OBJECTIVE: To estimate the prevalence of caseous lymphadenitis (CLA), determine the current usage of vaccines against CLA and to measure the effectiveness of these vaccines on sheep farms. DESIGN AND POPULATION: A survey was undertaken on 223 sheep flocks in New South Wales, Victoria and Western Australia. METHOD: The prevalence of CLA was measured by conventional inspection techniques at abattoirs in lines of sheep that could be traced back to a farm. Managers of the flocks were sent a questionnaire about their vaccine practices, management practices and knowledge of CLA. RESULTS: The average prevalence of CLA in adult sheep in these flocks was 26% and varied from 20% in Western Australia to 29% in New South Wales. About 43% of sheep producers used CLA vaccines; only 12% used them as recommended. Awareness of CLA was highest in Western Australia. More producers would use CLA vaccine if they knew the prevalence of CLA in their flock and producers obtained most information about CLA from vaccine resellers. CONCLUSIONS: Only 10 to 15% of producers are currently achieving effective CLA control through the use of recommended CLA vaccination programs. In Western Australian flocks more than 25% of effectively vaccinated ewes will be sent to abattoirs in the 2 to 3 years after this study. However, large decreases in the prevalence of CLA can be achieved by about 70% of producers by either making adjustments to their vaccination programs or buying a vaccine with a CLA component. Two or three key facts on effective CLA vaccination could be made available at the point of sale of vaccines and from abattoirs that reported the prevalence of CLA to farmers. 相似文献

Enzyme-linked immunosorbent assays (ELISA) with Corynebacterium pseudotuberculosis cell wall and bacteria-free supernatant with exotoxin preparations as antigens, and hemolysis inhibition tests were used to detect antibodies in the sera of adult range sheep with naturally acquired caseous lymphadenitis (CL). The extent and severity of lesions were quantitated on the basis of a lesion score, derived from an examination of the carcass (peripheral lymphoid tissue) and viscera (including internal lymphoid tissue) at the time of slaughter. The overall prevalence of C pseudotuberculosis-positive CL lesions in 104 sheep was 31.7%. The cell wall ELISA detected antibodies in 96.9% (32/33) of sheep with C pseudotuberculosis-positive CL lesions. The exotoxin ELISA detected antibodies in 84.8% (28/33) of positive sheep in the same group. Both ELISA resulted in a high number of apparent false-positives, with 64.7% and 49.2%, respectively, positive optical density (OD) values in sheep with no gross CL lesions and no apparent C pseudotuberculosis infection. There was no significant relationship between the extent of lesion development (lesion score) and OD values in both cell wall (r = 0.472) and exotoxin (r = 0.464) ELISA. Similarly, there was no significant relationship between the titer of antitoxin antibodies, as measured by the hemolysis inhibition test, and the extent of disease. These investigations indicate that those ELISA that use crude C pseudotuberculosis antigens are of questionable utility in the field, where C pseudotuberculosis infection is endemic in many sheep populations. Furthermore, these studies suggest that antibodies that are reactive with components of C pseudotuberculosis and that develop in response to infection may have little impact on the recovery of the host. 相似文献

A total of 8,711 sheep in 3 distinct age/sex groups were examined to determine for occurrence and extent of caseous lymphadenitis after slaughter at a major Western Australian abattoir. An adaptation of current mean inspection procedures to provide a numerical weighted evaluation according to number, site, and size of lesions was used to determine extent of involvement. Frequency in slaughter populations was estimated to be 3.4 +/- 0.6% for lambs, 41.8 +/- 3.9% for mixed-age rams, and 53.7 +/- 1.5% for adult ewes at 95% confidence intervals. The results confirmed that frequency increased with age, but also revealed increases in extent of involvement and occurrence of visceral lesions, particularly in association with lesions of the body. Nevertheless, lesions occurred more frequently in the body than in the viscera. However, the mean visceral involvement was greater than mean body involvement in all groups, and the ratio was found to be more or less constant. Visceral involvement was found to be more extensive in rams than in ewes. Difficulties associated with complexity of current inspection procedures were also recorded. The relevance of these findings to pathogenesis and postmortem disposition after slaughter is discussed. It is suggested that current postmortem inspection criteria should be reevaluated. 相似文献

Constituents of Corynebacterium pseudotuberculosis were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Analysis of sonicated whole bacterial cells and ether-extracted cells revealed more than 35 bands in silver-stained gels. SDS-PAGE analysis of concentrated culture filtrates with exotoxin activity demonstrated more than 15 bands. Sera from sheep with C. pseudotuberculosis-induced disease of variable severity were used to probe immunoblots of electrophoresed ether-extracted cells and culture filtrates. Twenty or more corynebacterial molecules, ranging in molecular weight from 20 to 112 kDa, in ether-extracted cells were recognized by antibodies in the sera of naturally exposed sheep with positive ELISA titers. These sera also recognized up to six molecules, ranging from, 20 to 68.1 kDa, on immunoblots of ammonium sulfate-concentrated culture filtrate. There was no apparent relationship between the stage of disease and the response to specific corynebacterial antigens in these animals. 相似文献

Pregnant ewes were vaccinated twice, seven weeks and three weeks before lambing, with a multivalent formalin-killed Escherichia coli vaccine containing an added K99, F41 antigen preparation. Lambs born to vaccinated and unvaccinated ewes were exposed to oral infection with E coli B44 (09:K30, K99, F41). All 10 lambs from vaccinated ewes were protected whereas all 10 control lambs developed severe diarrhoea and five died or were killed in extremis. In the following year, previously immunised ewes were given a single dose of the vaccine two weeks before lambing. Eleven of their 12 lambs were protected against a similar challenge, which caused the death of six of eight control lambs and severe diarrhoea in the two survivors. Higher levels of antibody to the K99, F41 preparation were detected by enzyme-linked immunosorbent assay in the serum and colostrum from vaccinated ewes and in the serum of their lambs when compared with similar samples from control ewes and lambs. 相似文献

Pregnant ewes were vaccinated 1 month before parturition with mono or bivalent adenovirus vaccines. Vaccination resulted in increased levels of homologous and heterologous antibody in ewes, with corresponding increases in passive immunity of lambs. Challenge of lambs with homologous or heterologous virus at 21 days of age was associated with significant resistance to development of lesions in lambs challenged with homologous virus, and partial resistance in those challenged with heterologous virus. The progress of caseous lymphadenitis in sheep and goats is described as involving primary wound infection, lymphatic and haematogenous dissemination, and secondary infection of lymph nodes and various visceral organs. This is followed by elimination or containment of infection, the latter being represented by the characteristic caseous lesions. Resistance to infection is complex involving components of both the specific and non-specific host response. The role of these in pathogenesis is discussed. Bacterial factors including the phospholipase exotoxin and cytotoxic lipid contribute to pathogenesis at a local level with little likelihood of systemic effect in the natural disease. In a disease such as caseous lymphadenitis, lesion site, size or number do not indicate severity of disease but rather, reflect the ability of the host to mount a cellular response to infection. 相似文献

Two hundred Merino wether hoggets were used to examine the effect of Corynebacterium pseudotuberculosis infection (caseous lymphadenitis) on wool production and bodyweight. Sheep which were challenged with C. pseudotuberculosis (artificially infected) and not vaccinated against this disease produced 0.20 kg less clean wool than unchallenged controls during the following 12 months. The incidence of sheep with lesions in the group that was vaccinated prior to challenge was 55% lower than in unvaccinated challenged sheep but their wool production was not significantly different from either the controls or the unvaccinated challenged sheep. Vaccinated sheep were also heavier than unvaccinated sheep 12 months after challenge. These results indicate that caseous lymphadenitis infection may reduce wool production. 相似文献

In Alberta, caseous lymphadenitis (CLA) is one of the leading causes of lamb and mutton carcass condemnation. In this study, serologic results confirmed a high (50-94%) incidence of exposure to Corynebacterium pseudotuberculosis, the causative agent of CLA, in mature, unvaccinated sheep in southern Alberta. To assess the efficacy and impact of vaccination with 2 commercial (Glanvac-6 and Case-Vac) and 1 experimental (WC+ MDP-GDP) CLA vaccines, a series of 3 field trials in 3249 ewes and lambs was conducted in affected flocks from 1992-1996. Efficacy was assessed from the serological response to vaccination, prevalence and size of injection site reactions by treatment, and the incidence of CLA abscesses. Overall, agglutinating antibody titres to C. pseudotuberculosis in lambs vaccinated with WC+MDP-GDP and Case-Vac remained significantly elevated above nonvaccinated control lambs for the 12 mo period after the initial vaccination. Lambs vaccinated with the WC/MDP-GDP maintained higher titres (P < 0.06) than those vaccinated with Case-Vac for the period from 6 to 12 mo after vaccination. Agglutinating antibody titres for lambs vaccinated with Glanvac did not differ from those of controls at any point during the 12 mo period after vaccination. The number of injection site reactions was elevated in lambs vaccinated with Glanvac as compared to those vaccinated with WC+MDP-GDP but the size of injection site reactions did not significantly differ. Sheep vaccinated with WC+ MDP-GDP also had a reduced incidence of putative CLA abscesses, although confirmation of the presence of C. pseudotuberculosis was only successful in a small number of instances. 相似文献

A total of 2,920 feral goats from pastoral areas of Western Australia were examined and the prevalence of caseous lymphadenitis was estimated to be 7.8 +/- 0.9% in goats presented for slaughter. Head, body and visceral lesions were present in 49.3%, 46.7% and 12.3% of affected goats, respectively. A comparison with previously reported data from sheep has been made and significant differences are discussed. Lung lesions were relatively uncommon in goats, whereas the distribution of carcase lesions was similar between the species. Multiple lesions appear to be less common in goats than in sheep. 相似文献

Numbers of lymphoid cells bearing markers for peanut agglutinin, wheat germ agglutinin, and anti-immunoglobulin G in healthy goats and in goats with caseous lymphadenitis were determined by use of immunofluorescent techniques. Results indicated that, when compared with healthy goats, diseased goats had normal numbers of anti-immunoglobulin G-positive cells (B cells), had significantly reduced numbers of peanut agglutinin-positive cells (T cells), and had fewer cells with receptor sites for Lens culinaris agglutinin and concanavalin A. Compared with healthy goats, numbers of peanut agglutinin-positive cells in goats with caseous lymphadenitis indicated that the goats may have had a compromised cell-mediated immunity. 相似文献

This study was carried out of 4,419 animals raised in an extensive system on local breed sheep of the Tiaret region during 2 years (2010–2011) to determine some clinical and pathological aspects associated to caseous lymphadenitis (CL). Clinical examination revealed a prevalence rate of 5.7 %. CL prevalence rate was significantly higher in females (6 %) than in males (4.8 %). The most affected superficial lymph nodes were mandibular lymph nodes (46.1 %). Gross examination of 1,679 carcasses of sheep at the abattoir revealed the presence of abscesses on 250 carcasses (14.9 %). Out of 105 cases showing lesions of CL, Corynebacterium pseudotuberculosis (53.6 %) was isolated as mixed infection with Staphylococcus aureus (43 %) and Escherechia coli (17.9 %). CL prevalence was significantly higher in adult than in young sheep and in female than in male ones. Visceral form of CL was associated to “thin ewe syndrome” responsible for significant economic losses in Algeria. 相似文献

A total of 876 sheep from five flocks in north Jordan were selected to study the effect of shearing on the incidence of caseous lymphadenitis (CLA). The animals were divided into two age groups, sheep aged 1-2 years and those aged > or = 3 years. Blood samples were collected from the animals at the time of shearing and again 6 months later. A toxin enzyme-linked immunosorbent assay was used to identify sheep that had been infected with Corynebacterium pseudotuberculosis. The point prevalences of CLA were 6.59% and 21.06% in the 1-2-year and > or = 3-year age groups, respectively, and were significantly higher (P < 0.01) in the > or = 3-year age group. The overall prevalence among all ages was 15.3%. In the shorn sheep, the incidence of CLA was 22.46% and 9.47% in the 1-2-year and > or = 3-year age groups, respectively, and was significantly higher (P < 0.05) in the 1-2-year age group. In the control animals, the incidence was 8% and 5.26% in the 1-2-year and > or = 3-year age groups, respectively, and was different (P < 0.01) between the shorn (22.46%) and control (8%) animals of the 1-2-year age group. An epidemiological survey of 35 sheep farms revealed the prevalence of CLA, shearing wounds and unhygienic conditions during shearing in all farms. In conclusion, the prevalence of CLA increases with age and the incidence increases only in young sheep after shearing. Sheep are sheared under unhygienic conditions, which may be a contributing factor in increasing both the prevalence and the incidence of CLA. 相似文献

AIM: To determine whether short-term progesterone supplementation post mating or shearing of ewes in early pregnancy affected either the proportion of ewes that lambed or that had multiple lambs, or the birthweight of lambs. METHODS: Romney ewes (n=457) were synchronised in oestrus using controlled internal drug-releasing (CIDR) devices containing progesterone, and mated to Romney rams over a 5-day period. The mid-point of mating (Day 0) occurred 2 days after the withdrawal of CIDR devices. Ewes mated (n=397) were randomly allocated to one of four treatment groups: shearing at Day 5, shearing at Day 30, no shearing, and no shearing plus progesterone supplementation using a CIDR device inserted on Day 3 for 6 days. During the period from Day 5 to Day 27, six harnessed Suffolk rams were placed with the ewes and matings recorded. At Day 48, all ewes that did not return to the Suffolk rams were scanned for pregnancy using ultrasound. At Day 140, single- and multiple-bearing ewes were set-stocked at 15.1 and 12.2 ewes/ha, respectively, and equivalent numbers of ewes from each treatment group were placed in each paddock. Blood samples from 10 unshorn and 10 progesterone-supplemented ewes were collected on Days 3, 6 and 9, and analysed for plasma progesterone concentrations. Lambs were identified to their dam and weighed within 12 h of birth, and again at 27 and 93 days after the mid-point of lambing. The ewes were weighed at regular intervals throughout the trial. RESULTS: Plasma progesterone concentrations of supplemented ewes were higher than those of unsupplemented ewes (3.28 vs 1.75 ng/ml) on Day 6 (p=0.02) but not on Day 9 (4.58 vs 4.63 ng/ml). Treatment of ewes had no effect on either the proportion of ewes which lambed to the synchronised mating period or that had multiple lambs. Lambs born to ewes shorn at Day 30 tended (p=0.09) to be heavier at birth (by 0.28 kg) than those born to unshorn ewes but this effect was not evident when data were corrected for length of gestation. Neither shearing at Day 5 nor progesterone supplementation had any effect on the birthweight of lambs, and the treatment of ewes had no effect on the survival rate of lambs to weaning. CONCLUSIONS: Progesterone supplementation for 6 days beginning 3 days post mating did not increase either the proportion of ewes that lambed or that had multiple lambs, or the birthweight of lambs. Shearing 5 days after mating had no significant effect on the reproductive performance of ewes and need not be avoided, but is unlikely to result in an increase in lamb birthweight. Shearing ewes at Day 30 may result in an increase in the birthweight of lambs but, ideally, ewes should be further advanced in pregnancy before shearing is undertaken. 相似文献
