SUMMARY: Common carp Cyprinus carpio were administered intraperitoneally with 10 μmol epinephrine per 100 g bodyweight, and enzyme activities and metabolic intermediate concentrations were determined in the hepatopancreas and muscle. Glycogen phosphorylase a (GPase a) activity together with cyclic AMP (cAMP) concentration was increased, and glycogen content was decreased in the hepatopancreas and muscle at 2 h after the administration. The epinephrine administration also increased hepatopancreatic glucose-6-phosphatase and fructose-1,6-biphosphatase activities as well as serum glucose, lactate, and free amino acid concentrations. Furthermore, its administration increased phosphofructokinase activity together with lactate, fructose-6-phosphate, adenosine-5'-monophosphate, and adenosine-5'-diphosphate concentrations and decreased citrate concentration in the muscle. Thereafter, almost all parameter concentrations in the hepatopancreas and muscle recovered to the pre-administered levels during 24 h after the administration. These results suggest that epinephrine administration enhanced glycogenolysis and gluconeogenesis in the hepatopancreas, and released glucose into the bloodstream to supply it to the muscle. The blood glucose together with muscle glycogen seems to be metabolized through enhanced glycolysis in the muscle.  相似文献

This study was conducted to investigate the effect of dietary phosphorus on the intestine and hepatopancreas antioxidant capacity of juvenile Jian carp (Cyprinus carpio var. Jian). Jian carp, with an average initial weight of 7.17 ± 0.01 g, were fed with diets containing graded concentrations of available phosphorus, namely 1.7 (control), 3.6, 5.5, 7.3, 9.2 and 11.0 g kg?1 diet for 9 weeks. Results showed that, in intestine and hepatopancreas, content of malondialdehyde (MDA), protein carbonyl (PC) and glutathione (GSH), capacity of anti‐superoxide anion (ASA) and anti‐hydroxyl radical (AHR), and glutathione reductase (GR), catalase (CAT), glutathione S‐transferase (GST), superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities were significantly affected by dietary phosphorus levels (P < 0.05). Regression analysis showed that significant quadratic responses occurred in MDA content and ASA, GST, GPx and AHR activities in intestine, GSH content and CAT and SOD activities in hepatopancreas (P < 0.05). These results indicate that optimal level of dietary phosphorus prevented oxidative damage and increased antioxidant enzyme activities in the intestine and hepatopancreas of juvenile Jian carp. The phosphorus requirement estimated from MDA using quadratic regression analysis was 5.7 g kg?1 diet.  相似文献

草鱼样品体重(1543±213)g。采用离体消化培养和茚三酮比色方法,以草鱼前肠、中肠、后肠和肝胰脏的粗酶液作为酶源,对鱼粉、豆粕、菜粕和棉粕进行酶解,并测定酶解14h内,豆粕酶解液中氨基酸生成量随时间的变化,以及0至4h,内4种饲料蛋白质酶解液中氨基酸的生成量。结果表明,(1)在0至4h内,4种饲料酶解液OD570随时间变化的线性关系较好。(2)对于同种蛋白质饲料原料,以中肠组织粗酶液的酶解速度最大,其余依次为前肠、后肠及肝胰脏粗酶液。表明草鱼中肠对饲料蛋白质的酶解能力强于前肠和后肠,同时,肠道酶液的酶解速度均大于肝胰脏。(3)对于4种不同的蛋白质饲料原料,以鱼粉酶解氨基酸生成速度最大,其他依次为豆粕、菜粕、棉粕。(4)草鱼肝胰脏粗酶液对豆粕的酶解氨基酸生成速度最大,表明草鱼对豆粕进行酶解消化的能力较强。实验证实,饲料原料种类的差异和饲料蛋白质组成与性质的差异使其酶解氨基酸生成量和生成速度有差异。  相似文献

采用膨化加工的家禽下脚粉替代不同比例鱼粉配制四组试验饲料,分别饲喂同批孵育的幼鲤(平均体长5.45±0.78cm,体重1.38±0.21g)76天,水温22~25℃,日投喂量占鱼体重5%。实验结束时各组受试鱼肝胰脏、肠道蛋白酶活性和生长测定结果表明:不同试验饲料对幼鲤肝胰脏、肠道蛋白酶活性有一定影响;添加家禽下脚粉各组明显高于鱼粉(对照)组P<0.05);幼鲤生长和蛋白质效率与蛋白酶活性显示正相关。  相似文献

The influence of ontogeny and nutrition on metabolic enzyme activities in larvae of the African catfish, Clarias gariepinus, was studied. After start of exogenous feeding, the larvae were reared for 10 days under three different nutritional conditions: Artemia nauplii, a dry starter diet, and starvation. The live feed gave the best growth (96 mg within 10 days) whereas the dry diet resulted in low growth (33 mg). This growth difference was reflected in larval RNA and DNA concentrations, but not in the levels of soluble protein. Enzymes representing the following aspects of metabolism have been analysed: NADPH generation (G6PDH, ME), glycolysis (PFK, PK), gluconeogenesis (FDPase), amino acid catabolism (GOT, GPT) and oxidative catabolism (CS). All enzymes were present from the start of exogenous feeding onwards, but their maximum specific activities displayed different developmental patterns. In catfish larvae fed on Artemia, G6PDH and ME activities steadily increased with age and weight of the larvae. CS levels remained, after an immediate enhancement upon onset of exogenous feeding, on a rather stable plateau. The amino acid-degrading enzymes GOT and GPT showed maximum levels at days 3–5 of feeding or at a body weight of 10–20 mg, but decreased thereafter. Activities of PFK, PK and FDPase showed low initial levels, and increased significantly with age and size. Based on the ontogenetic patterns of metabolic enzymes, in C. gariepinus larvae an early and a late developmental phase can be distinguished. During the early phase, the glycolytic and gluconeogenetic capacities are low, whereas they are enforced during the later phase. The oxidative capacity is high both during the early and the late phase. The metabolic changes in catfish development coincide with other major ontogenetic events, e.g., alterations of muscle organization, gill morphology, respiration and stomach structure and function. Rearing catfish larvae on a dry diet instead of Artemia partly altered the developmental pattern described: The ontogenetic elevation of CS, PFK and FDPase was delayed and the early peak in GOT and GPT activities was not realized. The present study evaluated the effect of dietary polysaccharide from fermented wheat bran on growth performance, muscle composition, digestive enzyme activities and intestinal microbiota of juvenile common carp (Cyprinus carpio L.). Four diets supplemented with 0 (D0), 1.0 (D1), 2.0 (D2) and 4.0 (D3) g/kg fermented wheat bran polysaccharide (FWBP) were prepared. Fish were randomly assigned into four groups, each group having four replicates (30 fish/replicate) and fed with experimental diets for 8 weeks. Compared with the D0 group, the D3 group had higher final body weight and weight gain (p < .05). The amylase activity in the intestine and hepatopancreas of D1 and D2 groups and pepsin and trypsin activities in the hepatopancreas of D2 and D3 groups were significantly higher than D0 group. Significantly increased muscle protein and lipid content while reduced moisture content was observed in D1 group; additionally, D3 group had significantly lower muscle lipid content. The population of Bifidobacterium spp. in the intestine digesta of D1 and D2 groups was significantly increased, whereas lactic acid bacteria (LAB) and Escherichia coli were unaffected. Collectively, dietary FWBP supplementation could enhance digestive enzyme activities and Bifidobacterium population and change the muscle composition and improve growth performance of common carp.  相似文献

In this study, we established and characterized a cell line derived from the kidney of black carp (Mylopharyngodon piceus), which is an important freshwater aquaculture species. The cell line was designated as MPK and subcultured for more than 70 passages in DMEM medium containing 10% fetal bovine serum (FBS) at 28°C. MPK had a modal diploid chromosome number of 48. Moreover, a transient MPK transfection efficiency was up to 18% using a green fluorescent protein plasmid by a modified electroporation. In addition, the MPK cells showed susceptibility to spring viremia of carp virus (SVCV), as demonstrated by the presence of severe cytopathic effects (CPEs) and increased viral RNA. Unexpectedly, the MPK cells expressed pluripotency‐associated genes such as nanog, oct4 and vasa, indicating that these are possibly adult stem cells. Taken together, we have established a stable cell line from kidney that may potentially be utilized as an in vitro platform for genetic modifications and host–pathogen analysis in black carp.  相似文献

A 60‐day indoor trial was conducted to study the effect of periphyton supplementation on metabolic and immune responses in tiger shrimp, Penaeus monodon. Periphyton developed over bamboo substrate in outdoor tanks (15 m2) was used as dietary supplement for P. monodon (2.02 ± 0.04 g) reared in 1000 L FRP tanks. Graded levels of periphyton were included in shrimp basal diets: 0% (P0), 3% (P3), 6% (P6), 9% (P9) and P0 diet with natural periphyton (NP) over bamboo substrate. At the end of the trial, P6 and NP showed significantly higher (P < 0.01) growth rate, 23.9% and 20%, respectively, compared with control, P0. Comparatively, lower level of metabolic enzymes, such as lactate dehydrogenase, malate dehydrogenase, aspartate aminotransferase and alanine aminotransferase, was recorded in treatments P3, P6 and NP compared with control, P0. The periphyton‐supplemented group, P3 had significantly higher (P < 0.05) superoxide dismutase (15.83 ± 0.96) and catalase activity (15.73 ± 0.69) compared to 6.88 ± 2.84 and 9.15 ± 0.67 unit mg?1 protein min?1, respectively, in P0. Similarly, higher total haemocyte counts, 32.58 ± 1.30, 28.51 ± 3.12 and 27.26 ± 4.43 × 106 cells mL?1, were recorded in P6, NP and P3, respectively, compared to P0, 23.57 ± 1.80 × 106 cells mL?1. After challenge with Vibrio harveyi, P3 recorded the highest relative percentage survival 67% followed by NP (58%) and P6 (42%) compared with control. However, treatment with highest periphyton inclusion (P9) did not differ significantly with P0 on growth and immunological parameters. This study indicates that periphyton supplementation at 3–6% level improves growth, immune response and metabolic activities in P. monodon.  相似文献

Effects of exercise on the distribution of phosphofructokinase (PFK), fructose-1,6-biphosphatase (FBPase), and AMP-deaminase between free and particulate-bound fractions was analyzed in white skeletal muscle of rainbow trout Oncorhynchus mykiss. With a widely used technique for the separation of free and bound enzyme fractions (homogenization in low ionic strength, high sucrose buffer), the data showed that the amount of bound PFK increased from 64 to 95% during burst swimming whereas other enzymes were unaffected. Since this data for AMP-deaminase contrasted with earlier reports, different methods of separating free and bound enzyme were evaluated. A clear effect of exercise on AMP-deaminase binding occurred when high ionic strength media (either KCl or KF) were used; in extraction media containing 150 mM KCl, the percent bound rose from 30% in controls to 97% after 1 min burst swimming. Exercise also produced stable changes to AMP-deaminase kinetic properties, including for the bound enzyme (compared with the free) a 2-fold higher Km AMP, a 3-fold higher Ki for inorganic phosphate, and a 60% increase in Ka ADP after 1 min burst exercise. The data suggest that AMP-deaminase in working skeletal muscle is subject to combined controls by allosteric effectors, post-translational modification, and distribution between free and bound states.  相似文献

A 60‐day indoor experiment was conducted to study the effect of dietary supplementation of biofloc on metabolic enzyme activities and immune responses in Penaeus monodon juveniles. Biofloc developed in indoor fibreglass‐reinforced plastic (FRP) tanks (1000 L) was used as dietary supplement in P. monodon (2.90 ± 0.10 g) reared in 1000‐L FRP tanks. Graded level of dried biofloc was included in shrimp basal diets, 0% (control, B0), 4% (B4), 8% (B8) and 12% (B12). The level of metabolic enzymes like malate dehydrogenase (MDH), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) was not significantly different with control up to 8% dietary supplementation. A higher level of total haemocyte count (THC) was noticed in B8 (22.16 ± 2.17 × 106 cells mL?1) and B4 (21.11 ± 0.56 × 106 cells mL?1) compared with control, C (14.61 ± 2.74 × 106 cells mL?1). Biofloc‐supplemented groups recorded significantly higher (< 0.05) serum SOD and catalase activity (P < 0.01) in comparison with control. The groups fed with 4% dietary biofloc supplement recorded highest relative percentage survival (RPS), 45% after challenge with Vibrio harveyi followed by 36% and 27% RPS in B8 and B12 groups. Based on these results, it can be concluded that supplementation of biofloc even at 4% level in the feed improves immune responses and metabolic activities in black tiger shrimp juveniles.  相似文献

Studies of the enzyme fructose-1,6-bisphosphatase (FBPase) of rainbow trout (Oncorhynchus mykiss) have been undertaken in order to illuminate aspects of skeletal muscle gluconeogenesis in these animals. Maximal activities in crude homogenates of several organs suggest that the liver possesses the greatest FBPase activity on a unit g–1 tissue basis but that the white muscle, owing to its bulk, contributes substantially to whole body FBPase activity. Studies of fructose-6-phosphate-1-kinase (PFK) and FBPase in crude homogenates of several organs suggests an important role for intracellular pH in regulating the relative carbon flux through the FBPase/PFK locus in vivo. Furthermore, a three-step purification scheme is described for trout white muscle FBPase by which a stable and homogeneous (by SDS PAGE) enzyme preparation (isoelectric point = 7.2; molecular weight = 37.6 kd) was obtained. Kinetic studies of the purified enzyme were undertaken at 20°C under conditions reflective of "rest" and "exercise/recovery" intramuscular pH in vivo. Affinity for substrate (F-1,6-P2) was increased (Km = 6.88 versus 2.44 mol 1-–1 as was enzyme activity when pH was lowered from 7.0 to 6.5. Various inhibitor metabolites are identified including F-2,6-P2 (mixed-type inhibitor, Ki = 0.201 mol 1–1, pH 7.0) and AMP (non-competitive inhibitor, Ki = 0.438 mol 1–1, pH 7.0). Inhibition by F-2,6-P2 was strongly alleviated by a reduction in pH from 7.0 to 6.5 (I50 increased from 0.14 to 0.32 mol 1–1). AMP on the other hand was a more potent inhibitor at pH 6.5 but this inhibition was totally reversed under conditions of citrate, NH4 + and AMP typical of muscle during recovery from exercise in vivo. In purified white muscle enzyme preparations, FBPase demonstrated maximal activity at pH 6.5 whereas the optimal pH of PFK was 7.0 or greater. Indeed, it appears from these in vitro data that regulation by metabolite levels as well as pH are required for net FBPase flux in vivo. It is concluded, therefore that trout white muscle FBPase demonstrates the potential to play an important enzymatic role in the control of intramuscular gluconeogenesis in these animals. The results are discussed in relation to present knowledge regarding the metabolic responses of trout white muscle to, and its subsequent recovery from, exhaustive exercise.  相似文献

A 56‐day feeding trial was carried out to investigate the effect of dietary glutamate (Glu) supplementation on growth performance, digestive enzyme activities and intestinal antioxidant capacity of grass carp (Ctenopharyngodon idella). A total of 540 grass carp with average initial weight of 370 ± 3.8 g were fed three diets supplemented with 0, 8 and 16 g kg?1 Glu. The per cent weight gain, specific growth rate, feed conversion ratio, and protein and lipid productive value were significantly improved by dietary Glu supplementation. Feed intake, condition factor, visceral somatic index and perivisceral fat ratio were not significantly different between dietary groups. Hepatosomatic index, intestinal somatic index, intestine length index, intestine protein content, trypsin activity in hepatopancreas and intestine, lipase activity in proximal and mid‐intestine (MI) and hepatopancreas, alkaline phosphatase and gamma‐glutamyl transpeptidase activities in intestine, creatine kinase activities in the mid‐ and distal intestine, intestinal folds height in the proximal and MI were significantly increased with increased Glu. Malondialdehyde and protein carbonyl contents in the intestine decreased with increased Glu. Superoxide anion and hydroxyl radical scavenging ability, glutathione reducase, catalase and total superoxide dismutase activities, and glutathione content in the intestine were increased with increased Glu. These results suggested that dietary Glu supplementation increased intestinal antioxidant capacity, digestive and absorptive ability and improved fish growth.  相似文献

This study investigated the effects of phenylalanine on growth, digestive and absorptive ability and antioxidant status of young grass carp (Ctenopharyngodon idella). Young grass carp were fed diets containing 3.4 (basal diet), 6.1, 9.1, 11.5, 14.0 and 16.8 g phenylalanine kg?1 diet with a fixed of 10.7 g tyrosine kg?1 diet for 8 weeks. Percent weight gain (PWG), feed efficiency and feed intake of fish were the lowest in fish fed the basal diet (< 0.05). Trypsin, lipase and amylase activities in the hepatopancreas, and antioxidants including glutathione contents and glutathione reducase activities in the hepatopancreas and intestine were all the highest in fish fed 11.5 g phenylalanine kg?1 diet (< 0.05). Trypsin, chymotrypsin and amylase activities in whole intestine, and creatine kinase, Na+, K+‐ATPase and alkaline phosphatase activities in the proximal intestine, and superoxide dismutase activities in the hepatopancreas and intestine were all the highest when phenylalanine at level of 9.1 g kg?1 diet (< 0.05). In conclusion, phenylalanine improved growth, digestive and absorptive ability, and antioxidant capacity of young grass carp. The phenylalanine requirement of young grass carp (256–629 g) based on PWG was 10.4 g kg?1 diet or 3.44 g 100 g?1 protein.  相似文献

To investigate the effects of niacin on growth, digestion and absorption capacity, and the potential mechanism for digestive and brush border enzyme activities, grass carp (Ctenopharyngodon idella) (256 ± 0.41 g) were fed diets containing 3.95 (basal diet group), 14.92, 24.98, 35.03, 44.97 and 55.01 mg niacin kg?1 diet for 8 weeks. Results indicated that percentage weight gain (PWG), feed intake and feed efficiency were the lowest in basal group (< 0.05). Similarly, niacin deficiency decreased hepatopancreas trypsin, chymotrypsin, lipase and amylase activities (< 0.05), intestinal Na+, K+‐ATPase, alkaline phosphatase, γ‐glutamyl transpeptidase and creatine kinase (CK) activities, the cholecystokinin (CCK) content in proximal intestine (PI) and growth hormone content in serum (< 0.05). Furthermore, niacin deficiency downregulated gene expression of hepatopancreas trypsinogen 1, trypsinogen 2, chymotrypsinogen and amylase, intestinal Na+, K+‐ATPase alpha subunit isoform 1, Na+, K+‐ATPase alpha subunit isoform 8 and CK, and target of rapamycin (TOR) and S6 kinase 1 (S6K1) of hepatopancreas and intestine (< 0.05), whereas upregulated eIF4E‐binding protein (4EBP) gene expression (< 0.05). The niacin requirement for young grass carp (256–689 g) based on PWG, hepatopancreas trypsin activity and Na+, K+‐ATPase in PI was 34.01, 35.10 and 42.08 mg kg?1 diet, respectively.  相似文献

An 8-week feeding trial was conducted to evaluate the effect of dietary carbohydrate sources on the growth performance and hepatic carbohydrate metabolic enzyme activities of juvenile cobia. Six experimental diets were formulated to contain 20% glucose, sucrose, maltose, dextrin, corn starch and wheat starch respectively. The results indicated that fish fed the wheat starch and dextrin diets showed significantly better weight gain, specific growth rate and protein efficiency ratio compared with those fed the other diets. However, fish fed the glucose diet had a significantly lower survival and condition factor than those fed the other diets. There were significant differences in the total plasma glucose and triglyceride concentration in fish fed diets with different dietary carbohydrate sources. Haematocrit, haemoglobin, red blood cell and leucocytes were significantly affected by the dietary carbohydrate sources. The activities of glucose-6-phosphate dehydrogenase (G6PD), 6-phosphofructokinase (PFK) and fructose-1,6-bisphosphatase (FBPase) were significantly affected by the dietary carbohydrate sources, while fish fed the glucose diet showed higher G6PD, PFK and FBPase activities than those fed the other diets. These data indicated that dextrin and wheat starch were the most optimal carbohydrate sources for juvenile cobia.  相似文献

Ovulation stimulation was carried out in carp (Cyprinus carpio L.) females of Lithuanian line B, using carp pituitary (0.3 + 2.7 mg kg?1), Aquaspawn (0.5 mL kg?1) and Aquaspawn + carp pituitary (0.3 mL kg?1 + 2.7 mg kg?1). The poorest results of reproduction (the lowest percentage of ovulating females, the lowest weight of eggs and their poorest quality) were obtained after double hypophysation. The best results were induced by the application of Aquaspawn alone. The effect of the applied stimulators was statistically non‐significant with respect to the weight of eggs, being highly significant (P ≤ 0.01) with respect to the percentage of fertilized eggs and that of living embryos after 24‐ and 36‐h incubation. The multiple‐regression equations were calculated for the three groups separately. The percentage of living embryos after 36‐h incubation constituted a dependent variable while the mass of females, the mass of eggs, the fertilization percentage and the percentage of living embryos after 24‐h incubation were independent variables. The regression was only significant (P ≤ 0.01) for the group of fish treated with Aquaspawn.  相似文献

A total of 1400 juvenile Jian carp (Cyprinus carpio var. Jian) (8.93 ± 0.03 g) were fed diets containing graded levels of vitamin K at 0.027 (basal diet), 1.52, 3.02, 4.51, 6.02 and 7.52 mg kg?1 diet for 60 days to investigate the effects of vitamin K on growth, enzyme activities and antioxidant capacity in the hepatopancreas and intestine. Percentage weight gain (PWG), feed intake and feed efficiency of fish were improved by vitamin K. Activities of trypsin, chymotrypsin, amylase and lipase in the intestine and hepatopancreas and Na+, K+‐ATPase, creatine kinase, alkaline phosphatase and gamma‐glutamyl transpeptidase in the intestine were increased by vitamin K. Malondialdehyde and protein carbonyl contents in the hepatopancreas and intestine were decreased with vitamin K supplements. Certain level of vitamin K increased antihydroxyl radical, antisuperoxide anion, superoxide dismutase, catalase, glutathione‐S‐transferase, glutathione peroxidase and glutathione reductase activities and glutathione contents in the hepatopancreas and intestine. Intestinal Lactobacillus, Ecoli and Aeromonas were changed with vitamin K supplements. Together, these results indicate that vitamin K improved fish growth, digestive and absorptive ability, and anti‐oxidant capacity. The dietary vitamin K requirement of juvenile Jian carp (8.93–73.7 g) based on PWG was 3.13 mg kg?1 diet.  相似文献

The effect of protein quality and of supplementation of corn-glutenprotein with lysine on the growth, feed conversion and protein turnoverrates in white muscle was investigated in carp (Cyprinus carpio) acclimatedto either 18 or 25 °C. Fish fed the lysine-deficient diet showed asignificantly lower food intake, weight gain and feed-conversion efficiencythan animals fed the lysine-sufficient diets, regardless of environmentaltemperature. Coated lysine, compared with free lysine, proved to be asignificantly better way of supplementing dietary protein, as shown byfeed-conversion indices at 18 and 25 °C. White-muscle protein, RNA andDNA contents were not altered by dietary treatment or water temperature.Supplementation with coated lysine, but not with free lysine, significantlyincreased the protein-synthesis rate (KS) at 18 or 25 °Cin comparison to lysine deficiency, although not to control values. Theeffiiency of protein synthesis (KRNA) and retention (PRE)obtained for fish fed the coated-lysine diet, at 18 °C, reached controlvalues. At 25 °C PRE and protein accumulation rate (KG)showed the following significant differences: lysine-deficient diet D, KRNA or protein-related growth, at bothtemperatures.  相似文献
