To investigate the usefulness of culture for the confirmation of brucellosis in cattle, a comparison of culture and serology was undertaken on 248 animals in four dairy herds where the disease was active. Paired supramammary (SM), retropharyngeal (RP), and internal iliac (IL) lymph nodes were cultured, and five serological tests were deployed: the microserum agglutination test (MSAT), complement fixation test (CFT), the indirect (iELISA) and competitive ELISA, and the fluorescence polarisation assay (FPA). Brucella abortus was isolated from 86.8% of animals on combined culture of all three lymph nodes. Individually, the highest isolation rate was from the RP (90.5% of culture positives). Of culture positive animals, 13.7% and 6.2% were positive from the RP and SM alone, respectively.Approximately half of the positive cultures yielded <10 colonies/culture plate. Although 80.9% of animals were positive in at least one serological test, only 45.2% were positive in all five. For culture-positive animals, the MSAT was the most sensitive test (71.8%). Of the culture-negative animals 67.7% were positive in at least one test, while 12.9% were positive in all five. Titres were higher in animals culture-positive from the SM, and there was a direct correlation between higher titres and higher colony counts in SM cultures. Only 8.9% of animals were both culture-negative and seropositive (in at least one test), while 16.5% were culture-positive and seronegative in all five tests. The results highlight and validate the sensitivity of bacteriological culture in confirming a diagnosis of bovine brucellosis. While the MSAT and FPA were the most sensitive serological tests, a significant percentage of infected animals were undetectable using these standard serological assays.  相似文献

验证牛结核菌素阳性反应牛的结核病变及其病变的组织分布及病变特征,追踪感染源及传播途径,为奶牛场结核检疫净化提供技术支持。在新疆某规模化奶牛场应用牛结核菌素皮内变态反应(SIT)、IFN-γ试验、PPD点眼反应、ELISA抗体检测4种方法联合检疫,综合判定感染牛23头,全部扑杀并进行系统剖检,采集病料,进行病理组织学和细菌学诊断。剖检结果表明,23头牛中14头牛在肝、脾、淋巴结等肺外组织器官发现眼观结核病变而未见肺脏结核病变,仅有2头发现肺脏及肺门淋巴结病变,7头牛在全身组织器官未能发现眼观结核病变;病理组织学检查结果表明,剖检的23头牛均呈现早、中、后期典型结核结节病变,11头牛的病料分离培养到牛分枝杆菌。该奶牛场结核主要表现为肺外结核,经典肺结核较少,这可能是该奶牛场在以往扑杀的阳性牛中很少发现肺脏结核病变的原因。  相似文献

上海市某奶牛养殖专业合作社检出14头结核病阳性牛。为查明疫病发生状况,调查疫病可能的发生原因,提出针对性的防控措施,通过现场座谈、问卷调查、实地查看和档案资料查阅等方式开展了暴发调查。结果发现本次暴发调查与奶牛引进有关,历年来检测发现的结核病阳性牛和疑似牛未采取有效的隔离、淘汰处置措施,从而导致了结核病的场内循环。建议立即扑杀和无害化处理结核病阳性牛,加强环境消毒,加大结核病监测频率和力度,同时做好工作人员的结核病健康检查。  相似文献

Using PCR on aborted foetal material, Coxiella burnetii infection was confirmed as the cause of abortions in a dairy goat herd with over 1000 adults. Ninety-five (22%) abortions and 355 normal births were recorded from 440 goats over 2months. The herd was sampled three times in 6months to look at the within-herd seroprevalence, with the 1st visit done 24days after the last recorded abortion. The true seroprevalence in the herd was 79.2%, 66.5% and 45.7% on each of these visits, but introduction of a group of young goats prior to the 3d visit influenced these results. Using PCR, widespread environmental contamination was demonstrated in surface dust, bedding, muck heaps, milk, bird droppings and drinking water in the goat shed. MST and MLVA analysis showed the C. burnetii from this outbreak to be of a genotype previously observed in the UK and different from those recorded in the Netherlands outbreak of 2007-2011.  相似文献

Bovine tuberculosis is an ongoing problem in Ireland, and herd incidence has remained at approximately 5% for some years. Spillover of infection from cattle to people remains an ever-present possibility, given the ongoing pool of infection in the Irish cattle population. This paper describes an outbreak of tuberculosis affecting cattle and people on a dairy farm in southeastern Ireland following the consumption of milk from a seven-year-old cow with tuberculous mastitis. Twenty-five of 28 calves born during autumn 2004 and spring 2005 were subsequently identified as TB reactors, and five of six family members were positive on the Mantoux test. During 2005, milk from this cow had mainly been used to feed calves, and was added only occasionally to the bulk tank. Therefore, the calves each received infected milk on an almost continuous basis between birth and weaning. The family collected milk from the bulk milk tank, and consumed it without pasteurisation. This case highlights the risks associated with the consumption of raw milk. In this family, TB has had a very significant impact on the health of two young children. These risks are well recognised, and relevant information for farmers is available. It is of concern, therefore, that raw milk consumption remains prevalent on Irish farms. New strategies are needed, in partnership with industry, to address this important issue. A severe outbreak of enteric and respiratory disease associated with bovine coronavirus (BCoV) infection is described. The outbreak occurred in a dairy herd of southern Italy in the first decade of September 2006, when summer temperatures were still recorded, affecting calves, heifers and adult cows, with a marked decrease in milk production. By virus isolation and RT-PCR targeting the S gene, BCoV was identified as the etiological agent of the outbreak, whereas bacteriological, parasitological and toxicological investigations failed to detect other causes of disease. BCoV strains with 99-100% nucleotide identity in the S gene were isolated from nasal, ocular and rectal swabs, thus proving the absence of separate clusters of virus on the basis of tissue tropism. Sequence analysis of the haemagglutination-esterase and spike proteins of the strain detected in one rectal sample (339/06) showed a high genetic relatedness with recent BCoV isolates (98-99% amino acid identity), with several unique amino acid substitutions in the S protein. The BCoV outbreak described in this paper presents interesting aspects: (i) the occurrence of a severe form of disease in the warmer season; (ii) the simultaneous presence of respiratory and enteric disease; (iii) the involvement of young as well as adult cattle.  相似文献

The aim of the present study was to describe the strategies of the control of an outbreak of leptospiral infection in dairy cattle in Maranhão State, Northeastern Brazil. In the period from January to July 2015, 18 (17%) out of 106 cows presented abortion, six (5.7%) stillbirth, and 12 (11.3%) repeated estrus, totaling 24 animals with reproductive problems. The diagnosis of leptospirosis was based on serology (microscopic agglutination test—MAT), bacteriological culture, and polymerase chain reaction (PCR). Antibiotic therapy, vaccination protocols, and changes in management practices were suggested as control measures. Of all animals on the farm (n?=?280), 136 (48.6%) were seropositive for at least one serovar of Leptospira sp. No pure leptospiral culture was obtained. Eight of the animals with reproductive problems yielded positive PCR results (vaginal fluid of seven animals and urine and vaginal fluid of one animal). Genetic sequencing of a vaginal fluid/urine PCR-positive sample revealed Leptospira borgpetersenii. One year after the adoption of control measures, no reproductive problems were observed. Thus, leptospirosis probably caused the reproductive failures in the herd, and the control and prevention measures implemented were efficient in controlling the disease.

OBJECTIVE: To evaluate the use of a polymerase chain reaction (PCR) assay in detecting bovine leukosis virus (BLV) in adult dairy cows. DESIGN: Prospective study. ANIMALS: 223 adult dairy cows. PROCEDURE: Cows were tested for BLV status by use of an ELISA and a PCR assay. Sensitivity, specificity, predictive values of positive and negative tests, and the percentage of cows correctly classified by PCR assay were calculated. Ninety-five percent confidence intervals were calculated for sensitivity and specificity. RESULTS: Sensitivity and specificity were 0.672 and 1.00, respectively. Prevalence of BLV in this herd was 0.807. Predictive value of a positive test was 1.00, and predictive value of a negative test was 0.421. The percentage of cows correctly classified by PCR assay was 73.5%. CONCLUSIONS AND CLINICAL RELEVANCE: A positive PCR assay result provided definitive evidence that a cow was infected with BLV. Sensitivity and negative predictive value for PCR assay were low. Consequently, PCR assay alone is unreliable for routine detection of BLV in herds with high prevalence of the disease.  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed and was compared with the complement fixation test (CFT) in a bovine brucellosis eradication program. The ELISA detected significantly more reactors than the CFT in both strain 19 vaccinated infected herds (1.79% versus 1.14%) and non-vaccinated infected herds (4.2% versus 3.59%) but not in either vaccinated or non-vaccinated brucella-free herds. The specificity for both tests in brucella-free herds was greater than 0.998. The specificity and sensitivity of the ELISA were compared with those of 3 other tests (the Rose Bengal test; the indirect haemolysis test [IHLT] and the CFT) on serum from 151 animals cultured at slaughter. The calculated specificity of the ELISA in this infected group was lower than for both the CFT and the IHLT (0.58 versus 0.67 versus 0.75). The sensitivity however was much greater (1.0 versus 0.73 versus 0.71). The value of the ELISA when used in an eradication program is discussed.  相似文献

OBJECTIVE: To investigate the seroprevalence of Neospora caninum infection in a commercial dairy cattle herd, 15 months after detection of an abortion outbreak. PROCEDURE: Sera from the whole herd (n = 266) were examined for N caninum antibodies by indirect fluorescent antibody test (IFAT) and immunoblot analysis. Herd records were reviewed to collate serological results with abortion history, proximity to calving, and pedigree data. RESULTS: The seroprevalence of N caninum infection was 24% (63/266) for IFAT titre > or = 160, 29% (78/266) for immunoblot positive (+ve), and 31% (82/266) for IFAT > or = 160 and/or immunoblot +ve; 94% (59/63) of animals with IFAT > or = 160 were immunoblot +ve. The association between seropositivity (IFAT > or = 160 and/or immunoblot +ve) and history of abortion was highly significant (P < 0.001); the seroprevalence was 86% (18/21) in aborting cows, compared with 30% (50/164) in non-aborting animals. The abortion rate for seropositive cows was 26% (18/68) compared with 3% (3/117) for seronegative animals. IFAT titres of infected cows were higher within 2 months of calving than at other times (P < 0.001). The association between seropositivity in dams and daughters was highly significant (P = 0.009). CONCLUSIONS: The abortions were associated with N caninum infection and there was evidence of reactivation of latent infection close to calving and congenital transmission of infection. Immunodominant antigens identified by immunoblots may prove useful for improved diagnostic tests.  相似文献

The aim of the present study was to explore the prevalence and potential risk factors associated with infestation by Psoroptes spp. mites in buffalo at smallholder farms in the Nile Delta region, Egypt. A total of 318 out of 1234 buffalo in 77 small scale herds were examined. The age of buffalo varied from three months to seven years. In all, 53 (16.66%) buffalo were recorded to be infested; 51 (16.35%) with Psoroptes spp. mites and two cases (0.31%) with Chorioptes spp. mites. Multivariate logistic regression was performed for significant risk factors at univariate analysis on both animal and herd level. The results of multivariate analysis showed that, on the animal level, the prevalence was affected by animal age (P?<?0.01; OR: 0.71; CI 95%: 0.441–1.11), season (P?<?0.01; OR: 1.20; CI 95%:0.541–1.15), indoor management (P?<?0.001; OR: 6.625; CI 95%:2.489 –17.631) and rearing with other animals (P?<?0.01; OR: 2.22; CI 95%:1.340 –7.132). However, on the herd level, the prevalence was affected by indoor rearing (P?<?0.05; OR: 22.4; CI 95%:2.75–16.431), mixed rearing with other animal species (P?<?0.05; OR: 4.5; CI 95%:1.66–7.941), and season (P?<?0.01; OR: 2.3; CI 95%:0.575–2.426). Clinically, mild skin lesions with mild pruritis were significantly prevalent in buffalo with the infestation by Psoroptes spp. mites (P?<?0.001), where 33/51 cases showed mild lesions. Also, psoroptic mites had significant association with inappitence (P?<?0.001). The result of the present study indicates that Psoroptes spp. mites are the most prevalent in buffalo in the Nile Delta Region, Egypt. Moreover, recognition of risk factors associated with mange mites in buffalo may enable the practitioner to establish the most appropriate control measures.  相似文献

The incidence and direct financial loss caused by babesiosis were evaluated in 121 Holstein Friesian female cattle that formed eight cohorts (1981–1988) of a dairy farm located approximately 24° 55′S 65° 29′W in Salta, Argentina. Female calves born in 1986 and 1987 (n=32) were vaccinated with a live Babesia vaccine at six months of age.

No cases of babesiosis occurred in the vaccinated cattle. The incidence of babesiosis in the six non-vaccinated cohorts was 23.6% (21/89). Eighteen of the cases were the result of Babesia bovis, one to Babesia bigemina and two to a mixed infection. Two cattle died of B. bovis infection in spite of drug treatment (diaminazene, 3.5 mg kg−1). No disease occurred in cattle younger than seven months or older than 24 months. The number of cases according to age of cattle was: 7–9 months, 5; 10–12 months, 6; 13–24 months, 10.

Financial loss for the six cohorts that suffered clinical cases amounted to US $ 1624.6 (prices in October 1990)—62% were the result of physical losses and 38% to costs of control. A benefit-cost analysis of vaccination was carried out assuming that 95% of the mortality and morbidity losses of the six non-vaccinated cohorts was prevented after a single inculation of a live vaccine (cost of a dose plus administration was US $ 4.2. The benefit-cost ratio was 4:1 for each US dollar expended.  相似文献

Neonatal calf diarrhea is a significant health problem in dairy industry in the whole world. Although the aetiological agents of neonatal diarrhea are diverse, it is obvious that viruses play an important role. One of them is bovine coronavirus (BCV). Since BCV was discovered (MEBUS et al., 1969), there have been many published reports describing the virus and its importance in calves. In China, we first investigated the seroepidemiological status of BCV infections in cattle (YAO et al., 1990). The results showed that the incidence of infections with BCV in cattle in different chinese regions are high. Further work on direct diagnosis of BCV as a causative agent of diarrhea was therefore necessary. In the last years we have developed diagnostic methods including a monoclonal antibody-based ELISA to detect BCV and other diarrheal viruses. In this study, the diagnosis of an outbreak of BCV in chinese dairy cattle is described.  相似文献

From August to October 1991 bovine ephemeral fever (BEF) occurred sporadically in two localities in Israel. The morbidity and mortality rates reached 2.6% and 0.1%, respectively. Only 12/50 dairy cattle herds were clinically infected with BEF in the dairy community. The total morbidity rate reached 0.8%. The lowest morbidity rate was recorded in young heifers (5.5%) and the highest in adult cows (75%). Only heifers over the age of three months were clinically affected. The spread of the disease apparently followed the local prevailing night winds, which blow from east to west, i.e., from the land toward the sea. The morbidity period lasted 61 days. The low incidence and morbidity rates were possibly due to the low virulence of the virus strain involved in the 1991 epidemic. Retrospective analysis indicates that vectors - apparently mosquitoes - infected with BEF virus could have been overwintering.  相似文献

From July 1999 till November 2000, a longitudinal study was conducted on a dairy farm in The Netherlands to study within herd prevalence and types of verocytotoxin producing Escherichia coli (VTEC) of serogroup O157 over time, and determine environmental reservoirs and possible transmission routes. Faeces, blood, milk and environmental samples were collected 14 times with intervals varying from 4 to 10 weeks during the study period. Faecal samples were selectively cultured for Escherichia coli O157. Isolates were tested by PCR for the most common virulence genes, VTI, VTII and eae, and typed by pulsed field gel electrophoresis. In total, 71 isolates were obtained, of which 49 from dairy cows, 8 from young stock, 5 from other animals and 9 from the environment. Positive samples were all detected in summer and early fall. VT- and eae-genes were found in all tested isolates, except in one. DNA typing showed that three clusters of O157 isolates could be identified. One of these clusters contained samples of two shedding seasons, indicating persistence on the farm during winter and spring. Repeated measures analysis of variance showed that cows with O157 VTEC infection had higher daily milk production in the period preceding sampling (p = 0.0055). There was no significant association between the results of the LPS-ELISA on serum samples from dairy cows and their O157 status.  相似文献
