Potatoes (Solanum tuberosum L.) supplemented with increasing amounts of sodium selenate were analyzed for glycoalkaloid (GA) content. GAs were extracted with 5% acetic acid from freeze-dried tubers of two potato cultivars, Satu and Sini, harvested 10 weeks after planting as immature. The GAs alpha-solanine and alpha-chaconine were quantified by reverse-phase high-performance liquid chromatography (RP-HPLC) with diode array detection. Two independent experiments were performed. In the first experiment, the total GA concentration +/- standard error of the tubers ranged between 105 +/- 9 and 124 +/- 10 mg kg(-1) fresh weight in Satu and between 194 +/- 26 and 228 +/- 10 mg kg(-1) fresh weight in Sini. The ratio of alpha-solanine to alpha-chaconine was 0.2 in Satu and 0.5-0.6 in Sini. In the second experiment, the total GA concentration +/- standard error was 75 +/- 4 to 96 +/- 11 mg kg(-1) fresh weight, and the ratio of alpha-solanine to alpha-chaconine was 0.3-0.4 in Satu. A high sodium selenate supplementation (0.9 mg of Se kg(-1) quartz sand) slightly decreased the GA content in Satu, but this decrease was not statistically significant. Furthermore, at this addition level the Se concentration increased to a very high level of 20 microg g(-1) dry weight, which cannot be recommended for human consumption. In both experiments, the Se concentration in tubers increased with increasing sodium selenate application levels. Our results show that acceptable application levels of selenate did not have an effect on the GA concentration in immature potato tubers.  相似文献

Effects of Se fertilization on potato processing quality, possible changes in Se concentration and form in tubers during storage, and retransfer of Se from seed tubers were examined. Potato plants were grown at five selenate (SeO4(2-)) concentrations. Tubers were harvested 16 weeks after planting and were stored at 3-4 degrees C prior to analysis. The results showed that the Se concentration did not decrease during storage for 1-12 months. In tubers, 49-65% of total Se was allocated in protein fraction, which is less than found in plant leaves in a previous study. The next-generation tubers produced by the Se-enriched seed tubers had increased Se concentrations, which evidenced the relocation of Se from the seed tubers. At low levels, Se improved the processing quality of potato tubers by diminishing and retarding their raw darkening. The value of Se-enriched potato tubers as a Se source in the human diet was discussed.  相似文献

The calystegines detected in tubers from 17 Phureja ( S. tuberosum Group Phureja) lines and five Tuberosum ( S. tuberosum Group Tuberosum) cultivars were identified as the A 3 and B 2 structural types. Their concentration in whole tubers was of a similar order of magnitude in both species, as was the variability in the ratio of B 2 to A 3. On average, calystegine concentrations in the peel were about 13 times that found in the flesh for the five Tuberosum cultivars, and 4 times higher for four Phureja lines. Removal of the peel reduced the calystegine content by an average of over 50% in Tuberosum but by only 30% in Phureja, despite the latter having the greater proportion of peel. The calystegine content of sprouts was also determined for five Tuberosum cultivars and four Phureja lines and was found to include small amounts of four additional types, B 3, B 4, N 1, and X 2, in addition to the more abundant A 3 and B 2. Concentrations in the sprouts of Tuberosum were on average 100 times higher than that in the tuber flesh and 8 times higher than in the peel, whereas for Phureja, the equivalent values were 30 and 7 times higher, respectively. No correlation was found between sprout concentration and either flesh or peel calystegine concentration.  相似文献

The total ascorbate content of potato tubers from 33 Solanum tuberosum genotypes grown at three geographically diverse sites in Europe in each of two years was determined immediately postharvest and after approximately 4 months of storage at 4 degrees C. Statistically significant differences in total ascorbate concentration were observed between genotypes both at harvest and after storage. In all genotypes, the levels of ascorbate decreased during storage. These results are discussed in terms of their implications for diet and health as well as in terms of breeding for improved vitamin C content in potatoes.  相似文献

Abstract

Experiments with foliar application of selenite to barley crops were carried out on a number of Danish farms during the years 1975 to 1979. The plants were treated at the growth stage of Feekes 4–6 with 2.5, 5 and 10 g Se/ha, and analysed for selenium at harvest. The conclusion from the results is that spraying of 3–5 g Se with a detergent at about Feekes 5 is a safe, cheap, and effective way of preventing an outbreak of selenium deficiency in livestock mainly feeding on cereals.  相似文献

Four related phenolic amides previously undescribed from the species were revealed during metabolic profiling of potato (Solanum tuberosum) tubers. N(1),N(12)-Bis(dihydrocaffeoyl)spermine (kukoamine A) and N(1),N(8)-bis(dihydrocaffeoyl)spermidine were positively identified by comparison with authentic standards, while the structures N(1),N(4),N(12)-tris(dihydrocaffeoyl)spermine and N(1),N(4),N(8)-tris(dihydrocaffeoyl)spermidine are proposed for the other two metabolites. Each amide was present at several tens of micrograms per gram of dry matter. Several of these compounds were subsequently detected in other solanaceous species, such as tomato (Lycopersicon esculentum) and Nicotiana sylvestris. They appeared not to be present in Arabidopsis thaliana or Beta vulgaris. Bis(dihydrocaffeoyl)spermine isomers have previously been identified in only a single plant, the Chinese medicinal species Lycium chinense (Solanaceae), where they may account for some of the described biological activity. The other compounds have not until now been reported in vivo, though some of the equivalent hydroxycinnamoyl derivatives are known. The surprising discovery of kukoamine and allies in a range of solanaceous species including potato, a common food crop that has a long history of scientific investigation, provides exemplary evidence for the potential of the nontargeted techniques of metabolomics in studying plant metabolites.  相似文献

A method of hydride generation atomic fluorescence spectrometry was applied to the determination of the selenium concentration of regular polished rice in China and selenium-enriched polished rice obtained by foliar application of selenium-enriched fertilizer in the forms of selenite and selenate. The average selenium content of regular rice was 0.025 +/- 0.011 microg g(-)(1). On the basis of a daily dietary rice intake of 300-500 g suggested by the China Nutrition Society, the total selenium intake from regular rice was calculated to be 7.5-12.5 microg per person per day for an adult. The selenium contents of rice were significantly increased to 0.471-0.640 microg g(-)(1) by foliar application of selenium-enriched fertilizer at rate of 20 g of Se ha(-)(1) in the forms of sodium selenite and sodium selenate. The selenium content of rice by application of a fertilizer of selenate was 35.9% higher than that by a fertilizer of selenite, which showed that Se-enriched fertilizer in selenate exhibited greater efficiency in increasing Se content in rice products. The Se-enriched rice products can increase daily Se intake on average by 100-200 microg of Se per day by the consumption of 400 g of rice products if the Se level of rice products is controlled at 0.3-0.5 microg of Se g(-)(1). Because rice is a staple food in China, selenium-enriched rice obtained by bioenrichment of selenium to increase the Se content of rice could be a good selenium source for the population in selenium-deficient regions.  相似文献

Abstract

The objective of this sand culture experiment was to determine how fertilization methods (i.e., fertigation rates of 0.5, 1, and 2?mg Se·pot?1, foliar rates of 5, 10, and 20?mg Se· L?1) and fertilizer type (i.e., selenate or selenite) affected wheat Se concentrations. The results showed that the fertigation and foliar treatments both increased wheat Se content. In the fertigation and foliar treatment total Se content of wheat was greatest in the selenate treatments. In the fertigation treatment, the Selenite had mainly accumulated in the roots, while the Selenate was majority transported to shoot. In the foliar treatment, we found that most of Se was transported to grain. In addition, the organic Se concentration was the most than other Se fractions. In conclusion, selenate was more effective than selenite in both the fertigated and the foliar application treatments. And the foliar application was better than fertigation.  相似文献

The influence of 2 weeks and 3 months of dark storage upon light-induced chlorogenic acid accumulation within tuber tissue of four potato cultivars and upon 5-, 4-, and 3-caffeoylquinic acid concentrations within cv. King Edward was determined. Storage period significantly affected (P < 0.05) the magnitude of the light-induced chlorogenic acid response with accumulation rates 3-4 times higher in tubers exposed to light after 2 weeks compared with those placed under light after 3 months. Comparison of chlorogenic acid concentrations in controls after 2 and 3 months of dark storage indicated that tuber chlorogenic concentrations decline during prolonged cold store at 5 degrees C. Rates of accumulation in response to light were cultivar-dependent with cv. Fianna the most light-sensitive and cv. Maris Piper relatively light-insensitive. In virtually all cases exposure to sodium and fluorescent light promoted higher rates of accumulation than did exposure to high-pressure mercury light sources. Chlorogenic acid values steadily increased over 15 days of illumination with, in the majority of cases, no indication of cessation. Light exposure increased 5-, 4-, and 3-caffeoylquinic acid accumulation rates in cv. King Edward. Irrespective of storage period and light source, ratios of 5-:4-:3-caffeoylquinic acid were ca. 85:15:0 at day 0 and 52:42:6 by day 15.  相似文献

A four-year study of a number of compositional parameters of potato tubers during low-temperature storage was conducted to examine the compositional differences between cold-tolerant (low sugar-accumulating) and cold-sensitive (high sugar-accumulating) tubers in relation to potato chip processing quality. Compositional parameters analyzed included sucrose, reducing sugars, nitrogen, protein, ascorbic acid, and dry matter content. Pearson correlation analysis of the data illustrated that chip color was most closely correlated with reducing sugar concentration. Multiple regression analysis revealed that the relative contribution of each parameter to chip color varied greatly among the cultivars and selections evaluated and from season to season. This analysis demonstrates that the quantitative relationships between the measured compositional parameters and chip color were not sufficient to provide a general predictive index of chip color quality for tubers processed directly from low-temperature storage.  相似文献

Secondary metabolites in potato tubers include both phytonutrients and plant defense compounds. The extent these small molecules vary among different potato genotypes is not well characterized. LC-MS analysis of tuber extracts from seven potato genotypes showed that one large source of small molecule variation is the glycoalkaloids. Glycoalkaloids are involved in the resistance of potatoes to pathogens and pests, but they also have implications for human health and nutrition. This study focused on glycoalkaloids with solanidane or solanidane-like aglycones, of which over 50 were tentatively identified, many of which appeared to be novel glycoalkaloids. Results suggested the variety of glycoalkaloids in potatoes is considerably greater than previously thought. Dissecting the role of these many glycoalkaloids in human health or pest and pathogen resistance will be a formidable undertaking.  相似文献

【目的】探究在茶树不同生育时期叶面喷施不同硒肥对夏茶产量、品质及硒含量的影响,为生产富硒茶提供技术依据。【方法】田间试验在浙江嵊州进行,供试茶树品种为‘中茶108’。试验采用二因素列区设计,主处理为硒肥种类(A因素),副处理为硒肥喷施时期(B因素)。主处理设喷施清水对照(A0)、硒酸钠(A1)、亚硒酸钠(A2)和酵母硒(A3);副处理设夏茶顶芽萌发前(5月12日,B1)与1芽1叶期(5月20日,B2)两个喷施时期。硒肥喷施浓度均为Se 50 mg/L,硒肥溶液喷施量为1.8 L/m2。当茶树蓬面1芽2叶占比达30%左右时,每个小区随机选取30 cm×30 cm茶蓬,调查蓬面新梢总数、1芽2叶数量、1芽2叶长度和百芽重。同时,取1芽2叶新梢样品,测定茶多酚、儿茶素、咖啡碱、游离氨基酸、花青素以及硒含量。【结果】与A0B1处理相比,A3B1处理茶树萌展值显著降低了0.14,但对茶树蓬面新梢总数无明显影响;A2B1处理茶树1芽2叶新梢长度和百芽重分别显著降低了1.04 cm和1.94 g。A1B2和A3B2处理茶树蓬面新梢总数分别显著降低了17.66和22.33,但不影响其萌展值;A1B2显...  相似文献

Total and individual fatty acid contents were determined in raw tubers of four genotypes from each of the two species Solanum phureja and S. tuberosum. The four S. phureja genotypes contained statistically significantly higher concentrations of total fatty acids, with their average value being 37% greater than that for the mean of the four S. tuberosum cultivars. In both species, a total of 17 fatty acids were detected in quantifiable amounts, and in all genotypes the predominant fatty acid was linoleic followed by alpha-linolenic and palmitic acids. Unusually, 15-methyl hexadecanoate was present as a minor acid in both species. Although a number of statistically significant differences in the fatty acid percentage compositions were found between and within the two species, these were generally small. Averaged over all species and genotypes, tuber storage resulted in an initial small but statistically significant increase in total fatty acid content, but prolonged storage resulted in a fall to the initial values detected close to harvest. The same trend was evident for S. phureja alone (for mean values of all genotypes), but for S. tuberosum the total fatty acid content remained constant over the whole storage period. For both species, the contents (both as absolute levels and as percent compositions) of linoleic acid decreased and alpha-linolenic acid increased in tubers over the whole storage period, and possible mechanisms are discussed. Also, the absolute levels of these two acids were greater in S. phureja than in S. tuberosum, and this is discussed in relation to the development of flavor-related compounds during cooking. The polar lipids of one representative of S. tuberosum and of S. phureja were qualitatively similar. There were only minor differences in the polar lipid percentage compositions and in the corresponding fatty acid compositions of the individual polar lipids between the two species, although the absolute levels of the total, and of some individual, polar lipids were higher in S. phureja.  相似文献

The effect of selenium (Se) treatments on potato growth and Se, soluble sugar, and starch accumulation was investigated. Potato plants were cultivated in quartz sand without or with sodium selenate (0, 0.075, 0.3 mg Se kg(-1) sand). In young potato plants, Se treatment resulted in higher starch concentrations in upper leaves. The tuber yield of Se-treated potato plants was higher and composed of relatively few but large tubers. At harvest, the starch concentration in tubers did not differ significantly between treatments. The higher Se addition (0.3 mg Se kg(-1)) may have delayed the aging of stolons and roots, which was observed as high concentrations of soluble sugar and starch. Together with the earlier results showing elevated starch concentration in Se-treated lettuce, the findings of this research justify the conclusion that Se has positive effects also on potato carbohydrate accumulation and possibly on yield formation.  相似文献

The total carotenoid content was determined of tubers from 38 Solanum phureja lines grown in field plots over 3 years. The results indicated a significant difference between years, but the ranking was similar from year to year and the interaction between season and variety was small. Postharvest storage significantly reduced the carotenoid content of the tubers, and reducing the storage temperature further lowered the carotenoid content. Examination of the individual carotenoids revealed that lutein was the most stable and least likely to be reduced, while the levels of the carotenoids derived from beta-carotene were significantly reduced during storage at either temperature. Exposure of the tubers to either mercury or sodium lights resulted in a significant increase in total carotenoid content, concomitant with elevated chlorophyll. Although both types of radiation produced a broadly similar increase in total carotenoid contents, differential effects on the individual carotenoid profile of the light-induced carotenoids were observed.  相似文献

Abstract

The proportionate distribution of fertilizer P amongst all organs of the potato plant was measured by frequent sampling of a field grown crop which had received labelled P fertilizer. The observed values of the proportions of P derived from the fertilizer in each organ were compared to the values predicted by three models. The results of the comparison indicated that differences in the proportionate distribution of fertilizer P amongst organs are to be expected whenever the proportion of fertilizer derived P in P entering the plant exceeds the proportion of fertilizer derived P in the P already present in the plant.  相似文献

Regardless of variable environmental conditions and changing pathogen populations, the Estonian potato cultivar (cv.) Ando has shown elevated field resistance to Phytophthora infestans for over 40 years. One of cv. Ando’s parents, the Latvian cultivar Agra, is considered a cultivar of medium to low resistance, the other parent, line 382-48, has a well-known source of resistance, Solanum demissum, in its pedigree. Segregation of late blight disease severity in a population of 95 hybrid plants from cv. Ando’s backcross to cv. Agra (cv. Ando x cv. Agra BC1) in field trials of three consecutive years (2013?2015) and in a test of a single pathogen isolate fits a model of additive action of multiple unlinked race-specific resistance genes responsible for the field resistance of cv. Ando. At least some of the resistance genes in cv. Ando’s genome do not function in juvenile plants and are developmentally upregulated.  相似文献

Earlier attempts to measure potato (Solanum tuberosum) leaf glycoalkaloids indicated variability among similar plants, suggesting that a single small sampling of a young plant might not be a reliable measure of composition. It was also suggested that freeze-dried leaf samples might be less variable than fresh ones. In the present work, variability was minimized by comparing single leaves from the same stem position of each plant. Comparisons involving other leaves indicated that the glycoalkaloid content was not constant with respect to either time or position on the stem. In addition, the possible influence of differences in growing conditions at different times suggests that repeated plantings should include a known variety as a control to which other plants are compared. Variability was reduced by calculating glycoalkaloid concentrations on a dry weight rather than fresh weight basis. The method of drying the samples, however, had no influence on the variability of data. These considerations should be generally applicable to the sampling of replicate leaves of any plant for analysis of any components.  相似文献

Abstract

Phosphorus (P) is an essential element and its efficient use is of global importance. This study evaluated the effect of growing potato under legume intercrops on P uptake and use efficiency indices: P harvest index (PHI), P uptake efficiency (PuPE), P partial factor productivity (PPFP) and P partial balance (PPB). The experiment was carried out for four consecutive seasons with treatments comprising potato cultivated under legume intercrops: none (T1), dolichos (Lablab purpureus L) (T2), peas (Pisum sativum L) (T3) and beans (Phaseolus vulgaris L) (T4). Across the seasons, the mean haulm P uptake for T2 (6.7?kg P ha?1), T4 (5.5) and T3 (4.5) were 6%, 23% and 36% lower than that observed in T1 (7.1?kg P ha?1), respectively. On the other hand, tuber P uptake was highest in T1 (21.8?kg P ha?1) and T2 (21.3?kg P ha?1) and were significantly higher than 13.2?kg P ha?1 in T3 and 15.1?kg P ha?1 in T4. This had a profound effect on PuPE, which was equally highest in T1 (0.26?kg total P uptake kg?1 P supply) and T2 (0.25) and lowest in T3 (0.16) and T4 (0.18). Similarly, PPFP, PHI and PPB followed a similar trend, with highest values in T1 (57?kg tuber dry matter yield kg?1 P supply, 76.4?kg tuber P uptake kg?1 total plant’s P uptake and 0.20?kg tuber P uptake kg?1 P supply, respectively). Among the tested legume intercrops, dolichos competed least for P with the main crop (potato) hence it can be integrated into potato-based cropping systems without compromising potato tuber yield.  相似文献
