The structure and mechanical properties of onions are important factors affecting their textural quality. The onion bulb consists of several layers of pigmented, papery scales surrounding fleshy storage scales that comprise an upper epidermis, an intermediate parenchyma tissue, and a lower epidermis. The purpose of this study was to examine the chemical composition of cell walls from the papery scales and outer fleshy scales of onion (Allium cepa L. cv. Sturon) in relation to their mechanical properties. Cell-wall material (CWM) was prepared from the component tissues and analyzed for its carbohydrate and phenolic composition. The CWMs were rich in uronic acid and glucose, with smaller quantities of arabinose, galactose, and xylose. In the fleshy scales, the lower epidermis contained relatively more galactose-rich pectic polysaccharides, whereas the upper epidermis and the papery scales contained virtually no galactose. Analysis of mechanical properties showed that the order of strength of the tissues was papery scales > fleshy scales, which were in the order lower epidermis > upper epidermis > intermediate parenchyma. The upper epidermis of fleshy scales was stronger in the vertical than the horizontal direction, and both orientations showed negligible notch sensitivity. Cyclohexane-trans-1,2-diaminetetraacetate-induced vortex-induced cell separation of the intermediate layer of fleshy scales indicated that calcium cross-linking may play an important role in cell-cell adhesion. A small but significant amount of ferulic acid was found in the walls, predominantly in the thick cuticle of the lower epidermis of fleshy scales. Alkali-labile wall-bound flavonoids were also detected.  相似文献

The following organic acids (glutamic, oxalic, pyruvic, malic, tartaric, citric, and fumaric), pungency, Brix degree, acidity, and pH were determined in onion cultivars (Texas, Guayonje, San Juan de la Rambla, Carrizal Alto, Carrizal Bajo, and Masca) harvested in the same agroclimatic conditions. Glutamic acid was the most abundant organic acid (325 +/- 133 mg/100 g) followed by citric acid (48.5 +/- 24.1 mg/100 g) and malic acid (43.6 +/- 10.4 mg/100 g). There were significant differences between the onion cultivars in the mean concentrations of all of the analyzed parameters. The San Juan de la Rambla and Masca cultivars presented, in general, higher concentrations of the organic acids than the other cultivars. Significant differences in most of the analyzed parameters were observed between the two seed origins for the Masca and San Juan de la Rambla cultivars. The onion samples tended to be classified according to the cultivar and, in the case of San Juan de la Rambla cultivar, according to the precedence of the seeds after applying discriminant analysis.  相似文献

High-performance liquid chromatography-diode array detector (HPLC-DAD) coupled with electron spray mass spectrometry (ESI-MS-MS) was used to determine the flavonol profile in southern Italian red onions (Allium cepa L.). This on-line technique allowed the identification of seven flavonols in southern Italian red onion, quercetin 4'-glucoside and quercetin 3,4'-diglucoside being the most abundant components. Five minor flavonols have been recognized, offering a characteristic profile of such compounds in red onions under study. Quercetin 3-glucoside, quercetin 7,4'-diglucoside, quercetin 3,7,4'-triglucoside, and isorhamnetin 4'-glucoside have been previously reported as minor flavonoid components in Allium cepa, while isorhamnetin 3,4'-diglucoside was previously found in Allium ascalonicum. Traces of isorhamnetin 3-glucoside and free quercetin were also detected.  相似文献

The purpose of this study was a comparative examination of the fructan and fructooligosaccharide (FOS) content of different varieties of onions (Allium cepa L. cv. Sturon, Hysam, Durco, Grano de Oro, and Caribo) and the changes produced during their commercial storage. In fresh onions, the Grano de Oro variety presented a remarkably different behavior, showing low contents of total fructans and FOS and high levels of reducing sugars. In the other varieties, Sturon, Hysam, Durco, and Caribo, fructans were the main carbohydrates, the lowest polymerized FOS being the major oligomer. Storage period caused in these varieties important increased levels of free fructose attributed to fructan hydrolysis. Maleic hydrazide treatment had no significant effect in avoiding the hydrolysis of fructans during storage conditions for the Sturon variety. Varieties with >16% dry matter or 15% soluble solids contents could be stored for 6 months at 0 degrees C and 60-65% relative humidity.  相似文献

Bulb samples from a range of onion cultivars grown over three consecutive years were freeze-dried and the resulting powders extracted using three previously reported methods. The extracts were analyzed for fructose, glucose, and sucrose content using HPLC coupled with ELSD, and for fructans using MALDI-MS. The three methods gave differing results, indicating that the extraction procedure is crucial in the determination of the concentration and ratios of nonstructural carbohydrates in onion bulbs. O'Donoghue et al.'s method (O'Donoghue, E. M.; Somerfield, S. D.; Shaw, M.; Bendall, M.; Hedderly, D.; Eason, J.; Sims, I. J. Agric. Food Chem. 2004, 52, 5383-5390), which utilized a more polar solvent (62.5% (v/v) aqueous methanol) and also had the benefit of shorter extraction times and lower temperatures, was far superior to 80% (v/v) ethanol-based methods in extracting significantly greater amounts of fructose, glucose, and sucrose from all onion bulbs tested. Discrepancies between and within cultivars tested also demonstrated that the ratio of monosaccharides to sucrose was affected by extraction method, such that some caution should be given to interpreting some previous work on elucidating the nonstructural carbohydrate composition in onion.  相似文献

Rhizospheric pH changes induced by arbuscular mycorrhiza formation in onion plants fertilized either with NO3? or NH4+ were studied. The pH changes promoted by either mycorrhizal or non-mycorrhizal roots were studied by means of a non-destructive technique using the pH indicator bromocresol purple. Results showed that the pH changes observed depended on i) the symbiotic status of the root and ii) the N form amended to the soil. When growing in a NH4+-supplied soil, mycorrhizal onion roots produced more intense and wider acidification halos than non-mycorrhizal plants did. These differences were maintained throughout the whole experiment (60 days). NO3?-supplied mycorrhizal roots initially promoted a more intense alkalinization on their surface, compared to the control roots (30 days); however, at the end of the experiment (60 days), intense acidification halos were observed in the mycorrhizosphere, whereas this acidification was almost absent in the non-mycorrhizal rhizosphere. The link between these mycorrhiza-induced pH changes in the soil and the higher efficiency in the exploitation of nitrogen in the rhizosphere by the arbuscular-mycorrhizal plants is discussed.  相似文献

Onion tissues of three varieties were evaluated for dietary fiber (DF) composition. Insoluble (IDF) and soluble (SDF) dietary fibers were subjected to acid hydrolysis, and the resultant neutral sugars, uronic acids, and Klason lignin were quantified. Brown skin exhibited the highest total dietary fiber (TDF) content (65.8%) on a dry matter basis, followed by top (48.5%) and bottom (38.6%), IDF being the main fraction found. The SDF:IDF ratio decreased from inner to outer tissues. Brown skin and outer leaves byproducts appear to be the most suitable sources of DF that might be used in food product supplementation. The chemical composition reveals that cellulose and pectic polysaccharides were the main components of onion DF in all tissues, although differences between them were noticed. An increase in the uronic acids/neutral sugars ratio from inner to outer tissues was found, suggesting that the galactan side chain shows a DF solubilization role.  相似文献

 N2O emissions were measured from three contrasting onion (Allium cepa L.) production systems over an 8.5-month period. One system was established on soil where a clover sward had 3 months earlier been ploughed in (ploughed clover site). This production system followed conventional production management practices. The other two systems were established on soil where a mixed herb ley had 3 months earlier been either ploughed or rotovated. These last two production systems followed the guidelines of the International Federation of Organic Agriculture Movements (IFOAM). Cumulative N2O emissions were significantly greater from the ploughed clover site compared to the ploughed ley site (3.8 and 1.6 kg N2O-N ha–1, respectively), while cumulative N2O emissions from the ploughed ley and rotovated ley sites were not significantly different from each other. Emissions from all sites were dominated by episodes of high N2O flux activity following seedbed preparation and drilling, when soil water suction (SWS) was shown to be the rate-controlling variable. The decline in the N2O fluxes after these peak emissions followed clear exponential relationships of the form F=Ae kt (r≥0.91), where F is the daily flux and A is the y-intercept. First-order decay constants (k) during these periods of declining N2O fluxes (corresponding to half-lives of 2.6–3.0 days) were not significantly different in magnitude from the first-order rate constants that characterised the increasing SWS. Gross differences in cumulative emissions between the clover and ley sites were attributed to the influence of differing soil pHs at the two sites on the N2O:(N2O+N2) ratio in the denitrification products. It also appeared that fertiliser applications to the clover site had both direct and indirect effects on N2O emissions by: (1) enhancing N2O emissions via potential nitrification, (2) increasing the NO3 supply for enhanced N2O emissions via denitrification, and (3) influencing the N2O:(N2O+N2) ratio by lowering soil pH and increasing NO3 concentrations. Onion crop yields were greater at the clover site, mainly due to the higher density of planting made possible under a conventional production philosophy. Expressing the yield on the basis of net N2O emissions, 23 t onions kg–1 N2O-N was obtained from the ploughed clover, which was double that obtained for the two systems based on the ley site. However, when the N2O emissions from the cultivation of the soils prior to the sowing of the onions was included, all three systems produced a similar yield per kilogram of N2O-N emitted, averaging 10 t kg–1. Freeze-dried onion sprout was steam-distilled, and the distillate was extracted with dichloromethane (volatile sample). Water sample I was obtained from the residual aqueous solution in the extractor. The filtrate and the methanol extract of filtrand from the residual aqueous solution in the steam distillation flask were named water sample II and methanol sample, respectively. Among the total of 71 components identified in the volatile sample, 24 were sulfur-containing compounds, which comprised 36.87% of the total volatile chemicals identified. The volatile sample inhibited hexanal oxidation for 40 days by >99% at levels >100 microg/mL. The volatile sample and water sample II exhibited moderate antioxidant activity in a malonaldehyde/gas chromatography assay and thiobarbituric acid assay, whereas water sample I did not show appreciable activity. The volatile sample, water sample I, and water sample II exhibited anti-inflammatory activity with a dose-related response in the lipoxygenase inhibitor screening assay. However, the methanol sample did not show appreciable activity in either antioxidant or anti-inflammatory tests. The results suggest that onion sprouts can be an excellent food source.  相似文献

One gram of onion added to the food of rats inhibits significantly (p < 0.05) bone resorption as assessed by the urinary excretion of tritium released from bone of 9-week-old rats prelabeled with tritiated tetracycline from weeks 1 to 6. To isolate and identify the bone resorption inhibiting compound from onion, onion powder was extracted and the extract fractionated by column chromatography and medium-pressure liquid chromatography. A single active peak was finally obtained by semipreparative high-performance liquid chromatography. The biological activity of the various fractions was tested in vitro on the activity of osteoclasts to form resorption pits on a mineralized substrate. Medium, containing the various fractions or the pure compound, was added to osteoclasts of new-born rats settled on ivory slices. After 24 h of incubation, the tartrate-resistant acid phosphatase positive multinucleated cells, that is, osteoclasts, were counted. Subsequently, the number of resorption pits was determined. Activity was calculated as the ratio of resorption pits/osteoclasts and was compared to a negative control, that is, medium containing 10% fetal bovine serum only and to calcitonin (10(-12) M) as a positive control. Finally, a single peak inhibited osteoclast activity significantly (p < 0.05). The structure of this compound was elucidated with high-performance liquid chromatography-electrospray ionization-mass spectrometry, time-of-flight electrospray ionization mass spectrometry, and nuclear magnetic resonance spectroscopy. The single peak was identified as gamma-L-glutamyl-trans-S-1-propenyl-L-cysteine sulfoxide (GPCS). It has a molecular mass of 306 Da and inhibits dose-dependently the resorption activity of osteoclasts, the minimal effective dose being approximately 2 mM. As no other peak displayed inhibitory activity, it likely is responsible for the effect of onion on bone resorption.  相似文献

The L-cysteine derivatives (R)-2-amino-3-(methyldisulfanyl)propanoic acid (S-methylthio-L-cysteine), (R)-2-amino-3-(propyldisulfanyl)propanoic acid (S-propylthio-L-cysteine), (R)-2-amino-3-(1-propenyldisulfanyl)propanoic acid (S-(1-propenylthio)-L-cysteine), and (R)-2-amino-3-(2-propenyldisulfanyl)propanoic acid (S-allylthio-L-cysteine) were prepared from 3-[(methoxycarbonyl)dithio]-L-alanine, obtained from the reaction of L-cysteine with methoxycarbonylsulfenyl chloride. The occurrence of these S-(+)-alk(en)ylthio-L-cysteine derivatives in onion (Allium cepa L.) was proven by using UPLC-MS-ESI(+) in SRM mode. Their concentrations in fresh onion were estimated to be 0.19 mg/kg S-methylthio-L-cysteine, 0.01 mg/kg S-propylthio-L-cysteine, and 0.56 mg/kg (S-(1-propenyllthio)-L-cysteine, concentrations that are about 3000 times lower than that of isoalliin (S-(1-propenyl-S-oxo-L-cysteine). These compounds were treated with Fusobacterium nucleatum, a microorganism responsible for the formation of mouth malodor. These L-cysteine disulfides were demonstrated to predominantly produce tri- and tetrasulfides. Isoalliin is almost entirely consumed by the plant enzyme alliin lyase (EC 4.4.1.4 S-alk(en)yl-S-oxo-L-cysteine lyase) in a few seconds, but it is not transformed by F. nucleatum. This example of flavor modulation shows that the plant produces different precursors, leading to the formation of the same types of volatile sulfur compounds. Whereas the plant enzyme efficiently transforms S-alk(en)yl-S-oxo-L-cysteine, mouth bacteria are responsible for the transformation of S-alk(en)ylthio-L-cysteine.  相似文献

A field experiment was conducted to investigate the role of boron and zinc on growth, yield and quality of onion. There were eight treatment combinations consisted of four levels of boron (0, 0.1, 0.2 and 0.5%) and 4 levels of zinc (0, 0.1, 0.2 and 0.5%) were applied as foliar spraying. application of 0.5% boron significantly increased the growth (plant height, 63.93cm and number of leaves per plant, 7.25), yield (30.74 t ha?1) and quality (total soluble solids, 13.45 0B and pyruvic acid 5.94 µmol g?1) of onion. Among various levels of zinc 0.5% exhibited the best growth (plant height, 67.25cm and number of leaf per plant, 7.75), yield (33.34 t ha?1) and quality (total soluble solids, 14.57 0B and pyruvic acid, 5.86 µmol g?1) attributes of onion. These results suggest that the foliar application of boron and zinc significantly influenced the growth, yield and quality of onion.  相似文献

The rapid quantification of flavonoid compounds in onions by attenuated total reflectance (ATR) Fourier transform infrared (FT-IR) spectroscopy combined with multivariate analysis was evaluated as a possible alternative to high-performance liquid chromatography (HPLC) analysis. Quercetin content in onion varieties (yellow, red, and sweet) was quantified using ATR FT-IR (4000 to 400 cm?1) spectroscopy and HPLC methods. Quercetin-3,4'-O-diglucoside (3,4'-Qdg) and quercetin-4'-O-glucoside (4'-Qmg) comprised >80% of the total flavonol content detected in the studied varieties. The quercetin compounds (3,4'-Qdg and 4'-Qmg) and total flavonol conjugates were quantified by HPLC, and results correlated closely with ATR-IR values (R > 0.95). Cross-validated (leave-one-out) partial least-squares regression (PLSR) models successfully predicted concentrations of these quercetins. The standard errors of cross-validation (SECV) of 3,4'-Qdg and 4'-Qmg, total quercetin, and total flavonol contents of onions were 20.43, 21.18, and 21.02 mg/kg fresh weight, respectively. In addition, supervised and unsupervised segregation analyses (principal component analysis, discriminant function analysis, and soft independent modeling of class analogue) were performed to classify onion varieties on the basis of unique infrared spectral features. There was a high degree of segregation (interclass distances > 3.0) for the different types of onion. This study indicated that the IR technique could predict 3,4'-Qdg, 4'-Qmg, total quercetin, and total flavonol contents and has advantages over the traditional HPLC method in providing a valid, efficient, and cost-effective method requiring less sample preparation for the quantification of quercetins in onion.  相似文献

The bulb onion (Allium cepa L.) was historically seed propagated by open pollination (OP). Cytoplasmic-genic male sterility (CMS) and protandry encourage outcrossing among individual onion plants. The most common source of CMS in onion is conditioned by the interaction of sterile (S) cytoplasm with a single nuclear male-fertility restoration (Ms) locus. We previously reported that the majority of OP onion populations possess normal (N) male-fertile cytoplasm and varying frequencies of the dominant Ms allele. It was unclear why N-cytoplasmic onion populations often possess relatively high frequencies of the Ms allele, which has no obvious function. We used computer simulations to estimate changes in allelic frequencies at Ms for onion populations possessing S-cytoplasm or a mixture of N- and S-cytoplasms, and to determine if frequencies of the Ms allele stay constant or change due to failure of male gamete production from male-sterile (S msms) plants. The models revealed selection against the recessive ms allele over generations in onion populations possessing S-cytoplasm and varying amounts of self pollination and inbreeding depression. These models were consistent with field and molecular analyses documenting that N-cytoplasm and the dominant Ms allele predominate in OP onion populations.  相似文献

Phosphorus uptake by plant roots and arbuscular mycorrhizae (AM) hyphae can be described based on morphological and physiological uptake characteristics and parameters affecting P transport in the soil to the uptake surface. The current study aimed to quantify the relative contribution of hyphae and roots to P uptake of onion and to investigate the associated mechanisms using a mechanistic simulation model. Onion was grown in a growth chamber under controlled conditions at five levels of P supply with (M) or without (NM) AM. Uptake by onion roots and AM hyphae was simulated separately using a nutrient‐uptake model, which was developed for roots. Model parameters describing P transport in the soil, as well as morphological and physiological uptake characteristics of roots and hyphae were derived from soil and nutrient‐solution experiments. The model fairly described P uptake of M and NM plants at the highest level of P supply (28 µM P in soil solution) where roots and hyphae contributed nearly equally to P uptake of M plants. However, at lower P levels (≤0.41 µM P), hyphae accounted for nearly the whole P uptake whereas roots hardly explained any uptake. The effectiveness of AM hyphae at low P supply was well explained by their favorable morphological and physiological uptake properties. Hyphae were characterized by a diameter of 3 × 10–4 cm, a Km of 0.25 µM, and a Cmin of 0.04 µM, values that are about 200, 40, and 10 times lower, respectively, compared to those of roots. Not more than 60% of the uptake at lower levels of P supply could be explained by the sum of predicted uptake of roots and hyphae suggesting that hyphae and roots may have used mechanisms additional to those described by the model to mobilize P. This work demonstrated that mechanistic models can be useful tools to study the role of AM in P uptake of plants, although reliable estimation of model parameters, especially physiological uptake properties of AM, is still limiting this approach.  相似文献

Genetic lines and example cultivars are used as markers to define variation when describing or evaluating wild or cultivated germplasm. Occasionally genotypes previously described in the literature are no longer available. This was thought to be the case for chartreuse skin colour in Onion. However, registration authorities undertaking Distinctness, Uniformity and Stability tests recently observed this skin colour in the cultivar Greenella and have provided seed to a genebank for long-term conservation. This characteristic is suitable both for use as a genetic marker and as an example cultivar for describing Onion skin colours.  相似文献

A phytochemical analysis of the polar extract from the red bulbs of Allium cepa L. var. Tropea, typical of Calabria, a southern region of Italy, was performed extensively for the first time, leading to the isolation of four new furostanol saponins, named tropeoside A1/A2 (1a/1b) and tropeoside B1/B2 (3a/3b), along with the respective 22-O-methyl derivatives (2a/2b and 4a/4b), almost certainly extraction artifacts. High concentrations of ascalonicoside A1/A2 (5a/5b) and ascalonicoside B (6), previously isolated from Allium ascalonicum Hort., were also found. This is the first report of furostanol saponins in this A. cepa variety. The chemical structures of the new compounds were established through a combination of extensive nuclear magnetic resonance, mass spectrometry and chemical analyses. High concentrations of quercetin, quercetin 4(I)-glucoside, taxifolin, taxifolin 7-glucoside, and phenylalanine were also isolated. The new saponins were found to possess antispasmodic activity in the guinea pig isolated ileum; such an effect might contribute to explaining the traditional use of onion in the treatment of disturbances of the gastrointestinal tract.  相似文献

During onion processing, the outer dried protective layer (outer paper layer) and first two fleshy leaf layers are removed. This coproduct material is a potential commercial source of flavonoids especially quercetin. In the following study, the flavonoid composition was determined in coproduct materials and the press cake (material generated after juice extraction) in several commercially important onion varieties grown in California. Flavonoids were characterized and quantified using LC-(ESI)MS/MS and HPLC. The long-term stability of quercetin glycosides was assessed in dried coproduct materials stored at 4 and 22 °C over a 12 month period. In all varieties, the predominant forms of quercetin were the quercetin 3,4'-O-glucoside and 4'-O-glucoside. The first layer had significantly higher levels of flavonoids than the outer paper, second, and inner flesh layers on a DW basis (p < 0.05). Allium cepa "Milestone" contained the highest levels (p < 0.05) of flavonoids (1703 mg/100 g on a dry weight basis (DW). Onion press cake had significantly higher levels of total quercetin as compared with fresh onions (p < 0.05). The levels of 4'-O-glucoside significantly decreased during the first month of storage and remained stable for 12 months of storage at either 4 or 22 °C (p < 0.05).  相似文献
