美国红鱼和大黄鱼β-珠蛋白基因的克隆和遗传进化分析

根据已发表的其他脊椎动物β-珠蛋白基因序列设计并合成1对特异性引物,通过RT-PCR方法首次克隆到美国红鱼和大黄鱼β-珠蛋白cDNA,其开放读码框均由447个碱基组成,编码148个氨基酸的多肽,预测的相对分子量分别为16 333和16 148 Da,两者核苷酸序列和推导的氨基酸序列同源性分别为90.4%与81.2%.将推导的美国红鱼和大黄鱼β-珠蛋白氨基酸序列与其他鱼类及人β-珠蛋白氨基酸序列进行同源性比较,其同源性在35.5%～70.3%,它们与同属真口鱼纲的真鲷、黄尾鲱鱼、鲤鱼、虹鳟等的同源性较高,而与肉鳍鱼纲的肺鱼及板鳃纲的鲨鱼的同源性较低;二级结构预测表明,美国红鱼和大黄鱼β-珠蛋白存在类似哺乳动物的8个α-螺旋区,在近端与远端与血红素结合的组氨酸高度保守.     

牦牛α1-珠蛋白基因的克隆和序列分析

根据α-珠蛋白基因起始密码子上游及终止密码子下游的保守序列设计引物，以牦牛基因组DNA为模板克隆并测定了α-珠蛋白基因序列。结果表明，所测的氨基酸序列与牦牛α-珠蛋白链的氨基酸序列完全相同，即为牦牛α1-珠蛋白基因，长706bp，编码141个氨基酸。通过与其他物种的同源性比较，发现牦牛α1-珠蛋白基因的核苷酸序列与黄牛和水牛的同源性分别为99．71％和98．58％，与山羊、绵羊、人和马的同源性只有96．0：3％、95．57％、68．55％和51．13％；氨基酸序列与黄牛和水牛的同源性分别为99．29％和96．45％，与山羊、绵羊、人和马的同源性只有92．90％、91．49％、87．23％和87．23％，说明牛亚科α-珠蛋白基因在进化上高度保守。同时发现第254位的碱基A为牦牛α1-珠蛋白基因所特有。牦牛的两个α1-珠蛋白基因长度相等，都为706bp，且间距为2．47kb，距离比山羊的远。     

中国牦牛β-珠蛋白基因的克隆和序列分析

根据黄牛β-珠蛋白基因的序列设计引物,扩增了中国牦牛的β-珠蛋白基因,并对其进行了克隆测序和氨基酸序列比较分析。结果显示,其与黄牛β-珠蛋白基因的同源性很高,达到97%以上;检测到中国牦牛β-珠蛋白基因的2个等位基因7β3A sn和1β35A sn,其中67号牦牛个体中检测到2个等位基因7β3A sn和1β35A sn,64号牦牛个体中检测到等位基因1β35A sn,1078号牦牛个体中检测到等位基因7β3A sn;等位基因1β35A sn是中国牦牛特有的一个等位基因,推测该等位基因编码的第135位氨基酸天冬酰胺,可能会降低牦牛血红蛋白的氧亲和力。     

油茶14-3-3蛋白基因的cDNA克隆及序列分析

以构建的油茶近成熟种子cDNA文库和EST文库为基础,采用分子克隆技术,分离克隆1个14-3-3蛋白的全长cDNA序列,由1 156个核苷酸组成,5'非编码区57 bp,3'非编码区301 bp,开放阅读框长777 bp,编码259个氨基酸.该基因编码蛋白的分子质量为29.466 ku,等电点4.78,无信号肽序列,是非分泌蛋白,命名为Co-14-3-3a.推测的二级结构有9个α-螺旋,1个反平行的β-折叠,位于αC和αD之间.     

枸杞胆碱合成关键酶基因PEAMT的克隆及生物信息学分析

利用RT-PCR和RACE结合的方法,获得了枸杞磷酸乙醇胺N-甲基转移酶PEAMT基因cDNA全序列(命名为LbPEAMT).LbPEAMT cDNA全长1 863 bp,开放读码框1 497 bp,编码一个由498个氨基酸构成的多肽,理论分子量为56.53 kDa,等电点pⅠ为5.50.通过多种序列比对,该基因编码的氨基酸序列与番茄PEAMT氨基酸的相似性为93％.二级结构预测LbPEAMT蛋白由44.98％的α-螺旋、17.67％的延伸链、6.63％的β-转角和30.72％的不规则卷曲组成.LbPEAMT是一个亲水蛋白.含有2个S-腺苷甲硫氨酸依赖催化活性结构域,分别位于N端65-164 aa和C端290-392aa.每个活性区都包含有4个部分基序,分别为Ⅰ、post Ⅰ、post Ⅱ和post Ⅲ.LbPEAMT的这2个活性区在蛋白、磷脂和小分子甲基转移酶中都是相对保守的.     

大肠杆菌志贺样毒素Ⅱ型基因的分子克隆

本文克隆了含大肠杆菌志贺样毒素Ⅱ型基因的2．0kbSau3AⅠ酶切片断，对插入片段进行了核苷酸序列测定，1298bp的核苷酸序列包含2个完整的开放读框，其中SLT ⅡA亚基基因编码区长957bp，编码319个氨基酸多肽，SLTⅡB亚基基因编码区长267bp，编码89个氨基酸多肽，与已知人源的SLTⅡ基因核苷酸序列比较具有99％的同源性，与SLTⅠ结构基因相比，A亚基同源序列为56．43％，B亚基为60．15％。     

牦牛2个α-珠蛋白基因在染色体上排列顺序的确定

本试验扩增并克隆测定了牦牛2个α-珠蛋白基因间区域的序列.根据所测结果及人、鼠、山羊、马和黄牛的α-珠蛋白基因序列比对中的保守区设计了2对特异性引物,扩增并克隆测定了牦牛α-珠蛋白的上、下游基因序列.将测定的牦牛α-珠蛋白基因间区域序列进行同源性搜索,发现没有任何一个基因与它同源.上、下游基因的测序结果表明,上游基因为牦牛α1-珠蛋白基因,下游基因为α2-珠蛋白基因.这表明牦牛的2个α-珠蛋白基因在染色体上紧密排列,其中α1-珠蛋白基因在前,α2-珠蛋白基因在后.     

梅花鹿β-防御素-1(siBD-1)cDNA的克隆及序列分析

从梅花鹿的舌黏膜上皮组织中提取RNA,根据已获得的驯鹿β-防御素reBD-1基因序列设计并合成引物,采用RT-PCR技术进行梅花鹿β-防御素siBD-1基因的扩增,将获得的300bp的cDNA进行双向克隆及序列测定分析。结果表明所克隆出的cDNA为梅花鹿β-防御素siBD-1,该cDNA包含由192个碱基组成的开放读码框(ORF),编码64个氨基酸残基的前原防御素,该前原防御素含有β-防御素特征性结构即6个在特定位置上的保守半胱氨酸残基。siBD-1 cDNA的获得为以后更好地研究梅花鹿黏膜防御机制奠定了基础。     

3种食用菌漆酶活性比较及其基因的克隆与序列分析

[目的]研究食用菌漆酶基因的差异以及漆酶的活性。[方法]利用编码杏鲍菇漆酶基因序列（GenbankNo．AY686700）设计特异性引物,采用PCR方法获得3种典型木腐食用菌（杏鲍菇、平菇、白灵菇）漆酶的基因序列。[结果]扩增得到的片段长度均为2606bp,通过对基因序列的内含子／外显子分析,获得漆酶编码区的cDNA,该基因的开放阅读框由1596个核苷酸组成,编码一个由531个氨基酸组成的多肽。3种食用茵漆酶基因的核苷酸序列、氨基酸序列同源性均为99．81％,说明漆酶的一级结构序列具有很高的保守性。[结论]序列的差异性、氨基酸跨膜区的不同及蛋白质的构象不同可能导致漆酶活性的高低。     

桂花β-胡萝卜素羟化酶基因的克隆与序列分析

为揭示桂花不同品种β-胡萝卜素羟化酶(β-carotene hydroxylase,HYB)基因的序列差异,利用已构建的桂花花瓣转录组数据库中Unigene序列信息,结合RT-PCR技术对桂花品种玉玲珑、金球桂、堰虹桂和日香桂中2个β-胡萝卜素羟化酶基因(HYB1和HYB2)的最大阅读框进行扩增,并测序验证。生物信息学分析结果表明,4个桂花品种HYB1基因均含有长为909 bp的开放阅读框,编码302个氨基酸残基; HYB2基因均含有长为915 bp的开放阅读框,编码304个氨基酸残基。4个桂花品种HYB1和HYB2推导的氨基酸序列均具备保守的组氨酸结构域。HYB1基因核苷酸序列存在27个变异位点,其氨基酸序列存在8个变异位点; HYB2基因核苷酸序列存在12个变异位点,其氨基酸序列存在3个变异位点。进化树分析结果发现,4个桂花品种的HYB1和HYB2蛋白与茄科植物番茄和辣椒HYB蛋白亲缘性最高。     

Activation of developmentally mutated human globin genes by cell fusion

Human fetal globin genes are not expressed in hybrid cells produced by the fusion of normal human lymphocytes with mouse erythroleukemia cells. In contrast, when lymphocytes from persons with globin gene developmental mutations (hereditary persistence of fetal hemoglobin) are used for these fusions, fetal globin is expressed in the hybrid cells. Thus, mutations of developmental origin can be reconstituted in vitro by fusing mutant lymphoid cells with differentiated cell lines of the proper lineage. This system can readily be used for analyses, such as globin gene methylation, that normally require large numbers of pure nucleated erythroid cells, which are difficult to obtain.     

高产和低产奶牛几项血清生化指标的比较

对高产和低产奶牛(305天泌乳期的产乳量分别为6000公斤以上和4000公斤以下)的几项血液生化指标进行比较研究,结果表明:高产奶牛在血清总蛋白、球蛋白、血糖,谷草转氨酶活性值等方面,显著高于低产奶牛(p<0.005),而血清钙、磷、甘油三酯的含量及谷丙转氨酶活性值等,则低于低产奶牛,但差异不显著(p>0.005)。     

Autonomous developmental control of human embryonic globin gene switching in transgenic mice

The mechanisms by which expression of the beta-like globin genes are developmentally regulated are under intense investigation. The temporal control of human embryonic (epsilon) globin expression was analyzed. A 3.7-kilobase (kb) fragment that contained the entire human epsilon-globin gene was linked to a 2.5-kb cassette of the locus control region (LCR), and the developmental time of expression of this construct was studied in transgenic mice. The human epsilon-globin transgene was expressed in yolk sac-derived primitive erythroid cells, but not in fetal liver or bone marrow-derived definitive erythroid cells. The absence of epsilon gene expression in definitive erythroid cells suggests that the developmental regulation of the epsilon-globin gene depends only on the presence of the LCR and the epsilon-globin gene itself (that is, an autonomous negative control mechanism). The autonomy of epsilon-globin gene developmental control distinguishes it from the competitive mechanism of regulation of gamma and beta-globin genes, and therefore, suggests that at least two distinct mechanisms function in human hemoglobin switching.     

将透明颤菌血红蛋白基因(vgb)转移到链霉菌染色体整合载体的构建

链霉菌抗生素的生物合成对培养环境中氧的供应相当敏感,为了改善这一溶氧问题,本研究通过将透明颤菌血红蛋白基因(vgb)克隆到含ФC31int,attP基因的诱导型表达载体pU8600上,构建了可接合转移到链霉菌的整合型表达vgb基因的质粒pHZ1276。pHZ1276通过接合转移导入变铅青链霉菌(Streptomyces lividans),所得重组子经Southern杂交验证后,进行诱导表达,菌体经破碎后所得蛋白粗提物经Western杂交和CO结合试验表明vgb基因在该菌中表达出了有活性的VHb蛋白。     

Heterozygous Beta thalassemia: balanced globin synthesis in bone marrow cells

In two patients with heterozygous beta thalassemia the rates of synthesis of the alpha and beta chains of hemoglobin were equal in nucleated red cell precursors, although beta chain synthesis was reduced in peripheral blood reticulocytes. This finding suggests a relative instability of beta chain messenger RNA in beta thalassemia.     

Simultaneous expression of globin genes for embryonic and adult hemoglobins during mammalian ontogeny

The dominant hemoglobin of the adult hamster was detected in yolk-sac erythroid cells, and its identity was confirmed by peptide mapping and by analysis of relevant peptides. Both the presence and active synthesis of two embryonic hemoglobins presumed to exist only in yolk-sac erythroid cells were detected in neonatal liver and spleen. Thus the time span of expression of both embryonic and adult globin genes during mammalian ontogeny may be considerably broader than presently believed.     

Heme and globin synthesis control: observations in vivo in beta thalassemia

After administration of glycine-2-(14)C to a patient with thalassemia, the specific activities of heme and globin of F hemoglobin were consistently higher than those of hemoglobin A. After reaching a maximum, the ratio of the specific activity of heme to that of globin remained constant within each hemoglobin. Explanations considered include dilution by preformed subunits, differential turnover of hemoglobins, and possibly more than one heme-synthesizing pool.
