We evaluated the hypoglycemic and antioxidant effects of the total alkaloids of leaves and twigs of Catharanthus roseus Linn.(CTA) in streptozotocin(STZ)-induced diabetic rats. The hypoglycemic effect was measured by blood glucose and plasma insulin level. Oxidative stress was measured in heart, liver and kidney by levels of antioxidant markers, free radical scavengers and lipid peroxides i.e. superoxide dismutase(SOD), catalase(CAT), glutathione(GSH) and thiobarbituric acid reactive substances(TBARS). Biochemical parameters, i.e. aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphate(ALP) were observed in diabetic control and treated rats. Oral administration of CTA for30 days was followed by a significant(P \ 0.05) decrease in fasting blood glucose and increase in insulin level as compared with untreated diabetic rats. Also it significantly(P \ 0.05) reduced ALT, AST and ALP. The treatment also resulted in significant(P \ 0.05) reductions in GSH,SOD, CAT, and decrease in TBARS in the heart, liver and kidney of diabetic rats. The results suggest that CTA can effectively normalize the impaired antioxidant status in STZ-induced diabetes in a dose-dependent manner.CTA exerted rapid protective effects against lipid peroxidation by scavenging of free radicals and reducing the risk of diabetic complications.  相似文献

To explore the physiological and biochemical mechanism of the occurrence of vitrified shoots of Populus suaveolens in tissue culture, the changes in water, chlorphyll, lignin, H2O2, phenylalanine ammonialyase (PAL), malonaldehyde (MDA), protective enzymatic systems, and some key enzymes involved in the ascorbate- glutathione cycle were comparatively studied in both normal and vitrified shoots of P. suaveolens. The results show that the lower activities of peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), glutathione reductase (GR) and PAL, and the less contents of chlorphyll, lignin, ascorbate (ASA) and reduced glutathione (GSH) as well as the lower ratios of ASA / DHA and GSH / GSSG are observed in vitrified shoots than in normal ones during the whole culture period. While in comparison with normal shoots, the higher activity of superoxide dismutase (SOD) and the more concentrations of water, H2O2, MDA, dehydroascorbate (DHA) and oxidized glutathione (GSSG) are found in vitrified shoots. Statistical analysis indicates that the enhanced activity of SOD and the decreased activities of CAT and POD as well as some enzymes involved in the ascorbate-glutathione cycle might be closely correlated to the accumulation of H2O2. The less regeneration of ASA and GSH and the lower capacity of the ascorbate-glutathione cycle observed in vitrified shoots might be due to a significant decrease in APX, MDAR, DHAR and GR activities and a decline in redox status of ASA and GSH. The decreases in chlorphyll content might result in a decline in photosynthesis. The lower activities of POD and PAL could result in the decrease of lignin synthesis and cell wall ligination, which might be the key factor leading to the increase in water content. It is concluded that the deficiency of detoxification capacity caused by the lower capacity of the ascorbate-glutathione pathway and the decreased activity of protective enzymatic system might lead to the large accumulation of H2O2 and the enhancement of membrane lipid peroxidation, which might be the main cause leading to the occurrence of vitrifying shoots of P. suaveolens in tissue culture. We investigated the changes in the contents of H2O2, malonaldehyde (MDA) and endogenous antioxidants, the activities of protective enzymes and some critical enzymes involved in the ascorbate-glutathione (ASA-GSH) cycle as well as freezing resistance (expressed as LT50) and correlations mentioned above, in detail using Populus suaveolens cuttings. The purpose was to explore the physiological mechanism of the enhancement of freezing resistance induced by freezing acclimation at –20°C, and to elucidate the physiological mechanisms by which trees adapt to freezing. The results showed that freezing acclimation enhanced the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), monodehydroascorbate reductase (MDAR), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR). And it increased the contents of reduced ascorbate (ASA), reduced glutathione (GSH), dehydroascorbate (DHA) and oxidized glutathione (GSSG). However, H2O2 and MDA contents and LT50 of cuttings were decreased. LT50 in cuttings was found to be closely correlated to the levels of SOD, POD, CAT, APX, DHAR, MDAR, GR, H2O2, MDA, ASA, GSH, DHA and GSSG during freezing acclimation. This suggested that the enhancement of freezing resistance of cuttings induced by freezing acclimation may relate to the distinct increase for the levels of SOD, POD, CAT, APX, DHAR, MDAR, GR, ASA, GSH, DHA, and GSSG. In addition, the observed levels of APX, DHAR, MDAR, GR, ASA, DHA, GSH and GSSG were higher than those of SOD, POD and CAT during freezing acclimation. It indicated that a higher capacity of the ASA-GSH cycle is required for H2O2 detoxification, and growth and development of cuttings. Based on the obtained results, it can be concluded that the ASA-GSH cycle plays an important role in enhancement of freezing resistance of P. suaveolens cuttings during freezing acclimation.  相似文献

The hexane extract of Phyllanthus maderaspatensis (200 and 100 mg/kg) showed significant hepatoprotection on carbon tetrachloride and thioacetamide induced liver damage in rats. The protective effect was evident from serum biochemical parameters and histopathological analysis. Rats treated with P. maderaspatensis remarkably prevented the elevation of serum AST, ALT and LDH and liver lipid peroxides in CCl(4) and thioacetamide treated rats. Hepatic glutathione levels significantly increased by the treatment with the extracts. Histopathological changes induced by CCl(4) and thioacetamide were also significantly reduced by the extract treatment. The activity of hexane extracts of P. maderaspatensis was comparable to that of silymarin, the reference hepatoprotective drug.  相似文献

Stigmasterol, isolated from the bark of Butea monosperma was evaluated for its thyroid hormone and glucose regulatory efficacy in mice. Its administration at 2.6 mg/kg/d for 20 days reduced serum triiodothyronine (T3), thyroxin (T4) and glucose concentrations as well as the activity of hepatic glucose-6-phophatase (G-6-Pase) with a concomitant increase in insulin indicating its thyroid inhibiting and hypoglycemic properties. A decrease in the hepatic lipid peroxidation (LPO) and an increase in the activities of catalase (CAT), superoxide dismutase (SOD) and glutathione (GSH) suggested its antioxidative potential. The highest concentration tested (5.2 mg/kg) evoked pro-oxidative activity.  相似文献

Spirulina fusiformis given by oral route to mice at doses of 250, 500 and 1000 mg kg(-1) significantly inhibit the genotoxicity induced by cisplatin and urethane. In addition, a significant reduction in the extent of lipid peroxidation with concomitant increase in the liver enzymatic (GPx, GST, SOD, CAT) and non-enzymatic (reduced glutathione) antioxidants were observed.  相似文献

The ethanolic extract of Clerodendrum inerme leaves were screened for its hepatoprotective activity in CCl(4) (0.5 ml/kg, i.p) induced liver damage in Swiss albino rats at a dose of 200 mg /kg bw. The ethanolic extract of C. inerme significantly (P<0.001) decreases the serum enzyme alanine amino transferase (ALT), asparate amino transferase (AST), alkaline phosphates (ALP), triglycerides (TGL), total cholesterol (TC) and significantly increased the glutathione level. Silymarin (25 mg/kg), a known hepatoprotective drug used for comparison exhibited significant activity (P<0.001). The extract did not shown any mortality up to a dose of 2000 g/kg bw.  相似文献

The hepatoprotective effects of polyprenols from Ginkgo biloba L. leaves were evaluated against carbon tetrachloride induced hepatic damage in Sprague-Dawley rats. The elevated levels of serum ALT, AST, ALP, ALB, TP, HA, LN, TG, and CHO were restored towards normalization significantly by GBP in a dose dependent manner. The biochemical observations were supplemented with histopathological examination of rat liver sections. Meanwhile, GBP also produced a significant and dose-dependent reversal of CCl4-diminished activity of the antioxidant enzymes and reduced CCl4-elevated level of MDA. In general, the effects of GBP were not significantly different from those of the standard drug Essentiale.  相似文献

The present study was undertaken to examine the inhibitory effect of garlic (Allium sativum) on 4-nitroquinoline 1-oxide (4NQO)-induced tongue carcinogenesis in male rats, both in the initiation and post-initiation phases. Lipid peroxidation, reduced glutathione (GSH), glutathione peroxidase (GPx), and glutathione S-transferase (GST) were used to monitor the chemopreventive potential of garlic. Biochemical estimations were carried out on tumour and normal tongue tissues. Diminished lipid peroxidation in the tumour tissue was accompanied by a significant increase in the levels of GSH, GPx and GST. Administration of garlic (250 mg/kg, p.o., three times a week) effectively suppressed 4NQO-induced tongue carcinogenesis as revealed by the absence of carcinomas in the initiation phase and their reduced incidence in the post-initiation phase. The results of the present study suggest that garlic may exert its chemopreventive effects by modulating lipid peroxidation and enhancing the levels of GSH, GPx and GST.  相似文献

本研究报道沙棘(胡颓子科)对雄性白化病白鼠由铬诱导氧化胁迫的抗氧化活性。氧化胁迫通过对小白鼠进行强制性喂养30 d,以浓度为30 m g/kg的铬相对体重比例的重铬酸钾盐实现。铬促使体重下降,而却明显增加了器官与体重比例。铬处理明显减少了谷胱甘肽的降低,增加了丙二醛和肌氨酸磷酸激酶的水平;而且它还加强了血清中谷氨酸草酰乙酸转移酶和谷氨酸丙酮酸转移酶的浓度。用不同剂量的沙棘叶子提取物(乙醇提取)对保护铬元素诱导的氧化胁迫进行了评估,结果表明叶子提取物在浓度为100到250 m g/kg铬与体重比的情况下可以明显保护动物避免由铬所诱导的氧化伤害。  相似文献

The chemopreventive efficacy of lycopene on 7,12-dimethylbenz[a]anthracene (DMBA)-induced hamster buccal pouch (HBP) carcinogenesis was examined using lipid peroxidation, reduced glutathione (GSH), glutathione peroxidase (GPx), glutathione S-transferase (GST) and glutathione reductase (GR) as biomarkers of chemoprevention. Twenty four male Syrian hamsters were divided into four groups of six animals each. The right buccal pouches of the animals in group 1 were painted with a 0.5% solution of DMBA in liquid paraffin three times a week. The animals in group 2 were painted with DMBA as in group 1 and in addition received 2.5 mg/kg body weight lycopene orally three times a week on days alternate to DMBA application. Group 3 animals received lycopene as in group 2. Animals in group 4 received neither DMBA nor lycopene and served as control. The hamsters were killed after an experimental period of 14 weeks. Biochemical measurements were carried out in tumour and normal tissues. All hamsters painted with DMBA alone for 14 weeks developed well-differentiated squamous cell carcinomas. Diminished lipid peroxidation in the oral tumour tissue was accompanied by a significant increase in the levels of GSH, GPx, GST and GR. Administration of lycopene significantly suppressed DMBA-induced oral carcinogenesis as revealed by the absence of carcinomas. The results of the present study suggest that lycopene may exert its chemopreventive effects by modulating lipid peroxidation and enhancing the activities of the enzymes in the glutathione redox cycle.  相似文献

The suspensions of chloroform extract of leaves in 0.3% carboxy methyl cellulose (CMC) was evaluated for hepatoprotective activity in Wistar albino rats by inducing hepatic injury with d-galactosamine (400 mg/kg). The chloroform extract of Polygala arvensis at an oral dose of 200 mg/kg and 400 mg/kg exhibited a significant (P<0.001, P<0.01 and P<0.05) protection effect by normalizing the levels of aspartate amino transferase (ASAT, GOT), alanine amino transferase (ALAT, GPT), alkaline phosphatase (ALP), total bilirubin (TB), lactate dehydrogenase (LDH), total cholesterol (TC), triglycerides (TGL), albumin, total protein (TP) which were significantly (P<0.001) increased in rats by treatment with 400 mg/kg i.p. of d-galactosamine. The effects of lycopene on blood oxidant-antioxidant balance during N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced gastric carcinogenesis in the presence of saturated sodium chloride (S-NaCl) as promoting agent were investigated. Enhanced lipid peroxidation in the blood of tumour-bearing animals was accompanied by significant decreases in the levels of reduced glutathione (GSH), ascorbic acid and vitamin E and the activities of glutathione peroxidase (GPx), glutathione-S-transferase (GST) and glutathione reductase (GR). Administration of lycopene significantly lowered the concentrations of lipid peroxides and enhanced antioxidant levels. We suggest that the modulatory effects of lycopene on the blood oxidant-antioxidant balance may be responsible for its chemopreventive potential.  相似文献

[目的]研究菲胁迫下活性氧和抗氧化物质的变化规律,探究蒿柳抗氧化系统的防御机制,为提高其对多环芳烃(PAHs)的抗性及加强植物修复的研究提供理论依据。[方法]以蒿柳扦插苗为试验材料,采用水培方式,研究其在0、1.0 mg·L^-1菲处理下活性氧、抗氧化酶、抗氧化剂以及丙二醛(MDA)的动态变化,处理时间为16 d。[结果]研究表明:(1)菲处理后第4天,H2O2含量和氧自由基(O2^·-)生成速率迅速增加,MDA含量升高,过氧化氢酶(CAT)活性显著上升;第8天超氧化物歧化酶(SOD)和过氧化物酶(POD)活性显著升高;第16天时,H2O2含量下降到与对照无显著差异,O2^·-和MDA的增加量下降。(2)还原型谷胱甘肽(GSH)和谷胱甘肽还原酶(GR)在处理后第4天即迅速上升,谷胱甘肽-S-转移酶(GST)呈缓慢上升趋势。(3)还原型抗坏血酸(AsA)含量在处理后第4天低于对照,但随着处理时间的延长呈上升趋势,在第16天时高于对照。[结论]菲胁迫下,O2^·-是造成细胞膜脂过氧化的主要活性氧,SOD活性一直高于对照,但不足以清除增加的O2^·-,CAT和POD的升高可以清除过量的H2O2;GSH是抵御菲胁迫的有效抗氧化剂,并通过GST的催化参与菲的解毒。  相似文献

A herbal preparation “Compound Codyceps-TCM-700C (CC-700C)” was tested for hepatoprotective effect against the carbon tetrachloride induced liver damages in Sprague-Dawley rat model for a period of 6-weeks. Two dosage levels of CC-700C, respectively 286.2 mg/kg-bw (L-TCM) and 2862 mg/kg-bw (H-TCM), and a positive control Silymarin (Sigma) were used to compare their therapeutic effect. Both CC-700C's and Silymarin showed nontoxic in nature, as evidenced by body weight gain, organ weights and appearance including liver, spleen, and kidney. The activities of aspartate aminotransferase (AST) and alanine-aminotransferase (ALT) were more effectively suppressed by CC-700C than Silymarin. In addition, all levels of serum bilirubin, serum albumin, triglyceride (TG), total cholesterol (TC), platelet count (PLT), and prothombin time (PT) except TG were shown effectively restored to normal values by CC-700C and Silymarin. Moreover, although levels of glutathione (GSH), glutathione reductase (GSH-Rd), and superoxide dismutase (SOD) were equally maintained by these three preparations, glutathione peroxidase (GSH-Px) was suppressed only by H-TCM, and SOD only by Silymarin. In contrast, the activity of catalase efficiently recovered to control level on administration of CC-700C, being far better than Silymarin. Finally the liver collagen content, an indication of fibrosis, was also significantly suppressed by CC-700C, better effect was by L-TCM, but both levels were superior to Silymarin. Conclusively, the herbal preparation “Compound Cordyceps TCM-700C” is a potent hepatoprotective preparation. For therapeutic use, a dosage of 286.2 mg/kg-bw would be sufficiently effective.  相似文献

Lupeol, a pentacyclic triterpene and its ester derivative, lupeol linoleate, were investigated for their possible hepatoprotective effect against cadmium-induced toxicity in rats. Cadmium intoxicated rats showed elevated levels of malondialdehyde (basal and induced), and decreased levels of antioxidants and antioxidising enzymes in the liver. The oral administration of triterpenes (150 mg/kg, once a day for 3 days before injection of cadmium chloride) changed the tissue redox system by scavenging the free radicals and by improving the antioxidant status of the liver. Lupeol linoleate had a better effect on the antioxidant status of the liver, when compared to lupeol.  相似文献

The cytoprotective effect of piperine on benzo[a]pyrene (B[a]P) induced experimental lung cancer was investigated in male Swiss albino mice. Oral administration of piperine (100 mg/kg body wt.) effectively suppressed lung cancer initiated with B[a]P as revealed by the decrease in the extent of lipid peroxidation with concomitant increase in the activities of enzymatic antioxidants (superoxide dismutase, catalase and glutathione peroxidase) and non-enzymatic antioxidant (reduced glutathione, vitamin E and vitamin C) levels when compared to lung cancer bearing animals. Our data suggest that piperine may extend its chemopreventive effect by modulating lipid peroxidation and augmenting antioxidant defense system.  相似文献
