Anthracnose of cashew (Anacardium occidentale) was studies on various genotypes and locations in Mozambique. Colletotrichum gloeosporioides was identified as the anthracnose causal agent using polymerase chain reaction. The relationships between incidence and severity of anthracnose on cashew genotypes were statistically analyzed by regression. Anthracnose leaf incidence, which is practically easy to evaluate, was consistently associated with leaf severity, and their relationships can be estimated using the restricted exponential function across locations, crop seasons, genotype and fungicide trials. Pooled data enabled estimation of initial incidence of 1.43% with percentage variance accounting for 83.2 and standard error of 8.3. By computing incidence data into the summary equation, 24 changes of 0, 1, 5, 10 and 40%, resulted in changes of severity estimates of 0.01, 0.05, 0.10, 0.50 and 1.00%, respectively. The maximum disease incidence was estimated as 80% when the severity reached only 5%. Increase in severity was observed afterward, approached a maximum of 25% when leaf detachment is observed. The use of incidence data for epidemic comparisons, genotype and fungicide evaluation in cashew orchards is recommended. Anthracnose incidence on leaves however, could not predict incidence on nuts.  相似文献

Summary Increased virulence associated with fungicide resistance and variablePhytophthora infestans populations has been recorded in many potato growing regions with enormous economic effects. The current emphasis on disease management in East Africa includes the use of potato varieties with durable resistance to late blight. Seven promising clones from Population B with quantitative resistance (no R-genes), two advanced clones from Population A (with Rgenes) and three control varieties were grown for three cropping seasons in order to determine their reaction and stability of late blight resistance. Late blight occurrence was detected in all cropping seasons. The analysis of variance of disease data (AUDPC) for genotypes, locations x seasons was highly significant (P<0.001) indicating the differential response of the geno-types and the need for stability analysis. The Additive Main Effects and Multiplicative Interaction (AMMI) statistical model, showed that the most stable genotypes were 392127.256, 381471.18, 387121.4 (resistant) and 391049.255 (susceptible to late blight). Within environments, the ranking of genotypes was not consistent. The variety Kabale was ranked as the most susceptible genotype while clone 381471.18 and Rutuku were ranked the most resistant. Selective deployment of resistant and stable varieties is critical in minimizing economic loss and damage attributed to late blight in low input farming systems.  相似文献

Potato varietal resistance to bacterial wilt disease caused by Ralstonia solanacearum Yabuuchi et al., (Microbiology and Immunology 39:897–904, 1995) is the best management of the disease. Because the causal pathogen exhibits strong host-pathogen-environment interaction, screening the potential parents for resistance under the target growing environmental conditions is the first important step for effective resistance breeding. The objective of this study was to determine the response to bacterial wilt of selected potato genotypes currently grown by farmers in Kenya and candidate clones from the International Potato Center to identify parents that can be used in the local breeding program to develop resistant cultivars. A study was carried out at Kabete, Kenya for three consecutive seasons between November 2011 and February 2013. Thirty six potato genotypes were planted on an inoculated field at the Kenya Agricultural Research Institute (KARI), Kabete using alpha lattice experimental design with three replications. Data collected were days from planting to onset of wilting (DTOW), bacterial wilt incidence (BWI), total tuber weight (ton ha?1) (TTW), total tuber numbers/hectare (TTN), proportion of ware sized tubers (PWTTW), proportion of symptomatic tubers based on weight (PSTTW), proportion of symptomatic tubers based on tuber numbers (PSTTN) and latent infection (LI) of the tubers. Almost, all the potato genotypes evaluated in this study were susceptible to bacerial wilt. Ranking of genotypes based on resistance differed among the three seasons. On average, the three most resistant genotypes were Kenya Karibu, Kenya Sifa and Ingabire. The study identified eight potato genotypes (Meru, Ingabire, Kenya Karibu, Sherekea, Kihoro, Tigoni, Bishop Gitonga and Cangi) to be used as promising parents for subsequent crosses. The chosen genotypes are prolific in pollen production and popularly grown by Kenyan farmers.  相似文献

Pseudocercospora macadamiae causes husk spot of macadamia. Husk spot control would be improved by knowledge of the persistence of the pathogen in orchards between seasons, and by characterising macadamia cultivars for susceptibility to infection, premature fruit abscission, and spatial escape from inoculum sources. Field observation of seven macadamia cultivars found a significant exponential relationship between husk spot incidence and the prevalence of fruit pericarp that failed to abscise (sticktights) in the canopy. Viable conidia of P. macadamiae were readily (>100 conidia per fruit) produced on sticktights for 30 months. The incidence of husk spot was up to four times greater in trees with sticktights than trees without sticktights. Assessment of the propensity to form sticktights, husk spot incidence and fruit abscission after the insertion/removal of sticktights, demonstrated differences in macadamia cultivars; cv. A16 was susceptible to infection and formation of sticktights, but tolerant of premature abscission; cv. 246 was susceptible to infection, tolerant of premature abscission, and spatially escapes P. macadamiae inoculum due to a lack of sticktights; cv. A38 was susceptible to infection, premature abscission, and sticktight formation. Our findings suggest that disease could be reduced by removing sticktights from trees, and that fungicide inputs could be reduced in trees with naturally low numbers of sticktights. In addition we show that sticktights can be used as an inoculum source for field assays to better characterise macadamia cultivars for better informed cultivar selection, and as a tool in breeding programs to screen for germplasm with high levels of resistance.  相似文献

Fusarium basal rot (FBR) caused by Fusarium oxysporum f. sp. cepae (Foc) is one of the most significant production constraint to shallot. Field experiment was conducted in a naturally Foc infested soil at Debre Zeit Agricultural Research Center during 2006 and 2007 cropping seasons to evaluate the level of resistance of sixteen shallot genotypes against FBR disease. Treatments were arranged in randomized complete block design with four replications. The genotypes significantly varied in their susceptibility to FBR and yield. They were grouped into tolerant, moderately and highly susceptible types. Five genotypes (DZ-Sht-168-1A, DZ-Sht-157-1B, Huruta, Negelle and DZ-Sht-169-1b) were identified to be tolerant as they had reduced disease severity levels from 26.8 to 32.5% and increased mean yield by more than 5 t ha−1 compared to highly susceptible genotypes (DZ-Sht-076-4, DZ-Sht-201-1C and DZ-Sht-054-3A). Among the tolerant genotypes, DZ-Sht-169-1b had greatly reduced bulb rot incidence by 48% in ground storage and 30% in wire mesh shelf as compared to highly susceptible genotype DZ-Sht-201-1C. The tolerant genotypes have high yielding characteristic, and farmers could adopt them for cultivation where FBR is a problem.  相似文献

Tests were made on lines ofSolanum tuberosum spp.andigena to determine 1) whether resistance to potato virus Y exists in the Andigena germplasm, 2) the nature of its inheritance, and 3) the type of resistance. Results of the isolation and identification studies indicated that the pathogen involved was a common strain of potato virus Y. Under field conditions susceptible plants frequently escaped infection. However, field exposures over two seasons resulted in the same ratios as tests in which the same progenies were mechanically inoculated. Mechanical inoculation at the seedling stage proved to be a reliable means of transmission and resulted in accurate screening for resistance. Of 641 tub × tub clones tested, all were susceptible to the virus. Of 366 tub × adg clones tested, 170 were resistant and 196 were susceptible. This fits a 13:15 ratio, assuming random chromatid segregation and a single dominant gene conferring resistance. Plants that were resistant following mechanical transmission were also resistant when inoculated by aphids, indicating the reliability of mechanical transmission as a means of screening for resistance. To determine the type of resistance, top-graft and approach-graft tests were made. Failure to recover the virus from resistant plants inoculated by either grafting method suggests that immunity is the type of resistance involved.  相似文献

This study aimed to identify new sources of resistance to Verticillium wilt in olive. We evaluated various types of genotypes: wild olive trees (Olea europaea subsp. europaea var. sylvestris), genotypes belonging to related subspecie (Olea europaea subsp. guanchica) and genotypes coming from crosses between Picual cultivar and wild olive trees. Fifty-six genotypes were inoculated by dipping roots and then screened under controlled conditions to test their resistance to a highly virulent Defoliating isolate of Verticillium dahliae. Picual (susceptible) and Frantoio (resistant) were control cultivars. Wide variability and significant differences were obtained in the evaluated disease parameters. The Relative Susceptibility Index (RSI), summarizing the disease parameters, was used for final classification of genotypes. Thirteen genotypes were categorized as resistant: eight wild olives from different locations, two genotypes belonging to subsp. guanchica populations from Canary Islands and three genotypes obtained from one of the crosses Picual x wild. The identification of high levels of resistance to V. dahliae among wild olive genotypes may be helpful for the management of this disease. The resistant genotypes could be used as rootstocks for susceptible olive cultivars or parents in future breeding programs.  相似文献

Narrow-leafed lupin (Lupinus angustifolius L.) is a grain legume well adapted to sandy soils of a Mediterranean climate and is the major grain legume in Australia. Improved herbicide tolerance of lupin cultivars is now considered as top priority by the industry for effective weed management. This paper reports induced mutation by seed mutagenesis in metribuzin-sensitive but anthracnose resistant cv. Tanjil, selection for metribuzin tolerant mutants in the field and evaluation of the tolerant mutants to metribuzin and anthracnose resistance. Two highly tolerant mutants Tanjil-AZ-33 and Tanjil-AZ-55 were identified and they were the first reported metribuzin tolerant mutants. The estimated mutation rate was 2.5 × 10−5. Tanjil-AZ-33 was six times and Tanjil-AZ-55 was four times more tolerant to metribuzin than the original cultivar Tanjil and they were also more tolerant than the tolerant cv. Mandelup. Tanjil-AZ-33 was the most tolerant lupin germplasm. Disease nursery assessment over 2 years revealed that both mutants also retained anthracnose resistance. Seed yield under irrigation was 4.2 t/ha for Tanjil-AZ-33 and 1.9 t/ha for cv. Tanjil when the seedlings were treated with metribuzin at 300 g/ha at the six-leaf stage. Clearly the mutation process has created new tolerance to metribuzin, but did not alter the anthracnose resistance. The mutant Tanjil-AZ-33 has been used as a parent in the lupin breeding program as a source of both metribuzin tolerance and anthracnose resistance. Seed mutagenesis followed by field selection in M2 proved to be an effective method to develop new tolerance to herbicides.  相似文献

The plasmodiophorid pathogen Spongospora subterranea f. sp. subterranea causes powdery scab disease of potatoes (Solanum tuberosum L.), which severely affects tuber quality. Adequate chemical control of the disease is lacking, so breeding resistant potato cultivars is important. Little is known about the mechanisms or genetics of host resistance, as the epidemiology of the pathogen makes research on the disease difficult. An environmentally controlled disease screen that allows the accurate and repeatable assessment of diseased tubers (rather than a ranking system) is required to study host resistance in more detail. A high-throughput greenhouse disease assay is described that has been used to assess varying levels of tuber disease resistance of potato genotypes for three consecutive Southern Hemisphere growing seasons (2001–2002, 2002–2003, 2003–2004). This assay allows the evaluation of symptoms on tubers to verify resistance, rather than inferring the tuber infection on the basis of root galling or zoosporangial root infection. The assay was used to determine the level of resistance of genotypes of both known and unknown resistance status. The assay clearly distinguished the susceptible and resistant standards, and the results were highly correlated over the three seasons. The most efficient ways of assessing tuber disease susceptibility of genotypes are discussed. The consistency of the results and the ability to screen many genotypes confirms the value of the assay as a tool for early-generation selection in a plant breeding programme.  相似文献

Summary The interaction betweenErwinia carotovora subsp.atroseptica (Eca) andVerticillium dahliae and its effect on symptom development in potato cultivars showing different degrees of resistance to them was examined over two seasons in irrigated fields in a hot, dry climate. Four cultivars were used: Cara, highly resistant to blackleg and tolerant to Verticillium wilt; Pentland Crown also resistant to blackleg but susceptible to Verticillium wilt; Désirée, moderately susceptible to blackleg but tolerant to Verticillium wilt; and Maris Bard, susceptible to both diseases. Seed tubers were inoculated with Eca immediately after harvest in Scotland, to simulate natural infection, and then planted in Israel in soil naturally infested byV. dahliae. In Verticillium tolerant cultivars, Eca infection increased the rate of colonization byV. dahliae and significantly enhanced symptom development, as assessed by suppression of plant height and wilt symptoms. Changes in the fungal pathogenPhytophthora infestans in the United States pose a significant threat to potato production. Sources of resistance to these new genotypes of P.infestans need to be identified for potato breeders to have parental materials for crossing, and the phenotypic stability of late blight resistance in these potato clones needs to be determined. Sixteen potato clones which reportedly have some resistance to late blight were evaluated at eight locations: Florida (FL), Maine (ME), Michigan (MI), Minnesota (MN), North Dakota (ND), New York (NY), Pennsylvania (PA) and Wisconsin (WI) in 1996. Percent infected foliage was recorded at approximately weekly intervals following the onset of the disease at each location. Area under the disease progress curve (AUDPC) was calculated. Clones were ranked for mean AUDPC within location and the nonparametric stability statistics, mean absolute rank differences and variance of the ranks, were analyzed for phenotypic stability. Neither of these statistics was significant, indicating a lack of genotype x environment interaction on the rankings of these clones across locations in 1996. The four clones with lowest AUDPC scores were U.S. clones AWN86514-2, B0692-4, B0718-3 and B0767-2. These clones should be useful parental materials for breeders seeking to incorporate genes for late blight resistance into potatoes.  相似文献

Rhizoctonia solani causes economically important root and hypocotyl diseases in common bean throughout the world. Root health is a vital factor in plant development and root diseases would negatively influence water and nutrient uptake as well as cause direct stand reduction and root rot damage to the crop. An efficient common bean screening method to evaluate damping-off and early root/hypocotyl damage from R. solani was developed and used to identify dry bean lines with levels of resistance to this disease. Two sets of 163 and 111 lines previously evaluated for drought tolerance in Nebraska and Puerto Rico were evaluated for damping-off resistance and early root/hypocotyl damage under greenhouse conditions. Disease severity on plants was identified based on above-ground symptoms, seedling survival and root lesions using a rating scale of 1 (resistant) to 9 (susceptible). In the first set of lines representing commonly grown dry bean cultivars, germplasm and sources of damping-off resistance, the Rhizoctonia mean rating ranged from 1.7 to 3.9; Phaseolus vulgaris lines PI 310668 and PI 533249 had the highest damping-off resistance. In the second set of the best lines from a drought tolerance shuttle breeding program the Rhizoctonia mean rating was between 2.6 and 5.7. The availability of drought tolerant dry bean lines allowed the testing of the hypothesis that there was a correlation between selecting for drought tolerance and R. solani damping-off resistance. No correlation between mean disease rating and drought tolerance was found, but adapted dry bean lines such as NE14-08-176 released as SB-DT1, and NE14-08-225 were identified with moderate damping-off resistance and drought tolerance. Lines with both traits and other attributes will facilitate development of resistant bean cultivars to manage damping-off caused by R. solani.  相似文献

Wild potatoes are important sources of genes for resistance to disease and insect pests. A collection of wild Mexican and South AmericanSolarium species from the US potato Genebank was evaluated under laboratory and/or field conditions for their reaction to late blight (Phytophthora infestans), Colorado potato beetle (CPB,Leptinotarsa decemlineata Say), and blackleg (Erwinia carotovora subsp.atroseptica (van Hall) Dye) in order to identify individual genotypes with multiple resistance genes. Late blight inoculations using aggressive isolates (US-8/A2 and US-11/A1 mating types) of P.infestans revealed a wide range of variation for resistance between and within the accessions of the wild species tested. For late blight, susceptible as well as moderately to highly resistant genotypes were observed in all the species tested. However, at least one accession from the three Mexican and one South American wild diploid species tested showed a relatively uniform high level of resistance toP. infestans. These includedS. bulbocastanum, S. pinnatisectum, S. cardiophyllum, andS. circaeifolium. Two accessions from South American speciesS. commersonii were highly susceptible to late blight. For the Colorado potato beetle test, only one species,S. pinnatisectum appeared uniformly resistant to CPB under field conditions. Results of screening for blackleg resistance showed that there were major differences between genotypes in the wild species. Accessions ofS. circaeifolium PI 498119 andS. bulbocastanum PI 243504 were identified as having significantly higher blackleg resistance than cultivated potato and the other wild species tested. However, genotypes from these two accessions were more susceptible to late blight and CPB. Characterization of theP. infestans isolate P1801C.16 used for late blight evaluation and multi-locus isolate tests using US-8/A2 and US-11/A1 races revealed that the resistance inS. pinnatisectum genotypes tested corresponded to a race-non-specific genetic system, which was different from any existing R genes.Solanum pin-natisectum genotypes with both high levels of late blight and CPB resistance as well as blackleg resistance genotypes identified in the present study represent a diverse gene pool that may be useful for development of new potato cultivars with multiple disease and insect resistance. The potential utilization of these valuable sources for improvement of cultivated potato is discussed.  相似文献

The objective of this work was to assess the effect of the incidence of cassava anthracnose disease (CAD) on the performance and yield of cassava cultivars planted in Owerri, Imo state. Eighteen cultivars were planted in a randomized complete block design and replicated three times over a period of 3 years. Cultivars TMS 30211, TMS 30001, 98/0510 and TMS 4(2)1425 were regarded as resistant due to the possession of more than three desirable characteristics such as the least size of cankers and absence of cankers in the first and second trials (2003/04 and 2004/05) respectively, lowest severity score and highest weight of healthy fresh tuber and stems. They are recommended for use as parent materials for breeding for resistance to CAD. Cultivar Akwakwuru had the largest size of cankers on whole plant, highest fresh weight of infected tubers and stems which led to low yield and less planting materials. It is regarded as susceptible and may lead to spread of CAD. The correlation of weight of infected fresh tubers on size of canker on whole plant, young stem and maturing stem were positively and highly significantly correlated (r = 0.645, r = 0.470, r = 0.583 and r = 0.622, respectively).  相似文献

A rapid method of testing direct-seeded rainfed rices (Oryza sativa L.) for drought reaction, concurrently with, but separately from, evaluation for agronomic traits and reaction to various stresses, is needed for early identification of desirable genotypes. Thus, a technique which quantifies soil moisture stress for screening thousands of cultivars and breeding lines for drought tolerance during the vegetative stage was developed. Seeds were drilled on dry granular clay soil (Andaqueptic Haplaquoll) in the field, then sprinkler-irrigated every 4 days for 37 days. As the soil dried, each entry was visually scored for drought reaction at 0.1–0.2, 0.4–0.5 and 0.8–1.0 MPa soil moisture tension (smt) at 20-cm soil depth. Entries were visually scored for drought recovery 10 days after the field was reirrigated. A total of 38 691 rices was tested in the dry seasons from 1978 to 1985. The 8-year screening test showed reproducibility of results in the field as indicated by the consistent scores of drought-tolerant and susceptible control cultivars within and across years. Ten germplasm accessions repeatedly performed similar to, or better than, the drought-tolerant control. There were ten outstanding drought-tolerant breeding lines, six of which were progenies of moderately tolerant Nam Sagui 19 from Thailand. The 20 outstanding selections likewise had consistent drought-tolerant scores across years, irrespective of duration of stress, which ranged from 31 to 54 days.  相似文献

Late blight, caused byPhytophthora infestans, is the most important disease of potato worldwide and foliar resistance is an important component of managing late blight in the field. The objective of this research was to identify germplasm for use in breeding cultivars with foliar resistance toP. infestans. More than 500 clones were tested from 1997 to 2002 in inoculated (US8 genotype) field experiments conducted at the Michigan State University Muck Soils Research Farm in Bath, Michigan. All of the current commercial cultivars tested were classified as susceptible toP. infestans. The most resistant clones were A90586-11, AWN86514-2, B0718-3, Jacqueline Lee (MSG274-3), MSI152-A, MSJ307-2, MSJ317-1, MSJ453-4Y, MSJ456-2, MSJ456-4, MSJ461-1, MSK101-2, MSK128-1, NY121, LBR8, LBR9, Tollocan, and Torridon. Some of these resistant selections were from crosses with B0718-3, Jacqueline Lee, and Tollocan suggesting that the resistance toP. infestans was transmissible. These resistant clones will provide the opportunity to breed late-blight-resistant cultivars from a diverse pool of cultivated germplasm. Consistent foliar reaction toP. infestans over years suggested that the Michigan State University Muck Soils Research Farm is a valuable location for North American breeders to assess the reaction of potato germplasm to the US8 genotype of late blight.  相似文献

Bacterial soft rot is a serious disease in potato (Solanum tuberosum L.), causing rapid tuber tissue maceration and, consequently, marketable yield loss. Soft rot bacteria, including Pectobacterium carotovorum subsp. carotovorum (Pbc), are favored by moist conditions, which are prevalent in large potato storage facilities. However, although most potatoes in North America are stored before use, there are no published surveys of soft rot resistance in cultivars exposed to long-term storage conditions. Thus, we tested 65 cultivars and 13 breeding lines for soft rot resistance after 6 months of storage. There was a significant effect of cultivar and production environment on soft rot resistance score. During 6 months of storage, tuber soft rot resistance in resistant clones did not change, while it changed in susceptible clones. The three most resistant cultivars to soft rot were Freedom Russet, Anett, and Alaska Red Eye.  相似文献
