Protection tests in guinea pigs indicate that vaccines prepared from virus propagated in chick embryo cell cultures are as effective as the purified whole chick embryo vaccines which are currently used for human immunization against eastern equine encephalomyelitis.  相似文献

When cultures of cells derived from the entoderm of the chick embryo were infected with trachoma virus, cytoplasmic inclusion bodies composed of viral particles were easily demonstrated. The inclusions are similar to those found in stained smears from trachomatous eyes and they developed in a sequence characteristic for this group of viruses. This method of culture appears to offer a valuable additional tool for study of the trachoma agent.  相似文献

Several mutants resistant to the herbicides chlorsulfuron and sulfometuron methyl were isolated form cultured cells of Nicotiana tabacum. Resistance was inherited as a single dominant or semidominant mutation in all cases. Linkage analysis of six mutants identified two unlinked genetic loci. Studies of plants homozygous for one mutation showed the mutant plants to be completely resistant to treatment with a concentration of chlorsulfuron 100 times higher than that which produces symptoms of phytotoxicity on normal plants.  相似文献

Cyclic changes in enzyme activity in synchronized mammalian cell cultures   总被引:8,自引:0,他引:8  
Activity of the enzymes glucose-6-phosphate dehydrogenase and lactate dehydrogenase increases intermittently in synchronized cultures of Chinese hamster cells. During G(1) phase (3 hours after mitosis), midway through S phase (7 hours after mitosis) and again in late S phase (10 hours after mitosis), rapid increases in activity were observed and correlated with a net increase in cell size and total protein. The evidence suggests that this is a general phenomenon which affects a whole population of proteins and may be the expression of an endogenous cellular rhythm.  相似文献

S-49 mouse lymphoma cells undergo lysis when treated with glucocorticoids; the mechanism of this effect is not understood. A protein was detected in the plasma membrane of these cells by means of direct immunofluorescent labeling with a monoclonal antibody to the soluble glucocorticoid receptor. Cellular heterogeneity in the content of this glucocorticoid receptor-like molecule was evident. By immunoadsorption to antibody-coated tissue culture plates, the cells were separated into populations positive (100%) and depleted (38%) for this membrane antigen. Gel electrophoresis, specific immunoblot, and autoradiographic (binding of [3H]dexamethasone mesylate) analysis of the membrane proteins from the membrane antigen-positive group revealed multiple protein bands ranging in size from 85 to 145 kilodaltons. Furthermore, comparison of the glucocorticoid sensitivity of these groups of cells showed complete lysis of the membrane antigen-positive cells and only partial lysis of the antigen-deficient group, which suggests that the lysis response of cells to glucocorticoids is mediated by a glucocorticoid receptor-like molecule located in the plasma membrane.  相似文献

Signals propagated through the B cell antigen receptor (BCR) are vital for the development and survival of B lymphocytes in both the bone marrow and the periphery. These signals not only guide maturation and activation but also affect the removal of potentially self-reactive B lymphocytes. Interestingly, these signals are known to be either ligand-independent ("tonic" signals) or induced by ligand (antigen) binding to the BCR. We focus on the problems that occur in B cell development due to defects in signals emanating from the BCR. In addition, we present the B Cell Antigen Receptor Pathway, an STKE Connections Map that illustrates the events involved in B cell signaling.  相似文献

Epidermal growth factor and fibroblast growth factor inhibited follicle-stimulating hormone-dependent induction of luteinizing hormone receptor in cultured ovarian granulosa cells of the rat. In contrast, platelet-derived growth factor potentiated the induction of luteinizing hormone receptor by follicle-stimulating hormone. These results indicate that growth factors, well known for their effects on mitosis and DNA synthesis in cultured mammalian cells, are also able to modulate hormone-dependent differentiation in an endocrine target cell.  相似文献

Mature male and female gametocytes of Eimeria tenella, a coccidium of the chicken ceca, developed in primary cultures of chick embryonic kidney cells inoculated with sporozoites. Immature gametocytes appeared in the cultures after approximately 144 hours of incubation at 41 degrees C. Dendritic cells (DCs) process and present self and foreign antigens to induce tolerance or immunity. In vitro models suggest that induction of immunity is controlled by regulating the presentation of antigen, but little is known about how DCs control antigen presentation in vivo. To examine antigen processing and presentation in vivo, we specifically targeted antigens to two major subsets of DCs by using chimeric monoclonal antibodies. Unlike CD8+ DCs that express the cell surface protein CD205, CD8- DCs, which are positive for the 33D1 antigen, are specialized for presentation on major histocompatibility complex (MHC) class II. This difference in antigen processing is intrinsic to the DC subsets and is associated with increased expression of proteins involved in MHC processing.  相似文献

Lability of host-cell DNA in growing cell cultures due to Mycoplasma   总被引:11,自引:0,他引:11  
In HeLa-cell cultures chronically infected with Mycoplasma, (PPLO) host-cell DNA is unstable as detected by incorporation of H(3)-or C1 i-thymidine into DNA and subsequent release into the medium as acidsoluble radioactivity. This characteristic can be transmitted to PPLO-free cultures of strain L cells by inoculation with preparations of PPLO from the HeLa cells, although chronically infected cultures of L cells continue to multiply. In addition, virus preparations also may carry PPLO contamination through numerous passages.  相似文献

Cultures of white cells derived from peripheral blood of individuals homozygous and heterozygous for the inherited mucopolysaccharidoses revealed a distinct intracellular metachromatic staining with toluidine blue O. These short-term cultures circumvent the technical problems of skin fibroblast cultures and provide a simple screening procedure to detect the heterozygous state for the mucopolysaccharidoses, as well as offering an opportunity to study the heterozygous state of various inherited storage diseases.  相似文献

Alpha-aminoisobutyric acid transport in hepatoma cells in culture was increased by insulin but not by hydrocortisone. Both of these agents induce tyrosine aminotransferase activity in this system. The apparent increase in alpha-aminoisobutyric acid transport and tyrosine aminotransferase activity produced by glucagon is probably caused by insulin contamination. Insulin did not increase transport in this system until after tyrosine aminotransferase activity had reached maximum levels. The mechanisms underlying increased alpha-aminoisobutyric acid transport appear to differ from those for tyrosine aminotransferase induction with hydrocortisone despite their close association in previous whole animal experiments.  相似文献

以烟草(Nicotiana tabacum L.)细胞作为材料,用改良后的培养基进行悬浮培养,在培养周期内不同的培养阶段,对烟草细胞生长和培养基中碳源、硝态氮、磷酸盐等无机元素含量的变化及烟草细胞内CoQ10合成的情况进行了测定.结果表明,在细胞培养周期内,细胞生长与次生代谢产物的合成基本上同步;培养基中碳源、硝态氮和磷酸盐的消耗曲线与细胞生长的3个阶段基本上一致.  相似文献
