Objective To determine whether a drug detected in the blood or urine of a racing animal could have penetrated through the skin from a topically applied preparation.
Design Blood and urine of dogs and horses were analysed after topical administration of three common nonsteroidal anti-inflammatory preparations.
Experimental method Dimethylsulphoxide was analysed using a gas chromatograph with a flame photometric detector. Phenylbutazone, its metabolites and lignocaine were analysed using a gas chromatograph with a mass selective detector.
Results Dimethylsulphoxide, phenylbutazone and ligno-caine were detected in dog urine after muliple applications of the preparations. The maximum concentration of dimethyl-sulphoxide in dog urine correlated with the concentration of dimethylsulphoxide in the preparation. Phenylbutazone penetrated the skin more effectively from the cream than from the solution or gel preparations. This penetration was independent of the concentration of dimethylsulphoxide.
Conclusion The superior penetration of phenylbutazone from the cream can be explained by it being present as a neutral molecule in an hydrophobic medium. It is proposed that phenylbutazone penetrates the skin of greyhounds most effectively by a hydrophobic lipid route which is likely to be different from the path by which dimethylsulphoxide penetrates the skin.  相似文献

A total of 292 field investigative reports of drug residues in food animals for 1983 to 1988 were analyzed. The investigations had been conducted by the Food and Drug Administration (FDA) and the Virginia State Veterinarian's Office, in cooperation with the Center for Veterinary Medicine of the FDA, to trace residues reported by the USDA Food Safety and Inspection Service to the source of the animal and the administration of the drug. The analysis disclosed the following. (1) Antibiotic residues were most often associated with streptomycin, penicillin, oxytetracycline, and neomycin. Sulfamethazine was, by far, the most frequently cited sulfonamide. (2) Residues are being found predominantly in cows, veal calves, and market hogs (barrows and gilts). (3) The cause of drug residue most frequently cited by the field investigators was failure to observe the withholding time for the drug. Almost half of these investigations revealed that the individual responsible for the sale of the animal did not know the proper withholding time for the drug. Failure to maintain adequate records was also a contributing factor. (4) The producer was considered to be the responsible party in over 80% of the cases for which responsibility was determined. (5) Residues associated with injectable drugs were investigated most frequently. Long-acting and sustained-release products were most often associated with penicillin and oxytetracycline residues. (6) The 2 most common sources of purchase for the drugs involved in the investigations were the feed/farm supply store and the veterinarian. (7) Unapproved drug use was not a major cause of residues.  相似文献

A screening and confirmation procedure for drugs and metabolites in the blood serum and urine of racing animals was developed. Equine blood serum was spiked with low concentrations of several drugs of interest. Canine blood serum and urine were collected following oral doses of diethylcarbamazine, procaine, and phenylbutazone. Serum, urine, and extracts of each were analyzed, using a triple quadrupole mass spectrometer. Simultaneous screening of up to 50 drugs was possible in a single sample, in less than 2 minutes. Detection limits for most compounds were in the ng/ml to microgram/ml range, using 1-microliter samples. This procedure provided fast, sensitive screening for selected drugs and metabolites in blood serum and urine.  相似文献

The objective of the study was to determine if there were adverse effects on animal health and performance when a range of ruminant animal species were fed at least 10 times the maximum permitted European Union (EU) Se dietary inclusion rate (0.568 mg of Se/kg of DM) in the form of Se-enriched yeast (SY) derived from a specific strain of Saccharomyces cerevisiae, CNCM I-3060. In a series of studies, dairy cows, beef cattle, calves, and lambs were offered a control diet that contained no Se supplement or a treatment diet that contained the same basal feed ingredients plus a SY supplement that increased total dietary Se from 0.15 to 6.25, 0.20 to 6.74, 0.15 to 5.86, and 0.14 to 6.63 mg of Se/kg of DM, respectively. The inclusion of the SY supplement increased (P < 0.001) whole-blood Se concentrations, reaching maximum mean values of 716, 1,505, 1,377, and 724 ng of Se/mL for dairy cattle, beef cattle, calves, and lambs, respectively. Seleno-methionine accounted for 10% of total whole-blood Se in control animals, whereas the proportion in SY animals ranged between 40 and 75%. Glutathione peroxidase (EC 1.11.1.9) activity was greater (P < 0.05) in SY animals compared with controls. A range of other biochemical and hematological parameters were assessed, but few differences of biological significance were established between treatment groups. There were no differences between treatment groups within each species with regard to animal physical performance or overall animal health. It was concluded that there were no adverse effects on animal health, performance, and voluntary feed intake with the administration of at least 10 times the EU maximum, or approximately 20 times the US Food and Drug Administration permitted concentration of dietary Se in the form of SY derived from a specific strain of Saccharomyces cerevisiae CNCM I-3060.  相似文献

Trimethoprim (TMP) was administered in combination with either sulphadiazine or sulphadimidine to broilers, and plasma concentrations were determined simultaneously by newly developed thin-layer and/or high-performance liquid-chromatographic procedures, which also allowed quantification of the N4-acetyl metabolites of the sulphonamides. After i.v. injection of TMP (20 mg/kg body wt) and sulphadiazine (100 mg/kg body wt), both compounds were rapidly eliminated from plasma with half-lives of 1 and 2.7 h, respectively. Apparent volumes of distribution (3.3 and 0.96 l/kg, respectively) indicated that the tissue distribution of TMP was more extensive than that of the sulphonamide. After oral administration of the same dosages, elimination appeared to be slower compared to the i.v. injection, but this was obviously related to delayed absorption. Bioavailability after oral administration was approximately 100% of sulphadiazine, but only about 60% for TMP. Oral dosing of TMP in combination with sulphadimidine yielded similar maximum plasma concentrations of both compounds to those obtained with the combination of TMP with sulphadiazine, but the plasma concentration decline of sulphadimidine appeared to be more rapid than that of sulphadiazine after oral administration. During prolonged administration of different dosages of TMP-sulphadiazine combinations via drinking water, only low plasma concentrations were attained by the recommended dosage of the combination. Up to 10-fold higher dosages were tolerated by the animals without side-effects. In view of the fact that the sensitivity of bacterial strains to TMP-sulphonamide combinations differs widely, the plasma concentrations determined in the present study during prolonged drinking-water medication with different dosages of a TMP-sulphadiazine combination can be used to select effective doses for treatment of different poultry diseases.  相似文献

Dimethyl sulfoxide (DMSO) had been postulated to be a 'masking agent' when used concurrently with therapeutic or prohibited drugs in racing animals. Eight drugs (flunixin, furosemide, caffeine, apomorphine, phenylbutazone, lidocaine, cocaine, and acepromazine maleate) were administered to six horses singly and with concurrent intravenous DMSO. Urine samples were analyzed for the presence of the drugs and/or their metabolites by thin layer chromatography. Direct comparison of thin layer chromatograms of extracts of positive urine samples with and without DMSO verified that DMSO did not interfere with the detection of these drugs.  相似文献

Levels of erythromycin in the blood and organs of rabbits, rats and mice were determined after the i. m. implantation of the drug Erythromycin inj. ad usum vet. at doses of 10 mg kg-1 live weight (rabbits), and/or 25 mg kg-1 (rats) and 1 mg pro toto--approximately 40 mg kg-1 (mice). A diffusion plate method and the germ Sarcina lutea CCM 552 were used to find out the antibiotic in samples. Sensitivity of the method used was 0.025 microgram ml-1 (g). In rabbits the level of the antibiotic was maintained in the blood and examined organs and tissues (lungs, liver, kidney, muscles and the wall of the small intestine) by the above dose for more than 12 hours--except liver. The values obtained for the organs and tissues were mostly higher than those obtained for the blood. The highest levels were found in the kidneys and lungs. Neither did the dose of 20 mg kg-1 l. w. maintain the significant level of erythromycin in the blood serum in the course of 24 hours. The results obtained in rats and mice almost coincided with the distribution of erythromycin in the rabbit organisms, and the affinity of this antibiotic with the pulmonary tissue was proved.  相似文献

The purpose of this report is to describe the results of epidemiological surveys of racing-related fractures in Thoroughbred horses in Japan. In the period 1987-2000, a total of 10,203 fractures were diagnosed in 556,705 runners, resulting in an overall incidence of 1.83%. The annual incidence of fractures in flat racing during the 14-year period fluctuated between 1.44% and 2.19%. The majority of fractures affected the forelimbs. We found significant effects of track condition on injury incidence. The incidence of fractures decreased as track conditions on turf became softer and increased as track conditions on dirt became muddier. Because of the general trend for the incidence of fractures to vary with track condition, we examined the relationship between track conditions and racing times for winning horses in 4117 races and for 50,564 overall runners for the period 1990-1994 on two tracks. For turf courses, racing times became longer as track conditions became softer. In contrast, for dirt courses, racing time tended to become shorter in muddier conditions. The variation in the incidence of fracture with track condition might be due to the corresponding difference in racing time.  相似文献
