Vitellaria paradoxa C. Gaertn. (shea butter tree) is an indigenous African tree species that is widely distributed in the dry areas of northern and eastern Uganda. The species is widely known for its oil which is used in cooking, cosmetics and traditional medicine. Local folk classification recognises the presence of different ethno-varieties on the basis of fruit and nut characters. In the present study, 176 trees representing 44 ethno-varieties from three farming systems of Uganda were assessed to determine the patterns of morphological variation and establish the congruence between morphological variation and folk classification. The results show high variation in pulp weight (CV = 35.9 %), stem diameter (CV = 28.48 %), fruit weight (CV = 27.81 %) and canopy diameter (CV = 26.69 %). There was a strong positive correlation between pulp and fruit weight (r = 0.963, p < 0.001), leaf length and leaf width (r = 0.652, p < 0.001) and between petiole length and leaf length (r = 0.788, p < 0.001). There was no underlying quantitative morphological structuring among the 44 ethno-varieties. Hierarchical cluster analysis using quantitative morphometric data produced three groups without clear aggregation based on ethnographic or geographic separation. However, a combination with qualitative traits as perceived by farmers provided good congruence with folk classification. Quantitative morphological data alone does not resolve any discrete forms of V. paradoxa that are related to folk classification. There is need to utilise biochemical and molecular markers to unravel the underlying variation for use in selection and improvement of shea butter tree ethno-varieties.  相似文献

The effect of the addition of Lactococcus lactis subsp. lactis INIA 415, a strain harboring the structural genes of nisin Z and lacticin 481, on the formation of volatile compounds in Hispánico cheese manufactured with a mesophilic starter or with the mesophilic starter and a thermophilic starter was investigated. Addition of bacteriocin-producing L. lactis subsp. lactis INIA 415 to milk enhanced the formation of 2-methyl-propanal, 2-methylbutanal, 3-methylbutanal, 2-methyl-1-propanol, 3-methyl-1-butanol, 1-octanol, 2-butanone, and 2,3-butanedione. On the other hand, addition of thermophilic starter enhanced the formation of acetaldehyde, ethanol, 3-methyl-2-buten-1-ol, ethyl butanoate, ethyl hexanoate, 2-butanone, and 2,3-butanedione in Hispánico cheese. Stepwise discriminant analysis using the relative abundances of volatile compounds classified cheeses by type of starter, with function 1 related to thermophilic starter and function 2 to bacteriocin producer.  相似文献

Lactococcus lactis subsp. lactis INIA 415, a strain harboring the structural genes of bacteriocins nisin Z and lacticin 481, was used as adjunct culture in the manufacture of Hispánico cheese with a mesophilic starter and a thermophilic starter of high aminopeptidase activity. Addition of the bacteriocin producer promoted early lysis of mesophilic and thermophilic starter bacteria. Extracellular aminopeptidase activity in 7-day-old cheese made using mesophilic and thermophilic starters plus bacteriocin producer was 3.0-fold the level reached in cheese made without the bacteriocin producer. Proteolysis in cheese made with mesophilic and thermophilic starters plus bacteriocin-producing adjunct culture after 25 days of ripening was 1.5-fold the level reached in cheese made without the bacteriocin producer, and the level of total free amino acids was 2.9-fold the level found in cheese made without the bacteriocin producer. Cheese made with mesophilic and thermophilic starters plus bacteriocin producer received the highest scores for flavor quality and flavor intensity and reached in 25 days the flavor intensity score of a 75-day-old cheese made without the bacteriocin producer.  相似文献

The lacZ gene from Lactobacillus delbrueckii subsp. bulgaricus DSM 20081, encoding a β-galactosidase of the glycoside hydrolase family GH2, was cloned into different inducible lactobacillal expression vectors for overexpression in the host strain Lactobacillus plantarum WCFS1. High expression levels were obtained in laboratory cultivations with yields of approximately 53000 U of β-galactosidase activity per liter of medium, which corresponds to ~170 mg of recombinant protein per liter and β-galactosidase levels amounting to 63% of the total intracellular protein of the host organism. The wild-type (nontagged) and histidine-tagged recombinant enzymes were purified to electrophoretic homogeneity and further characterized. β-Galactosidase from L. bulgaricus was used for lactose conversion and showed very high transgalactosylation activity. The maximum yield of galacto-oligosaccharides (GalOS) was approximately 50% when using an initial concentration of 600 mM lactose, indicating that the enzyme can be of interest for the production of GalOS.  相似文献

Narcissus cv. “Garden Giant” bulbs were grown in N, P, K, Ca, Mg, or Fe-deficient solutions and compared with bulbs grown in the control solution containing all these nutrients. Plants were sampled at 4 stages: (I) at planting, (II) at sprouting, (III) after flower senescence and/or after visible deficiency symptoms developed, and (IV) at lifting. Observation of visible deficiency symptoms showed that leaves of narcissus displayed chlorosis in the —N, —Mg, and —Fe treatments, while roots were most susceptible to Ca-deficient conditions. Root tips in the —Ca treatment showed brown in followed by root rot. In the —N treatment, shoot growth was markedly retarded and leaves were small and yellow. On the other hand, visible deficiency symptoms were not evident in the —P treatment except for early senescence. K deficiency symptoms were also not evident. Narcissus flowers were not affected by the mineral deficiencies and mineral contents in full-bloom flowers were not different between samples.

In the control plants, there was a large accumulation of N in the roots at sprouting and the content decreased thereafter. Large amounts of K accumulated in roots more than in any other organs. On the other hand the Ca content was high in the tunic and Ca in the scales was hardly mobilized thoughout the growth period. A large proportion of each element eventually accumulated in new inner scales whereas only a small fraction in the old outer scales. The -N, -Ca, and -Mg treatments severely depressed dry matter accumulation, unlike the -P, -K, and -Fe treatments. The -N treatment did not affect the concentration of other minerals, but the -P treatment tended to decrease N, K, Ca, and Mg concentrations. On the other hand, the -K treatment increased Ca and Mg concentrations. -Ca caused an increase in Mg concentration and -Mg raised Ca concentration. These results may be due to compensation effects of cation absorption for the maintenance of the cation/anion balance.  相似文献

The equilibrium adsorption and binding of the active toxin from Bacillus thuringiensis subsp. kurstaki on complexes of montmorillonite–humic acids–Al hydroxypolymers, as well as the biodegradation and the insecticidal activity of the bound toxin, were studied. Seventy percent of the total adsorption occurred within the first hour, and maximal adsorption occurred in <8 h. Adsorption of the toxin on a constant amount of the complexes increased as the amount of the toxin added increased, and equilibrium adsorption isotherms of the L-type were obtained. There was essentially no desorption of the toxin after extensive washing of the toxin–organomineral complexes with double distilled H2O and 1 M NaCl. The bound toxin was resistant to utilization by mixed microbial cultures from soil and to enzymatic degradation by Pronase E. Free and bound toxin were active against the larvae of Manduca sexta; the bound toxin retained the same activity after exposure to microbes or Pronase, whereas the toxicity of the free toxin decreased significantly. The results of these studies indicate that the release of transgenic plants and microorganisms expressing truncated genes that encode active insecticidal toxins from B. thuringiensis could result in the accumulation of these toxins in soil as a consequence of binding on surface-active soil particles. This persistence could pose a hazard to nontarget organisms, enhance the selection of toxin-resistant target species, and increase the control of target insect pests.  相似文献

The concentrations of mercury, lead, cadmium, and arsenic were evaluated in 96 samples, 12 by each one of the following eight fish species: snook (Centropomus undecimalis), crevalle jack (Caranx hippos), Serra Spanish mackerel (Scomberomorus brasiliensis), southern red snapper (Lutjanus purpureus), blue runner (Caranx crysos), Atlantic tarpon (Megalops atlanticus), ladyfish (Elops saurus), and Atlantic goliath grouper (Epinephelus itajara), which were collected during 1 year in the Atrato River Delta in the Gulf of Urabá, Colombian Caribbean. Three fish were caught from each of the following sites the community usually uses to catch them (known as fishing grounds): Bahía Candelaria, Bahía Marirrío, Bocas del Roto, and Bocas del Atrato. The quantification of metals was performed by microwave-induced plasma-optical emission spectrometry. The Pb concentration fluctuated from 0.672 to 3.110 mg kg?1, surpassing the maximum permissible limit (MPL?=?0.3 mg kg?1) for human consumption for all species. The Hg concentration ranged between < Limit of detection and 6.303 mg kg?1, and in the crevalle jack and Atlantic tarpon, concentrations exceeded the MPL (0.5 mg kg?1). The levels of Cd and As were not significant in the studied species and did not exceed the MPL (0.05 mg kg?1).  相似文献

The aim of the study was to investigate how the quality of snow varies within a city. The study area is situated in the Luleå municipality, in northern Sweden (lat. 65°35′N and long. 22°10′E). For this study, the city was divided into three different types of areas: housing areas, city centre and outskirts. In one area of each type, sampling sites were selected. Snow samples were taken at nine occasions, once every fourteenth day in the city centre and in the housing area. Analyses were carried out for pH, conductivity, suspended solids and both the total and dissolved concentration of phosphorus and selected metals. It was concluded that traffic or activities related to traffic were a major source of heavy metals and phosphorus in urban snow, both in the city centre and in the housing area. Also, the type of area and the design of the street were important for the quality of snow. A clear relation was found between the pH and the quantity of suspended solids in the snow. Higher pH values were found in snow samples from sites with higher traffic loads, and lower pH values at sites, which were the no-traffic sites, with small quantities of particles. The particulate and dissolved substances in the snow behaved in different ways.  相似文献

Abstract

The rationale for this experiment was to determine forage nutrient concentrations as affected by biosolids fertilization. We studied the effects of single applications of two exceptional quality biosolids to bahiagrass (Paspalum notatum) pasture with regard to satisfying beef cattle nutrient requirements. Twenty‐five 0.8‐ha pastures were divided into five blocks. Two biosolids were applied as normal and double agronomic rates. The control plot received NH4NO3. Forages were analyzed for calcium (Ca), phosphorus (P), magnesium (Mg), potassium (K), sodium (Na), crude protein (CP), and in vitro organic matter digestibility (IVOMD), and soils were analyzed for Mehlich I extractable Ca, P, Mg, and K. Single (agronomic or twice this) applications of biosolids to pastures had little effect on Ca, P, Na, and K forage concentrations, but forage Mg was elevated in several treatments late in the season. Crude protein concentrations were elevated above the control for all biosolids treatments late in the season, whereas only small differences were observed at early sampling times. Trends were similar for IVOMD. In general, all treatments were associated with soils with adequate Ca, P, and Mg concentrations, while soil K was uniformly low. In relation to grazing beef cattle requirements, all treatments resulted in generally adequate forage levels of Ca, P, Mg, K, CP, and IVOMD, however, Na (<0.06%) was deficient.  相似文献

Respiration and mineralisation of N were measured in a set of contrasting soils that had either been autoclaved, air-dried, fumigated (with chloroform or methyl bromide) or exposed to gamma radiation. The soils used were a manured and an unmanured arable soil, an acid and a neutral woodland soil, an arable sandy soil and an organic soil under grass. With the exception of the acid woodland soil, the flushes of decomposition (i.e. the increases in O2 consumption, CO2 evolution and N mineralisation that occurred when the treated soil was inoculated and incubated for 10 days) were in the order: air-drying < CH3Br ? CHCl3 < irradiation < autoclaving. All of the treatments, except air-drying, decreased the ratio (C mineralised after treatmcnt)/(N mineralised after treatment). All of the treatments increased the amount of 1N K2SO4 extractable organic C, autoclaving causing by far the greatest increase.Neither of the fumigants increased respiration in the acid soil over the whole 10 day period, although N mineralisation was slightly increased. Irradiation, air-drying and autoclaving did, however, produce a flush in the acid soil, the order being: irradiation < air-drying < autoclaving. A soluble substrate, extracted from yeast cells by ultrasonic disintegration, decomposed to about the same extent in neutral and in acid soil. When 14C labelled glucose was added to the acid soil and incubated for 52 days, the retention of labelled C was slightly greater (31·6%) than in a comparable near-neutral soil (28·8%). However, the flush that followed fumigation of the acid soil was only half that in the near-neutral soil, suggesting that less biomass is formed under acid conditions. Liming increased the size of the flush in an acid soil.For soils from the same field but under different management, the size of the flush caused by CHCl3 is in the order: grassland > cropped arable > bare fallow. The flush is much more sensitive to differences in soil management than is the total amount of soil organic matter; a fallowed soil lost half its organic C in 10 yr whereas the increase in respiration that followed fumigation fell to one-seventh its original value. Two Nigerian soils behaved similarly; a soil that had been 2 years under cultivation contained only 16% less total organic C than an adjacent soil still under secondary forest, yet the flush in the cultivated soil was half that in the forest soil. The amount of substrate metabolised during the flush is thus very sensitive to changes in soil management that alter the amount of fresh organic matter entering the soil each year.  相似文献

Purpose

This work studies the implications of different traffic patterns for heavy metal and solid pollution generation processes following rainfall events with contrasting antecedent meteorological conditions, at a periurban catchment. The aim is to provide information on the pollution processes and their potential environmental impacts for urban areas.

Materials and methods

Seven campaigns were performed covering winter, spring, and summer conditions, for rainfall events with different antecedent conditions. Four types of roads were monitored: low traffic, average traffic, heavy traffic with demanding driving situations (break and turning), and heavy traffic with high vehicle speed (motorway profile). Samples were taken at the beginning, middle and end of the events to measure within event variation in concentration. Analytical standard procedures were used to quantify pH, conductivity, turbidity, total solids, volatile solids, suspended solids, volatile suspended solids and heavy metals (Cd, Cu, Pb and Zn) in the total and dissolved forms (as to infer the particulate fraction), namely copper, zinc, cadmium and lead.

Results and discussion

The collected data show a direct relation among the number of vehicles and/or the driving manoeuvres performed by them and the amount of solids and heavy metals present in the wash out overland flow collected. An important fraction of the heavy metals is washed off in the particulate form, which represents an increased problem since the road overland flow is directed to green/brown areas and for the local aquatic ecosystems. Maximum copper values recorded exceed 0.6 mh L?1, zinc exceeds 5 mg L?1, lead 0.1 mg L?1 and cadmium 0.01 mg L?1. Values are higher after long dry spells and reduce concentration throughout the rainfall events.

Conclusions

An important part of the heavy metals (with relevance for zinc and lead) are washed off in the particulate form, pollutants are typically related to the amount of traffic, and especially to the existence of driving manoeuvres. The summer events show the highest values, due to the accumulation of pollutants during the long dry spells.
