Longer-term partial stagnant flooding, particularly when it occurs following transient complete submergence causes severe damage to modern rice varieties. Progress was made in developing varieties tolerant of complete submergence through transfer of the Submergence-1 (SUB1) gene into popular varieties. However, SUB1 may not be effective under partial stagnant flooding (SF), as the new varieties may not elongate and continue growth when fully or partially submerged because of the SUB1-mediated suppression of elongation. We tested a set of rice genotypes, including a pair of near-isogenic lines (NILs), Swarna and Swarna-Sub1, under either SF or SF following complete submergence of 12 d. Swarna-Sub1 had higher survival and yield than Swarna following 12 d of submergence, but survival and grain yield of all lines decreased substantially when SF of 15-30 cm followed complete submergence, with the sensitive lines experiencing greater reductions in growth and yield. This suggests the importance of combining SUB1 with tolerance of SF in areas where both stresses are expected during the season. Swarna and Swarna-Sub1 are more sensitive to long-term partial SF than IR49830 and IR42 because of their short stature, and Swarna-Sub1 showed slightly higher reduction in tillering than did Swarna when subjected to deeper SF alone, possibly because of further inhibition of elongation by SUB1 if induced in submerged tissue. The results suggest that tolerance of these genotypes to SF depends less on SUB1 introgression and more so on the genetic background of the recipient genotype, with better performance of the genotypes that are inherently taller, such as IR42 and IR49830. The SUB1 donor landrace FR13A and its derivative breeding line IR49830 had better survival and relatively less reduction in grain yield under SF following complete submergence and under longer-term partial SF, indicating that these genotypes may have genes other than SUB1 for submergence and SF tolerance. For better adaptation to prolonged SF, SUB1 should be introgressed into genotypes that tolerate partial SF through better tillering ability and taller shoots, because SUB1 may not be effective in shorter genotypes, as it promotes survival of submerged plants by hindering shoot elongation to conserve energy reserves. Varieties combining tolerance of prolonged SF with SUB1 will have broader adaptation in flood-prone areas and greater impacts on yield stability.  相似文献

A set of near-isogenic lines for blast resistance genes was developed by using an Indica-type elite rice variety, IR49830-7-1-2-2, suitable for the rainfed lowland conditions in the tropics, as a genetic background. Initially, we revealed that IR49830-7-1-2-2 harbors five blast resistance genes - Pia, Pib, Pik-s, Pita, and Pi11(t) - by using a differential system involving 19 selected standard blast isolates from the Philippines. Based on this result, we developed nine near-isogenic lines (NILs) targeting eight resistance genes - Pik, Pi7(t), Pi3, Pi5, Pita-2, Piz-5, Pish, and Pi9 - by recurrent backcrossing. The introgression of each resistance gene in the NILs was confirmed by reaction patterns to the blast isolates, allelism tests, and DNA marker analysis. In addition, a genome-wide DNA marker survey revealed that most of the chromosome regions in each NIL were of the IR49830-7-1-2-2 type. The agricultural characteristics of most of the developed NILs were almost the same as those of IR49830-7-1-2-2. Moreover, with one exception, they showed submergence tolerance similar to IR49830-7-1-2-2. The developed NILs could be used as a multiline variety suitable for the rainfed lowland in the tropics.

Background

Understanding the responses of rice to environmental stresses such as unscheduled submergence is of pressing important owing to increasing severity of weather thought to arise from global climate change. When rice is completely submerged, different types adopt either a quiescence survival strategy (i.e., minimal shoot elongation) or an escape strategy (i.e., enhanced shoot elongation). Each strategy can prolong survival depending on the circumstances. While submergence responses have been studied in rice typical of lowland and flood-prone areas, few studies have explored the physiological and molecular properties of upland rice under submergence. Here, we use seedlings of the upland rice ‘Tung Lu 3’ (‘TL3’) to analyze physiological and molecular responses to submergence. We compare them with those of ‘FR13A’, a lowland rice that tolerates submergence by adopting the quiescence strategy.

Results

Plant height and distance between leaf sheaths, increased rapidly in ‘TL3’ under submergence. Although this indicated a strong escape strategy the seedlings remained totally underwater for the duration of the experiments. In contrast, ‘FR13A’ elongated much less. Consequently, after 4 days complete submergence followed by drainage, ‘TL3’ lodged much more severely than ‘FR13A’. After 10 d complete submergence, 55% of ‘TL3’ seedlings survived compared to 100% in ‘FR13A’. Chlorophyll a, b and total chlorophyll concentrations of the 2nd oldest leaves of ‘TL3’ were also significantly above those of ‘FR13A’ (but were lower than ‘FR13A’ in the 3rd oldest leaves) and less hydrogen peroxide accumulated in ‘TL3’. Peroxidase activity in submerged ‘TL3’ was also greater than in ‘FR13A’ 1 day after submergence. Quantitative RT–PCR showed increased expression of sucrose synthase 1 and alcohol dehydrogenases 1 after 2 days complete submergence with significantly higher levels in ‘TL3’ compared to ‘FR13A’. Expression was also higher in ‘TL3’ under non-submerged conditions.

Conclusions

The upland rice line ‘TL3’ gave a stronger elongation response than ‘FR13A’ to complete submergence. This escape strategy is widely considered to prejudice survival when the plant remains totally submerged. However, contrary to expectations, ‘TL3’ survival rates were substantial although below those for ‘FR13A’ while physiological, biochemical and molecular parameters linked to adaptation differed in detail but appeared to be broadly comparable. These findings highlight that submergence tolerance is determine not only by the adoption of quiescence or escape strategies but maybe by metabolic and physiological properties unrelated to the underwater elongation rate.

The present study evaluated submergence responses in 88 lowland indigenous rice(Oryza sativa L.) landraces from Koraput, India, to identify submergence-tolerant rice genotypes. In pot experiments, variations in survival rate, shoot elongation, relative growth index, dry matter, chlorophyll, soluble sugar and starch contents were evaluated in two consecutive years under well-drained and completely submerged conditions. Principal component analysis showed that the first three axes contributed 96.820% of the total variation among the landraces, indicating wide variation between genotypes. Major traits such as survival rate, relative growth index, soluble sugar and starch contents appeared to be important determinants of phenotypic diversity among the landraces. Phenotypic coefficient of variance was higher than genotypic coefficient of variance for all the traits and all showed high heritability(90.38%–99.54%). Five rice landraces(Samudrabali, Basnamundi, Gadaba, Surudaka and Dokarakuji) were the most tolerant to submergence. When submerged for up to 14 d, Samudrabali, Basnamundi and Godoba were notable for having greater survival rates than a standard submergence tolerant variety FR13 A, and also notable for elongating more vigorously and accumulating more biomass. These three landraces may therefore be especially useful in lowland rice growing areas that are affected by both moderate stagnant water and flash flooding. Molecular genotyping revealed that the submergence tolerance of Samudrabali, Basnamundi and Godoba is linked to the presence of one or more different Sub1 loci and it may well prove useful for breeding improved submergence tolerant rice varieties, thereby assising to improve yield stability in the rainfed lowland agro-ecosystem.  相似文献

Submergence tolerant high yielding rice variety was developed using BR11 as a recipient parent applying foreground, phenotypic and background selection approaches. Recombinant selection was found essential to minimize linkage drag by BC2F2 generation. Without recombinant selection, the introgression size in the backcross recombinant lines(BRLs) was approximately 15 Mb on the carrier chromosome. The BRLs were found submergence tolerance compared to the check varieties under complete submergence for two weeks at Bangladesh Rice Research Institute, and produced higher yield compared to the isogenic Sub1-line under control ed submerged condition. The BRL IR85260-66-654-Gaz2 was released as BRRI dhan52 in 2010, which was the first high yielding submergence tolerant variety in Bangladesh. BRRI dhan52 produced grain yield ranging from 4.2 to 5.2 t/hm2 under different flash flood prone areas of Bangladesh in three consecutive seasons. The study demonstrated the efficiency of recombinant selection and better adaptability of the newly released submergence tolerant high yielding variety in flash flood prone different areas of the country with respect to submergence tolerance and yield potential.  相似文献

Summary

The cell elongation-promoting activities of brassinolide (BR) and indoleacetic acid (IAA) in the lamina-inclination test using etiolated rice seedlings were compared at a low temperature. IAA promoted the lamina joint-cell elongation at 30°G, but the effect was markedly lowered with decreasing temperature and reached null at 15°C. On the other hand, BR and castasterone (CAS), a brassinosteroid, promoted this cell elongation even at 15°G. Homobrassinolide (HBR), a brassinosteroid, also promoted this cell elongation, although the activity was weak. The combination of BR and IAA synergistically promoted this cell elongation at a low temperature (15°G).

Immersion of the dry seeds in 2 X 10-8 M BR for the first 24 h promoted the early growth of rice seedlings. This treatment also enhanced the germination rate and the growth after direct sowing in submerged paddy pots in a greenhouse or phytotron at a low temperature (15°C). Leaf spraying of BR on the rice seedlings at the 4th leaf stage increased plant height and the fresh weights of tops and roots even at a low temperature. BR sprayed on the completely expanded 4th leaves did not increase their blade length, but that sprayed on the expanding 5th and 6th leaves strikingly increased their blade length, and the effect tended to be stronger at a low temperature.

These results suggest that BR promotes cell elongation in young rice seedlings under low-temperature stress, although IAA had no such effect, and that BR may promote germination and the early growth of rice seedlings at a low temperature in direct sowing in the submerged paddy field and in the rice nursery.

The inhibitory effect of the herbicide dalapon on germination and shoot elongation was evaluated in two perennial ryegrass cultivars, Melino RvP and Cropper. These cultivars had been shown to be dalapon resistant and susceptible, respectively, in a previous comparison of 14-week-old plants. Dalapon (10-2 and 10-3 m) significantly reduced rates of germination and shoot elongation to the same extent in both cultivars. It is suggested that the effects of dalapon on germination and shoot elongation during the first 9 days of development are not indicative of levels of resistance in more mature plants.  相似文献

Summary

To characterize the protection effect of silicon against excessive transpiration in rice plants (Oryza sativa L.), we investigated the factors responsible for transpiration, such as cuticular conductance (gc), leaf surface wax content (WC), and stomatal movement in response to environmental stimuli, using leaves of plants grown in nutrient solution with silicon (+Si) and without silicon (–Si). The rate of transpiration of whole plants (E) under field conditions was clearly reduced by application of silica. The gc of –Si leaves under precisely regulated conditions was 33% higher than that of +Si leaves, reflecting higher adaxial gc. Silicon has little effect on WC, a factor responsible for regulation of cuticular resistance to water flux in rice leaves. These results suggest that silicon is one of the main factors responsible for regulation of gc, but that its effect was not due to an increase in the WG. Leaf conductance (gl) of –Si leaves tended to decrease more slowly than that of +Si leaves over time after changes from high to low light intensity. Moreover, it was always higher over the entire duration of the experiment, irrespective of irradiation intensity. The gl of –Si leaves was higher at lower relative humidity (RH) and the initial response to a subsequent increase in RH was slower than that of +Si leaves. A comparison of gl and gc between +Si and –Si leaves showed that the higher gl of –Si leaves was due to higher stomatal conductance. These results indicated that the reduction in E by application of silica was mainly attributable to reduction in the rate of transpiration through stomatal pores.  相似文献

Oilseed rape seedlings (Brassica napus L.) treated with uniconazole were transplanted at the five-leaf stage into specially designed experimental containers, and then exposed to waterlogging for 3 weeks. Pre-treatment of rape seedlings with uniconazole significantly increased seedling height, shoot width, number of green leaves and leaf area per plant, and consequently the shoot, root, and total dry weight after waterlogging. Following waterlogging stress, the uniconazole treated plants had a significantly improved growth including plant height, length and width of leaves, number of green leaves, leaf area per plant, and canopy width at the stem elongation stage, and plant height, stem width, and root, shoot and total dry weight at the flowering stage. At harvest, uniconazole treatment increased the number of primary and secondary branches, seeds per pod, and number of effective pods in branches and in terminal raceme after waterlogging treatment. The uniconazole-induced increase in the number of pods per plant and number of seeds per pod were the two yield components responsible for the significantly greater seed and oil yields obtained from the uniconazole plus waterlogging treated plants, over either the control or waterlogged plants. Uniconazole also reduced waterlogging-induced rise in the erucic acid content of the seeds. The modification of GA3, zeatin, ABA and ethylene levels due to pre-treatment of rape seedlings with uniconazole might have helped to delay the chlorosis and senescence induced by waterlogging. Uniconazole treatment also increased the leaf photosynthetic rates of waterlogged plants, in part, due to the changes in leaf conductance and hormone levels which ultimately affected various physiological processes.  相似文献

Morpho-Physiological Changes in Roots of Rice Seedling upon Submergence   总被引:1,自引:0,他引:1
Submergence is a serious environmental condition that causes large loss in rice production in rain fed lowland and flood affected area. This study evaluated morphological and physiological responses of rice roots to submergence using two tolerant rice genotypes FR13A and Swarna-Sub 1 and two sensitive ones Swarna and IR42. The tolerant genotypes had higher survival rate and less shoot elongation but greater root elongation during submergence than the sensitive ones. After submergence, the tolerant genotypes also had higher root dry weight and more active roots than the sensitive ones.Tolerant genotypes exhibited less root injury, with less malondialdehyde production and slower electrolyte leakage after submergence. Tolerant genotypes also maintained higher concentrations of soluble sugar and starch in roots and shoots and higher chlorophyll retention after submergence than the sensitive ones. Our data showed that root traits such as root activity and root growth are associated with survival rate after submergence. This is probably accomplished through higher energy supply, and membrane integrity is necessary to preserve root function and reduce injury during submergence. These root traits are important for submergence tolerance in rice.  相似文献

Abstract

Eleocharis retroflexa (Poir.) Urban ssp. chaetaria (Roem. & Schult.) T. Koyama, an amphibious leafless sedge, grows not only under terrestrial conditions but also under completely submerged aquatic conditions. We investigated the photosynthetic traits and structural features of the culms, which are the photosynthetic organs, in the terrestrial and submerged forms of this species and compared them with those of other amphibious species of Eleocharis which are known to change the photosynthetic modes. The culms of the terrestrial form had Kranz anatomy with well-developed Kranz (bundle sheath) cells and high levels of C4 enzyme activity typical of the NAD-malic enzyme (NAD-ME) subtype of C4 metabolism. They also had a δ 13C value typical of C4 plants, indicating that the terrestrial form fixes carbon through the C4 pathway. The culms of the submerged form had not only a Kranz-like anatomy but also revealed anatomical traits typical of leaves of submerged aquatic plants. The activities of the C4 enzymes in the submerged form were lower than those in the terrestrial form, but were still in the range typical of G4 plants, 14C pulse-12C chase experiments with the submerged form indicated that almost all of the fixed 14C was incorporated into G4 compounds, and subsequently the raioactivity was transferred into C3 compounds and sucrose. The submerged form showed no diurnal fluctuation in malate level. These data demonstrate that a C4 metabolism is operative even in the submerged form. This unique amphibious C4 plant provides an intriguing example of the physiological and ecological adaptability of C4plants.  相似文献

Abstract

The objective of this study was to clarify the effects of soil temperature in the stage from late tillering to panicle initiation (SI) and during the grain-filling stage (SII) on grain setting, dry matter production, photosynthesis, non-structural carbohydrate (NSC), xylem exudation and abscisic acid (ABA) levels in rice (Oryza sativa L. cv. Koshihikari). Rice plants were exposed to four different soil temperatures during SI or SII: 17.5, 25, 31.5 and 36.5°C (ST18, ST25, ST32 and ST37, respectively). The yield, yield components, grain filling and quality in SI were negatively influenced by high soil temperature of 37°C. On the other hand, there was no significant difference in those characters among temperature treatments in SII. The root/shoot ratio was smallest in the ST37 plants in both SI and SII, mainly due to their lighter root weight. At 7 days after initiation of treatment (DAT) in both SI and SII, the photosynthetic and xylem exudation rate tended to increase slightly as soil temperature increased up to 32°C. At 21 DAT, however, the photosynthetic rate was lowest in ST37, with concurrent decrease of diffusion conductance and SPAD value. In addition, decrease of NSC concentration in stem and xylem exudation rate, and increase of ABA level in leaves and xylem exudate were observed in ST37 plants at 21 DAT. These results suggested that high soil temperature before heading especially influenced yield, grain quality and plant growth. Possible mechanisms of the effect of soil temperature are discussed.  相似文献

Abstract

We measured the concentration of polyols (pinitol, ononitol, and myo-inositol), which are known to have health-promoting and/or disease-preventing functions, in the common ice plant (Mesembryanthemum crystallinum L.) cultured under salt- and drought-stressed treatments. In NaCl-treated plant the concentration of pinitol/ononitol increased with increasing NaCl concentration in culture solution. The maximal concentration was 3.6 mg g-1 FW, which was foundin the shoot top, followed by small side shoots (2.1 mg g-1 FW) of mature plants grown with 400 mM NaCl for 35 ds. The drought stress also accelerated the accumulation of pinitol/ ononitol. The maximal concentration was 1.2 mg g-1 FW, which was found in the shoot top of plants under the stress for 25 ds. The myo-inositol increased in salt-stressed plants at 3 ds after the start of the treatment and then decreased with the lapse of time during stress. The concentration of polyols in the ice plant was comparable to that in the other species reported to accumulate polyols at high levels. Radical scavenging activity evaluated by DPPH assay was increased two-fold by 400 mM NaCl treatment, which was twice as high as that in the leaves of lettuce (Lactuca sativa L.). These results indicated the high potential of the ice plant as a polyol-rich high-functional food.  相似文献

Sugarcane (Saccharum spp.) is an economical crop in the tropical and subtropical countries. However, because of global climate change, flooding has become problematic, particularly during the rainy season, in Thailand. We investigated the effects of floods on three commercial sugarcane cultivars, namely NiF8, U-thong 6 (UT6), and U-thong 9 (UT9), as well as Erianthus spp. Growth was assessed using a pot experiment in a glasshouse with two treatments: (1) control and (2) 60 d of flooding followed by 30 d of normal conditions. In comparison with control, during prolonged flooding, Erianthus showed greatly decreased CO2 assimilation, whereas NiF8, UT6, and UT9 showed slightly declined CO2 assimilation. Growth in plants subjected to 60 d of flooding was less influenced by floods while sucrose content was not affected except in UT6. During flooding, some roots died, resulting in plants compensating adventitious roots to offset the negative effects of root death and to assist them in maintaining their growth, which appeared from the submerged nodes, with different characteristics for each cultivar. However, 30 d after draining, roots remained damaged, while adventitious roots died, resulting in lesser growth as compared with the control, but it did not significantly affect sucrose content and sugar yield. This study suggests that sugarcane plants need to produce the adventitious roots to compensate their roots’ death during flooding and require time to recover their root system after flooding for obtaining the optimum yield and quality at harvest.  相似文献

In vitro direct plant regeneration of lucerne was achieved by simultaneous application of thidiazuron (TDZ) and 6‐benzyladenine (BA) in Murashige and Skoog (MS) medium. Seedlings were germinated and grown for 6 d on growth regulator–containing MS medium. The shoot tip, consisting of the apical meristem along with parts of the cotyledonary leaves and hypocotyl, was then cultured on a medium containing the growth regulator(s). Adventitious budding of the shoot tip was promoted synergistically by treatment with TDZ and BA, and a maximum of thirty‐five shoots per explant was obtained on a medium supplemented with 2 mg L?1 TDZ and 1 mg L?1 BA. Plant regeneration frequency varied from 67 to 93%, and five Indian lucerne cultivars responded well to the regeneration protocol. The Agrobacterium‐mediated transformation frequency from co‐cultivated explants was 13% following multiple shoot induction. Southern analysis of the T0 plants and T1 progenies confirmed stable inheritance of the hpt marker gene. Agrobacterium infection of the explant caused a significant reduction in the plant regeneration frequency (23%) and the number of shoots induced (11) when compared with uninfected explants. A single shoot tip provided sufficient material to regenerate and establish twenty‐seven lucerne plants, whereas only nine plants could be regenerated from an Agrobacterium co‐cultivated explant. This transformation protocol could represent a valuable improvement over existing ones for lucerne.  相似文献

Prolonged submergence is a major constraint to rice production, affecting over 15 million ha in South and Southeast Asia and causing an annual yield loss of over US$ 600 million. This is because all the modern high-yielding rice varieties are sensitive to complete submergence. A major quantitative trait locus (QTL), SUB1, associated with submergence tolerance, was mapped on chromosome 9 and recently bred into popular rainfed lowland rice varieties sensitive of complete submergence, using marker-assisted backcrossing (MABC). Here, we assessed the impact of introgressing SUB1 into three rice varieties popular in farmers’ fields of South and Southeast Asia, Swarna, Sambha Mahsuri, and IR64, under control conditions as well as following submergence in the field. Under control conditions, no differences were observed between Sub1 introgression lines and their recurrent parents in grain yield or quality aspects. Submergence substantially delayed flowering and maturity, and reduced grain yield, shoot biomass, harvest index, and yield components across cultivars. The reduction in yield was more drastic in the sensitive varieties, particularly when submerged for longer duration (17 d). The significant decline in grain yield was mainly attributed to reductions in grain filling, number of grains per panicle, and grain weight. The SUB1 QTL is effective in conferring tolerance of submergence for about 12–17 d, depending on floodwater conditions. Sub1 introgression lines showed a yield advantage of up to 3.8 t ha−1 and slightly better grain quality after submergence compared with their parents. Apparently, the introgression of SUB1 into popular varieties did not have any negative impact on their performance under control conditions but considerably enhanced their yield and grain quality following short-term submergence.  相似文献

Abstract

Effects of nitrogen (N) deficiency on photosynthetic gas exchange and photosystem II (PSII) photochemistry of flag leaves during grain-filling stage were investigated in six rice cultivars, Kasalath (a conventional indica), IR36 (an improved indica), Shirobeniya (a conventional japonica), Nipponbare (an improved japonica), Akenohoshi (an improved japonica-indica intermediate type) and BSI429 (an improved tropical japonica, a new plant type line) grown hydroponically in N-sufficient (NS) and N-deficient (ND) solution. From 3 to 24 days after heading (DAH), net photosynthetic rate (Pn), maximum quantum yield of photosystem II (PSII) (Fv/Fm), quantum yield of PSII electron transport ( Φpsii), and contents of chlorophyll (Chl) and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) in the flag leaves decreased, particularly under the ND condition in all six cultivars. A substantial difference was observed among the ND plants for the sustainability index (SI, the ratio of the value at 24 DAH to that at 3 DAH) of Pn, Fv/Fm, Φpsii, Chl content and Rubisco content; SIs of those parameters of Akenohoshi, BSI429, Nipponbare and Shirobeniya were higher than those of IR36 and Kasalath. The SI of Pn showed significant positive correlations with those of Fv/Fm, Φpsii, and the contents of Chl and Rubisco under the ND condition. It was concluded that the sustainability of photosynthesis in the flag leaves was mainly due to those of PSII photochemistry and electron transport, which was associated with the maintenance of Chl and Rubisco under the ND condition.  相似文献

Summary We have evaluated the effect that the, silencing of two genes specifically expressed in conditions of dormancy (A2-1) and sprouting (G1-1) had on tuber dormancy. For this purpose potato plants (Solanum tuberosum L. cv. Désirée) were transformed with the antisense of the genes G1-1 and A2-1 under the control of constitutive 35S CaMV promoter. A first generation of transgenic plants was propagated from axenic stem cuttings and a second generation by tuber planting. The plants obtined were analyzed for the length of dormancy, plant morphology and agronomic characteristics. Statistical analysis of dormancy in lines obtained from the original transformants for the antisense of G1-1 gene showed a significant increase in length as compared with different types of control plants, with few effects on plant vegetative habit and tuber production. In contrast, results obtained on A2-1 antisense transformed plants did not reveal any significant change on the length of dormancy. Here we report small-scale field trials performed with the aim to select and regenerate commercially exploitable potato plants with a stable transgene-dependent phenotype, affecting the length of dormancy.  相似文献
