An enzyme-linked immunosorbent assay (ELISA) for detection of Bordetella avium infection in turkey poults was developed. One-week-old poults challenged intratracheally with 10(12) colony-forming units of B. avium had detectable titers (greater than or equal to 11), with an average of 13.6% positive samples when the birds were 6 to 11 weeks old. The method was sensitive enough to detect maternal antibodies to B. avium in poults up to 3 weeks of age. The same poults challenged at 1 week of age had 100% tracheal infection up to 3 weeks of age, which dropped to 0% by 6 weeks. The method resulted in no false-positive samples (titer = 0) from birds not infected with B. avium and tested weekly between 4 and 11 weeks of age. Antibodies in turkey flocks infected with Newcastle disease virus, hemorrhagic enteritis virus, and Mycoplasma meleagridis, and birds infected with Escherichia coli had no apparent cross-reactivity to the B. avium antigens used in the ELISA. The percentages of B. avium-positive serum samples collected from different turkey flocks did not significantly differ (P greater than 0.05) when samples were tested by the developed ELISA at different times, an indication of the reproducibility of the method.  相似文献

The role of pili in protection against Bordetella avium infection in turkey poults was studied. An isolate that produced the largest number of pili under growth conditions developed in our laboratory was used for preparation of pili and bacterin and for challenge. The pili were isolated, purified, examined by electron microscopy, and tested for purity by gel electrophoresis. Poults were vaccinated with oil-adjuvant pili, formaldehyde- or merthiolate-inactivated bacterins, or a commercial bacterin. Poults were vaccinated once or twice subcutaneously at different ages and challenged intranasally with a pathogenic B. avium isolate 5 days following the last vaccination. A few vaccinated birds had very mild clinical signs. B. avium was isolated from the sinuses of a few vaccinated birds, and growth was scanty. The mean colony counts from tracheal sections was significantly higher (P less than 0.1) in unvaccinated challenged poults than in vaccinated challenged poults. It is postulated that B. avium pili are important immunogens in turkey poults.  相似文献

Convalescent serum given to 1-day-old poults delayed clinical signs of turkey coryza by several days and reduced mortality on infected farms. Turkey breeders immunized with cell-culture-adapted infectious bursal disease virus (IBDV) or turkey infectious bursal disease virus (TIBDV) had a marked increase in virus-neutralization (VN) antibody titers. The VN antibody titer was significantly higher in progeny poults than in poults from unimmunized breeders. Clinical turkey coryza and mortality was considerably less in poults from IBD- or TIBD-vaccinated breeders than in control poults. They also responded more favorably to hemorrhagic enteritis and fowl cholera vaccination.  相似文献

Groups of white leghorn hens were vaccinated twice with a Mycoplasma gallisepticum (MG) bacterin, once with bacterin, or left unvaccinated. Four weeks after vaccination, they were challenged with virulent R strain MG. Egg production was significantly higher in challenged vaccinated groups than in the challenged control group. Four challenged control hens went out of production, whereas only one twice-vaccinated hen did. MG was first isolated directly from eggs 5 days postchallenge (PC) in twice-vaccinated hens, 4 days PC in once-vaccinated hens, and 2 days PC in controls, and the hens continued to lay positive eggs till the end of the experiment 7 weeks PC. MG was found in 17.65%, 38.55%, and 45.90% of eggs cultured in twice-vaccinated, once-vaccinated, and control groups, respectively. Nine of 16 twice-vaccinated hens were found to be shedding MG through their eggs, whereas 15 of 17 once-vaccinated hens and 14 of 16 controls were shedding MG through their eggs.  相似文献

Poults 3 weeks and older developed temporary tracheal resistance to intranasal challenge following inoculation of either Artvax vaccine or formalin-inactivated Bordetella avium bacterin by the intranasal and eyedrop routes. Resistance usually persisted for 3-4 weeks after B. avium challenge. However, with constant exposure to infected controls, the vaccinated birds eventually developed tracheal infection. Day-old poults did not respond to either the Artvax or the bacterin and were completely susceptible to challenge. Two-week-old poults responded to some degree, but poults 3 weeks old and older responded best. Poults inoculated with bacterin by aerosol or by drinking water did not respond as well as those inoculated by the intranasal and eyedrop routes. When poults were given a single subcutaneous injection at 3 weeks of age and challenged 2 weeks later, three of five resisted infection for 18 days.  相似文献

The efficacy of Salmonella enteritidis (SE) oil-emulsion bacterin (a commercially available vaccine) was evaluated in an intravaginal challenge model in hens producing a high rate of SE-contaminated eggs. Hens were vaccinated at 38 wk of age. A second (booster) bacterin injection was administered 4 wk later. Two weeks after the second vaccination, all hens were challenged intravaginally with 10(7) colony-forming units of SE. After challenge, 36 of 189 eggs (19.0%) in the vaccinated hens were positive for SE, and this contamination rate was significantly (P < 0.01) lower than that in the unvaccinated hens (61 of 165 eggs, 37.0%). SE was highly recovered from the cloacal and vaginal swabs of the unvaccinated and vaccinated hens, but the number of SE from the cloaca of the vaccinated hens was significantly (P < 0.05) lower than that in the unvaccinated hens at 7 days post-challenge (PC). The recoveries of SE from the spleen and ovary in the vaccinated hens were significantly (P < 0.05) lower than those in the unvaccinated hens at 7 days PC. At necropsy, SE was recovered from 2 of 15 forming eggs (13.3%) taken from the oviducts of the unvaccinated hens, whereas no SE was recovered from 17 forming eggs in the vaccinated hens. After vaccination, serum antibodies for SE in the vaccinated hens were significantly higher than those in the unvaccinated hens. Antibodies from the oviductal washing, especially immunoglobulin G isotype, in the vaccinated hens were higher than those in the unvaccinated hens after challenge. This intravaginal challenge model produced frequent contaminated eggs and clearly demonstrated the ability of the bacterin to protect against egg contamination. The present model may be a useful tool for further studies to evaluate the protective effect against SE contamination of eggs by potential vaccine candidates.  相似文献

The California poultry industry experienced an outbreak of H6N2 avian influenza beginning in February 2000. The initial infections were detected in three commercial egg-laying flocks and a single noncommercial backyard flock but later spread to new premises. The vaccination of pullet flocks with a commercially prepared, killed autogenous vaccine prior to their placements on farms with infected or previously infected flocks was used as a part of the eradication programs for some multiage, commercial egg production farms. The purpose of this study was to follow three vaccinated flocks on two commercial farms to track the immune responses to vaccination. The antibody-mediated responses of the three flocks followed in this study were markedly different. One flock achieved 100% seroconversion at 12.5 wk of age, but by 32 wk of age, all of the hens were seronegative by agar gel immunodiffusion (AGID). In contrast, at 32 wk of age, flocks from the other farm (flocks 2A and 2B) were 95% and 72% seropositive by AGID, respectively. Of the differences that were identified between the vaccination protocols on the two farms, the distinction that could explain the level of disparity between responses is the delivery of the second dose of vaccine with a bacterin on the first farm, which may have interfered with the persistence of immunity in this flock. Hens from flocks 2A and 2B were experimentally challenged at 25 wk of age with H6N2 avian influenza virus. Hens from flock 2A did not transmit virus to naive contact-exposed hens, but hens from flock 2B did. At 34 wk of age, hens from flock 2A were again challenged and naive contact-exposed hens were infected in this second trial. These challenge experiments served to demonstrate that despite detectable antibody responses in flocks 2A and 2B, the birds were protected from infection for less than 21 wk after the second vaccination.  相似文献

Electrocardiograms (ECGs) were recorded from turkey poults infected with the W isolate of Bordetella avium. Strip chart data (amplitudes and intervals) were measured at 7, 14, 21, and 28 days postinoculation and compared with data from uninoculated controls. B. avium infection altered P-, RS-, and T-wave amplitudes, as well as P-R and S-T intervals throughout the 4-week experimental period. Heart rates were similar over the 4-week period in control poults, while rates in the infected birds were variable. Alterations in ECGs associated with B. avium infection appeared to be the result of disturbances in the cardiovascular system and may be associated with the pathogenicity of the organism in turkey coryza.  相似文献

Six hours post-hatch, large white turkey poults were inoculated intranasally with 5 x 10(7) cells of the W isolate of Bordetella avium. Three hours after inoculation and subsequently on days 7, 10, 14, 21, and 28 postinoculation, poults from infected and control groups were killed by cervical dislocation. Thymus, spleen, and bursa of Fabricius were removed, weighed, and frozen until assayed for norepinephrine (NE), dopamine (DA), and serotonin (5HT). B. avium infection caused a reduced concentration of NE, DA, and 5HT in the spleen, thymus, and bursa of Fabricius. The reduced concentrations of these monoamines in lymphoid tissues of diseased poults may be a normal response during the course of a disease or during the mounting of an immune response.  相似文献

Leghorn hens vaccinated twice with an inactivated Mycoplasma gallisepticum (MG) bacterin before egg production and subsequently challenged with virulent MG were protected against transmission of MG through the egg. Unvaccinated control hens transmitted MG through the egg at a high rate. When unvaccinated hens were vaccinated with MG bacterin 2 weeks after challenge with MG, there was no significant decrease in egg transmission. Hens vaccinated twice before laying did not suffer as severe egg-production drops as unvaccinated hens did when challenged with virulent MG. In a natural MG outbreak in a leghorn breeder flock, 50 hens were separated from the remainder of the flock and vaccinated with MG bacterin approximately 3 weeks after initial exposure. The unvaccinated hens transmitted MG through the egg at a rate three times higher than the rate of transmission of the vaccinated hens.  相似文献

We designed this study to compare the replication potential of turkey coronavirus (TCV) and its effect in chickens and turkeys and to study the effect of singleand combined infection of turkey poults with TCV and astrovirus. We studied the pathogenicity of TCV in experimentally inoculated turkey poults and chickens by observing the dinical signs and gross lesions. Two trials were conducted with 1-day-old and 4-wk-old specific-pathogen-free turkey poults and chickens. One-day-old turkey poults developed diarrhea at 48 hr postinoculation. Poults euthanatized at 3, 5, and 7 days postinoculation had flaccid, pale, and thin-walled intestines with watery contents. The 4-wk-old turkeys had no clinical signs or gross lesions. One-day-old and 4-wk-old chicks developed no clinical signs or gross lesions although the TCV was detected in gut contents of the birds throughout the experimental period (14 days). In another experiment, mean plasma D-xylose concentrations in 3-day-old turkey poults inoculated with TCV, turkey astrovirus, or a combination of both viruses were significantly lower than in the uninoculated controls.  相似文献

The feasibility of producing salmonella-free turkeys was investigated over a 5-year period. In Phase 1, a hatchery-breeder flock operation was monitored extensively for 4 years. Hatching eggs from a primary breeder over this period (1978-81) resulted in salmonella-free day-old poults from which 7500 hens and 600 tons were selected for breeders each of the 4 years. Approximately 2.5 million poults were produced over the 4 years. Salmonella arizonae was isolated from the hatchery debris over a 2-week period in 1980. The pelleted feed contained no animal protein products except fish solubles. A sample of feed from each delivery was cultured with no salmonella isolations. Environmental samples of dust and litter remained negative for salmonella. Phase 2 involved monitoring seven grow-out flocks initiated with salmonella-free poults with extra precautions directed at the feed and environment. The intestinal tracts of five of seven flocks at the time of marketing were negative for salmonella. Phase 3 involved a primary breeder-hatchery that had a 10-year history of S. sandiego infection in its breeder flocks and poults. A vaccination program using an autogenous oil-adjuvant bacterin supplementing other sanitation and management efforts resulted in elimination of S. sandiego. Because the breeder went out of business, it was not possible to determine if the freedom from salmonella could be sustained over a period of years.  相似文献

The reproductive tracts of turkey breeder hens from five flocks were examined grossly and histologically. Hens from one flock had a normal reproductive history, but hens from the four other flocks had poor records in both egg production and hatchability. Nodular growths occurred in the oviducts of birds in all five flocks. The incidence of lesions varied from flock to flock and from bird to bird. In four flocks, lesions were small and consisted of areas of dysplasia with adenomatous change. Histologically, the lesions in some birds in the fifth flock were adenocarcinomas. An acute upper respiratory disease was observed in two broad-breasted white (BBW) turkey primary breeder flocks. Associated clinical signs included sneezing, depression, and a deep dry cough originating from large conducting airways. Morbidity reached approximately 15-20% of the hens in an affected house. None of the turkeys died, and total feed consumption was not affected. A minimal effect upon egg production was noticed. Sera from an acutely affected flock exhibited a marked rise in titer to Bordetella avium compared with preinfection sera samples. In Case 1, B. avium was isolated in pure culture from affected birds. In Case 2, B. avium was diagnosed by serological results and clinical signs; bacteriological examination was not attempted. The findings presented here are consistent with an acute clinical outbreak of B. avium-induced turkey rhinotracheitis (turkey coryza) in BBW turkey breeder hens.  相似文献

Six hours post-hatch, large white turkey poults were inoculated intranasally with 5 X 10(7) organisms of the "W" isolate of Bordetella avium. Three hours after inoculation and subsequently on days 7, 10, 14, 21, and 28 postinoculation, 30 infected and 30 uninfected control poults were injected intramuscularly with alpha methylparatyrosine (AMPT, 250 mg/kg), and an additional 30 of each group received a saline vehicle. Three hours after AMPT injection, whole brains from treated and untreated poults from both infected and control groups were removed, weighed, and frozen until assayed for norepinephrine (NE), dopamine (DA), and serotonin (5-HT). B. avium-infected poults had a significant reduction in steady-state NE and Da and a greater depletion of NE and DA after AMPT treatment compared with control poults. Infected poults had significantly reduced whole brain 5-HT concentrations, which persisted through 21 days postinoculation. Altered brain NE, DA, and 5-HT concentrations suggest that the B. avium-infected poults may be less able to cope with additional stressors.  相似文献

Antibody responses of hens and their progeny were studied in commercial broiler nuclear lines. Starting at 168 days of age, individually housed pullets from lines A and B were fed a 16% crude protein and 2752 kcal metabolizable energy/kg mash diet supplemented with either 10 or 300 IU/kg of vitamin E fed as dl-alpha-tocopherol acetate. Fifty-eight days later (226 days of age), 12 hens per line-vitamin E subclass were inoculated i.v. with 0.1 ml of a 2.5% suspension of sheep red blood cells (SRBC). Plasma antibody titers were measured 6, 20, 40, 54, 70, and 88 days after inoculation. Hens from both lines were artificially mated to males from line C, and progeny from eggs collected 9-15, 25-30, and 65-70 days after inoculation were tested for antibodies to SRBC. Hens were reinoculated i.v. with 0.1 ml of 0.25% SRBC 88 days after the first inoculation, and their antibody levels were measured 3, 6, and 20 days later. Eggs laid 10-13 days after reinoculation were incubated, and antibody titers of chicks were measured at hatch. Antibody response of hens to an initial inoculation of SRBC was line-diet-time after inoculation specific. In line A, titers were greater for hens fed the lower than the higher vitamin E diet, whereas diet had no effect on the antibody levels in line B. Line effects (A > B) were observed on days 6 and 20 after inoculation but not thereafter. After the second inoculation, dietary vitamin E level had no effect on antibody levels of hens within lines, whereas a between-line difference (A > B) was observed for the lower but not the higher level of dietary vitamin E. Although there was no difference between diets for antibody transferred to progeny by line B, there was a difference (lower > higher) for line A. After reinoculation of their dams, antibody titers of chicks from line A, but not line B, reached levels similar to those after the first inoculation. Antibody levels were higher for chicks at hatch than in 16-day embryos or 10 days posthatch. The results of this research suggest genetic variation in response to immune stimulation by dietary vitamin E.  相似文献

The outbreak of the disease occurred in a large multiple-age farm with about 50,000 meat turkeys, where groups of 6-8000 one-day-old birds were stalled up every 14 days. All the turkey poults housed were affected mostly in the 1.-3. week of the life. The respiratory disease spread rapidly within the flocks and were characterised clinically by inclination of huddle, ruffled feathers, anorexia, stunted growth, swelling of the infraorbital sinus and nasal discharge. The clinical apparent disease lasted 3 to 4 weeks on the average in the affected flocks and were associated with a mortality from 7-20 percent. The main pathoanatomical lesions were catarrhal-fibrinopurulent rhinitis, sinusitis, tracheitis, bronchopneumonia and air sacculitis as well as atrophy of the thymus. Fibrinous adhesive peri- and epicarditis, perihepatitis, miliary necrotic foci in the liver and diarrhea have been found less frequently. The results of cultural and serological examinations of moribund and dead turkey poults of 6 different flocks indicate that Bordetella avium and Chlamydia psittaci are the primary inciting agents of the respiratory disease. However, the following severe course of the disease were mainly caused by concurrent infections with Klebsiella pneumoniae subsp. pneumoniae, Escherichia coli and Pseudomonas fluorescens. In some cases coccidiosis with lesions in ceca were additionally diagnosed. Campylobacter jejuni could be always isolated culturally from the liquid cecal content of diseased birds.  相似文献

The effects of Mycoplasma gallisepticum (MG) vaccination on egg transmission of MG and egg production were evaluated. Leghorn hens vaccinated with live MG (strain F), with strain F plus MG bacterin, with one dose of MG bacterin, or with two doses of MG bacterin all transmitted MG through the egg at a significantly lower level than unvaccinated controls. Hens vaccinated with two doses of MG bacterin had the longest lag before detectable transmission of MG through the egg. All vaccinated groups were protected against the egg-production drop seen in unvaccinated hens challenged with virulent MG.  相似文献

To assess the potential protective efficacy of a Salmonella enteritidis bacterin, an acetone-killed oil-emulsion vaccine was prepared from a phage type 13a S. enteritidis strain and administered subcutaneously to hens in two experiments. Hens were housed individually, and every other hen was vaccinated (at 23 weeks of age in one experiment and at 45 weeks in the other). A second (booster) bacterin injection was administered 6 weeks later in both experiments. Three weeks after the second vaccination, all hens were challenged with an oral dose of approximately 10(9) cells of a heterologous (phage type 14b) S. enteritidis strain. In both trials, S. enteritidis was isolated from fewer internal organs (spleens, ovaries, and oviducts) and pools of egg contents from vaccinated hens than from unvaccinated control hens. Vaccination did not, however, affect the percentage of hens that shed S. enteritidis in feces in either experiment.  相似文献

Turkey viral hepatitis (TVH) was experimentally reproduced in two experiments in 1-day-old poults. In the first experiment, an infectious inoculum was prepared from filtered yolk materials harvested from dead embryonating chicken eggs (ECE) previously inoculated with suspensions of liver and pancreas tissues collected from TVH-affected birds in commercial turkey flocks. One-day-old poults given a yolk-sac inoculation or oral gavage with this preparation developed lesions in the liver and pancreas characteristic of TVH at 20 days postinoculation (PI) in 60% and 14% of the experimentally infected birds, respectively. With the identical inoculum, embryo mortality occurred at 8 and 10 days PI in embryonating turkey eggs (ETE) inoculated into the yolk sac. In the second experiment, an infectious inoculum was prepared from filtered yolk materials from dead ETE harvested in the first experiment. One-day-old poults given a yolk-sac inoculation with this filtered yolk material developed lesions in the liver and pancreas within 5 days PI. At 20 days PI, 67% of the experimentally infected birds had similar lesions. With the inoculum given to these poults, embryo mortality occurred at 6, 8, and 10 days PI in ETE inoculated into the yolk sac. Virus particles 26-28 nm in diameter with icosahedral morphology typical of picornaviruses were identified by EM in the yolk sacs of ETE that died in both experiments, and inoculated ETE that died following passage of filtered suspensions of pancreatic tissues collected from affected birds in the first experiment.  相似文献
