OBJECTIVE: To compare the use of gonadotrophin releasing hormone (GnRH) and oestradiol benzoate (ODB) administered following a synchronised pro-oestrus on reproductive performance of lactating dairy cows and the submission rates of non-pregnant cows following resynchronisation. DESIGN: Cohort study. PROCEDURE: Lactating Holstein cows enrolled in a controlled breeding program were first treated with an intravaginal progesterone releasing insert (IVP4) for 8 days, 2.0 mg of ODB intramuscular (i.m.) at device insertion (Day 0), an analogue of PGF2alpha at device removal and either 1.0 mg of ODB i.m., 24 h after device removal (ODB group, n = 242), or 0.25 mg of a GnRH agonist (GnRH group, n = 152) injected i.m. approximately 34 h after device removal. Every cow was artificially inseminated between 49 and 56 h after removal of its insert (Day 10). Cows detected in oestrus 1 day after artificial insemination (AI) that were not detected in oestrus on the previous day were re-inseminated on that day. All cows treated on Day 0 were resynchronised for reinsemination by insertion of a used IVP4 device on Day 23. Oestradiol benzoate at a dose of 1.0 mg was administered i.m. at the time of device insertion. Inserts were removed 8 days later (Day 31) and 1.0 mg of ODB was injected i.m. 24 h later. Those cows detected in oestrus between Days 31 and 35 were artificially inseminated. On Day 46 these cows were resynchronised for a third round of AI by insertion of an IVP4 device, used previously to synchronise cows for the first and second rounds of AI, and administration of 1.0 mg of ODB i.m.. Eight days later inserts were removed. Cows detected in oestrus between Days 54 and 58 were artificially inseminated. Bulls were run with the herd between rounds of AI and removed after 21 weeks of mating. RESULTS: Treatment with ODB or GnRH at the first synchronised pro-oestrus did not significantly alter the reproductive performance over three rounds of AI or over a 21-week breeding period. Treatment also did not alter submission rates at the second round of AI or the proportion of non-pregnant and non-return cows ('phantom' cows) detected and did not result in significant differences in concentrations of progesterone in plasma 10 and 18 days after removal of inserts at the first round of AI. Treatment with GnRH reduced the proportion of cows detected in oestrus at the first round of AI (36.2 vs 97.5%; P < 0.001). CONCLUSION: Administration of GnRH compared to ODB at a synchronised pro-oestrus results in similar reproductive performance. Treatment with GnRH reduced the proportion of cows detected in oestrus following treatment. This may offer advantages to the way AI is managed by enabling insemination at a fixed-time and removing the need for the detection of oestrus. 相似文献

AIM: To compare 2 treatments for anovulatory anoestrus (AA) in postpartum dairy cows. The treatments were combinations of gonadotropin-releasing hormone (GnRH) and prostaglandin F2 (PG) or progesterone (P4) and oestradiol benzoate (ODB). METHODS: Forty AA cows from each of 5 herds were blocked by age (2 or 2 years old) and randomly assigned to 1 of 2 treatments. The first group (GPG) were treated with 250 mug of a GnRH analogue, gonadorelin, followed 7 days later by 15 mg of the PG analogue, luprostiol. Two days later the cows were injected with 250 mug of gonadorelin. Cows were artificially inseminated 16-24 h after the second GnRH injection. The second group (P4+ODB) were treated with an intravaginal P4 releasing device for 6 days, followed 24 h after device removal by injection of 1 mg of ODB. Cows were pregnancy tested 35-40 days after the initial insemination and twice again at 6-8 week intervals thereafter. RESULTS: There was no significant difference between P4+ODB and GPG groups in the percentage of cows submitted for insemination in the first 7 days (94.0% vs 100% for P4+ODB vs GPG, respectively; p>0.3), in conception rate to first insemination within the first 7 days (43.6% vs 35.0% for P4+ODB vs GPG, respectively; p>0.2), in the percentage of cows conceiving in the first 28 days of the breeding period (68.0% vs 58.3%, P4+ODB vs GPG, respectively; p>0.1), or in median interval from the end of treatment to conception (20 vs 21 days; p>0.1). CONCLUSIONS: No differences in the reproductive performance of AA cows treated with either P4+ODB or GPG were detected. However, given the small number of animals enrolled, further data are required before the GPG protocol can be recommended for treatment of AA cows. 相似文献

OBJECTIVE: To compare the effects of two doses of oestradiol benzoate (ODB) administered as part of a treatment designed to resynchronise returns to oestrus on the reproductive performance of cows in a controlled breeding program. DESIGN: Cohort study. PROCEDURE: Lactating dairy cows on two farms were treated to synchronise three successive oestrous cycles. An intravaginal progesterone releasing insert (IVP4) was used to synchronise the first oestrous cycle. The cows were then treated 15 days after the first treatment by reinsertion of an IVP4 that had been used to synchronise the first oestrus and administration of 1.0 mg of oestradiol benzoate (ODB) i.m. at device insertion. The IVP4 device was removed 8 days later and either 0.5 (n = 421) or 1.0 mg of ODB (n = 446) was administered 24 h later. Injections of (ODB) with or without the use of an IVP4 were used to synchronise the third oestrous cycle. Different synchronisation treatments were used to synchronise first and third oestrous cycles but differences were included in statistical models to account for variation in the data. This enabled examination of effects due to differences in the dose of ODB used to synchronise the second synchronised oestrus. RESULTS: The dose of ODB (0.5 or 1.0 mg) administered just before the second synchronised oestrus did not significantly (P > 0.10) affect the cumulative pregnancy rates over three successive rounds of artificial insemination, the mating start date to the conception intervals, the conception rates to the first or second insemination, the proportion of cows submitted for insemination at the second synchronised oestrus or the proportion of cows that were not pregnant yet failed to show signs of oestrous (phantom cows) identified within each herd. CONCLUSION: There was no difference in reproductive performance between cows receiving either 0.5 mg or 1.0 mg ODB after removal of used IVP4 devices that had been inserted to resynchronise them for a second insemination. 相似文献

Abstract Extract Recent studies have shown that the anoestrous condition of post-partum dairy cows can be treated with a combination of progesterone (for 5–7 days) and oestradiol benzoate (ODB, 1 mg injected at 24 or 48 h after termination of the progesterone treatment)(1). However, published data on the performance of this anoestrus treatment programme in commercial dairy herds is limited(2). During the 1994–95 and 1995–96 breeding seasons, performance of this anoestrus treatment programme was determined in nine and six spring-calving herds, respectively. In each herd, cows that had not been detected in oestrus using the tailpainting technique by Day -7 (Day 0 = start of the breeding season) were examined per rectum by each herd owner's attending veterinarian for the presence of a palpable corpus luteum (CL) on the ovary. Cows with a palpable CL or with an enlarged or infected uterus were excluded. Throughout the trial period, anoestrous cows were managed together with the cycling cows. 相似文献

The aim of this study was to investigate whether a significant relation exists between the presence of a negative energy balance (NEB) in cows early in lactation and the reproductive performance after treatment with gonadotrophin-releasing hormone of cows with clinical cystic ovarian disease. Reproductive performance after treatment was assessed from the interval between treatment and first insemination (ITFI) and the interval between treatment and conception (ITC). Based on the outcome of the daily change of milk fat yield (Deltafatg) between the first and second milk recordings post-partum, cows were considered to have passed the NEB nadir (positive Deltafatg) or not (negative Deltafatg). Lactations (n = 430) were divided into four groups according to the interval between calving and first milk recording (ICMR): (i) 0-9 days; (ii) 10-19 days; (iii) 20-29 days; (iv) 30-49 days. The relation between a NEB-proxy parameter (Deltafatg) and reproductive performance was determined. A significant interaction existed for groups 1 and 3 (ICMR on 0-9 and 20-29 days post-partum respectively) with an increased and a decreased probability of being inseminated the first day after treatment (ITFI) respectively. AIMS: (a) To compare the reproductive performance of anovulatory anoestrous (AA) postpartum dairy cows following treatment with 1 of 2 progesterone (P4) and oestradiol benzoate (ODB)-based treatment regimens; (b) To determine whether resynchronisation of cows initially treated for AA would improve reproductive performance and; (c) to determine whether cows not detected in oestrus but with a corpus luteum (CL) present (NDO/CL+), treated with P4 and ODB, would conceive earlier than untreated controls. METHODS: Cows (n=1386) from 11 herds, that had not been detected in oestrus before the start of the seasonal mating period (PSM) and in which a CL was not detected were diagnosed AA, blocked by age (2 or >2 years old), then randomly assigned to be treated with an intravaginal P4-releasing device for either 6 (6-Day group) or 8 days (8-Day group). Cows in the 8-Day group were injected intramuscularly (IM) with 2 mg ODB at device insertion and all cows were injected with 1 mg ODB 24 h after device removal (Day 0). Cows detected in oestrus from Days 0-3 were subsequently assigned to be either resynchronised or left as untreated controls. Resynchronised cows had a used P4-releasing device reinserted on Day 14 for 8 days and were injected with 1 mg ODB at device reinsertion and again 24 h after device removal. NDO/CL+ cows were assigned to be either treated the same as the 8-Day group or left as untreated controls. All cows were inseminated on detection of oestrus and pregnancy tested approximately 10 weeks after the PSM and again 6-8 weeks after the end of the mating period. RESULTS: For AA cows, the 14-day submission rate was similar between 6-Day and 8-Day groups (p0.1). However, the pregnancy rate by Day 14 was higher for the 8-Day than the 6Day group (43.0% vs 35.2%; p=0.006). Resynchrony treatment decreased the 14-day pregnancy rate compared with untreated controls (35.2% vs 42.5%; p=0.026). The resynchrony treatment increased the submission rate between Days 14-28 for non-pregnant cows compared with untreated controls (80.6% vs 57.4%; p=0.049). However, conception rate to resynchronised heats was lower than for cows that returned to oestrus naturally (56.6% vs 67.9%; p=0.025). Neither initial treatment type nor resynchrony treatment increased the 28-day pregnancy rate (p>0.1). There were no differences between treatment groups in the final non-pregnant rate (4.5% vs 4.6%; p>0.1). Treated NDO/CL+ cows had a higher 14-day submission rate (88.1% vs 49.4%; p>0.001), higher 14-day and 28-day pregnancy rates (42.9% vs 20.7%, p>0.001 and; 56.0% vs 42.5%, p=0.094, respectively) and conceived earlier (21 vs 36 days from PSM to median day of conception; p>0.05), than untreated NDO/CL+ cows. CONCLUSIONS: The 8-Day, ODB-P4-ODB treatment regimen resulted in a higher pregnancy rate by 14 days but not 28 days than the 6-Day, P4-ODB treatment. The resynchrony treatment increased the proportion of non-pregnant cows inseminated on days 14-28, but did not increase the 28-day pregnancy rate or final pregnancy rate. Treatment of NDO/CL+ cows with the 8-Day, ODB-P4-ODB treatment improved reproductive performance compared with no treatment. 相似文献

Fertility data were collected every four weeks for 10 years from 40 herds of Holstein-Friesian dairy cattle. The data collected during 925 lactations from cows with cystic ovarian disease which were treated with 500 microg gonadorelin were compared with data from a control group of 13,869 normal lactations. The intervals between parturition and first insemination and between parturition and conception were significantly shorter (P<0.001) in the normal cows, but the interval between first insemination and conception was not. The overall conception rate tended to be higher (P<0.10) in the normal cows, but the conception rate after the first insemination was significantly higher (P<0.001) and the number of services per conception was significantly lower (P=0.008) in the normal cows. 相似文献

The objective of this study was to examine the effects of increasing circulating progesterone in multiparous Holstein cows, using an intravaginal progesterone implant between days 3 and 7 following fixed-time artificial insemination (FTAI). The animals were randomly assigned into two groups: (1) treatment group and (2) control (no treatment). The experiment took place during periods of high and mild temperatures. Pregnancy rates were evaluated by ultrasound at 30 and 60 days after FTAI; embryonic and fetal mortality rates were also evaluated on these dates. The temperature and humidity index on the day of FTAI were different (P?<?0.05) during the times of interest, but it did not affect conception rates. Neither the conception rates at days 30 and 60 nor the embryonic and fetal mortality rates differed between the groups during either the high- or mild-temperature periods (P?>?0.05). Throughout the experimental period, the serum progesterone concentration was higher in the treatment group than in the control (P?<?0.05). We conclude that the intravaginal progesterone implant inserted from days 3 to 7 following FTAI had no effect on conception rates at days 30 and 60 or on embryonic or fetal mortality rates but enhances the serum progesterone concentrations following treatment in multiparous Holstein cows during high- and mild-temperature periods. 相似文献

Eighty multiparous Holstein cows were assigned randomly at calving to receive either 100 microg of GnRH or saline 13 or 14 d postpartum (PP). From 4 to 28 d PP the cows' reproductive organs were palpated weekly per rectum, and cows were subclassified within each group as undergoing slow (delayed) cervical and uterine involution (abnormal) or as normal cows. Last milk obtained after removing the milking machine was assayed for progesterone 3 times a week for 120 d PP. Fourteen of the 80 cows were removed from the experiment because of culling or various veterinary treatments of pathologic conditions that could confound analysis of the GnRH treatment effects. As expected, the treatment of normal cows with GnRH had no significant effects on the first estrus or the first estrous cycle PP, on services per conception, days open, or any other reproductive trait measured. However, in the abnormal group of cows receiving saline, first rebreeding after calving was delayed (81 vs. 67 d), fewer were pregnant by 105 d PP (23 vs. 64%), and number of days open was greater (121 vs. 87 d) compared with those receiving GnRH; all were significant (P<.05). Treated abnormal cows were equivalent to the control normal cows. Thus, GnRH given 13 to 14 d PP to cows characterized as undergoing slow involution of the reproductive system, but with no other clinical problems, seems to assist in promoting rapid normal reproductive function. The objective of this study was to determine the effect of timing of artificial insemination on pregnancy rates, calving rates, abortion rates, twinning rates, and calf gender ratio after synchronization of ovulation with Ovsynch protocol in Holstein dairy cows. The ovulation of 219 lactating Holstein dairy cows was synchronized using the Ovsynch protocol. Therefore, cows received an injection of GnRH followed by an injection of prostaglandin F2α 7 days later and a second treatment with GnRH 2 days later. Cows were artificially inseminated at 0, 12, or 24 h after the second injection of GnRH. Reproductive performance did not differ between cows inseminated at 0 h (n?=?82), 12 h (n?=?66), or 24 h (n?=?71) after the last injection of GnRH (pregnancy rate: 0 h 48 %, 12 h 47 %, 24 h 52 %; abortion rate: 0 h 5 %, 12 h 0 %, 24 h 11 %; calving rate: 0 h 43 %, 12 h 47 %, 24 h 41 %; twinning rate: 0 h 2 %, 12 h 0 %, 24 h 0 %; calf gender ratio (F/M): 0 h 61:39 %, 12 h 48:52 %, 24 h 39:61 %; P?>?0.05). Pregnancy rates for cows inseminated in postpartum times of 50–75, 76–100, and >100 days within the first and ≥3 parities were statistically similar (P?>?0.05), but pregnancy rates for cows inseminated at different postpartum times of 50–75, 76–100, and >100 days within the second parity were different (P?<?0.01). In general, pregnancy rates of cows inseminated at different postpartum times (P?<?0.01) and parities (P?<?0.001) differed. The findings of the current study showed that rates of pregnancy, abortion, calving, and twinning of Holstein dairy cows subjected to artificial insemination at different times after synchronization were similar. These results also indicate that the timing of artificial insemination after synchronization did not influence calf gender ratio. Furthermore, pregnancy rates of Holstein dairy cows inseminated after synchronization were significantly influenced by postpartum time and parity number. 相似文献

Induction of premature calving contributes to subsequent failure of some dairy cows to become pregnant and calve. Of a sample of 276 cows 16.7% failed to calve the following year despite the occurrence of oestrus relatively early after calving, and several matings. The cows which did conceive had a normal first-mating calving rate of 53.9%, requiring 1.7 matings per pregnancy. Two of the 18 survey herds under consideration suffered lengthy calving-to-first-mating oestrus intervals, presumably because the induced cows were in poor condition at the time of calving. 相似文献

Sixteen days after the PSM, all cows that had not been mated were presented for veterinary examination, and those which were still classified as anoestrus were treated with the previously described CIDR regimen. Pregnancy status and approximate date of conception were determined by palpation per rectum 10 -13 weeks after the PSM or 6 weeks after the end of the mating period. RESULTS: Treatment of anoestrous cows 8 days before the PSM significantly increased the number of cows detected in oestrus (95.0% vs 63.1%; p<0.001) and conceiving (59.5% vs 38.8%; p<0.001) during the first 21 days of mating, and reduced the interval from PSM to conception by 7.5 days (p<0.001). There was no significant difference between the conception rate of cows mated following the CIDR treatment regimen compared to cows mated at their first spontaneous oestrus after calving (52.4% vs 58.3%; p = 0.143). CONCLUSION: Diagnosis and treatment of anoestrous dairy cows prior to the start of mating significantly improves their reproductive performance under the seasonal mating conditions typical of spring-calving New Zealand dairy herds. 相似文献

The effects of gonadotrophin releasing hormone (GnRH) on the reproductive performance of dairy cows with retained placenta were studied. Three hundred and seventy-eight cows diagnosed as having retained placenta received intramuscular injections of either 2 mL sterile water or 200 micrograms of GnRH in 2 mL sterile water between day 8 and day 14 postpartum. Rectal palpation was performed at the time of treatment and ten to 20 days after treatment in order to determine the rate of uterine involution. Thereafter, monthly rectal examinations were carried out until insemination. Pregnancy diagnosis was made by rectal palpation at 40 days or more after breeding. Using the entire experimental population, there were no significant differences between GnRH-treated and control cows for the rate of uterine involution, the occurrence of reproductive problems, the interval from parturition to first observed estrus, the interval from parturition to first insemination, the interval from parturition to conception, the number of services per conception, the total number of services per cow regardless of conception and the incidence of culling for infertility. When the data for herds in which breeding began earlier in the postpartum period (herds having a mean less than or equal to 80 days from parturition to first service for retained placenta cows) were considered, the GnRH treatment resulted in a significantly shorter (p less than or equal to 0.01) calving to conception interval as compared to control cows. The relative merits of three hormone treatments of dairy cows: (1) intravaginally administered progesterone and oestradiol benzoate; (2) intravaginally administered progesterone and injected cloprostenol; and (3) injected cloprostenol; begun 35-75 days after calving and designed to synchronize oestrus and ovulation and allow successful artificial insemination (AI) at fixed times, have been assessed utilizing information from progesterone concentrations in milk. From this it was concluded that 89% of the cows had ovulated one to three times between calving and the beginning of treatment. Treatment (2) was more effective than (1) in synchronizing ovulation. This was due to the fact that when treatments began early in the ovulation cycle, the requirement for a rapidly effective luteolytic agent was provided by cloprostenol but not by oestradiol benzoate. Treatment (2) was also more effective than (3) in synchronizing ovulation. This is interpreted as meaning that progesterone treatment for 12 days had a beneficial effect in restoring normal cyclic ovarian function in the cows after calving. Whilst cloprostenol administered alone did not have this beneficial effect, there is no evidence that it had a detrimental effect. Based on all cows in treatment groups, the proportion that became pregnant to the fixed-time AI was significantly greater after treatment (2) than after (1), but when based on numbers of cows with synchronized ovulation, there were no significant differences among treatments in the proportions becoming pregnant. The progesterone/cloprostenol treatment had a disadvantage in that when begun during the 11-22 day period of the ovulation cycle, so resulting in a long, total period of suppression of ovulation (mean, 32.1 days), fertility to the fixed-time AI was poor despite effective synchronization of ovulation. Ovulation cycles immediately following the failed, fixed-time AI were normal, both in length and in maximum, luteal-phase progesterone concentration and indicated normal corpus luteum function. Thus the infertility could be ascribed neither to poor timing of AI nor to gross degeneration of follicles prior to their synchronized ovulation following the prolonged suppression of ovulation. The 12-day progesterone treatments when given to anovulatory cows gave, within 5.5 h of their beginning, a concentration of progesterone in milk that was not significantly different from the maximum reached. This concentration declined during the 12 days of the treatment but remained above pretreatment level until 5.5 h after treatment withdrawal; the maximum reached was about half that in normal ovulation cycles.(ABSTRACT TRUNCATED AT 400 WORDS) 相似文献

This study analysed the effects of the use of bovine herpesvirus 1 (BHV1) marker vaccine on the performance of dairy cattle. In Spring of 1999, vaccination of 12 herds with the BHV1 marker vaccine resulted in severe animal health problems and mortality. The vaccines used on these farms were all from a batch that appeared to be contaminated with bovine virus diarrhoea virus type 2. This led to a general call to farmers and veterinary practitioners to report side-effects of this vaccine. As a result, more than 7000 farmers reported symptoms. The information was obtained by means of a questionnaire; there was no control group. To determine the effects of the use of the marker vaccine, it was necessary to perform a study based on objectively acquired information. The information collected by the Royal Dutch Cattle Syndicate and the office of Identification and Registration was complied into herd indices on production, udder health, reproduction, and culling. Two groups of dairy farms that had used the BHV1 marker vaccine (attenuated and inactivated vaccine) were compared with farms that were certified BHV1-free. The analyses were performed based on intra-herd comparisons, meaning that per herd each index calculated over a certain period of time after the use of the marker vaccine was compared to a similar period of time prior to the use of the marker vaccine. A total of 144 comparisons were made. Seven comparisons were statistically significant. In two comparisons, the results were in favour of the BHV1-free farms and in five comparisons, the result were in favour of the vaccinated farms. Thus use of the BHV1 marker vaccine could not be proven to affect herd performance. The sensitivity of the tests was very high, so with a high level of probability even very small differences in indices between groups would have been detected. 相似文献

Gonadotrophin releasing hormones have become widely used hormonal compounds in veterinary medicine, particularly with respect to bovine reproduction. The character and physiological actions of gonadotrophin releasing hormone are briefly reviewed and its clinical applications are summarized. The endocrinological research concerned with the use of gonadotrophin releasing hormone in the early postpartum period is discussed. Field trials which have been conducted to assess the effects of postpartum gonadotrophin releasing hormone administration on reproductive performance have varied widely in both design and interpretation of results. These experiments are reviewed, including the clinical trials using normal cows as well as those on cows with retained placenta. 相似文献

AIM: To determine if the reproductive performance of dairy cows not previously detected in oestrus but with a detectable corpus luteum before the planned start of mating (PSM), could be improved by treatment with progesterone, oestradiol benzoate (ODB) and prostaglandin F2alpha (PGF). METHODS: Cows in 18 herds which had not been detected in oestrus, but which had a detectable corpus luteum present at veterinary examination 7 days prior to the PSM (Day -7), were allocated to 1 of 2 groups. Treated cows (n=232) received an injection of 2 mg ODB and an intravaginal progesterone releasing device (CIDR insert) on Day -7, and an injection of PGF on the day of insert removal 7 days later (Treated group). The Control group (n=243) remained untreated. Cows were mated to detected oestrus from Day 0, and conception dates confirmed by manual palpation or transrectal ultrasonography. RESULTS: During the first 7 days of mating, 37.4% of Control cows and 65.9% of Treated cows were inseminated on detection of oestrus (p<0.001). Pregnancy rates for this period were 20.4% and 36.3%, respectively (p=0.001). Conception rates to first insemination, pregnancy rates after 21 days of mating and at the end of the mating period were similar between groups (p>0.1). Median interval from the PSM to conception did not differ between treatment groups (24 and 23 days for Control and Treated, respectively, p>0.1). CONCLUSION: Treating postpartum dairy cows which had not previously been detected in oestrus but which had a detectable corpus luteum, with progesterone, ODB and PGF did not significantly improve their reproductive performance compared with no hormonal intervention.
