近年来, 番茄病毒病, 特别是番茄黄化曲叶病毒(TYLCV)病在北京地区空前暴发, 给番茄生产造成严重威胁, 使番茄的产量和品质显著降低。2012-2013年在北京周边7个区县, 采集疑似感染病毒的番茄植株样品325份, 分别针对TYLCV、烟草花叶病毒(TMV)、黄瓜花叶病毒(CMV)3种病毒进行了PCR或ELISA 检测。检测结果表明, 目前在北京地区大棚或温室内发生的番茄病毒病以TYLCV为主; 露地番茄病毒复合侵染现象比较普遍, 病毒检出率100%, 其中TYLCV检出率达到75%以上。在TYLCV侵染的样品中, TYLCV和CMV复合侵染占20%左右, TYLCV和TMV复合侵染占15%左右。部分样品检测到TYLCV、CMV 和TMV 3种病毒复合侵染的现象。  相似文献

To identify viruses in Henan tobacco-planting areas, from 2015 to 2017 and 2019, 288 symptomatic tobacco samples were collected and then subjected to small RNA sequencing. Results showed that at least 7 viruses were detected from these samples which including four previously reported viruses, cucumber mosaic virus (CMV), tobacco mosaic virus (TMV), potato virus Y (PVY) and tobacco vein banding mosaic virus (TVBMV). Other three viruses, wild tomato mosaic virus (WTMV), brassica yellows virus (BrYV), and cycas necrotic stunt virus (CNSV) were firstly detected in Henan province. However, tobacco etch virus (TEV) and tobacco ringspot virus (TRSV) were not detected from these samples. In addition, CMV, TMV, PVY, TVBMV, and BrYV were the dominant viruses infecting tobacco in Henan Province.  相似文献

Commercial capsicum fields in Lusaka Province were surveyed for the presence of viruses during 1996-01/06. The survey demonstrated the occurrence of five viruses in 13 fields. Virus identification was based on field disease syndrome, host range studies, DAS-ELISA and electron microscopy. The identified viruses were alfalfa mosaic alfamovirus (AMV), potato Y potyvirus (PVY), tobacco mosaic tobamovirus (TMV), cucumber mosaic cucumovirus (CMV) and pepper mild mottle tobamovirus (PMMV). The most prevalent and widespread virus, recovered from 70% of the surveyed fields, was AMV, and this was followed by PVY and TMV, which were detected in 50% and 40% of the fields respectively. CMV and PMMV were less common and occurred in 30% of fields. The study also showed that PVY could infect capsicum plants alone or in combination with CMV or TMV. The average plant infections caused by AMV was 80–100% and by TMV and PVY 50% and 60% respectively. The disease incidence in the case of CMV was 15–60% and for PMMV 30–40%. This is a first report on the identification of viruses that infect capsicum in Zambia.  相似文献

Viruses in the northern potato-producing regions of Saudi Arabia   总被引:2,自引:0,他引:2  
Agar double diffusion tests and, later, ELISAs were used to detect viruses associated with potato in 242 samples collected in 16 trips to Tabuk and Hail, northern regions of Saudi Arabia, in four consecutive growing seasons (autumn 1989, spring and autumn 1990 and spring 1991). Eleven different viruses were detected in Tabuk and 12 in Hail. The viruses detected in Tabuk were alfalfa mosaic (AMV), cucumber mosaic (CMV), tobacco mosaic (TMV), potato leaf roll (PLRV), tomato spotted wilt (TSWV), tobacco ringspot (TRSV) and potato A, M, S, X and Y. The same viruses, plus potato yellow dwarf (PYDV), were detected in Hail. AMV was most frequently and CMV least frequently detected in Tabuk, whereas in Hail the most and least common were PVA and PLRV respectively.  相似文献

Tomato yellow leaf curl begomovirus (TYLCV) severely invaded tomato plantations in Egypt (Lower and Middle Egypt) in 1989. This study aimed to discover the relationship between TYLCV and other epidemic-associated factors in the Fayium area. The rate of TYLCV infection was inspected visually for three successive years (1994/1996) in the Fayium area. During the same period, whiteflies were collected for virus detection using bait-plant and DNA hybridization techniques. DAS-ELISA was used to detect mixed virus infections in tomato plants. TYLCV infection was prevalent (60–68%) and severe (2.1–3.0) in the Fayium fields. Cucumber mosaic cucumovirus (CMV) was found in some fields (5–28%) with moderate severity (1.0–20). Potato Y potyvirus (PVY) and potato leaf roll polerovirus (PLRV) were found in few fields (5–19% and 5% respectively) at very low severity. There was a negative correlation between TYLCV occurrence and distance from the source of infection, and a positive correlation (98%) between TYLCV intensity and percentage of viruliferous whiteflies in 1994 and 1995. There was no positive correlation between TYLCV and the total population of whiteflies caught during the same period. Five percent of viruliferous whiteflies, as proved by cDNA hybridization, led to 46% TYLCV infection. The same percentage of whiteflies, as shown by bioassay, led to 68% TYLCV infection. Monitoring of viruliferous whiteflies could be used for early prediction of TYLCV infection.  相似文献

Serological tests have been used to detect viruses associated with tomato in 257 samples collected in different regions in Tunisia, Cap-Bon, Sahel and South during successive seasons. The viruses detected were cucumber mosaic cucumovirus (CMV) and tomato aspermy cucumovirus (TAV), potato Y potyvirus (PVY), tobacco etch potyvirus (TEV) and pepper veinal mottle potyvirus (PVMV), tomato mosaic tobamovirus (ToMV) and tobacco mosaic tobamovirus (TMV), tobacco rattle tobravirus (TRV), alfalfa mosaic alfamovirus (AMV), tomato spotted wilt tospovirus (TSWV), tomato ringspot nepovirus (TomRSV) and potato X potexvirus (PVX). Some were detected in all three regions surveyed, at variable frequencies: TMV, CMV, TEV, PVY, ToMV, AMV, TAV, TSWV and TRV. Others were only detected in two regions (PVMV in Cap-Bon and Sahel and PVX in Sahel and in the south) or one region (TomRSV in Cap-Bon). Movement of individual viruses from one region to another may be due to movement of specific vectors, as in the case of the thrips-transmitted TSWV moving from the south to the north. Some of these viruses were found for the first time in Tunisia.  相似文献

Studies on polymer size, concentration and mode of application, either as foliar spray or soil drench, in relation to the induction of resistance to tobacco mosaic virus (TMV) in Nicotiana tabacum cv xanthi-nc by poly(acrylic acid) (PA) are reported. PA also induced resistance to TMV in N. glutinosa, to pelargonium leaf curl virus in Datura stramonium, to cucumber mosaic virus in Vigna sinensis and to tobacco ring-spot virus in N. tabacum cv White Burley. No TMV was detected in PA-treated tomato cv Virocross 11 days after inoculation; but the susceptible cultivar Craigella became infected. PA treatment had no effect on TMV replication in White Burley tobacco but resistance was induced to Peronospora tabacina, a fungal pathogen of N. tabacum cv xanthi-nc. The potential of PA-induced resistance as a control measure for viruses and fungi is discussed.  相似文献

 苏南地处我国东南沿海长江三角州中心。为明确该地蔬菜作物病毒病种类和发生趋势,采用分子生物学核酸检测的方法,对2018年5月至2019年12月,采自江苏南京、无锡、镇江、苏州和常州主要蔬菜作物的232份疑似病毒病样品进行检测鉴定,共检出12种病毒。其中,南京检出7种病毒,无锡检出4种,镇江检出2种,苏州检出6种,常州检出7种。茄科蔬菜中检出11种病毒,葫芦科蔬菜中检出9种,豆科蔬菜中检出7种,以辣椒轻斑驳病毒(pepper mild mottle virus, PMMoV)、番茄褪绿病毒(tomato chlorosis virus, ToCV)、番茄花叶病毒(tomato mosaic virus, ToMV)、瓜类褪绿黄化病毒(cucurbit chlorotic yellows virus, CCYV)、番茄黄化曲叶病毒(tomato yellow leaf curl virus, TYLCV)和辣椒脉黄化病毒(pepper vein yellow virus, PeVYV)检出率最高。鉴定的南方番茄病毒(southern tomato virus, STV)、烟草扭脉病毒(tobacco vein distorting virus, TVDV)、烟草轻型绿花叶病毒(tobacco mild green mosaic virus, TMGMV)和PeVYV在江苏省属首次报道。检测发现,苏南五地(市)蔬菜作物中普遍存在多种病毒复合侵染现象,有的多达5种病毒复合侵染,PMMoV与其他病毒的复合侵染出现频率高且分布范围较广。研究结果有助于了解苏南五地(市)蔬菜作物主要病毒病的种类、分布和发生趋势,为病害防控提供科学依据。  相似文献

Field surveys were carried out in the main vegetable-growing areas of Western and Central Albania to evaluate the sanitary status of open-field and protected cultivations of capsicum, tomato, potato, watermelon, cucumber, courgette, aubergine, lettuce, cabbage, chicory, leek and celery. The following viruses were detected: Alfalfa mosaic alfamovirus (AMV), Cucumber mosaic cucumovirus (CMV), Potato Y potyvirus (PVY), Tomato spotted wilt tospovirus (TSWV) and Watermelon mosaic potyvirus 2 (WMV-2). The virus found most frequently was CMV and all the isolates identified were of subgroup IA. AMV was also detected in several areas and all isolates were of subgroup II, suggesting a French origin. Finally, at the time of this survey, TSWV infections appeared to be moderately relevant and absent in protected crops. This is the first documented record of AMV, CMV, PVY, TSWV and WMV-2 in Albania. No infection by Tomato yellow leaf curl begomovirus (TYLCV) or Zucchini yellow mosaic potyvirus (ZYMV) was detected during this survey.  相似文献

Tomato yellow leaf curl begomovirus (TYLCV), transmitted by the whitefly Bemisia tabaci , is epidemic in Africa, the Middle East and South-East Asia. It is also reported in some European countries and the American continent. In Lebanon, it is the major limiting factor for summer and autumn production of tomato. Comparison of the nucleotide sequence in the intergenic region with other reported leaf curl viruses showed the Lebanese TYLCV isolate to be closely related to Egyptian, Israeli and Jamaican isolates (94–96% identity). However, it is not closely related to isolates from Sardinia, Spain and Thailand, or to tomato leaf curl isolates from India, Taiwan and Australia. In field and greenhouse screening tests conducted for 5 years on 67 tomato lines, several were identified as promising. TY-Carla, PSR and RS lines were among the most promising with determinate growth, while S&G 143 and the DR lines were the most promising with semi-determinate and indeterminate growth, respectively. Virus concentrations in most, but not all, tolerant tomato lines were significantly lower than in the susceptible lines. None of the lines tested was immune to the virus. A survey of TYLCV alternative hosts on at least 58 plant species, using nucleic acid hybridization, showed that Amaranthus sp., Malva sp., Sonchus oleraceus , Plantago sp., Solanum melongena , Phaseolus vulgaris and Mercurialis annua may play an important role in the epidemiology of TYLCV in Lebanon. Nine tobacco fields of small- and large-scale farmers in Central, Lusaka and Southern provinces of Zambia with an experimental area in the range of 4–52 ha were surveyed for the incidence, prevalence and identification of virus diseases during the growing season of 1997. Samples were collected from three tobacco fields in each of the three provinces, and a total of 72 samples was analysed. Virus identification was based on field disease syndrome, host range studies, DAS-ELISA and electron microscopy of virus particles in some cases. The study demonstrated the occurrence of Tobacco mosaic virus (TMV), Potato virus Y (PVY), Alfalfa mosaic virus (AMV) and Tobacco ringspot virus (TRSV). TMV and PVY occurred widely and were common in all three provinces, while AMV and TRSV were relatively less common. The prevalence of the four viruses was TMV 78%, PVY 67%, AMV 33% and TRSV 22%. Serological tests for Tomato spotted wilt virus (TSWV) and Cucumber mosaic virus (CMV) showed that these viruses were not present in the tobacco samples analysed.  相似文献

 本文报道胶体金免疫电镜技术的建立:铜网捕获病毒后,用其稀释约200倍的同源抗血清修饰处理3~5分钟,羊抗兔IgG-金复合物标记3~5分钟,经磷钨酸负染,电镜下可见病毒粒子周围金颗粒特异性吸附,而背景上非特异性吸附很少。用此技术成功地快速检测和鉴定了病汁液中线状病毒(大麦黄花叶病毒,大麦温和花叶病毒,小麦梭条斑花叶病毒,芜菁花叶病连和马铃薯Y病毒)、棒状病毒(烟草花叶病毒、长叶车前草花叶病毒和土传小麦花叶病毒)以及球状病毒(水稻矮缩病毒、黄瓜花叶病毒和大麦黄矮病毒)。用0.5M PBSpH7.0制备病汁液,可大幅度提高同源抗血清对黄瓜花叶病毒的捕获能力。  相似文献

Abstract

An intensive survey of cultivated plants throughout the island of Montserrat resulted in the identification of a number of viruses. Squash mosaic virus was found in cantaloupe, pumpkin and squash. There were cases of multiple infection of pumpkin and squash by cucumber and squash mosaic viruses. Potato virus Y was recovered from pepper and tomato. Bean yellow mosaic and cowpea mosaic viruses were found in red kidney bean and string bean while cowpea mosaic virus was found in cowpea and broad bean (Jamaica bean). Other viruses that were previously identified on the basis of field symptomatology were confirmed by host range and serology. Viruses were not recovered from sea island cotton or from sweet potato, although field symptoms on the sweet potato are similar to those caused by sweet potato mosaic virus. The leaf tissues of crop plants were inoculated to a wide range of indicator plants. Some of those that reacted systemically were examined by electron microscopy for virus particles and the sap extracted from them was used in serodiagnostic assays for virus identification. This is the first attempt to identify positively the viruses of crop plants on the island.  相似文献

双生病毒是一类在全世界范围内广泛发生的单链环状DNA病毒。本文对从湖南采集到的6例(洋姜、番茄、萝卜、赛葵、甘薯、牵牛花)疑似双生病毒侵染的植物叶片进行了分子鉴定。利用滚环扩增技术(RCA)对样品DNA进行扩增,分别对其RCA产物进行酶切,并将酶切得到的片段测序后进行BLAST比对,结果显示番茄样品中的病毒分离物与番茄黄化曲叶病毒相似性最高(99%),牵牛花样品中的病毒分离物与甘薯卷叶病毒相似性最高(99%),证明这两个分离物是单组分DNA-A双生病毒。这是在湖南省首次发现并报道双生病毒的全核酸序列。  相似文献

Begomoviruses were detected in Nicaraguan fields of tomato ( Lycopersicon esculentum ) and adjacently growing plants of pepper ( Capsicum annuum ), chilli pepper ( C . baccatum ), cushaw ( Cucurbita argyrosperma ) and Mexican fireplant ( Euphorbia heterophylla ) using polymerase chain reaction (PCR) and universal begomovirus primers. All tomato and Mexican fireplant plants showing symptoms were infected with begomoviruses, while only 30–46% of the pepper, chilli pepper and cushaw plants showing symptoms tested virus-positive. No begomoviruses were found in potato. The virus species were provisionally identified by sequencing 533 bp of the viral coat protein gene ( AV1 ). Tomato severe leaf curl virus (ToSLCV), Tomato leaf curl Sinaloa virus (ToLCSinV) and Pepper golden mosaic virus (PepGMV) were found to infect both tomato and pepper. A new provisional species designated Tomato leaf curl Las Playitas virus (ToLCLPV) was detected in a tomato plant. Squash yellow mottle virus (SYMoV) and PepGMV were found in cucurbits, the latter for the first time in this host. Euphorbia mosaic virus (EuMV) was detected in Mexican fireplant. Sequencing of a larger number of PCR-amplified clones from selected plants revealed intraspecific viral sequence variability, and also multiple begomovirus infections which could represent up to three species in a single tomato or cushaw plant. Phylogenetic grouping of virus sequences did not correlate with the host of origin.  相似文献

At a concentration of 0.4 μg purified pokeweed antiviral protein (PAP)/ml, the formation of local lesions on tobacco leaves caused by tobacco mosaic virus infection was completely inhibited and at 25 ng PAP/ ml. 68% inhibition was still obtained. PAP protected plants from infection by viruses from seven virus groups-five RNA viruses: tobacco mosaic virus, cucumber mosaic virus, alfalfa mosaic virus, potato virus X and potato virus Y; and two DNA viruses: African cassava mosaic virus (ssDNA) and cauliflower mosaic virus (dsDNA). Virus infection was probably blocked by PAP at a very early stage. PAP infiltrated into the intercellular spaces through the lower surfaces of leaves inhibited infection by virus inoculated on the upper leaf surface, and partially prevented PVY transmission by aphids. However. PAP did not show any activity against two bacterial and six fungal pathogens.  相似文献

 采用抗原直接包被和双抗体夹心酶联免疫吸附测定法(ELISA)对采自云南、福建、湖南烟区烟草花叶病样品进行了病毒种类检测,利用三抗体夹心ELISA对黄瓜花叶病毒(Cucumber mosaic virus,CMV)的亚组类型进行了鉴定。在云南采集的520个花叶病样品中,烟草花叶病毒(Tobacco mosaic virus,TMV)、CMV和马铃薯Y病毒(Potato virus Y,PVY)总检出率分别为71.74%、55.01%和6.35%;在福建采集的150个花叶病样品中,TMV、CMV和PVY的总检出率分别为94%、24.66%和8.00%;在湖南采集的74个花叶病样品中,TMV、CMV和PVY的总检出率分别为58.11%、51.35%和2.70%。部分样品为2种以上病毒复合侵染。云南、福建和湖南采集的64个CMV阳性样品中,属亚组Ⅰ的样品为57个,占89.1%;属亚组Ⅱ的样品为10个,占15.6%;其中3个样品为亚组Ⅰ和亚组Ⅱ的复合侵染。  相似文献

Eradication of plant pathogens and nematodes during composting: a review   总被引:5,自引:0,他引:5  
The effects of temperature–time combinations and other sanitizing factors during composting on 64 plant pathogenic fungi, plasmodiophoromycetes, oomycetes, bacteria, viruses and nematodes were reviewed. In most cases pathogen survival was determined by bioassays of unknown sensitivity and minimum detection limits of 5% infection or more. For 33 out of 38 fungal and oomycete pathogens, all seven bacterial pathogens and nine nematodes, and three out of nine plant viruses, a peak temperature of 64–70°C and duration of 21 days, were sufficient to reduce numbers to below the detection limits of the tests used. Shorter periods and/or lower temperatures than those quoted in these tests may be satisfactory for eradication, but they were not always examined in detail in composting systems. Plasmodiophora brassicae (clubroot of Brassica spp.), Fusarium oxysporum f.sp. lycopersici (tomato wilt) and Macrophomina phaseolina (dry root rot) were more temperature-tolerant, as they survived a peak compost temperature of at least 62°C (maximum 74°C) and a composting duration of 21 days. Synchytrium endobioticum (potato wart disease) survived in water at 60°C for 2 h, but was not examined in compost. For Tobacco mosaic virus (TMV), peak compost temperatures in excess of 68°C and composting for longer than 20 days were needed to reduce numbers below detection limits. However, TMV and Tomato mosaic virus (TomMV) were inactivated over time in compost, even at temperatures below 50°C. Temperatures in excess of 60°C were achieved in different composting systems, with a wide range of organic feedstocks. The potential survival of plant pathogens in cooler zones of compost, particularly in systems where the compost is not turned, has not been quantified. This may be an important risk factor in composting plant wastes.  相似文献

Plants of four species obtained from various sources were raised and compared for their suitability as test plants for a number of viruses. The species comprisedChenopodium amaranticolor (local lesion host of red clover mottle virus),Nicotiana glutinosa (local lesion host of tobacco mosaic virus (TMV)),N. rustica (systemic host of TMV, cucumber mosaic virus and tomato spotted wilt virus) andN. tabacum ‘White Burley’ (systemic host of TMV). Some sources proved to be more attractive as test plants or experimental hosts because of, e.g., production of more local lesions per unit of leaf area (C. amaranticolor andN. glutinosa), distinctness of lesions (C. amaranticolor), development of more pronounced systemic symptoms (N. rustica andN. tabacum ‘White Burley’), better growth characteristics (particularlyC. amaranticolor andN. rustica), smoother leaves being easier to rub (N. glutinosa) or more and bigger leaf formation (particularlyN. rustica). The implications for the interpretation of host range studies and the quest for propagation and assays hosts are discussed.  相似文献

 制备南方菜豆叶花叶病毒、大豆花叶病毒、烟草花叶病毒、番茄不孕病毒、苜蓿花叶病毒的兔抗血清和鼠腹水抗体,组合成ELISA异种动物抗体双夹心试验。抗体球蛋白的适宜工作浓度为4微克/毫升;未经纯化的特异抗血清(抗体)的适宜工作浓度为10-4(即1/10,000)稀释度。检测以上5种病毒的灵敏度:提纯抗原为10-0.64毫微克/毫升;病叶澄清液为1/10,000-1/100,000稀释度。本法灵敏度与ELISA双抗体夹心法相当或略高。一般8小时内可得出结果。适用于大量样品的检测。  相似文献
