Bursas from specific-pathogen-free white leghorn chickens of both sexes were examined at several intervals from hatching to 28 weeks of age. No histologic alterations other than scattered atrophic or cystic follicles were observed through 20 weeks. Obvious involution, first noted at 24 weeks, was in early stages in females and quite advanced in males. Involution was essentially complete by 26 weeks, and only cicatrized vestiges of bursas were present at 28 weeks of age. Gross manifestations included bursal atrophy, variable yellowish discoloration of the mucosa, and matting or total loss of identity of the mucosal plicae. Histologic characteristics of involution are summarized by the following approximate sequence: atrophy and exfoliation of plica epithelium; subepithelial stromal fibrosis; fusion and ultimate collapse of plicae; liquefactive necrosis of first medullary then cortical elements of follicles, which seemed to progress from basal to apical portions of the plicae; progressive proliferation of stromal connective tissue and infiltration of macrophages into areas occupied by necrotic follicles; and, finally, complete fibrous organization of luminal debris, leaving a firm nodule formed by a contracted muscularis surrounding the cicatrized remains of the mucosa.  相似文献

Within the cloacal bursa (bursa of Fabricius), there is a diffusely infiltrated area (DIA) of lymphoid cells just dorsal to the bursal duct opening. Because this region displayed certain histologic characteristics of thymus-dependent areas found in mammals, such as high endothelial venules, heavy reticular fiber components, and epithelium infiltrated with lymphocytes, experiments were conducted to determine whether thymus-derived cells (T cells) were present. Two independent methods substantiated the presence of T cells in the DIA. After the acid alpha-naphthyl acetate staining technique, the characteristic T cell focal esterase activity could be demonstrated in significant numbers of cells found in the DIA. Also autologous radioactively labeled thymocytes could be identified in the DIA by autoradiography a few hours after IV injection. This establishment of a definite T cell area within the bursa gives additional credence to the contention that the bursa functions as a secondary lymphatic organ.  相似文献

Light-microscope and electron-microscope studies of a coccidial organism found in the bursa of Fabricius from 3 chickens clearly established the parasite as belonging to the family Cryptosporiidae. Hyperplasia and heterophil infiltration were associated with the presence of organisms attached to the microvillus border of epithelial cells lining the plicae of the bursa of Fabricius. Although there were no clinical signs or gross lesions common to the 3 cases described, all had similar histologic lesions in the epithelium lining the bursa of Fabricius.  相似文献

This article reports genetic and phenotypic parameters of monthly egg production and the influence of Box‐Cox transformation on the parameters from a population of White Leghorns, selected for feed efficiency. A total of 6450 daughters of 180 sires and 1335 dams were analysed by restricted maximum likelihood (REML) using a multivariate animal model. The traits considered were monthly egg productions, cumulative production of the first 5 months (S5), cumulative production of first 10 months (S10), and survivor egg production in the first cycle (S12). Two sets of data were analysed: the original data and with the Box‐Cox method transformed data. The results indicated that there were no great differences in the estimates between untransformed and transformed data. The estimates of heritability for monthly egg production were high for the first period, decreased to reach the lowest during peak production, and increased to the end of lay. The estimates of heritability for cumulative records were generally higher than monthly records. Genetic and phenotypic correlations among monthly egg production totals were generally high for contiguous periods and then decreased as the interval between months increased. The highest genetic correlation between monthly records and S5 was for the second month of production, whereas the correlations between monthly production totals and S10 and S12 reached their peak at the sixth and eighth months of production, respectively.  相似文献

Single-comb white leghorn hens infected with either B4 or C2 strain of Eimeria mitis produced watery droppings as early as day 5 PI. E. mitis infection did not affect egg weight. However, specific gravities of the eggs produced by the hens infected with E. mitis were lower than those produced by the controls. Egg production was significantly reduced temporarily, but most birds returned to production within 14 days. Many of the birds that ceased to lay went through a complete-body molt (56% of the C2-infected hens and 20% of the B4-infected hens). Hens that ceased to lay regained considerable amounts of pigment in their skin, beaks, and shanks.  相似文献

The present study compares embryonic mortality between lines selected for different production traits, assesses the effects of inbreeding of the hen and embryo on embryonic mortality, and estimates genetic parameters of embryonic mortality. The experiment covered 10 generations of selection for increased egg number (EN), egg weight (EW), egg mass (EM) and a control line (C). The data included age at 1st egg, egg number and egg weight. Percent fertile eggs (PF), percent hatched of fertile eggs (PHF) and percent dead chick at hatch (PDH) were also recorded for the selected parents. PDH was higher in the selected lines than in the control line. Among the selected lines, the EW line had the highest embryonic mortality. Inbreeding of the hen and embryo had no significant effect on PDH in any of the lines. Estimates of heritability for PDH were 0.10+/-0.05, 0.02+/-0.02, 0.03+/-0.02 and 0.02+/-0.02 for lines EN, EW, EM and C, respectively. There was a positive genetic correlation between egg weight and PDH in line EW indicating that selection for increased egg weight was associated with high embryonic mortality. A negative genetic correlation between PDH and reproductive traits in line EN was observed, which is favourable.  相似文献

The developmental process of a Cryptosporidium isolated in Japan in the chicken intestine was investigated by scanning (SEM) and transmission electron microscopies (TEM). The parasites were detected in the ileum, cecum, colon, cloaca and bursa of Fabricius (BF). The intensity of infection tended to peak later in the BF than ileum. Trophozoites and schizonts were detected in all the portions of intestine, and were dominant in the developmental stages. Although macrogamonts were the secondary dominant stage, they were absent in the ileum and cecum at 60 hr postinoculation (PI). A few microgamonts were detected in the ileum at 36 hr PI and in the BF on day 19 PI. Oocysts were observed in the ileum at 48 hr PI and in the BF on day 19 PI.  相似文献

Among the chicken breeds, White Leghorn (WL) and White Plymouth Rock (WR) are major breeds and have different history in their establishments. Whole mitochondrial DNA of the breeds were sequenced in order to elucidate the genetic relationship between the breeds. The lengths of the two WL and two WR mitochondrial DNA were found to be 16 788 and 16 785 base pairs, respectively. When the DNA sequences were compared, the similarity was found to be 99.96% (six nucleotide differences). In addition, the present study conformed the existence of an extra nucleotide ‘C’ in NADH dehydrogenase subunit 3 (ND3) of the chicken mitochondrial DNA, which has been consistently observed in Galliformes.  相似文献

An outbreak of subcutaneous sarcomas in commercial White Leghorn egg layers was observed in the northeastern United States during late 2004. Subcutaneous tumors were confined to three flocks distributed in two locations and belonging to the same company. The tumors were first observed grossly by farm personnel at approximately 7 wk of age and persisted throughout the economic life of the flocks. Most of the tumors observed during the growing period were present on the facial region or around the head, wings, and legs. There was no gross evidence of bursal or visceral involvement. Microscopically, most tumors were undifferentiated sarcomas and myxomas. There was no microscopic evidence of Marek's disease or lymphoid leukosis. Reticuloendotheliosis virus proviral DNA was not detected by polymerase chain reaction either in tumors or in cell cultures. Egg production and mortality rates were within normal limits in the affected flocks and many of the chickens exhibiting tumors seemed healthy otherwise, albeit approximately one-half of the daily mortality exhibited tumors. Avian myeloblastosis-associated virus type 1 (MAV-1) was isolated from tumors, plasma, and serum. Upon initial virus neutralization, the viruses isolated seemed at least partially related antigenically to avian leukosis virus (ALV) subgroups A and B but not to subgroup J (ALV-J). Sequencing of the variable and hypervariable regions of gp85 in the envelope gene revealed that the viruses involved are closely related to MAV-1. Attempts to reproduce subcutaneous sarcomas with MAV-1 isolated from White Leghorn chickens in the case herein reported produced exclusively myelocytomas indistinguishable histologically from those induced by ALV-J in meat type chickens.  相似文献

Specific-pathogen-free 10-week-old chickens were inoculated via the air sac with Escherichia coli and showed lymphocytic depletion of bursa of Fabricius and thymus. In experiment I, chickens were necropsied at 12 and 24 hours, 2, 3, and 5 days after inoculation. At 12 hours after inoculation there was lymphocytic depletion in the medulla of lymphoid follicles of the bursa. At 24 hours after inoculation there was lymphocytic depletion also in the cortex of follicles and edema in interfollicular interstitium and follicular medulla. At 2 and 3 days after inoculation there were more marked lymphocytic depletion in medulla and cortex, and fibrosis in interfollicular interstitium. Partial repopulation of follicles with lymphocytes was seen at 5 days after inoculation. In the thymus, lymphocytic depletion occurred in the cortex. At 12 hours after inoculation, lymphocytic necrosis increased in number more than that of control chickens. The width of the cortex and medulla decreased. At 24 hours after inoculation, lymphocytic necrosis increased further. At 2 to 5 days after inoculation, the boundary between the cortex and medulla of lobules was obscure and cellular elements of the cortex and medulla were mingled. In experiment II, chickens were necropsied as in experiment I and also at 8 and 14 days after inoculation. The relative weights of the bursa and thymus reduced rapidly to minimal relative weights at 8 days after inoculation. At 14 days after inoculation, both bursa and thymus had normal relative weights and histological structures. These findings indicate that E. coli infection may induce transient lymphocytic depletion of lymphoid tissues in the chicken.  相似文献

Crossbreeding parameters of immune response traits were estimated from a set of well characterized crossbred populations derived from three chicken lines selected over 12 generations for three different general immune response traits and their F1, F2 and backcrosses. The three traits investigated were the selection criteria from each of the lines, i.e. antibody response to the Newcastle disease virus vaccine 3 weeks after vaccination (ND3), cell-mediated immune response (response to phytohemagglutinin, PHA) and phagocytic activity measured as carbon clearance (CC). Crossbreeding parameters included direct and maternal additive line effects, direct and maternal heterosis as well as direct epistatic recombination loss. They were estimated as linear combinations of genetic group effects estimated using animal model methodology. Significant line differences were obtained for ND3 and, to a lesser extent, CC. They were mainly due to direct effects, maternal effects being significant for none of the 3 traits. Significantly negative direct heterosis effects were also observed for ND3 and CC, but not for PHA. Maternal heterosis effects were not estimated for CC. They were non significant for PHA, and negative and significant (− 0.78 ± 0.24) for ND3. The significant favourable recombination gain estimated for ND3 (3.21 ± 0.88) indicates that epistatic interactions could be important for this trait.The present work shows that it was worthwhile to complete second generation crosses to be able to assess to what extent immunity gained by selection is maintained in advanced crossbred generations, and to compare the transmission of immune traits implicated in different aspects of immunity.  相似文献

1. The rate of inbreeding and its effect on reproduction and egg production traits were studied in White Leghorn lines selected for egg production traits. The experiment was carried out for 10 generations in a control line (C) and in lines selected for increased egg number (EN), egg weight (EW) and egg mass (EM). 2. Data were available on reproduction traits, such as percent fertile eggs (PF), percent hatched of fertile eggs (PHF) and percent hatched of total eggs set (PHT), and on egg production traits such as age at 1st egg (AFE), egg number and egg weight. 3. The rate of increase in average inbreeding per generation was 1.50, 1.24, 1.14 and 0.18% for the line EN, EW, EM and C, respectively. The effect of inbreeding on reproduction and production traits was estimated by including the inbreeding coefficient of the hen (Fh), embryo (Fe) and mate (Fs) as a partial linear regression in the model. 4. There was a significant effect of inbreeding on reproduction traits in line EW attributable to the inbreeding of the hen, embryo and mate. No such effect was observed in the other lines. 5. In all lines inbreeding tended to reduce egg number and delay sexual maturity. In general, all lines reacted differently to inbreeding.  相似文献

1. The effects of restricted feeding in the rearing and laying periods on the performance of three White Leghorn cockerel by Australorp pullet crossbred strains and one White Leghorn strain cross were studied in two experiments.

2. After feeding ad libitum in the rearing period, mean body weights of the three crossbred strains at 20 weeks of age were 1.75, 1.66 and 1.55 kg and that of the White Leghorn strain cross was 140 kg. The differing restrictions during rearing reduced mean body weight at 20 weeks by 14 to 33% and increased the time to sexual maturity by 8 to 23 d.

3. All restriction regimes during rearing increased mean egg weight and tended to reduce mortality in the laying period. Optimum hen‐day egg number over 64 weeks of lay was obtained following mild restriction (14% body weight reduction) of the two heavier strains or ad libitum feeding of the lighter strains. Over 48 or 64 weeks of lay, egg number per hen housed (at 18 to 20 weeks of age) was optimised after moderately restricting (14 to 21% body weight reduction) the crossbred strains or feeding the White Leghorn strain cross ad libitum.

4. Over 64 weeks of lay, mortality was reduced from 19.3% to 10.5% in the lightest crossbred strain, in which a high proportion of deaths were associated with Marek's disease and lymphoid leucosis, by restriction during rearing.

5. Restricting food intake by 7 or 8% throughout the laying period reduced hen‐day and hen‐housed egg number and mean egg weight of all four strains. The reduction in hen‐day egg number associated with food restriction during lay was less for the heavier strains that had also been restricted during rearing.

6. Metabolisable energy intakes required to support maximum production over 64 weeks of lay were 1.23 to 1.36 MJ/d.  相似文献
