Determination of free phenolic compounds in potato samples was optimized using a high-performance liquid chromatographic (HPLC) method with on-line diode array detection. This method was applied to samples of four cultivars of potatoes harvested in Tenerife (Canary Islands). The free phenolic compounds found in the potato samples were (+)-catechin, chlorogenic acid, caffeic acid, p-coumaric acid, and ferulic acid. Potato samples belonging to Colorada cultivar, ssp. andigena, had mean concentrations of total phenolic compounds and chlorogenic acid higher than those found for Kerr's Pink and Cara cultivars, ssp. tuberosum, and for Negra cultivar, S. x chaucha. In contrast, p-coumaric acid was not detected in any potato samples of the Colorada cultivar. Traditional potatoes presented a higher mean concentration of ferulic acid than recently imported potatoes. A significant and negative correlation was established between (+)-catechin and p-coumaric acid. A considerable contribution to the daily intake of flavonoids was observed with the actual consumption of potatoes.  相似文献

A statistical study of correlation, factorial, and discriminant analysis on the metal composition (Se, Fe, Cu, Zn, Na, K, Ca, Mg) of different types of milks (human, cow, goat, pasteurized, and powdered infant formula) was carried out to establish the relationships between the metal concentrations and, therefore, differentiate the samples according to the type of milk. A large number of significant intermetallic correlations were found in all samples, which could be due to biological relationships between the metals studied. After the factorial analysis, the dimension space was reduced from eight variables to two factors, accounting for approximately 71.4% of the total variance. After an orthogonal rotation, the first factor was positively correlated with Ca and the second factor with Fe. The representation of the scores makes it possible to separate not only human milk from powdered infant formula but also to separate both of these from the other milks. In the discriminant analysis, four discriminant functions were obtained, which are linear combinations of the quantitative variables that best explain the differences among the different milks analyzed. These functions make it possible to classify 98% of the samples analyzed within each type of milk correctly. Therefore, discriminant functions obtained here can be used to identify the origin of any milk sample.  相似文献

During the storage season changes in the chemical composition of the pectin moiety of the cell walls of raw potatoes were studied. This compositional analysis was performed for the cultivars Nicola and Irene, which represent two extremes with regard to sensory-perceived texture. Both cultivars were divided into three size categories. From each size category a dry matter (DM) distribution was made. From these distributions potatoes at the low and high ends of this distribution were selected for further analysis. In total 12 different samples were analyzed three times during the storage season. The analysis comprised a pectin fractionation study. Pectic fractions were extracted from the cell wall material (CWM) by increasing the harshness of the extraction procedure. This resulted in a calcium-complexed pectic fraction, two pectic fractions weakly bound to the CWM, and a residue fraction, respectively. It was shown that no statistically significant differences (p > or = 0.95), either in yield or in chemical composition, could be observed between the two cultivars studied (Nicola and Irene), between sizes (large, medium, and small), and between potatoes with either high or low DM contents. However, statistically significant effects of storage both on the yield and on the chemical composition of the pectic moiety of the CWM could be observed, irrespective of cultivar, size, and DM content. Despite the substantial changes in the composition of the pectic moiety of the CWM of the raw material, no to minimal changes in the sensory-perceived texture of the cooked potatoes were observed upon storage. This suggests that the observed changes in pectin composition upon storage are overruled by other aspects that contribute more importantly to the sensory-perceived texture of steam-cooked potatoes.  相似文献

Anthocyanins gain more and more importance as natural colourants. Besides many other plant species, coloured potatoes could serve as a potential source. Therefore, 27 potato cultivars and four breeding clones of different origin were analysed for their anthocyanin contents in fresh weight (fw). On average the highest amounts of anthocyanins were found in the skin (0.65 g kg−1 fw) of potatoes. The corresponding values of samples taken from whole tubers (0.31 kg−1 fw) and flesh (0.22 kg−1 fw) were significantly lower (p<0.03). The analysis revealed considerable differences in the amounts of anthocyanins between the 31 cultivars/breeding clones. Among them ȁ8Peru Purpleȁ9 revealed the highest anthocyanin content in the skin with 2.96 g kg−1 fw. A similar high value was reached by ȁ8Violettfleischigeȁ9 and clone 1.81.202–92 N. Also for the other two types of samples, flesh and whole tubers, these three genotypes exhibited the highest level of anthocyanins. Interestingly, different rates of nitrogen fertilization at 100 and 200 kg ha−1 had no significant effect on the pigment content of potatoes. The same was observed with respect to the year or the location of plant growth. There were also no significant changes in the anthocyanin contents of tubers during storage for 135 days. In dry matter, starch and protein contents the coloured potato cultivars/breeding clones were comparable with traditional cultivars. Further tests have shown that the glycoalkaloids were mainly localized in the skin of coloured potatoes.  相似文献

This work has focused on discriminating extra virgin olive oils from Sabina (Lazio, Italy) by olive fruit variety (cultivar). A set of oils from five of the most widespread cultivars (Carboncella, Frantoio, Leccino, Moraiolo, and Pendolino) in this geographical area was analyzed for chemical composition using only the Official Analytical Methods, recognized for the quality control and commercial classification of this product. The obtained data set was converted into a computer-compatible format, and principal component analysis (PCA) and a method based on the Fisher F ratio were used to reduce the number of variables without a significant loss of chemical information. Then, to differentiate these samples, two supervised chemometric procedures were applied to process the experimental data: linear discriminant analysis (LDA) and artificial neural network (ANN) using the back-propagation algorithm. It was found that both of these techniques were able to generalize and correctly predict all of the samples in the test set. However, these results were obtained using 10 variables for LDA and 6 (the major fatty acid percentages, determined by a single gas chromatogram) for ANN, which, in this case, appears to provide a better prediction ability and a simpler chemical analysis. Finally, it is pointed out that, to achieve the correct authentication of all samples, the selected training set must be representative of the whole data set.  相似文献

Red and white wheats must be segregated for marketing purposes because they have different end uses. Identification of wheat color is not straightforward, and currently there is interest in characterizing red and white wheats using spectroscopic methods and chemical tests. The kernels of both red and white wheats exhibit natural fluorescence that can be readily viewed under UV light, although it is not possible to differentiate the fluorescence spectra of red and white wheats by visual inspection only. Fluorescence emission spectra in the wavelength range of 370–670 nm for 91 wheat samples consisting of 48 red (from 30 cultivars) and 43 white (from 18 cultivars) were analyzed by partial least squares (PLS) and neural networks analyses (NNA). Samples included cultivars that were difficult to classify visually as well as wheat harvested after rainfall. Classification accuracies were ≈85% for calibration and ≈72% for the validation samples by both analyses. A plot of β‐coefficient vs. wavelength in PLS analysis indicated that fluorescence of red wheat cultivars was greater than that for white wheat cultivars at 425 (±20) nm wavelength. Fluorescence of white wheat cultivars was greater than that for red cultivars at 587 (±35) nm. Fluorescence emission at ≈450 nm from wheat samples increased in intensity after treatment with NaOH. The increase was greater for red than for white wheat. Wheat harvested after rainfall also exhibited a slight increase in fluorescence.  相似文献

Metabolite fingerprints, obtained with direct injection mass spectrometry (MS) with both positive and negative ionization, were used with analysis of variance-principal components analysis (ANOVA-PCA) to discriminate between cultivars and growing treatments of broccoli. The sample set consisted of two cultivars of broccoli, Majestic and Legacy, the first grown with four different levels of Se and the second grown organically and conventionally with two rates of irrigation. Chemical composition differences in the two cultivars and seven treatments produced patterns that were visually and statistically distinguishable using ANOVA-PCA. PCA loadings allowed identification of the molecular and fragment ions that provided the most significant chemical differences. A standardized profiling method for phenolic compounds showed that important discriminating ions were not phenolic compounds. The elution times of the discriminating ions and previous results suggest that they were common sugars and organic acids. ANOVA calculations of the positive and negative ionization MS fingerprints showed that 33% of the variance came from the cultivar, 59% from the growing treatment, and 8% from analytical uncertainty. Although the positive and negative ionization fingerprints differed significantly, there was no difference in the distribution of variance. High variance of individual masses with cultivars or growing treatment was correlated with high PCA loadings. The ANOVA data suggest that only variables with high variance for analytical uncertainty should be deleted. All other variables represent discriminating masses that allow separation of the samples with respect to cultivar and treatment.  相似文献

The potential of combining solid-phase microextraction (SPME) with gas chromatography and chemometric data analysis to differentiate between pure strawberry samples (Fragraria ananassa) and strawberry samples adulterated with 10, 40, and 70% (v/v) apple purée was investigated. The method involved the extraction of aroma volatiles from the headspace of the purée samples using a SPME fiber followed by GC analysis with flame ionization detection. The principal component analysis (PCA) data matrix consisted of the relative percent peak areas of 37 compounds deemed to be significant in the differentiation of the samples on the basis of adulteration. The PCA results clearly showed that differentiation of the adulterated and unadulterated samples was possible, particularly at the higher levels of adulteration. Partial least-squares regression (PLSR) using a dummy set of Y variables (set to 0 for unadulterated and 1 for adulterated samples) resulted in clear discrimination between unadulterated purées and those containing 40 and 70% (v/v) apple. PLSR using a second set of Y variables, consisting of the actual level of adulteration, enabled quantification of apple purée with a standard error of prediction of 11.6%, implying a minimum detectable level of 25% (v/v) apple. GC-MS analysis enabled identification of the compounds with the greatest influence on sample differentiation. These compounds were identified as hexanoic acid, 2-hexenal, and alpha-farnesene, all of which are key aroma compounds in apples.  相似文献

Three Chamaecyparis species (C. formosensis, C. obtusa, and C. obtusa var. formosana) are difficult to distinguish by the naked eye. Therefore, from the chemotaxonomic point of view, it would be valuable to find a simple and rapid method to differentiate these three Chamaecyparis species. In this study, the chemical compositions of biogenic volatile organic compounds (BVOCs) from mature leaves were analyzed using solid-phase microextraction-gas chromatography/mass spectrometry (SPME-GC/MS). Then cluster analysis (CA) and principal component analysis (PCA) were conducted for the BVOC constituents to reveal the differences among these three species. Results from SPME-GC/MS showed that the compositions of BVOCs from the three species were distinctly different. Moreover, these species were clearly differentiated according to the results of CA and PCA. In conclusion, the findings of this study suggest that SPME-GC/MS coupled with CA and PCA is a feasible and rapid technique to differentiate Chamaecyparis species with similar morphological characteristics.  相似文献

Diverse procedures have been reported for the separation and analysis by HPLC of the two major glycoalkaloids present in potatoes, alpha-chaconine and alpha-solanine. To further improve the usefulness of the HPLC method, studies were carried out on the influence of several salient parameters on the analysis of the two potato glycoalkaloids. Effects on retention (elution, separation) times of the (a) composition and pH of the mobile phase (acetonitrile and phosphate buffer), (b) concentration of the phosphate buffer, (c) capacity values of column packing of four commercial HPLC amino columns, (d) column temperature were studied. Except for pH, all of the variables significantly influenced the retention times. The results make it possible to select analysis conditions that produce well-separated as well as symmetrical peaks of the two glycoalkaloids. This improved HPLC method (limit of detection of approximately 150 ng) was evaluated with extracts from the cortex of one whole potato variety (May Queen) grown in Japan and the freeze-dried peel and flesh from the following eight cultivars grown in the United States: Atlantic, Dark Red Norland, Ranger Russet, Red Lasoda, Russet Burbank, Russet Norkota, Shepody, and Snowden. In addition, the same samples were analyzed by GC-MS for the presence of two water-soluble nortropane alkaloids, calystegine A(3) and calystegine B(2), reported to be potent glycosidase inhibitors. The following ranges for the eight varieties of total glycoalkaloid and calystegine levels were observed: dry flesh, 5-592 and 6-316 mg/kg; dry peel, 84-2226 and 218-2581 mg/kg; dry whole potatoes, 40-883 and 34-326 mg/kg; wet flesh, 1-148 and 1-68 mg/kg; wet peel, 12-429 and 35-467 mg/kg; wet whole potatoes, 7-187 and 5-68 mg/kg. The possible significance of the results to plant and food sciences is discussed.  相似文献

采用加标回收率的方法,分析并比较了硝酸银滴定法、紫外分光光度法和自动电位滴定法测定甘薯(Ipomoea batatas L.)幼苗Cl-含量的结果,并初步探明不同甘薯品种地上部和根系C1-吸收和分布规律.结果表明:(1)硝酸银滴定法重复性差且难以判断滴定终点;紫外分光光度法测定Cl-回收率低(52.9%~81.4%),且受样品浸提液颜色影响;自动电位滴定法Cl-回收率为93.9%~105.5%,相对标准偏差(RSD)为0.6%~ 6.0%,该方法操作简单、重复性好且精确度高,因此是测定甘薯氯含量的最佳方法;(2)利用自动电位滴定法测定不同甘薯品种Cl-积累和分布,结果显示:甘薯幼苗地上部Cl-含量为0.067~0.096 mmol g-1DW,根系Cl-含量为0.038~0.087 mmol g-1 DW,品种间甘薯幼苗地上部和根系Cl-含量皆存在显著差异.不同品种向地上部转运Cl-选择性比率均大于1.0,表明Cl-向地上部聚集,以菜用型福薯7-6比率最高,紫心型徐紫20-1和浙紫1号次之,淀粉型泉薯9号最低.  相似文献

Chestnut (Castanea sativa) is an important basic food in rural diets and a major starch crop used in a similar way to potatoes. Chestnuts are a fundamental economic resource in the "chestnut regions" not only for the fruit but also for the chestnut wood. Chestnuts have become increasingly important with respect to human health, for example, as an alternative gluten-free flour source. Chestnuts are also a rich source of other beneficial compounds, but there have been few studies on the composition during processing. In this study, we analyzed the chemical composition of three Portuguese cultivars at different stages of industrial processing. The chestnut cultivars were Longal, Judia, and Martaínha. All three cultivars had high moisture contents but were low in ash, crude fat, and crude protein contents, with high starch and low fiber contents. The free amino acid contents, including various essential amino acids, varied depending on the cultivar. All three cultivars also had a significant content of polyphenolics with gallic acid; ellagic acid was predominant among hydrolyzable and condensed tannins. Many of these compounds are known to exert significant positive effects on human health. The one-way analysis of variance for fresh chestnut shows significant differences among the three cultivars for most of the studied parameters. The same statistical analysis applied to each one of the two cultivars (Judia and Longal) sampled for the four processing steps analyzed indicates a significant effect of this factor in practically all of the constituents. On the other hand, the two-way analysis of variance shows that, besides the residual, the processing step and the interaction cultivar x processing step were the factors that more contributed for the total variation observed in the constituents analyzed, while the contribution of cultivar was much less significant.  相似文献

Ninety-one young varietal wines from the Valencian community (Spain), made from Cabernet Sauvignon, Tempranillo, Monastrell, and Bobal grapes, were tested on the basis of 33 variables: 9 conventional parameters, 10 alcohols and polyols, and 14 esters. Discriminant analysis was used to identify and explain the differences among samples, as well as to determine whether it is possible or not to differentiate among varieties. This differentiation (100% of the samples) has been possible due to the new discriminant analysis based on only 11 main variables: total acidity, cis-3-hexenol, methanol, glycerol, 2,3-butanediol, isobutyric alcohol, 1-pentanol, acetaldehyde, ethyl propionate, ethyl decanoato, and gamma-butyrolactone, which allow differentiating 100% of the 1994 vintage and 97% of the 1995 vintage.  相似文献

UV spectral fingerprints, in combination with analysis of variance-principal components analysis (ANOVA-PCA), can differentiate between cultivars and growing conditions (or treatments) and can be used to identify sources of variance. Broccoli samples, composed of two cultivars, were grown under seven different conditions or treatments (four levels of Se-enriched irrigation waters, organic farming, and conventional farming with 100 and 80% irrigation based on crop evaporation and transpiration rate). Freeze-dried powdered samples were extracted with methanol-water (60:40, v/v) and analyzed with no prior separation. Spectral fingerprints were acquired for the UV region (220-380 nm) using a 50-fold dilution of the extract. ANOVA-PCA was used to construct subset matrices that permitted easy verification of the hypothesis that cultivar and treatment contributed to a difference in the chemical expression of the broccoli. The sums of the squares of the same matrices were used to show that cultivar, treatment, and analytical repeatability contributed 30.5, 68.3, and 1.2% of the variance, respectively.  相似文献

Efficient selection of potato varieties with enhanced nutritional quality requires simple, rapid, accurate, and cost-effective assays to obtain tuber chemical composition information. Our objective was to develop simple protocols to determine phenolics, anthocyanins, and antioxidant capacity in polyphenolic extracts of potatoes using Fourier transform infrared spectroscopy combined with multivariate techniques. Lyophilized potato samples (23) were analyzed. Polyphenolic compounds were extracted from potatoes and applied directly applied onto a three-bounce ZnSe crystal for attenuated total reflectance measurements in the infrared region of 4000 to 700 cm (-1). Robust models were generated (r > or = 0.99) with standard error of cross-validation values of 4.17 mg gallic acid equivalent/100 g (total phenolics), 0.87 mg pelargonidin-3-glucoside/100 g (monomeric anthocyanins), and 130.8 mumol Trolox equivalent/100 g (antioxidant capacity) potato powder. In addition, classification models discriminated potato samples at the species and variety level. Application of a simple infrared spectroscopic protocol allowed simultaneous rapid quantification of specific nutritional components in potatoes and efficient selection of value-added potato varieties.  相似文献

Isozyme studies of genetic diversity and evolution in Hippophae   总被引:4,自引:0,他引:4  
Summary To provide information on the genetic variation, differentiation and evolution in Hippophae seed samples of 25 populations from China, Finland and Russia were electrophoretically analyzed. Of six loci investigated, four were good genetic markers for identifying species and subspecies. The percentages of polymorphic loci per population were 40.3% and 62.5% at 0.95 and 0.99 polymorphic criteria respectively. The mean number of alleles per locus per population was 2.1. Total genetic diversity in the material was 0.4614. Genetic diversity partitioning showed that there was a large amount of diversity residing within geographical populations (0.1354), between subspecies within species (0.1046) and between species (0.2566) but not between geographical populations (0.0114). There were nearly twice as many negative fixation indices as positive ones in Hippophae populations. The phylogenetic tree agreed very well with botanic classifications of the species and subspecies and their geographical distributions, and quantitatively presented the genetic relationships of 25 populations. A detailed view of the evolutionary stages in Hippophae showed clearly a general decline of similarity as evolutionary divergence continued, which further explained the evolution process in Hippophae.  相似文献

Volatile compounds of cooked rice from scented (Aychade, Fidji) and nonscented (Ruille) cultivars grown in the Camargue area in France were compared to that of a marketed Asian scented one (Thai) by gas chromatography-olfactometry (GC-O) and gas chromatography-mass spectrometry (GC-MS). GC-O analyses of the organic extracts resulted in the perception of 40 odorous compounds. Only two compounds, oct-1-en-3-one and 2-acetyl-1-pyrroline, were almost always perceived. Hierarchical cluster analysis showed that most of the difference between rice odors was linked to quantitative differences with only 11 compounds being specific to some of the rice. Sixty compounds were identified and quantified by GC-MS, including a few new odor-active components. Principal component analysis enabled us to differentiate scented cultivars from a nonscented one, and scented rice cultivars from Camargue from a Thai sample. Calculated odor-active values evidenced that the Thai sample odor differed from that of scented Camargue cultivars because of the degradation of lipids and of cinnamic acid compounds.  相似文献
