The effect of early and late sowing dates on the establishment of Orobanche crenata Forsk. (crenate broomrape) in resistant and susceptible cultivars of faba bean (Vicia faba L.) and vetch (Vicia sativa L.) were studied over four seasons in southern Spain. Differences in establishment, development and shoot emergence of the parasite were compared. With resistant faba bean and vetch cultivars, there was reduced attachment and shoot emergence of O. crenata with all sowing dates. Susceptible cultivars were more severely affected by the parasite with early sowing. Orobanche crenata development was also delayed in resistant cultivars. Crop yield, estimated by the number of pods per plant, decreased with late sowings. The combined use of resistant cultivars and early sowing is a useful tool as part of an integrated control strategy. Resistant cultivars allow early sowing (with low O. crenata attack), thus avoiding yield losses due to the short crop cycle with late sowing.  相似文献

为明确我国热带和亚热带地区蚕豆Vicia faba和豌豆Pisum sativum锈病的病原菌种类,通过致病性测定和ITS序列系统发育分析对来自我国云南省玉溪市的4份豌豆锈菌分离物及云南、广西、重庆和四川省(区、市)的5份蚕豆锈菌分离物进行系统鉴定。结果显示,分离自豌豆的锈菌WX1分离物对蚕豆和豌豆均具有高致病性,在侵染叶片上产生大量锈子器;分离自蚕豆的锈菌CX3分离物仅对蚕豆具有高致病性,能在叶片上产生大量夏孢子,而对豌豆的致病性相对较低,仅产生少量的夏孢子堆;分离物WX1和CX3对小扁豆和鹰嘴豆不具有致病性。基于ITS序列系统发育分析表明,所有不同寄主来源的蚕豆单胞锈菌分离物均聚类于一个系统发育组,但分离自蚕豆和豌豆的分离物分别聚类在不同的亚组。表明分离自云南省玉溪市豌豆上的蚕豆单胞锈菌Uromyces viciae-fabae应为豌豆专化型,定名为U. viciae-fabae ex P. sativaum,而来源于云南、广西、重庆和四川省(区、市)的蚕豆锈病病原菌为蚕豆专化型U. viciae-fabae ex V. faba。  相似文献

Field studies were carried out to evaluate the efficacy of imazaquin and glyphosate in controlling broomrape (Orobanche crenata Forsk. and O. aegyptiaca Pers.) in faba bean (Vicia faba L.) in Northern Syria. Several rates and times of application of the herbicides were tested. Nearly complete control of broomrape was achieved with both the herbicides when they were sprayed twice (when the broomrape attachments to the faba bean roots were at tubercle stage of develoment and 15 days later) at a rate of application of 80 g a.i. ha?1 for glyphosate and 10 g a.i. ha?1 for imazaquin. However, the yield of faba bean did not increase with these treatments, suggesting that the herbicides at these rates were still causing some physiological disorder in the plant that counteracted the beneficial effect of broomrape control on the performance of faba bean plants.  相似文献

Various Orobanche species are weedy and cause severe reduction in the yields of many important crops. The seeds of these parasitic weeds may remain dormant in the soil for many years until germination is stimulated by the release of a chemical signal from a host plant. In order to determine the effects of fenugreek root exudate on the induction of Orobanche crenata, Orobanche ramosa and Orobanche foetida seed germination, root exudate was collected from hydroponically grown fenugreek seedlings. Fractionation patterns obtained from column and thin layer chromatography of the fenugreek root exudate showed a set of metabolites differing in their polarity with stimulatory activity on Orobanche seed germination. The crude root exudate stimulated both O. ramosa and O. crenata seed germination to the same level caused by the synthetic germination stimulant GR24 at 10 mg L?1. It also stimulated O. foetida seed germination which did not respond to GR24. Active fractions of root exudate stimulated the germination of Orobanche species differentially.  相似文献

A new strain of broad bean mottle virus, isolated from faba bean (Vicia faba L.) in the Sudan, is described. It differs considerably from known isolates by its nearly symptomless infection of faba bean in spite of high concentrations of the virus in infected plants. It does not differ from regular isolates in gel-diffusion serology, light and electron microscopy, host range and symptoms in major hosts other than faba bean. It may constitute a potential threat to other food legumes in the region.  相似文献

Uromyces viciae-fabae, rust of faba bean, parasitizes other legume crops such as lentils (Lens culinaris) and field peas (Pisum sativum) in some environments. In this study we examined the host range of two Australian isolates of U. viciae-fabae collected and purified from a faba bean crop and classified as U. viciae-fabae ex V. faba. Field pea (P. sativum), chickpea (Cicer arientinum), lupin (Lupinus spp.), lentil (L. culinaris), and mung bean (Vigna radiata) genotypes were tested with these isolates, as well as resistant and susceptible genotypes of the faba bean host. Race specificity for these two pathogen isolates was observed on Vicia faba, with two faba bean genotypes showing partial resistance. Both U. viciae-fabae isolates also colonized field pea seedlings and successfully produced uredinia under glasshouse conditions, despite this fungus not being known as a pathogen of Australian field pea crops. No sporulation of either isolate of U. viciae-fabae ex V. faba was observed on any of the remaining legume species tested. However, obvious differences in fungal growth were observed, ranging from small infection sites with very rare haustorium formation in mung bean to more extensive growth and the development of potential uredinial structures in chickpea. These observations are discussed in relation to the phylogenetic relationship of these host and nonhost species.  相似文献

Orobanche foetida is widely distributed in western Mediterranean countries infecting wild legumes. Recently it has been reported in Morocco infecting common vetch, presenting a further constraint for legume production in this area. Comparative studies between Orobanche populations attacking wild species and Orobanche populations growing on crops from the same region may help to clarify whether a host specialisation process occurred. In the present study, the genetic variability of five O. foetida populations, four infecting wild plants and one infecting cultivated vetch in Morocco was characterised using amplified fragment‐length polymorphism (AFLP) markers to elucidate the existing genetic relationship between populations and to suggest a potential origin for the recently detected vetch‐infecting population. Analysis of molecular variance suggested the existence of genotypic differentiation among populations. The most genetic divergent population by cluster analysis was the population collected on Ornithopus sativus. The vetch‐infecting O. foetida population was closer to the three populations infecting Scorpiurus muricatus. The possibility of the presence of a reservoir of diversity is balanced against direct genetic adaptation and completely new introduction. The potential for this Orobanche species to shift host and become a threat to agriculture exists and has to be taken into account for future legume breeding for these areas.  相似文献

A virus, isolated from faba bean (Vicia faba) obtained from Algeria, was readily recognized as a tobravirus by its particle sizes and morphology. Pea (Pisum sativum) and French bean (Phaseolus vulgaris) characteristically reacted to the isolate like pea early-browning virus (PEBV), but faba bean,Antirrhinum majus, Nicotiana rustica, andN. tabacum reacted with line-pattern symptoms which were unusually brilliant on theNicotiana species. In electronmicroscope decoration tests, the isolate did not react with an antiserum to the Dutch type strain of PEBV, but with one to the broad bean yellow band (BBYB) serotype from Italy. It resembles this serotype in reaction on faba bean, but seems to differ appreciably onN. rustica, N. tabacum, andPetunia hybrida. It is described as a deviant isolate of the BBYB serotype of PEBV.All thirteen faba-bean genotypes tested were found to be susceptible to the Algerian isolate and two Dutch type strain isolates of the virus, and to react with erratic line-pattern symptoms to the Algerian isolate only. All ten genotypes of chickpea (Cicer arietinum) tested reacted hypersensitively, and four out of ten genotypes of lentil (Lens culinaris) were susceptible to the virus but reacted differentially to the three isolates. Seed transmission of PEBV, including the new isolate, in faba bean is confirmed (9% for the Algerian isolate, and over 45% for one of the Dutch type strain isolates), and seed transmission of the virus in a non-legume (N. rustica, 4%) is herewith first reported. This is the first report on the occurrence of the BBYB serotype of PEBV outside Italy, and of PEBV outside Morocco in North Africa.  相似文献

Broomrapes (Orobanche spp.) are parasitic weeds that cause significant losses of crop yield. Experiments were conducted to investigate the seed response to the artificial germination stimulant GR24 in three species of Orobanche subjected to preconditioning under various temperatures, water potentials and with plant growth regulators. The highest germination percentages were observed in Orobanche ramosa, Orobanche aegyptiaca and Orobanche minor seeds conditioned at 18°C for 7 days followed by germination stimulation at 18°C. With the increase of the conditioning period (7, 14, 21 and 28 days), the germination percentage of O. ramosa and O. aegyptiaca progressively decreased. When conditioned at −2 MPa, the germination percentage was lower than at 0 and −1 MPa, especially at 13 and 28°C. Orobanche minor seeds could retain relatively high germination if conditioned at 18, 23 or 28°C, even after significantly extended conditioning periods (up to 84 days). GA3 (30–100 mg L−1), norflurazon and fluridone (10–100 mg L−1), and brassinolide (0.5–1.0 mg L−1) increased seed germination, while 0.01 mg L−1 uniconazole significantly reduced germination rates of all three Orobanche spp. The promotional effects of GA3 and norflurazon and the inhibitory effect of uniconazole were evident, even when they were treated for 3 days. Germination of Orobanche seeds was much lower when the unconditioned seeds were directly exposed to GR24 at 10−6 m . This early GR24-induced inhibition was however alleviated or even eliminated by the inclusion of GA3 or norflurazon (10–50 mg L−1) in the conditioning medium. On the contrary, the inclusion of uniconazole increased the inhibitory effect of GR24, particularly in the case of O. ramosa.  相似文献

There is an increasing interest in the legume species Medicago truncatula as a model plant for structural and functional genomic studies that can be used to identify agronomically important genes in legumes. Field screening has shown very high levels of resistance to Orobanche crenata in M. truncatula. However, in vitro studies with O. crenata, Orobanche foetida, Orobanche ramosa and Orobanche minor showed useful variation among accessions at early stages of the parasite–host interaction. Significant differences were observed in the levels of germination of O. crenata and O. foetida seeds induced by different accessions of M. truncatula. Only limited germination was observed on accession SA‐4327. All accessions induced little O. ramosa and O. minor germination. Accessions also varied in the number of O. crenata and O. foetida attachments supported, with few developing on accession SA‐27774. The variation observed for induction of germination and of subsequent attachment will be useful to isolating and characterizing genes involved in the early stages of Orobanche–host plant interaction and for the study of the biosynthetic pathways of production for germination stimulants.  相似文献

Orobanche species (broomrapes) are parasitic weeds which dramatically decrease the yields of many economically important dicotyledonous crops, including pea (Pisum sativum), in Mediterranean areas. Previously, we identified some Rhizobium leguminosarum strains, including P.SOM, which could both promote pea development and significantly reduce infection by Orobanche crenata, notably through induction of necrosis of attached parasites. In the present study, induced resistance against broomrape in the nodulated pea was shown to be associated with significant changes in rates of oxidative lipoxygenase (Lox) and phenylpropanoid/isoflavonoid pathways and in accumulation of derived toxins, including phenolics and pisatin (pea phytoalexin). Changes were followed for 5 weeks after inoculation and attack by Orobanche. In contrast to non‐inoculated plants or Orobanche only infected plants, polyphenoloxidase (PPO) activity and hydrogen peroxide content increased in response to bacteria inoculation indicating the involvement of oxidative processes. In parallel, the nodulated roots displayed high Lox activity related to the overexpression of the lox1 gene. Similarly, the expression of phenylalanine ammonia lyase (PAL) and 6a‐hydroxymaackiain 3‐O‐methyltransferase (Hmm6a) genes were induced early during nodule development, suggesting the central role of the phenylpropanoid/isoflavonoid pathways in the elicited defence. As a consequence, the derived products, phenolics and pisatin, accumulated in response to rhizobacteria and conferred mechanical and chemical barriers to the invading parasite. These results highlight the likely role of signalling pathways in induced resistance and suggest these mechanisms should be enhanced through integrated Orobanche management practices.  相似文献

Chickpea chlorotic dwarf virus (CpCDV; genus Mastrevirus, family Geminiviridae) is one of the most important legume-infecting viruses with a wide host range and geographic distribution in Africa and Asia. In Iran, CpCDV is common in chickpea (Cicer arietinum), but there is limited information about diversity and infections in plants of other legume species. In the current study, a total of 1671 leaf samples from different pulse crops with symptoms were collected in nine provinces of Iran, and the CpCDV infection status was tested by PCR and/or rolling circle amplification (RCA), resulting in the detection of CpCDV in samples of chickpea, lentil (Lens culinaris) and faba bean (Vicia faba) from different regions. Sequence analysis of complete genomes of 18 isolates recovered by digestion of RCA products revealed infection with isolates of the strains CpCDV-A and CpCDV-F in chickpea, lentil and faba bean. Phylogenetic analysis showed that the Iranian isolates of CpCDV were closely related to previously sequenced isolates of CpCDV-A and CpCDV-F. To the authors' knowledge, this is the first report of CpCDV-F in Iran. Using agroinoculation with infectious clones for one isolate each of CpCDV-A and CpCDV-F, infectivity was confirmed in both faba bean and chickpea, with plants developing leaf curling and/or yellowing. Both infectious clones also successfully infected Nicotiana benthamiana resulting in mild yellowing and intensive leaf curling for CpCDV-A, and dark-green mosaic, dwarfing and mild leaf curling for CpCDV-F.  相似文献

A study on the population dynamics of broom-rape (Orobanche crenata Forskal) in faba bean (Viciafaba L.) was conducted in two locations for 8 years. The O. crenata seed bank increased every year in the 20-cm depth arable layer until it reached approximately 4 million (M) seeds m?2. Seed viability ranged between 53% and 68%, approximately half of the seeds remaining dormant. About 3 × 10?3% of the seed bank became attached to the root system of the faba beans. Only 9% of the attached broomrapes developed and emerged from the soil, possibly reflecting high levels of intraspecific competition. Maximum broomrape seed production for a population of 53 emerged broomrapes per m2 was approximately 4 M seeds m?2. About 43% of the seeds produced were not incorporated into the soil, most probably as a result of their degradation, decomposition or dispersion. Long-term continuous monocropping of faba beans increases the incidence of faba bean wilt, while faba bean–wheat intercropping can effectively control it. This study aimed to understand the underlying mechanism of faba bean–wheat intercropping for the control of Fusarium oxysporum and vanillic acid (VA)-promoted occurrence of faba bean wilt. The occurrence of faba bean wilt was investigated among the monocropped and intercropped plants of faba beans in a field experiment. The contents and types of phenolic acids were examined in the rhizosphere soil. Monocropped and intercropped faba beans were examined under the dual stress of F. oxysporum and different concentrations of VA (0, 50, 100, 200 mg/L) to understand the alleviating mechanism of faba bean–wheat intercropping. Exogenous addition of high concentrations of VA significantly inhibited the growth and reproduction of F. oxysporum, but under the dual stress of F. oxysporum and different concentrations of VA, it significantly inhibited the defence enzymes of faba bean roots, stems, and leaves, and rhizosphere soil enzymes. Interestingly, faba bean–wheat intercropping alleviated VA stress and thereby the incidence and disease index of faba bean Fusarium wilt by improving plant resistance and soil enzyme activity. The dual stress of F. oxysporum and VA promotes the occurrence of Fusarium wilt by damaging the defence system of the faba bean root system and rhizosphere soil environment. However, faba bean–wheat intercropping effectively alleviates the autotoxicity of VA by improving the physiological and biochemical resistance of faba beans and soil enzyme activities, and thus controls the occurrence of Fusarium wilt.  相似文献

The broomrape (Orobanche crenata Forsk) susceptibility to glyphosate applied on faba-bean (Vicia faba L.) as affected by the parasite growth stages at the time of application was studied under field conditions. Glyphosate treatments delayed O. crenata emergence. Single glyphosate application to faba-bean infected with O. crenata predominantly at the stage (a) (small nodule) and (b) (nodules with initial vestigial roots) resulted in a moderate to low control. When the stages (c) (shoot bud already visible) or (d) (shoot and vestigial roots well developed) were the predominant stages, an excellent control was achieved with a single glyphosate application at 60 g ha?1. Increased development stages, with the shoot emerged from the shoot bud, decreased its susceptibility to glyphosate. Orobanche amethystea Thuill.,O. loricata Reichenb. andO. pubescens D’Urv. were found to parasitize the roots of cultivated plants in Israel.Orobanche pubescens is known from the local flora, and was now found to cause damage in parsley fields and toTropaeolum majus L. (nasturtium) in ornamental gardens.Orobanche amethystea andO. loricata are known as occasional weeds in Europe and were recently found for the first time in Israel.Orobanche amethystea densely populated vetch fields in Israel, andO. loricata was found in ornamental gardens. Virus transmission studies were conducted under glasshouse conditions using the vector Bemisia tabaci biotype B to determine how effectively isolates of the begomoviruses Tomato yellow leaf curl virus (TYLCV) and Tomato leaf curl Bangalore virus (ToLCBV) could be transmitted to phaseolus bean, capsicum and tomato test plants, the latter host used as a positive control for transmission. Diagnostic detection of viruses in these host crops and vector was also evaluated. Polymerase chain reaction (PCR) detection of TYLCV in bean cv. Wade and capsicum cv. Bellboy was achieved 4 weeks after fumigation in asymptomatic plants. Detection of TYLCV in tomato controls was achieved 2 weeks after fumigation with improved frequency of detection at 4 weeks. PCR was found to be a more sensitive method than triple‐antibody sandwich enzyme‐linked immunosorbent assay (TAS‐ELISA) for the detection of TYLCV isolates in all hosts. ToLCBV was detected by PCR and TAS‐ELISA in bean. TYLCV was also detected by PCR in the vector, with a novel internal positive control. A 2‐year comprehensive field survey was conducted across major tomato‐growing areas of Iran. Two hundred and thirty‐four tomato fields and six tomato‐producing greenhouses were surveyed for the potential presence of bacterial spot disease. Five hundred and ninety‐six tomato samples with and without symptoms were analysed. While Xanthomonas spp. were found in association with tomato plants both with and without symptoms from five surveyed counties, the bacterial spot disease was observed only in plants from three of them. Only strains isolated from plants with symptoms induced disease symptoms on tomato, while those isolated from symptomless plants caused symptoms only on cabbage and common bean. None of the isolates caused disease symptoms on pepper and eggplant. Phylogenetic analysis showed that X. perforans is the causal agent of tomato bacterial spot in Iran, although X. campestris and X. axonopodis were also associated with symptomless tomato plants. All X. perforans isolates in this study were sensitive to streptomycin, copper sulphate and copper oxychloride at concentrations of 50 mg L?1, 200 mg L?1 and 0.8 g L?1, respectively. Unlike the type strain of X. perforans, isolates in this study did not produce bacteriocin against other Xanthomonas spp., nor were they detected using the usual species‐specific primer pair Bs‐XpF/Bs‐XpR. This suggests an atypical nature of X. perforans strains in Iran, which leads to the hypothesis that X. perforans strains in Iran may have a separate origin to those causing disease epidemics elsewhere. The aggregated dispersal pattern of the diseased tomato fields signifies the seedborne introduction of the pathogen into the country.  相似文献

A total of 188 fungal isolates was obtained from the rhizosphere of Vicia faba grown in an Egyptian soil heavily infested with Orobanche species. Agar cultures of 58 isolates inhibited the germination of conditioned seed of Orobanche crenata exposed to the germination stimulant, GR24. Filtrates of inhibitory fungi grown in liquid medium for 9–15 days were also assayed and those of five isolates, which were morphologically similar, inhibited germination even when diluted 16-fold. The fungus was identified as Myrothecium verrucaria (Alb. & Schwein.) Ditmar by its morphology and the nucleotide sequence of the ITS1 and ITS2 regions of the ribosomal repeat unit. Purification of the inhibitor to homogeneity was accomplished by solvent partitioning, flash chromatography on silica gel, semi-preparative HPLC on a reversed phase C18 column, solid phase extraction and tlc on silica gel. The inhibitor was identified as verrucarin A by nuclear magnetic resonance spectroscopy and comparison of the spectra with those of an authentic sample of the compound. A preliminary experiment demonstrated that infection of V. faba by O. crenata could be prevented by addition of spores of the fungus to soil infested by the parasite.  相似文献

Localized placement of prometryne, linuron and diuron in the soil at the first or second shoot internodes of dwarf broad bean (Vicia faba L.) equally reduced aerial plant growth, whereas simazine and atrazine had no effect. Growth reduction also occurred when the first shoot internode of scarlet runner bean (Phaseolus multiflorus L.) in the soil was treated with all five herbicides, especially with diuron. Localized placement of these herbicides at the first or second shoot internodes of vetch (Vicia sativa L.) in the soil equally reduced aerial plant growth. Foliar injury to vetch due to placement of these herbicides in the shoot zone of the soil was markedly reduced by simultaneous treatment with trifluraiin or nitralin which prevented adventitious root development on the shoot without otherwise affecting plant growth. This lack of root development on the shoots treated with trifluraiin was associated with a marked decrease in 14C-labelled atrazine uptake, which probably accounted for the reduction in atrazine phytotoxicity. A similar explanation may account for the reduced phytotoxicity of the other herbicides in the presence of trifluraiin or nitralin.  相似文献
