The tuberous roots of Decalepis hamiltonii are consumed as pickles and beverages and are believed to possess health-promoting properties. We have investigated the antioxidant potential of the roots. The methanolic extract of the root showed a high antioxidant activity. The methanolic extract was fractionated on a silica gel column, which showed three major fractions with good antioxidant activity. The active fractions were further subjected to preparative thin layer and silica gel column chromatography, which yielded six pure compounds. The purified compounds were characterized by MS, 1H NMR, 13C NMR, and two-dimensional NMR spectroscopic techniques and identified as 2-hydroxy-4-methoxybenzaldehyde, p-anisaldehyde, vanillin, borneol, salicylaldehyde, and bis-2,3,4,6-galloyl-alpha/beta-D-glucopyranoside. The latter compound, named decalepin, is a new antioxidant molecule from the plant kingdom. The purified compounds showed antioxidant activities in in vitro assays such as inhibition of lipid peroxidation, hydroxyl radical, superoxide anion, and 1,1-diphenyl-2picrylhydrazyl radical scavenging. This is the first report of the antioxidant constituents of the roots of Decalepis hamiltonii.  相似文献

The chemical composition of the essential oils obtained from the species restricted to Greece and the eastern Mediterranean region, Satureja spinosa L. and Thymus longicaulis L.; species endemic to central and south Greece, Satureja parnassica ssp. parnassica Heldr. and Sart ex Boiss.; species endemic to the island of Crete, Origanum dictamnus L.; and species widely distributed in the Mediterranean region, Satureja thymbra L. and Origanum vulgare L. subsp. hirtum, were determined by gas chromatography (GC) and GC/mass spectrometry (MS) analysis. The in vitro antibacterial activities of the essential oils were evaluated against a panel of five foodborne bacteria (Escherichia coli 0157:H7 NCTC 12900, Salmonella enteritidis PT4, Staphylococcus aureus ATCC 6538, Listeria monocytogenes ScottA, and Bacillus cereus FSS 134). The analytical data indicated that various monoterpene hydrocarbons and phenolic monoterpenes constitute the major components of the oils, but their concentrations varied greatly among the oils examined. The antibacterial assay results showed that 5 muL doses of the essential oils extracted from the endemic Satureja species in Greece possess remarkable bactericidal properties, which are clearly superior as compared to those of Origanum and Thymus species essential oils. Therefore, they represent an inexpensive source of natural mixtures of antibacterial compounds that exhibit potentials for use in food systems to prevent the growth of foodborne bacteria and extend the shelf life of the processed food.  相似文献

Roots of Decalepis hamiltonii are consumed as pickles and as a health drink in southern India for their health benefits. The antioxidant properties of the root extracts have been shown previously; this paper reports the isolation of antioxidant compounds from the aqueous extract of the roots of D. hamiltonii. Five novel antioxidant compounds were isolated and characterized by NMR and MS. The compounds exhibited free radical scavenging activity in vitro and inhibited low-density lipoprotein oxidation. This study demonstrates that the root extract of D. hamiltonii is a cocktail of several antioxidant compounds with health implications.  相似文献

为了探究低温等离子体活化水 (cold plasma activated water, PAW) 对丁香假单胞杆菌猕猴桃致病变种 (Pseudomonas syringae pv. actinidiae, PSA) 的抗菌活性和潜在机制。该研究通过介质阻挡放电(dielectric barrier discharge, DBD) 低温等离子体发生装置制备不同激活时间的PAW,考察放电时间与杀菌效果之间的关系。此外,通过评估PSA形态特征,细胞粒径、DNA、细胞膜损伤情况和胞内活性氧积累 (reactive oxygen species,ROS) 情况探究PAW对PSA的杀菌机制。结果表明:PAW对PSA的杀灭效果与PAW激活时间呈依赖性,与对照相比PAW处理120 s后,PSA显著 (P<0.05) 减少了4.38 lg (CFU/mL)。扫描电子显微镜 (field emission scanning electron microscope, FESEM) 结果清楚地表明,由于PAW处理,PSA细胞发生了明显的质壁分离。荧光染色结果显示,PSA细胞DNA、膜渗透屏障被破坏程度、内容物泄漏量和ROS积累量与PAW激活时间表现为正相关关系。因此,推测PAW由于自身酸性、较高的氧化还原电位 (oxidation-reduction potential, ORP),以及水性活性物质导致细胞的氧化损伤,而且这可能是杀灭PSA的主要原因。研究结果可为PAW控制猕猴桃细菌性溃疡病提供参考。  相似文献

Helicobacter pylori is linked to a majority of peptic ulcers and to some types of gastric cancer, and resistance of the microorganism to antibiotic treatment is now found worldwide. Virgin olive oil is an unrefined vegetable oil that contains a significant amount of phenolic compounds. Under simulated conditions, we have demonstrated that these substances can diffuse from the oil into the gastric juice and be stable for hours in this acidic environment. In vitro, they exerted a strong bactericidal activity against eight strains of H. pylori, three of them resistant to some antibiotics. Among the phenolic compounds, the dialdehydic form of decarboxymethyl ligstroside aglycon showed the strongest bactericidal effect at a concentration as low as 1.3 microg/mL. Although the experimental conditions are different from other reported works, this bactericidal concentration is much lower than those found for phenolic compounds from tea, wine, and plant extracts. These results open the possibility of considering virgin olive oil a chemopreventive agent for peptic ulcer or gastric cancer, but this bioactivity should be confirmed in vivo in the future.  相似文献

The aim of the study presented here was to gain knowledge about the vapor-phase antimicrobial activity of selected essential oils and their major putatively active constituents against a range of foodborne bacterial and fungal strains. In a first step, the vapor-phase antimicrobial activities of three commercially available essential oils (EOs)-cinnamon (Cinnamomum zeylanicum), thyme (Thymus vulgaris), and oregano (Origanum vulgare)-were evaluated against a wide range of microorganisms, including Gram-negative bacteria (Escherichia coli, Yersinia enterocolitica, Pseudomonas aeruginosa, and Salmonella choleraesuis), Gram-positive bacteria (Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus, and Enterococcus faecalis), molds (Penicillium islandicum and Aspergillus flavus), and a yeast (Candida albicans). The minimum inhibitory concentrations (MICs) were generally lower for oregano EO than for the thyme and cinnamon EOs, especially against the relatively resistant Gram-negative. The persistence of the EOs' antimicrobial activities over time was assessed, and changes in the composition of the atmosphere they generated over time were determined using single-drop microextraction (SDME) in combination with gas chromatography-mass spectrometry (GC-MS) and subsequent analysis of the data by principal component analysis (PCA). More relevant chemicals were selected. In addition, the vapor-phase activities of putatively key constituents of the oils were screened against representative Gram-positive (L. monocytogenes) and Gram-negative (S. choleraesuis) bacteria, a mold (A. flavus), and a yeast (C. albicans). Of the tested compounds, cinnamaldehyde, thymol, and carvacrol showed the strongest antimicrobial effectiveness, so their MICs, defined as the minimum vapor concentrations that completely inhibited detectable growth of the microorganisms, were calculated. To check for possible interactions between components present in the EOs, cinnamon EO was fortified with cinnamaldehyde and thyme EO with thymol, and then the antimicrobial activities of the fortified oils were compared to those of the respective unfortified EOs using fractional inhibitory concentration (FIC) indices and by plotting inhibition curves as functions of the vapor-phase concentrations. Synergistic effects were detected for cinnamaldehyde on A. flavus and for thymol on L. monocytogenes, S. choleraesuis, and A. flavus. In all other cases the fortification had additive effects, except for cinnamaldehyde's activity against S. choleraesuis, for which the effect was antagonistic. Finally, various microorganisms were found to cause slight changes over time to the atmospheres generated by all of the EOs (fortified and unfortified) except the fortified cinnamon EO.  相似文献

The essential oil content and the composition of subterranean parts of two valerian (Valeriana officinalis, L.) cultivars Select and Anthose, from certified commercial organic fields, were determined by hydrodistillation, followed by gas chromatography (GC) and GC/mass spectrometry analysis. Eight and fourteen month old cv. Select had 0.67 and 0.87% essential oil, while similar aged cv. Anthose contained 0.97 and 1.1% essential oil. Forty-three and fifty-three components from cv. Select and cv. Anthose oils were detected, respectively. The oil composition significantly varied due to the cultivar type, plant age, and/or harvesting time. The major components for cv. Select were valerenal, bornyl acetate, 15-acetoxy valeranone, valerenic acid, and camphene, while cv. Anthose had valerenal, (-)-bornyl acetate, alpha-humulene, camphene, 15-acetoxy valeranone, and valerenic acid. With further aging of the plants, the valerenal, valerenic acid, and alpha-humulene contents increased. The oil of cv. Select had a strong antimicrobial effect against Aspergillus niger, Escherichia coli, Staphylococcus aureus, and Saccharomyces cerevisiae, while cv. Anthose showed low or no activity against all test microbes, including Pseudomonas aeruginosa, suggesting that the inhibitory activity of valerian oil depends on the cultivar and its developmental stage. The oil profile of our cultivars did not match the literature proposed chemotype profiles.  相似文献

Acaricidal activities of components derived from Foeniculum vulgare fruit oil against Dermatophagoides farinae and Dermatophagoides pteronyssinus were examined using direct contact application and compared with that of the commercial repellent benzyl benzoate. The major biologically active constituent of Foeniculum fruit oil was characterized as (+)-fenchone by spectroscopic analyses. On the basis of LD(50) values, the compound most toxic to D. farinae was p-anisaldehyde (11.3 mg/m(2)) followed by (+)-fenchone (38.9 mg/m(2)), (-)-fenchone (41.8 mg/m(2)), benzyl benzoate (89.2 mg/m(2)), thymol (90.3 mg/m(2)), and estragol (413.3 mg/m(2)). Against D. pteronyssinus, p-anisaldehyde (10.1 mg/m(2)) was much more effective than benzyl benzoate (67.5 mg/m(2)), thymol (68.5 mg/m(2)), and estragol (389.9 mg/m(2)). These results indicate that the acaricidal activity of F. vulgare fruit oil likely results from (+)-fenchone and p-anisaldehyde. (+)-Fenchone was 20.3 times more abundant in the oil than p-anisaldehyde. (+)-Fenchone and p-anisaldehyde merit further study as potential house dust mite control agents or as lead compounds.  相似文献

In this study, antimicrobial activity of aminoreductone (AR), a product formed during the initial stage of the Maillard reaction, against methicillin-resistant Staphylococcus aureus (MRSA) was evaluated. The significant growth inhibition of all 51 MRSA isolates irrespective of drug susceptibility by AR was observed. The minimum inhibitory concentration (MIC) of AR ranged from 13 to 26 mM. The bactericidal activity of AR was evaluated by a killing assay with multiples of MIC, and it was recognized to depend on its dose. The combined effects of AR and antibiotics frequently used for the treatment of infections caused by Gram-positive and Gram-negative bacteria, such as amikacin (AN), ciprofloxacin, imipenem and levofloxacin, were examined. As a result, AR did not interfere with these antibiotic activities against 12 MRSA isolates selected and showed the advanced effect of growth inhibition in combination with antibiotics. Moreover, the inhibitory effects of AR were similar to those of AN, an antibiotic with known adverse effects, some serious. These findings show that AR is a naturally formed antimicrobial agent present in thermally processed foods with potential health benefits in medical practice.  相似文献

The phenolic constituents in the roots of Rheum officinale and Rubia cordifolia were identified with the aid of high-performance liquid chromatography and liquid chromatography-mass spectrometry and by comparison with authentic standards. A total of 17 hydroxyanthraquinones, gallic acid, and tannins were separated, and 14 of them were identified, being the main phenolic constituents present. Their antioxidant activity (Trolox equivalent antioxidant capacity) was evaluated using the improved 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt method. Hydroxyanthraquinones were the predominant antioxidant phenolic constituents in the roots of R. cordifolia, and tannins and gallic acid were the predominant antioxidant phenolic constituents in the roots of R. officinale. The structure-radical scavenging activity relationships of the tested hydroxyanthraquinones were systematically demonstrated as follows: Hydroxy groups on one benzene ring of the anthraquinone structure were essential for hydroxyanthraquinones to show activity, the ortho-dihydroxy structure in the hydroxyanthraquinone molecules could greatly enhance their radical scavenging effect, and glycosylation of the hydroxyanthraquinones reduced activity.  相似文献

The screening for bioactive principles from several Chinese medicinal herbs showed that the essential oil of Cymbopogon distans aerial parts possessed strong repellency against the booklouse, Liposcelis bostrychophila , and the red flour beetle, Tribolium castaneum . A total of 36 components of the essential oil were identified by GC and GC-MS. trans-Geraniol (16.54%), (R)-citronellal (15.44%), (+)-citronellol (11.51%), and α-elemol (9.06%) were the main components of the essential oil followed by β-eudesmol (5.71%) and (+)-limonene (5.05%). From the essential oil, four monoterpenes were isolated by bioassay-guided fractionation. The compounds were identified as limonene, citronellol, citronellal, and trans-geraniol. Geraniol and citronellol were strongly repellent against the booklouse, L. bostrychophila, whereas citronellal and limonene exhibited weak repellency against the booklouse. Geraniol and citronellol exhibited comparable repellency against the booklouse relative to the positive control, DEET. Moreover, geraniol and citronellol exhibited stronger repellency against the red flour beetle than DEET, whereas the two other compounds showed the same level of repellency against the red flour beetle compared with DEET.  相似文献

Sorgoleone, 2-hydroxy-5-methoxy-3-[(8'Z,11'Z)-8',11',14'-pentadecatriene]-p-benzoquinone (1), and its corresponding hydroquinone are the major components of the root exudate of Sorghum bicolor. The name sorgoleone includes minor analogues differing in the length or degree of unsaturation of the 3-alkyl side chain. These compounds are known to be phytotoxic, probably through inhibition of photosystem II (PSII) driven oxygen evolution, as previously demonstrated for 1. Isolation of these sorgoleone congeners was achieved by C(8) column chromatography and argentation thin-layer chromatography, and the purified compounds were structurally characterized. The abilities of the minor sorgoleones to inhibit PSII were similar to that of the major compound, suggesting that all of these sorgoleone congeners contribute to the overall allelopathy of sorghum.  相似文献

Radical scavenging activities of extracts and constituents in Cornus capitata adventitious root cultures were evaluated by using 1,1-diphenyl-2-pycrylhydrazyl (DPPH) and superoxide anion radicals. Inhibitory activity against peroxidation of linoleic acid was assayed by using the thiobarbituric acid (TBA) method. Ethyl acetate and aqueous fractions were prepared from adventitious roots cultured in Murashige-Skoog liquid medium with 0.1 microM Cu(2+) (0.1CuMS) or 10 microM Cu(2+) (10CuMS). The highest scavenging activities on DPPH and superoxide anion radicals were observed in the ethyl acetate fraction from 0.1CuMS. In the inhibitory activity against linoleic acid oxidation, the ethyl acetate fraction from 10CuMS was highest among the fractions tested. The ethyl acetate fraction of adventitious roots cultured in 0.1CuMS contained mainly galloylglucoses (1,2,3,6-tetragalloylglucose and 1,2,3,4,6-pentagalloylglucose). The ethyl acetate fraction of adventitious roots cultured in 10CuMS contained mainly ellagic acid derivatives [3,3'-di-O-methylellagic acid 4-(5"-acetyl)-alpha-L-arabinofuranoside and stenophyllin H1]. Aqueous fractions prepared from both media contained iridoid glycosides (dihydrocornin and cornin). Tetra- and pentagalloylglucoses showed strong inhibitory activities (61.9 and 85.2%, respectively) against linoleic acid oxidation relative to those of butylated hydroxytoluene (BHT) (91.1%) or alpha-tocopherol (49.5%) at 50 microM concentration. Although both ellagic acid derivatives had weak activities (<50%) on DPPH and superoxide anion radical scavenging, 3,3'-di-O-methylellagic acid 4-(5"-acetyl)-alpha-L-arabinofuranoside was stronger (74.7%) than alpha-tocopherol (49.5%) in inhibiting linoleic acid oxidation at 50 microM concentration. Iridoid glycosides exhibited little activity against DPPH and superoxide anion radicals or against oxidation of linoleic acid.  相似文献

This study evaluated the protective effects of purple rice ( Oryza sativa L.) bran extract (PRE) and its major anthocyanidins (cyanidin and peonidin) against light-induced retinal damage. In an in vitro experiment, cultured murine photoreceptor cells (661W) were damaged by a 24 h exposure to light. Viability of 661W after light treatment, assessed by the tetrazolium salt (WST-8) assay and Hoechst 33342 nuclear staining, was improved by the addition of PRE, cyanidin, and peonidin. Intracellular radical activation in 661W, evaluated using the reactive oxygen species (ROS) sensitive probe 5-(and 6)-chloromethyl-2,7-dichlorodihydrofluorescein diacetate acetyl ester (CM-H(2)DCFDA), was reduced by PRE and its anthocyanidins. Electron spin resonance (ESR) measurements showed that PRE, peonidin, and cyanidin all exhibited radical scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion radical ((?)O(2)(-)), and hydroxyl radical ((?)OH). In an in vivo mouse experiment, intravitreous injection of PRE significantly suppressed photoreceptor degeneration induced by exposure to light as revealed by histological analysis using hematoxylin-eosin staining. These findings suggest that PRE and its anthocyanidins possess protective effects with antioxidation mechanism in both in vitro and in vivo models of retinal diseases.  相似文献

The hydrodistilled essential oils of the aerial parts of wild-growing Salvia sclarea originated from two localities in Greece were analyzed by GC-MS. Sixty-six compounds, representing 93.26-98.19% of the oils, were identified. Linalyl acetate (19.75-31.05%), linalool (18.46-30.43%), geranyl acetate (4.45-12.1%), and alpha-terpineol (5.08-7.56%) were the main components. The antifungal activity of the oil of one locality and of the main components, linalyl acetate and linalool, was evaluated in vitro against three soil-borne pathogens.  相似文献

A tool for integrated pest management is the use of essential oils (EOs) and plant extracts. In this study, EOs from Tagetes lucida , Lepechinia betonicifolia , Lippia alba , Cananga odorata , and Rosmarinus officinalis , species grown in Colombia, were analyzed by gas chromatography-mass spectrometry. These oils as well as several of their constituents were tested for repellent activity against Tribolium castaneum , using the area preference method. The main components (>10%) found in EOs were methylchavicol, limonene/α-pinene, carvone/limonene, benzyl acetate/linalool/benzyl benzoate, and α-pinene, for T. lucida, L. betonicifolia, L. alba, C. odorata, and R. officinalis, respectively. All EOs were repellent, followed a dose-response relationship, and had bioactivity similar to or better than that of commercial compound IR3535. EOs from C. odorata and L. alba were the most active. Compounds from EOs, such benzyl benzoate, β-myrcene, and carvone, showed good repellent properties. In short, EOs from plants cultivated in Colombia are sources of repellents against T. castaneum.  相似文献

Essential oil of Haplopappus greenei A. Gray was obtained by hydrodistillation of aerial parts, which were subsequently analyzed by gas chromatography and gas chromatography-mass spectrometry. Major components were identified as carvacrol (8.7%), beta-pinene (7.6%), trans-pinocarveol (6.2%), and caryophyllene oxide (5.8%), respectively. In total, 104 components representing 84.9% of the investigated essential oil were characterized. Furthermore, the essential oil was evaluated for antimalarial, antimicrobial, and antifungal activities. However, only antifungal activity was observed against the strawberry anthracnose-causing fungal plant pathogens Colletotrichum acutatum, Colletotrichum fragariae, and Colletotrichum gloeosporioides using the direct overlay bioautography assay. Major essential oil components were also evaluated for antifungal activity; the carvacrol standard demonstrated nonselective activity against the three Colletotrichum species and the other compounds were inactive.  相似文献

One of the main olive oil phenolic compounds, hydroxytyrosol (3,4-DHPEA), exerts in vitro chemopreventive activities (antiproliferative and pro-apoptotic) on tumor cells through the accumulation of H(2)O(2) in the culture medium. However, the phenol composition of virgin olive oil is complex, and 3,4-DHPEA is present at low concentrations when compared to other secoiridoids. In this study, the in vitro chemopreventive activities of complex virgin olive oil phenolic extracts (VOO-PE, derived from the four Italian cultivars Nocellara del Belice, Coratina, Ogliarola, and Taggiasca) were compared to each other and related to the amount of the single phenolic constituents. A great chemopreventive potential among the different VOO-PE was found following this order: Ogliarola > Coratina > Nocellara > Taggiasca. The antiproliferative and pro-apoptotic activities of VOO-PE were positively correlated to the secoiridoid content and negatively correlated to the concentration of both phenyl alcohols and lignans. All extracts induced H(2)O(2) accumulation in the culture medium, but this phenomenon was not responsible for their pro-apoptotic activity. When tested in a complex mixture, the olive oil phenols exerted a more potent chemopreventive effect compared to the isolated compounds, and this effect could be due either to a synergistic action of components or to any other unidentified extract constituent.  相似文献

Broad-spectrum antimicrobial activity of a synthetic peptide, D4E1, is documented in this paper. D4E1 inhibited the growth of several fungal phytopathogens belonging to four classes-Ascomycetes, Basidiomycetes, Deuteromycetes, and Oomycetes, and two bacterial pathogens, Pseudomonas syringae pv. tabaci and Xanthomonas campestris pv. malvacearum race 18. The minimum inhibitory concentration (MIC) of D4E1 required to completely inhibit the growth of all fungi studied ranged from 4.67 to 25 microM. Fungal pathogens highly sensitive to D4E1 include Thielaviopsis basicola, Verticillium dahliae, Fusarium moniliforme, Phytophthora cinnamomi, and Phytophthora parasitica. Comparatively, the least sensitive fungal pathogens were Alternaria alternata, Colletotrichum destructivum, and Rhizoctonia solani. The two bacterial pathogens, P. syringae pv. tabaci and X. campestris pv. malvacearum race 18, were most sensitive to D4E1 with MIC values of 2.25 and 1.25 microM, respectively. Microscopic analysis of D4E1 effects on fungal morphology of Aspergillus flavus and R. solani revealed abnormal hyphal growth and discontinuous cytoplasm. After 8 h of exposure to 25 microM D4E1, A. flavus spore germination was reduced by 75%. The suitability of peptide D4E1 to enhance disease resistance in transgenic crop plants is discussed.  相似文献

Cinnamomum burmannii Blume (cinnamon stick) from Indonesia is a little-investigated spice. In this study, the antibacterial activity, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) of cinnamon stick extract were evaluated against five common foodborne pathogenic bacteria (Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, and Salmonella anatum). Cinnamon stick extract exhibited significant antibacterial properties. Major compounds in cinnamon stick were tentatively identified by gas chromatography-mass spectrometry (GC-MS) and liquid chromatography (LC-MS) as a predominant volatile oil component ((E)-cinnamaldehyde) and several polyphenols (mainly proanthocyanidins and (epi)catechins). Both (E)-cinnamaldehyde and proanthocyanidins significantly contributed to the antibacterial properties. Additionally, scanning electron microscopy was used to observe morphological changes of bacteria treated with the crude extract of cinnamon stick and its major components. This study suggests that cinnamon stick and its bioactive components have potential for application as natural food preservatives.  相似文献
