The germination of freshly collected Juniperus polycarpos (K. Koch) seeds is very low and attributed to the large proportion of nonviable seeds in the seed lots. Thus, the aim of this study was to improve seed lot quality by removing nonviable seeds using two flotation techniques: incubation-drying-separation (IDS) and modified specific gravity (MSG) separation. We examined different IDS conditions (the specific incubation time, subsequent drying time and sorting media) for effectively sorting out nonviable seeds; and tested the feasibility of modified SG separation, which involved soaking seeds in water for a certain period before sorting in different concentrations of sucrose solution. Viable seeds were expected to absorb and metabolically bind more water during soaking than dead seeds could, and hence be sorted effectively depending on the viscosity of the sucrose solution. The viability of the seeds that floated or sank was determined in a topographical tetrazolium chloride (TTC) test. For the IDS trial, 7 days incubation followed by 9 h of drying and sedimentation in pure water or 200 g·L?1 sucrose solution identified, respectively, 75 and 82 % of the seeds as viable (sunken) seeds. For the MSG trail, 77 % viable seeds were recovered in the sunken fraction when seeds were soaked for 48 h then immediately sorted in 600 g·L?1 sucrose solution. In both cases, the loss of viable seeds in the discarded floating fraction was only 4 %. The results demonstrate that both IDS and MSG separation techniques substantially improved seed lot viability, but MSG separation is simple and needs no modern seed handling facility, and its efficacy relies on seed mass (due to the initial soaking) and the specific density and viscosity of the flotation medium.  相似文献

If low absolute pressure is first applied to and then released from seeds of Pinus sylvestris L. lying in water, the seeds with mechanical damage, i.e. with seed coat cracked, with part of seed coat missing or with part of seed coat and gametophyte missing, quickly lose their buoyancy and sink and can thus be separated from the floating undamaged seeds (PREVAC method). The most crucial of the tested physical properties was the level of pressure. The time of treatment (1–20 min) was of less importance, and the water temperature (5–40°C) was unimportant. Treatment at approximately 97 kPa below atmospheric pressure (3–5 kPa absolute pressure) for 1 up to 20 min resulted in removal of almost all mechanically damaged seeds in the four studied seed lots (containing up to 26% damaged seeds). In total, less than 5% undamaged seeds of the whole seed lots were removed. The germination percentage of the fractions remaining after separation increased with increasing number of damaged seeds in the seed lots and vice versa for the removed fractions.  相似文献

To study the effects of seed soaking on seedling emergence and development in Norway spruce [Picea abies (L.) Karst.] two commercial seed lots were soaked for 15 h in aerated water. After soaking, floating and sunken seeds were separated and analyzed prior to being sown in peat-filled containers. Seedling development was followed for one growing season. All sunken seeds were viable according to radiography whereas the floating seeds contained a considerable number of larvae filled and empty seeds. Mean time of emergence of the sunken seeds was approximately 1.5 days shorter than control (i.e. dry) seeds and floating seeds. Soaking had no effect on the time taken for most germinants to emerge in both seed lots, thus failing to synchronize seedling emergence. The later a seedling would emerge, the more likely it was for it to die during the growing season or to be culled at the end of the growing season. Soaking had an inconsistent effect on stem diameter at the end of the growing season and no effect on stem or root biomass. Seedling size uniformity was equal in all treatments. This study suggests that soaking and seed sorting does not guarantee a more uniform seedling crop, but this technique may be useful in upgrading poor quality seed lots of Norway spruce containing a high number of empty or larva infested seeds.  相似文献

The IDS (Incubation, Drying and Separation) method was tested on Schinus molle L. seeds collected from Ethiopia in an attempt to sort non-viable from viable seeds. Seeds were incubated for 24 h followed by one, three, four, five, six, nine and twelve hours of drying and then separated in water. All IDS treatments gave 80.5–93.5% germination in the sunken fraction compared with 61% for untreated seeds. The mean germination time of sunken seeds reduced from 11.2 (control) to 8.9–9.2 days (IDS-treated). Specific gravity separation in water of untreated seeds was also tested, and the germination capacity of sunken fraction was 83.5% with a mean germination time of 9.3 days. IDS method can, therefore, be used to enhance both percentage and rate of germination of a seed lot of Schinus molle, which is desirable to produce uniform and vigorous seedlings in the nursery or out in the field.  相似文献

We report results of preliminary evaluations of marked, biased, filter (MBF), a method for both estimating and improving the effectiveness of separating conifer seeds. MBF integrates elements of X-radiographic seed quality assessment and mark-recapture methods from wildlife population sampling. Given a sample drawn from a population, individual seeds with attributes targeted for separation are labelled with a mark which biases the subsequent removal probability of other, non-marked seeds in the lot during grading and processing, after returning the sample to the population. Bias in the marking filter, in the form of extra mass or density added to seeds, minimizes the likelihood that uncertainty due to sampling error in the prior estimate will result in empty seeds remaining in the population during processing. In two experiments, a ponderosa pine seed lot was partitioned into empty and filled fractions with an air column separator. Powder dye talc was used for marking in the first trial, with low bias (<1%) and a small sample size (n = 150) with dry (2% moisture content) and soaked (26% moisture content) seed lots. A second trial conducted with two coats of spray paint used for marking and a larger sample size (n = 400) resulted in removal of 100% of non-filled seeds in the population. A simulation was conducted to evaluate sample statistics upon which to base the value of threshold bias, τ, below which seeds in an X-rayed sample should be marked to achieve consistent separation. Estimating the marking threshold based on quantiles of filled seeds most consistently resulted in removal of the non-filled fraction. MBF is a useful method for statistically linking one or more individual seed quality attributes extracted from X-ray image analysis with population-level processing of seed lots.  相似文献

Juniperus procera Endl. is economically important timber species, but its populations are extremely small and fragmented in its natural habitat, thus, calling for immediate ex situ conservation. Here we examined the effects of seed sources and storage temperature on the longevity of Juniperus procera seed lots through collection and preservation of seeds in seed banks. Seeds were collected from nine sites across the species natural distribution in Ethiopia and stored in four warehouses: modern cold room (5℃), mud house (15℃), concrete block house (17℃ or corrugated iron house (20℃) for 42 months. Every three months, a random sample of stored seeds were drawn and tested for germination. A highly significant variation (p < 0.01) in germination of stored seeds was observed among different storage environments, seed lots, and duration of storage. Over the storage period, seeds stored in the cold room had the highest mean percentage germination, followed by the mud house, corrugated house and blocket house. The cold room (41%) and the mud house (38%) maintained the same level of germination as the intitial germination of the seedlotds (42%). The variation in longevity of stored seeds was significnatly correlated with the initial germination of seed lots (r > 0.80; p < 0.01). Cold storage also resulted in enhancement of germination through its stratification effect that terminated the non-deepphysiological dormancy of juniper seeds. In conclusion, seed lots with good initial germination can be effectively stored in cold room (5℃) up to four years. In the absence of modern cold stores, mud houses can be used as a good alternative to store seeds at local level.  相似文献

Abstract

The effect of soaking on germination and occurrence of fungal infections on Norway spruce (Picea abies (L.) Karst.) seeds and germinants was studied in three commercial seed lots. Treatments in which seeds were soaked in water for 24 h or in which the water was changed during the soak did not have an effect on the species diversity or on the abundance of the fungi isolated from the seeds. Different fungi were found in different seed lots. Most of those isolated are saprophytic or weakly pathogenic, but pathogens such as Sirococcus conigenus (D.C.) P.F. Cannon &; Minter and Trichothecium roseum (Pers.) Link were also isolated. Soaking increased germination energy but had no effect on final germination percentage. The number of mouldy seeds and germinants with disease symptoms was different among seed lots when seeds were germinated on water agar. When germinated in low humified Sphagnum peat, no differences in the emergence of disease symptoms were observed among seed lots. The frequency of disease was lower in peat than in water agar. Soaking had no effect on disease emergence in germination trials on either water agar or in Sphagnum peat.  相似文献

Seeds of Juniperus procera collected from five provenances across its geographic range in Ethiopia were subjected to cold-moist stratification at 5°C or 10°C for 6–12 weeks. The effect of aqueous smoke solution in overcoming the light requirement for germination, and the potential of visible (VIS) and near infrared reflectance (NIR) spectroscopy for sorting sound and insect-damaged seeds were also investigated. Highly significant differences in germination were detected among provenances (P < 0.0001) and stratification periods (P < 0.0001), but not between temperature regimes (P=0.111). Seeds from the south and southeast distribution ranges had higher percentage germination after 6 weeks of stratification than seeds collected from north, northwest and central ranges of distribution. The smoke treatment did not affect germination regardless of whether the seeds were exposed to light. Exposure to light increased germination capacity three fold. Sound and damaged seeds were distinguished with 90% accuracy using VIS + NIR spectroscopy. It can be concluded that dormancy in juniper seeds varies with provenances, and cold stratification for 6 weeks alleviates dormancy in some seed lots. Tentatively, smoke treatment seems ineffective in overcoming photo-dormancy in juniper seeds. VIS + NIR spectroscopy has demonstrated a great potential for sorting damaged seeds, thereby upgrading seed lot purity.  相似文献

Acer velutinum Boiss is a valuable tree species native to Iran, and its seeds possess physiological dormancy that hampers seedling production in the nursery for large-scale reforestation efforts. The aim of this study was to determine the optimal dormancy breaking treatments for A. velutinum seeds. We conducted a factorial experiment involving six seed lots collected along an elevation gradient from 300 to 1800 m at 300 m interval and four cold-moist stratification periods (0, 4, 8 and 16 weeks) at 4℃ and 70% relative humidity. The result shows that the germination of cold-moist stratified seeds was significantly (p < 0.0001) higher than the control for all seed lots. The highest germination capacity was recorded after 16 weeks of cold-moist stratification for all seed lots (68% 88% depending on the seed lot) except those collected from mid altitude sites (600 and 900 m) that germinated equally well (≥ 75%) after 4and 8-week of clod-moist stratification compared to the other seed lots. The mean germination time was significantly shorter (12 to 19 days, depending on the seed lot) for seeds stratified for 16 weeks than for untreated seeds. It can be concluded that: (1) cold-moist stratification for 16 weeks is the best pre-sowing treatment for breaking dormancy in A. velutinum seeds; and (2) seeds should be collected from mid altitude sites (600 and 900 m) to get more than 80% germination within 15 days, and these seed lots even required shorter cold-moist stratification period(eight weeks) than other seed lots.  相似文献

Douglas-fir seedlots were treated with the Incubation Drying Separation (IDS) method to test whether seeds infested with the seed chalcid, Megastigmus spermotrophus Wachtl, could be separated from non-infested seeds. Seeds were soaked in distilled water for 24 h, drained, placed in plastic bags and incubated at 15 °C for 3 days. The seeds were dried for either 0.5, 1, or 2 h at 25 °C, and then separated into floaters and sinkers in a water column. An average of 97% of the infested seed floated. The drying period did not affect the separation of infested seeds but significantly fewer sound seeds floated in the seedlots dried for 0.5 h than those dried for 1 h. Germination capacity of IDS-treated seeds did not differ from that of untreated seeds but the germination rate was significantly faster for IDS-treated seeds when all lots were stratified.  相似文献

Prunus cerasoides has poor regeneration in its natural habitat due to its hard seed coat. The information about maturity time of fruit/seed is scanty and studies on seed germination enhancement lacking. In the present study, the main focus has been given to the physical fruits/seeds attributes which are reliable indicators of maturity and seed pretreatments which help to enhance germination. The fruits/seeds were collected from low and high elevational sites covering the altitudinal range of the species during the period of fruit/seed development (last week of February–mid April) for two consecutive years (2003–2004). The change in fruit colour from dark green to pale red or red was a useful indicator of seed maturity in P. cerasoides. Fruit moisture content between 46.57 ± 0.45% and 56.81 ± 1.14% and seed moisture content between 29.8 ± 1.35% and 34.13 ± 1.50% coincided with maximum germination (41.11 ± 13.96% and 59.99 ± 12.05%) across sites. The removal of seed coat (mechanical treatment) enhanced the germination of seeds to 70 ± 0% at low elevation and 100 ± 0% at high elevation in comparison of control or untreated seeds (germination range between 5.83 ± 0.83% and 31.68 ± 25.02%). Predispersal seed predation among individual Acacia macrostachya trees over two sites in Burkina Faso was assessed. In addition, the effects of seed predation on seed viability; germination responses to scarification (mechanical, hot water at 60, 70 and 80°C, and sulphuric acid for 10, 20 and 30 min) and dry heat (at 60, 70 and 80°C for 15, 30 and 60 min each) treatments were investigated under laboratory conditions. The results indicated a large difference in predation intensity among individual trees, as well as between sites. Predispersal predation significantly decreased seed germination, particularly when the number of insect larvae per seed increased. Scarified and unscarified seeds germinated equally well, except for hot water treatments of high temperature. This indicates lack of physical dormancy and dispersed seeds can readily germinate provided that conditions for germination are conducive. Seeds exposed to dry heat treatments also germinated close to 97% under low intensity and short exposure times. This suggests that fire, under natural condition, plays a key role in triggering germination of A. macrostachya seeds.  相似文献

Ponderosa pine (Pinus ponderosa Dogl. ex. Laws.) is the most planted conifer species in the forest-steppe ecotone of Patagonia, Argentina, because of its adaptability and excellent growth rates. In spite of this, and the increasing demand for this species, local commercial seed lots showed low quality, making hazardous seedling production. Aiming to upgrade germinability of a local seed lot, we set an experiment to determine the duration of the prechilling period (0, 10, 20, 30, 40 and 60 days) that promoted the highest seed germination speed (GE) and percentage (GP). Moreover, part of that lot was IDS treated, in an attempt to separate empty and dead filled seeds from viable seeds. Results showed that after 40 days prechilling, GE reached 62%, and GP 70%, both higher than the values obtained under customary conditions. The application of the IDS technique, after 40 days prechilling, 8 h drying at ambient conditions (16 ± 2°C and 50% HR), and 25% seed moisture content (mc), increased GE and GP to 68% and 89%, respectively. Optimal prechilling periods and the application of the IDS technique successfully improved germinability of ponderosa pine seeds from Patagonian stands.  相似文献

South American Polylepis mountain forests are recognised as being one of the most endangered forest ecosystems in the world. Reforestation measures have been strongly recommended but may be hampered due to the very low seed germination reported for several Polylepis species. In order to facilitate reforestation we analysed the influence of seed mass on germination probability for Polylepis australis seeds in the Córdoba mountains (central Argentina). We collected seeds from 43 trees distributed throughout five woodland fragments located within two regions differing in size, topographical position, and altitude (1,900 m a.s.l. and 2,200 m a.s.l.). Seeds of Polylepis australis exhibited a great variation in terms of mass and percent seed germination among individual trees and among geographical regions. The results of logistic regression showed that germination probability was highly correlated with seed mass. However, the explained deviance significantly increased by including the region, the woodland fragment and especially the individual tree in addition to seed mass in the regression models. We conclude that selecting seeds on the basis of mass is an appropriate way to enhance germination prospects for reforestation projects. However, no absolute mass values are applicable in this context as the highest germination probabilities were reached at varying seed mass values depending on geographical region, woodland fragment or individual tree. We suggest collecting the relatively heaviest available seeds, even though the absolute seed mass may be low.  相似文献

We investigated the effects of litter accumulation by an alien tree, Casuarina equisetifolia, on the germination and seedling establishment of Rhaphiolepis wrightiana, a shrub native to the Ogasawara (Bonin) Islands, in the northwestern Pacific Ocean, in a field experiment. We compared the emergence of seedlings in forests dominated by C. equisetifolia with that in native forests, with and without litter removal. More than 75% of seedlings emerged during a year except at the C. equisetifolia sites with litter removal (approximately 45%). Thus, seeds of R. wrightiana can germinate under C. equisetifolia if they are dispersed on the ground.  相似文献

Abstract

The response of common alder [Alnus glutinosa (L.) Gaertn.] and downy birch (Betula pubescens Ehrh.) seeds to germination temperature was examined following chilling and priming. Seeds of two seed lots of each species were subjected to combinations of chilling (4±1°C) and priming (20°C) treatments in fully imbibed (FI) state or a lower target seed moisture content (TMC) level (30% and 35% in alder and birch, respectively). After treatment, the seeds were allowed to germinate for 56 days at constant temperatures of 7.5, 10, 15, 20, 25 or 30°C. The response to temperature and pretreatment differed between species, but the effect was consistent in each seed lot within each species. In alder, the TMC seeds germinated well across the full range of temperatures, whereas there was an optimum temperature (22–23°C) for seeds given the FI pretreatment. Priming had no significant effect on the germination response of the TMC seeds in alder, but priming greatly improved germination in the FI seeds, especially at the lower germination temperatures (optimum 18–19°C). In contrast, in birch, the TMC seeds germinated better across the full temperature range, but the optimal germination temperature (15°C) was the same for all seed pretreatments. Priming improved germination in both the FI and TMC seeds in birch.  相似文献

以南美油藤种子为对象,研究不同品种、气温、播种基质对其发芽情况的影响。结果显示:细沙以及珍珠岩+泥炭两种基质不利于南美油藤种子发芽,轻型基质(黄心土:稻壳:珍珠岩=3:4:3)可使其发芽率超过90%;不同品种的南美油藤种子发芽率差异显著,C和H两个大果种子不发芽,V2和V4发芽率低于15%,V3发芽率可达90%;不同气温影响南美油藤种子发芽率,以气温25~35℃播种为宜。  相似文献
