本文通过研究妊娠后期母猪和仔猪补饲外源精胺对初生和28日龄仔猪肠道形态结构和二糖酶活性的影响,初步探讨干预“仔猪早期断奶综合征”的技术措施.试验第1阶段,选择6头体重和膘情相近、胎次为3、已怀孕91 d的健康“长×大”母猪,随机分为Ⅰ、Ⅱ和Ⅲ3个组,每个组设2个重复,每个重复1头母猪.Ⅰ、Ⅱ和Ⅲ组妊娠母猪饲粮中外源精胺的添加量为0、1.5和3.0 mg/kg,饲喂至分娩结束;第2阶段,分娩后母猪采食同一种不含外源精胺的饲粮,Ⅰ、Ⅱ和Ⅲ组哺乳仔猪于7日龄起相应补饲外源精胺添加量为0、3.0和6.0 mg/kg的哺乳仔猪饲粮至28日龄.在仔猪初生和28日龄时,分别从每窝仔猪中选1头接近平均体重的健康仔猪进行屠宰,用于胃肠发育指标的测定.结果表明:与未添加外源精胺相比,妊娠母猪饲粮添加3.0 mg/kg外源精胺显著提高了初生仔猪十二指肠和回肠的柱状细胞数量、十二指肠和空肠的杯状细胞数量(P<0.05),显著增加了空肠隐窝深度(P<0.05),显著提高了麦芽糖酶和蔗糖酶比活力(P<0.05);哺乳仔猪饲粮添加6.0 mg/kg外源精胺显著增加了28日龄仔猪空肠黏膜重(P<0.05),显著提高了十二指肠和空肠的麦芽糖酶和蔗糖酶比活力(P<0.05),极显著增加了空肠柱状细胞和杯状细胞数量(P<0.01),极显著增加了十二指肠和空肠隐窝深度(P<0.01).因此,在妊娠后期母猪和哺乳仔猪饲粮中添加外源精胺有利于初生和28日龄仔猪肠道形态结构的改善和二糖酶活性的提高.  相似文献

Brush border membrane-bound digestive enzymes such as disaccharidases (lactase, sucrase, and maltase), leucine aminopeptidase N, and alkaline phosphatase were measured in jejunum from pigs experimentally infected with porcine epidemic diarrhea virus (PEDV). Three piglets from the infected and control groups were euthanized by electrocution and subjected to necropsy at 24, 36, 48, 60, and 72 hours post-inoculation (hpi). The infection of PEDV to jejunum resulted in significant decreases in brush border membrane-bound digestive enzymes such as disaccharidases (lactase, sucrase, and maltase), leucine aminopeptidase N, and alkaline phosphatase. PEDV replication results in massive destruction of villous enterocytes leading to a marked reduction of intestinal epithelial surface and brush border membrane-bound digestive enzyme activity. Reduced enzymatic activity and villous atrophy in the small intestine is thought to result in a maldigestive and malabsorptive diarrhea.  相似文献

Disaccharidase activity was studied in microvilli membranes of enterocytes isolated from duodenum, jejunum and ileum of lambs at an age of 1, 7 and 23 days. A maximum of lactase activity was observed on the 7th day compared to activity on 1st and 23rd day after parturition and a decrease in enzyme activity on the 23rd day compared to activity on the 7th day. Differences, however, were significant only in respect to activity in ileum. A significant increase of maltase activity in ileum and jejunum was established on the 7th and 23rd day compared to activity on the 1st day after parturition. Trehalase activity attained a maximum in all the three intestine divisions, consequently it subsided with no statistically significant differences. Lactase activity in ileum was significantly lower compared to activity in jejunum of lambs at an age of 1 and 23 days. A marked decrease of maltase activity in distal direction was found only on 1 day-old lambs. No significant alterations were observed in trehalase activity along the small intestine of lambs of the three age groups.  相似文献

The activities of the enterocyte brush border enzymes lactase (beta-D galactoside galactohydrolase, EC 3.2.1.23) and sucrase (sucrose alpha-D glucohydrolase, EC 3.2.1.48) were measured at set percentage lengths along the small intestines of 112 piglets killed between 21 and 32 days of age. The influences on these activities of consumption of creep feed and of weaning were recorded. Weaning at three weeks old resulted in large, rapid reductions in lactase activity at most sites along the small intestine; sucrase activity declined temporarily and then recovered. Minimum values were recorded about four to five days after weaning. Similar changes were observed whether or not creep feed was consumed before weaning. Continued consumption of creep feed by unweaned pigs over the 21 to 32 day period also produced small but significant reductions in lactase activities. The large loss of digestive enzyme activities at brush borders in weaned animals coincided with a reduced ability to absorb xylose and to checks in growth rate in otherwise healthy piglets.  相似文献

Gnotobiotical one-day-old piglets were infected with 100,000 Isospora suis coccidia oocysts, and were immediately killed. In piglets killed on the 3rd to 11th day after infection (DAI), the morphometric analysis of alkaline phosphatase activity was performed in the area of the microvillus zone of small intestine. In the control 2-7 days old animals, the small intestine was not equally supplied with alkaline phosphatase. In duodenum the activity reached 88.37 per cent of the active length of absorbent surface (% LAac), in the middle jejunum 95.98 per cent LAac, in the dorsal jejunum 78.63% LAac and the ileum 90.55 % LAac. The width of the active area was more balanced and ranged from 5.003 microM in the ileum to 6.129 microM in the dorsal jejunum. In infected gnotobiotical piglets the lowest activity was found out on the 3rd to 4th DAI, with a greater decline on the 9th day after infection. The range from 25.99 to 40.50 per cent LAac with minimum in the duodenum and maximum in the ileum was observed on the 3rd DAI. In the middle and dorsal ileum the activity was nearly equal (28.34 and 27.69 per cent LAac). nI the dorsal jejunum a moderate increasing was up to 47.13% LAac on the 4th DAI, with the exception of the ileum, where the activity of alkaline phosphatase decreased to 24.96% LAac. On the 9th DAI the activity of alkaline phosphatase was nearly equal in the whole small intestine (from 55.70 to 60.01% LAac) with the maximum in the middle jejunum. In the width of the reaction product a direct dependence on the total activity of alkaline phosphatase was evident only in the segment of the middle and dorsal jejunum and ileum, but merely on the period of the 3rd to 4th DAI. The lowest values were measured in the middle jejunum (0.982 micron on the 3rd DAI and 0.709 micron on the 4th DAI). No dependence was observed between the total activity and the reaction product in the middle jejunum (0.982 micron on the 3rd DAI and 0.709 micron on the 4th DAI), there was no general stabilisation of the activity of alkaline phosphatase.  相似文献

The purpose of this study was to examine the short-term effects of 75, 100 and 150 mg of acarbose mixed in 100 g standard laboratory chow on specific intestinal disaccharidase activities and on hyperglycaemia in diabetic CBA strain mice on standard diet. The small intestine was excised and divided into three segments, from pylorus to duodenum, and two equal lengths of the jejunum and ileum of control and diabetic mice with or without added acarbose. Specific maltase and sucrase activities were determined using maltose and sucrose as substrates respectively. Increased specific activities of maltase and sucrase were detected in the intestines of CBA mice on standard laboratory diet seven days after alloxan-induced diabetes. Feeding for 7 days with 75, 100 or 150 mg acarbose uniformly mixed in 100 g standard laboratory chow, induced a decrease in the specific maltase and sucrase activities, compared with diabetic mice on standard laboratory diet. Feeding with 75 mg acarbose mixed in 100 g standard laboratory chow caused a statistically significant decrease of maltase in the duodenum and of sucrase in duodenum and jejunum, without a antihyperglycaemic effect. Feeding with 100 or 150 mg caused statistically significant decreases in specific maltase and sucrase activities in duodenum, jejunum and ileum. An antihyperglycaemic effect was observed only in the group of diabetic mice fed with 100 mg acarbose. This indicates that the antihyperglycaemic effect of acarbose involves factors other than these, related only to its inhibitory effect on disaccharidase activities.  相似文献

试验旨在建立猪流行性腹泻病毒(PEDV)感染幼龄仔猪肠道损伤模型。试验选用16头7日龄健康幼龄仔猪(杜×长×大),随机分为两个处理组:对照组和PEDV组,每个处理8个重复,每个重复1头猪。试验期为10 d,试验期间两个处理组饲喂相同的基础日粮。于试验第7天晚上对PEDV组仔猪口腔灌服PEDV病毒(剂量为104.5 TCID50),对照组灌服等量的生理盐水。于试验第10天早上空腹灌服D-木糖(0.1 g/kg体重),1 h后前腔静脉采血,屠宰取样,取十二指肠、空肠、回肠等组织样品,测定平均日增重(ADG)、腹泻率(DR)、肠道形态结构、肠道黏膜损伤相关基因mRNA水平。试验结果表明:①PEDV感染显著降低了仔猪ADG(P<0.05),极显著提高了仔猪腹泻率(P<0.01);②PEDV感染极显著降低了血浆D-木糖含量、空肠和回肠绒毛高度、十二指肠、空肠和回肠绒毛高度与隐窝深度的比值(P<0.01),显著提高了十二指肠和空肠隐窝深度(P<0.05);③PEDV感染极显著提高了仔猪空肠黏膜PEDV M基因的相对表达量(P<0.01),极显著降低了肠绒毛蛋白(villin)和肠型脂肪酸结合蛋白(i-FABP)基因的相对表达量(P<0.01)。以上结果表明,口腔灌服PEDV可以成功诱导建立幼龄仔猪肠道损伤模型。  相似文献

试验旨在研究发酵床饲养模式下饲粮中添加抗菌肽对仔猪生长性能、胰腺和小肠内容物消化酶及小肠黏膜二糖酶和垫料蛋白酶、脲酶、铵态氮的影响。选用108头35日龄、体重13kg左右的健康苏钟猪随机分为3组,每组3重复,每重复12头,分别饲喂基础饲粮(对照组)、基础饲粮+40mg/kg杆菌肽锌+20mg/kg硫酸黏杆菌素(抗生素组)和基础饲粮+300mg/kg抗菌肽(抗菌肽组)的试验日粮,试验期49d10结果表明:(1)各组间仔猪生长性能差异不显著(P〉0.05)。(2)与对照组相比,饲粮中添加抗菌肽能够显著提高仔猪胰腺、十二指肠和空肠内容物淀粉酶及空肠内容物胰蛋白酶活性(P〈0.05);并显著提高仔猪十二指肠黏膜麦芽糖酶和空肠黏膜蔗糖酶、乳糖酶及回肠黏膜蔗糖酶活性(P〈0.05);抗菌肽组十二指肠黏膜麦芽糖酶活性比抗生素组提高(P〈0.05)。(3)各组间发酵床垫料蛋白酶和脲酶活性及铵态氮含量差异不显著(P〉0.05)。综上所述,本试验条件下,饲粮添加300mg/kg的抗菌肽能够在一定程度上提高仔猪生长性能;提高胰腺、小肠内容物及小肠黏膜消化酶活性;同时不影响垫料蛋白酶、脲酶活性和垫料铵态氮含量。  相似文献

In gnotobiotical and conventional piglets infected a day post partum (DPP) with oocysts of the coccidium Isospora suis, densitometrical analysis of the activity of aminopeptidase M (EC.3.4.11.2; APM) was performed in the area of microvillous zone of the small intestine. Piglets were infected with different infection doses of oocysts (100,000 oocysts) in gnotobiotical piglets and 200,000 oocysts in conventional piglets). In infected gnotobiotical piglets, the APM activity was studied in the period from the 3rd to 11th day after infection (DAI) and in infected conventional piglets in the period of to the 2nd to 10th day after infection (DAI). Control piglets, in the group of the gnotobiotical animals at the age of 2 to 5 days in the group of the conventional animals at the age of 4 to 7 days, had different APM activity in the microvillous zone of the intestinal mucosa. It was stated that the microvillous zone of the intestinal mucosa gained higher values in control conventional piglets (+7.01 mean values of density). In infected gnotobiotical piglets the density fall of the reaction product of APM was demonstrated already on the third day with further marked reduction of APM density on the 4th day after infection in the whole small intestine with predominance of the persisting APM activity in ileum. Even despite the slight increase in the density of the reaction product of APM in the period from the 5th to 7th DAI (the highest increase in APM density on the 6th DAI), a further decrease of the activity was recorded again on the 8th and namely the 9th DAI in the whole small intestine (the lowest value of density was found in the rear jejunum), the ileum mucosa being affected, too. A slightly higher density of the reaction product of APM was found in the duodenum. On the 10th DAI the APM density started to change and on the 11th DAI in the duodenum and in the middle jejunum it even reached higher values in comparison with the control data. Some differences were proved in the infected conventional piglets in comparison with the development of the APM activity in the small intestine mucosa in the infected gnotobiotical piglets. On the 3rd and 4th DAI APM defect occurred in the whole small intestine, with APM density prevailing in the ileum mucosa (like in the group of infected gnotobiotical piglets). The intestinal morphology of 7-week-old pigs was investigated by light (LM) and scanning electron microscopy (SEM). The piglets were fed either a semisynthetic or a cereal-based diet. The shapes of the intestinal villi and crypts of the duodenum, jejunum and ileum were examined. The villi were predominantly tongue-shaped. In the duodenum they were also ridged, branched and folded, and in the jejunum they were also leaf-like and ridged. At places with lymph follicles, the surface of the ileum was rugged with meandering fold-like villi. The crypts of the three segments of the small intestine were mainly coiled and sometimes branched. A novel morphometric evaluation method was introduced using the enlargement factors of each villus and crypt surface. The enlargement factor for the villus surface of the duodenum, jejunum and ileum was 3.13, 3.72 and 2.71, respectively. The factor for the crypt surface of the duodenum, jejunum and ileum was 9.07, 8.94 and 6.53, respectively. Furthermore, the relative proliferation rate and the epithelial renewal index were calculated for the first time. The relative proliferation rate of the duodenum, jejunum and ileum was 32.88, 34.78 and 50.77 proliferations per mm crypt perimeter, respectively. The diets consumed had an influence on the epithelial renewal index being higher for piglets fed the cereal-based diets.  相似文献

In the small intestine mucosa of 24 gnotobiotical piglets experimentally infected the first day post partum with oocysts of the coccidium Isospora suis, the activities of dipeptidylpeptidase IV (EC.3.4.14.5.; DAP IV) and gamma-glutamyl transferase (EC.2.3.2.2.; GGT) in the microvillous zone of enterocytes were evaluated by scanning densitometry. The tissue of the small intestine in piglets infected with a dose of 100,000 oocysts of the coccidia of I. suis was examined in the period from the first till the eleventh day post infection (DPI). In the control piglets at the age of 2-5 days it was found that most of the DAP IV activity was located in the microvillous zone of the enterocytes of the middle jejunum, rear jejunum and ileum. The DAP IV activity of duodenum mucosa was lower; as compared with the activity in the mucosa of the jejunum and ileum it reached 53-57%. In the case of GGT activity, the highest density values of the reaction product were recorded in the microvillous zone of enterocytes of the duodenum and the whole jejunum, the lowest in the ileum mucosa (86-89%) of the activity found in the duodenum and jejunum). During the experimental infection the infected piglets had a significant deficit of both peptidases, especially DAP IV (the whole studied period). The development of GGT activity was slightly different with the onset of the marked decline of the enzyme activity only on the fifth DPI. The lower GGT activity persisted till the eighth DPI. The density of the GGT reaction product began to return to the normal on the ninth to eleventh DPI. No predisposition in the location of the deficit was observed in the peptidases studied during the infection. The decline of the activity of both enzymes influenced also the mucosa of all studied parts of the small intenstine. The difference lay in the relevance of lowering of the density of reaction product of DAP IV and GGT on other DPI and in the different intensities of the return of the activity to the physiological normal.  相似文献

Forty eight 21 days old piglets were used to compare the effect of prebiotic or probiotic supplementation and ileo rectal anastomosis on the morphology of the small intestine. Half of the piglets were maintained intact and the other half was subjected to an ileo rectal anastomosis (IRA). Each group of piglets received one of the following diets: 1) basal diet (C), 2) basal diet supplemented with a Xylo-oligosaccharide (XOS), 3) basal diet supplemented with a Saccharomyces cerevisiae (SC) and 4) basal diet supplemented with XOS and SC. Villus height was greatest with XOS and with XOS + SC, only in the ileum, as compared to controls. In the duodenum, crypt width was highest in the control group, but no significant differences were found in the jejunum and ileum. The IRA piglets had longer villi in the jejunum and shorter villi in the ileum. The crypt depth was greater in the duodenum and in the ileum of IRA piglets. Villus height/crypt depth was lower in the duodenum and in the ileum, in the IRA piglets. In conclusion, the XOS, but not the SC, moderately modified the intestinal morphology. The IRA modified the intestinal villus and crypt architecture but its consequence on the absorption of nutrients needs to be investigated.  相似文献

【目的】 通过研究槲皮素对猪流行性腹泻病毒(PEDV)感染仔猪肠道形态、肠道抗氧化功能及空肠脂质代谢相关基因相对表达量的影响,旨在探讨槲皮素对PEDV感染仔猪肠道的保护作用。【方法】 选取18头健康的7日龄断奶仔猪,随机分为3组:对照组、PEDV组和槲皮素+PEDV组,每组6个重复,每个重复1头猪。试验期8 d,于试验第0~7天,对槲皮素+PEDV组仔猪每天口腔灌服10 mg/kg BW的槲皮素,其余组灌服等体积的人工奶。于试验第5天晚上给PEDV组和槲皮素+PEDV组仔猪口腔灌服104.5TCID50的PEDV,对照组仔猪灌服相同体积的PBS溶液。于试验第8天早上屠宰,取仔猪十二指肠、空肠、回肠及结肠组织样品进行检测。【结果】 与对照组相比,PEDV组仔猪空肠和回肠的绒毛高度以及绒毛高度与隐窝深度比值极显著降低(P<0.01),空肠、回肠和结肠中谷胱甘肽过氧化物酶(GSH-Px)和总超氧化物歧化酶(T-SOD)的活力显著降低(P<0.05),十二指肠、空肠、回肠和结肠中过氧化氢酶(CAT)的活力显著降低(P<0.05),回肠和结肠中丙二醛(MDA)的含量显著升高(P<0.05),空肠中载脂蛋白A1(APOA1)、载脂蛋白B (APOB)、载脂蛋白C2(APOC2)、脂肪酸结合蛋白2(FABP2)、酰基辅酶A合成酶长链家族成员3(ACSL-3)和脂肪酸合酶(FASN)基因的相对表达量极显著下调(P<0.01)。与PEDV组相比,槲皮素+PEDV组仔猪空肠和回肠的绒毛高度以及绒毛高度与隐窝深度比值极显著提高(P<0.01),空肠、回肠和结肠中GSH-Px和T-SOD的活力显著提高(P<0.05),十二指肠、空肠和结肠中CAT的活力显著提高(P<0.05),回肠和结肠中MDA的含量显著降低(P<0.05),空肠中APOB、FABP2和FASN基因的相对表达量极显著上调(P<0.01)。【结论】 添加槲皮素可有效缓解PEDV感染导致的仔猪肠道损伤,提高仔猪肠道抗氧化能力以及空肠脂质代谢的能力。  相似文献

The effect of segregated early weaning (SEW) on postweaning small intestinal development was investigated in SEW and control (CON) pigs. Small intestines were collected from a total of 15 pigs killed at 11 (preweaning), 15 (3 d postweaning), and 34 d of age. At 3 d postweaning, the SEW and CON pigs had shorter villi (P<.01), deeper crypts (P<.01), and reduced (P<.01) ratios of villus height:crypt depth (V:C) compared with preweaning. Weaning also reduced specific activities of lactase (P<.01) in duodenum and ileum and alkaline phosphatase (ALP) (P<.05) in duodenum and jejunum. Sucrase activity in the three regions of the small intestine marginally decreased in both groups at 3 d postweaning. The mucosal protein:DNA ratio in duodenum and jejunum increased (P<.05) in SEW and CON pigs at 3 d postweaning compared with preweaning pigs. The SEW and CON treatments resulted in differences in postweaning gut development. At 15 d of age in SEW pigs, the mucosal protein:DNA ratio in duodenum and jejunum were 20 and 25.5% (P<.05) less, respectively, than those in CON pigs. However, at 34 d, these ratios in duodenum, jejunum, and ileum were 43.5 (P<.05), 24.3, and 32.9% (P<.05) greater, respectively, in SEW pigs than in CON pigs. Longer villi, shorter crypts (P<.01), and higher V:C ratios (P<.01) in jejunum and ileum were observed in SEW pigs vs CON pigs at 34 d of age. The specific activities of lactase in duodenum (P<.01) and jejunum (P<.05) and of ALP in duodenum (P<.01) were higher in SEW pigs. Sucrase activity in duodenum, jejunum, and ileum was 21.7, 46.3 (P<.05), and 11.2% greater in SEW pigs at 34 d of age. These results demonstrate differences in postweaning gut development between SEW and CON pigs. Furthermore, the number of intraepithelial lymphocytes in jejunum was greater (P<.001) in 34-d-old SEW pigs compared with CON pigs. Microscopy revealed a thick mucus coating over epithelial cells in the ileum of 34-d-old CON pigs that was not apparent in the SEW pigs. These observations are consistent with reduced pathogen exposure associated with SEW. We suggest that segregated early weaning advances postweaning gut maturation, which is consistent with improved growth and feed efficiency observed in SEW pigs.  相似文献

The objective of this study was to compare the effect of three Chinese herbal medicine (medicated leaven, hawthorn, fructus hordei germinates) on histomorphology of different parts of small intestine in weaning piglets.40 28-day old ternary pigs (Duroc×Landrace×Yorkshire) with the body weight (8.13±1.32)kg were divided into four groups, control group, medicated leaven group, hawthorn group and fructus hordei germinates group, and feeding with 1% Chinese herbal medicine, there were two repeats in each group and each repeat had 5 piglets, slaughtering and sampling at 7th and 14th day morning, respectively, sections were used for HE staining.The staining tissues were taken pictures under a microscope camera (200×) and measured VH, CD, IWT and V/C in different parts of small intestine with Image-Pro Plus.The result showed that the IWH of ileum in medicated leaven group was significantly or extremely significantly higher than control group (P<0.05;P<0.01).The VH and IWT of duodenum and jejunum at 7th day in hawthorn group were extremely significantly lower than control group (P<0.01);The IWT of ileum and the VH of ileum at 14th day were extremely significantly higher than control group (P<0.01).In fructus hordei germinates group, the VH of duodenum and jejunum at 7th day and IWT of jejunum were extremely significantly lower than control group (P<0.01);The CD of ileum at 7th day, the IWT of duodenum and ileum at 7th day were extremely significantly higher than control group (P<0.01).The effects of different simple Chinese herbal medicine on histomorphology of different parts of small intestine in piglets were different, because of the different digestive functions of small intestine, the most effects of Chinese herbal medicine an duodenum, jejunum and ileum were fructus hordei germinates, medicated leaven and hawthorn, respectively.  相似文献

本研究旨在比较神曲、麦芽、山楂3种助消化中草药对断奶仔猪小肠不同部位组织形态学的影响。本试验以杜长大三元猪为研究对象,选取40头28日龄、体重为(8.13±1.32)kg断奶仔猪,添加1%的中草药饲喂,分空白组、神曲组、麦芽组、山楂组,每组两个重复,每重复5头,分别在试验第7和14天早晨空腹屠宰取样,经HE染色,200×显微镜下照相,用Image-Pro Plus软件测定小肠各段绒毛高度(VH)、隐窝深度(CD)、肠壁厚度(IWT)、绒毛高度与隐窝深度的比值(V/C)。结果显示,神曲组在回肠段的IWT显著或极显著高于空白组(P<0.05;P<0.01);第7天时山楂组的十二指肠和空肠的VH和IWT均极显著低于空白组(P<0.01),回肠段的IWT和第14天回肠段VH均极显著高于空白组(P<0.01);第7天的麦芽组十二指肠和空肠VH及空肠IWT均极显著低于空白组(P<0.01),第7天的回肠段CD及十二指肠和回肠段IWT均极显著高于空白组(P<0.01)。不同单味中草药对仔猪小肠不同部位组织形态学的影响不同,源于肠道消化物质的功能不同,对十二指肠、空肠、回肠影响最大的分别是麦芽、山楂、神曲。  相似文献

试验旨在研究表达耐热肠毒素(STa)的重组大肠杆菌对7日龄仔猪肠道形态结构及抗氧化功能的影响。选取24头7日龄仔猪,随机分在4个日粮处理组,分别为对照组(人工乳),STa组(人工乳+2×109 CFU重组菌LMG194-pBAD-STa),LMG194组(人工乳+2×109 CFU大肠杆菌LMG194),K88组(人工乳+2×109 CFU大肠杆菌K88)。试验第5天进行攻毒,第7天屠宰取样,测量小肠黏膜的绒毛高度和隐窝深度,检测空肠、回肠、结肠及血清中的过氧化氢酶(CAT)、总一氧化氮合成酶(TNOS)与诱导型一氧化氮合酶(iNOS)的活力及血清中丙二醛(MDA)与过氧化氢(H2O2)的含量。试验结果表明,与对照组相比,STa组仔猪各肠段绒毛高度均显著降低(P<0.05),十二指肠隐窝深度及绒毛高度与隐窝深度的比值显著降低(P<0.05),十二指肠、空肠、回肠绒毛表面积显著降低(P<0.05);同时,STa组回肠、结肠CAT活力显著降低,血清、空肠、回肠和结肠中的TNOS活力显著降低(P<0.05),STa组结肠iNOS活力显著降低(P<0.05),STa组血清中的MDA与H2O2含量显著提高(P<0.05)。结果显示,表达STa的重组大肠杆菌可导致7日龄仔猪肠道结构损伤和抗氧化能力下降。  相似文献

This experiment was conducted to investigate the effects of recombinant E.coli expressing heat-stable enterotoxin(STa) on intestinal absorption and barrier function, intestinal morphology and antioxidant capacity of 7 days old piglets. Twenty-four 7 days old piglets were allotted to four treatments:control group (artificial milk), STa group (artificial milk +2×109 CFU E.coli LMG194-pBAD-STa), LMG194 group (artificial milk +2×109 CFU E.coli LMG194), and K88 group (artificial milk +2×109 CFU E.coli K88).The pigs were treated with E.coli on the 5th day and slaughtered on the 7th day. The results showed that, compared with the control group, villus height in jejunum, ileum and duodenum, crypt depth and villus height/crypt depth in duodenum, and small intestine villi surface area were significantly decreased in STa group (P<0.05). The activities of catalase (CAT) in ileum and colon, total nitric oxide synthase (TNOS) in serum, ileum, jejunum and colon, inducible nitric oxide synthase (iNOS) in colon were significantly decreased in STa group (P<0.05). STa group also had a higher levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in serum (P<0.05). These results suggested that recombinant E.coli expressing STa could lead to intestinal injury and oxidative stress of 7 days old piglets.  相似文献

通过用加味葛根芩连汤治疗仔猪的湿热泄泻,探讨其对湿热泄泻仔猪肠道的修复作用。选取8~15日龄未断奶仔猪(大白×长白)30头,其中健康仔猪6头,为对照组;有粪色黄褐而臭、小便短赤、舌质红等症状的泄泻仔猪24头,分为湿热泄泻组、加味葛根芩连汤低剂量组、加味葛根芩连汤中剂量组和加味葛根芩连汤高剂量组,每组6头。对照组和湿热泄泻组仔猪灌服生理盐水,药物组仔猪灌服加味葛根芩连汤(按生药量,低、中、高剂量组药物添加量分别为2.5、5.0和10.0 g·d-1),连续5 d。在服药的第0、3、5天记录各组仔猪的粪便评分。取对照组、湿热泄泻组、加味葛根芩连汤中剂量组仔猪十二指肠、空肠、回肠和结肠进行组织切片和H.E.染色,取十二指肠进行增殖性细胞核抗原(PCNA)免疫组化染色;用荧光定量PCR检测对照组、湿热泄泻组、加味葛根芩连汤中剂量组仔猪空肠TLR4、TNF-αIL-1β和IL-6 mRNA表达量变化。湿热泄泻严重破坏了仔猪的十二指肠、空肠、回肠和结肠结构,尤其是十二指肠和空肠,十二指肠隐窝处PCNA表达量显著升高(P<0.05)。与对照组相比,湿热泄泻组仔猪空肠TLR4(P<0.05)、TNF-αP<0.01)、IL-1β(P<0.01)和IL-6(P<0.05)mRNA表达量显著升高。相比于湿热泄泻组,加味葛根芩连汤显著降低了仔猪的粪便评分(P<0.05)。中剂量加味葛根芩连汤组仔猪的小肠和结肠的组织结构得到显著改善,十二指肠的肠腔中有不断脱落的绒毛团,绒毛中的PCNA表达量极显著升高(P<0.01)。与湿热泄泻组相比,加味葛根芩连汤中剂量组空肠TLR4(P<0.05)、TNF-αP<0.01)、IL-1β(P<0.01)和IL-6(P<0.05)mRNA表达量显著降低。综上表明,加味葛根芩连汤可通过促进隐窝干细胞的增殖和降低炎症反应对湿热泄泻造成的仔猪肠道损伤进行修复。  相似文献

对小尾寒羊羔羊小肠不同部位(十二指肠、空肠和回肠)pH及淀粉酶、胰蛋白酶、糜蛋白酶、脂肪酶和乳糖酶的活性进行了测定,结果表明:羔羊小肠不同部位内容物pH不同,回肠pH显著高于空肠,空肠和回肠pH显著高于十二指肠,羔羊1月龄后,小肠各段pH变化不大;小肠不同部位淀粉酶和胰蛋白酶、糜蛋白酶及脂肪酶活性不同,空肠段淀粉酶、胰蛋白酶、糜蛋白酶及脂肪酶活性高于十二指肠和回肠段,随着羔羊日龄的增长,小肠淀粉酶、胰蛋白酶、糜蛋白酶及脂肪酶活性增加,3月龄后各消化酶活性变化差异不显著(P<O.05);羔羊小肠不同部位乳糖酶活性不同,空肠乳糖酶活性显著高于十二指肠和回肠,随着羔羊日龄的增长,小肠乳糖酶活性逐渐降低。  相似文献
