Uncharged tRNA and sensing of amino acid deficiency in mammalian piriform cortex

Recognizing a deficiency of indispensable amino acids (IAAs) for protein synthesis is vital for dietary selection in metazoans, including humans. Cells in the brain's anterior piriform cortex (APC) are sensitive to IAA deficiency, signaling diet rejection and foraging for complementary IAA sources, but the mechanism is unknown. Here we report that the mechanism for recognizing IAA-deficient foods follows the conserved general control (GC) system, wherein uncharged transfer RNA induces phosphorylation of eukaryotic initiation factor 2 (eIF2) via the GC nonderepressing 2 (GCN2) kinase. Thus, a basic mechanism of nutritional stress management functions in mammalian brain to guide food selection for survival.     

Membranous structures associated with translation and transcription of poliovirus RNA

Poliovirus RNA and proteins are synthesized in association with distinct membranous structures that were separated by means of Isopycnic centrifugation of cytoplasmic extracts in discontinuous sucrose-density gradients. Viral RNA is replicated in a structure that contains rapidly labeled replicative intermediate RNA and viral RNA polymerase associated with the smooth membrane fraction. In sucrose gradients this viral RNA replication complex is distributed at densities in the range of 1.12 to 1.18 grams per cubic centimeter. Viral proteins are synthesized on polyribosomes bound to membranes and sediment with polyribosomes at densities of less than 1.25 grams per cubic centimeter.     

拟南芥翻译起始因子eIF5B-1调控种子萌发的分子机制

为明确eIF5B-1基因在拟南芥种子萌发过程中的生理作用。以拟南芥翻译起始因子的T-DNA插入突变体eif5b-1为材料。测定突变体eif5b-1种子的萌发速率,同时利用多聚核糖体谱和qRT-PCR的研究方法,对拟南芥野生型(WT)和eif5b-1突变体的萌发过程进行了研究。结果表明:1)eif5b-1突变体种子的萌发率下降;2)eif5b-1突变体种子萌发过程中多聚核糖体的形成受到影响;3)eif5b-1突变体中ABI1、ABI3、ABI4、ABI5及DOG1基因的转录受到影响;4)eif5b-1突变体选择性地影响ABI4和DOG1基因的翻译效率。

Structure of the 70S ribosome complexed with mRNA and tRNA

The crystal structure of the bacterial 70S ribosome refined to 2.8 angstrom resolution reveals atomic details of its interactions with messenger RNA (mRNA) and transfer RNA (tRNA). A metal ion stabilizes a kink in the mRNA that demarcates the boundary between A and P sites, which is potentially important to prevent slippage of mRNA. Metal ions also stabilize the intersubunit interface. The interactions of E-site tRNA with the 50S subunit have both similarities and differences compared to those in the archaeal ribosome. The structure also rationalizes much biochemical and genetic data on translation.     

L-dopa: disaggregation of brain polysomes and elevation of brain tryptophan

One hour after administration of L-dopa (50 to 300 milligrams per kilogram), there is a marked disaggregation of brain polysomes in immature rats. Adult animals show a similar response, but require larger doses of the amino acid (500 milligrams per kilogram). Single doses of L-dopa significantly elevate amounts of tryptophan in the brain; hence their effect on polysomes does not result from the unavailability of this amino acid.
