采用磁珠富集法筛选适合漠斑牙鲆遗传多样性分析的微卫星分子标记。筛选共获得43条序列,其中完美型26个,占60.5%;非完美型14个,占32.6%;复合型3个,占6.9%。选取其中14对特异性好且扩增效率高的微卫星引物,对采自美国北卡罗来纳州沿海的漠斑牙鲆野生群体和养殖群体进行遗传多样性及遗传结构比较分析。研究结果表明,12对引物的扩增产物具有多态性,其中7个位点为高度多态(PIC>0.5)。两个群体中共检测到90个等位基因。12个多态性微卫星位点在两个群体中的平均观察杂合度(Ho)和期望杂合度(He)分别为0.36和0.57。9个位点在整个群体中呈现出不同程度的偏离遗传平衡(P<0.05),且偏离平衡的位点均表现为杂合子缺失(Fis>0)。野生群体和养殖群体间的遗传距离为0.1115,群体间的遗传分化微弱(Fst=0.0438)。  相似文献

Three experiments were conducted to evaluate the effects of micro‐bound diets (MBD) on southern flounder larvae. In experiment 1, four MBDs were formulated with different protein sources as follows: MBD 1: herring meal, MBD 2: menhaden meal, MBD 3: menhaden and squid meal; MBD 4: menhaden, squid and herring meal. In experiment 2, four MBDs were formulated as follows: MBD 5: menhaden, squid and herring meal; MBD 6: menhaden, squid, herring and attractants; MBD 7: menhaden, squid, herring and casein, and MBD 8: menhaden, squid, herring, casein and attractants. In experiment 3, three groups were maintained as follows: Group 1: live feed; Group 2: co‐fed with MBD 6; and Group 3: MBD 6. In experiment 1 on 35 dph, survival and body weight (BW) of the fish fed MBD 4 was significantly higher than the MBDs 1 and 2. In experiment 2 on 34 dph, fish fed MBD 6 had significantly higher BW than the commercial microdiets. In experiment 3 on 21 dph, fish receiving only MBD had significantly lower survival than the other groups. Growth, survival and larval fatty acid composition suggested that co‐feeding MBD 6, a mixture of marine protein sources plus attractants was more effective than the other MBDs.  相似文献

The inclusion of soybean meal and soy concentrates in finfish feeds may introduce endocrine disrupting compounds in the form of phytoestrogens. Genistein, which may dually act as an oestrogen receptor agonist/antagonist, has been shown to elicit both masculinizing and feminizing effects in several teleost species. The objective of this investigation was to examine the effects of dietary genistein administration on reproductive development, growth and survival in southern flounder (Paralichthys lethostigma). Southern flounder received one of three experimental diets which contained varying concentrations of genistein (control – 0, low dose –100 and high dose –1000 mg genistein kg?1 feed) from 84 to 153 days post hatch (DPH). Fish remained in culture until 285 DPH at which time sex ratios were determined. Growth and survival were evaluated at 153 and 285 DPH. Greater numbers of female fish resulted from the low and high dose genistein treatments (98 ± 2% and 78 ± 12% respectively), as compared to the control group (21 ± 10%). Reduced growth at 153 DPH and poor survival (28%) at 285 DPH were also observed for the high dose genistein treatment. Collectively, these results demonstrate the feminizing effects of genistein in southern flounder, but reduced growth and survival with high dietary administration.  相似文献

A feeding trial with five crude protein levels (549, 513, 472, 445 and 399 g kg?1) was conducted to investigate the optimum protein level in diets of juvenile southern flounder (Paralichthys lethostigma). Budgets of nitrogen and energy were discussed. Fish (initial weight 32.9 ± 0.5 g fish?1, mean ± SD) were fed the experimental diets to satiation twice daily for 61 days. Protein levels affected specific growth rate in wet weight (SGRW) and protein (SGRP) significantly. SGRW and SGRP were highest at 512.5 g kg?1 protein level. SGRW was positively correlated to growth nitrogen (GN), growth energy (GE), nitrogen digestibility, energy digestibility, amount of digestible nitrogen and amount of digestible energy. Faecal nitrogen (FN) and faecal energy (FE) were affected significantly with trends contrary to SGRW. The nitrogen budget was described by the equation 100CN = 2.1FN + 34.4UN + 63.5GN (CN, nitrogen intake; UN, excretion nitrogen). The energy budget was 100IE = 4.04FE +3.32UE + 54.35GE + 38.30ME (IE, gross energy intake; UE, excretion energy; ME, metabolizable energy). The average proportion of GE and ME in assimilated energy (AE) was described by the equation 100AE = 58.65GE + 41.35ME.  相似文献

In Sendai Bay, stone flounder larvae settle and spend their juvenile period in either shallow exposed inshore nursery grounds or estuarine nursery grounds. The purpose of this study is to examine the relative contributions of these two kinds of nursery grounds to the flounder population using otolith strontium:calcium ratios. Stone flounder juveniles were collected from both nursery grounds, and one- and two-year-old flounder were caught deeper in Sendai Bay. Sr and Ca content in the otoliths were measured by electron probe micro analysis. The Sr:Ca ratios in the otolith section corresponding to the early postsettlement period ranged from 3.06 to 3.85 for the exposed inshore areas with stable low temperature and high salinity conditions, and from 3.81 to 5.32 in brackish estuaries with high temperature and low salinity conditions but with large diel and tidal cyclical fluctuations. Values from an estuarine site with stable salinity ranged from 3.58 to 4.15 overlapping with both the above ranges. Rearing experiments supported our inference that the high otolith Sr:Ca ratios of juveniles inhabiting estuarine nursery grounds are attributable to higher temperature and physiological stress caused by the large diel temperature and salinity fluctuations within the estuaries. Estimation of the Sr:Ca ratio of recruited fish using the otolith section formed while in the nursery area showed that at least 20 out of 42 individuals examined originated from estuarine nursery grounds. The present study indicates that estuaries play an important role as nursery grounds for stone flounder, producing about half of the stock in spite of the small and restricted area compared with the wide expanse of the exposed inshore area.  相似文献
