Earthworms are being used as bio-indicators to assess terrestrial pollution. However, it is often not known whether their populations possess a uniform genetic structure, which would allow comparison of residues or biological properties of earthworms from different sampling locations. In order to investigate this point, random amplified polymorphic DNA (RAPD) variation was surveyed in earthworms (Lumbricus terrestris) from five different sampling sites in Germany. Forty oligonucleotide RAPD primers (10 base pairs in length) were screened, three of which produced high polymorphic band patterns. A total of 61 DNA fragments were detected in 90 individuals of L. terrestris from five sampling sites with 49 (80.3%) RAPD markers being polymorphic. The genetic similarities within (band sharing rates between 0.756 and 0.795) and among the L. terrestris populations (0.635) were similar even at widely separate locations. Inter-population variation in the RAPD pattern for all five earthworm populations accounted for 37.9% of the total variation, while intra-population variation for three adjacent Saarland populations accounted for only 18.0% of the total variation. Principal component analysis (PCA) and the genetic distances of the populations confirm these results. Twenty-four percent of the genetic distance is caused by geographical isolation as shown by a test for isolation by distance. These results show that L. terrestris fulfils the genetic qualifications for a bio-indicator particularly at closely located sampling sites. However, the results also suggest that earthworm studies of widely separated locations should include genetic characterisation of the earthworm samples. 相似文献

The basic ecology of well-researched earthworm species, such as Lumbricus terrestris, remains relatively unknown. In order to contribute to the limited body of knowledge, the present work aimed to investigate if specific behaviours could be detected between an adult and the cocoons it produced by comparison with a non-parental adult and cocoons. Modified Evans’ boxes (0.8×0.2×0.008 m) with recently mated adults allowed observation of their habits under what was essentially a 2-D system. Experimental manipulation (at 81 days) involved four treatments: (1) L. terrestris removal (LtRm); (2) L. terrestris replacement (LtRp), where individuals were replaced by others; (3) control, with no manipulation and (4) L. terrestris removal control (CLtRm) where individuals were removed and then reintroduced in the same box, to account for any effects of the removal method. Burrowing patterns of adults and later, hatchlings, were recorded throughout the duration of the experiment (191 days). Cocoon production across treatments did not differ significantly (p>0.05) and averaged 26±6.4 cocoons worm−1 over a 26 week period. In the LtRm treatment mean cocoon production was 12.6±2.7 cocoons worm−1 over 11 weeks. Forty one percent of cocoons were deposited in the upper 0.05 m of the soil profile. Cocoons below that depth were found deposited in side burrows (1–4 cm in length) and were coated with earthworm casts. Hatchling number and survival rate did not differ significantly across treatments (p>0.05) with means±s.d. of 2±2.2 and 62±29%, respectively. Hatchling mass in the LtRm treatment (0.4±0.38 g), was found to be significantly greater (p<0.01) than that of all other treatments (0.12±0.12 g). In this experimental set up, the presence of parental earthworms and other con-specifics had no significant effect on cocoon viability and hatchling survival, however, results indicated a negative effect of adult presence on hatchling growth. 相似文献

A laboratory experiment was carried out to test the hypothesis that the earthworms Lumbricus terrestris and Aporrectodea caliginosa are able to maintain their populations and reproduce in the acid forest soil of a deciduous forest where no lumbricids were found in the field. The experiment was conducted in 45-l containers in which layers of mineral subsoil, humus and organic topsoil collected from the site were established. Both species survived and at least L. terrestris reproduced during the 60 weeks’ incubation. Burrows and middens of L. terrestris were recorded and quantities of litter were consumed. The presence of lumbricids increased the organic matter content of humus, reduced the acidity of the topsoil and humus layers, and suppressed the population of the enchytraeid Cognettia sphagnetorum. A dense population of Enchytraeus albidus was found in L. terrestris middens. It is concluded that edaphic factors do not explain the absence of earthworms, but isolation from cultural landscapes and lack of opportunity to colonize the site from the surroundings is the decisive factor. 相似文献

A field experiment was conducted to elucidate ecosystem services provided by earthworms on the repression of phytopathogenic and toxinogenic fungi. The study focussed on decomposing Fusarium culmorum-infected and deoxynivalenol (DON)-contaminated wheat straw remaining on the soil surface as part in conservation tillage. Mesocosms were established in the topsoil of a winter wheat field located in Northern Germany, where conservation tillage has been practised for 20 years. Besides a non-earthworm treatment, two earthworm species were inoculated in the mesocosms either separately or combined: Lumbricus terrestris (anecic, detritivorous) and Aporrectodea caliginosa (endogeic, geophagous). The earthworms were exposed either to artificially Fusarium-infected wheat straw highly contaminated with DON or to non-infected straw serving as a control. The experiment was conducted during an eight week period after harvest from mid August to mid October. For both species, the artificially Fusarium-infected and DON-contaminated wheat straw was a more attractive food source than the non-infected control. In contrast to A. caliginosa, L. terrestris incorporated infected straw faster into the soil compared to control straw. Furthermore, the reduction of Fusarium biomass and DON concentration in wheat straw was significantly higher in the presence of L. terrestris than in treatments with A. caliginosa and without earthworms. Here, no significant differences could be measured between the Fusarium biomass and DON concentration in wheat straw. A. caliginosa seems not to be relevant for the reduction of Fusarium biomass and DON concentration. We concluded that amongst earthworms, anecic detritivorous species are the drivers to compensate possible negative consequences (like crop infection) of conservation tillage. They take an important role in the control of phytopathogenic and toxinogenic fungi surviving on plant residues and in the degradation of their mycotoxins. 相似文献

Effects of heavy metals on the litter consumption by the earthworm Lumbricus rubellus in field soils
Aim of this study was to determine effects of heavy metals on litter consumption by the earthworm Lumbricus rubellus in National Park the “Brabantsche Biesbosch”, the Netherlands. Adult L. rubellus were collected from 12 polluted and from one unpolluted field site. Earthworms collected at the unpolluted site were kept in their native soil and in soil from each of the 12 Biesbosch sites. Earthworms collected in the Biesbosch were kept in their native soils. Non-polluted poplar (Populus sp.) litter was offered as a food source and litter consumption and earthworm biomass were determined after 54 days. Cd, Cu and Zn concentrations were determined in soil, pore water and 0.01 M CaCl2 extracts of the soil and in earthworms. In spite of low available metal concentrations in the polluted soils, Cd, Cu and Zn concentrations in L. rubellus were increased. The litter consumption rate per biomass was positively related to internal Cd and Zn concentrations of earthworms collected from the Biesbosch and kept in native soil. A possible explanation is an increased demand for energy, needed for the regulation and detoxification of heavy metals. Litter consumption per biomass of earthworms from the reference site and kept in the polluted Biesbosch soils, was not related to any of the determined soil characteristics and metal concentrations. 相似文献

Fusarium head blight (FHB) is an important disease which has been causing damage to wheat and barley crops in western Canada. Because crop residues are an important source of inoculum, it is important to know the ability of Fusarium spp. to colonize and survive in different residue types, and how their populations might be affected by agronomic practices. Sampling of residue types on producers’ fields for quantification of Fusarium and other fungi was conducted in 2000–2001 in eastern Saskatchewan. Fusarium spp. were isolated from most fields, whereas their mean percentage isolation (MPI) was over 50% for cereal and pulse residues, and under 30% for oilseed residues. The most common Fusarium, F. avenaceum, had a higher MPI in pulse and flax (45–48%) than in cereal or canola (10–22%) residues. This was followed by F. equiseti, F. acuminatum, F. graminearum, F. culmorum and F. poae which were isolated from all, or most, residue types. Factors affecting Fusarium abundance in residues included the current crop, cropping history, and tillage system. In cereal residues, the MPI of F. avenaceum was higher when the current crop was another cereal (24%) versus a noncereal (4–8%). When the current crop was another cereal, the lowest MPI of F. avenaceum and F. culmorum occurred when the field had been in summerfallow (SF) two years previous (F. avenaceum: 17% for SF, 28% for a crop; F. culmorum: 1% for SF, 4% for a crop); in contrast, F. equiseti and Cochliobolus sativus were most common in residues of cereal crops preceded by SF (F. equiseti: 16% for SF, 10% for a crop; C. sativus: 22% for SF, 13% for a crop). The MPI of F. graminearum was higher when the crop two years previous was an oilseed (7%) versus a cereal (4%). In regards to tillage effects, when the current crop was a cereal, the MPI of F. avenaceum was higher under minimum (MT) and zero tillage (ZT) (22–37%) than conventional tillage (CT) (15%), that of F. graminearum was lowest under ZT (3% for ZT, 7–11% for CT-MT), whereas that of C. sativus was highest under CT (27% for CT, 6–11% for MT-ZT). Under ZT, previous glyphosate applications were correlated positively with F. avenaceum and negatively with F. equiseti and C. sativus. These observations generally agreed with results from previous FHB and root rot studies of wheat and barley in the same region. Percentage isolation of F. avenaceum from noncereal and of F. graminearum from cereal residues were positively correlated with FHB severity and percentage Fusarium-damaged kernels of barley and wheat caused by the same fungi. 相似文献

Subsurface drainage induces systematic spatial variability in soil properties which may be reflected in the abundance and distribution of soil organisms. We compared the population density of the deep burrowing earthworm Lumbricus terrestris L. above and between tile subdrains in 41 sample pairs on an eight hectare grass field. Above the drains the median number of individuals was twice as high and their total fresh mass five times as high as between the drains (4.5 vs. 2.1 individuals m-2 and 9.6 vs. 1.9 g m-2, respectively). The mean difference (above drain – between drains) was 2.5 individuals m-2 (95% CI = 1.0 to 4.0) and 6.6 grams m-2 (95% CI = 3.6 to 9.6). The relatively larger difference in fresh mass was due to a high proportion of adult individuals above drains. One likely explanation for the pattern of abundance is that the lowered water table level near the drains provides an environment beneficial for the population growth of L. terrestris. Due to the role of L. terrestris burrows as flow paths of percolating water the observations may have implications on subdrain function. 相似文献

利用微生物消除农药污染是一项安全、经济、有效的方法,降解动力学模型的构建有助于理解污染物的生物降解行为和估测系统中特征污染物的浓度变化,菌株对高浓度污染物的降解效果是降解菌在受污染水体中实际应用的关键。本研究采用基础培养基中定量添加毒死蜱和定时取样分析毒死蜱残留浓度的方法,探讨两株蜡状芽孢杆菌(HY-1和HY-2)的接种体培养时间、接种量和降解菌对毒死蜱的降解动力学,同时研究了降解菌对高浓度毒死蜱的降解率。结果表明:HY-1和HY-2最适接种体培养时间分别为10 h和19 h,接种体培养时间对菌株降解毒死蜱的影响较大。两菌株最适接菌量为8%(v/v),接种量从4%增至8%时,接种量对HY-1降解毒死蜱的影响大于HY-2。当毒死蜱的初始浓度为40 mg.L 1、80 mg.L 1、100 mg.L 1和120 mg.L 1时,一级动力学方程ln(C0/Ct)=kt可以用来拟合两菌株对毒死蜱的降解动力学及确定降解动力学参数,当毒死蜱初始浓度再次增加时,仅HY-2对毒死蜱的降解符合一级动力学方程。当毒死蜱初始浓度为40~120 mg.L 1时,菌株HY-1对毒死蜱的降解速率常数分布在0.013 5~0.015 7;当毒死蜱初始浓度为40~200 mg.L 1时,菌株HY-2的降解速率常数分布在0.008 0~0.015 3。菌株HY-2比HY-1可以在较高的毒死蜱浓度下发挥降解作用,且降解率较高。因此,两菌株在毒死蜱污染水体的净化去毒方面具有重要意义。 相似文献

The effects of fertilization with N, P, K, and organic manure (alone or in combination) on earthworm populations, biomass, and casting activity were measured in a cultivated soil (organic C 1.5%, annual rainfall 2000–2300 mm). These applications of fertilizer caused significant increases in earthworm numbers, biomass, and casts. N alone or in combination with P and K also influenced these earthworm parameters significantly. The inorganic NPK fertilizer in combination with organic manure had a significantly greater effect on earthworm activities than NPK fertilizer alone, and therefore the addition of organic matter appears advisable in order to obtain maximum benefits from NPK fertilizer in this soil. 相似文献

香蕉枯萎病由病原真菌尖孢镰刀菌4号生理小种引起,其根际微生物组是抵御病原菌入侵的关键屏障。根际原生动物群落在根际微生物群落调控以及植物健康方面有不可忽视的作用。利用Illumina MiSeq高通量测序技术分析连作蕉园中植株发病前后根际土壤原生动物群落特征变化以及其与病原菌的相互关系。结果表明:相较于可培养细菌与真菌,原生动物的数量对病原菌的种群数量变化的影响更大;根际原生动物的群落多样性与丰富度都随植株生长而降低,且在发病植株中更低;抽蕾前后的健康与发病植株中根际原生动物群落结构存在显著差异;总体而言,吞噬型原生动物的相对丰度显著高于其他功能类型的原生动物,且在健康与发病植株中差异显著;吞噬型原生动物中与病原菌具有显著相关性的多为丝足虫门;在抽蕾期的健康植株中,丝足虫门的高丰度属中Group-Te与Cercomonas属与病原菌呈显著负相关;在抽蕾期的患病植株中,相对丰度较高的植物致病型原生动物Pythium属与病原菌呈显著正相关。综上所述,根际土壤中原生动物对病原菌的影响较大,在植株生长过程中,原生动物的群落特征发生改变,且在发病与健康植株中存在差异。与病原菌具有显著负相关的吞噬型原生动物,尤其是丝足虫门的部分高丰度属,可能会在防控香蕉枯萎病上产生一定潜力。 相似文献

The earthworm population in a winter cereal field in Ireland was studied over a 3-year-period and its effects on soil and N turnover were assessed. The mean annual population density was 346–471 individuals m-2 and the mean biomass was 56.9–61.2 g m-2. Twelve species were recorded, the most abundant being Allolobophora chlorotica followed by Aporrectodea caliginosa, and 242 mg at 5°C to 713 mg at 10°C in the case of juvenile Lumbricus terrestris. Gut contents (dry mass of soil) comprised 6.7–15.5% of the A. caliginosa live mass, and 9.7–14.7% of the Lumbricus terrestris mass. Annual soil egestion by the field population was estimated as 18–22 kg m-2. Tissue production ranged from 81.7 to 218.5 g m-2, while N turnover resulting from mortality was calculated as 1.5–3.9 g m-2 depending on the year and the method of calculation. Earthworms were estimated to contribute an additional 3.4–4.1 g mineral N to the soil through excretion, mucus production, and soil ingestion. Independent estimates of N output via mucus and excretion derived from 15N laboratory studies with Lumbricus terrestris were 2.9–3.6 g m-2 year-1. 相似文献

Summary When earthworm casts were used as a casing, an early fructification, a greater stimulation in fruitbody formation, an increase in the carpophore protein content and in L-14C-leucine incorporation were observed inAgaricus bisporus. The protein-synthesizing capacity of the carpophore and its protein content were tested on samples collected 28 and 35 days after the casing. Summary To assess the ability of endogeic earthworms to tolerate variations in soil water potential, groups of these worms were subjected to different, constant levels of soil water suction () over a period of 17 days. At water suctions varying from 0.3 kPa (pF 0.5) to 1990 kPa (pF 4.3), the earthworms showed no physiological ability to a maintain constant internal water content as assessed by the fresh weight. The relationship between weight loss and the increase in water, suction was modelled and the following critical thresholds were identified: <60 kPa did not affect earthworm weight; >620 kPa led to diapause; between these two values, earthworm weight was closely governed by the variation in . At the intermediate suction of 167 kPa, the exchange of water between the earthworms and the soil was at a maximum, and this value is therefore proposed as the level at which the diapause is induced. In parasite-host dynamics, parasites exert frequency-dependent selection on their hosts by favouring rare alleles that may confer resistance against infection. Therefore host populations that suffer strong parasite stress should maintain higher levels of genetic variability. We studied the Lumbricus terrestris-Monocystis sp. host-parasite system at a microgeographical scale. Using three polymorphic microsatellite loci on one large earthworm population sampled at 26 different sites (281 genotypes), we tested the relationship between parasite load and genetic variation in natural samples of the common earthworm L. terrestris. Our analysis yielded the following: (1) parasite load varied significantly across sites in this population; (2) there was no consistent evidence for heterozygote deficiency (observed heterozygosities ranged between 0.74 and 0.87), indicating a low level of inbreeding; (3) there was no significant genetic structuring among sample sites; (4) we could not identify a significant association between parasite load and population genetic diversity; (5) there was considerable population differentiation (15.17%) between our German samples and a Canadian L. terrestris reference population. Our study provides insight into the population genetics of one of the most economically important soil organisms on a microgeographic scale. 相似文献

Summary Adult earthworms (Aporrectodea longa) were maintained for 199 days in soil columns (h=30 cm; ø=10 cm) where the water potential ranged from -7 to-65 kPa and compaction from 50 to 350 kPa. The weight of casts on the soil surface was measured at the end of two periods of activity (spring and autumn). Cast production increased with bulk density, but the activity of earthworms was limited both by the mechanical strength of the soil and by decreasing water potential. The results obtained in the laboratory conditions of this study were consistent with field observations on casting and burrowing activities. The effects of water potential and soil compaction on these activities were estimated. 相似文献

Rates of carbon dioxide production of cocoons, juveniles and matures of Lumbricus rubellus were measured at five constant (2°C, 5°C, 10°C, 15°C, 20°C) and three diurnally fluctuating temperature regimes (0–10°C, 5–15°C, 10–20°C) covering the whole range of temperature conditions experienced by this species in forests in Central Germany. Respiration rates of developmental stages significantly increased with temperature both under constant and fluctuating regimes. Overall, at constant temperature regimes Q10 values for cocoons, juvenile and mature earthworms were 1.6, 2.7 and 2.0, respectively. At fluctuating temperature regimes Q10 values were generally higher with 2.4, 3.6 and 3.5 for cocoons, juvenile and mature earthworms, respectively. Summary Although various authors have contributed to our knowledge of the biology of Eudrilus eugeniae some basic facts about the life cycle and reproduction are still not fully documented. Quantitative observations were made of the cocoons at 25 °C in different substrates. The incubation period for 166 cocoons was 16.89 days, based on cocoons produced by worms between the ages of 70 and 100 days. These cocoons produced a mean 2.12 hatchlings per cocoon after incubation in cattle manure, moist filter paper and distilled water. A smaller batch of cocoons incubated in cattle manure produced a mean of 2.7 hatchlings per cocoon. The hatching success of the cocoons was 84% in cattle manure, 50% in distilled water, and 48% on moist filter paper. The reproductive capabilities of E. eugeniae and Eisenia fetida were compared. 相似文献
