Ten enzymatic systems were analyzed to determine allozyme genetic differentiation among three hatchery strains (A, B and C) of white shrimp, Penaeus (Litopenaeus) vannamei, commonly used in shrimp farming in northwest Mexico. A wild population from northern Sinaloa was used as a reference. Fifteen loci were detected, nine of which were polymorphic (ACP-1*, ACP-2*, AKP-2*, EST-2*, EST-3*, EST-4*, EST-5*, LAP*, and LDH*). Polymorphism of A, B and C were 53, 53, and 40%. The mean observed heterozygosity per locus was 0.071, 0.093, and 0.050 without any significant difference among them or with respect to the wild population (0.056). In all samples observed heteroxygosity was smaller than expected. The mean number of alleles per locus was 1.8 in A and 1.87 in both B and C. Strain A was the only sample without rare alleles. Only EST-3* and LAP* of strain A were in Hardy-Weinberg equilibrium; the other loci were in disequilibrium in all samples. High inbreeding values and heterozygote deficiencies were detected in all samples. Distribution of allelic frequencies was heterogeneous among the samples (G-test), involving all polymorphic loci and suggesting a genetic differentiation. According to Fst, a moderate genetic differentiation (7.4%) was detected among the samples. Greater differences were between strains A and C. Based on genetic distance, the samples were grouped into two pairs, B-C and A-wild. Strain A is a young strain related to the wild sample, whereas strains B and C have a different geographic origin than the wild sample.  相似文献

对虾WSSV病是亚洲对虾养殖业中的一个棘手问题。本研究采用Kimura引物 ,用PCR技术对不同生长期的中国对虾 (Penaeuschinensis)进行了WSSV的检测 ,同时也检测了对虾发病时养殖池中多见的野生厚蟹 (Helicesp .)和矛尾刺虎鱼 (Acanthogobiushasta)。检测结果表明 :分别在检测的 5尾亲虾中的 1尾 ,6尾仔虾中的 1尾 ,5尾稚虾中的 3尾及所检测的 5尾病虾和 2只厚蟹中获得到 982bp的PCR扩增产物 ,说明为WSSV感染阳性。在检测的 2尾矛尾刺虎鱼中均未获得PCR扩增产物 ,说明为WSSV感染阴性。在亲虾、虾苗以及虾池内的野生厚蟹中检测到WSSV感染的阳性结果表明 :WSSV感染的亲虾有可能是病毒的储主 ,WSSV感染的野生厚蟹有可能是病毒中间宿主或病毒的携带者 ,它们在对虾WSSV病的感染、传播中起了重要的作用  相似文献

White spot syndrome virus (WSSV) is one of the most important pathogens of penaeid shrimp. It is widely distributed in most Asian countries where penaeid shrimp are cultured, as well as in the Gulf of Mexico and SE USA. The virulence of six geographic isolates of WSSV was compared using Litopenaeus vannamei postlarvae and Farfantepenaeus duorarum juveniles. The six geographic isolates of WSSV originated from China, India, Thailand, Texas, South Carolina, as well as from crayfish maintained at the USA National Zoo. For challenge studies, virus infected tissues were given per os to L. vannamei postlarvae and Fa. duorarum juveniles. Resultant WSSV infections were confirmed by histological examination. The cumulative mortality of L. vannamei postlarvae reached 100% after challenge with each of the six geographic isolates of WSSV. However, the Texas isolate caused mortalities more rapidly than did the other shrimp isolates; the crayfish WSSV isolate was the slowest. In marked contrast, cumulative mortalities of juvenile Fa. duorarum reached only 35–60%, and varied among the geographic isolates of WSSV. Interestingly, in Fa. duorarum, the Texas WSSV isolate was also the most virulent, while the crayfish WSSV was the least virulent. The findings suggest that slight differences in virulence exist among geographic isolates of WSSV, and that susceptibility may vary with species and lifestages of the host.  相似文献

In recent years, the shrimp industry has turned to inland freshwater culture as one method to avoid problems such as the introduction of possible vectors of viral pathogens into seawater ponds. Our experiments evaluated susceptibility to white spot syndrome virus (WSSV) in Litopenaeus vannamei held under different salinity regimens. Juvenile L. vannamei that were conditioned at salinities of 35, 25, 15, 5 and 2 g L−1 were challenged with WSSV. In order to assess the severity of white spot disease, histological analysis and nested polymerase chain reaction (PCR) tests were carried out on the challenged shrimp every 4 h after 48 h post challenge. The results indicated that significantly more severe infections resulted at 15‰ than at other salinities. Mortality could not be compared due to the sampling design and because severe WSSV infections occurred in all test groups such that few shrimp remained alive in each challenged group at the end of the test. Despite this, the results suggest that salinity may affect the course and outcome of WSSV infections.  相似文献

从感染白斑综合症病毒(Whitespotsyndromevirus,WSSV)的对虾中提取和纯化WSSV,对其蛋白酶活力进行分析。结果表明,WSSV蛋白酶具广泛的pH稳定性,当pH达7.5时,蛋白酶活性最大;pH高于10 0时,酶活力很低,其蛋白酶偏碱性。丝氨酸蛋白酶抑制剂苯甲基磺酰氟(PMSF)对酶活性有抑制作用。Ca2 、Mn2 、Fe2 和Cu2 可降低WSSV蛋白酶活性,但Mg2 有轻微的激活作用。胰蛋白酶抑制剂在浓度为12.5~25.0mg/L时,对WSSV蛋白酶活性无影响。Leupeptin使蛋白酶活性降低12.29%。Chymostatin在质量浓度为12.5~25.0mg/L时,对WSSV蛋白酶活性有强烈的抑制作用,表明WSSV蛋白酶类属胰凝乳蛋白酶。蛋白质修饰剂对WSSV蛋白酶活性影响的研究结果表明,组氨酸残基为WSSV蛋白酶活性基团,而巯基为非必需基团,说明WSSV蛋白酶为非巯基依赖型的蛋白酶。  相似文献

Three hybridoma clones secreting monoclonal antibodies (MAbs) were produced from mouse myeloma and spleen cells immunized with white spot syndrome virus (WSSV) isolated and purified from Penaeus monodon (Fabricius), collected from north-eastern Taiwan. By sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS–PAGE), the protein profile of this isolate contained four major proteins with sizes of approximately 35 (VP35), 28 (VP28), 24 (VP24), and 19 kDa (VP19). Western blot analysis revealed that two MAbs (1D7 and 6E1) recognized epitopes on VP28 and one MAb (3E8) recognized an epitope on VP19. The MAb 6E1 isotyped to the IgG1 class was used in both an indirect immunofluorescence assay (IFA) and in an immunochemical staining protocol for successful identification and localization of WSSV in infected shrimp tissues. Antigenic similarity of isolates from Indonesia and Malaysia to the Taiwan isolate was illustrated by IFA with MAb 6E1. A MAb (2F6) which bound specifically to two shrimp proteins, 75 and 72 kDa, and reacted to the healthy and non-target tissues of WSSV in infected shrimp, such as hepatopancreas, is also described here and shows the necessity for specific identification of antibodies.  相似文献

In the field, moulting and salinity drop in the water due to excessive rainfall have been mentioned to be risk factors for WSSV outbreaks. Therefore, in this study, the effect of an acute change in environmental salinity and shedding of the old cuticle shell on the susceptibility of Penaeus vannamei to WSSV was evaluated by immersion challenge. For testing the effect of abrupt salinity stress, early premoult shrimp that were acclimated to 35 g L?1 were subjected to salinities of 50 g L?1, 35 g L?1, 20 g L?1, 10 g L?1 and 7 g L?1 or 5 g L?1 and simultaneously exposed to 105.5 SID50 mL?1 of WSSV for 5 h, after which the salinity was brought back to 35 g L?1. Shrimp that were transferred from 35 g L?1 to 50 g L?1, 35 g L?1 and 20 g L?1 did not become infected with WSSV. Shrimp became infected with WSSV after an acute salinity drop from 35 g L?1 to 10 g L?1 and lower. The mortality in shrimp, subjected to a salinity change to 10 g L?1, 7 g L?1 and 5 g L?1, was 6.7%, 46.7% and 53.3%, respectively (P < 0.05). For testing the effect of moulting, shrimp in early premoult, moulting and post‐moult were immersed in sea water containing 105.5 SID50 mL?1 of WSSV. The resulting mortality due to WSSV infection in shrimp inoculated during early premoult (0%), ecdysis (53.3%) and post‐moult (26.72%) demonstrated that a significant difference exists in susceptibility of shrimp during the short moulting process (P < 0.05). The findings of this study indicate that during a drop in environmental salinity lower than 10 g L?1 and ecdysis, shrimp are at risk for a WSSV infection. These findings have important implications for WSSV control measures.  相似文献

2002-2007年在人工感染白斑综合症病毒(white spot syndrome virus, WSSV)的基础上进行一代个体选育(G1)后,对凡纳滨对虾连续进行了4代家系选育,共建立120个抗WSSV家系,感染实验结果表明:G2~G5选育家系对虾平均成活率分别为5.57%±9.83%, 8.66%±11.52%, 9.52%±8.84% 和 13.79%±12.86%;G2~ G5选育家系对虾平均成活率的变异系数分别为1.77、1.40、0.97和0.87。根据每个家系对虾的成活情况可分为敏感、中等抗性和高抗性家系,G2至G5敏感家系在各代选育家系中的比例逐年下降,分别占76.5%、55.2%、51.4%和33.3%,抗病成活率分别为0.44%±1.09%、0.78%±1.70%、2.27%±2.76%和2.44%±3.09%,感染WSSV后2~3 d出现1个急性死亡高峰;中等抗病家系在各代选育家系中的比例逐年上升,分别占0、20.7%、31.1%和38.5%,抗病成活率分别为0、9.08%±1.46%、10.7%±1.41%和11.36%±3.30%,感染WSSV后出现2个死亡高峰,第1死亡高峰值大于第2高峰;高抗家系在各代选育家系中的比例逐年上升(G4除外),分别占23.5%、24.1%、17.1%和28.2%,抗病成活率分别为22.23%±5.21%、22.7%±12.30%、24.45%±6.56%和28.98%±8.09%,感染WSSV后出现2个死亡高峰,第1死亡高峰至小于第2高峰。经连续的定向选育,选育对虾抗病性状一代比一代强,表现出明显的抗病性能,特别是高抗对虾不仅死亡率低且其死亡高峰推迟2~3d,延缓了对虾WSSV暴发的时间,但是每代每尾对虾平均产卵量逐年下降(P<0.05)。  相似文献

White spot syndrome virus (WSSV) is a major threat for farmed shrimp worldwide. RNA interference (RNAi) is the most recent tool against viral diseases. Rab7 silencing effectively inhibited virus infections in juvenile shrimp, but the antiviral effect in brooders remains unknown. This study found a homologue Penaeus monodon Rab7 gene in Litopenaeus vannamei brooders from Mexico. Sequence identity was >99% to a Thai LvRab7 sequence and >94% to Rab7 sequences from P. monodon or Marsupenaeus japonicus. Animals treated with a partial (494 bp) or a complete (618 bp) LvRab7 dsRNA sequences and challenged 48 h post treatment (hpt) with a high WSSV dose showed 80–88% mortality respectively. Shrimp treated with 4 or 20 μg LvRab7 dsRNA and challenged with a WSSV high dose had 80% mortality each, but it was reduced to 33% and 40%, respectively, with a low dose. Efficacy of dsRNA to reduce shrimp mortality was dependent on virus dose used regardless of dsRNA concentration. A significant reduction in LvRab7 mRNA levels was observed at 120 hpt. In conclusion, silencing LvRab7 in brooders showed a mild antiviral effect against a WSSV challenge at 48 hpt.  相似文献

The simultaneous presence of monodon baculovirus (MBV) and white spot syndrome virus (WSSV) in apparently healthy postlarvae of Penaeus monodon from different hatcheries in India was studied by nested polymerase chain reaction (PCR). MBV could be detected in 54% of the samples. However, only 15% of samples were positive by non-nested reaction. WSSV could be detected in 75% of samples, 19% being positive by non-nested reaction. The results show simultaneous presence of WSSV and MBV in many samples at various degrees of infection. Only 14% of the samples analysed were negative for both viruses.  相似文献

为研究新型抗菌物质凹凸棒土对凡纳滨对虾生长性能、消化能力、抗氧化能力和肠道菌群的影响。实验选取初始体质量为(0.17±0.01)g的幼虾,分别投喂0% (对照)、0.1% (S1)、0.2% (S2)、0.4% (S3)、0.8% (S4)含量的凹凸棒土饲料,以及含有0.2%恩诺沙星抗生素对比组饲料(E),养殖8周。凹凸棒土处理组对虾增重率均未见显著提升,但S1、S2组对虾存活率显著高于对照组,与E组没有显著差异。相比于对照组,凹凸棒土添加组提高了幼虾肠道淀粉酶活性,以及肝胰腺中脂肪酶活性。S2组对虾血清酸性磷酸酶和碱性磷酸酶活性显著高于对照组和E组。S3和S4组对虾的肝胰腺超氧化物歧化酶活性显著高于其他组。肠道菌群α多样性分析显示,S2组肠道微生物丰富度Chao1指数有所下降,但是覆盖度显著提高。与对照组相比,S2组部分属的丰度调节作用与恩诺沙星类似。根据代谢通路差异分析与种间关系作图,与E组相比,凹凸棒土能够上调牛磺酸、谷胱甘肽、半乳糖和脂肪酸代谢的4条通路,并增加肠道微生物种间合作能力。在饲料中添加0.1%~0.2%的凹凸棒土能够显著提高凡纳滨对虾的成活率、消化能力和肝胰腺抗氧化能力,并在调节对虾肠道部分菌群丰度方面起到类似于恩诺沙星的作用,能改善肠道菌群结构和相应功能。  相似文献

在已有对虾白斑综合症病毒(WSSV)蛋白酶活性研究的基础上,从分子水平上确定WSSV呈蛋白酶活性的肽段。采用凝胶电泳技术的蛋白酶酶谱分析法确定WSSV蛋白酶活性,研究非变性条件下WSSV多肽裂解的条件,确定低温(20℃)裂解WSSV多肽可满足实验的要求。同时使用SDS-PAGE分离胶切割法结合蛋白洗脱分离出Vp19、Vp22、Vp24、Vp26和Vp28共计5个肽段,并对分离的5个组分进行蛋白酶活性的检测,初步结果表明,Vp19具有蛋白酶催化活性。  相似文献

The effect of plants and probiotics on the survival and immune response of Litopenaeus vannamei challenged with the white spot syndrome virus (WSSV) was evaluated. A probiotic mixture (PM), plant extract (PE) or powdered plants (PP) were added to feed with the attractant Dry Oil®. An experiment was conducted with five treatments in triplicate. Shrimp (weighing 11.70±2.5 g) were cultured in 120 L plastic tanks and fed twice a day with commercial feed plus additives or with commercial feed plus WSSV. Animals were monitored for the occurrence of WSSV using single‐step and nested PCR. The PM and PP added to the commercial feed showed high survival, a decrease in WSSV prevalence in shrimp and an increase in the activity of lysosomal enzymes, N‐acetyl‐β‐glucosaminidase and acid phosphatase. The total haemocyte count in shrimp treated with PM was significantly higher than that in the control group (treatment I) and in shrimp fed with PE. The results of the present work indicate that PP and PM are good candidates for use as feed additives against WSSV in shrimp cultures.  相似文献

白斑综合征病毒WSSV(White Spot Syndrome Virus,WSSV)是引起对虾大规模死亡的主要病毒性病原,且该病毒感染的宿主广泛。自WSSV基因组全序列公布以来,国内外的学者对WSSV的研究热点转移到该病毒基因产物功能的相关研究上面来。弄清WSSV感染机理是有效防治病毒的基础,而对其感染机理的阐释则主要是搞清病毒各蛋白在感染中的作用,这些是WSSV蛋白质组学的研究内容之一。分析WSSV蛋白同时可以在分子水平上为研究海洋和陆地生物病毒的进化趋势提供有力的基础。  相似文献

Cell membranes from the rotifer, Brachionus urceus, were obtained by centrifugation and found to specifically bind white spot syndrome virus (WSSV) in vitro. This finding suggests that there is likely a WSSV receptor on the rotifer cell membrane and provides evidence that rotifers may be a host for WSSV.  相似文献

Infectious hypodermal and haematopoietic necrosis virus (IHHNV) and white spot syndrome virus (WSSV) are two widespread shrimp viruses. The interference of IHHNV on WSSV was the first reported case of viral interference that involved crustacean viruses and has been subsequently confirmed. However, the mechanisms underlying the induction of WSSV resistance through IHHNV infection are practically unknown. In this study, the interference mechanisms between IHHNV and WSSV were studied using a competitive ELISA. The binding of WSSV and IHHNV to cellular membrane of Litopenaeus vannamei was examined. The results suggested that there existed a mutual competition between IHHNV and WSSV for binding to receptors present on cellular membrane of L. vannamei and that the inhibitory effects of WSSV towards IHHNV were more distinct than those of IHHNV towards WSSV.  相似文献

将VP110基因的部分序列克隆到pET-28a载体中构建pET28a-vp110b重组质粒并进行原核表达,获得重组表达的蛋白rVP110-B;用rVP110-B注射凡纳滨对虾Litopenaeus vannamei后,经WSSV感染,实验表明,该蛋白注射使凡纳滨对虾感染WSSV的半数死亡时间比对照组延长了20%。用表达纯化的该重组蛋白制备了兔抗rVP110-B多克隆抗体,该抗体用于凡纳滨对虾鳃细胞膜蛋白与rVP110-B的Far-western分析显示,凡纳滨对虾鳃细胞膜蛋白中除90 kDa左右的血蓝蛋白外,在41.7 kDa存在结合条带,经质谱分析表明这条鳃细胞膜蛋白是肌动蛋白。  相似文献
