Efficacy of a 1% solution of sodium carboxymethylcellulose (CMC) infused into the peritoneal cavity of ewes was evaluated for prevention of intraperitoneal adhesions resulting from surgery of the reproductive tract. Six ewes were assigned to each of 4 groups. Group-1 ewes were controls that underwent ventral midline celiotomy and exploration of the abdominal viscera. Group-2 ewes were treated similarly to group-1 ewes, except that a 1% solution of CMC (14 ml/kg of body weight) was infused into the peritoneal cavity. This group was studied to determine whether CMC would cause changes in the peritoneal cavity. Group-3 comprised ewes representing a uterine trauma model. Ewes underwent abdominal exploration, but in addition had a standard embryo collection technique performed on 1 uterine horn and hysterotomy performed on the opposite uterine horn. Group-4 ewes were treated like group-3 ewes, except that, similar to treatment of group-2 ewes, CMC was infused into the peritoneal cavity. All ewes were euthanatized and necropsied 12 to 14 days after surgery. Abdominal adhesions were evaluated, and an adhesion severity score was assigned to each ewe on the basis of number and severity of the adhesions. Ewes of all groups had abdominal adhesions. Significantly (P less than 0.05) lower adhesion score was observed in ewes given CMC (groups 2 and 4) than in the adhesion model (group 3). Significant difference was not observed in adhesion score when groups 1, 2, or 4 were compared. Though not statistically significant, fewer adhesions were observed in ewes of groups 2 and 4 than in group-1 ewes.  相似文献

Ewes were treated with a medroxyprogesterone acetate (MAP) sponge for 8 d followed, at sponge removal, with 500 IU pregnant mare serum gonadotropin (PMSG) at d 30, 40 or 50 (d 0 = lambing) to induce estrus. Dry and lactating ewes were divided into equal numbers at each postpartum day and bred at estrus. Conception rates and number of accessory sperm were determined by flushing the oviducts 3 d after mating and examining the recovered ova. There was no effect (P greater than .05) of lactational status on conception rates. Conception rates increased (P less than .05) from d 30 (10%) to d 40 (45%) and from d 40 to d 50 (80%). There were fewer (P less than .05) ova with accessory sperm (5/26) in d-30 ewes compared with d-40 (10/27) or d-50 (12/24) ewes. In Exp. 2, ewes were assigned to two groups after receiving PMSG on d 30: 1) mated naturally or 2) inseminated during laparotomy near the uterotubal junction (UTJ). Dry and lactating ewes were divided evenly within each of the two treatments. Oviducts were flushed and ova were examined for cleavage. The conception rate was 60% in ewes that were inseminated in the UTJ vs 10% in ewes mated to rams (P less than .05). Lactational status had no effect on results. In conclusion, conception rates in postpartum ewes treated with MAP sponge and PMSG increased from postpartum d 30 to d 50 with natural breeding, and d-30 conception rates were increased over natural mating by insemination into the uterine horn near the UTJ.  相似文献

A 6-year-old Hereford embryo donor cow was referred to Auburn University College of Veterinary Medicine for a mass in the tip of her left uterine horn. The cow had recently undergone an embryo collection which yielded unfertilized, degenerated ova. Transrectal palpation and ultrasound revealed a multi-locular mass enveloped by two separate compartments that resembled an amniotic and allantoic cavity within the uterus. Tissue was collected via a uterine flush and submitted for histopathology. The tissue was determined to be placenta, confirming the diagnosis of a molar pregnancy. Following treatment, the cow was able to produce numerous viable embryos. Molar pregnancies are rare and characterized by abnormal growth of trophoblastic cells leading to formation of intrauterine cystic masses. It is important to routinely perform an ultrasonographic examination of the cow's reproductive tract approximately 30 days following non-surgical in vivo embryo collections to detect and treat unwanted conditions such as pregnancy and cystic conditions.  相似文献

Synchronization of development between the embryo and uterus is required for successful pregnancy establishment.Transfer of early embryos requires synchrony with the recipient uterus of 2 days or less in sheep,because asynchrony of 3 days or more results in failure of pregnancy recognition signaling for maintenance of corpus luteum (CL) and progesterone (P4) production and/or uterine support of the embryo.The objective was to determine if P4 treatment of recipient ewes would obviate the need for pregnancy recognition signaling and maintain a uterine environment conducive to embryo survival after asynchronous transfer,thereby establishing a universal recipient.Embryos (morulae/blastocysts) were recovered on day 6 from super-ovulated donor ewes.Recipient ewes received 25 mg P4 daily from day 6 post-estrus until 60 days after embryo transfer.Embryos were transferred into recipients on day 6,9,12,18,or 30 post-estrus.The pregnancy rate on day 22 post-transfer was 60% for synchronous transfers to day 6 ewes,44% and 22% for asynchronous transfers to day 9 and 12 ewes,and 0% for asynchronous transfers to day 18 and 30 ewes.On day 39 post-transfer,pregnancy rates remained 60% for day 6 ewes,33% for day 9 ewes,and 0% for day 12,18,and 30 ewes.The P4 treatment did extend the window of uterine receptivity to early embryos in ewes by one day,but did not create a universal recipient.Available results support the idea that a window of uterine receptivity to the conceptus exists in sheep that is independent of pregnancy recognition signaling.  相似文献

The present study aimed to investigate the fertility of ewes artificially inseminated with three different methods using a synthetic semen extender, AndroMed. The three methods of artificial insemination (AI) were cervical AI with fresh-diluted or frozen-diluted semen at observed estrus, and an intrauterine AI with frozen-thawed semen. A total of 80 ewes were treated with a controlled internal drug release (CIDR) containing 0.3 g progesterone per device for 12 days. In Experiment 1 (26 Suffolk ewes), superovulation was induced with 20 mg follicle-stimulating hormone and 250 IU equine chorionic gonadotropin (eCG) two days and one day before CIDR removal, respectively, during the non-breeding season. In Experiment 2 (54 Suffolk and Suffolk crossbred ewes), an intramuscular injection of 500 IU eCG was administered one day before CIDR removal to synchronize estrus and ovulation during the breeding season. In Experiment 1, fresh-diluted or frozen-thawed semen was deposited into the cervical orifice after estrus detection, and an intrauterine AI with frozen-thawed semen was performed by laparoscopy at a fixed-time basis without estrus detection. Embryos were recovered by uterine flushing 6 days after AI, and the rates of recovered, fertilized (cleaved) ova and embryos at the morula or blastocyst stage were compared among the three AI methods. In Experiment 2, the pregnancy rates after the three AI methods were compared. In Experiment 1, the rates of recovered ova were not significantly different among the three AI methods (52.5-56.7%). The rate of fertilized ova (81.0%) by laparoscopic AI with frozen-thawed semen was significantly higher compared with cervical AI of fresh-diluted (25.5%) or frozen-thawed (3.5%) semen, but the rate of embryos at the morula or blastocyst stage (17.6%) was significantly lower than that of the cervical AI with fresh-diluted semen (69.2%). The rates of ewes yielding fertilized ova were not significantly different among the three groups (44.4, 11.1 and 62.5% for cervical AI with fresh-diluted and frozen-thawed semen and intrauterine AI with frozen-thawed semen). In Experiment 2, the pregnancy rate of ewes intrauterinally inseminated with frozen-thawed semen (72.2%) was significantly higher than those of ewes inseminated cervically with fresh-diluted (5.5%) or frozen-thawed (0.0%) semen. The present results showed that acceptable fertilization and pregnancy rates could be obtained by an intrauterine AI with frozen-thawed semen using a synthetic semen extender (AndroMed), but not sufficient by the cervical AI with either fresh or frozen semen.  相似文献

Records of 79 cats determined to have endometrial hyperplasia (EH) with or without pyometra or endometritis were studied. Forty-eight cats (61%) were spayed or died because of complications relating to reproductive tract disease. All other cats were free of clinical signs of disease, and lesions were found incidentally during routine ovariohysterectomy or at necropsy. Prevalence of uterine lesions was greater in cats greater than or equal to 5 years old. Breeding, age at first breeding, and queening could not be correlated with clinical or pathologic signs of disease. Seventy-one percent (34/48) of cats with clinical signs referable to the reproductive tract had histologic evidence of pyometra or endometritis, whereas only 19% (6/31) of cats without clinical signs had histologic evidence of either disease. Only 23% (9/39) of cats without pyometra or endometritis had corpora lutea (CL) in at least one ovary, whereas 40% (16/40) of cats with either pyometra or endometritis had CL. We concluded that the prevalence of EH in cats increases with age in sexually intact female cats, and that most cases of pyometra or endometritis, but not EH, in cats are associated with retained CL.  相似文献

A study was conducted to determine whether the antifertilization of intrauterine devices in ewes could be overcome by the use of hormones or drugs that either stimulate or inhibit uterine motility. 44 parous, western ewes were unilaterally ovariectomized, and a plastic spiral was inserted into the uterine horn opposite the remaining ovary. Spirals were immediately removed from 5 ewes. At the time of mating, 2 weeks to 3 months post surgery, sham-operated and spiral control ewes were injected with saline solution. Ewes with spirals were injected with either estradiol, epinephrine, oxytocin, oxytocin and acetylcholine, or oxytocin, acetylcholine, and physostigmine. Other ewes with spirals were laparotomized, and either the uterus was massaged or the cervical lumen was cleaned. 3 days postmating, 8 ova recovered from 5 sham-operated ewes were all cleaved, with a mean of 90 accessory sperm in the zonae pellucidae. 3 of 12 ova recovered from 8 spiral control ewes were cleaved. Only 1 of 40 ova recovered from 28 treated ewes was cleaved. Accessory sperm were not found in either cleaved or uncleaved ova from any spiral ewe. None of the treatments overcame the antifertilization effect of the spiral.  相似文献

The hypothesis that subnormal luteal function after induced ovulation in anestrous ewes was the result of uterine influences exerted during the periovulatory period was tested. Crossbred ewes (n = 27) in seasonal anestrus were induced to ovulate by administration of 12 doses of 250 ng of LHRH at 2-h intervals, followed immediately by a bolus injection of LHRH (250 micrograms; d 0). Ewes were unilaterally hysterectomized on either d -3 (PRELHRH) or 2 (POSTLHRH). Daily blood samples were collected and assayed for progesterone (P4) and 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM). All ewes were slaughtered on d 10, and corpora lutea (CL) were collected, weighed, and assayed for concentration of P4. All ewes that ovulated exclusively in the ovary ipsilateral to the remaining uterine horn had a transient increase in plasma P4 of 2 to 3 d (short luteal phase). In ewes with at least one CL in the isolated ovary, elevated plasma P4 was maintained after hysterectomy but was consistently lower (P less than .05) in POSTLHRH ewes than in PRELHRH ewes. Concentrations of PGFM did not differ between treatments. The CL ipsilateral to the remaining uterine horn weighted less (P less than .01) and contained less P4 (P less than .01) than contralateral CL. These data confirm the hypothesis that premature regression of subnormal CL is uterine-dependent in a local fashion. Presence of the uterus during the follicular and(or) early luteal phase inhibited subsequent luteal function in seasonally anestrous ewes.  相似文献

In Exp. 1, 21 first-service cattle and seven repeat-breeder cattle, averaging 4.7 infertile services, were brought into estrus and superovulated by treatment with follicle-stimulating hormone and prostaglandin F2 alpha. At insemination, semen was deposited in the greater curvature of one uterine horn, about midway between the utero-cervical junction and the utero-tubal junction. Cattle were necropsied 2 to 7 d after estrus and ova were recovered and examined. The fertilization rate for first-service cows was 74% of 362 intact ova and for repeat-breeders, 43% of 128 intact ova (P less than .001). Fertilization rate in first-service cows was 81% on the side of semen deposition and 68% on the opposite side (P less than .01); the rates in repeat-breeders were 54% and 32% (P less than .025). Differences between sides were due mostly to four cows that averaged 93% fertilization on the side of semen deposition and 19% on the opposite side. The proportion of fertilized ova with accessory sperm (17%) did not differ between sides of the reproductive tract. In Exp. 2, 60 first-service and 32 repeat-breeder cows in natural estrus had semen deposited in the uterine body or in the greater curvature of one uterine horn, either on the side of impending ovulation or on the opposite side. At necropsy, 55 ova were recovered from first-service cows, of which 42 (76%) were intact and 13 (24%) were ruptured or fragmented. Of the 42 intact ova, 41 (98%) were cleaved. From the 32 repeat-breeders, 30 ova were recovered, of which 26 (87%) were intact and 4 (13%) were ruptured; 23 of the 26 intact ova (88%) were cleaved. Site of semen deposition had no significant effect on either fertilization rate or number of accessory sperm in either type of cow. First-service cows averaged more accessory sperm (40) than did repeat-breeders (19, P less than .01). Overall results indicated that sperm deposited deep in one uterine horn fertilized ova nearly as frequently in the opposite oviduct as in the adjacent oviduct except in 14% of superovulating cattle.  相似文献

In the autumn oestrus season, 20 Slovak Merino ewes were exposed to synchronization of oestrus, treated with the PGF2alfa at doses 125 micrograms (Oestrophan, inj. Spofa). followed by an injection of PMSG at doses 1000 IU (Antex Leo Denmark) and 50,000 IU of Vitamin A (Axerophtol Spofa). 23 anoestrus ewes were synchronized with an intravaginal sponges containing 20 mg of chlorsuperlutine (Agelin, Spofa) for 12 days and after sponge withdrawal, the ewes were injected with 750 and 1000 IU of PMSG (Antex Leo Denmark). Ovulatory response was observed and the possibility of ova recovered from the genital organs in ewes after synchronization of oestrus and superovulation in oestrus season. Higher values of the total follicular response (CFO), and the average number of ovulation (PO) after administering equal doses of PMSG were found out both in anoestrus ewes (CFO 6.62 +/- 4.24; PO 4.25 +/- 4.52) and in oestrus ewes (CFO 2.70 +/- 2.10; PO 2.60 +/- 1.74; resp. CFO 2.80 +/- 1.83; PO 3.4 +/- 3.0), if the ewes were treated with PMSG together with vitamin A. The average number of ova flushed was higher in anoestrus ewes (3.0-0.5) than in oestrus ewes (1.67-3.75). In both trials the equal ratio in the number of released ova was gained from ewes of experimental groups (83-88% of the total number). After ova flushing from the genital organs in ewes of the experimental groups most ovas were found in the isthmatal part of the uterine tube (36-60%). On the basis of gained results it was concluded, that synchronized oestrus ewes on receiving PMSG in anoestrus season the ovarial response was more significant than in autumn breeding season.  相似文献

To determine if a transient increase in uterine blood flow (BF) and estradiol-17 beta (E2 beta) secretion occurs during maternal recognition of pregnancy in ewes (as previously observed for sows and cows), 40 nonpregnant (NP) ewes were assigned in equal numbers to surgery on d 9, 11, 13 or 15 postestrus (d 0 = day of estrus). For 20 NP ewes (five/day), each uterine horn (UH) was flushed with saline and uterine flushings (UF) collected. For the remaining 20 ewes, BF was determined for each UH using electromagnetic transducers, and samples of uterine arterial (UA) and uterine venous (UV) blood were obtained from each UH. After an intervening cycle, each ewe was mated, subjected to surgery on the same day postmating as during her previous nonmated cycle, and BF measurements and UA and UV samples were obtained. In addition, each UH of pregnant (P) ewes was flushed and the location of conceptuses was determined. Concentrations of E2 beta and progesterone (P4) in UA and E2 beta in UV and UF were determined by radioimmunoassay. For NP ewes, BF (ml/min) was not different for UH ipsilateral or contralateral to the ovary bearing the corpus luteum (CL), and did not differ across days, averaging 6.5 +/- .4. For P ewes, BF to UH contralateral to the ovary bearing the CL on all days and BF to UH ipsilateral to ovaries bearing CL on d 9 was similar to BF of either UH of NP ewes, averaging 6.8 +/- .6. The objectives of the study were to characterize an ultrasound reproductive tract scoring (URTS) system to assess suitability for breeding in dairy cows, to describe the prevalence of these scores in commercial dairy herds and to examine their relationship with subsequent fertility. Ultrasound examinations (7797) were performed on 5751 Holstein–Friesian cows prior to breeding in 62 seasonally calving herds over 2 years. Data recorded from images of both ovaries and the uterus were combined into a six point scoring system and the prevalence of cows with cystic ovarian follicles and uterine abscesses and adhesions was recorded separately. The prevalence of ovulatory cows which had completed uterine involution (score 1), or had mild (2) or moderate endometritis (3) was 62.5%, 21.7% and 2.2%, respectively. The prevalence of anovulatory cows with moderate endometritis (4), ovulatory cows with pyometra (5) and anovulatory cows which had completed uterine involution (6) was 3.3%, 2.2% and 8.1%, respectively. The interval between calving and examination differed between cows with each of the scores 1, 2, 5 and 6 (61, 46, 53 and 50 days, respectively, p < 0.05) but not between cows with scores 3 and 4 (37 and 35 days, respectively). Ovulatory cows which had completed uterine involution (score 1) had a higher likelihood of pregnancy to first service than ovulatory or anovulatory cows which had not completed uterine involution (p ≤ 0.10). The prevalence of cows with cystic ovarian follicles or abscesses or adhesions of the reproductive tract was 3.9% and 1.2%, respectively. In conclusion, 29% and 11% of cows in seasonally calving and breeding dairy herds had not completed uterine involution or were anovulatory prior to the mating start date, respectively. Both conditions, detected using a URTS system, significantly reduced first service pregnancy rate in these pasture-based dairy herds.  相似文献

Abnormalities of the reproductive tract of female sheep were studied by examining 9970 reproductive tracts from cull ewes and 23,536 tracts from nulliparous sheep (prime lambs) over a period of 12 months in abattoirs in south-west England. Overall, 3.37 per cent of the tracts were pregnant (8.11 per cent of cull ewes, and 1.36 per cent of nulliparous sheep), with a peak incidence between September and December. A total of 655 ewes (6.57 per cent) and 459 nulliparous sheep (1.95 per cent) had acquired abnormalities of the reproductive tract. Within these totals, abnormalities of the ovaries accounted for 3.51 per cent (for the ewes) and 10.68 per cent (for the nulliparous sheep) of all the abnormalities, and abnormalities of the ovarian bursa and uterine tube accounted for 42.1 per cent (for the ewes) and 5.23 per cent (for the nulliparous sheep). In addition, uterine lesions (hydrometra and metritis) accounted for 9.92 per cent (for the ewes) and 13.51 per cent (for the nulliparous sheep); lesions of the cervix and vagina (total of 1.44 per cent) and Cysticercus tenuicollis cysts associated with the reproductive tract (total of 3.05 per cent) were less common. Among the ewes the most common ovarian lesions were ovulation tags, and follicular cysts were the most common in nulliparous animals. Lesions such as bursitis, parametritis and abscesses of the reproductive tract were much more common in cull ewes than in nulliparous sheep, probably having arisen from peripartum infections. Hydrosalpinx and hydrometra, in which the intraluminal fluid was clear, were present at relatively high incidence in nulliparous animals, but not in cull ewes. The proportion of tracts containing macerated fetal remnants (2.14 per cent of all abnormalities in cull ewes) was lower than expected. It was considered that the functional significance of many of the lesions, such as ovulation tags and C tenuicollis cysts, was likely to be low, although in some cases of the latter calcification of the cyst had occluded the uterine tubes. Other lesions, notably hydrosalpinx, bursitis and metritis were likely to have made the affected animals sterile. The acquired abnormalities were therefore more significant in terms of individual animal infertility than as a major cause of infertility in flocks.  相似文献

The local uteroovarian pathway for uterine-induced luteolysis was studied in 34 ewes. Bilaterally ovulating ewes were used to study the role of the main uterine vein (uterine branch of uteroovarian vein) and unilaterally ovulating ewes were used to study the role of the ovarianartery. Surgical operation was done on day 7, 8, or 9 of diestrus and ewes were necropsied on day 20. In 4 bilaterally ovulating control ewes (unilaterally hysterectomized), mean weight of corpus luteum (CL) was less (P smaller than 0.05) on the side of the intact uterine horn (55 mg) than on the side of unilateral hysterectomy (548 mg). In 4 treated ewes (unilaterally hysterectomized and with surgical anastomosis of uterine vein from the intact side to the hysterectomized side), mean weight of CL on the hysterectomized side (153 mg) was less (P smaller than 0.05) than on the uterine-intact side (391 mg). The mean weight of CL on the hysterectomized side in the treated group was not significantly different from that of the uterine-intact side in the control group. Unilaterally ovulating ewes were randomized into 5 groups: (1) controls which were uterine intact, (2) controls in which a unilateral hysterectomy was done ipsilateral to CL, (3) unilateral hysterectomy done ipsilateral to CL and anastomosis of the ovarian branch of the ovarian artery on the uterine-intact side to the ovarian branch of the ovarian artery on the hysterectomized side, (4) controls in which unilateral hysterectomy was done contralateral to CL and the ovarian vascular pedicle on the uterine-intact side was isolated, and (5) unilateral hysterectomy done contralateral to CL, isolation of ovarian vascular pedicle on the uterine-intact side, and anastomosis of the ovarian branch of the ovarian artery on the unilaterally hysterectomized side to the ovarian branch of the ovarian artery on the uterine-intact side. Mean weight of CL was less (P smaller than 0.05) in groups 1 (56 mg), 3 (40 mg), and 4 (120 mg) than in groups 2 (408 mg) or 5 (473 mg). The mean weight of CL was not significantly different among groups 1, 3, and 4 or between groups 2 and 5. Results demonstrate that the local luteolytic effect of the uterus in ewes involves a venoarterial pathway and indicate that the main uterine vein is an adequate uterine outlet and the ovarian artery is an adequate ovarian inlet for the venoarterial pathway without the necessity of other possible concomitant local routes.  相似文献

OBJECTIVE: To evaluate topical application of 1% sodium carboxymethyl cellulose (SCMC) for prevention of postoperative adhesions in a laparoscopic model of uterine trauma in sheep. STUDY DESIGN: Experimental study. ANIMALS: Fourteen non-pregnant ewes. METHODS: Ewes were randomly assigned to 1 of 2 groups: control (saline solution) or 1% SCMC treatment. By left flank laparoscopy, traumatic forceps were used to create serosal trauma (1.5 x 5 cm) and hemorrhage on the left uterine horn. Either 30 mL saline solution or 30 mL 1% SCMC was applied topically to the traumatized uterine horn. Adhesion formation was evaluated by repeat laparoscopy at days 14 and 21. Sheep were euthanatized on day 28 for necropsy evaluation of adhesions. RESULTS: Five control sheep had adhesions of the uterine horn by day 14, but only 4 had adhesions at day 21, and 2 at day 28. Adhesions did not occur in SCMC-treated sheep. No adhesions occurred elsewhere in the abdomen. CONCLUSIONS: Laparoscopically created uterine trauma is an effective method for induction of uterine adhesions, and laparoscopy is an excellent method for serial evaluation of adhesion formation. SCMC (1%) was effective at preventing adhesion formation in sheep and no inflammatory response was noted. CLINICAL RELEVANCE: SCMC (1%) should be considered for prevention of adhesions in abdominal surgery in sheep.  相似文献

An oocyte recovery procedure was developed and evaluated to determine whether a transcervical embryo recovery procedure is feasible with our method, which includes estradiol-17beta (E2) and oxytocin (OT) treatments, for dilating the cervix in ewes. On d 6 of an estrous cycle, oocytes were recovered either transcervically or with a laparotomy procedure. In the laparotomy group, ovulation rate was determined during the procedure and was used to calculate the percentage ofoocytes recovered. The laparotomy procedure was a standard uterine flush, and 12 mL of PBS was used to flush each uterine horn. In the transcervical group, the ovaries in each ewe were evaluated ultrasonically to determine ovulation rate. For transcervical recovery, 100 microg of E2 were injected i.v. on d 5 to increase cervical OT receptors, and 100 USP units of OT were injected i.v. 10 to 12 h later to dilate the cervix. Approximately 25 min after OT, ewes were placed in dorsal recumbency in a Commodore cradle, and a modified Foley catheter was passed through the cervix and into the uterus for injection (80 to 210 mL) and aspiration of PBS. The PBS was aspirated with a vacuum pump. The percentage of PBS recovered was greater (P<.01) at laparotomy than with the transcervical procedure (85.8 vs. 36.2%). Despite that difference, oocyte recovery did not differ significantly between the two groups (67% for laparotomy vs. 50% for transcervical; [oocytes recovered/number of corpora lutea] x 100), and there was no evidence that the transcervical procedure damaged the oocytes; the zona pellucida remained intact around all of the oocytes. In conclusion, a procedure that includes E2-OT-induced cervical dilation, passage of a modified Foley catheter into the uterus, and incremental infusion and aspiration of media through the catheter can be used to recover oocytes transcervically from ewes. This procedure may make transcervical embryo recovery feasible for sheep.  相似文献

Sheep ova are normally obtained from the reproductive tract using the flushing techniques described by Hunter et al. (1955) Cumming, I. A. and McDonald, M. F. 1967. The production of ova by New Zealand Romney ewes following hormonal stimulation. N.Z. Jl agric. Res., 10: 226236. [Taylor &; Francis Online], [Web of Science ®] [Google Scholar]. Oviduct flushes, in the writers' experience, give a higher recovery of ova than uterine flushes, but they result in more adhesions of the tract, which can cause infertility. Reduced pregnancy rates often occur in ruminants grazing endophyte-infected (EI) tall fescue. The objectives were to characterize basal and oxytocin-induced PGF concentrations in serum and reproductive function in ewes fed tall fescue seed and to determine whether addition of fish meal (FM) to a diet of EI fescue would alter PGF production. Ewes were fed a diet with novel or non-toxic endophyte-infected (NE) or EI tall fescue seed containing either corn gluten meal (CG) or FM. Serum concentrations of prolactin, a measure of severity of fescue toxins, were reduced in ewes fed EI compared with NE forage seed (forage × day, P < 0.02) and were greater in NEFM than NECG-fed ewes (P < 0.03). Size and number of corpora lutea (CL), determined by trans-rectal ultrasonography, were similar between diets (P > 0.10). Serum concentrations of progesterone were reduced in ewes with two CL fed EI compared with NE seed (forage × CL number × day, P < 0.001). Oxytocin-induced PGFM concentrations during the luteal phase were determined as a measure of uterine function. On the day of oxytocin administration, peak plasma concentrations of PGFM were reduced in EI compared with NE-fed ewes (forage × time, P < 0.003), but FM did not influence PGFM concentrations. Estrous cycle length was more variable in EI than NE-fed ewes. There appears to be some asynchrony between NE and EI-fed ewes leading to changes in uterine responsiveness to oxytocin. Inclusion of FM did not alter uterine responsiveness to oxytocin.  相似文献

[目的] 评价引进品种肉羊的连续重复超数排卵(超排)、冲胚、胚胎移植等繁育技术集成模式的应用效果。[方法] 于2016—2018年,对选取的1 575只澳洲白(n=230)、白头杜泊(n=228)、黑头杜泊(n=541)、白头萨福克(n=200)、黑头萨福克(n=376)5个品种母羊,进行间隔30 d以上的连续同期发情(孕酮海绵栓)与超排(330~450 IU FSH+200~300 IU PMSG+0.1 mg PG)处理;经过腹腔内窥镜输精后进行子宫角冲胚,回收胚胎后计数并评估质量,挑取部分胚胎进行移植;对比分析品种、季节、重复超排次数等对胚胎生产指标的影响。[结果] 共获得18 496枚胚胎,有14 971枚可用胚;胚胎移植后的受胎率为69.8%,产羔率为93.2%;只均总胚数和只均可用胚数2项指标显示,胚胎生产特性以白头萨福克、黑头萨福克、黑头杜泊较好,澳洲白次之,白头杜泊最差;不同品种母羊连续重复超排冲胚后回收的只均总胚数和只均可用胚数持续增长,直到第4或第5次;澳洲白的超排冲胚效果在春季较好,而其他品种在冬季较好。[结论] 连续重复超数排卵、冲胚技术对于扩繁引进肉羊品种群体是可行的,应根据不同品种及其表现出的季节特性科学、适时应用。  相似文献
