The hormone GnRH has a stimulatory effect on gonadotropin synthesis and secretion. The objective of the first study was to evaluate concentrations of FSH and LH in plasma of boars after successive treatment with SB75, a GnRH antagonist. Thirteen boars greater than 1 yr of age (eight White Composite [WC] and five Meishan [MS]) were injected once daily with SB75 (10 microg/kg of body weight) for 4 d. Plasma concentrations of LH and testosterone (T) decreased after 1 h from the first dose of SB75. After 12 h of treatment, LH gradually returned to pretreatment concentrations, but T remained suppressed (< 2 ng/mL) until after the last injection of SB75. There was a modest, but significant, reduction in FSH during treatment with SB75. The prolonged inhibitory effect of SB75 on suppression of plasma T concentrations, in the presence of pretreatment concentrations of LH, implied direct effects of SB75 at the testis. In the second experiment, testicular tissue from adult boars was incubated in the presence of three doses of human chorionic gonadotropin (hCG; 0, .5, and 5 IU) with SB75 (250 ng/mL) or with Deslorelin, a GnRH agonist (500 ng/mL). Samples of media were collected every hour for 3 h, and concentrations of T and estrone (E1) were determined by RIA. Concentrations of T and E1 increased with time in response to treatment with hCG. Co-treatment with SB75 decreased media concentrations of T (P < .01) and E1 (P < .03) compared to controls (77.9 vs 85.7 +/- 2.0 and 4.7 vs 5.3 +/- .2 ng/g). In contrast, treatment with Deslorelin had no effect on the amount of T (P > .50) or E1 (P > .26) released with all dosages of hCG. These results indicate that a GnRH antagonist has a direct effect on the testis, decreasing amounts of T and E1 released from the Leydig cells; however, treatment with a GnRH agonist had no direct effect on release of these gonadal steroids. Thus, it remains unresolved whether the site of action of GnRH antagonist on testicular steroidogenesis is through a testicular GnRH receptor or through some other mechanism. 相似文献

The inverse relationship of testicular size and circulating follicle-stimulating hormone (FSH) concentrations has been documented, and accompanying this relationship is the change in color of the parenchymal tissue of the testes. Large testes (300 to 400 g) are pink to light red and small testes (100 g) are dark maroon with color gradations for weights in between. It was hypothesized that this color most likely represented an iron protein. Chromatographic analysis of testicular tissue indicated that the Fe was associated primarily with ferritin, and immunohistochemistry showed that Leydig cells were the primary location of ferritin storage within the testes. Concentrations of Fe and ferritin were higher in small testes and decreased as testes weight increased (P < 0.05). As testicular Fe concentrations increased, daily sperm production (DSP) and total DSP declined (P < 0.05). Genotyping six generations of Meishan x White composite boars (n = 288) for a quantitative trait locus that is indicative of elevated FSH and small testes in boars indicated that the Meishan genotype had elevated testicular iron concentrations and darker color in conjunction with reduced total DSP (P < 0.01). It is not thought the elevated iron concentrations affect testicular weights but are probably a result of elevated FSH and FSH inducement of Fe transport. The storage of Fe in Leydig cells may provide a reservoir of Fe for easy access by Sertoli and germ cells, but still provide a degree of protection to germ cells from ionic iron. 相似文献

Testicular tissue and blood samples (V. spermatica interna) were taken from 32 boars during castration. The animals were of different age groups. Against this background, comparative studies were conducted into the relationships between testicular and plasma testosterone. A very close correlation was found to exist between the androgen values in testicular tissue and those in the blood plasma of V. spermatica interna (r = 0.9795), wich appeared to support the conclusion that by determination of blood plasma the androgen content in the testes can be established with high probability. 相似文献

Duroc, Meishan, Fengjing, and Minzhu boars were mated to crossbred gilts during two breeding seasons. From each sire breed group each season, six pens of approximately eight barrows each were slaughtered. A pen of pigs from each sire breed group was slaughtered at 7-d intervals from 168 to 203 d of age each season. Breed of sire effects were significant for all age-adjusted carcass traits except carcass length, fat thickness at the last rib, color score, and firmness score. At 184 d of age, Duroc crosses had the heaviest (P less than .05) slaughter and carcass weights; Minzhu crosses were lighter (P less than .05) than Meishan crosses but not lighter than Fengjing crosses. Differences among age-constant traits reflect differences in BW. After adjustment to a constant carcass weight of 78 kg, the three Chinese breeds had very similar carcass characteristics. Carcasses sired by Durocs had significantly less backfat and larger longissimus muscle area than carcasses sired by the Chinese breeds. Weight of each trimmed wholesale lean cut and their total weight were significantly higher for Duroc crosses than for Chinese crosses. Breed of sire means did not differ significantly for belly weight, but Duroc crosses had less (P less than .05) weight of leaf fat. Relative to Chinese crosses, longissimus muscles from Duroc crosses had more marbling (P less than .05). Sire breed groups did not differ significantly for color or firmness score. Pigs sired by Meishan, Fengjing, and Minzhu produced carcasses with significantly less lean content at a carcass weight of 78 kg than did pigs sired by Duroc. 相似文献

Meishan, Fengjing, Minzhu, and Duroc boars were mated by AI to crossbred gilts to compare the sire breeds for effects on productivity of their mates and performance of their progeny. Gilts were derived from a four-way cross of Chester White, Landrace, Large White, and Yorkshire. At weaning, litters sired by Meishan boars averaged 1.69 and 1.32 more pigs than those sired by Duroc and Minzhu boars, respectively (P less than .05). Pigs sired by Meishan and Fengjing boars averaged 1.2 to 1.8 more nipples than pigs sired by Duroc and Minzhu boars (P less than .05). Minzhu-sired pigs had a lower (P less than .05) survival rate to 14 and 28 d of age than did those sired by Meishan, Fengjing, and Duroc. Fengjing-sired pigs were heavier than Minzhu-sired pigs at birth and heavier than Minzhu- and Duroc-sired pigs at 14, 28, and 56 d of age (P less than .05). At 70 d of age, Meishan-sired pigs were heaviest, followed in order by Fengjing-, Duroc-, and Minzhu-sired pigs. At 98 d of age, Meishan-sired pigs were significantly heavier than all other breed groups and Duroc-sired pigs were significantly heavier than Minzhu-sired pigs. At 126 and 154 d of age, Duroc-sired pigs were heaviest, followed in order by Meishan-, Fengjing-, and Minzhu-sired pigs; all differences were significant. Average backfat thickness of gilts at 99.7 kg was least for Duroc-sired gilts and highest for Meishan-sired gilts. Sire breed groups did not differ significantly in feed efficiency from 70 to 98 d of age. Duroc-sired pigs were the most efficient (P less than .05) from 98 to 126 and 126 to 154 d of age. Results indicate that use of these Chinese breeds in crossbreeding programs will decrease performance traits. This decrease would have to be offset by increases in reproductive efficiency of Chinese sows, use of repartitioning agents, or other management techniques if Chinese pigs are to improve overall production efficiency of a crossbreeding program. 相似文献

Mature boars were subjected to chronic treatment with a gonadotropin-releasing hormone (GnRH) agonist, goserelin (D-Ser[But]6, Azgly-NH210), and serum luteinizing hormone (LH) and testosterone concentrations were measured. Ten sexually mature boars were randomly assigned to treatment (n = 5) or control (n = 5) groups. On day 0, boars were implanted sc (day 0) with 2 GnRH agonist implants (1 mg of GnRH/implant) or sham implants. Blood samples were collected at 12-hour intervals on days -2 and -1, at 6-hour intervals on days 0 through 4, and at 12-hour intervals on days 5 through 8. In addition, blood samples were collected at 15-minute intervals for 6 hours on days -1, 0, 4, and 8. Serum testosterone and LH concentrations were determined by radioimmunoassay. Maximal LH (7 +/- 1 ng/ml) and testosterone (26 +/- 3 ng/ml) concentrations were observed at 5 and 18 hours, respectively, after GnRH agonist treatment. Subsequently, LH and testosterone concentrations decreased to pretreatment values (0.3 +/- 0.1 ng/ml and 1.8 +/- 0.4 ng/ml, respectively) by 24 and 48 hours, respectively, after GnRH agonist implantation. Few differences in the characteristics of pulsatile LH release were observed between the groups. Testosterone and LH concentrations in samples collected at 6- and 12-hour intervals and pulsatile LH release did not change after sham treatment of control boars. Whereas previous reports indicated that chronic GnRH administration suppressed serum LH and testosterone concentrations in rams, rats, and dogs, our results indicate that chronic GnRH agonist treatment induced transitory increases, without subsequent suppression, in LH and testosterone concentrations in mature boars. 相似文献

It is well known that heat stress has a detrimental effect on testicular functions. In addition to the alteration of semen quality and testicular damage, reproductive hormone secretion can be altered. The objective of this study was to describe changes in plasma concentrations of testosterone, LH and cortisol, as well as in testicular ultrasonographic appearance after induced testicular degeneration. Four Swedish Red and White bulls, aged 3 years, were used. They were fed according to Swedish standards. The scrotum was covered with an insulation device during 96 h. Semen was collected weekly 3 times before and up to 4 months after insulation. Testicular ultrasonography and clinical genital examination were performed with the same intervals. Heparinized blood samples were taken from the jugular vein at 2 h interval during 24 h every 2 weeks during the study. Blood samples were tested for the content of testosterone, LH and cortisol. Data were analysed, using one way analysis of variance of seminal data, clinical examination data as well as 24 h hormonal output data as percentage of mean individual pretreatment values. The use of a 5 MHz B-mode ultrasound unit did not contribute with an objective estimation of the degree of testicular degeneration. In 3 of the bulls testosterone levels had a tendency to decrease and LH to increase during the time of severe degeneration, whereas an opposite trend was seen during the regenerative phase, changes becoming significant 15 weeks after scrotal insulation. Variation between animals was big. Effects of chicken thymic stromal cells on the growth and differentiation of thymocytes in vitro 总被引:2,自引:0,他引:2
We examined contact-mediated effects of chicken thymic stromal cells (TSC) on thymocyte differentiation by co-cultivation of these cell populations. The primary cultures of TSC isolated from thymus mainly have consisted of epithelial cells which were polygonal in shape, possessed long processes and expressed MHC class II antigen. When thymocytes were co-cultured with TSC, 60% to 70% of thymocytes attached to TSC and some of them engulfed underneath TSC. These attached thymocytes were CD4-CD8- and CD4+CD8+ subsets and expressed alpha/beta TCRhigh or gamma/delta TCRlow. Some of the thymocytes attaching to TSC showed an increase of intracellular and nuclear density, fragmentation of cytoplasm and nuclei, and DNA fragmentation. And also, thymocytes attaching to TSC contained a higher percentage of cycling (S and G2 + M phase) cells than nonattaching cells. These results indicate that specific subsets in thymocytes selectively bind to TSC and undergo apoptotic death or proliferation because of interaction with TSC. Chicken TSC may play an important role in thymic differentiation by direct contact within the thymus as in mammals. 相似文献

Nine blood samples were taken at 30-min intervals from 36 Landrace X Large White boars at each of eight ages (42, 56, 70, 84, 98, 112, 126 and 140 d). Serum concentrations of luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone (T) and estradiol-17 beta (E2) were quantified by radioimmunoassay procedures. The maximum concentration of LH and the age at maximum concentration were predicted for each boar. Variability of LH samples was described for each boar by the pooled within age variance among LH samples and by the number of LH peaks. Measurements of testicular development taken at 140 d of age included: in situ testis width and length, excised testis weights and histological traits of excised testes (seminiferous tubule diameter, percentage of tubules with a lumen and percentage of tubules with active spermatogenesis). Pooled within line correlations were calculated with data from boars selected for either high or low testis weight. Correlations among the testicular traits ranged from .45 to .88. Luteinizing hormone concentration (mean over all ages) was related to measures of testicular development (r = .24 to .49). Mean concentrations and the occurrence of pulsatile release of luteinizing hormone (LH) were determined in 14-wk-old crossbred boars (50.5 +/- 1.5 kg) after bilateral or unilateral castration at 10 wk of age. Blood was collected at 10-min intervals for 5 h. Then gonadotropin releasing hormone (GnRH; 40 micrograms) was given and sampling was continued at 5-min intervals for 1 h. Compared with intact boars, bilateral castration increased (P less than .001) mean LH (982 +/- 56 vs 389 +/- 56 pg/ml), pulsatile releases of LH (7.0 +/- .6 vs 2.0 +/- .6 pulses/5 h) and LH pulse amplitude (617 +/- 29 vs 360 +/- 58 pg/ml). Unilaterally castrated boars did not differ from intact boars in any of the above measures of LH secretion. Testis weight increased more between 10 and 14 wk of age in the unilateral castrates than in the intact boars (432 +/- 42 vs 245 +/- 34%; P less than .05). Thus, compensatory hypertrophy occurred within 4 wk of castration. Plasma testosterone was lower for bilateral castrates than for intact animals (.1 +/- .8 vs 3.6 +/- .9 ng/ml; P less than .05) while unilateral castrates (3.8 +/- 1.0 ng/ml) and intact boars did not differ. Crossbred boars were used to evaluate the influence of exposure to 8 or 16 hr of light daily from 75 to 175 days of age on growth rate, testicular characteristics and endocrine function. At 160 days of age, concentrations of testosterone in serum (P<.10), the areas under plotted 12 hr testosterone profiles (P<.10) and the number (P<.05) and magnitude (P<.10) of testosterone secretory spikes were increased in boars exposed to 16 hr of light compared to boars in 8 hr light, but concentrations of LH in serum were similar in boars exposed to both treatments. Treatment with GnRH resulted in similar concentrations of LH in serum for both groups of boars. Testosterone in serum after GnRH-mediated LH release was greater at .5 (P<.05) and 1.0 (P<.10) hr following GnRH in boars exposed to 16 hr of light compared to boars at 8 hr, but concentrations of testosterone were similar for both treatments from 1.5 to 4.0 hr after GnRH. Growth rate and testicular and epididymal weights and sperm reserves at 175 days of age were not significantly altered by duration of photoperiod. Boars exposed to 8 hr of light had more hair per unit area than boars exposed to 16 hr of light. We conclude that exposure of prepubertal boars to longer daily photoperiods results in increased concentrations of testosterone in serum at 160 days of age. 相似文献

Investigations were made to identify the causal agent of an acute outbreak of abortions in a domesticated herd of wild boar. Only porcine parvovirus (PPV) was isolated from samples of organs from the still-born sucklings and mummified aborted fetuses. The isolated virus hemagglutinated erythrocytes of guinea pig, murine, rat, and chicken. Identity of the virus, designated the BQ strain, was confirmed by the production of a specific cytopathic effect on susceptible cells and by the results from ELISA, PCR, immunofluorescence assay, and electron microscopy. PPV BQ strain was adapted to growth in a swine testicular cell line. When inoculated into healthy sows, PPV BQ caused the same reproductive disorder observed in the affected herd. 相似文献

Steroid test substance 547 and Norgestrel were used in a combination treatment of parts of the hypothalamo-adenohypophyseo-gonadal axis of boars. The quantifiable parameters of the morphokinetic effect thus produced (cell nucleus volumes of Nucleus praeopticus medialis hypothalami and its neurons as well as those of Leydig's cells testicle weight, length and diameter of tube, percentage of Leydig's cells, testicle weight, length and diameter of tube, percentage of Leydig's cells) did not differ significantly from the findings obtained from boars treated with Norgestrel only (cf. Dorst and co-workder, 1978). The same combination treatment, however, differed from exclusive Norgestrel treatment by causing the occurrence of socalled castration cells, which led to the assumption of an anti-androgenic effect of such combination treatment. 相似文献
