Abstract

Growth response of a halophyte species, Suaeda salsa (L.) Pall, to graded NaCl concentrations was examined under water culture conditions. Growth increased with increasing NaCl concentration from 2 to 200 mol m?3, but decreased at NaCl concentrations above 200 mol m?3. Maximum growth was attained at 50 to 200 mol m?3. The role of Na and Cl in the growth stimulation by NaCl was examined by growing S. salsa in nutrient solutions with or without Na and Cl separately at 5 and 50 mol m?3. The growth stimulation induced by Cl was greater than that induced by Na, and Na did not significantly induce growth stimulation. The effect of Na or Cl on O2 evolution from leaves was examined under 5 and 50 mol m?3 concentrations using an oxygen electrode. Oxygen evolution from leaves in –Cl treatments was smaller than that in +Cl treatments both at 5 and 50 mol m?3. The O2 evolution in Na treatments with Cl was similar to that at NaCl. These results indicated that the mechanism of growth stimulation induced by Cl was mainly an increased photosystem II of photosynthesis in leaves. The contribution of Na on the growth stimulation of S. salsa by NaCl was smaller than Cl.

The shoot and root growth response of three species of Sesbania to 0, 50 and 100 mM NaCl in a hydroponic culture was studied. Absorption and translocation of Na and Cl in 15 day old seedlings were studied using 22Na and 36Cl labelled NaCl. Shoot growth was significantly reduced at 100 mM NaCl in the nutrient solution for all three species. The salt tolerance of the three species was in the order S. rostrata > S. aculeata > S. speciosa. The transport of Na to stem and leaf was less compared to uptake in S. rostrata but greater proportion of the absorbed Cl was translocated to the shoot. Salinity reduced the nitrogen content in root and stem significantly.

ABSTRACT

Salt tolerance of Arthrocnemum macrostachyum (Moric.) C. Koch (Chenopodiaceae), a stem-succulent halophyte most commonly found in the intertidal regions of the provinces of Sind and Balochistan, Pakistan, was investigated. Plants were grown for 125 d at six sodium chloride (NaCl) concentrations from 0 to 1000 mM to determine the effects of salinity on ion accumulation, plant water status, and biomass. Shoot biomass was greatest at 200 to 400 mM NaCl, but it was inhibited at salinities of 600 mM NaCl or higher. Tissue water content (g g?1dry mass) of shoots under 200 to 600 mM NaCl treatments was higher than under the control nutrient solution, equal to the control at 800 mM NaCl, but significantly lower at 1000 mM NaCl than under all other treatments, indicating an increase in shoot succulence at salinity levels up to that of seawater. Ash content increased with added salt, but was about 60% of plant dry mass under all salinity treatments. The Na+ and Cl? concentrations of shoots were significantly higher under 1000 mM NaCl than under the control treatment. These results indicate that A. macrostachyum is salt tolerant and capable of accumulating large quantities of Na+ and Cl? when treated with from 200 to 1000mM NaCl.

Osmotic and specific ion effects are the most frequently mentioned mechanisms by which saline substance reduces plant growth. However, the relative importance of osmotic and specific ion effect on plant growth seems to vary depending on the salt tolerance of the plant under study. Tall wheatgrass (TW), perennial ryegrass (PR), African millet (AM) and Rhodesgrass (Rh) were grown in nutrient solution with sodium chloride (NaCl), sodium sulfate (Na2SO4), potassium chloride (KCl), and potassium sulfate (K2SO4) salinity up to electrical conductivity (EC) 27 dS m?1. Growth of all plant species decreased significantly at high level (EC 27 dS m?1) of NaCl and Na2SO4 salts. However, the growth of none of the plant species was affected significantly by KCl and K2SO4 at any level. Even leaf and shoot fresh weights were enhanced by K2SO4 in all plant species, except AM. Chlorine (Cl) was taken up in similar quantities from KCl and NaCl solutions and the content of the respective cations was similar to each other. Further sensitivity to sulfate and chloride was equal when sodium concentrations in shoots were equal, regardless of the anion composition of the media. The sodium (Na) concentration of the leaves of the plant species increased with increased NaCl and Na2SO4 levels in the nutrient solutions. The leaf Na concentration of TW was lower than that of the other plant species. However, the root Na concentration of TW was higher than that of the other plant species. Increased NaCl and Na2SO4 concentrations had a marked effect on leaf water potential of all plant species, and the TW showed higher leaf water potential at all levels of salts. Tall wheatgrass adjusted osmotically by accumulating electrolytes from the nutrient solution and by accumulation of glycinebetaine. Sodium was generally found more injurious than Chloride in all the four forage species. Salt tolerance could be ascribed as greater exclusion of Na ion.

A greenhouse experiment was conducted using a Soil-N Uptake Monitoring (SUM) system to determine nitrogen (N) and water uptake dynamics of citrus (CIT), perennial peanut (PP) (Arachis glabrata Benth), and common bermudagrass (BG) (Cynodon dactylon L.) over time. We also assessed the competition for water and N uptake between citrus and groundcover species and compared citrus N uptake measured using the SUM system with the 15N method. Nitrogen uptake followed cyclic patterns and was greatest for bermudagrass-based systems, while values were similar for PP and citrus systems. Competition for N uptake did occur during the summer months between citrus and BG, while no obvious competition for N uptake occurred between citrus and PP. Water uptake was greatest for the mixed systems and BG monoculture. Citrus, PP, and BG competed for water during the spring and summer seasons. None of the groundcovers significantly affected overall citrus tree growth. Nitrogen use efficiency was greatest for mixed systems and bermudagrass.

Summary The response of two-week-old seedlings of 35 cultivars of lucerne (Medicago sativa L.) to increasing NaCl concentrations was assessed in solution culture under growth room conditions. Shoot length, used as the criterion for assessing salt tolerance, was found to decrease significantly with increasing NaCl concentration for all cultivars, but there was considerable variation in response between, and within cultivars. It is concluded that selection between and within cultivars should lead to increased salt tolerance in this species.

Salt stress has become a major menace to plant growth and productivity. The main goal of this study was to investigate the effect of inoculation with the arbuscular mycorrhizal fungi (AMF; Rhizophagus intraradices) in combination or not with plant growth‐promoting rhizobacteria (PGPR; Pseudomonas sp. (Ps) and Bacillus subtilis) on the establishment and growth of Sulla coronaria plants under saline conditions. Pot experiments were conducted in a greenhouse and S. coronaria seedlings were stressed with NaCl (100 mM) for 4 weeks. Plant biomass, mineral nutrition of shoots and activities of rhizosphere soil enzymes were assessed. Salt stress significantly reduced plant growth while increasing sodium accumulation and electrolyte leakage from leaves. However, inoculation with AMF, whether alone or combined with the PGPR Pseudomonas sp. alleviated the salt‐induced reduction of dry weight. Inoculation with only AMF increased shoot nutrient concentrations resulting in higher K+: Na+, Ca2+: Na+, and Ca2+: Mg2+ ratios compared to the non‐inoculated plants under saline conditions. The co‐inoculation with AMF and Pseudomonas sp. under saline conditions lowered shoot sodium accumulation, electrolyte leakage and malondialdehyde (MDA) levels compared to non‐inoculated plants and plants inoculated only with AMF. The findings strongly suggest that inoculation with AMF alone or co‐inoculation with AMF and Pseudomonas sp. can alleviate salt stress of plants likely through mitigation of NaCl‐induced ionic imbalance, thereby improving the nutrient profile.

Calcium (Ca) has an important role in plant physiology, including involvement in the responses to salt stress, and controls numerous processes. To overcome the negative impact of high salinity, the addition of supplemental Ca to the growth medium as an ameliorative agent could be necessary. Atriplex halimus subsp. schweinfurthii and Atriplex canescens subsp. linearis were grown in hydroponic conditions to investigate the effectiveness of supplementary calcium chloride (CaCl2) applied into nutrient solution on plants grown at high (400 mM) sodium chloride (NaCl) concentration. Treatments were: 1) nutrient solution alone [control (C)]; 2) nutrient solution plus 400 mM sodium chloride (NaCl); and 3) nutrient solution and 400 mM NaCl plus supplementary 40 mM CaCl2 supplied in nutrient solution (NaCl + CaCl2). The experiment was set up as a completely randomized design, consisting of two species (A. halimus and A. canescens), three treatments (control, NaCl, and NaCl + CaCl2), and five replicates. Dry weight and chlorophyll content of plants grown at high NaCl were lower than those at normal nutrient solution. Supplementary CaCl2 ameliorated the negative effects of salinity on plant growth in both species. Root hydraulic conductivity (L 0) decreased with elevated NaCl and increased with supplementary CaCl2 compared to the stressed plants. Membrane permeability increased with high NaCl application and these increases in root membrane permeability decreased with supplementary CaCl2 compared to the NaCl treatment. Sodium (Na) concentration in plant tissues increased in both species in high NaCl level. Application of supplementary CaCl2 lowered Na concentration. Concentrations of calcium (Ca) and potassium (K) were at deficient ranges in the plants grown at high NaCl levels and these deficiencies were corrected by supplementary CaCl2.

ABSTRACT

This study investigated how sequentially exposing plants to sodium chloride (NaCl) would affect growth and the flavor quality of onion (Allium cepaL.) bulbs at harvest. In a greenhouse experiment beginning 74 d before harvest, 100 mM concentrations of NaCl were applied at biweekly intervals to onions growing in nutrient solutions. At harvest, fresh weights (FW) were measured and the bulbs were analyzed for soluble solids content (SSC), bulb pungency as measured by total pyruvate (TPY), bulb sulfur (S) and sulfate (SO4 ?2) accumulation, flavor precursors, and their biosynthetic intermediates. Bulb and leaf FW decreased linearly the earlier NaCl was added during plant growth and development. While total bulb S was significantly affected by sequential addition of NaCl, bulb SO4 ?2 was unaffected. Bulb pungency was significantly reduced by NaCl, especially when NaCl was introduced during early bulb development. NaCl generally reduced flavor precursor accumulation in a quadratic response, with the greatest depression occurring when plants were exposed to NaCl beginning in the early stages of bulbing. Significant reductions in plant growth and changes in the S-compounds associated with flavor suggested that the duration and timing of NaCl exposure are important in onion.

Leucaena leucocephala is a leguminous, deep rooting tree, increasingly utilized in “Low Input” mixed cultures under arid conditions to reduce mineral N requirements and to improve nutrient cycling. This work was carried out to evaluate the growth and N yield of Leucaena and Maize in sand culture with low (0,6 mM) nitrogen supply under saline conditions (NaCl; NaCl + CaCl2; NaCl + CaSO4). While the low N nutrient solution failed to sustain maize growth in monoculture, both Leucaena and maize grew well in mixed culture with considerable N yields suggesting significant transfer of N from Leucaena to maize. NaCl depressed the uptake of Ca and S and reduced growth and N yields of both crop species. CaSO4 counteracted partly the NaCl-effects.

Thirty eight accessions of brown mustard (Brassica juncea (L.) Czern. and Coss.) were screened after two weeks growth in solution culture containing 120 mol m‐3 NaCl. Considerable variation for salt tolerance was observed in this set of germplasm, since some accessions showed relatively vigorous growth in saline medium.

In order to determine the consistency of degree of salt tolerance at different growth stages of crop life cycle two salt tolerant accessions, P‐15 and KS‐51 and two salt sensitive 85362 and 85605 were tested at the adult stage in 0(control), 100 and 200 mol m‐3 NaCl. Both the tolerant accessions produced significantly greater fresh and dry biomass and had considerably higher seed yield than those of the salt sensitive accessions. Analysis of different ions in the leaves showed that salt tolerant accessions contained greater amounts of Na+, K+ and Ca2+ than the salt sensitive accessions, although they did not differ significantly for leaf Cl. Only one salt tolerant accession P‐15 had greater leaf K/Na ratio and K+ versus Na+ selectivity compared with the tolerant KS‐51 and the two salt sensitive accessions.

From this study it was established that there is a considerable variation for salt tolerance in B.juncea which can be exploited by selection and breeding for improvement of its salt tolerance. Since the degree of salt tolerance in B.juncea does not change at different growth stages of the crop life cycle, selection for salt tolerance at the initial growth stages could provide individuals that would be tolerant at all other growth stages. Accumulation of Na+, K+ and Ca2+ in the leaves are important components of salt tolerance in B.juncea.

Glutathione (GSH) and phenolics play an important role in plant defense against metal‐ion toxicity. The antioxidant activity and metal‐binding capacity of these compounds can account for the protective effects. In contrast to animal‐cell models, however, the possible interplay among these substances in stress defense of plants is poorly investigated. This study compares the influence of cadmium (Cd) on the profiles of both soluble phenolics and GSH in shoots of different Thlaspi and Noccaea species: two ecotypes of the nonhyperaccumulator T. arvense differing in Cd resistance (ecotype Aigues Vives, Cd‐sensitive, and ecotype Jena, Cd‐resistant) and two Cd‐tolerant Cd‐Zn hyperaccumulators N. praecox and N. caerulescens (formerly Thlaspi praecox and T. caerulescens). To reveal the possible influence of Cd‐induced sulfur (S) shortage on the stress response, plants receiving normal S concentrations (500 μM MgSO4) and plants treated with surplus S (500 μM MgSO4 + 500 μM K2SO4) were analyzed. Our working hypothesis was that species differences in tolerance to high tissue Cd concentrations should be reflected by differences in endogenous levels of GSH and phenolic compounds. The results reveal clear species‐dependent differences in both the constitutive patterns and the Cd‐ and S‐induced changes in shoot concentrations of GSH and phenolics. However, no simple relationship between these shoot concentrations and Cd accumulation and tolerance can be established.

Dent corn, as a catch crop used for salt removal, was cultivated at different densities, i.e., 7.3 (low density), 59.7 (normal density), and 119.5 plants m-2 (high density), during a 50 d fallow period after cultivation of a commercial crop in a greenhouse, to analyze the characteristics of nutrient salt (N, K, Mg, and Ca) uptake by roots and to study the effect of plant density on the characteristics associated with crop water use. Leaf area index for the high and normal density treatments reached extremely high values of 24.3 and 14.9, respectively. These values induced higher transpiration rates that were estimated using the Penman-Monteith model with the incorporation of specific parameters for crop and greenhouse conditions. The total N, K, Mg, and Ca contents in the crop canopy at harvest were 26.8, 13.0, 1.0, and 1.7 g m-S, respectively, under the high density treatment. The dynamics of salt uptake rates for high, normal, and low density treatments were evaluated by assessing weekly changes in salt content, and were subsequently compared against the transpiration rate. A positive linear relationship was obtained between these 2 parameters for all 3 density treatments and all tested salts. Hence, higher transpiration rates caused higher salt uptake rates through water absorption. On the other hand, salt uptake efficiency per unit water use by cultivation was lower in the low density treatment. Therefore, management procedures with dense planting that induce higher transpiration rates and lower evaporation rate are extremely important for the effective cultivation of corn catch crops.

Salinity and low nitrogen availability are important growth‐limiting factors for most plants. Our objective was to assess the influence of nitrogen (N) and salt levels on the growth and mineral nutrition of three forage grasses of varying salt resistance which are widely found in Tunisian salt lands, Aeluropus littoralis, Catapodium rigidum, and Brachypodium distachyum. Their response to salt and N interaction has not been studied and further investigations are necessary. Twenty day–old plantlets were hydroponically cultivated in Hewitt's nutrient solution. Half the plants were then exposed to 100 mM NaCl and the other half to no NaCl, and N was supplied at 0.5 or 5.0 mM N as NH4NO3. Plants were harvested after 60 d growth. Saline treatment (100 mM NaCl) decreased growth of B. distachyum (a relatively salt‐sensitive plant), but no significant effect was noted for A. littoralis (a relatively salt‐resistant plant) in both low– and high–N availability treatments. However, the effect of 100 mM NaCl on growth of C. rigidum (a moderately salt‐resistant plant) depended on N level. Increasing N availability and NaCl did not influence phosphate, sulfate, calcium, and magnesium concentrations in both A. littoralis and C. rigidum, but increased N supply reduced shoot sodium and chloride (Cl) accumulation. Potassium acquisition in A. littoralis and C. rigidum plants was severely depressed by increasing N availability under saline and nonsaline conditions, respectively. In these species, the increase of nitrate accumulation via N was attenuated by salinity. In contrast, total N content and allocation toward shoots were enhanced in these conditions, especially in A. littoralis, the most resistant species. It appears that increasing N availability at moderate salt levels has a beneficial effect on growth of species with high and moderate salt resistance, but not on species with low resistance to salinity.

Two experiments were conducted to determine if improved nutrient uptake increases salinity tolerance of cotton (Gossypium hirsutum L.). A transgenic cotton line (CMO3) with increased salt tolerance and its wild line (SM3) were grown in pots containing substrate (peat:vermiculite = 1:1, v/v) in the first experiment, while cotton (‘SCRC 28’) was cultured in hydroponics with a split-root system in the second experiment. Contents of essential nutrient elements and Na+ in plant tissues, leaf photosynthesis (Pn) and chlorophyll (Chl) concentration and plant biomass were determined after salinity [sodium chloride (NaCl)] treatment in both experiments. In the first experiment, salinity stress with 150 mM NaCl reduced plant biomass and photosynthesis (Pn) of both SM3 and CMO3 compared with their non-stressed controls, but the CMO3 suffered significantly lower reductions than SM3, suggesting an increased salinity tolerance of CMO3 relative to SM3. Total uptake and contents of main nutrient elements [nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), iron (Fe), manganese (Mn), copper (Cu), and zinc (Zn)] in CMO3 were higher than those in SM3. Also, less sodium (Na+) accumulation and lower extreme ratios of Na/N, Na/P, Na/K, Na/Ca, Na/Mg, Na/Fe, Na/Mn, Na/Cu, and Na/Zn were observed in CMO3 than in SM3. Increased salt tolerance in transgenic AhCMO cotton was probably attributed to its superior nutrient uptake compared with SM3. In the second experiment, the non-stressed root half fed with moderate level of nutrient solution and salt-stressed half fed with low level of nutrient solution (CMN/SLN) exhibited higher salinity tolerance than salt-stressed root half fed with moderate level of nutrient solution and non-stressed root half fed with low nutrient solution (CLN/SMN). Plants absorbed more nutrients but less Na+ under CMN/SLN than CLN/SMN. The overall results suggest that improved nutrient uptake played an important role in the enhanced salt tolerance of cotton.

ABSTRACT

The present work was aimed at determining the limits of tolerance to sodium chloride (NaCl) of a halophyte, Beta macrocarpa Guss (wild Swiss chard). Five week-old plants were cultivated with a nutritive solution to which was added 0, 100, 200, and 300 mM NaCl. Plants were harvested after four weeks of treatment. The growth (fresh and dry weight, leaf surface area, and leaf number), water contents, and the mineral composition (meq · g?1 DW) of roots and leaves (reduced nitrogen (N), K+, Ca2 +, Na+, Cl?) were determined on individual plants. Results show that Beta macrocarpa can tolerate up to 200 mM NaCl. A significant decrease in biomass production (to 50% of control) was observed only for 300 mM NaCl. In the latter treatment, leaf mean surface area was 25% of control. The shoot-to-root ratio was not changed. Leaf hydration was not modified by salt treatment. This ability of the plant to maintain the hydric equilibrium of its leaves seemed associated with an efficient intracellular compartmentalization of Na+ and Cl? ions. Salt treatment had little effect on N content (80% of control), but decreased significantly K+ and Ca2 + contents. These three essential elements could be limiting for growth of leaves and roots of plants challenged by NaCl.

Plants in arid or semiarid areas often experience simultaneous salt and boron (B) stress. Interactive effects on stress responses have been clearly established, but results are inconsistent and variably indicate antagonistic or synergistic interactions even within the same plant species. In this study, five differently B‐ and salt‐resistant wheat genotypes were grown hydroponically at low and high B supply. The effect of increasing NaCl salinity on plant growth, boron uptake rates, shoot B concentrations, and transpiration was determined under both B regimes. The interactive effect of salt and B was different under low and high B supply. Boron‐uptake rates were reduced with increasing salt concentration only under high B supply, and reductions correlated significantly with decreases in leaf area and shoot B concentrations. Under low B supply, however, salt‐induced effects on B‐uptake rates were variable and not significantly correlated with leaf‐area reductions. These results suggest that under high B supply, when B uptake is predominantly passive by diffusion or channel‐mediated via aquaporins, transpiration‐driven water flow is the dominant factor for B accumulation in arial plant parts. Under low B supply, when a significant portion of B can be taken up via active pathways, transpiration is not the decisive factor for B accumulation. Under these conditions, the salt sensitivity of a genotype is a modifying factor of salt–B interactions, because salt‐induced growth inhibition can result in a concentration effect, offset the reduction of B‐uptake rates, and result in increased shoot B concentrations. Contradictory reports on the nature of salt–B interactions might in part be related to low levels of B supply chosen as control treatments and concomittant differences in predominant B‐uptake pathways.

The resistance of most plants to salt can be impaired by concurrent waterlogging. However, few studies have examined this interaction during germination and early seedling growth and its implications for nutrient uptake. The aim of the study was to examine the response of germination, early growth, and nutrient uptake to salt (NaCl) and hypoxia applied to barley (Hordeum vulgare L. cv. Stirling), in solution culture. Hypoxia, induced by covering seeds with water, lowered the germination from 94% to 28% but salinity and hypoxia together lowered it further to 13% at 120 mM NaCl. While the germination was 75% at 250 mM NaCl in aerated solution, it was completely inhibited at this NaCl concentration under hypoxia. Sodium ion (Na+) concentrations in germinated seedlings increased with increasing salinity under both aerated and hypoxic conditions during germination, while K+ and Mg+ concentrations were decreased with increasing salinity in 6 d old seedlings. After 20 d, control seedlings had the same dry weights of the roots and shoots with and without hypoxia but at 10 mM NaCl and higher, shoot and root dry weight was depressed with hypoxia. Sodium ion increased in roots and shoots with increased NaCl under both aerated and hypoxic conditions while K+ was depressed when salinity and hypoxia were applied together and Ca2+ was mostly decreased by NaCl. In general, hypoxia had greater effects on nutrient concentrations than NaCl by decreasing N, P, S, Mg, Mn, Zn, and Fe in shoots and by increasing B concentrations. The threshold salinity levels decreased markedly for germination, uptake of a range of nutrients, and for seedling growth of barley under hypoxic compared to well‐aerated conditions.
