The morphological variation and the systematical status of 28 traditional melon including two ancient varieties collected from different geographical and bioclimatic areas of Tunisia were assessed based on 34 qualitative and quantitative traits. Yellow Canary was used as a reference variety. Significant differences among accessions within and among sites of collection were revealed for the majority of traits. The accessions from the Sahel were more polymorphic than those from Tozeur’s oasis. However, the classification of the local accessions according to Munger and Robinson’s varietal groups did not allow clear segregation of several of them because of their high heterogeneity. The principal component analysis (PCA) performed on all measured traits, allowed a distinction among the introduced varieties clustered according to their varietal group, and the local accessions grouped separately according to their geographical origin and bioclimatic zone. Mahalanobis distances among PCA’s groups were significant. The correlation found between Mahalanobis distance matrix and geographical distance matrix among origins corroborated isolation per distance. Conservation strategies should be made appropriately according to origins. Accessions from the Sahel showing the highest polymorphism within and among sites of collection should be firstly preserved.  相似文献

Earthworms are being used as bio-indicators to assess terrestrial pollution. However, it is often not known whether their populations possess a uniform genetic structure, which would allow comparison of residues or biological properties of earthworms from different sampling locations. In order to investigate this point, random amplified polymorphic DNA (RAPD) variation was surveyed in earthworms (Lumbricus terrestris) from five different sampling sites in Germany. Forty oligonucleotide RAPD primers (10 base pairs in length) were screened, three of which produced high polymorphic band patterns. A total of 61 DNA fragments were detected in 90 individuals of L. terrestris from five sampling sites with 49 (80.3%) RAPD markers being polymorphic. The genetic similarities within (band sharing rates between 0.756 and 0.795) and among the L. terrestris populations (0.635) were similar even at widely separate locations. Inter-population variation in the RAPD pattern for all five earthworm populations accounted for 37.9% of the total variation, while intra-population variation for three adjacent Saarland populations accounted for only 18.0% of the total variation. Principal component analysis (PCA) and the genetic distances of the populations confirm these results. Twenty-four percent of the genetic distance is caused by geographical isolation as shown by a test for isolation by distance. These results show that L. terrestris fulfils the genetic qualifications for a bio-indicator particularly at closely located sampling sites. However, the results also suggest that earthworm studies of widely separated locations should include genetic characterisation of the earthworm samples.  相似文献

An analysis of the variability of genes encoding six isozyme systems (15 loci) and two storage proteins (2 loci) in landrace barley from Ethiopia is reported. The materials consisted of populations collected from sites as low as 1650 and as high as 3750 meters, covering a wide range of agro-ecological conditions and geographical areas. Of the 17 loci 7 were polymorphic and 10 monomorphic when the 95% criterion of polymorphism was applied. Despite the disproportionate monomorphic loci, polymorphism was detected in all populations when this criterion is used. The populations were found to possess fairly low mean number of alleles per locus (A = 1.5), low mean value of expected heterozygosity (H = 0.134) and a fairly high mean percentage of polymorphic loci (P = 35.3%). The mean FST= 0.474 for the populations is typical of inbreeding species. The result indicated that allelic richness is concentrated in altitude class 3 (2500–3000 m) followed by altitude class 1 (<2000 m). Altitude class 2 (2000–2500) holds an intermediate place though it is the highest in terms of expected heterozygosity (H = 0.245). Higher genetic diversity is concentrated in some geographical regions such as Shewa, Arsi, Bale compared to others (Welo, Gamu Gofa, Gojam). Genetic differentiation among the agro-ecological zones was more profound than both among the altitudes and among regions. Correlation analysis between phenotypic diversity (Shannon-Weaver diversity index) and expected heterozygosity (H) for isozyme/hordein loci revealed non significant associations except with respect to agro-ecological zones. In general, it was detected that sites in highland areas in central and northern regions may be more desirable for in situ conservation than sites in peripheral regions in terms of isozyme/hordein diversity and current rate of varietal replacement.  相似文献

Twenty natural populations of F. cymosum, an insect-pollinated allogamous wild species of Fagopyrum, were investigated for their chromosome number and allozyme variation at 10 Ioci encoding 8 enzymes. Diploid populations were obtained in Sichuan, Yunnan and Tibet in China, whereas tetraploid populations were obtained in Tibet, the Himalayan hills, Thailand and southern China. Both diploid and tetraploid populations maintained a large amount of allozyme variation. The average heterozygosity, He, ranged from 0.045 to 0.389 (0.213 on average). Tetraploid populations showed higher He values than diploid populations. Natural populations of F. cymosum were locally well differentiated (GST = 0.322), probably due to reproductive isolation between the two ploidy levels and to distribution over wide areas from southern China to the Himalayan hills. The phylogenetic tree constructed by the neighbor-joining method based on allozyme variation clarified two distinct groups of diploid populations, the Sichuan and Yunnan groups. As for the tetraploid populations, polyploidization occurred twice independently, once in Yunnan and once in eastern Tibet. Tetraploid F. cymosum diffused to warmer areas such as southern Yunnan and Thailand as well as to cooler areas of Tibet and the high Himalayan hills and became the most widely distributed of the Fagopyrum species.  相似文献

Fifty grass pea (Lathyrus sativus L.) populations collected from different administrative regions and altitude classes in Ethiopia, were evaluated for variations of five morphological traits. Shannon-Weaver diversity index was calculated for traits, populations and altitude classes. Monomorphism was recorded in flower color (blue) for most of the populations. Olive seed colour was dominantly distributed in all regions with mean frequency of 50% while black seed colour is rare with mean frequency of 4% in Ethiopia. Much variability was observed within populations. High diversity values for pattern of testa colour and leaflet size were recorded in populations of highland origin (>2550 m.a.s.l.). Shannon's diversity index is highest in Gondar (H = 0.65) followed by Tigray region (H = 0.64) even though there was no significant variation in H values between regions. These regions with high diversity are recommended for immediate in situ and ex situ conservation.  相似文献

Genetic diversity was investigated in 348 accessions and subaccessions of grasspea (Lathyrus sativus L.) from 10 geographical regions. Polymorphism for 20 isozymes of 13 enzyme systems was studied to estimate the genetic diversity. The Near East and North Africa regions included the most variability for these isozyme systems, suggesting that the center of diversity (center of origin) for grasspea is in this general area. The lowest variability was found in accessions and subaccessions from South America, followed by those from Sudan–Ethiopia. Diversity was measured for individual loci over regions and EST-1 and SKDH had the highest genetic diversity. The closest genetic diversity was observed for LAP-2, followed by AAT-1 and PGM. The closest genetic distance existed between populations from the Near East and North Africa. Populations from South Asia and Sudan–Ethiopia, though geographically widely separated, exhibited a closer genetic distance from each other than from other regions.  相似文献

Chloroplast Microsatellites to Investigate the Origin of Grapevine   总被引:5,自引:0,他引:5  
The origin of the grapevine, Vitis vinifera ssp . sativa L., has been investigated with archaeobotanical–archaeological, cultural and historical data indicating a unique domestication centre located in the Caucasian and Middel-East regions about 6–7000 years ago, but, events leading to the domestication of this species are still an open issue. In this work, eight universal chloroplast microsatellites are used to assess genetic relationships among varieties selected as representatives of four distinct geographical groups from Middle-East to Western European regions. Results show that two out of the eight analysed chloroplast loci are polymorphic within the 142 individuals. Allele variants of the cpSSR loci combine in a total of six different haplotypes. The analysis of haplotypes distribution and haplotype diversity (HD) suggest that only three out of the six haplotypes are represented in the Caucasian and Middle-East samples, with 90% of individuals sharing the same haplotype. Moreover, the presence of all six haplotypes in the European accessions, with a high level of haplotype diversity, suggests varietal influx in these areas. Concerning the Western European varieties, especially in Spanish accessions, half of the individuals share haplotype VI which is completely absent in the Caucasian and Middle-East cultivars. This result opens the discussion about the existence of a unique and common domestication centre, located in the Caucasian and Middle-East area, for all the European cultivars. This work suggests the usefulness of chloroplast genome markers to provide information on haplotype distributions that could help to identify further geographical areas for grapevine varietal evolution.  相似文献

Genetic diversity in forest coffee (Coffea arabica L.) was estimated using inverse sequence-tagged repeat (ISTR) markers. One hundred ninety two samples representing 16 populations of C. arabica from four regions of Ethiopia were analyzed with 12 pairs of forward and backward ISTR primer combinations. A total of 144 reproducible bands were generated out of which 37 (25%) were polymorphic and scored as present (1) or absent (0) data matrix. This data was used to compute Jaccard coefficient to estimate genetic variability among all possible pairs of samples. The proportion of polymorphic bands within populations ranged from 19% for Bale-3, to 54% for Walega-2 populations. Un-weighted pair group method with arithmetic mean (UPGMA) based cluster analysis generated two clusters at 56% similarity value. The samples were clustered on the basis of their geographical origin, which could be attributed to a few region specific banding patterns detected. However, within regions most of the samples failed to cluster on the basis of their respective populations, which may be due to the presence of substantial gene flow between local populations in the form of seedlings carried out by farmers. The results may provide information to develop strategies for in situ conservation.  相似文献

Genetic variability, based on prolamines, was studied among 40 populations of Agropyron mongolicum Keng that were collected from the extensive geographic areas of northern China. A high level of genetic diversity was detected in these A. mongolicum populations. In total, 81 electrophoretic bands were observed following acid polyacrylamide gel electrophoresis (A-PAGE), among which 20, 16,␣10, and 35 allelic variants were separated into α-, β-, γ-, and ω-fractions of prolamine. Each A. mongolicum population had unique electrophoretic pattern, which exhibited enough genetic diversity for identification and classification of populations in A. mongolicum. The proportion of diversity among and within geographic regions from which the populations of A. mongolicum originated indicated that 52.1% of the total variation was found within regions, and 47.9% among regions. According to the dentrogram based on the Nei’ genetic distance matrix, the A. mongolicum populations from different regions were distinctly clarified. At Nei’s distance of 0.75, the populations of A. mongolicum were divided into two groups. The UPGMA phenogram and the principal coordinate analysis (PCA) indicated that the populations from similar eco-geographical situations were clustered together.  相似文献

Ethiopia is at the edge of the distribution for African wild rice, Oryza longistaminata. Here, chloroplast (cp) and simple sequence repeat (SSR) markers were applied to wild rice accessions in Ethiopia to evaluate how they differ from control O. longistaminata, O. barthii and O. glaberrima accessions which originated from African countries. Based on the cp genomes of African wild rice species, maternally inherited cpINDEL markers were developed. The cp indels helped to elucidate 20 plastid types. African cultivated rice shared a particular plastid type with one of annual O. barthii. Parts of northern wild rice in Ethiopia shared Type 6 with control O. longistaminata. The north group shared another type with parts of the south group. The 16 SSR markers amplified a total of 155 alleles in 215 rice accessions, with mean allelic richness of 9.688 per locus, observed heterozygosity of 0.241, expected heterozygosity of 0.724, polymorphic information content of 0.700, and a significant genetic differentiation of 0.215. Both cpINDEL and nuclear markers analyses suggested that wild rice in Ethiopia belongs to O. longistaminata. However, they carry both a unique plastid type and different population structure from control O. longistaminata collected from other areas in Africa. We concluded that the edge of its distribution maintains unique variation. These populations are regarded as valuable genetic resources for future rice breeding.

Isozyme studies of genetic diversity and evolution in Hippophae   总被引:4,自引:0,他引:4  
Summary To provide information on the genetic variation, differentiation and evolution in Hippophae seed samples of 25 populations from China, Finland and Russia were electrophoretically analyzed. Of six loci investigated, four were good genetic markers for identifying species and subspecies. The percentages of polymorphic loci per population were 40.3% and 62.5% at 0.95 and 0.99 polymorphic criteria respectively. The mean number of alleles per locus per population was 2.1. Total genetic diversity in the material was 0.4614. Genetic diversity partitioning showed that there was a large amount of diversity residing within geographical populations (0.1354), between subspecies within species (0.1046) and between species (0.2566) but not between geographical populations (0.0114). There were nearly twice as many negative fixation indices as positive ones in Hippophae populations. The phylogenetic tree agreed very well with botanic classifications of the species and subspecies and their geographical distributions, and quantitatively presented the genetic relationships of 25 populations. A detailed view of the evolutionary stages in Hippophae showed clearly a general decline of similarity as evolutionary divergence continued, which further explained the evolution process in Hippophae.  相似文献

Amplified fragment length polymorphism (AFLP) markers were used to characterize the genetic diversity within and among natural populations and cultivars of Hedysarum coronarium. Twelve populations within Tunisia were evaluated with three AFLP primer combinations. A total of 207 reproducible bands was detected of which 178 (86%) were polymorphic. The great discriminative power of AFLP markers and their ability to represent genetic relationships among Hedysarum plants was demonstrated. Genetic diversity within and among populations was assessed through Principal Component Analysis (PCA) and cluster analysis by using the Neighbor-joining clustering algorithm. AFLP technology has provided evidence of a high degree of intra- and inter-population genetic diversity in H. coronarium. AFLP banding patterns provided molecular markers correlated with the plants’ geotropism. In addition, AFLP markers can differentiate wild accessions from cultivars. Moreover, geographical origins did not correspond to population clustering.  相似文献

In this study, we investigated the genetic diversity and relationships in C. olitorius by analysing populations representing different distribution areas, and developed a hypothesis on the origin and spread of the species in the pantropics. We employed amplified fragment lengths polymorphism (AFLP) and morphometric analyses in a total of 101 C. olitorius accessions. Results of both data sets are mostly congruent. The molecular analysis indicated generally low genetic diversity within populations and the Nei’s gene diversity (He) ranged from 0.0457 to 0.0955 with a mean of 0.0763. Qualitative traits, especially related to leaf morphology, branching habit and stipule color were the taxonomically most informative characters. The highest morphological variability occurred within African accessions, indicating that this species originally evolved in Africa. In both analyses, the Asian materials were nested within African populations, especially with those from North and East Africa. This indicates an African origin of the species and we hypothesise that dispersal occurred via the Mediterranean–Indian trade routes instead of natural migration along the coasts from western Africa to the Indian subcontinent. Both analyses revealed materials from Ethiopia to be quite distinct. The highest number of private bands (N?=?35), percent polymorphism (29.29%), and gene diversity (0.0955) were also detected in wild accessions collected from Ethiopia, suggesting a long-term spatial isolation of C. olitorius populations in the country. Germplasm samples from this region could therefore be a useful source of genetic variation in jute breeding programs.  相似文献

Changes in the diversity of landraces in centres of diversity of cultivated plants need to be assessed in order to monitor and conserve agrobioversity—a key-element of sustainable agriculture. This notably applies in tropical areas where factors such as increased populations, climate change and shifts in cropping systems are hypothesized to cause varietal erosion. To assess varietal erosion of staple crops in a country subjected to various anthropogenic and natural environmental changes, we carried out a study based on a comparison of the diversity of pearl millet and sorghum varieties collected in 79 villages spanning the entire cereal-growing zone of Niger over a 26 year period (1976–2003). For these two crops, the number, name and type of varieties according to important traits for farmers were considered at different spatial scales (country, region, village) at the two collection dates. The results confirmed the high diversity of millet and sorghum varieties in Niger. No erosion of varietal diversity was noted on a national scale during the period covered. Some changes were observed but were limited to the geographical distribution of certain varieties. This highlights that farmers’ management can preserve the diversity of millet and sorghum varieties in Niger despite recurrent and severe drought periods and major social changes. It also indicates that rainfed cereal cropping systems in Niger should remain to be based on millet and sorghum, while reinforcing farmers’ seed systems.  相似文献

Fifty-five Greek Vicia faba L. populations, collected from diverse areas, were planted at two dry and low fertility sites for evaluation and classification. Yield evaluation, which was carried out by Principal Component Analysis (PCA) on the basis of seven yield traits, showed the number of pods per plant, number of ovules and seeds per pod, and branching from the basal nodes to be the most important traits for population evaluation regarding yield. For population classification, four dissimilarity coefficients (Manhattan, Average Taxonomic Distance, Euclidean distance and squared Euclidean distance) and four multivariate methods (PCA, UPGMA, Neighbor-joining and Principal Coordinate Analysis) were evaluated using fifteen morphological and seven yield traits. Neighbor-joining was chosen as the most suitable multivariate method. This method combined with PCA for the seven yield traits, placed the populations into six groups. As revealed by the application of PCA on all twenty-two traits the grouping was based mainly on pod characteristics, stem thickness, plant height, 1000 seed weight and branching from basal nodes. Based on the results of the present study, a model is proposed for conserving cross-pollinated species, such as faba bean.  相似文献

RAPD markers were used to investigate the distribution of genetic variability in a germplasm collection of the dioecious tropical tree Aroeira (Myracrodruon urundeuva) composed of open pollinated maternal half-sib families collected from trees at least 300 meters apart and grouped in nine collection areas throughout its geographical range in Brazil. Open pollinated families from nearby trees from two natural populations were also included in the study. A Principal Coordinate Analysis (PCO) based on genetic similarities estimated with 83 RAPD markers showed no defined clustering among individuals from the same collection areas. However, a matrix correlation test showed low, but significant correlation between genetic and geographic distances (r = 0.17; p < 0.02). An AMOVA showed that 92% of the genetic variation is found within collection areas. Significant variation was found both between regions and between areas within regions although only 3.54 and 4.44% of the variation was found at these levels respectively. An AMOVA on the individuals from the two natural populations resulted in a pattern of distribution congruent to the one found for the collection areas, with 97% of variability within populations. An analysis of 10 open-pollinated half-sib families revealed that between 75% and 89% of the variability is contained within families, suggesting that seeds from one tree represent a significant proportion of the variability found in the population. These results indicate that, either for the establishment of in situ genetic reserves or for ex situ conservation, efforts should be directed to the conservation of several individual trees or collection of open pollinated seed families in a few distant sites. In addition, if resources are available to enlarge the genetic base of the existing collection, expeditions should prioritize collection sites located in areas at significant distances to those already sampled.  相似文献

Jacaratia mexicana A. DC. (Caricaceae) is a tropical tree distributed throughout Mexico and Mesoamerica. Some evidence in Mexico indicates the presence of an incipient domestication process in this species. Phylogeographical analyses can potentially determine contemporary patterns of gene flow, isolation between population lineages, as well as historical processes such as population bottlenecks or expansions on their geographical areas. In this study we reconstruct the phylogeographical patterns in populations of J. mexicana A. DC., in order to find differences between genetic variation among wild and cultivated populations utilizing chloroplast DNA and nuclear DNA sequences. We generate a Bayesian phylogenetic tree, to estimate the divergence time between clades using calibrated mutation rates. We also infer the demographic history of these populations using neutrality tests among wild and cultivated accessions. We identified higher levels of haplotype and nucleotide diversity for the cpDNA and ITS types in wild populations than in domesticated populations. These results indicate a reduction of genetic diversity derived from human selection on domestication traits. Neutrality test suggests population expansion detected by the significant negative values of Fu’s Fs in the cultivated populations of this specie. These process results in an excess of rare polymorphism with the fixation of certain advantageous mutation throughout time, this implication are in accordance with the role of the strong selection in the fruit traits of J. mexicana. The dated phylogeny constructed with BEAST program indicated a dispersion pattern for the J. mexicana ancestors across the South Pacific and South Eastern populations during the late Pliocene. Posterior dispersion and divergence in the clades from Central Mexico and North Pacific are in agreement with the episodes of mountain-building in different regions of Mexico.  相似文献

For effective varietal improvement of horticultural crops peach (Prunus persica) and nectarine (Prunus persica var. nucipersica), information about their population structure and genetic relatedness plays an important role. In this study we used retrotransposon-based markers (iPBS) to estimate the genetic diversity and population structure of 48 peach and nectarine genotypes from various distinct geographical regions of Punjab and Khyber Pakhtunkhwa, Pakistan. A total of 461 alleles were identified from PCR amplicons derived from nine iPBS primer pairs with an average of 8.5 alleles/locus. Among all four groups the genotypes collected from Swat and Hunza had the highest and the lowest expected heterozygosity, unbiased expected heterozygosity and Shannon’s information index, respectively. We constructed a Neighbour-Joining dendrogram and performed principal coordinate analysis based on the distance matrices, and both forms of analysis grouped the 48 genotypes into two distinct clusters. The STRUCTURE software distributed the forty-eight genotypes into two main populations (k?=?2) indicating a low diversity between genotypes collected from Chakwal, Swat, Mansehra and Hunza.

Proton high‐resolution magic angle spinning nuclear magnetic resonance (1H HR‐MAS NMR) has been applied for the analysis of two cultivars of durum wheat produced in different Italian geographical areas. Furthermore, on the same samples, isotopic ratios were measured by mass spectrometry (IRMS). The application of chemometrics to these results permitted the discrimination of semolina by cultivar and geographical origin. A similar approach has been applied to the results obtained from chemical analyses. The comparison showed that NMR spectroscopy can provide a faster method for the detection of differences among the durum wheat semolina according to geographical and varietal origin. Furthermore, IRMS data are suitable to provide information about the geographical origin of samples. This present investigation is part of an extensive research project to find a scientific method capable of classifying wheat foods for the assignment of a “denomination of origin” trademark.  相似文献

The Ethiopian region is characterised by a wide range of agro-climatic conditions, which accounted for the enormous resources of agro-biodiversity that exist in the country. The most important of these resources is the immense genetic diversity of the various crop plants in the country. Of these, one of the most on farm genetically diverse crops is sorghum. Since the advent of formal breeding in particular after green revolution, genetic diversity of most crops has been threatened worldwide. In order to assess on farm genetic erosion (GE), various research methodologies were employed. These were focused group interviews with 360 farmers, on farm monitoring and participation with 120 farmers, key informant interviews with 60 farmers and development agents, and semi-structured interviews with 250 farmers. Besides, diversity fairs were done with over 1200 farmers. Notwithstanding the complexity of assessing GE, it was assessed by various methods; namely, by temporal method (comparing 1960 and 2000 collections), area method, and semi-structured interview method at individual, community or wereda level and causes of varietal loss from other various perspectives. Farmers perceived GE as the reduced importance of the variety as indicated by lower proportion in the varietal portfolio. The five most important factors for varietal loss at individual farmers’ level were reduced benefit from the varieties, drought, Khat expansion, reduced land size and introduction of other food crops respectively. GE was not affected by wealth groups and ecological regions. Farmers do not make simple replacement as a strategic mechanism for genetic resources management. GE at regional level was quantified by temporal and spatial method. There was a complementation not rivalry between farmer varieties (FVs) and improved varieties (IVs). The whole process of GE is explained by three models, namely: Bioecogeographic enhanced genetic erosion model, Farmer induced genetic erosion model and Farmer-cum-bioecogeographic genetic erosion model. As aforementioned, sorghum genetic erosion behaviour is completely different from other food crops such as tetraploid wheat. The prediction in the late seventies that complete erosion of FVs by IVs by the end of the eighties, the principle of GE that competition between IVs and FVs, favours the former and results in the replacement of the latter is not valid in the context of sorghum in Ethiopia. Hence, maintenance of the on farm genetic diversity of sorghum is a reality but GE is rhetoric.  相似文献
