The concentration of testosterone in the blood and its response to administration of chorionic gonadotropin was examined in two bovine intersexes (testicular pseudo-hermaphroditism) and two control bulls without disorders in sexual differentiation. The levels of testosterone in the blood were markedly lower in both intersexes than in the control bulls. Both intersexes, unlike the control bulls, also showed a response of the blood testosterone level to the administration of chorionic gonadotropin. The conclusion is that in the given type of intersexuality the incretion function of the testes is mardedly deficient as to androgen production and functional hormonal reserve.  相似文献

SUMMARY: Data were collected from 1982 through 1992 from 100 sires and 891 Duroc boars. Testosterone production was measured from peripheral blood samples before (PRE) and after (POST) GnRH challenge. Additionally, data were collected on testes volume at 168 d (TVOL), days to 104 kg (DAYS104), number born alive (NBA) and backfat adjusted to 104 kg body weight (FAT). Realized heritabilities were estimated from the regression of response on cumulative selection differentials. Heritabilities for POST were .15 ± .18 and .24 ± .08 in the low and high lines, respectively. This compares with the estimate of .26 ± .21 from son-sire regressions. The regression of other traits on cumulative selection differentials can be viewed as realized correlated responses to selection. After 10 generations, the high line was approximately three times greater than the low line for both PRE and POST levels of testosterone. Although not significant, high line pigs required fewer days to reach 104 kg, had more backfat and larger testes than low line pigs. Litter size was significantly larger for high line than for low line. Zus?tzlich wurden Angaben über Hodenvolumen am Tag 168 (TVOL), Zeitraum bis 104 kg Lebendgewicht (DAYS104), Zahl lebend geborener (NBA) und auf 104 kg K?rpergewicht korrigierte Rückenspeckdicke (FAT) erhoben. Realisierte Heritabilit?tswerte wurden aus der Regression von Selektionserfolg auf kumulative Selektionsdifferenzen gesch?tzt. Heritabilit?t für POST waren 0,15 ± 0,18 und 0,24 ± 0,08 in der Minus- und in der Pluslinie. Aus Sohn-Vater-Regressionen ergab sich 0,26 ± 0,21. Die Regression anderer Eigenschaften auf kumulative Selektionsdifferenzen k?nnen als realisierte korrelierte Selektionserfolge betrachtet werden. Nach 10 Generationen hatte die Pluslinie etwa dreimal h?here PRE- und POST-Spiegel von Testosteron als die Minuslinie. Obwohl nicht signifikant, brauchten Schweine der Pluslinie weniger Tage bis 104 kg, waren fetter und hatten gr??ere Hoden als Schweine der Minuslinie. Effects of zeranol on the maturation of the adenohypophyseal-gonadal axis were studied in beef bulls. Calves were implanted with 36 mg of zeranol at 3-month intervals from birth through 6 months of age (group 2, n = 10) or were not treated (control group 1, n = 10). After 9 months, group-2 calves were given implants of 36 mg of zeranol at 3-month intervals through 18 months of age (group 2B, n = 5) or were not reimplanted (group 2A, n = 5). Areas under the curves outlined by concentrations of luteinizing hormone (LH), follicle-stimulating hormone (FSH), and testosterone for 6 hours after the administration of 100 micrograms of gonadotropin-releasing hormone (GnRH) were calculated. Gonadotropin-releasing hormone was administered at 3-month intervals from 1.5 through 19.5 months of age. Areas under the curves for concentrations of testosterone for 4 hours after the administration of 10,000 IU of human chorionic gonadotropin (HCG) at 4.5, 7.5, and 10.5 months or 1,000 IU at 13.5 and 16.5 months of age also were calculated. The amount of FSH released was greater (P less than 0.05) for group-2 than for group-1 calves at 4.5 and 7.5 months of age. The amount of FSH released in groups 2A and 2B tended (P less than 0.10) to be greater than that for group 1. Significant differences between groups 2A and 2B were not observed. Thirty-six Landrace x Large White cross boars were selected from litters with either high or low estimated breeding values for 150-d paired testis weight. Blood samples were taken via jugular venipuncture at eight ages (42, 56, 70, 84, 98, 112, 126 and 140 d). At each sampling age, nine blood samples were taken at 30-min intervals. Luteinizing hormone (LH) was determined on the individual serum samples. Serum samples from each boar at each age were pooled and concentrations of follicle-stimulating hormone (FSH), estradiol-17 beta (E2) and testosterone (T) were determined. Paired testis width, testis length and body weight were measured at 98, 112, 126 and 140 d of age. Backfat probe, weights of excised testes and histological data on testes were obtained at 140 d of age. Boars with high testis weight (HTW) were heavier (P less than .05), had higher adjusted backfat probes (P less than .01) and had consistently larger in situ testis measurements (P less than .01) than did low testis weight (LTW) boars. Boars with HTW had heavier (P less than .01) testes and epididymides at 140 d of age. They also had a higher percentage of seminiferous tubules in which spermatogenesis was present (P less than .05), a larger percentage of tubules with a lumen (P less than .05) and tubules had a larger mean diameter (P less than .01) than did those of boar with LTW. Adjustment of in situ testis measurements and excised testis weights for body weight reduced line differences by less than 20%. A rise in LH concentrations occurred at approximately 100 d of age. Boars with HTW had higher (P less than .05) and more variable (P less than .01) LH concentrations than did boars with LTW. Boars with HTW also had higher maximum concentrations of LH during the pubertal rise (P less than .01) and these concentrations tended to reach maximum levels at younger ages. Concentrations of T increased in a fashion that was nearly linear with age (P less than .01) and tended to be higher for the boars with HTW (P less than .10). Concentrations of E2 changed little from 42 to 84 d of age but increased steadily thereafter. Ovarian sensitivity to exogenous gonadotropin stimulation (equine chorionic gonadotropin [eCG] and human chorionic gonadotropin [hCG]) following pre-treatment with a progestin (levonorgestrel) versus GnRH antagonist (antide) was studied in cats known to be induced versus spontaneous ovulators. Queens were assigned to one of three treatments: (1) levonorgestrel implants+eCG/hCG (n=7 cats); (2) antide injections+eCG/hCG (n=7) or (3) eCG/hCG alone (control; n=7). Hormonal metabolites were assessed in fecal samples collected daily for 60 days before and during the 37 days inhibitory pre-treatment and for more than 60 days after eCG/hCG. Fecal metabolites of estradiol and progesterone were measured by radioimmunoassay. Females that maintained baseline progesterone were considered induced ovulators, whereas cats that exhibited a luteal phase before inhibition treatment were classified as spontaneous ovulators. Based on fecal hormone profiles, levonorgestrel thoroughly inhibited ovarian activity, whereas antide synchronized follicular phases but did not induce complete ovarian down-regulation. Both treatments prevented ovulation in spontaneous ovulators, but neither caused regression of existing corpora lutea (CL). Levonorgestrel, but not antide, pre-treatment resulted in a quiescent ovary at the time of eCG injection, yet endocrine responses to eCG/hCG were not different among treatments. Interestingly, spontaneously ovulating females exhibited a prolonged estradiol response to gonadotropin stimulation compared to induced ovulators, and this prolonged estradiol surge was replicated by levonorgestrel pre-treatment. Thus, the progestin levonorgestrel effectively suppresses follicular and luteal activity in the cat, resulting in a more consistent response to gonadotropin stimulation, even in females prone to spontaneous ovulation.  相似文献

Background

The aim of the present study was to investigate the efficacy of the Improvac on testosterone concentration in blood serum, sexual behavior and sperm quality in matured AI boars. A total of nine Danish Landrace AI boars were included in the analysis.

Methods

The trial period lasted for 15 weeks and was divided into four periods: Control period: three weeks before vaccination; Period I – four weeks after first vaccination; Period II – four weeks after second vaccination, Period III – four weeks after third vaccination. Blood and sperm samples were collected at weekly intervals. Freshly collected sperm samples were analyzed.

Results

Testosterone concentration correlated with libido (r = 0.531; p < 0.001), volume of ejaculate (r = 0.324; p < 0.001) and the percentage of morphologically normal spermatozoa (r = 0.207; p < 0.05). Testosterone concentration rised significantly (p < 0.05) in 5–6 week of trial, e. i. after the first dose of Improvac and after this peak the level of testosterone further progressively decreased (p < 0.05).

Conclusions

Results from this study indicate that active immunization of sexually matured boars against GnRH has negative impact on testosterone concentration, sexual behavior, volume of ejaculate and total number of normal spermatozoa in ejaculate.  相似文献

Following weaning at 24 +/- 1 d of age, crossbred boars were subjected to removal of the cranial cervical ganglia (GX, n = 8) or sham surgery (SHAM, n = 8). At 213 +/- 1 d of age, a catheter was inserted into a jugular vein or vena cava, and all boars were housed in environmentally controlled rooms at 22 degrees C with an equatorial photoperiod. After 2 wk of exposure to this photic environment, samples of plasma and serum were collected at hourly intervals for 24 h. The plasma was assayed for adrenocorticotropic hormone (ACTH) and the serum was assayed for cortisol and melatonin by RIA. The time-trends for all three hormones were described by regression models, which were tested for heterogeneity of regression between SHAM and GX treatments. The circadian rhythm of cortisol in serum was similar for SHAM and GX treatments. The profiles of ACTH were also similar between the two treatments, but circadian changes in concentrations of ACTH paralleling those of cortisol were not evident in either treatment. Overall, concentrations of ACTH were reduced (P = .06) for GX boars compared to SHAM boars. The time-trends of melatonin in serum differed (P less than .001) for GX and SHAM treatments, with a nocturnal rise in melatonin evident in some SHAM boars but not in GX animals. Four SHAM boars had profiles of melatonin that obviously entrained to the light-dark cycle. Three experiments were conducted to determine the effects of n-methyl-D,L-aspartate (NMA), an agonist of the excitatory amino acid glutamate, on secretion of hormones in boars. In Exp. 1, boars (185.0+/-.3 d of age; mean +/- SE) received i.v. injections of either 0, 1.25, 2.5, 5, or 10 mg of NMA/kg BW. There were no effects of NMA (P>.1) on secretion of LH and testosterone. Treatment with NMA, however, increased (P<.01) circulating GH concentrations in a dose-dependent manner. In Exp. 2, boars (401 d of age) received an i.v. challenge of NMA at a dose of 10 mg/kg BW or .9% saline. Treatment with NMA, but not saline (P>.1), increased serum concentrations of LH (P<.01), GH (P <.01), and testosterone (P<.06). In Exp. 3, boars that were 152, 221, or 336 d of age were treated i.v. with NMA (10 mg/kg BW). Across ages, treatment with NMA increased circulating concentrations of LH (P<.07) and testosterone (P<.01). However, NMA increased (P<.01) mean GH concentrations in only the oldest boars. Treatment with NMA had no effect (P>.1) on circulating concentrations of estradiol or leptin; however, estradiol concentrations increased (P<.03) with age. In summary, NMA increased secretion of LH, GH, and testosterone in boars. However, endocrine responses to treatment with NMA may be influenced by age of the animal. Finally, NMA did not influence circulating concentrations of estradiol or leptin.  相似文献

The levels were assessed of testosterone in the blood plasma before and 90 minutes after i.v. application of 1 mg of synthetic LH-releasing hormone to 57 boars with disorders of sexual functions and to 43 boars without sexual dysfunctions. The group of animals with sexual disorders included boars with inferior ejaculate quality and low fertility (24 animals) and cases with disturbed sexual potency (33 boars). In animals with the studied changes of sexual functions, compared with boars without sexual dysfunctions, no statistically significant difference was found in the basal concentration of testosterone in the blood. LH-releasing hormone application increased significantly the testosterone levels in the group of boars without sexual disorders by 99.5% on an average and in the whole group of animals with changes in sexual functions approximately by only 65.8%. At the same time in the subgroup of inferior ejaculate quality and low fertility the post-application increase of testicular incretion reached 60.4% and in potency disorders 61.6% and was statistically insignificant in the latter. On the basis of these findings it was derived that in boars with reproduction deviations there existed a decreased incretion reserve of the system hypophysis - testicle and the involvement of this factor in the formation of the studied sexual disorders is assumed. The obtained results are discussed in view of the earlier findings about the incretion reserve of the testicles in boars with changes in sexual functions.  相似文献

The hormone GnRH has a stimulatory effect on gonadotropin synthesis and secretion. The objective of the first study was to evaluate concentrations of FSH and LH in plasma of boars after successive treatment with SB75, a GnRH antagonist. Thirteen boars greater than 1 yr of age (eight White Composite [WC] and five Meishan [MS]) were injected once daily with SB75 (10 microg/kg of body weight) for 4 d. Plasma concentrations of LH and testosterone (T) decreased after 1 h from the first dose of SB75. After 12 h of treatment, LH gradually returned to pretreatment concentrations, but T remained suppressed (< 2 ng/mL) until after the last injection of SB75. There was a modest, but significant, reduction in FSH during treatment with SB75. The prolonged inhibitory effect of SB75 on suppression of plasma T concentrations, in the presence of pretreatment concentrations of LH, implied direct effects of SB75 at the testis. In the second experiment, testicular tissue from adult boars was incubated in the presence of three doses of human chorionic gonadotropin (hCG; 0, .5, and 5 IU) with SB75 (250 ng/mL) or with Deslorelin, a GnRH agonist (500 ng/mL). Samples of media were collected every hour for 3 h, and concentrations of T and estrone (E1) were determined by RIA. Concentrations of T and E1 increased with time in response to treatment with hCG. Co-treatment with SB75 decreased media concentrations of T (P < .01) and E1 (P < .03) compared to controls (77.9 vs 85.7 +/- 2.0 and 4.7 vs 5.3 +/- .2 ng/g). In contrast, treatment with Deslorelin had no effect on the amount of T (P > .50) or E1 (P > .26) released with all dosages of hCG. These results indicate that a GnRH antagonist has a direct effect on the testis, decreasing amounts of T and E1 released from the Leydig cells; however, treatment with a GnRH agonist had no direct effect on release of these gonadal steroids. Thus, it remains unresolved whether the site of action of GnRH antagonist on testicular steroidogenesis is through a testicular GnRH receptor or through some other mechanism.  相似文献

AIM: To investigate ovarian follicular and luteal activity during the postpartum period of cows genetically selected for high or low mature bodyweight, in relation to metabolic and reproductive endocrine parameters, to determine whether there are differences between strains that could affect fertility outcomes.

METHODS: The presence of follicles ≥5 mm diameter and luteal structures was mapped in the ovaries of 12 high (heavystrain) and 12 low (light-strain) mature bodyweight cows by daily trans-rectal ultrasonography from Day 7 postpartum until the end of their first normal oestrous cycle. Blood samples were collected daily, for measurement of concentrations of follicle stimulating hormone (FSH), progesterone, growth hormone (GH), insulin-like growth factor-1 (IGF-1), and insulin. Intervals to first ovulation were calculated from ultrasonography data.

RESULTS: Heavy-strain cows had shorter intervals than light-strain cows from calving to the emergence of the first (9.0 (SE 0.9) vs 12.4 (SE 1.3) days) and second (16.4 (SE 1.8) vs 20.6 (SE 1.6) days) dominant follicles (p<0.05). Concentrations of FSH in heavy-strain cows prior to the emergence of the second, third and fourth dominant follicles were higher than in light-strain cows (p<0.05). Heavy-strain cows were more likely to have a large (>15 mm diameter) follicle earlier than light-strain cows (p<0.01). Concentrations of insulin and IGF-1, but not those of GH, were higher in heavy- than light-strain cows during the postpartum period (p=0.01 and p=0.02, respectively), and concentrations of both on Day 6 were inversely related to the time of emergence of the first dominant follicle (p>0.01).

Concentrations of progesterone were similar in both strains of cow until Day 10 of the first oestrous cycle, but thereafter were higher in light- than heavy-strain cows until Day 16. Progesterone concentrations in heavy-strain cows declined earlier and more rapidly than in their lighter counterparts.

CONCLUSION: These results indicate that there is a rapid postpartum resumption of follicular activity in both heavy-and light-strain cows, but that there is an earlier emergence of dominant follicles and ovulation in the former. Differences in luteal function, in terms of lower dioestrus progesterone concentrations and an earlier onset of luteolysis, in heavy- than light-strain cows might be sufficient to impair the fertility of the former.  相似文献

The purpose of the present study was to test if plasma growth hormone (GH) concentrations in juvenile male and female cattle before or after intravenous stimulation with secretagogues was affected by selection for high (H) vs. low (L) milk yield in lines of Norwegian cattle. In the first of two experiments (A), 32 yearling heifers (16 H and 16 L, at 307–424 days of age) were tested by use of four doses of growth hormone releasing factor (GRF); 0.02, 0.10, 0.50 and 2.50 μg/kg live weight, on 4 consecutive days. The animals were fed ad libitum on a silage-based ration before and during the experiment. Growth hormone was assayed in plasma from blood samples taken at ?15, ?5, 0, 5, 10, 15, 20, 30, 45 and 60 min from stimulation. Plasma GH concentrations were log transformed before statistical analyses. Response variables were; PRIOR (mean of ?15, ?5 and 0 min samples) and PEAK (mean of 10, 15 and 20-min samples). In experiment B, 37 calves (19 H+18 L, 22 males and 15 females, age 114–259 days) were subjected daily to one of three intravenous stimulation tests (GRF, 0.10 and 0.50 μg/kg or thyrotrophin releasing hormone (TRH) at 0.20 μg/kg live weight) on each of 3 consecutive days. Feeding was restricted to cover estimated maintenance requirements only. Rations were given once daily during the test days and 3 days prior to first test. Blood sampling and variables followed those of experiment A. Selection line did not significantly affect GH variables in experiments A or B at any dose of GRF or TRH. GH response increased with increasing dose of GRF up to 0.50 μg/kg. At the highest GRF dose, the response was delayed and persisted longer. Doses giving intermediate to large response increased repeatability of GH measurements. It is concluded that GH secretion in juvenile cattle can be accurately assessed using GRF based stimulation tests combined with restricted and controlled feeding, but it is not affected by selection for milk yield in Norwegian cattle.  相似文献

Mature boars were subjected to chronic treatment with a gonadotropin-releasing hormone (GnRH) agonist, goserelin (D-Ser[But]6, Azgly-NH210), and serum luteinizing hormone (LH) and testosterone concentrations were measured. Ten sexually mature boars were randomly assigned to treatment (n = 5) or control (n = 5) groups. On day 0, boars were implanted sc (day 0) with 2 GnRH agonist implants (1 mg of GnRH/implant) or sham implants. Blood samples were collected at 12-hour intervals on days -2 and -1, at 6-hour intervals on days 0 through 4, and at 12-hour intervals on days 5 through 8. In addition, blood samples were collected at 15-minute intervals for 6 hours on days -1, 0, 4, and 8. Serum testosterone and LH concentrations were determined by radioimmunoassay. Maximal LH (7 +/- 1 ng/ml) and testosterone (26 +/- 3 ng/ml) concentrations were observed at 5 and 18 hours, respectively, after GnRH agonist treatment. Subsequently, LH and testosterone concentrations decreased to pretreatment values (0.3 +/- 0.1 ng/ml and 1.8 +/- 0.4 ng/ml, respectively) by 24 and 48 hours, respectively, after GnRH agonist implantation. Few differences in the characteristics of pulsatile LH release were observed between the groups. Testosterone and LH concentrations in samples collected at 6- and 12-hour intervals and pulsatile LH release did not change after sham treatment of control boars. Whereas previous reports indicated that chronic GnRH administration suppressed serum LH and testosterone concentrations in rams, rats, and dogs, our results indicate that chronic GnRH agonist treatment induced transitory increases, without subsequent suppression, in LH and testosterone concentrations in mature boars.  相似文献

In boars and rabbits, administration of adrenocorticotropic hormone (ACTH) results in a testis-dependent, short-term increase in concentrations of testosterone in peripheral plasma. This experiment was designed to assess the short-term effects of a single ACTH injection on plasma concentrations of testosterone, luteinizing hormone (LH), follicle stimulating hormone (FSH) and cortisol in stallions. Eight light horse and two pony stallions were paired by age and weight and then one of each pair was randomly assigned to the treatment (ACTH, .2 IU/kg of body weight) or control (vehicle) group. Injection of ACTH increased (P<.01) plasma concentrations of cortisol by approximately twofold in the first 60 minutes; control stallions showed no change (P>.10) in concentrations of cortisol during the blood sampling period. Control stallions exhibited a midday increase (P>.05) in concentrations of testosterone similar to that reported previously; ACTH treatment prevented or delayed this increase such that concentrations of testosterone in treated stallions were lower (P<.05) than in controls 4 to 5 hours after injection of ACTH. Treatment with ACTH had no effect (P<.10) on plasma concentrations of LH or FSH up to 12 hours after injection.  相似文献

为探讨铝胶佐剂与白油Span佐剂的GnRH疫苗对公猪睾丸发育和血清睾酮的影响,12头初生公猪随机分为GnRH疫苗铝胶佐剂免疫组和GnRH疫苗白油Span佐剂免疫组(下简称铝胶组和白油组).9周龄初免,17周龄加强免疫,25周龄屠宰.游标卡尺测量猪阴囊直径和睾丸大小,石蜡切片观察睾丸组织发育,ELISA法和放射免疫分析(RIA)法分别检测GnRH抗体水平和血清睾酮水平.结果表明,铝胶组与白油组GnRH抗体效价差异不显著(P0.05).铝胶组血清睾酮水平于17,21周龄(P<0.05)和25周龄(P<0.01)显著地低于白油组,阴囊直径,睾丸直径、横径、周长及睾丸血量均显著地低于白油组(P<0.05).铝胶组与白油组猪睾丸曲精小管均出现不同程度的空泡化,白油组零星分布少量精子,铝胶组无精子产生.这表明在公猪免疫去势的效果上,辅以铝胶佐剂的GnRH疫苗比辅以白油Span佐剂的GnRH疫苗好.  相似文献

This study was designed to clarify the characteristics of changes in plasma concentrations of reproductive hormones in heifers from birth to puberty. Weekly or daily hormonal changes were observed in 39 heifers. Daily changes in the concentration of follicle-stimulating hormone (FSH) demonstrated a consistent cycle of hormone changes over a 7- to 8-day period in heifers from approximately 10 days to 9 months old. Weekly changes in reproductive hormones showed that there were three brief periods in heifers between birth and puberty in which dramatic changes occur. The first period was the first week after birth, during which a reciprocal relationship between steroid hormones and gonadotropins was observed. At birth, the concentrations of steroid hormones were higher than those at any other age. These hormone levels rapidly decreased within the first week after birth. Gonadotropin levels, however, increased from birth to 1 week of age. The second period of major change was at approximately 4 weeks of age when there was an increase in the concentrations of luteinizing hormone (LH), estradiol-17beta, testosterone, and immunoreactive inhibin. The third period was the last 5 weeks before the first ovulation, when there was an increase in the concentrations of estradiol-17beta followed by an increase in (LH). These results suggest that regular hormone changes start from 10 days after birth and that the periods from birth to 4 weeks of age and the last 5 weeks before the first ovulation in heifers are important to the development of reproductive functions before puberty.  相似文献

In dairy cattle, increased circulating growth hormone has been associated with selection for greater milk yield. This study tested the hypothesis that beef cows divergently selected for milk production would have differing GH responses to a challenge dose of GHRH. Growth hormone response to a challenge of GHRH was measured in 36 Angus-sired cows ranging from 6 to 10 yr of age. The cows were classified as high milking (n = 16) or low milking (n = 20), on the basis of their sires' milk EPD. Mean milk EPD (in kilograms) were 16.6 and -14.4 for high and low milking cows, respectively. Milk production was estimated by the weigh-suckle-weigh procedure. Blood samples were taken immediately before and 10 min after a clearance dose of 4.5 microg of GHRH/100 kg BW (injected i.v.) and, 3 h later, immediately before and 10 min after a challenge dose of either 1.5 or 4.5 microg of GHRH/100 kg BW. Each animal received both challenge doses, and the doses were randomly assigned across 2 d of blood collection. Serum concentrations of GH and IGF-I were measured by RIA. Serum IGF-I was measured in the baseline blood sample on d 1 of blood collection. A positive relationship (r = 0.35; P = 0.03) was observed between the cows' rankings for each dose of GHRH; that is, high responders to the low dose were high responders to the high dose. Growth hormone response to the 4.5 microg/100 kg BW challenge dose of GHRH was positively related to sire milk EPD (R2 = 0.09; P = 0.03). Response of GH to the 1.5 microg GHRH/100 kg BW challenge dose also tended to be related (P = 0.08) to sire milk EPD of high milking cows. In addition, IGF-I concentrations of high milking cows were inversely related (R2 = 0.24; P = 0.04) to sire milk EPD. Growth hormone response to GHRH challenge may have potential as an additional tool in the evaluation of milk production in beef cattle.  相似文献

The objective was to determine if the endocrine status of the animal dictates the responsiveness of gonadotrophs to estradiol, activin, inhibin and follistatin; hormones implicated in the differential release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Bovine pituitaries were obtained at 13 (n=8), 30 (n=24) and 66 (n=8) h after the onset of estrus, corresponding to before, during and the end of the first FSH increase of the estrous cycle which follows the pre-ovulatory gonadotropin surge in heifers. Heifers slaughtered at 30 h received no treatment, or were treated with progesterone with or without estradiol before slaughter to suppress the first transient FSH increase. Secretion of FSH from cultured pituitary cells, reflecting the prior in vivo status, was greater (P<0.01) at 30 h than 13 or 66 h, whereas, LH secretion was less (P<0.01) at 13 h compared with 30 h. Treatment with exogenous steroids decreased (P<0.05) the pituitary gland's ability to subsequently secrete FSH and LH. Inhibin and, to a greater extent, estradiol decreased (P<0.01) mean FSH secretion but increased (P<0.05) mean LH secretion. These findings suggest that estradiol and inhibin both have the ability to differentially modulate basal gonadotropin secretion during the first FSH increase of the bovine estrous cycle. Differential regulation of LH and FSH is mediated via an alteration in gonadotropin biosynthesis and basal secretion. Furthermore, the secretory capability of cultured pituitary cells and basal gonadotropin secretion reflect the prior endocrine status of the animal from which pituitaries were obtained.  相似文献

In two sexually mature boars subjected to bilateral vasectomy the content of testosterone was followed in the semen and blood plasma prior to the operation and in the course of 26 days after the operation. In the third boar only the operation without actual vasectomy as the control was made to eliminate the possible effect of narcosis and of the surgical stress. Bilateral vasectomy induced an average decrease in the initial levels of testosterone in the semen plasma of the first and second animal by 63.24% and 47.67% respectively, and 47.17% and 56.46% respectively in the blood plasma. In the control boar no analogous post-operation drop of testosterone concentration in the semen plasma was observed, the content of testosterone in the blood plasma decreased over the followed period on an average only by 14.38%. The results imply that bilateral vasectomy in boars results, in the first followed month after the surgical procedure, in a decrease of semen and plasma testosterone, which proves that the operation under investigation affects testicular incretion.  相似文献
