Samples of Manila clam (Ruditapes philippinarum Adams & Reeve 1850) were collected from May 2004 to April 2005 monthly, and plankton net trawling of planktonic larvae and bottom sediment sampling surveys were further conducted from May to October 2006 in Jiaozhou Bay. Based on the data collected, growth, mortality and reproduction of the transplanted Manila clam and the environmental effects were examined. The results showed that the enhanced clams grew well and showed a growth trend similar to the local wild ones. The main growth periods lasted from April to September, with the water temperature being the main factor affecting the growth, which was the same as that of the wild clams. There were also two reproduction cycles for the farmed Manila clams each year in Jiaozhou Bay and the main breeding period was from May to June. The phenomenon of delayed metamorphosis was quite common through larval development. The farmed clams could spawn when they reached sexual maturity, but they could not perform effective recruitment as many planktonic larvae died during metamorphosis and settlement. A preliminary study indicated that sediment perturbation and marine environment pollution were the main factors causing the death of larvae in the development process.  相似文献

Abundance, length‐frequencies and distribution of Ruditapes philippinarum (Manila Clams) and Tellina (Quidnipagus) palatum were measured at two beaches in Kāne'ohe Bay, O'ahu, Hawai'i in June, 2010. Abundances had decreased from 866.2 m?2 to 3.4 m?2 for Ruditapes and from 75.5 m?2 to 1.5 m?2 for T. palatum since 1977. Distribution of both species was patchy, but both were most commonly found >40 m from shore. Size frequencies of live clams compared with empty shells suggest that few Ruditapes survive to sexual maturity. A similar trend was not detected for T. palatum. Aquaculture trials of R. philippinarum were conducted at the He'eia and Moli'i traditional Hawaiian fishponds in the same bay. The clams failed to thrive, although triploid and diploid Crassostrea gigas performed well in concurrent trials in the same fishponds. Current lower abundances for wild R. philippinarum could be due to factors related to predation or nutrient limitations. Previously, two large sewage outfalls existed at the surveyed clam bed areas which may have temporarily increased nutrient availability for both wild and cultured clams. Current nutrient levels may inhibit Manila clam growth and recruitment in Kāne'ohe Bay.  相似文献

We investigated the effects of infection by the protozoan Perkinsus olseni on the reproduction of female Manila clams, Ruditapes philippinarum, from a population in Gomso Bay, Korea. The reproductive effort of the clams was assessed by ELISA using a clam egg-specific antibody and was expressed as a weight-based gonadosomatic index (GSI). The number of Perkinsus infecting each clam was estimated from the gills using Ray's fluid thioglycollate medium (RFTM) along with a NaOH digestion assay. We found that reproductive effort was negatively correlated with the intensity of the Perkinsus infection: more heavily infected clams produced fewer eggs during the spawning period from May to August. Frequency of spawning was also negatively correlated with the level of Perkinsus infection; heavily infected clams (HIC) exhibited a single spawning pulse in late July, whereas lightly infected clams (LIC) showed three spawning peaks in mid-May, late July, and late August. Egg production of HIC was only 30–75% of LIC during spawning. The level of total protein in LIC was also higher year round than in HIC. In conclusion, our investigation demonstrates that a high level of Perkinsus infection affects spawning frequency and reduces egg production, which may have long-term impacts on clam recruitment and population growth.  相似文献

Mass selection for shell length was conducted in the orange strain of the Manila clam, Ruditapes philippinarum, in this study. Based on the size‐frequency distribution, the clams constituting the largest 10% of the distribution were selected as parents for the selected line. An equal number of clams of the orange strain were randomly chosen as parents for the control line prior to the size selection. The responses to selection and realized heritability were 2.37 and 0.61, respectively. In addition, 97.3% of the progenies inherited the orange shell color. Our results suggest that mass selection for fast growth of shell length in this orange strain of the Manila clam is effective.  相似文献

2004年5月至2005年4月对胶州湾菲律宾蛤仔(Rudimpes philippinarum)底播增殖区进行了逐月定点采样,将样品带回放于实验室内水族箱暂养,每站随机取样,共对3269个个体进行了基本生物学特性测定,研究了移植底播菲律宾蛤仔的生长、死亡等渔业生物学特性。结果表明,底播增殖菲律宾蛤仔苗种的平均个体质量0.26g、平均壳长11.1mm;底播蛤仔与野生蛤仔在生长规律上基本一致,春末至秋初(4~9月)是其主要生长期;1~3龄期间个体生长速度较快,1~2龄、2~3龄,个体质量分别增长3.88g和4.02g;1~2龄生物量增长最快,2~3龄由于死亡率增大,其生物量增长缓慢;1~3龄蛤仔软体部的生长速度快于贝壳,5~6月是菲律宾蛤仔的繁殖肥育期。根据个体生长特性,3龄为最佳采捕年龄;根据目前的养殖状况,2龄蛤仔已达到商品规格,从生物量上分析,采捕2龄蛤仔收益最高;6月是最佳的捕获时期。蛤仔生长具有明显的季节变化,水温是影响菲律宾蛤仔生长的主要环境因子。[中国水产科学,2006,13(4):642—649]  相似文献

The protein expression profiling in clam haemocytes and plasma in response to Perkinsus olseni was addressed. Adult Manila clams from a P. olseni‐free bed were experimentally challenged with parasite zoospores to analyse immune response. In another experiment, the effects of longer term infection were assessed in adult clams collected from a P. olseni‐affected bed, by comparing moderate to very heavily infected clams with non‐infected ones. Haemocyte and plasma proteins were separated by two‐dimensional electrophoresis; spot patterns were qualitatively compared between treatments within each experiment and the spots indicating differential protein expression associated with P. olseni challenge or with field infection were processed for protein identification. Fifteen clam proteins (four in haemocytes and eleven in plasma) of which expression was markedly affected by P. olseni were identified. Some of the identified proteins have a well‐known role in clam immune response against the parasite, such as lysozyme and lectins. Rho GTPase‐activating protein 6 could be a marker of resistance against P. olseni, which should be further studied.  相似文献

Manila clam Ruditapes philippinarum is common and abundant on Japanese tidal flats, forming a commercially important clam fishery. However, annual catches of Manila clam have decreased drastically since 1975?C1985. To study larval recruitment processes of Manila clam, we carried out scanning electron microscopy (SEM) observations on larval and juvenile shells of clams reared at 20 and 24?°C. There was no significant difference in final shell length of trochophores between 20 and 24?°C. However, the larval duration was much longer and the shell length of settled size of pediveligers was much larger for clams reared at 20?°C than those reared at 24?°C. These findings suggest that larval duration and growth, as well as settlement size, may vary markedly depending on temperature (and probably on season). The larval shell morphology of Manila clam can provide essential information about larval recruitment processes.  相似文献

Infection with Perkinsus species, primarily P. olseni, is thought to be a major cause of the decline of Manila clam populations in Japan since the 1980s. However, the pathogenicity of the infection has not been sufficiently evaluated to estimate the impact of infection on wild Manila clam populations. We experimentally challenged juvenile (3- to 6-mm shell length) and adult (18- to 22-mm shell length) Manila clams with P. olseni at 18, 23, 28, and 30 °C. Mortality was significantly higher in challenged groups than in control groups. The difference in mean mortality between the challenged and control groups (all life stages and temperatures) was only significant above a threshold of infection intensity ~106 cells/g soft wet tissue (SWT). As temperature increased, the onset of mortality occurred more rapidly. The increase in mortality occurred earlier in juveniles than adults at 28 °C and lower. Our results suggest that the pathogenicity of P. olseni is higher in juveniles than in adults and at higher water temperatures. Given the infection intensities (ca. 106 cells/g SWT) previously reported in wild Manila clams, the parasite likely has considerable impact on wild Manila clam populations, particularly juveniles during periods of high temperature.  相似文献

In this study, the energy budget of the Manila clam, Ruditapes philippinarum, was evaluated after one-week acclimation periods at 5, 10, 15, 20, and 25°C. Small clams (151 ± 12 mg DW) and large clams (353 ± 16 mg DW) were fed with the microalgae, Isochrysis galbana. Filtration rate, ingestion rate, assimilation efficiency, oxygen-consumption rate, and ammonia excretion rate were measured. Both filtration rate and ingestion rate of small and large clams were found to be related to temperature. The highest Q 10 values were measured in the range 15–20°C for both small and large clams. Assimilation efficiency of both small and large clams was not significantly influenced by temperature, although the maximum mean values were detected at 20°C. Oxygen consumption rate and ammonia excretion rate of small and large clams were found to be related directly to temperature over the entire range, with a maximum being detected at 25°C. The highest Q 10 value was estimated in the range 10–15°C with regard to oxygen consumption rate, and in the range of 15–20°C with regard to ammonia excretion rate. Scope for growth (SFG) was positive at all temperatures, achieving a maximum value at 20°C in both small and large clams, primarily as a consequence of the enhanced ingestion rate which offset the concomitant elevation in the metabolic rate. 1. Margaritifera margaritifera populations are declining throughout its range, including Ireland, despite legislation designed to protect freshwater pearl mussels and their habitat. 2. A survey of freshwater pearl mussels was carried out on rivers in County Donegal, north-west Ireland, to determine the current distribution, size and density of M. margaritifera populations, as well as to identify potential threats to mussels there. 3. The survey revealed the freshwater pearl mussel to be widespread, particularly in the western half of the county. However, densities of mussels at most sites are low, with just two sites having mussel densities of over 5 m−2. Furthermore, the species appears to be absent from a number of sites from which it had been previously recorded. 4. According to the literature, there is a long history of pearl fishing in Co. Donegal and neighbouring counties. Evidence from heaps of shells found on the river bed and banks at several sites and recent anecdotal reports from local people suggest pearl fishing is being practised on all rivers investigated during the present study. 5. The main conservation requirements for M. margaritifera populations in Donegal are to maintain water quality at its present high standard and, as pearl fishing appears to be a widespread and immediate threat to the remaining mussel populations, to enforce existing legislation designed to protect M. margaritifera.  相似文献

Abstract The effectiveness of live-algae replacement diets for the conditioning and spawning success of adult Manila clam, Tapes philippinarum (Adams and Reeve), was assessed. Two dried diets were tested: a diet of dried Tetrasetmis suecica (Kylin) Butch, and a mixed diet of dried T. suecica (70%) and dried Cyclotella cryptica Reimann et al. (30%). These diets were compared with three live diets: Skeletonema costatum (Grev.) Cleve, Dunaliella tertiolecta (Butcher) and T. suecica. An unfed group was used as a control. The same ration (dry weight) of food was supplied for all groups at 6% of the dry meat weight per day. The broodstock were conditioned for 7 weeks. After 4 weeks none of the animals was ready to spawn. After 6 weeks all the diets promoted spawning and after 7 weeks the unfed group also spawned. It seems that dried algal diets are effective for conditioning and spawning of Manila clams, as the dry meat weight, condition index and fecundity of broodstock fed these diets were significantly greater than for unfed animals. However, the fecundity and proportion of parental lipid released in the eggs of clams conditioned with dried diets was lower than for the clams conditioned with live algae.  相似文献

In 1989 and 1990. triploid Manila clam, Tapes philippinarum Adams and Reeve, seed were reared to 15-20 mm at the Fisheries Laboratory, Conwy, and planted out in the Menai Strait, North Wales. In each of the summers of 1992 and 1993, three of these batches, at 2, 3 or 4 years old, were returned to the laboratory to assess ploidy, size, spawning potential and biochemical composition. Percentage triploidy at this time was similar to that in the seed. After 6 and 8 weeks of warmwater conditioning. Only 45 out of l21 triploids (37%) were induced to spawn by thermal shock, with only one spawning as a male. By comparison, 75% of diploid clams spawned with a 1:1 ratio of males to females. Mean fecundity of triploids was significantly lower than that for diploids, at 0.497 compared with 1.54 million eggs per female. Compared with eggs from diploid females, eggs from triploids were larger and significantly fewer of them developed into D-larvae when fertilized by sperm from diploid males. Triploid clams were heavier and had a higher condition index and carbohydrate content than diploids of the same age, but lipid levels were similar. Potential advantages of producing and cultivating 100% triploid batches of Manila clam seed are discussed.  相似文献

Soft shell clams, Mya arenaria, are found from Canada to North Carolina on the U.S. Atlantic Coast and from Canada to California on the U.S. West Coast. They are also found in several other parts of the world including Europe. The primary market for these clams on the U.S. East Coast is in New England. Clams are sold whole live or in one of the several cooked forms. Commercial soft clam shucking and processing is primarily by manual methods. However, physical properties data for these clams is lacking and is a constraint on the automation of processing. Several properties of soft shell clams harvested from five different harvest locations in the Chesapeake Bay are detailed in this paper and relationships between the components of the clam are defined mathematically through regression equations. The live weight and the clam length are related to shell weight, siphon weight, meat weight, total solids and free water in the clams. Meat yields under both standard processing methods and for clams steamed and consumed with only the shell removed are detailed. The effect of harvest area on the clam parts is also defined. Soft shell clams shucked manually result in a meat yield of approximately 29% of the live weight. If steamed and eaten the meat yield may be as high as 43% of the live weight, primarily because the siphon is often consumed in steamed clams.  相似文献

A novel sialic acid-specific lectin (MCsialec) was detected from an expressed sequenced tag (EST) sequence from Manila clam haemocytes infected with Perkinsus olseni. The cDNA of the lectin was cloned using gene-specific primers based on a previously determined EST and characterized. The full-length cDNA of MCsialec is 603 bp in length and encodes a polypeptide of 200 amino acids with a calculated molecular mass of 21.928 kDa. Sequence alignment and protein motif analyses showed that MCsialec shares identity with sialic acid-specific invertebrate lectins from Cepaea hortensis, Helix pomatia and Haliotis discus discus. The lectin was expressed in Escherichia coli M15 cells and purified using a Ni-NTA His-binding resin matrix for antibody production. The presence of the lectin in various tissues of Perkinsus-infected and uninfected Manila clams was analysed by both PCR and immunohistochemical localization assays. MCsialec was detected in each tissue of the clams; however, upon infection, the level of expression of the lectin increased in each tissue. Vibrio tapetis infection also induced high-level expression of MCsialec in the haemocytes. These data suggest that MCsialec plays a crucial role in the immune system of the Manila clam during pathogenic infection.  相似文献

Monitoring of persistent QPX infections in clams of Raritan Bay (New York) shows certain areas of the estuary have remained without any significant disease prevalence. This study was conducted to investigate the potential to mitigate QPX disease by relocating infected hard clams, Mercenaria mercenaria (Linnaeus), from enzootic areas to nearby sites with prevailing environmental conditions suggested to deter infection and favour remission and healing. Clams were collected from a location with consistent disease prevalence in central Raritan Bay and brought to near shore habitats subject to lower salinities and higher summer temperatures. A reduced host density treatment was included in the study to examine the common observation of high clam density in the most persistently infected locales. An additional treatment retained clams above the sediment, since sediments are suspected to represent a QPX reservoir. At the end of the 4‐month study all treatments displayed less QPX disease than the control group and the greatest contrast was provided by the disappearance of infections in a tidal creek.  相似文献

Mortality of the short-neck clam Ruditapes philippinarum exposed to the toxic dinoflagellate Heterocapsa circularisquama was studied under controlled conditions to clarify the mechanisms of recurrent mass deaths of clams occurring in western Japanese coastal areas. One-week mortality tests, involving three water temperatures, six H. circularisquama concentrations, and two clam body sizes, showed a significant increase in mortality with increasing temperature, H. circularisquama concentration, exposure duration, and body size (ANOVA, P < 0.01). Clam death was observed at concentrations as low as 50 cells/ml and temperatures as low as 15°C. Prior to death, clams showed an extreme retraction of their mantle edge and siphon, along with recurrent vomiting behavior before initiating a closure reaction followed by paralysis then death. Gills of paralyzed clams showed an important uptake of dye, implying gill damage. This study is the first laboratory evidence of bivalve mortality induced by H. circularisquama at low concentrations and low temperature, and the first report of differential effects according to the body size of bivalves.  相似文献

The virulence of two Vibrio strains, previously isolated from diseased corkwing wrasse Symphodus melops and identified as V. tapetis and V. splendidus, to corkwing and goldsinny wrasse Ctenolabrus rupestris and to Atlantic salmon Salmo salar, was studied under laboratory conditions. Both bacteria were shown to be opportunistically pathogenic to corkwing wrasse, causing significantly higher mortality in the challenged groups than in the controls. Bacterial cultivation of kidney samples and re-isolation of V. tapetis and V. splendidus from most mortalities confirmed the two strains as the probable cause of mortality in the challenged groups. The control group also suffered relatively high mortality, but no specific pathogens that were suspected to be the main cause of death were isolated, other than a mixture of Vibrio spp. and, in the case of one individual, atypical Aeromonas salmonicida. Following injection challenge with both bacterial strains, no mortality was recorded in Atlantic salmon. In bath challenge trials with goldsinny wrasse, only slight mortality was observed in the challenged groups and the unchallenged control group. Bacterial examination showed that atypical Aeromonas salmonicida was the probable cause of death in both bath challenged and control groups of goldsinny wrasse, and no indication of infection by any Vibrio sp. was found.  相似文献

With increasing awareness of the use of the coastal zone, it is necessary to understand the potential environmental effects of aquaculture practices. This is especially important when non-native species, which may be competitively superior to native species, are cultivated. A 5-year experiment was established to study the environmental effects of the various stages of Manila clam. Tapes philippinarum Adams & Reeve, cultivation, from seeding, through on-growing, harvesting and post-harvesting. The aim was to monitor changes in biological and physical variables in the sediment which may be useful in formulating an environmental management strategy for the cultivation of this species. This paper describes the biological and physical changes that occur in the sediment during the early phase of clam cultivation. We compared the changes in netted plots (with and without clams) and unnetted control areas, 6 months before and after laying the clams. The clams were planted in April 1992 under netting at a density of 500 m-2 (0.16 kg m-2), and in 6 months, had increased their weight to 3.2 kg m-2 but decreased their number to 410 m-2. A significant, but small increase in organic content (net only plots, 3.37%; control plots 2.42%) and in phaeopigment (netted plots, 8.6 mg m-2; control, 5.6 mg m-2) of the sediment in the netted plots relative to the control areas were seen. Short-term sedimentation rates on the netted plots were up to four times higher than in the control areas. The netting also encouraged the settlement of Enteromorpha sp. which, in turn, attracted Littorina littorea to feed on these plots. The infaunal community in the control areas was similar to that in samples 12 months earlier and continued to be dominated by the predatory polychaete Nephtys hombergii. Netted plots (with and without clams) had a greater abundance of deposit feeding polychaetes. particularly Ampharete acutifrons and Pygospio elegans, which were the dominant fauna in these plots. Within the clam treatments, the density of clams had a negative effect on the abundance of cirratulids, although mean abundance was generally greater than in the control areas. There were few physical changes to the experimental area after 6 months. The most important effect appears to be the increased sedimentation rate over plots with netting, which has led to an increase in productivity of those areas.  相似文献

Differences in taste and odour between three kinds of clam with the highest aquacultural production in China were investigated. Meretrix petechialis, Mactra veneriformis and Ruditapes philippinarum (fresh and dried product) were analysed firstly by electronic tongue and electronic nose. Fresh and dried clams could be easily distinguished, and there was little difference between uncooked fresh clams, while greater difference occurred between R. philippinarum and other fresh clams after cooking. The total free amino acid (FAA) content of uncooked clams increased after cooking but decreased in dried clam; the highest proportion was of sweet taste FAAs. Ala, Glu, Arg, 5′‐adenosine monophosphate and 5′‐inosine monophosphate were the most important active taste compounds. The umami intensity order was found to be fresh R. philippinarum, followed by M. petechialis and M. veneriformis, and finally dried R. philippinarum. The greatest number of volatile compounds was found in dried clam, while M. petechialis and M. veneriformis had the most compounds in common.  相似文献

To assess the potential for hatchery production of the venerid clam, Tapes dorsatus, diploid and triploid clams were produced and ongrown until the spat reached 5–8 mm in size. Triploidy percentages at metamorphosis that ranged from 56 to 85% were induced using a 15-min exposure to 1 mg/l cytochalasin B. No differences were observed in the growth rate of sibling diploid and triploid T. dorsatus larvae. Survival of diploid clams to pediveliger stage was however higher. Post-metamorphic growth and survival of diploid and triploid clams remained the same until the clams were removed from the hatchery to outdoor nurseries. Throughout the larval and early spat phases of production, T. dorsatus growth was similar to that reported for the Manila clam, Tapes philippinarum. Based upon its ease of culture, rapid growth and marketability, T. dorsatus are thought to have considerable aquaculture potential.  相似文献
