A positive energy balance in dairy cows pre‐partum may decrease hepatic carnitine palmitoyltransferase (CPT) enzyme activity, which might contribute to disturbances of lipid metabolism post‐partum. The purpose of this study was to investigate whether skeletal muscle CPT activity can also be downregulated during positive energy balance. Mid‐lactating dairy cows were maintained on intravenous infusion of either saline (control) or glucose solutions that increased linearly over 24 days, remained at the 24‐day level until day 28 and were suspended thereafter. Liver and skeletal muscle biopsies, as well as four diurnal blood samples, were taken on days 0, 8, 16, 24, and 32, representing infusion levels equivalent to 0%, 10%, 20%, 30% and 0% of the net energy for lactation (NEL) requirement respectively. Glucose infusion increased serum insulin concentrations on day 16 and 24 while plasma glucose levels were increased at only a single time point on day 24. Serum beta‐hydroxybutyric acid concentrations decreased between day 8 and 24; whereas changes in non‐esterified fatty acids were mostly insignificant. Total lipid contents of liver and skeletal muscle were not affected by treatment. Hepatic CPT activity decreased with glucose infusion (by 35% on day 24) and remained decreased on day 32. Hepatic expression levels of CPT‐1A and CPT‐2 mRNA were not significantly altered but tended to reflect the changes in enzyme activity. In contrast to the liver, no effect of glucose infusion was observed on skeletal muscle CPT activity. We conclude that suppression of CPT activity by positive energy balance appears to be specific for the liver in mid‐lactating dairy cows.  相似文献

The purpose of this study was to estimate the prevalence of Subclinical ketosis (SCK) between 4 and 19 days in milk (DIM) in a grazing production system and investigate the importance of potential risk factors for SCK. This cross‐sectional study was conducted in dairy cows (= 107), which had more of two parities. The concentration of β‐hydroxybutyric (BHB) in blood was quantified through a hand‐held meter. Potential risk factors evaluated were calving interval (CI), milk yield in previous lactation, metritis, dystocia, calf sex (male), parity (≤3 vs. ≥4) and pre‐partum body condition score (BCS ≤ 3.5 vs. ≥3.75). Prevalence of SCK was 10.3% (95% CI 4.7–15) between 4 and 19 DIM. Risk factors identified were the occurrence of both metritis and pre‐partum BCS ≥ 3.75. Cows with metritis had 4.9 (95% CI 1.17–20.98) times more risk of developing SCK than cows without metritis. And the cows with pre‐partum BCS ≥ 3.75 had 5.25 (95% CI 1.32–21.11) times more risk of developing SCK than cows with pre‐partum BCS ≤ 3.5. Metritis could induce a lower feed intake and promote the development of SCK. High pre‐partum BCS could induce a greater mobilization of body reserves altering liver function and aggravating post‐partum NEB. The results are indicative of the expected prevalence of SCK in grazing production system. Factors associated could help to identify cattle at risk of SCK and improve the management of strategies to limit the effects.  相似文献

A study was conducted to evaluate the effects of three energy supply (E) levels [low (L), medium (M), high (H)], both pre‐partum (PRE) and post‐partum (POST), and their interactions on metabolic parameters and energy balance (EB) in dairy cows of three breeds. In both phases, E levels applied to a total of 81 multiparous cows of breeds Simmental (SI), Brown Swiss (BS) and Holstein–Friesian (HF;= 27 for each breed) were 75%, 100% and 125% of recommendations of the German Society of Nutrition Physiology, using a 3 × 3 factorial arrangement of treatments. During the pre‐calving period, serum concentrations of non‐esterified fatty acids (NEFA) were higher for LPRE cows, and glucose concentrations were elevated for HPRE cows. During the lactation period, NEFA concentrations were greatest for treatment LPOST. Mean concentrations of β‐hydroxybutyrate (BHB) were highest for cows of the LPOST treatment, intermediate for MPOST and lowest for HPOST. Glucose concentrations were lower for LPOST cows. SI cows had lower BHB concentrations both pre‐ and post‐calving and higher glucose concentrations during early lactation than the other breeds. BHB concentration POST was highest for BS cows. Restricted feeding PRE resulted in a better energy status of cows fed above energy requirements POST (EPRE × EPOST interaction). HF cows had a higher EB pre‐calving, whereas SI cows had a less negative EB during early lactation, compared with the other breeds respectively. Correlations of serum NEFA, BHB and glucose concentrations with EB were strongest during the transition period. Results suggest that controlling energy intake during the dry period might be advantageous for the energy status of dairy cows after calving, whereas energy restriction in early lactation leads to metabolic stress. Evidence is provided of a clear relationship between EB and the blood metabolites NEFA and BHB, especially in the transition period.  相似文献

This study was performed to investigate the hypothesis that supplementation of conjugated linoleic acid (CLA) changes the concentrations of retinol and tocopherols in the milk of cows. To investigate this hypothesis, Holstein cows received daily from 3 weeks ante‐partum to 14 weeks post‐partum either 172 g of a CLA‐free rumen‐protected control fat (control group, = 20) or the same amount of a rumen‐protected CLA fat, supplying 4.3 g of cis‐9, trans‐11 CLA and 3.8 g of trans‐10, cis‐12 CLA per d (CLA group, = 20). Milk samples (collected at weeks 1, 3, 5, 8 and 11 of lactation) were analysed for retinol, α‐ and γ‐tocopherol concentrations. Milk of cows supplemented with CLA had higher concentrations of retinol (+34%), α‐tocopherol (+44%) and γ‐tocopherol (+21%) than milk of control cows (p < 0.05). The daily output of these vitamins via milk was also greater in cows of the CLA group than in cows of the control group (+36, 50 and 24% for retinol, α‐tocopherol and γ‐tocopherol, respectively, p < 0.05). In agreement with higher concentrations of tocopherols, concentrations of thiobarbituric acid‐reactive substances, determined in milk of week 5, were lower in cows of the CLA group than in control cows, indicative of a lower susceptibility of milk lipids to peroxidation. Plasma concentrations of retinol and α‐tocopherol, determined at 1 and 5 weeks post‐partum, were not different between the two groups of cows. In conclusion, this study shows that supplementing dairy cows with a moderate amount of CLA causes an increase of the concentrations of vitamins A and E in the milk and results in an increased output of those vitamins via milk. These effects might be beneficial with respect to the nutritional value of dairy products and the susceptibility of milk fat to oxidative deterioration.  相似文献

Undernutrition before and after calving has a detrimental effect on the fertility of dairy cows. The effect of nutritional stress was previously reported to influence gene expression in key tissues for metabolic health and reproduction such as the liver and the genital tract early after calving, but not at breeding, that is, between 70 and 90 days post‐partum. This study investigated the effects of pre‐ and post‐partum mild underfeeding on global gene expression in the oviduct, endometrium and corpus luteum of eight multiparous Holstein cows during the early and middle phases of an induced cycle 80 days post‐partum. Four control cows received 100% of energy and protein requirements during the dry period and after calving, while four underfed received 80% of control diet. Oestrous synchronization treatment was used to induce ovulation on D80 post‐partum. Oviducts, ovaries and the anterior part of each uterine horn were recovered surgically 4, 8, 12 and 15 days after ovulation. Corpora lutea were dissected from the ovaries, and the endometrium was separated from the stroma and myometrium in each uterine horn. The oviduct segments were comprised of ampulla and isthmus. RNAs from ipsi‐ and contralateral samples were pooled on an equal weight basis. In each tissue, gene expression was assessed on a custom bovine 10K array. No differentially expressed gene (DEG) in the corpus luteum was identified between underfed and control, conversely to 293 DEGs in the oviduct vs 1 in the endometrium under a false discovery rate (FDR) < 0.10 and 1370 DEGs vs 3, respectively, under FDR < 0.15. Additionally, we used dedicated statistics (regularized canonical correlation analysis) to correlate the post‐partum patterns of six plasma metabolites and hormones related to energy metabolism measured weekly between calving and D80 with gene expression. High correlations were observed between post‐partum patterns of IGF‐1, insulin, β‐hydroxybutyrate and the expression in the oviduct of genes related to reproductive system disease, connective tissue disorders and metabolic disease. Moreover, we found special interest in the literature to retinoic acid‐related genes (e.g. FABP5/CRABP2) that might indicate abnormalities in post‐partum tissue repair mechanisms. In conclusion, this experiment highlights relationships between underfeeding and gene expression in the oviduct and endometrium after ovulation in cyclic Holstein cows. This might help to explain the effect of mild undernutrition on fertilization failure and early embryonic mortality in post‐partum dairy cows.  相似文献

Fifty‐one dairy cows with subclinical ketosis were used to investigate the effects of butafosfan alone or in combination with cyanocobalamin on metabolism. Treatments included i.v. injection of 10 ml/100 kg of body weight with butafosfan (BUT) or combined cyanocobalamin with butafosfan (BUTCO) at a similar concentration as in Catosal®. Control cows (CON) received a 0.9% saline solution. Cows were injected on days 1–3 at 22.3 ± 0.7 days post‐partum. Milk production and composition were not affected by the treatments. In plasma, CON cows had a significantly higher plasma NEFA concentration (0.59 ± 0.03 mm ) across the study period than BUTCO cows (p < 0.05; 0.42 ± 0.03 mm ), whereas the plasma NEFA concentration of BUT was intermediate (0.52 ± 0.03 mm ) but not significantly different from CON. Both BUTCO and BUT cows had lower (p < 0.05) plasma BHBA concentrations (1.02 ± 0.06 mm and 1.21 ± 0.06 mm , respectively) across the study period than CON (1.34 ± 0.06 mm ). Plasma glucose was not different between treatments, but plasma glucagon concentrations were consistently high in BUT compared to BUTCO and CON. Lowest post‐treatment glucagon levels were observed in BUTCO. Hepatic mRNA abundance of liver X receptor α, a nuclear receptor protein involved in lipid metabolism, was higher in BUTCO compared to BUT and CON (p < 0.05) on day 7. Furthermore, on day 7, the mRNA abundance of beta‐hydroxybutyrate dehydrogenase 2 was higher in BUTCO compared to BUT and CON (p < 0.01). In conclusion, injections of combined cyanocobalamin with butafosfan post‐partum in early lactation ketotic dairy cows act on lipid metabolism with effects on plasma metabolites, most likely mediated via modified activity of key factors in the liver. Results indicate that the application of butafosfan only in combination with cyanocobalamin exhibits the expected positive effects on metabolism.  相似文献

Negative energy balance is considered as the pathological basis of energy metabolic disorders in periparturient dairy cows. Serum non‐esterified fatty acids (NEFA) are one of the most important indicators of energy balance status. Fibroblast growth factor 21 (FGF21) has been identified as a hepatokine involved in regulation of metabolic adaptations, such as promoting hepatic lipid oxidation and ketogenesis, during energy deprivation. However, the direct effects of NEFA on FGF21 expression and secretion in bovine hepatocytes are not entirely clear. The objective of this study was to investigate the effects of different NEFA concentrations on FGF21 expression and secretion in calf hepatocytes cultured in vitro. NEFA were added to the culture solution at final concentrations of 0.6, 1.2, 1.8 and 2.4 mmol/L. After 24 hr of continuous culture, FGF21 mRNA and protein expression levels in the hepatocytes were determined by real‐time PCR and Western blot respectively. FGF21 secretion in the supernatant was determined by enzyme‐linked immunosorbent assay (ELISA). The results showed that expression and secretion of FGF21 at 0.6 mmol/L NEFA‐treated hepatocytes was higher than that of the control group (< .05). The FGF21 expression and secretion were similar at 1.2, 1.8 and 2.4 mmol/L NEFA‐treated hepatocytes and significantly higher than those observed for controls (< .01). These data suggest that high concentrations of NEFA significantly promote FGF21 expression and secretion in bovine hepatocytes. In particular, this promotion occurs in a dose‐dependent manner and may be involved in the pathological processes of energy metabolism disorders of dairy cows in the peripartum period.  相似文献

Dry cow diets based on grassland forage from intensive production contain high amounts of K and could be responsible for a reduced ability to maintain Ca homoeostasis. The aim of this study was to determine whether a moderate anionic salt supplementation to a forage‐based pre‐calving diet with varying native K content affects the mineral and acid–base status in transition cows. Twenty‐four dry and pregnant Holstein cows, without antecedent episodes of clinical hypocalcemia, were assigned to two diets during the last 4 weeks before estimated calving date. Twelve cows were fed a hay‐based diet low in K (18 g K/kg DM), and 12, a hay‐based diet high in K (35 g K/kg DM). Within each diet, six cows received anionic salts during the last 2 weeks before the estimated calving day. After calving, all cows received the high K diet ad libitum. Blood samples were taken daily from day 11 pre‐partum to day 5 post‐partum. Urine samples were taken on days 7 and 2 pre‐partum and on day 2 post‐partum. The anionic salt did not alter feed intake during the pre‐partum period. Serum Ca was not influenced by the dietary treatments. Feeding pre‐partum diets with low K concentrations induced a reduced metabolic alkalotic charge, as indicated by reduced pre‐partum urinary base–acid quotient. Transition cows fed the low K diet including anionic salts induced a mild metabolic acidosis before calving, as indicated by higher urinary Ca, lower urinary pH and net acid–base excretion. Although serum Ca during the post‐partum period was not affected by dietary treatment, feeding a low K diet moderately supplemented with anionic salts to reach a dietary cation–anion difference close to zero permitted to obtain a metabolic response in periparturient cows without altering the dry matter intake.  相似文献

Dairy cows with high and low plasma non-esterified fatty acid (NEFA) concentrations in early lactation were compared for plasma parameters and mRNA expression of genes in liver and subcutaneous adipose tissue. The study involved 16 multiparous dairy cows with a plasma NEFA concentration of >500 μmol/l [n = 8, high NEFA (HNEFA)] and <140 μmol/l [n = 8, low NEFA (LNEFA)] in the first week post-partum (pp). Blood samples, adipose and liver tissues were collected on day 1 (+1d) and at week 3 pp (+3wk). Blood plasma was assayed for concentrations of metabolites and hormones. Subcutaneous adipose and liver tissues were analysed for mRNA abundance by real-time qRT-PCR encoding parameters related to lipid metabolism. Results showed that mean daily milk yield and milk fat quantity were higher in HNEFA than in LNEFA cows (p < 0.01), and the NEB was more negative in HNEFA than in LNEFA in +3wk too (p < 0.05). HNEFA cows had slightly lower (p < 0.1) insulin concentrations than LNEFA cows across the study period, and the body condition score decreased more from +1d to +3wk in HNEFA than in LNEFA (p = 0.09). The mRNA abundance of genes in the liver related to fatty acid oxidation (carnitine palmitoyltransferase 2 and very long chain acyl-coenzyme A dehydrogenase) and ketogenesis (3-hydroxy-3-methylglutaryl-coenzyme A synthase 2) were lower in HNEFA than in LNEFA cows. No differences between the two groups were observed for mRNA expression of genes in adipose tissue. The number of calculated significant correlation coefficients (moderately strong) between parameters in the liver and in adipose tissue was nearly similar on +1d, and higher for HNEFA compared with LNEFA cows in +3wk. In conclusion, dairy cows with high compared with low plasma NEFA concentrations in early lactation show differentially synchronized mRNA expression of genes in adipose tissue and liver in +3wk that suggests a different orchestrated homeorhetic regulation of lipid metabolism.  相似文献

The transition period represents the most critical period in the productive life of high‐yielding dairy cows due to both metabolic and inflammatory stimuli, which challenge the liver and predispose dairy cows to develop liver‐associated diseases such as fatty liver and ketosis. Despite the fact that all high‐yielding dairy cows are affected by marked metabolic stress due to a severe negative energy balance (NEB) during early lactation, not all cows develop liver‐associated diseases. Although the reason for this is largely unknown, this indicates that the capacity of the liver to cope with metabolic and inflammatory challenges varies between individual high‐yielding dairy cows. Convincing evidence exists that endoplasmic reticulum (ER) stress plays a key role in the development of fatty liver, and it has been recently shown that ER stress occurs in the liver of high‐yielding dairy cows. This indicates that ER stress may be involved in the development of liver‐associated diseases in dairy cows. The present review shows that the liver of dairy cows during early lactation is exposed to several metabolic and inflammatory challenges, such as non‐esterified fatty acids, tumour necrosis factor α, interleukin‐1β, reactive oxygen species and lipopolysaccharides, which are known inducers of ER stress. Thus, ER stress may represent a molecular basis for fatty liver development and account for the frequent occurrence of fatty liver and ketosis in high‐yielding dairy cows. Interindividual differences between dairy cows in the activation of hepatic stress response pathways, such as nuclear factor E2‐related factor 2, which is activated during ER stress and reduces the sensitivity of tissues to oxidative and inflammatory damage, might provide an explanation at the molecular level for differences in the capacity to cope with pathological inflammatory challenges during early lactation and the susceptibility to develop liver‐associated diseases between early‐lactating dairy cows with similar NEB and milk yield.  相似文献

In extensive systems, beef cows should be continuously with their calves to optimise pasture use but this practice can lengthen post‐partum interval (PPI). A study was conducted to determine the influence of suckling frequency on Parda de Montaña cow‐calf performance when cows are fed 70% energy requirements during lactation (outdoor winter conditions simulation). Thirty‐six autumn‐calving cows with similar body condition at calving (2.57) were assigned to three suckling systems [ad libitum (AL), twice (2D) or once daily (1D) for 30 min]. Blood samples were collected twice a week to analyse progesterone concentrations by RIA. Cows lost similar weight until start of mating period (day 52 post‐partum; ?0.583, –0.513, –0.520 kg/day in AL, 2D and 1D). Standard milk yield was higher in AL than the rest (8.9, 6.2, 7.0 kg; p < 0.05), which was reflected on greater calf gain (0.895, 0.752, 0.676 kg/day; p < 0.05) and larger cow weight loss within 90 days post‐partum in this treatment (–0.345, –0.188, –0.083 kg/day; p < 0.05). Suckling system did not affect either PPI (69.6, 89.1, 65.5 days) or cows cycling within 90 days post‐partum (55, 46, 58%), which may compromise the target calving interval. In Parda de Montaña breed fed moderately pre‐calving and undernourished during lactation, restricted suckling did not favour ovarian activity resumption, but post‐partum subnutrition delayed about 40 days PPI observed in similar body condition at calving and calf management (Sanz et al., 2003; Anim Reprod Sci 79: 57–69)  相似文献

Negative energy balance at the onset of lactation is unfavorably associated with fitness traits in high-producing dairy cows. Angiopoietin-like protein 4 (ANGPTL4) is an adipokine that has been associated with the regulation of lipid metabolism through the inhibition of lipoprotein lipase activity and regulation of lipolysis. Expression of ANGPTL4 messenger RNA (mRNA) increases during early lactation, but its regulation with changing energy status is currently unknown. Accordingly, the objective of this study was to determine whether ANGPTL4 mRNA abundance is responsive to declining energy balance induced by the transition from pregnancy to lactation, feed restriction, and GH administration in lactating dairy cows. The mRNA abundance of leptin, adiponectin, and adiponectin receptor 2 were also measured to compare adipokine mRNA profiles during changes in energy metabolism. Repeated adipose tissue biopsies were taken from different cows during transition from late pregnancy to lactation (n = 26), feed restriction (n = 19), and GH administration (n = 20). As expected, milk yield increased with the onset of lactation and GH administration (P < 0.01) but declined during feed restriction. Energy balance declined in each experiment, resulting in negative energy balance at the onset of lactation and after feed restriction. Abundance of ANGPTL4 mRNA expression increased 2- to 6-fold with declining energy balance in each experiment. Leptin mRNA declined with feed restriction, and adiponectin mRNA decreased with the onset of lactation. The consistency and magnitude of the increase in ANGPTL4 mRNA across multiple models of altered energy balance identifies it as an adipokine that is uniquely responsive to changes in energy balance in the lactating dairy cow.  相似文献

This study examined the effects of dry glycerol supplementation on follicular growth, post‐partum interval to first ovulation, concentration of serum metabolites and hormones related to fertility, body condition score (BCS) and body weight (BW) in primiparous Holstein dairy cows. Sixty primiparous Holstein dairy cows were randomly assigned to two groups (control: n = 30 and glycerol supplemented: n = 30). Dry glycerol (250 g/day/cow) was fed as a top dressing to the common lactating total mixed ration (TMR) from parturition to 21 days post‐partum. Ovaries were examined four times using ultrasonography on days 13, 19, 25 and 36 post‐partum to determine ovarian follicular growth. Concentration of serum metabolites and hormones was determined weekly. Body condition score was evaluated weekly from weeks 1 to 5 after parturition, and BWs were recorded three times on days 1, 11 and 21 during the experimental period. The cows fed dry glycerol had more large follicles (p < 0.0001) and corpora lutea (CL) (p = 0.02) compared with the control cows. Days to the first ovulation (p = 0.06), days to first oestrus (p = 0.05), services per conception (p = 0.06) and days open (p = 0.004) were positively affected by dry glycerol supplementation. Serum concentration of glucose and insulin was higher in dry glycerol‐supplemented cows (p = 0.1; p = 0.06, respectively). Feeding glycerol had no effect on mean serum concentrations of β‐hydroxybutyrate, non‐esterified fatty acids and IGF‐1 during the experimental period. However, significant differences were observed at concentration of BHBA and IGF‐1 (p = 0.02 and p = 0.04, respectively) between two groups on day 21 after calving. The cows in the glycerol‐fed group had higher serum progesterone concentrations on days 33 (p = 0.007) and 36 (p = 0.004) after calving. Supplemented cows had lower body condition loss during weeks 1–5 after calving compared with the control cows (0.34 vs 0.41 BCS). In week 13 post‐partum, the proportion of cycling cows was 83.3 and 69.9% for those which received supplemented or non‐supplemented diet, respectively. These results demonstrated that feeding dry glycerol as a glucogenic supply may be useful to improve negative energy balance and reproductive efficiency in young cows which calve with high requirement of energy.  相似文献

The energetic status of high‐yielding Holstein‐Friesian dairy cattle was studied during peripartum under field conditions using body condition score (BCS), glycemia, seric ß–hydroxybutyrate and adipose tissue cellularity. This last method was tested as a complementary tool for energetic status assessment. Biopsies of pericaudal subcutaneous adipose tissue were obtained from 25 multiparous animals at 28 days before and 21 days after parturition. Samples were routinely processed for histological examination and stained with haematoxylin and eosin (H&E). The mean diameter of adipocytes (MDA) was measured with the aid of a digital image processor. During the same period, blood samples were collected weekly for metabolite determinations. The MDA at 28 days pre‐partum and 21 days post‐partum were 72.1 vs. 66.2 μm respectively (p = 0.055), and the corresponding BCS at these moments was 3.32 vs. 3.19 (p = 0.068). At ?28 days pre‐partum, the BCS was positively correlated with MDA (Pearson’s r = 0.521, p = 0.016) and with glycemia (Pearson’s r = 0.404, p = 0.056). Correlations between BCS and MDA, and between BCS and glycemia, with ß–hydroxybutyrate although not significant, suggest that routine histological preparations of biopsies from subcutaneous adipose tissue could be included as an easy and valuable tool for research purposes to evaluate metabolic adaptation of dairy cows to peripartum, as well as the incidence of metabolic disorders and productive performance.  相似文献

The high energy demands of dairy cows during the transition period from late gestation into early lactation can place them at an increased risk for the development of metabolic and infectious diseases. Modification of the dry period diet has been investigated as a preventive means to minimize the detrimental aspects of metabolic shifts during the transition period. Studies investigating the impact of dry period diet on lipid parameters during the transition period have largely focused on markers of lipolysis and ketogenesis. Total cholesterol declines during the periparturient period and increases in early lactation. The impact total energy in the dry period diet has on the ability of the cow to maintain total serum cholesterol, as well as its natural high‐density lipoprotein‐rich status, during this metabolically challenging window is not clear. The impact of lipoproteins on inflammation and immune function may have a clinical impact on the cow's ability to ward off production‐related diseases. In this study, we hypothesized that the provision of adequate, but not excessive, total metabolizable energy, would better allow the cow to maintain total cholesterol and a higher relative proportion of HDL throughout the transition period. Cows were allocated to one of three dry period dietary treatment groups following a randomized block design. Total serum triglycerides, cholesterol and lipoprotein fractions were measured on a weekly basis from approximately 7 weeks pre‐calving to 6 weeks post‐calving. The cows on the high energy diet maintained total serum cholesterol as compared to the cows provided a lower energy diet, but there was no significant increase in the LDL fraction of lipoproteins between diet treatment groups.  相似文献

Blood plasma and hepatic parameters were identified that describe the differences between metabolically robust or vulnerable dairy cows grouped according to their past health status. Data from a field study on dairy cows were used from which metabolically challenged dairy cows were selected that had a milk fat percentage of >4.5 mg/g and a fat to protein ratio of >1.5 in their previous early lactation. The selected cows were either classified as metabolically robust or vulnerable based on the occurrence of various metabolic and (re)production disorders in their previous lactations. Blood and liver tissue samples were collected in week 3 ante partum (a.p.) (-3 wk), in week 4 (+4 wk) and in week 13 (+13 wk) post-partum (p.p.). Plasma concentrations of metabolites and hormones and mRNA expression of genes involved in metabolic pathways in the liver were used as variables for a two-group discriminant analysis (DA). Average discriminant scores (centroids) were different (p < 0.05) in -3 wk, +4 wk and in +13 wk. In -3 wk, significant variables that best explained the differences between metabolically robust and vulnerable cows were parity, plasma triglycerides, glucose and mRNA abundance of carnitine palmitoyltransferase 2 (CPT2). In addition, based on the classification matrix, 69% of the dairy cows were correctly classified. In +4 wk, identified significant parameters were parity, plasma glucose and urea, and 67% of the cows were correctly classified. In +13 wk, significant variables that explained the differences between the groups were parity, mRNA abundance of acyl-CoA synthetase long-chain 1 and CPT1, and 66% of the cows were correctly classified. In conclusion, the identified variables may distinguish from metabolically challenged cows, those cows that had a poorer health performance in their previous lactations.  相似文献

One hundred and twenty early lactating Holstein dairy cows were assigned to investigate the effect of dietary chromium (Cr) supplementation (0 or 6 mg Cr/head/day from organic preparation) on the productive and reproductive performance as well as on some blood serum parameters under heat stress (35–40 °C). Cows received treatment from 3 weeks pre‐partum through 12 weeks post‐partum. Chromium supplemented diet had lower body weight loss and improved dry matter intake with consequent reduction in the energy balance deficits during the first period after calving when compared with the control group. Chromium supplementation increased (p < 0.05) milk yield by 6.7%, 12.3% and 16.5% at 4, 8 and 12 weeks post‐partum, respectively, whereas milk composition and milk to feed ratio were unaffected (p > 0.05) when compared with the control. Moreover, dietary Cr supplementation had no effect (p > 0.05) on blood serum glucose, calcium and phosphorus concentrations. A reduction (p < 0.05) of non‐esterified fatty acids at 1 week pre‐partum, 2 and 4 weeks post‐partum was also observed. Serum insulin concentration increased whereas cortisol concentration decreased, when compared with the control group, throughout the whole experimental period. Also Cr supplementation showed a trend towards improving reproductive performance as indicated by increased percentage of pregnant cows in the first 28 days of breeding. It could be concluded that dietary Cr supplementation at level of 6 mg/head/day may offer a potential protective management practice to lessen the effect of heat stress in dairy cattle.  相似文献

Fatty liver or hepatic lipidosis is a major metabolic disorder of high-producing dairy cows that occurs rather frequently in early lactation and is associated with decreased health, production and fertility. A background section of the review explores reasons why high-producing dairy cows are prone to develop fatty liver post partum. Hepatic lipidosis and coinciding health and fertility problems seriously endanger profitability and longevity of the dairy cow. Results from a great number of earlier epidemiological and clinical studies made it clear that a different approach was needed for elucidation of pathogenesis and etiology of this complex of health problems. There was a need for an adequate animal model in which hepatic lipidosis and production, health and fertility problems could be provoked under controlled conditions. It was hypothesized that overconditioning ante partum and feed restriction post partum might induce lipolysis in adipose tissue and triacylglycerol accumulation in the liver following calving. This consideration formed the basis for the experiments, which resulted in the "Utrecht fatty liver model of dairy cows". In this model, post partum triacylglycerol-lipidosis as well as the whole complex of health and fertility problems are induced under well-controlled conditions. The experimental protocol based on this hypothesis produced in all cases (10 feeding trials with over 150 dairy cattle) the intended result, i.e. all experimental cows developed post partum higher hepatic triacylglycerol concentrations than did control cows. The model was evaluated in biochemical, clinical pathology, immunological, clinical and fertility terms. It turned out that in this model, post partum triacylglycerol-lipidosis as well as the whole complex of health and fertility problems were induced under well-controlled conditions.  相似文献

To study the effect of vitamin E (VE), copper (Cu) and zinc (Zn) supplementation on the in vitro phagocytic activity (PA) and lymphocyte proliferation response (LPR) of blood neutrophils and lymphocytes, thirty Sahiwal pregnant cows (six in each group) in their late gestation at 30 days before the expected date of calving were selected from the NDRI experimental herd and supplemented with various micronutrients from 30 days before calving to 45 days after calving. Cows were supplemented individually with VE (1000 IU/cow/day), Cu (20 ppm/cow/day) and Zn (80 ppm/cow/day) and also with a combination of VE, Cu and Zn to study cumulative effect of all micronutrients. One group without any supplementation acted as a control. Blood neutrophils and lymphocytes were isolated and studied for their PA and LPR. Supplementation of micronutrients like VE, Cu, Zn and a combination of all these nutrients significantly (p < 0.01) increased the PA of experimental cows as compared to control (unsupplemented) cows during the pre‐partum period. During post‐partum, all the micronutrients (VE, Cu, Zn and their combination) showed a significant (p < 0.01) increase in the PA of experimental cows as compared to control cows. Of all the groups, significant (p < 0.01) and maximum PA was observed in the combination group followed by Zn‐supplemented group during both the pre‐ and post‐partum period. A significant (p < 0.01) increase in LPR of B lymphocytes was observed in combination‐supplemented group during the pre‐partum period and during both the pre‐ and post‐partum period in the Cu‐supplemented group.  相似文献
