浙江省铁皮石斛根腐病病原真菌的鉴定   总被引:3,自引:0,他引:3  
为明确铁皮石斛根腐病病原真菌,于其主产地浙江省金华市武义县收集铁皮石斛根腐病病株,采用平板分离方法对病原真菌进行分离,使用镰刀菌种特异性引物并结合ITS和TEF序列分析及形态学鉴定确定该镰刀菌的分类地位。结果表明,共分离纯获得真菌117株,其中有105株镰刀菌;经分子生物学分析及形态学鉴定结果显示,分离出的镰刀菌为层出镰刀菌Fusarium prolife‐mum、茄病镰刀菌F. solani、尖孢镰刀菌F. oxysporum和厚垣镰刀菌F. chlamydosporum四个种,其中层出镰刀菌在数量上具有优势地位,占总镰刀菌数的44.8%;茄病镰刀菌、尖孢镰刀菌、厚垣镰刀菌分别占总镰刀菌数的21.0%、15.2%和19.0%。在致病性测定中发现层出镰刀菌和茄病镰刀菌并不具备致病性,尖孢镰刀菌的致病性明显弱于厚垣镰刀菌,表明厚垣镰刀菌为浙江省金华市武义县铁皮石斛根腐病的主要致病菌。  相似文献

Research on root rot pathogens of peas in the Netherlands has confirmed the prevalence ofFusarium solani, F. oxysporum, Pythium spp.,Mycosphaerella pinodes andPhoma medicaginis var.pinodella. Aphanomyces euteiches andThielaviopsis basicola were identified for the first time as pea pathogens in the Netherlands. Other pathogens such asRhizoctonia solani andCylindrocarpon destructans were also found on diseased parts of roots. F. solani existed in different degrees of pathogenicity, and was sometimes highly specific to pea, dwarf bean of field bean, depending on the cropping history of the field.A. euteiches was specific to peas, whereasT. basicola showed some degree of physiological specialization.  相似文献

Isolates of Verticillium dahliae were collected from affected trees (Acer spp., Tilia spp. and Robinia spp.) and soils in Belgian ornamental nurseries. Nitrate non-utilizing mutants were produced and vegetative compatibility groups (VCGs) were classified based on complementation tests with reference tester strains. Of the 30 isolates analysed, 12 were classified as VCG2B and 18 as VCG4B following the American classification. In order to distinguish VCG2B from VCG4B, specific polymerase chain reaction primers were designed based on the sequence of a VCG2B-associated Direct Amplification of Minisatellite-region DNA (DAMD) band generated with the core sequence of the phage M13 minisatellite DNA. Using this test, amplification products were generated for all the VCG2B isolates characterized in this study. In contrast, no signal was seen on ethidium–bromide agarose gel for VCG4B isolates. Pathogenicity tests were carried out in a glasshouse on maple-rooted cuttings inoculated with conidial suspensions of V. dahliae belonging to both groups (VCG2B/VCG4B). Some strains proved to be highly aggressive, while others did not. However, these different behaviours were not correlated with the VCGs.  相似文献

黄淮海夏玉米主产区穗腐病病原菌的分离鉴定   总被引:6,自引:0,他引:6  
为明确我国黄淮海夏玉米主产区玉米穗腐病的病原菌种类、优势种群及虫害、年度、省份对病原菌的影响,以形态学为基础,结合分子生物学方法对2013、2015年随机采自河南、河北、山东3省的155份玉米穗腐病样品进行分离鉴定。结果表明,引起黄淮海夏玉米主产区玉米穗腐病的主要致病菌为镰孢菌Fusarium spp.,包括拟轮枝镰孢F.verticillioides、禾谷镰孢F.graminearum、层出镰孢F.proliferatum、木贼镰孢F.equiseti及藤仓镰孢F.fujikuroi,分离频率分别为49.7%、28.4%、12.3%、3.9%和1.3%;其次为木霉菌Trichoderma spp.,包括哈茨木霉T.harzianum、绿色木霉T.viride和棘孢木霉T.asperellum,分离频率分别为8.4%、3.2%和5.2%;青霉菌Penicillium spp.分离频率较低,为14.2%;曲霉菌Aspergillus spp.包括黑曲霉A.niger和黄曲霉A.flavus,分离频率分别为2.6%和1.9%。研究表明,黄淮海主产区玉米穗腐病优势病原菌为拟轮枝镰孢、禾谷镰孢和木霉菌,不同省份不同年度间病原菌种类及优势病原菌存在差异,虫害能加重玉米穗腐病的发生。  相似文献

Fungi inhabiting Japanese pear were isolated from internal tissues of cv. Nijisseiki, and culture filtrates (CFs) of 100 isolates were evaluated for their inhibitory activity against infection by Alternaria alternata Japanese pear pathotype. CFs of 11 isolates inhibited lesion formation on the pear by the pathogen. Among these isolates, CFs of five isolates inhibited spore germination. CFs of the six other isolates inhibited appressorial formation, infection hypha formation, AK-toxin production, or a combination of these actions. Analysis of sequence homology in the rDNA ITS1 regions of these isolates showed that most isolates had high homology with some fungal endophytes.  相似文献

The main fungi affecting black locust(Robinia pseudoacacia L.) were recorded during a survey which took place in northern Greece in 1997. It was found thatPhomopsis oncostoma (Thüm.) v. Höhn,Aglaospora profusa (Fr.) de Not. andCucurbitaria elongata (@#@ Fr.) Grev. cause necroses of twigs, branches and even entire trees. The pathogenicity of these fungi was confirmed by inoculation experiments in which typical disease symptoms appeared with re-isolation of the respective fungi from the inoculated plants.  相似文献

Forty vineyards from four wine making regions of Spain were sampled at three different growth stages in 2002 and 2003. The aim was to study the fungi associated with grapes and their ability to produce ochratoxin A (OTA) on synthetic media. Among the total mycoflora, 464 (7.7%) and 648 (10.8%) Aspergillus section Nigri (black aspergilli) strains were isolated in 2002 and 2003, respectively, and were classified into three groups: isolates with uniseriate heads, A. niger aggregate and A. carbonarius. The latter presented the highest percentage of OTA-positive strains (82% in 2002 and 76% in 2003) and produced the highest levels of toxin (2.5–25 μg g−1). The sampling year, sampling date, the region and their interactions presented significant differences in the number of black aspergilli isolated. Most black aspergilli were found in 2003 and at harvest. A positive correlation between the number of black aspergilli found in grapes and the temperature in the field was found. Grapes from 2003, the warmest year, and from Costers del Segre, the warmest region, were significantly the most contaminated. No significant correlation between black aspergilli presence and other meteorological factors such as relative humidity or rainfall could be established. Musts from all the vineyards were also analysed in both years, although no OTA was found in either year.  相似文献

The most important tobacco producing areas in Greece were surveyed for virus presence, from 1997 to 2000. Tobacco seedlings or plants showing virus-like symptoms were randomly collected from seedbeds or fields, respectively, and tested by ELISA, and/or mechanical inoculation onto indicator plants. Potato virus Y (PVY), Cucumber mosaic virus (CMV) and Tobacco mosaic virus (TMV) were detected in all sampling areas, with TMV mainly found in oriental varieties. Tomato spotted wilt virus (TSWV) consisted a serious endemic virus in Northern Greece (Thrace, Central and Eastern Macedonia), whereas Alfalfa mosaic virus (AMV) was mainly found in regions, where alfalfa was cultivated in the vicinity of tobacco crops. Eggplant mottled dwarf virus (EMDV) was detected in several areas but always in very low incidence (<0.01%). Surveys were also conducted to assess the potential reservoir hosts of PVY, CMV and AMV among weeds collected from highly infected tobacco fields from 1998 to 2000. Among 3450 samples tested for PVY, plants from 17 species in 10 families were found infected. For CMV, 2891 weed samples were tested and 19 species in 12 families were positive. Assays for AMV infection were made on 961 samples and 12 species in 9 families were identified as hosts of this virus.  相似文献

为明确丛枝菌根(arbuscular mycorrhizal,AM)真菌对加拿大一枝黄花Solidago canadensis与本地菌根植物和非菌根植物种间竞争格局的调控作用,采用温室盆栽试验,通过接种摩西球囊霉Glomus mosseae(GM)、根内球囊霉G. intraradices(GI)及其混合菌种(GM+GI)3种处理,分析AM真菌对加拿大一枝黄花与本地菌根植物玉米Zea mays和非菌根植物油菜Brassica campestris种间作用的影响。结果表明:与对照相比,接种AM真菌均显著提高了加拿大一枝黄花和玉米的菌根侵染率,菌根侵染率为13.720%~50.015%,且前者的菌根侵染率明显高于后者。单独种植时,与对照相比,接种AM真菌尤其是接种混合菌种显著提高了加拿大一枝黄花的株高、叶片数和总干重。在加拿大一枝黄花与玉米混合种植时,与单独种植相比,加拿大一枝黄花的株高、叶片数、根长和总干重均较低;同时,与对照相比,接种AM真菌显著提高了玉米的相对竞争强度而对加拿大一枝黄花的相对竞争强度没有显著影响。在加拿大一枝黄花和油菜混合种植时,与对照相比,接种AM真菌则显著提高了加拿大一枝黄花的株高、叶片数、净光合速率和总干重;同时,接种AM真菌促进了入侵种的竞争优势而抑制了非菌根植物油菜的生长。说明加拿大一枝黄花与本地种的竞争格局受到与之混生物种的菌根依赖性强度以及AM真菌的种类差异影响。  相似文献

采用酸提碱沉的方法,从博落回中提取并制备了苯并啡啶类生物碱(QBAs)——血根碱(sanguinarine)和白屈菜红碱(chelerythrine)的硫酸氢盐;采用高效液相色谱法测定了2种生物碱的含量;初步测定了其硫酸氢盐对几种常见病原菌的抗菌活性,并对其急性毒性进行了研究。结果表明:所制备的QBAs硫酸氢盐中血根碱和白屈菜红碱的质量分数均在60%以上。生物测定结果表明,该生物碱的硫酸氢盐对葡萄叶斑病菌Cercospora viticola、葡萄黑豆病菌Elsinoe ampelina、葡萄炭疽病菌Colletotrichum gloeosporioides、葡萄褐斑病菌Cercospora viticola、稻瘟病菌Pvricularia oryzae、小麦赤霉病菌Gibberella zeae和辣椒疫霉Phytophythora capsici均具有较强的抑制作用,EC50值在3.35~10.08 μg/mL之间。其急性毒性属低毒,对皮肤有弱致敏性,但对眼睛有重度刺激性。博落回中QBAs硫酸氢盐制备方法稳定,具有较强的抗菌活性,可考虑开发成低毒、环境友好型植物源杀菌剂。  相似文献

The processes by which fungi and parasitic plants infect their plant host are compared. The steps considered are: location of the host by the pathogen, the concept of the haustorium, attachment of the pathogen to the host, mechanism of penetration into the host, responses of the host to infection by fungi and by parasitic plants, and the suppression of the host response by the pathogen. Although superficially similarities between fungal infection and infection by parasitic plants exist, the underlying mechanisms appear to be different and only in the penetration step are similar strategies used, including the use of lytic enzymes. These differences are important when seeking ways of combating the parasitic plants. Solacol®, a formulation of the antibiotic validamycin, at 0.33% in 2% malt extract agar, reduced the spread of fungi on dilution plates drastically and allowed twice as much incubation time before subculturing; this resulted in an elevated number of species isolated. Using pure cultures of 62 common soil fungi, it was shown that all fast-growing species (exceptPythium ultimum) were efficiently inhibited but not completely suppressed. Inhibition was comparable to that by 0.5% oxgall, though, while this substance completely suppressed several species, Solacol very strongly inhibited onlyGaeumannomyces graminis, Gerlachia nivalis, Harzia acremonioides, Verticillium biguttatum andRhizoctonia solani. In a further experiment each separate constituent of Solacol was tested against 22 fungi at equivalent concentrations. Validamycin strongly inhibitedChaetomium globosum and two Basidiomycetes, though hardly more than the non-ionic detergent which mainly inhibited the other fungi. A few species were, however, more inhibited by Solacol than by the detergent alone. Validamycine remde alleenChaetomium globosum en twee basidiomyceten behoorlijk, maar nauwelijks meer dan de niet-ionische uitvloeier, die in hoofdzaak de overige remeffecten veroorzaakte. Enkele soorten werden echter door het complete Solacol veel sterker geremd dan door de uitvloeier alleen. Fifteen isolates of rust fungi were collected in the United Kingdom and in the Netherlands from severalAllium species. The samples represented three putative rust species. The morphology of the telia, teliospores and urediniospores was investigated. From urediniospores infection structures were induced on leek, and their morphology was described. Telia and teliospores were not available on every sample. Morphology of the infection structures clearly differentiated between ‘leek’ type and ‘chive’ type isolates. The morphology of infection structures ofPuccinia mixta is very similar to that ofUromyces ambiguus, but clearly distinct fromPuccinia allii sensu Jennings from leek. Quantitative differences in urediniospore characters differentiated between the putative species, but there was overlap between the taxa. We conclude that morphology of infection structures of urediniosporelings is a useful trait for identification of rust isolates fromAllium. This is especially true where more than one rust species may occur on the same host species, as withAllium fistulosum.  相似文献

We isolated 629 fungi from 1296 berry seeds of solanaceous plants, including tomato (Lycopersicon esculentum), eggplant (Solanum melongena), bell pepper (Capsicum annuum), and red pepper (Capsicum annuum var. annuum) preserved for long and short terms. The isolates were classified into 22 genera excluding unidentified fungi, and the fungal floras were divided into two types: the tomato–eggplant and pepper groups. The results of cluster analysis with unweighted pair-group method with arithmetic average also supported these groups. Most tomato seeds infested with Geotrichum candidum germinated and grew the same as uninfested seeds. Cladosporium sphaerospermum and Arthrinium sp. isolated from eggplant seeds strongly suppressed germination, and Penicillium variabile suppressed seminal root elongation on eggplant. Alternaria alternata, Botrytis cinerea, and Myrothecium verrucaria detected from red pepper or bell pepper seeds were pathogenic to the fruits and the seedlings after artificial inoculation.  相似文献

A survey was carried out at nine locations in the Dutch coastal foredunes to identify the species of soil borne fungi and nematodes associated withAmmophila arenaria (Marram grass).Ammophila arenaria is a sand binding grass that is very important for the stabilization of coastal foredunes. Degeneration of the plants occurs at stabilized sites and is supposed to be caused by a combination of soil-borne fungi and nematodes. Canonical correspondence analysis (CCA) and two-way indicator species analysis (TWINSPAN) were used to examine which fungal and nematode species usually coexist in the rhizosphere of vigorous and early declining stands ofA. arenaria. In total, 47 species of fungi and 10 genera of plant-parasitic nematodes were found. According to CCA, the community of soil organisms of stands that were more than 10 years old was significantly different from recently established stands of 3 years old. Also, the community of soil organisms isolated from calcareous locations differed significantly from that of lime-poor locations. No relationship between the vigour of the plants (vigorous vs. early declining) and the soil borne species composition was found, although in roots of vigorous stands, the number of nematodes was higher than that of early declining stands. A relatively large group of soil organisms occurred generally. This group possibly contains an ubiquitous pathocomplex that cause the growth reducing effects of biotic origin which generally occur inA. arenaria. Analysis of this group of nematodes and fungi by TWINSPAN resulted in 9 different combinations of concurring soil organisms of which 5 combinations were present at all investigated locations. Two of the latter combinations contained both nematodes and fungi. The first contained three endoparasitic nematodes (Meloidogyne maritima, Heterodera spp. andPratylenchus sp.) that concurred with the fungusMucor hiemalis. The second group containedHeterodera spp.,Telotylenchus ventralis, Filenchus sp. together with the potentially plant-pathogenic fungiMicrodochium bolleyi and Fusarium culmorum, as well as the fungiMortierella sp. andTrichoderma harzianum, all in relatively high numbers.It is concluded that both CCA and TWINSPAN are valuable exploratory techniques, especially when used in combination, to detect possible combinations of soil organisms which may be involved in the degeneration ofA. arenaria. Further identifications of harmful organisms should be obtained from experiments.  相似文献

The host range of Polymyxa graminis isolates originating from peanut clump-infested areas in India (Andhra Pradesh and Rajasthan), Pakistan and Senegal was studied on monocotyledonous and dicotyledonous cultivated species, using known quantities of sporosori as inoculum. Profuse multiplication occurred only on some graminaceous species, but the various isolates showed different host specificity. All the isolates produced high infection on sorghum and pearl millet, and all but one isolate from Rajasthan infected maize. Wheat, rye and barley were susceptible to some of the tested isolates. The isolates from Rajasthan and Pakistan produced moderate to severe infection on at least one of these species. On rice, groundnut and sugar beet, only traces of infection by some isolates were detected, whereas no infection was observed on mustard and sunflower. Differences of susceptibility in Pennisetum spp. and Sorghum spp. were demonstrated. The variations in host specificity among isolates from peanut clump-infested areas may result from an adaptation of P. graminis populations to various biotopes. The implications of these results for the management of peanut clump disease are discussed. A comparison of the host ranges of isolates of P. graminis and P. betae from temperate areas demonstrated that distinct types of Polymyxa might be identified based on their relative ability to multiply on susceptible species. Nevertheless, overlapping in the host ranges among the different Polymyxa types, characterised by distinct ecological and genomic features, raises doubts about the host range as a classification criterion for the Polymyxa genus.  相似文献

Basal rot is a common disease in lettuce greenhouses. A 3-year study on the diversity of pathogens associated with basal rot in Belgium was carried out. A total of 150 isolates were collected originating from 56 greenhouses. Four pathogens appeared to be involved. Rhizoctonia solani was found to be the causal agent at 23 locations, Sclerotinia spp. at 14, Botrytis cinerea at 17 and Pythium spp. at seven. The isolates of R. solani were further characterised to anastomosis groups and subgroups using morphological characteristics, pectic zymogram and PCR-RFLP. Five anastomosis groups could be distinguished: AG1-1B, AG4 HGI, AG10, AG2-1, AG2-1 Nt and AG3, with isolates of AG4 HGI and AG1-1B being the most prevalent and the most aggressive. Sclerotinia sclerotiorum was found at 13 locations, while S. minor was found at only one location. Based on ITS-sequencing Pythium isolates were assigned to three different species. At 20°C, isolates of all pathogens were able to cause lesions on detached lettuce leaves, except isolates of R. solani AG3 and AG2-1 Nt. A correlation could be found between the occurrence of the pathogens and the growing season. Botrytis cinerea was the most common pathogen in winter, whereas R. solani was most frequently isolated in summer. Sclerotinia spp. and Pythium spp. were isolated in spring, summer and autumn. The information obtained in this study will be most useful in the development of an alternative control strategy for causal agents of basal rot.  相似文献
