This review considers the possible events that can occur when cattle are exposed to Mycobacterium bovis and, where appropriate, draws on principles accepted for tuberculosis infection in humans and laboratory animal models. Consideration is given to the many complex factors which influence the outcome of challenge with tubercle bacilli. These include features inherent to the mycobacterium, the host and the environment. It is apparent that clinical disease probably occurs only in a relatively small, but undetermined, proportion of cattle that are exposed to Al. bovis. The majority of animals may clear infection or control the bacilli, possibly in a condition of latency. It is concluded that a better understanding of the dynamics of the events following M. bovis exposure and subsequent infection in cattle would be of significant benefit in developing new tools appropriate for disease control and to designing optimal approaches for their application.

A case control study was conducted between October 2004 and April 2005 to determine the prevalence of bovine tuberculosis (BTB) in cattle in central Ethiopia relative to the tuberculosis status of their owners. A total of 174 farmers (87 with active tuberculosis and 87 with no active tuberculosis), and 1041 cattle (506 owned by farmers with active tuberculosis and 535 by farmers without active tuberculosis) were included. The comparative intradermal cervical tuberculin test was used in cattle while clinical symptoms, chest X-ray and Ziehl-Neelsen staining were used for the diagnosis of tuberculosis in the farmers. In addition, mycobacterial culture, biochemical tests, and drug susceptibility tests were performed for the identification Mycobacterium spp. from both humans and cattle. The prevalence of BTB was threefold higher (odds ratio [OR]=4.2, 95% confidence interval [CI]=2.79-6.2) in cattle owned by farmers with active tuberculosis (24.3%) than in those owned by farmers who did not have active tuberculosis (8.6%). Cattle owned by farmers with active tuberculosis were four times more likely to have tuberculosis than cattle owned by farmers with no active tuberculosis. Furthermore, cattle owners who consumed raw milk were at greater risk (chi2=14.1, P<0.001, OR=3.34) of having active tuberculosis than those who consumed boiled milk. Of the 42 human isolates, 31 (74%) were Mycobacterium tuberculosis, seven (16%) were Mycobacterium bovis while four (10%) were considered a typical mycobacteria on the basis of biochemical and drug sensitivity tests. Of the 11 cattle isolates, two (18%) were Mycobacterium tuberculosis, five (46%) Mycobacterium bovis, and four (36%) were atypical mycobacteria. The prevalence of tuberculosis was higher in cattle owned by farmers with active tuberculosis than in cattle owned by farmers who did not have active tuberculosis, which could suggest possible transmission of Mycobacterium spp. between cattle and their owners.

In a seroepidemiological survey using an indirect haemagglutination assay, the prevalence rate of toxoplasmosis in central Ethiopia was 22.9% of 899 sheep, 11.6% of 753 goats and 6.6% of 785 cattle. There were high titres of 1:256 or more which suggest current infections. These results indicate that toxoplasmosis may be an important cause of reproductive wastage in small ruminants. The public health significance of this disease is discussed. Improved hygiene and management could reduce the prevalence of the disease.

The prevalence of bovine tuberculosis (BTB) infection in cattle was investigated in extensive and intensive production systems in three districts of northwestern Ethiopia. Single comparative intradermal tuberculin test (SCIDTT) was used in the study. The prevalence of BTB infection as determined by SCIDTT was 9.7% whereas the non-specific infection prevalence was 10.8%. In the extensive system the prevalence was 8.2% and 11.3%; under intensive system the prevalence was 22.1% and 6.3% for BTB and non-specific infections respectively. The prevalence of BTB was significantly higher in the intensive than extensive production systems. Of the 75 herds tested 41 (54.7%) had BTB infections, 68.9% of the BTB positive herds were in the extensive system and 40% of herds in the intensive systems (small dairy farms) had BTB infections.

Mycobacterium tuberculosis is an old enemy of the human race, with evidence of infection observed as early as 5000 years ago. Although more host-restricted than Mycobacterium bovis, which can infect all warm-blooded vertebrates, M. tuberculosis can infect, and cause morbidity and mortality in, several veterinary species as well. As M. tuberculosis is one of the earliest described bacterial pathogens, the literature describing this organism is vast and overwhelming. This review strives to distill what is currently known about this bacterium and the disease it causes for the veterinary pathologist.

Bovine tuberculosis in domestic livestock such as cattle is an economically important disease with zoonotic potential, particularly in countries with emerging economies. We discuss the findings of recent epidemiological and immunological studies conducted in Ethiopia on host susceptibility differences between native zebu and the exotic Holstein-Friesian cattle that are increasingly part of the Ethiopian National herd, due to the drive to increase milk yields. These findings support the hypothesis that native Zebu cattle are more resistant to bovine tuberculosis. We also summarise the results of experimental infections that support the epidemiological data, and of laboratory experiments that suggest a role for the innate immune response, and in particular interleukin-6, in the outcome of bovine tuberculosis infection.

A 4-year-old male Boxer dog with a history of vomiting, diarrhea, and weight loss moved from West Africa to Lyon, France, where it was further evaluated. Radiographs revealed pleural effusion and enlargement of tracheobronchial lymph nodes and liver. Cytologic examination of the pleural effusion and a fine needle aspirate specimen of the liver showed mixed mononuclear inflammation with nonstaining rod structures within epithelioid histiocytes. At necropsy, the main gross pathologic findings were exudative pleuritis, nodular hepatitis, and infarcts and caseous nodules in the kidneys. The main histologic lesions were granulomatous hepatitis, granulomatous pneumonia, fibrinous leukocytic pleuritis, necrotic and fibro-calcified granulomatous lymphadenitis, and granulomatous nephritis. A Ziehl-Neelsen stain applied to both cytologic and histologic samples was positive for acid-fast bacilli. Bacterial culture of the pleural fluid was positive for Mycobacterium tuberculosis. Cytology is a valuable tool in the diagnosis of this important zoonotic disease.

The transmission of Mycobacterium bovis infection to cattle
The prevalence of Mycobacterium bovis infection in cattle is increasing rapidly in some countries, including the UK and Ireland. The organism infects a wide range of mammalian hosts, and eradication of the disease is difficult if there is an extensive reservoir in the wildlife population. Existing evidence suggests that wildlife vectors include the European badger in the UK and Ireland, the brush-tailed possum and ferret in New Zealand and ungulates in some other countries. Cattle grazing field boundaries or short swards are at particularly high risk, since the chance of contact with the intermediate host or their excreta is increased. There is evidence that the transmission of the disease between cattle following movement accounts for 10-15% of outbreaks in the British Isles and that transmission can occur across farm boundaries. The prevalence the prevalence of single reactors in herds suggested that within-herd transmission was not common. In herds with infected cattle, spreading slurry is a risk factor, which can be minimised by prolonged storage of the slurry, by spreading it on fields not used for grazing or by soil injection. M. bovis also survives in water and may enter the respiratory tract during drinking. It is concluded that M. bovis infection in cattle can be transmitted by a number of routes, some of which can be controlled by appropriate husbandry, but that circumstantial evidence suggests that the existence of a widespread intermediate host is the greatest contributor to infection in cattle.

This study shows a representative stratified cluster sample survey of the prevalence of comparative intradermal tuberculin test in cattle from four regions in Ethiopia. Using a cut-off for positivity of 2 mm, it assesses possible risk factors for tuberculin-positive reaction in cattle. Seventy-three villages in 24 kebeles (administrative units) were randomly selected, from which 2216 cattle from 780 owners were tested. In addition, 450 of these cattle owners were interviewed for risk factor assessment. Ninety-nine percent of the tested cattle in this rural livestock production system were traditional zebus. The individual overall prevalence of cattle bovine tuberculosis (BTB)e was 3%, with the highest found in Meskan Mareko, in Central Ethiopia (7.9%) and the lowest in Woldia, in the North East edge of the Rift Valley (1.2%). Generalised Linear Mixed Models (GLMM) with random effect on kebeles was used to analyse risk factors of cattle reactors and human tuberculosis (TB) infection. Purchase of cattle and presence of other livestock in the herd were statistically significant, with OR: 1.7, p-values of 0.03 and OR: 2, p = 0.05, respectively. Family members diagnosed with TB or showing clinical signs of extra-pulmonary TB (EPTB) were reported in 86 households (19%). None of the assessed potential risk factors of disease transmission between cattle and human (food consumption, livestock husbandry and presence of BTB-positive cattle) were statistically significant.

Fluctuations in the contamination of herbage with trichostrongylid nematode larvae are determined by a number of factors, the most important being climate and management. Practical measures to prevent calves from ingesting large numbers of larvae are based on detailed knowledge of the herbage contamination pattern and its epidemiological consequences. Intensification of animal husbandry methods including increased stocking rates and application of slurry to pasture may increase the hazards of gastrointestinal helminthiasis.

A cross-sectional study of bovine tuberculosis (BTB) was conducted in pastoral cattle herds in southern Ethiopia, from February to August 2008 using the comparative intradermal tuberculin test. The prevalence of BTB and the risk factors for having positive reactor herds were assessed in four pastoral associations in two districts of southern Ethiopia, namely Goro-Dola with 242 cattle in 16 herds and Liben with 231 cattle in 15 herds. A herd was considered positive if there was at least one reactor animal in a herd. The test results were interpreted based on the Office Internationale des Epizooties recommended 4-mm and a recently suggested 2-mm cut-off. The apparent individual animal prevalence of tuberculin reactors was 5.5% (95% confidence interval (CI), 4.0–8.0%) and 7.0% (95% CI, 5.0–10.0%), whereas the true prevalence estimate was 4.4% (95% CI, 0.8–8.0%) and 6.1% (95% CI, 2.6–9.5%), when using the 4-mm and the 2-mm cut-offs, respectively. The overall herd apparent prevalence of tuberculin reactor animals was 41.9% (95% CI, 24.9–60.9%) and 48.4% (95% CI, 30.2–66.9%) with the 4-mm and 2-mm cut-offs, respectively. A positive tuberculin test was associated with the age of animals and the main drinking water sources during dry seasons. In order to investigate the public health risks and the epidemiological importance of BTB in the area, we recommend to include other livestock species (camels and goats) as well as humans in future studies.

Investigations at the slaughterhouse of Limoges (Central France) were performed in 1994-1996 to determine the prevalence of Paramphistomum daubneyi infection in cattle. In 1994/1995, higher prevalences of P. daubneyi were recorded in May, October, and January. In 1996, smaller variations were recorded, and prevalence in April and May was higher. The prevalence of P. daubneyi in summer was lower. Breed and age of cattle were not significantly associated to P. daubneyi infection, but females were significantly more infected than males. P daubneyi infection was related to that of Fasciola hepatica, but not to that of Dicrocoelium lanceolatum.

Fifteen Holstein-Friesian bulls which grazed together for one season (April to November) in Schleswig-Holstein were necropsied after housing for 2 to 6 weeks. The approximately 12-month old bulls weighed 378 to 491 kg at slaughter and did not receive any anthelmintic on pasture. The most prevalent species of the gastrointestinal tract was Ostertagia ostertagi (prevalence; geometric mean worm count; minimum-maximum) (100%; 1,355; 130-4,590) followed by Cooperia (C.) oncophora (86.7%; 343; 20-14,800), Oesophagostomum (Oe.) radiatum (86.7%; 3.7; 1-42), Trichostrongylus axei (80%; 65; 50-910), C. punctata (73.3%; 14.8; 10-140), Trichuris (T.) discolor (73.3%; 4.8; 1-108) and Skrjabinagia lyrata (60%; 8.5; 18-260) which were recorded in more than half of the animals. Less frequently, C. surnabada (46.7%; 18; 10-4025), T. skrjabini (33.3%; 0.7; 2-14), Nematodirus helvetianus (13.3%; 0.5; 20-30), Spiculopteragia b?hmi (6.7%; 0.2; [26]), Capillaria bovis (6.7%; 0.2; [10]) and Oe. venulosum (6.7%; 0.2; [9]) were recovered. Inhibited fourth-stage larvae of the Ostertagiinae were recovered from the abomasal mucosae of all bulls (100%; 13,159; 5,800-56,950). The abomasum harboured the highest worm burden followed by the small intestine and large intestine (geometric means): 15,565, 461 and 10 nematodes, respectively. Inhibited fourth-stage larvae amounted to 90% of the abomasal Ostertagiinae. Dictyocaulus viviparus was recorded in 73.3% of the animals with a geometric mean count of 12.8 (1-822 per animal).

An outbreak of tuberculosis in pigs and cattle caused by Mycobacterium africanum produced lesions in the pigs similar to those caused by M tuberculosis, M bovis and M avium, with caseation in the lymph nodes of the head and in the jejunal lymph nodes. Bacteriological examination of the dysgonic mycobacteria isolated showed that they were M africanum I. The intradermal tuberculin test was very reliable in pigs, differentiating between mammalian and avian reactions, and the results of the test were in accordance with the lesions found at meat inspection. No clinical signs were observed during the outbreak, and the infection was neither serious nor progressive. There were no lesions in the tuberculin-positive cattle. The source of the infection remains unknown.

Dogs infected with Mycobacterium tuberculosis can develop clinical tuberculosis (TB) but there are currently no validated immunological assays for diagnosing this infection in this species. Using a post mortem survey we investigated the prevalence of non-clinical M. tuberculosis infection and clinical TB disease in a high-risk population of dogs and developed and utilised a novel interferon-gamma release assay to determine the risk of transmission of M. tuberculosis from TB patients to contact dogs. The prevalence of clinical TB in dogs from a high-risk setting was 1% (95% CI: 0-5%) while the prevalence of immunological sensitization to M. tuberculosis antigens in dogs living in contact with sputum smear-positive TB patients was 50%. The IGRA proved a useful test of M. tuberculosis infection in dogs and the high levels of transmission of this pathogen from humans to companion dogs should be considered when assessing the zoonotic risks associated with such animals.
