight, length, condition factor (K), muscle fibre growth and myogenic progenitor cells (MPCs) number were collected and results were analysed in relation to body growth and ploidy status. Diploids were significantly heavier than triploids throughout the trial (~10–20%) and had K in June and September samplings. Over the whole period, the rate of muscle fibres' recruitment was 318 fibres per day and 252 fibres per day for diploid and triploid cod respectively. The larger body weight of diploids resulted in a total number of fast fibre number of 114 979 compared to 91 086 in triploids. The average diameter of the 2.5% of the smallest fibres (2.5th percentile) was higher in diploids than triploids at the start of the trial, with a reversed picture for the average of the upper 2.5% (97.5th percentile) at the end of the trial. The probability density function of the estimated muscle fibre diameters showed similar fibre size distribution between size‐matched diploids and triploids at all sample points. The peak fibre diameter was approximately 25 μm in February and increased to approximately 50 μm in June and September, irrespectively of ploidy. Pax 7 were used as molecular markers for MPCs. A positive correlation between Pax 7+ cells and total body length was observed only among triploid fish at the onset of the experiment.  相似文献

The time course of appearance of catecholamine metabolites was studied in the spleen of cod,Gadus morhua, during perfusion with radioactively labelled noradrenaline and adrenaline at 10°C. The tlag for appearance of the metabolites ranged between 1.78 and 6.76 min after onset of perfusion for both amines, indicating a rapid disposition of catecholamines. Perfusion with noradrenaline resulted in mainly MOPEG, VMA and DOMA formation, while perfusion with adrenaline additionally resulted in MN formation. There was still formation of deaminated metabolites after denervation, which indicates an additional non-neuronal site of deamination. It is concluded from the study that the fate of noradrenaline and adrenaline within the cod spleen depends on their affinities for the two uptake mechanisms and an extraneuronal site of deamination of great importance cannot be excluded.  相似文献

Cod bite on aquaculture net cages has resulted in damages like frayed netting and holes, which in part can explain why cultured cod have escaped more frequently than salmon over the last years. We describe damages found on various netting materials subjected to cod bite through field experiments at commercial cod farms. Further, a method to test local cod bite resistance of traditional netting structures is suggested and initial results from a test jig prototype are given. Results from field experiments indicated that cod may have been attracted by types of netting that made it possible to draw filaments out of the twine, while stiff, coated netting structures and thick filaments showed no sign of bite damage during the test period. We concluded that netting materials for cod aquaculture must be resistant to cod bite or be repellent or uninteresting for cod. Based on the present findings, the better choice among the traditional netting materials seemed to be hard-laid netting materials, preferably with a primer that glues the filaments together.  相似文献
